Multiciliated ependymal (E1) cells line the brain ventricles and are essential for brain homeostasis. We previously identified in the lateral ventricles a rare ependymal subpopulation (E2) with only two cilia and unique basal bodies. Here we show that E2 cells form a distinct biciliated epithelium extending along the ventral third into the fourth ventricle. In the third ventricle floor, apical profiles with only primary cilia define an additional uniciliated (E3) epithelium. E2 and E3 cells ultrastructure, marker expression and basal processes indicate that they correspond to subtypes of tanycytes. Using sonic hedgehog lineage tracing, we show that the third and fourth ventricle E2 and E3 epithelia originate from the anterior floor plate. E2 and E3 cells complete their differentiation 2-3 weeks after birth, suggesting a link to postnatal maturation. These data reveal discrete bands of E2 and E3 cells that may relay information from the CSF to underlying neural circuits along the ventral midline.

The ventricular-subventricular zone (V-SVZ) is an extensive germinal niche containing neural stem cells (NSCs) in the walls of the lateral ventricles of the adult brain. How the adult brains neural activity influences the behavior of adult NSCs remains largely unknown. We show that serotonergic (5HT) axons originating from a small group of neurons in the raphe form an extensive plexus on most of the ventricular walls. Electron microscopy revealed intimate contacts between 5HT axons and NSCs (B1) or ependymal cells (E1) and these cells were labeled by a transsynaptic viral tracer injected into the raphe. B1 cells express the 5HT receptors 2C and 5A. Electrophysiology showed that activation of these receptors in B1 cells induced small inward currents. Intraventricular infusion of 5HT2C agonist or antagonist increased or decreased V-SVZ proliferation, respectively. These results indicate that supraependymal 5HT axons directly interact with NSCs to regulate neurogenesis via 5HT2C.

Two neurogenic regions have been described in the adult brain, the lateral ventricle subventricular zone and the dentate gyrus subgranular zone. It has been suggested that neural stem cells also line the central canal of the adult spinal cord. Using transmission and scanning electron microscopy and immunostaining, we describe here the organization and cell types of the central canal epithelium in adult mice. The identity of dividing cells was determined by 3D ultrastructural reconstructions of [(3) H]thymidine-labeled cells and confocal analysis of bromodeoxyuridine labeling. The most common cell type lining the central canal had two long motile (9+2) cilia and was vimentin+, CD24+, FoxJ1+, Sox2+, and CD133+, but nestin- and glial fibrillary acidic protein (GFAP)-. These biciliated ependymal cells of the central canal (Ecc) resembled E2 cells of the lateral ventricles, but their basal bodies were different from those of E2 or E1 cells. Interestingly, we frequently found Ecc cells with two nuclei and four cilia, suggesting they are formed by incomplete cytokinesis or cell fusion. GFAP+ astrocytes with a single cilium and an orthogonally oriented centriole were also observed. The majority of dividing cells corresponded to biciliated Ecc cells. Central canal proliferation was most common during the active period of spinal cord growth. Pairs of labeled Ecc cells were observed within the central canal in adult mice 2.5 weeks post labeling. Our work suggests that the vast majority of postnatal dividing cells in the central canal are Ecc cells and their proliferation is associated with the growth of the spinal cord.

The subventricular zone of many adult non-human mammals generates large numbers of new neurons destined for the olfactory bulb. Along the walls of the lateral ventricles, immature neuronal progeny migrate in tangentially oriented chains that coalesce into a rostral migratory stream (RMS) connecting the subventricular zone to the olfactory bulb. The adult human subventricular zone, in contrast, contains a hypocellular gap layer separating the ependymal lining from a periventricular ribbon of astrocytes. Some of these subventricular zone astrocytes can function as neural stem cells in vitro, but their function in vivo remains controversial. An initial report found few subventricular zone proliferating cells and rare migrating immature neurons in the RMS of adult humans. In contrast, a subsequent study indicated robust proliferation and migration in the human subventricular zone and RMS. Here we find that the infant human subventricular zone and RMS contain an extensive corridor of migrating immature neurons before 18 months of age but, contrary to previous reports, this germinal activity subsides in older children and is nearly extinct by adulthood. Surprisingly, during this limited window of neurogenesis, not all new neurons in the human subventricular zone are destined for the olfactory bulb--we describe a major migratory pathway that targets the prefrontal cortex in humans. Together, these findings reveal robust streams of tangentially migrating immature neurons in human early postnatal subventricular zone and cortex. These pathways represent potential targets of neurological injuries affecting neonates.

Type 2 diabetes (T2D) is among the most common and costly disorders worldwide (1). The goal of current medical management for T2D is to transiently ameliorate hyperglycemia through daily dosing of one or more antidiabetic drugs. Hypoglycemia and weight gain are common side effects of therapy, and sustained disease remission is not obtainable with nonsurgical approaches. On the basis of the potent glucose-lowering response elicited by activation of brain fibroblast growth factor (FGF) receptors (2-4), we explored the antidiabetic efficacy of centrally administered FGF1, which, unlike other FGF peptides, activates all FGF receptor subtypes (5). We report that a single intracerebroventricular injection of FGF1 at a dose one-tenth of that needed for antidiabetic efficacy following peripheral injection induces sustained diabetes remission in both mouse and rat models of T2D. This antidiabetic effect is not secondary to weight loss, does not increase the risk of hypoglycemia, and involves a novel and incompletely understood mechanism for increasing glucose clearance from the bloodstream. - Abstract
Objective: This dissertation aims to learn about the neural networks involved with using deep brain stimulation (DBS) to treat Parkinson’s disease (PD) and increase our understanding of the mechanism of DBS. The particular focus is on examining network dynamics in the cortico-basal ganglia (BG) circuit and how DBS modulates functional connectivity in the cortico-BG circuit. The overarching hypothesis of this work is that DBS changes electrophysiological environments to alter the brain’s activity on a network level, examined here in the cortico-BG network in PD. Background: Deep brain stimulation (DBS) is the therapeutic use of chronic electrical stimulation of the brain via an implanted electrode and is a treatment for a variety of neurological disorders. The brain is an electrically active organ, containing neurons that communicate in circuits using action potentials (APs) and local field potentials (LFPs). We can infer information about how neurons communicate by studying these signals, how they behave in PD, and how they respond to DBS. PD is a neurodegenerative disease that manifests motor symptoms of bradykinesia, tremor, and rigidity, as well as a host of non-motor symptoms including cognitive impairment. It is characterized by a loss of dopamine (DA) neurons in the substantia nigra (SN), which has downstream effects on the function of the cortico-BG network. The cortico-BG network is an intricate web of connections between anatomical areas that regulates movement. DBS is used to treat the motor symptoms of PD with widespread success and has long-lasting efficacy. The DBS electrodes are placed into subcortical structures in the cortico-BG network, most commonly the subthalamic nucleus (STN) and the globus pallidus internus (GPi). 49. Epithelial organization of the adult neural stem cell niche

The adult mammalian brain, for all of its complexity, develops from a single cell layered epithelium of neural stem cells. These stem cells are retained in the epithelium lining the embryonic brain ventricles throughout development. Postnatally however, this germinal epithelium is replaced by postmitotic multiciliated ependymal cells. Neural stem cells are retained in the adult brain, but they are displaced from the ventricular epithelium and it is generally thought that they do not maintain epithelial properties. In this dissertation, I challenge this notion. Using a wholemount imaging technique that allows en-face examination of the ventricular walls, I show that the adult brain lateral ventricle is lined by a mixed neuroepithelium of multiciliated ependymal cells and adult neural stem cells (type B cells). Type B cells have very small ventricle-contacting apical surfaces observed only in neurogenic niches of the adult ventricular system. At their apical surface, B cells extend a primary cilium into the ventricle and are surrounded by ependymal cells, forming pinwheel structures that appear as repeating units across the neuroepithelium. Type B cells also contain a previously unidentified long basal process ending on blood vessels. These adult neural stem cells therefore display apical-basal polarity. En-face analysis of the ventricular walls also reveals a third cell type in the ventricular epithelium, which I call E2 cells. These cells are bi-ciliated with a novel basal body structure containing elaborate lateral extensions. Finally, in addition to apical-basal polarity, the adult ventricular epithelium has planar polarity evident in the polarized beating of ependymal cilia, which propels cerebrospinal fluid through the ventricular system and is essential to normal brain function. I describe two reliable anatomical correlates to ependymal planar polarity: the rotational orientation of basal bodies and their translational position on the apical surface of ependymal cells. I show that this planar polarity does not require ependymal cilia. Altogether, this work reveals the epithelial organization of the adult neural stem cell niche, characterized by the apical surfaces of stem cells surrounded by ependymal cells in unique pinwheel structures.
