1. Development of Limpet aquaculture: Advances on settlement and early growth of Patella aspera Röding, 1798

Abstract

The limpet Patella aspera is an endemic species to the Macaronesian Islands. Its high economic value and vulnerability to exploitation has triggered the interest in the development of its aquaculture. Significant progress has been made in recent years, with the establishment of artificial fertilization and larval production protocols. However, there are at least two major challenges that need to be addressed before a commercial production can be achieved: larval settlement and juvenile grow-out. The aim of this research was to study the effect of different algae substrates (Ulvella lens, Amphora sp., Entomoneis sp., Navicula salinicola and Nitzschia sp.) on P. aspera settlement and to test the effect of two diets on juvenile growth: a mono diet based on U. lens versus a mixed diet composed by U. lens and the diatoms Nitzschia sp. and Navicula salinicola. Results showed that U. lens was the most efficient settlement cue, given that 84.6% of the post-larvae obtained were found on this algae substrate. In the following trial, U. lens was selected as the base diet for the grow-out phase. The rearing system used ensured juvenile survival percentages above 80%. The regular supply of the diatom mixture, as a dietary supplement, significantly enhanced juvenile growth performance. After 220 days post fertilization, limpets that grew on the mixed diet (U. lens plus diatoms) presented an average shell length of 19.280 ± 5.296 mm. Additionally, developed gonads were observed in females with less than 8 months. This study presents the first record of P. aspera juvenile grow-out, born from artificial fertilization, which represents a major step toward the development of limpet aquaculture.

Abstract

The production of cultured limpets is a recent research field contributing to aquaculture diversification, focusing on low trophic species while reducing the carbon footprint. Limpets are gastropods that colonize rocky substrates and are mostly present on tidal and subtidal shores. This animal group is in high commercial demand and is endangered in several regions. The aquaculture production of limpets has been traditionally challenging. The most successful reproduction method has been gonadal dissection, as artificial spawning induction has shown limited success to date. Moreover, methods for larval culture, settlement, and juvenile growth have been poorly developed and remain largely unknown. In recent years, advances in this field have led to the optimization of methods to enhance larval production, larval culture, settlement induction of competent larvae, and management of post-larvae and juveniles. The present manuscript reviews these advances, obtained within the framework of AQUAINVERT project, focusing on broodstock management, gametes release, larval production, larviculture, settlement, and grow-out of post-larvae, and providing an update on the actual state of the art in limpets’ aquaculture.

Abstract

Megabalanus azoricus giant barnacles are the most traditional seafood of the Azores archipelago (NE Atlantic). This valuable commercial species has been highly exploited in the past and it is considered one of the key species for the development of aquaculture in the region. Despite the importance for conservation and aquaculture there is still a lack of basic information about M. azoricus genetic diversity and population structure. Here we used seven microsatellites markers to analyse 300 samples collected at six out of nine islands of the Azores archipelago, including also different locations from a single island, to provide information on the scale of genetic diversity and population structure of this species. Parameters like heterozygosity, allelic richness and effective number of alleles indicated a high genetic diversity and variability among islands. Pairwise comparisons and PCoA analysis on F ST and Jost's D EST showed significant and evident differentiation among sampling locations. Additionally, AMOVA allocates a small (6.02%) but statistically significant portion of the variance to the among Island level revealing also a weak resolution (1.87%) at finer scale. Additionally Monte Carlo resampling methods indicated the most likely sources of the recruits were the local or adjacent populations. Genetic risks associated with the giant barnacle potential production scheme should be taken into account in a future management plan delimiting, as precautionary measure, this culture at a single island or at groups of islands here identified. Moreover a monitoring strategy should be implemented with the aim to evaluate possible changes in genetic parameters of native populations.

Abstract

A series of experiments were conducted to obtain an efficient larval rearing protocol for Megabalanus azoricus. The first part of this study investigates the effect of microalgae-based diets on survival and larval development. Mono and mixed-diets were tested at 20 ± 1°C, in a sequence of 11-day feeding experiments. The second part presents a preliminary study on the influence of a biofilm on recruitment and use of oyster spat collectors in a mass rearing system. A photographic record of larval development and a brief reference to the diagnostic features that enable quick larval staging are also presented, along with morphometric measurements. Of the microalgae tested (Chaetoceros sp., Chloromonas sp., Dunaliella sp., T-Isochrysis sp. and Skeletonema sp.) the mixed-diet Skeletonema sp. with T-Isocrysis sp. showed the highest survival percentages: total survival ranged from 79.7 to 85.7% and 69.7–80.0% of nauplii were in stage VI after 11 days of rearing. Cypris were also present, but only represented 5.3% of the survivors at most. In the mass rearing system juveniles were found settled in the collectors after 25 days, at 20 ± 1°C. However recruitment was less than 1%. Preliminary results showed no settlement preference towards collectors with biofilm. Nevertheless, this study provides the first record of M. azoricus settlement under laboratorial conditions and represent a starting point for future larval rearing studies.

Abstract

Fish community structure of the Island Lampedusa, with a special focus on the fish assemblage inhabiting the infralittoral rocky substrate, was studied from July to October 1995. Visual census techniques (strip transects and random censuses) and the application of an anaesthetic (quinaldine), to sample cryptic and benthic species, were used. A total of 77 species and 29 fish families were censused. The most common families were Labridae and Spari‐dae. Factor analysis applied to the 84 visual assessments, using the 9 most common species, divided the samples according to their position along the coast, depth, substrate, algae cover and presence of the vascular plant, Posidonia oceanica. In addition, discriminant analyses indicated the ecological preferences of some species. Size frequency distributions indicated the distribution of different life stages. Despite the shallow depths sampled, the present study shows how the fish community of this island has high species richness

Abstract

Eleven polymorphic microsatellite loci were isolated in the passeriform Petronia petronia using the enrichment protocol FIASCO (fast isolation by AFLP of sequences containing repeats). We detected three to 13 alleles per locus in 25 specimens collected from an Italian population. The level of expected heterozygosity ranged from 0.439 to 0.856. One locus is sex linked to the Z chromosome. The total exclusionary probabilities using these loci for the first and the second parent were 0.978 and 0.999, respectively. These are the first microsatellite loci characterized from the rock sparrow that can be used for estimating population structure and for large-scale parentage analysis.

Abstract

There are approximately a dozen species of commercially interesting barnacles worldwide, some of which have been cultured on a semi-industrial scale. These species are listed and information is provided with regard to geographical distribution, landings and prices. Traditionally, 'goose' barnacles (four species) are considered to be the most important for consumption. World production already stands at 500 tonnes year-1, but this species has not been cultured to date. Some 'acorn' barnacles are also consumed (seven species), with harvest levels per species that do not exceed 200 tonnes year-1 and selling prices that can reach US$17/kg. 'Acorn' barnacle culture on a world scale is still developing. Nevertheless, production has occurred on a semi-industrial scale; specifically, spat have been collected from the wild and grown in suspended systems. Farming trials have focused on two species of acorn barnacles: Austromegabalanus psittacus (Molina 1782) 'picoroco' in Chile and Megabalanus azoricus (Pilsbry 1916) 'craca' in Portugal. The large-scale production of these crustaceans will depend on the optimization of spat collection from the wild and/or the parallel development of mass production technologies for larvae (hatcheries). In addition, further development will be achieved by opening up new markets for commercialization.

International audience; This article documents the addition of 234 microsatellite marker loci to the Molecular Ecology Resources Database. Loci were developed for the following species: Acipenser sinensis, Aleochara bilineata, Aleochara bipustulata, Barbus meridionalis, Colossoma macropomum, Delia radicum, Drosophila nigrosparsa, Fontainea picrosperma, Helianthemum cinereum, Liomys pictus, Megabalanus azoricus, Pelteobagrus vachelli, Pleuragramma antarcticum, Podarcis hispanica type 1A, Sardinella brasiliensis and Sclerotinia homoeocarpa. These loci were cross-tested on the following species: Acipenser dabryanus, Barbus balcanicus, Barbus barbus, Barbus cyclolepis, Drosophila hydei, Drosophila melanogaster, Drosophila obscura, Drosophila subobscura, Fontainea australis, Fontainea fugax, Fontainea oraria, Fontainea rostrata, Fontainea venosa, Podarcis bocagei, Podarcis carbonelli, Podarcis liolepis, Podarcis muralis and Podarcis vaucheri.
