Artificial intelligence (AI) models have received considerable attention in recent years for their ability to identify optical coherence tomography (OCT) biomarkers with clinical diagnostic potential and predict disease progression. This study aims to externally validate a deep learning (DL) algorithm by comparing its segmentation of retinal layers and fluid with a gold-standard method for manually adjusting the automatic segmentation of the Heidelberg Spectralis HRA + OCT software Version 6.16.8.0. A total of sixty OCT images of healthy subjects and patients with intermediate and exudative age-related macular degeneration (AMD) were included. A quantitative analysis of the retinal thickness and fluid area was performed, and the discrepancy between these methods was investigated. The results showed a moderate-to-strong correlation between the metrics extracted by both software types, in all the groups, and an overall near-perfect area overlap was observed, except for in the inner segment ellipsoid (ISE) layer. The DL system detected a significant difference in the outer retinal thickness across disease stages and accurately identified fluid in exudative cases. In more diseased eyes, there was significantly more disagreement between these methods. This DL system appears to be a reliable method for accessing important OCT biomarkers in AMD. However, further accuracy testing should be conducted to confirm its validity in real-world settings to ultimately aid ophthalmologists in OCT imaging management and guide timely treatment approaches. The existence of crime-related racial stereotypes has been well documented. People tend to associate certain groups with specific crimes, which, in turn, impacts criminal-sentencing decisions through the perceptions of crime severity. This evidence calls for regular updating of rating norms combining these variables. With this objective, and given that most of the normative studies provide norms for a small number of crimes and/or with an insufficient number of participants, a new norming study was conducted. Furthermore, norms from European countries are absent, and the existing ones (mostly with USA-based populations) do not simultaneously examine crime stereotypicality and crime severity. The Crime Stereotypicality and Severity Database (CriSSD) presents normative ratings for a set of 63 crimes on three dimensions: White stereotypicality, Black stereotypicality, and crime severity. The crimes were selected according to a comprehensive procedure. A total of 340 Portuguese participants (72.6% female; Mage = 26.86, SD = 7.65) answered an online survey. Each crime was evaluated by a range of 46–60 participants. Data allowed us to identify a crime typology with three clusters. We present descriptive data (means, standard deviations, and 95% confidence intervals) for each crime. Crime evaluations were associated with sociodemographic characteristics. Additionally, this study gives input regarding the understudied link between crime stereotypes and crime severity, showing that crime severity is predicted by ratings of both Black and White stereotypicality. Trypanosoma cruzi is a flagellated protozoan parasite that causes Chagas disease, which represents a serious health problem in the Americas. Nucleoside diphosphate kinases (NDPKs) are key enzymes that are implicated in cellular energy management. TcNDPK1 is the canonical isoform in the T. cruzi parasite. TcNDPK1 has a cytosolic, perinuclear and nuclear distribution. It is also found in non‐membrane‐bound filaments adjacent to the nucleus. In the present work, X‐ray diffraction and in vivo studies of TcNDPK1 are described. The structure reveals a novel, multi‐hexameric, left‐handed helical oligomer structure. The results of directed mutagenesis studies led to the conclusion that the microscopic TcNDPK1 granules observed in vivo in T. cruzi parasites are made up by the association of TcNDPK1 oligomers. In the absence of experimental data, analysis of the interactions in the X‐ray structure of the TcNDPK1 oligomer suggests the probable assembly and disassembly steps: dimerization, assembly of the hexamer as a trimer of dimers, hexamer association to generate the left‐handed helical oligomer structure and finally oligomer association in a parallel manner to form the microscopic TcNDPK1 filaments that are observed in vivo in T. cruzi parasites. Oligomer disassembly takes place on the binding of substrate in the active site of TcNDPK1, leading to dissociation of the hexamers. This study constitutes the first report of such a protein arrangement, which has never previously been seen for any protein or NDPK. Further studies are needed to determine its physiological role. On the onset of the COVID-19 pandemic, the overcrowding in prisons led to efforts to decarcerate in order to prevent and control outbreaks in prisons. This study analyses how public support for such exceptional measures are determined by cognitive and ideological factors known to create and maintain racial biases in the criminal system. Participants were asked to express their level of agreement with the early-release of hypothetical prisoners. Results showed participants to be less favourable to the early-release of Black compared to White prisoners, when they had committed a stereotypically Black crime. As expected, the congruency between the crime stereotypicality and the colour of the prisoner's skin did not emerge for White prisoners. Moreover, the difference between the agreement with the release of the Black vs. the White prisoner when both committed a stereotypically Black crime was higher as the level of endorsement of Meritocracy increased. Contrastingly, Anti-egalitarianism only predicted an overall disagreement with prisoners' early-release. This paper highlights the cumulative explanation by different levels of analysis of this current problem and implications for the development of the public opinion on penal subjects. The behavior of microplastic particles (MPPs) is explored in a lab-scale direct contact membrane distillation (DCMD) system used to produce drinking water from seawater, while also analyzing the impact of their presence on the process performance. Commercial MPPs (LDPE, PET and uPVC) were first studied by temperature stress (TS) tests (using amber bottles) to mimic the exposure to the high temperatures commonly used in DCMD (ca. 80 °C), which led to uPVC being selected to be tested under different loads and aging degrees in the DCMD system. By analyzing uPVC MPPs samples before and after the TS and DCMD experiments by ATR-FTIR, UV–Visible spectroscopy and SEM, it was concluded that minor aging is expected to occur. High loads (>0.1 g L−1) of uPVC MPPs decreased the DCMD performance in terms of permeate flux (from 32.8 ± 0.3 with filtered seawater to 28.7 ± 0.3 kg m−2 h−1 with a load of 0.2 g L−1), although no signs of inferior treated water quality were found based on the conductivity and salinity results. The removal of MPPs (size > 1.2 μm) in DCMD applications with seawater was analyzed by μRaman, with all the results suggesting very high removal efficiencies (≥99 %). This paper presents an eye tracking study on the reading process of high school students while answering questions from the Enem-2017 Spanish Language Test. The research was aimed at identifying reading strategies and sources of difficulty of two groups of participants when taking the test. Group A (GA) had more previous study time in the Spanish Language and group B (GB) had less previous study time. The analyses considered eye movements (monitored during the task), accuracy and time of performance. Eye tracking data demonstrated that in both groups the preferred reading path is from the text to the multiple-choice alternatives and that textual complexity seems to affect the number and duration of fixations. There was a significant difference between the two groups, with more hits for GA than for GB. Therefore, the time devoted to studying the language seems to affect the performance of the participants in the reading comprehension. [ABSTRACT FROM AUTHOR]

Prolonged mechanical unloading in bedridden patients and concurrent hormonal dysregulation represents the cause of one of the severest forms of osteoporosis, a condition for which there are very few efficacious interventions available to date. Sclerostin, a Wnt antagonist, acts as a negative regulator of bone formation. Sclerostin antibody (Scl‐Ab)–mediated blockade of sclerostin can dramatically enhance bone formation and reduce bone resorption. This study was designed to investigate the therapeutic effect of the Scl‐Ab on severe bone loss induced by concurrent mechanical unloading and estrogen deficiency in a hindlimb‐suspended and ovariectomized rat model, and to study the cellular mechanisms underlying severe osteoporosis and Scl‐Ab action. Unloading and ovariectomy resulted in severe loss of trabecular and cortical bone mass and strength; Scl‐Ab can significantly counteract the deterioration of bone in unloaded and/or ovariectomized rats, with noticeably increased cortical bone formation. Scanning electron microscopy analysis revealed that unloading and ovariectomy lead to multiple morphological and structural abnormalities of osteocytes in cortical bone and the abnormalities were abolished by Scl‐Ab administration. This study extends our previous conclusion that Scl‐Ab represents a promising therapeutic approach for severe bone loss that occurs after being exposed to estrogen deficiency and prolonged mechanical unloading. A five-years-old boy was referred to the emergency department due to a 12- hour evolution of skin lesions similar to generalized varicella and concomitant herpes zoster in the C6 dermatome. Without history of prior disease or vaccination against varicella-zoster virus. There was an outbreak of varicella in nursery school. He was treated with oral acyclovir, with complete resolution. To our knowledge, it is the first published case of concomitant varicella and herpes zoster due to wild virus. There are only four similar cases described in the international literature, all related to vaccination against varicellazoster, which is not present in this case. The mechanism responsible for the simultaneous manifestation of varicella and herpes zoster is not known, however a theory under discussion is that the initial viremia during primary infection may cause immediate viral replication in the sensory ganglia, causing cutaneous manifestations and pain in the surrounding dermatomes. Trypanosoma cruzi, the etiological agent of Chagas disease, uses proline as its main carbon source, essential for parasite growth and stage differentiation in epimastigotes and amastigotes. Since proline is mainly obtained from extracellular medium by transport proteins, in this work we studied the regulation of the T. cruzi proline transporter TcAAAP069. Proline uptake and intracellular concentration presented oscillations during epimastigote growth phases, increasing during the early exponential phase (322 pmol/min) and decreasing to undetectable levels during the late exponential phase. Transporter expression rate correlated with proline uptake, and its subcellular localization alternated from both, the plasma membrane and close to the flagellar pocket, when the transport is higher, to only the flagellar pocket region, when the transport decreased until proline uptake and TcAAAP069 protein became undetectable at the end of the growth curve. Interestingly, when parasites were treated with conditioned medium or were concentrated to artificially increase the culture density, the proline transport was completely abolished resembling the effects observed in late exponential phase. Phytomonas are protozoan parasites from the Trypanosomatidae family which infect a wide variety of plants. Herein, Phytomonas Jma was tested as a model for functional expression of heterologous proteins. Green fluorescent protein expression was evaluated in Phytomonas and compared with Trypanosoma cruzi , the etiological agent of Chagas’ disease. Phytomonas was able to express GFP at levels similar to T. cruzi although the transgenic selection time was higher. It was possible to establish an efficient transfection and selection protocol for protein expression. These results demonstrate that Phytomonas can be a good model for functional expression of proteins from other trypanosomatids, presenting the advantage of being completely safe for humans. Tumor cells' differential mechanisms to get energy can be diverse, as occurs in certain tumors of high biological aggressiveness, causing varying degrees of impairment in nutritional status. For the identification of malnutrition, physical, dietary, social, subjective, anthropometric, laboratory and clinical functional parameters can be applied, providing information that will enable a better assessment of nutritional status (NS). Currently researches are using the Phase Angle (PA°) and the Standardized Phase Angle (SPA) as nutritional indicators of NS and severity of the disease. However, there are few studies evaluating these parameters across different types of malignancy. This study compared the averages of PA° and SPA in lung (LC), head and neck (HNC) and colorectal cancer (CRC), crossing them with anthropometric parameters, tumor stage, and the Karnofsky Performance Status scale (KPS). The total sample comprised 276 patients, 20% (n=55) LC, 64% (n=177) HNC and 16 % (n=44) CRC. The highest percentage of severe weight loss was observed in the group of LC, followed by patients with CRC. The mean arm muscle circumference (F (2; 67), p = 0.017) and arm muscle area (F (2.59), p = 0.036) differ between men with different malignancies, as do the average resistance (p = 0.005) and reactance (F (2;214) p = 0.02). In women there was difference between the means of the AFP, (F (2; 56) p = 0.033) in different malignancies. Patients with HNC and CCR with KPS ≤ 70% had a lower average SPA (p = 0.016). The study of humanness as a dimension of social judgment has received extensive attention over the past decade. Although the common reported finding is that people attribute more human characteristics to their ingroup than to the outgroup, similar tendencies are expected to be tempered for minority groups when judging the host society. In Study 1, carried out with Gypsy minority members, we tested the hypothesis that those group members who adopt an assimilative strategy identifying more with the host compared with the heritage culture will display the lowest levels of dehumanisation. In Studies 2 and 3, conducted with immigrants in Italy and in Portugal, respectively, the hypothesis was extended from an identification conceptualisation to an acculturation one. Despite significant variability in intergroup settings and measures, results confirmed our hypothesis that immigrants who choose to assimilate with the host culture dehumanise the outgroup less compared with those who adopt any of the other acculturation strategies. Trypanosoma cruzi, the etiological agent of Chagas' disease, has a metabolism largely based on the consumption of glucose and proline. This amino acid is essential for host cells infection and intracellular differentiation. In this work we identified a proline transporter (TcAAAP069) by yeasts complementation assays and overexpression in Trypanosoma cruzi epimastigotes. TcAAAP069 is mono-specific for proline but presents an unusual feature; the lack of stereospecificity, because it is competitively inhibited by the D- enantiomer. Parasites overexpressing TcAAAP069 have an increased intracellular proline concentration, 2.6-fold higher than controls, as a consequence of a higher proline transport rate. Furthermore, augmented proline concentration correlates with an improved resistance to trypanocidal drugs and also to reactive oxygen species including hydrogen peroxide and nitric oxide, emulating natural physiological situations. The IC50s for nifurtimox, benznidazole, H2O2 and NO. were 125%, 68%, 44% and 112% higher than controls, respectively. Finally, proline metabolism generates a higher concentration (48%) of ATP in TcAAAP069 parasites. Since proline participates on essential energy pathways, stress and drug resistance responses, these results provide a novel target for the development of new drugs for the treatments for Chagas' disease. Trypanosomatids' amino acid permeases are key proteins in parasite metabolism since they participate in the adaptation of parasites to different environments. Here, we report that TcAAP3, a member of a Trypanosoma cruzi multigene family of permeases, is a bona fide arginine transporter. Most higher eukaryotic cells incorporate cationic amino acids through a single transporter. In contrast, T. cruzi can recognize and transport cationic amino acids by mono-specific permeases since a 100-fold molar excess of lysine could not affect the arginine transport in parasites that over-express the arginine permease (TcAAP3 epimastigotes). In order to test if the permease activity regulates downstream processes of the arginine metabolism, the expression of the single T. cruzi enzyme that uses arginine as substrate, arginine kinase, was evaluated in TcAAP3 epimastigotes. In this parasite model, intracellular arginine concentration increases 4-folds and ATP level remains constant until cultures reach the stationary phase of growth, with decreases of about 6-folds in respect to the controls. Interestingly, Western Blot analysis demonstrated that arginine kinase is significantly down-regulated during the stationary phase of growth in TcAAP3 epimastigotes. This decrease could represent a compensatory mechanism for the increase in ATP consumption as a consequence of the displacement of the reaction equilibrium of arginine kinase, when the intracellular arginine concentration augments and the glucose from the medium is exhausted. Using immunofluorescence techniques we also determined that TcAAP3 and the specific lysine transporter TcAAP7 co-localize in a specialized region of the plasma membrane named flagellar pocket, staining a single locus close to the flagellar pocket collar. Taken together these data suggest that arginine transport is closely related to arginine metabolism and cell energy balance. Abstract: Nucleoside diphosphate kinases (NDPKs) are enzymes required to preserve the intracellular nucleoside phosphate equilibrium. Trypanosoma cruzi has four putative nucleoside diphosphate kinases with unidentified biological roles and subcellular localization. TcNDPK2 has an N-terminal domain (DM10) with unknown function, which defines a subgroup of NDPKs distributed in a wide variety of organisms. Digitonin extraction demonstrated that this isoform is distributed in detergent soluble and insoluble fractions. Fluorescence microscopy showed that TcNDPK2 alone or fused to GFP was localized in cytoskeleton and flagella. TcNDPK2 was also detected by Western blot in purified polymerized tubulin and flagellar samples. In parasites expressing DM10 fused with GFP, the fluorescence was localized in cytoskeleton and flagellum with an identical pattern to TcNDPK2. This constitutes the first report that could give insights on the role of DM10 domains in NDPKs and also the identification of the first T. cruzi peptide that contains a microtubule association domain. Abstract: Nucleoside diphosphate kinases (NDPKs) are multifunctional enzymes involved mainly in the conservation of nucleotides and deoxynucleotides at intracellular levels. Here we report the characterization of two NDPKs from the protozoan parasite Trypanosoma cruzi, the etiological agent of Chagas disease. TcNDPK1 and TcNDPK2 were biochemically characterized presenting different kinetic parameters and regulation mechanisms. NDPK activity was mainly detected in soluble fractions according to the digitonin extraction technique; however 20% of the activity remains insoluble at digitonin concentrations up to 5mgml−1. TcNDPK1 is a short enzyme isoform, whereas TcNDPK2 is a long one containing a DM10 motif. In addition, two other putative NDPK genes (TcNPDK3 and TcNDPK4) were detected by data mining at the T. cruzi genome database. The large number and diversity of NDPK isoforms are in agreement with those previously observed for other T. cruzi phosphotransferases, such as adenylate kinases. [Copyright &y& Elsevier]

Abstract: Trypanosoma cruzi arginine kinase is a key enzyme in cell energy management and is also involved in pH and nutritional stress response mechanisms. T. cruzi epimastigotes treated with hydrogen peroxide presented a time-dependent increase in arginine kinase expression, up to 10-fold, when compared with untreated parasites. Among other oxidative stress-generating compounds tested, only nifurtimox produced more than 2-fold increase in arginine kinase expression. Moreover, parasites overexpressing arginine kinase showed significantly increased survival capability during hydrogen peroxide exposure. These findings suggest the participation of arginine kinase in oxidative stress response systems. [Copyright &y& Elsevier]

Abstract: Adenylate kinases supply energy routes in cellular energetic homeostasis. In this work, we identified and characterized the adenylate kinase activity in extracts from the flagellated parasite Trypanosoma cruzi, the causative agent of Chagas'' disease. Adenylate kinase activity was detected in different subcellular fractions and the cytosolic isoform was biochemically characterized. Cytosolic adenylate kinase specific activity increases continuously during the epimastigote growth and is down-regulated when other soluble phosphotransferase, arginine kinase, is overexpressed. Six different genes of adenylate kinase isoforms were identified and the mRNA expression was confirmed by RT-PCR and Northern Blot. Three open reading frames coding for different enzyme isoforms named TzADK1, TzADK2 and TzADK5 were cloned and functionally expressed in E. coli. This work reports an unusually large number of genes of adenylate kinases and suggests a coordinated regulation of phosphotransferase-mediated ATP regenerating pathways in the unicellular parasite Trypanosoma cruzi. • A DPSIR analysis is presented on the topic of microplastics pollution. • EU regulatory and policy instruments concerning microplastics are identified. • Current and developing upstream and downstream responses are reviewed. This review organizes key information about microplastic pollution through a DPSIR (driving forces, pressures, states, impacts and responses) analysis, namely the current knowledge on the sources of microplastics in the environment, the abundance, mobility and fate of microplastics distributed across the different environmental compartments, as well as their socio-economic and environmental impacts. The available or developing upstream and downstream responses to the microplastic pollution are also reviewed as part of the DPSIR analysis. These include the regulatory and policy instruments, environmental education campaigns, product design, the development of biodegradable plastics, environmental cleanups, waste management, drinking water and wastewater treatment plants, and other treatment technologies and processes. Whenever possible, the current trends and discerning gaps in the research conducted so far by the scientific community are identified, giving some clues to what is going to be the future research on this topic and into new lines of research. Objectives: to calculate the prevalence and rate per 1,000 live births of sexually transmitted infections (STI) in pregnant women at a public maternity hospital in Salvador. Methods: this descriptive, cross-sectional study retrospectively collected data from compulsory notifications and medical records of pregnant women with STI seen at a maternity hospital in northeastern Brazil between 2014 and 2017 (n = 520). Prevalence and rate per 1,000 live births were estimated for hepatitis B, hepatitis C, HIV, and syphilis. Associations between STI and other clinical and sociodemographic variables were investigated. Results: most pregnant women were born and resided in Salvador, presented a mean age of 26.4 years, self-reported mixed-race and had unplanned pregnancies. Prevalence and rates per 1,000 live births were, respectively: 0.26% and 3.39 for hepatitis B, 0.06% and 0.79 for hepatitis C, 0.47% and 6.23 for HIV, and 2.46% and 32.2 for syphilis. Conclusion: higher prevalence and rates of infection per 1,000 live births were seen at the maternity hospital in northeastern Brazil compared to official data provided by the Brazilian government, notably with regard to HIV and syphilis. The appropriate epidemiological notification of STI, especially in pregnant women, enables the elaboration of effective preventive strategies incorporating specific sociodemographic and clinical characteristics. The Family Adaptability and Cohesion Evaluation Scale, developed by in the framework of the Circumplex Model of Marital and Family Systems, assess family functioning through a relational diagnosis. This instrument is subdivided into six subscales, allowing an assessment of the balanced and dysfunctional levels (extremely low and extremely high) of the dimensions of family cohesion and flexibility. The FACES IV Package also includes two scales that evaluate family communication and satisfaction. The present article analyzes the psychometric properties of the instruments included in the FACES IV Package with a sample of Portuguese adolescents in two studies. In the first study, we included a community group of 757 students and a clinical group of 67 adolescents attending psychiatric appointments. The results of this study revealed a factorial model of 24 items with satisfactory psychometric qualities, showing an acceptable adjustment to our sample. In the second study, the factorial solution obtained in the previous study was replicated with an independent sample of 707 young students. Vaccines are essential for the eradication of diseases. Yet for many reasons, individuals do not embrace them completely. In the COVID‐19 pandemic and with the possibility of the Brazilian population's immunization against the disease, both political and health‐related dimensions might have had a role in individual COVID‐19 vaccination acceptance. In two studies (n = 974), we tested the hypothesis that participants' vaccination acceptance is related to their past vote in the 2018 Brazilian presidential election (being or not being a Jair Bolsonaro voter) and their different levels of perceived vulnerability to disease (PVD). We further tested whether Bolsonaro's opposition or ambiguous messages towards vaccination (vs. control) increased vaccination rejection among those who have (vs. have not) voted for him and who are low (vs. high) in PVD. Results show that Bolsonaro (vs. non‐Bolsonaro) voters accepted less vaccination, with higher rejection rates when participants expressed low (vs. high) PVD. Also, when primed either with Bolsonaro's opposed or ambiguous messages towards COVID‐19 vaccination, such participants accepted less vaccines (vs. participants primed with neutral information). These findings are the first to show that the COVID‐19 vaccine acceptance is related to their past vote and leadership influence but also different levels of perceived vulnerability to disease. In two studies (n = 974), we tested the hypothesis that participants' vaccination acceptance is related to their past vote in the 2018 Brazilian presidential election (being or not being a Jair Bolsonaro voter) and their different levels of perceived vulnerability to disease (PVD). We further tested whether Bolsonaro's opposition or ambiguous messages towards vaccination (vs. control) increased vaccination rejection among those who have (vs. have not) voted for him and who are low (vs. high) in PVD. Results show that Bolsonaro (vs. non‐Bolsonaro) voters accepted less vaccination, with higher rejection rates when participants expressed low (vs. high) PVD. Also, when primed either with Bolsonaro's opposed or ambiguous messages towards COVID‐19 vaccination, such participants accepted less vaccines (vs. participants primed with neutral information). These findings are the first to show that the COVID‐19 vaccine acceptance is related to their past vote and leadership influence but also different levels of perceived vulnerability to disease. Vaccinia virus (VACV) is a notorious virus for a number of scientific reasons; however, most of its notoriety comes from the fact that it was used as a vaccine against smallpox, being ultimately responsible for the eradication of that disease. Nonetheless, many different vaccinia virus strains have been obtained over the years; some are suitable to be used as vaccines, whereas others are virulent and unsuitable for this purpose. Interestingly, different vaccinia virus strains elicit different immune responses in vivo, and this is a direct result of the genomic differences among strains. In order to evaluate the net result of virus-encoded immune evasion strategies of vaccinia viruses, we compared antiviral immune responses in mice intranasally infected by the highly attenuated and nonreplicative MVA strain, the attenuated and replicative Lister strain, or the virulent WR strain. Overall, cell responses elicited upon WR infections are downmodulated compared to those elicited by MVA and Lister infections, especially in determined cell compartments such as macrophages/monocytes and CD4+ T cells. CD4+ T cells are not only diminished in WR-infected mice but also less activated, as evaluated by the expression of costimulatory molecules such as CD25, CD212, and CD28 and by the production of cytokines, including tumor necrosis factor alpha (TNF-α), gamma interferon (IFN-γ), interleukin-4 (IL-4), and IL-10. On the other hand, MVA infections are able to induce strong T-cell responses in mice, whereas Lister infections consistently induced responses that were intermediary between those induced by WR and MVA. Polyamines play critical roles as regulators of cell growth and differentiation. In contrast with other protozoa, the human parasite Trypanosoma cruzi , the etiological agent of Chagas disease, is auxotrophic for polyamines. Therefore, their intracellular availability depends exclusively on polyamine transport and inhibition of these uptake processes can alter the viability of the parasite. The polyamine analogues used in this work were successfully tested as antiproliferative agents in cancer cells, bacteria, fungi and also showed a potent antiplasmodial effect. We evaluated the activity of these compounds on polyamine transport in T. cruzi and assessed the effects on parasite viability. Three polyamine derivatives, AMXT1501, Ant4 and Ant44, inhibited the putrescine transport in epimastigotes (the insect stage of T. cruzi ) with calculated IC 50 values of 2.43, 5.02 and 3.98 μM, respectively. In addition, only Ant4 and Ant44 inhibited spermidine transport with IC 50 of 8.78 μM and 13.34 μM, respectively. The Ant4 analogue showed a high trypanocidal effect on trypomastigotes (the bloodstream stage of T. cruzi ) with an IC 50 of 460 nM, (SI = 12.7) while in epimastigotes the IC 50 was significantly higher (16.97 μM). In addition, we studied the effect of the combination of benznidazole, a drug used in treating Chagas disease, with Ant4 on the viability of epimastigotes. The combined treatment produced a significant increase on the inhibition of parasites growth compared with individual treatments. In summary, these results suggest that Ant4, a putrescine conjugate, is a promising compound for the treatment of Chagas disease because it showed a potent trypanocidal effect via its inhibition of polyamine import. The Serra das Éguas Complex, in Brumado, state of Bahia, located in the northeastern region of Brazil, is a geological structure comprising volcanochemical, siliciclastic and carbonate units that contain layers and lenses of magnesite, important as a refractory in steel, cement, glass and copper and other applications. Considering the importance of such sort of deposit, this work aims to (i) study the physical properties of magnesite deposits via direct measurements of radiometric spectrometry and magnetic susceptibility associated with chemical analysis of the available borehole core samples; (ii) analyse the magnetic and radiometric data acquired by the combined airborne survey over the area with known magnesite deposits; (iii) estimate the magnetic response of the magnesite deposits and surrounding rock formations via 2D forward and 3D inverse magnetic modelling; (iv) evaluate and, if necessary, reconsider the stratigraphic sequences of the identified geological units of the Complex using the results of the 2D and 3D magnetic modelling. Through forward/inverse models, we noted an expressive magnetic signature likely related to the iron formations and metamafic and meta‐ultramafic rocks. The radiometric data analysis on the drill cores showed that the reddish magnesite has a well‐defined U/Th signature. Furthermore, the results show that the carbonate unit, which contains the magnesite deposits, has the lowest amplitude of magnetization values, and both signatures might be helpful for their identification. Additionally, the magnetic models show that the carbonate unit lies in the upper part of the stratigraphic sequence of the Serra das Éguas Complex. This conclusion drives us to propose a new stratigraphy sequence, where the volcanochemical unit that includes the iron formations lies at the bottom of this stratigraphic sequence. Trypanosoma cruzi is the etiological agent of Chagas disease, a major health problem in Latin America. Polyamines are polycationic compounds that play a critical role as regulators of cell growth and differentiation. In contrast with other protozoa, T. cruzi is auxotrophic for polyamines because of its inability to synthesize putrescine due to the lack of both, arginine and ornithine decarboxylase; therefore, the intracellular availability of polyamines depends exclusively on transport processes. In this work, the polyamine transporter TcPAT12 was overexpressed in T. cruzi epimastigotes demonstrating that growth rates at different concentrations of polyamines strongly depend on the regulation of the polyamine transport. In addition, parasites overexpressing TcPAT12 showed a highly increased resistance to hydrogen peroxide and the trypanocidal drugs nifurtimox and benznidazole, which act by oxidative stress and interfering the synthesis of polyamine derivatives, respectively. Finally, the presence of putative polyamine transporters was analyzed in T. cruzi, Trypanosoma brucei, and Leishmania major genomes identifying 3-6 genes in these trypanosomatids. Belief in a Just World research found evidence that one feels threatened whenever one witnesses an innocent victim suffering, often resorting to secondary victimization to neutralize the observed injustice. However, literature has neglected the explanatory power of adolescent deviant behavior in victimization processes. This study (n = 284 students) aims to determine the impact of the adolescents' deviant behavior, BJW and victim's innocence on secondary victimization. Additionally, we analyzed juvenile deviant behavior's impact on victim identification. Juveniles who committed more deviant behaviors identified less with the victim than those with lower deviance levels. The interaction effects show that juveniles who are strong just world believers and have higher delinquency engaged significantly more in secondary victimization when confronted with an innocent victim. These results clarify the role played by adolescent deviant behavior and BJW in secondary victimization judgments regarding situations with innocent and non-innocent victims. [ABSTRACT FROM AUTHOR]

Undergraduate students experience a form of circadian misalignment – known as “social jetlag” – that represents the discrepancy in timing between their circadian and social clocks. Whilst social jetlag is not dependent upon chronotype, the two phenomena tend to be related; evening types show a tendency to have a greater social jetlag, for example. Moreover, evening types have been found to be more likely to have inadequate eating habits than do morning types. The objective of this study was to examine the relationship between chronotype, social jetlag, perceived sleep debt and dietary intake in Brazilian undergraduate students. The chronotype was derived from mid-sleep time on free days (MSF) at the weekend. Social jetlag was calculated as the absolute difference between mid-sleep time on weekdays and weekends. Perceived sleep debt was calculated using the difference between students’ preferred weekday sleep duration and their self-reported actual weekday sleep duration. Correlations were found between chronotype and breakfast time (r= 0.24,p= 0.003) and lunch time (r= 0.19,p= 0.01). Multiple regression analyses showed that chronotype was positively associated with consumption of meat (β= 0.21;p= 0.003); social jetlag was negatively associated with consumption of beans (β= –0.16;p= 0.02) and perceived sleep debt was positively associated with consumption of beverages (β= 0.15;p= 0.02) and dairy products (β= 0.17;p= 0.01) and negatively associated with consumption of cereals and pasta (β= –0.16;p= 0.02). It is concluded that, in undergraduate students, chronotype (MSF), social jetlag and perceived sleep debt can influence the type and amount of some food groups consumed at mealtimes. Objective: The purpose of the study was to develop a novel diet based on standard AIN93G diet that would be able to induce experimental obesity and impair immune regulation with high concentrations of both carbohydrate and lipids. Methods: To compare the effects of this high sugar and butter (HSB) diet with other modified diets, male C57BL/6 mice were fed either mouse chow, or AIN93G diet, or high sugar (HS) diet, or high-fat (HF) diet, or high sugar and butter (HSB) diet for 11 weeks ad libitum. HSB diet induced higher weight gain. Therefore, control AIN93G and HSB groups were chosen for additional analysis. Regulatory T cells were studied by flow cytometry, and cytokine levels were measured by ELISA. Results: Although HF and HSB diets were able to induce a higher weight gain compatible with obesity in treated mice, HSB-fed mice presented the higher levels of serum glucose after fasting and the lowest frequency of regulatory T cells in adipose tissue. In addition, mice that were fed HSB diet presented higher levels of cholesterol and triglycerides, hyperleptinemia, increased resistin and leptin levels as well as reduced adiponectin serum levels. Importantly, we found increased frequency of CD4CD44 effector T cells, reduction of CD4CD25Foxp3 and Th3 regulatory T cells as well as decreased levels of IL-10 and TGF-β in adipose tissue of HSB-fed mice. Conclusion: Therefore, HSB represents a novel model of obesity-inducing diet that was efficient in triggering alterations compatible with metabolic syndrome as well as impairment in immune regulatory parameters. Adenylate kinases (ADK) are key enzymes involved in cell energy management. Trypanosomatids present the highest number of variants in a single cell in comparison with the rest of the living organisms. In this work, we characterized two flagellar ADKs from Trypanosoma cruzi, called TcADK1 and TcADK4, which are also located in the cell cytosol. Interestingly, TcADK1 presents a stage-specific expression. This variant was detected in epimastigotes cells, and was completely absent in trypomastigotes and amastigotes, while TcADK4 is present in the major life cycle stages of T. cruzi. Both variants are also regulated, in opposite ways, along the parasite growth curve suggesting that their expression depends on the intra- and extracellular conditions. Both, TcADK1 and TcADK4 present N-terminal extension that could be responsible for their subcellular localization. Twenty-four dairy cows, with daily average milk production of 18.1 kg, were fed diets containing different selenium (Se) sources. The purpose of this paper is to evaluate the effects of such diets on milk production and quality, on the occurrence of mastitis, and on physiological variables. During the experimental period (124 days), all the cows received the same diet: a total mixed feed with 0.278 mg.kg- 1 DM of selenium. In the inorganic Se treatment, the selenium source was sodium selenite and in the organic Se treatment the source was selenium yeast (Saccharomyces cerevisiae CNCM I-3060). There were no significant differences in milk yield or in Se concentration in the milk. No significant differences between the treatments were observed in protein, lactose, solids-not-fat andpercentage of total solids. The animals subjected to the organic Se treatment presented higher (P=0.013) percentage of milk fat and lower (P=0.014) somatic cell count (SCC) than those subjected to the inorganic Se treatment. There was no significant difference in subclinical mastitisas determined by the California Mastitis Test (CMT) between the treatments. However, both Se sources reduced the incidence of mastitis (subclinical positive mastitis and strongly positive mastitis) between the pre-experimental and experimental phases. There was no significant difference in rectal temperature (RT) between the treatments. Respiratory frequency (RF) was lower (P=0.027) in the inorganic treatment than in the organic one, whereas haircoat temperature (HT) was lower (P=0.007) in the organic treatment than in the inorganic one. Staphylococcus aureus is among the main etiologic agents of bovine mastitis. A total of 83 isolates of S. aureus from mammary glands of primiparous heifers were collected in the prepartum, calving and during lactation. For lactating cows, a total of 27 isolates of S. aureus from mammary glands were collected during lactation. The samples were taken in two dairy farms located in Sao Paulo State, Brazil. The highest frequency of S. aureus isolation in heifers was at the end of lactation. Strains were typified through Pulsed-field gel electrophoresis (PFGE) and grouped according to patterns of restriction enzyme SmaI. PFGE generated seven clonal profiles that were grouped into three different lineages, with the LA lineage being predominant and identified in heifers, as well as in the cows from the two regions studied. It was concluded that the cows showed a significant source of dispersion of S. aureus. At the first lactation the heifers were infected by the same clonal profiles of S. aureus which were isolated from multiparous lactating cows. Abstract: Adenylate kinases are key enzymes involved in cell energy management. Trypanosomatid organisms have the largest number of isoforms found in a single cell, constituting a major difference with the mammalian hosts. In this work we study an adenylate kinase, TcADK3, the only Trypanosoma cruzi protein harboring the putative peroxisomal (glycosomal) targeting signal, “-CKL”. Parasites expressing GFP fused to TcADK3 showed a strong fluorescence in the glycosomes. The same result was obtained when the tripeptide “-CKL” was added at the C-terminus of the GFP, demonstrating that this signal is necessary and sufficient for targeting proteins to glycosomes. When this tripeptide was removed from the GFP-TcADK3 fusion protein, the fluorescence was re-localized in the cytoplasm. The CKL signal could be used for targeting foreign proteins to the glycosomes. This model also provides a useful tool to study glycosomes dynamics, morphology or number in living parasites in any stage of the life cycle. [Copyright &y& Elsevier]

Abstract: Phytomonas are trypanosomatid plant parasites closely related to parasites that cause several human diseases. Little is known about the biology of these organisms including aspects of their metabolism. Arginine kinase (E.C. 2.7.3.3) is a phosphotransferase which catalyzes the interconversion between the phosphagen phosphoarginine and ATP. This enzyme is present in some invertebrates and is a homolog of another widely distributed phosphosphagen kinase, creatine kinase. In this work, a single canonical arginine kinase isoform was detected in Phytomonas Jma by enzymatic activity assays, PCR, and Western Blot. This arginine kinase is very similar to the canonical isoforms found in T. cruzi and T. brucei, presenting about 70% of amino acid sequence identity and a very similar molecular weight (40kDa). The Phytomonas phosphagen system seems to be very similar to T. cruzi, which has only one isoform, or T. brucei (three isoforms); establishing a difference with other trypanosomatids, such as Leishmania, which completely lacks phosphagen kinases, probably by the presence of the arginine-consuming enzyme, arginase. Finally, phylogenetic analysis suggests that Kinetoplastids'' arginine kinase was acquired, during evolution, from the arthropod vectors by horizontal gene transfer. [Copyright &y& Elsevier]

The purpose of this paper was to pre-test a set of characteristics associated with the moral dimension. For this reason, 102 characteristics associated with this dimension were evaluated in two dimensions of evaluation, desirability and uniquely humanness, by a sample of 106 university students. The means, medians, standard deviations, median absolute deviation and confidence intervals of 95% are presented for each trait in each dimension of evaluation. As are the uni-versus bi-modality of each distribution. The results allow the selection of the traits for desirability or distribution in the uniquely human dimension. We were able to verify a narrower distribution of the morality characteristics in the uniquely human dimension, contrary to what happens on the desirability dimension. [ABSTRACT FROM AUTHOR]

l-Cysteine and methionine are unique amino acids that act as sulfur donors in all organisms. In the specific case of Trypanosomatids,l-cysteine is particularly relevant as a substrate in the synthesis of trypanothione. Although it can be synthesized de novo,l-cysteine is actively transported in Trypanosoma cruzi epimastigote cells.l-Cysteine uptake is highly specific; none of the amino acids assayed yield significant differences in terms of transport rates.l-Cysteine is transported by epimastigote cells with a calculated apparent Km of 49.5 μM and a Vmax of about 13 pmol min−1 per 107 cells. This transport is finely regulated by amino acid starvation, extracellular pH, and between the parasite growth phases. In addition,l-cysteine is incorporated post-translationally into proteins, suggesting its role in iron–sulfur core formation. Finally, the metabolic fates ofl-cysteine were predicted in silico.
