In recent years, nanoparticles, especially carbon dots (CDs), exhibit many unprecedented applications for drug delivery. Several types of CDs and CD-ligand conjugates have been reported successfully penetrating the BBB, which shows a promising progress in the application of CD-based drug delivery system (DDS) for the treatment of CNS diseases. In this review, our discussion of CDs includes their classification, preparations, structures, properties, and applications for the treatment of neurodegenerative diseases, especially Alzheimer’s disease (AD) and brain tumor. Moreover, abundant functional groups on the surface, especially amine and carboxyl groups, allow CDs to conjugate with diverse drugs as versatile drug nanocarriers. In addition, structure of the BBB is briefly described, and mechanisms for transporting various molecules across the BBB and other biological barriers are elucidated. Most importantly, recent developments in drug delivery with CDs as BBB-penetrating nanodrugs and drug nanocarriers to target CNS diseases especially Alzheimer’s disease and brain tumor are summarized. 25. Development of Red-Emissive Carbon Dots for Bioimaging through a Building Block Approach: Fundamental and Applied Studies

Abstract

In recent years, many researchers have struggled to obtain carbon dots (CDs) that possess strong photoluminescence in the red region of light. Success in this area has been limited, although the past few years have brought several promising reports on this topic. The most successful efforts in this area still seem to struggle from a lack of dispersibility/reduced emission in water. This work endeavors to understand the formation process of CDs that do not possess strong performance in an aqueous environment and to improve their capabilities in bioimaging. o-Phenylenediamine (o-PDA) is used along with various precursors in several different solvents (varying acidic and oxidative strengths) to understand the formation process behind the structure leading to red emission that is sensitive to water. These results showed that the combination of acid properties and oxidation is essential for this process, and the important reactions are oligomerization of o-PDA and the crosslinking of these oligomers to form aromatic structural segments of CDs. These CDs are shown to be capable of quantitatively detecting water in organic solvents. Additionally, we have shown that conjugation with transferrin remarkably enhances the biocompatibility of these CDs. Transferrin-conjugated CDs with better biocompatibility were applied to bioimaging studies of neuroblastoma cell lines with N-mycand non-N-mycgene amplification, for the first time. Furthermore, CDs showed versatile bioimaging capability toward a highly aggressive neuroblastoma subgroup of tumors. The importance of creating red-emissive CDs has been well established, and this work is an important step toward understanding their formation and realizing their use in biological systems.

32. Optimization of carbon dots' emission wavelength for biological applications

Abstract

Carbon quantum dots (CDs) are a type of nanomaterial discovered in the first years of the 21st century. They have been widely studied due to their easily modified surface, bright and stable photoluminescence, and low toxicity. Due to these advantageous properties, CDs have been applied in fields such as drug delivery, bioimaging, light emitting diodes, and 3D-printing. One drawback of the use of CDs in biological systems is the normal blue or green wavelength of emission. Any attempt to track or image these particles through their emission in vivo is difficult as the CDsu2019 photoluminescence is reabsorbed by the tissue or interfered with through autofluorescence. To improve CDsu2019 capabilities, it would be highly advantageous to shift their emission to the red region of light (ca. 700 nm). Herein, is presented several strategies to accomplish this purpose. CDs have been prepared from a dye (Evans blue) and protein (bovine serum albumin) complex, through reaction in sulfuric acid and other acidic and oxidative species, and by coordination with lanthanide metals. These reactions all endeavor to produce CDs which possess red photoluminescence in aqueous solutions. From the results of these reactions, a great deal can be learned regarding how pyrolysis breaks down the structure of both dye and protein to produce CDs and how this impacts emission wavelength. Furthermore, the cooperative nature of acidic and oxidative properties has been observed in the preparation of CDs which possess red emission, but only in organic solvents. These preparation pathways present an opportunity to obtain CDsu2019 desired properties as well as understand the underlying principles relating to their formation and photoluminescence mechanism.

Abstract

A type of orange carbon dots (O-CDs) synthesized via an ultrasonication route with citric acid and 1,2-phenylenediamine as precursors was embedded into sodium polyacrylate (SPA) as the ink for 3D printing. Characterizations of these spherical O-CDs revealed an ultra-small size (~2 nm) and excitation-independent, but solvent dependent, emission. The O-CDs were evenly distributed with low degree of aggregation in sodium polyacrylate (SPA), which was achieved due to the property that SPA can absorb water together with O-CDs. The 3D printed photoluminescent objective with the ink revealed a great potential for high yield application of these materials for additive manufacturing. This also represents the first time, bare CDs have been reported as a photoluminescent material in 3D printing, as well as the first time SPA has been reported as a material for 3D printing.
