Abstract

This study aimed to evaluate the effects of ciplukan (Physalis angulata L.) leaf ethanol extract on L-NG-nitroarginine methyl ester (L-NAME)-induced hypertensive rats. We randomly divided twenty-five Wistar rats into five groups. The sham group was given a PBS injection. The hypertensive group was injected with L-NAME on days 1 to 28. Three groups of hypertensive rats were given the extract on days 4 to 28. Blood pressure was measured using the tail-cuff method on days 0, 4, 10, and 27. The endothelial progenitor cells (EPCs) in the blood were measured by flow cytometry as a percentage of circulating angiogenic cells (CACs, CD34+/CD309+/CD45+) and endothelial colony-forming cells (ECFCs, CD34+/CD309+/CD45-). Serum NO and MDA levels, as well as serum SOD activity, were measured colorimetrically. Serum TNF-a levels were measured by the ELISA method. The ciplukan leaf extract reduced systolic and diastolic blood pressure, reduced the percentage of EPCs in the blood, increased serum NO levels, reduced MDA levels, increased serum SOD activity, and reduced serum TNF-a levels in L-NAME-induced hypertensive rats. It is concluded that ciplukan ethanol leaf extract exerts protective effects on L-NAME-induced hypertensive rats. Abstract

Malaria infection in pregnant women can cause various clinical manifestations, one of them is fetus low birth weight. Sequestrations of infected erythrocytes and macrophages on intervillous space has been revealed by previous study, however the mechanism of this sequestration in placenta insufficiency is still unknown. We suggested that local immunity on placenta tissue can induce the production of reactive oxygen species (ROS) and reactive nitrogen species (RNS) that can cause the destructions of placenta tissue due to the fetus low birth weight. This research was conducted to explore the path of immunity response on malaria placenta which involved the expression of inducible nitric oxide synthase (iNOS) and the level of malondialdehyde (MDA). This experimental laboratory study was done in pregnant Balb/c strain mice that infected withPlasmodium bergheiin 9thday of pregnancy. Abstract

Non-streoidal anti-inflammatory drugs (NSAIDs) are commonly used despite their known gastrointestinal adverse effects. Centella asiatica (CA) is widely used for its anti-inflammatory due to its active components. This study was conducted to investigate the anti-inflammatory effects of CA ethanolic (CAE) extracts in gastric ulcer by measuring COX-2 expression. The effects of CAE were identified in mice (Rattus norvegicus) used as peptic ulcer models induced with Indometachin at 30 mg/kg BW. The experimental groups were divided into five groups: negative control group, positive control group day 8, and the treatment groups receiving three different doses of CAE (100, 200, and 400 mg/kg BW). CAE was administered once daily for seven days. The experimental models were sacrificed 24 hours after the last CAE administration, and COX-2 expression in the gastric mucosal layer was assessed using immunohistochemistry. Hematoxylin-eosin (HE) staining was used to examine gastric mucosal damage. There was a consistent significant decrease in COX-2 expression in all treatment groups compared to control groups (p = 0.006). The results showed that CAE displayed anti-inflammatory effect. CAE extracts could decrease COX-2 expression in all three treatment groups compared to control models. CAE has a potential to be anti-inflammatory herbal therapy for gastric ulcers. Abstract

Nonsteroidal Anti-inflammatory Drugs (NSAID) prescriptions have increased over the past decade to treat inflammatory diseases. Indomethacin is a non-selective NSAID that can cause gastric mucosal damage and often leads to acute gastric ulcers. TNF-α is a proinflammatory cytokine overproduced in gastric ulcers. Centella asiatica is a herb commonly found in Indonesia that have anti-inflammatory effects. This study aimed to determine the effects of Centella asiatica ethanolic extracts (CAE) on TNF-α expression in indomethacin-induced gastric ulcer model. This study had six experimental groups: negative control (C), positive control day-1 (P1), positive control day-8 (P8), treatment group 1 (CAE 100), 2 (CAE 200), and 3 (CAE 400). P1, P8, CAE 100, CAE 200, and CAE 400 groups were given 30 mg/kg BW of Indomethacin orally to induced gastric ulcer. Treatment groups were given CAE 8-hours following indomethacin administration at a dose of 100, 200, and 400 mg/kg BW. Gastric mucosal damage was examined with HE staining and TNF-α expression evaluated by immunohistochemical staining. Indomethacin has proven caused acute gastric ulcers in the histopathological evaluation and significantly upregulated TNF-α expression (p = 0.003). CAE has proven to have anti- inflammatory effects by downregulating TNF-α expression significantly in all treatment groups with the optimum dose at 100 mg/kg BW. [ABSTRACT FROM AUTHOR]

Abstract

Gastric ulcer is a disease that commonly occurs and spreads over the world, and one of the most common causes is the use of long-term non-steroid anti-inflammatory drugs (NSAIDs). Active compounds of bark and leaves of the cinnamon extract are cinnamaldehyde and eugenol, which are acting as gastroprotective by antioxidant and antiinflammatory properties and also flavonoids such as quercetin, kaempferol, and quercetrin as an antioxidant. This study aimed to determine the effect of cinnamon extract on the expression of SOD-1 and TNF-α in indomethacin-induced gastric ulcers. Indomethacin 30 mg/kg BW orally induced after 8 hour fasting period. Cinnamon extract administered in different doses daily for two days. The rats divided into s6 groups : nnegative control group, positive control group on first day, positive control group on third day, and three treatment groups (T1, T2, and T3) with cinnamon extract doses of 400, 200 and 100 mg/kg BW respectively. The level of SOD-1 measured by ELISA and the expression of TNF-α by immunohistochemistry from gastric mucosa. The results showed that cinnamon extract significantly increased SOD-1 and decrease of TNF-α. Histopathology analysis showed the improvement of gastric epithelial after cinnamon provision. These results demonstrate that the administration of cinnamon extract may protect indomethacin-induced gastric ulcer rats through their antioxidant and anti-inflammatory properties, and also it is improving gastric histopathology. Abstract

Objectives This study evaluated the protective effect of Physalis angulata leaf extract on L-NG-nitroarginine methyl ester (L-NAME)-induced preeclampsia rats. Methods Twenty-five pregnant Wistar rats were randomly divided into five groups. Preeclampsia rats were injected with L-NAME on the 5th to 17th day of gestation (G5-G17), while sham rats were injected with the same vehicle volume. Preeclampsia rats were administered with Physalis angulata leaf methanol extract at 500, 1500, or 2500 mg/kg body weight/day on G1-G17. The systolic blood pressure measurement was performed by the tail-cuff method. Placental VEGFR2 expression was determined by immunohistochemistry. The serum sFlt-1 level was evaluated using ELISA. Serum MDA level, SOD activity, and total nitrite level were measured by colorimetry. Results The L-NAME injection increased systolic blood pressure (1.44-fold), serum sFlt-1 level (1.22-fold), and serum MDA level (2.03-fold). It also decreased placental VEGFR2 expression (1.18-fold), serum SOD activity (2.11-fold), and serum nitrite level (1.25-fold) compared to the Sham group. The extract supplementation prevented the rise in systolic blood pressure and serum sFlt-1 level. It also tended to inhibit the reduction in the placental VEGFR2 expression. It prevented the increase in serum MDA level. It also prevented the decrease in serum SOD activity and nitrite level. Conclusions We concluded that Physalis angulata leaf methanol extract inhibits oxidative stress and an antiangiogenic factor in the rat preeclampsia model. Abstract

Stroke is a medical emergency that the patients of it ideally should have received stroke management <3 hours since the attack. Emergency situations can occur at anytime, thus the health cadres participation in minimizing pre-hospital delay through FAST method is needed. In order to make the cadres have knowledge, positive attitudes, and good practices about FAST, the rational model-based training was conducted. The aim of this study was to analyze the effectiveness of rational model-based training on the health cadres' changes on knowledge, attitudes, and practices in the stroke symptoms detection using FAST. This study used quasi-experimental research design through pretest-posttest control group design. 50 respondents were divided into intervention and control groups. Data analysis used independent sample t-test and mann whitney test. The results show that there were differences in attitude and practices changes between the intervention and the control groups after training, with p values 0.009 and 0.000. There was no any difference in the retention of knowledge, attitudes, and practices in both groups, the p value was 0.849; 0.626;0.456. [ABSTRACT FROM AUTHOR]
