Pumping surface water from a ditch into a denitrifying woodchip bioreactor could improve nitrate-nitrogen (N) removal by minimizing flow variabilities such as early flow cessation at a given subsurface drainage outlet and flashy drainage hydrographs. Few field-scale subsurface drainage bioreactors with pumping configurations have been assessed. Such evaluations would help better bound reasonable expectations of the benefits and drawbacks at these more advanced bioreactors. An underloaded "ditch diversion" bioreactor constructed in 2018 in Illinois, USA (LW: 4.6 × 9.1 m), was retrofitted with a solar-powered pumping system in 2021 and was then monitored for a 29-d period in 2022 and a 93-d period in 2023. The pumped bioreactor achieved N removal rates averaging 7.5 and 5.2 g N/m 3 -d and N removal efficiencies of 50 and 61% for the monitoring periods in 2022 and 2023, respectively. Pumping generally improved the bioreactor's performance compared to the same monitoring windows from the historic (non-pumped) 2019-2021 periods. Regression analysis indicated the addition of a pump slightly improved N load removal compared to what might be expected for a conventional bioreactor. The somewhat unintended diurnal batch mode operation resulting from the solar-powered pumping system boosted water temperature in the overnight batches. Adoption of edge-of-field conservation practices, such as denitrifying bioreactors, may be intrinsically linked to barriers associated with cost. However, most previous bioreactor cost efficiency assessments assumed values for either costs and/or nitrate removal. The objective of this work was to use actual construction costs as well as monitored nitrate removal to develop empirical cost efficiencies for eight full-size bioreactors in Illinois, USA. Capital construction costs were obtained via invoices or personal communications. A cash-flow discounting procedure was used to develop an equal annualized cost for each bioreactor assuming two media recharges over a 24-y planning horizon. These costs were combined with monitored nitrate removal based on one to six years of monitoring per site. Construction costs averaged $12,250 ± $7520 across the eight sites (or, $16,020 ± $9960 in 2023 price levels) but considering one of the sites was a paired bioreactor system, costs averaged $10,890 per bioreactor unit. Drainage treatment area-based cost averaged $132/ha-y and treatment area was strongly correlated with capital costs (R 2  = 0.90; p = 0.001). The bioreactors averaged $108/m 3 of woodchips and available federal government conservation programs could have offset an average of 70% of this cost. Monitored nitrate removal across 27 site-years resulted in a median of $33/kg N-y removed. This mass-based cost efficiency was higher than most previous assessments because the monitored nitrate removal for the study sites was lower than has been previously assumed or modeled. Future reporting about bioreactor recharge timing and cost will help guide assessment and planning. Water quality planning efforts should also consider the increasingly important engineering design costs, which were not included here. Fiorinia externa Ferris, elongate hemlock scale, was inadvertently introduced to North America from Japan. This insect is particularly problematic on hemlock, Tsuga spp., though it has been reported in association with several other conifers. The evidence that other conifers might be hosts, capable of supporting growing populations of the insect, has not been previously reviewed. Our review confirms that F. externa is an oligophagous pest of members of Pinaceae. Although species of Cupressaceae and Taxaceae have been reported as hosts of F. externa , they seem unable to support population growth of this pest. Evidence of the tree-killing potential of the insect, even on suitable hosts, is remarkably scant. Edge-of-field practices such as denitrifying woodchip bioreactors can be used to improve the water quality of agricultural effluents. This study evaluated the effectiveness of four field-scale woodchip bioreactors in removing nitrate‑nitrogen (nitrate-N) from subsurface drainage in eastern South Dakota. Four woodchip bioreactors were installed and monitored between 2014 and 2016 near Arlington, Baltic, Hartford, and Montrose, South Dakota. Results showed that reduction in nitrate-N concentration for the four bioreactors ranged from 7 % to 100 %, corresponding to removal rates of 5 to 27 g N/m 3 /day for the four bioreactors during the study period. Average Nitrate-N load reduction in the four bioreactors studied ranged from 39 % to 89 % during the study period. Reduction of nitrate-N in the four bioreactors decreased, on average, by 30 % when temperature dropped below 12 °C during the study period. Knowledge of a species' phenology can assist with timing accurate surveys to detect presence and density in a novel environment. Trichoferus campestris (Faldermann) (Coleoptera: Cerambycidae) has been found in the United States since the 1990s, but its biology and behavior remain poorly understood. This study investigated the phenology of T. campestris in Minneapolis and St. Paul, Minnesota, USA, based on local temperature data. In the summers of 2019 and 2020, 30 and 40 pheromone-baited traps, respectively, were deployed in trees in suburban parks to monitor capture of local individuals. Traps were suspended from branches of mature trees selected at random, with nine genera of trees selected over both years. Early, peak, and late adult abundance were characterized, and the impact of tree genus in which each trap was hung was evaluated. Abundance was found to be unimodal both summers with a peak around 650 degree days (base 10°C) in early July. Significantly more adult T. campestris were caught in traps hung in trees of the genus Tilia than in trees of the genus Quercus. This paper describes a multi-site and multi-decadal dataset of artificially drained agricultural fields in seven Midwest states and North Carolina, USA. Thirty-nine research sites provided data on three conservation practices for cropland with subsurface tile drainage: saturated buffers, controlled drainage, and drainage water recycling. These practices utilize vegetation and/or infrastructure to minimize off-site nutrient losses and retain water in the landscape. A total of 219 variables are reported, including 90 field measurement variables and 129 management operations and metadata. Key measurements include subsurface drain flow (206 site-years), nitrate-N load (154 site-years) and other water quality metrics, as well as agronomic, soil, climate, farm management and metadata records. Data are published at the USDA National Agricultural Library Ag Data Commons repository and are also available through an interactive website at Iowa State University. Background: Vitamin A test results have historically been notorious for poor repeatability and reproducibility. This problem has been discussed at length in Association of American Feed Control Officials Laboratory Methods and Services Committee meetings., Objective: The objective of this work was to assess the effect of test portion mass on the repeatability of vitamin A test results., Methods: The study was conducted in two parts. In Part I, fundamental sampling error (FSE) was determined experimentally through replicated (n = 16) vitamin A testing of three animal feed materials. The testing followed rigorous test portion selection for 10 g and 100 g test portions. In Part II, FSE calculations were made (1) using theoretical equations based on vitamin A as a liberated analyte and (2) on representing the particles in feed materials. Particle size characterization of vitamin A ingredients was estimated by microscopy and further evaluated by particle size analysis., Results: RSDs, % for vitamin A determinations ranged from 10.5-24.7, and 2.26-10.7 for 10 g and 100 g test portions, respectively. FSE calculated for Ingredient A ranged from 18.3-101% and 5.79-32.0% for 10 g and 100 g test portions, respectively, and for Ingredient B, ranged from 10.2-56.2% and 3.21-17.8% for 10 g and 100 g test portions, respectively., Conclusion: Test portion mass has a substantial impact on FSE and is an important factor in controlling the random error in vitamin A testing. Background: Norovirus is the etiology for about 60% of foodborne outbreaks identified in Minnesota. Contamination of food during preparation by food handlers is by far the most common cause of these outbreaks. Norovirus outbreaks due to commercially distributed foods are rarely reported in the United States, and only 2 have been previously identified in Minnesota, both due to oysters., Methods: In August 2016, we investigated an outbreak of norovirus gastroenteritis in Minnesota that was linked to consumption of commercially distributed ice cream at multiple venues. Sanitarians from local public health agencies visited the facilities involved for follow-up, and case-control studies were conducted. The outbreak was identified by linking multiple independent illness reports to a centralized foodborne illness complaint system and subsequently confirmed though genotyping of stool specimens., Results: A total of 15 cases from 4 venues were reported. Raspberry chocolate chip ice cream was statistically associated with illness in 2 analytic studies (6 of 7 cases vs 0 of 7 controls; odds ratio, undefined; P = .005). Norovirus GII.17[P17] (GII.17 Kawasaki) strains from case stool specimens matched norovirus found in frozen raspberries imported from China that were used to make the implicated ice cream., Conclusions: To our knowledge, this is the first norovirus outbreak due to commercially distributed frozen berries identified in the United States. Nontarget data acquisition for target analysis (nDATA) workflows using liquid chromatography-high-resolution accurate mass (LC-HRAM) spectrometry, spectral screening software, and a compound database have generated interest because of their potential for screening of pesticides in foods. However, these procedures and particularly the instrument processing software need to be thoroughly evaluated before implementation in routine analysis. In this work, 25 laboratories participated in a collaborative study to evaluate an nDATA workflow on high moisture produce (apple, banana, broccoli, carrot, grape, lettuce, orange, potato, strawberry, and tomato). Samples were extracted in each laboratory by quick, easy, cheap, effective, rugged, and safe (QuEChERS), and data were acquired by ultrahigh-performance liquid chromatography (UHPLC) coupled to a high-resolution quadrupole Orbitrap (QOrbitrap) or quadrupole time-of-flight (QTOF) mass spectrometer operating in full-scan mass spectrometry (MS) data-independent tandem mass spectrometry (LC-FS MS/DIA MS/MS) acquisition mode. The nDATA workflow was evaluated using a restricted compound database with 51 pesticides and vendor processing software. Pesticide identifications were determined by retention time ( t R ) of the precursor (RTP, δ M ) of the precursor (RTP, δ M ≤ ±10 ppm). The elution profiles of all 51 pesticides were within ±0.5 min among 24 of the participating laboratories. Successful screening was determined by false positive and false negative rates of <5% in unfortified (pesticide-free) and fortified (10 and 100 μg/kg) produce matrices. Pesticide responses were dependent on the pesticide, matrix, and instrument. The false negative rates were 0.7 and 0.1% at 10 and 100 μg/kg, respectively, and the false positive rate was 1.1% from results of the participating LC-HRAM platforms. Further evaluation was achieved by providing produce samples spiked with pesticides at concentrations blinded to the laboratories. Twenty-two of the 25 laboratories were successful in identifying all fortified pesticides (0-7 pesticides ranging from 5 to 50 μg/kg) for each produce sample (99.7% detection rate). These studies provide convincing evidence that the nDATA comprehensive approach broadens the screening capabilities of pesticide analyses and provide a platform with the potential to be easily extended to a larger number of other chemical residues and contaminants in foods.M ≤ ±10 ppm). Cactodera torreyanae Cid del Prado Vera & Subbotin, 2014 cysts were discovered during a Pale Potato Cyst Nematode (PCN) survey conducted by Minnesota Department of Agriculture as part of the Animal and Plant Health Inspection Service (APHIS) efforts to survey states for the presence of PCN. The soil samples were collected from a potato field, located in Karlstad, Kittson County, Minnesota, USA. Two out of 175 vials submitted for identification to the Mycology and Nematology Genetic Diversity and Biology Laboratory (MNGDBL) contained few cysts and juveniles of C. torreyanae . Cysts were dark brown in color, lemon-shaped to elongated with distinct vulval cone. Vulva with denticles present around fenestra, cyst length to width ratio between 1.6 and 2.3 and anus distinct. The juveniles had rounded stylet knobs, some sloping slightly posteriorly. The molecular analysis included sequence and phylogenetic analysis of ITS rRNA, D2-D3 expansion segments of 28S rRNA and COI of mtDNA genes. The nematode species was identified by both morphological and molecular means as Cactodera torreyanae . To the best of our knowledge this represents the first report of Cactodera torreyanae from the United States and first report of this cyst nematode species from potato fields. In 2019, Cactodera milleri cysts were discovered from soil samples collected from a Chenopodium quinoa field, located in Mosca, Alamosa county, Colorado, USA. Approximately 200 lemon shaped cysts and several hundred juveniles were recovered from the affected quinoa plants. The same species was also identified from several counties in Minnesota from samples submitted over the years by the Minnesota Department of Agriculture as part of the Animal and Plant Health Inspection Service (APHIS) efforts to survey states for the presence of Pale Potato Cyst Nematode. The cysts and juveniles (J2) were recovered from soil samples through sieving and Baermann funnel extraction. The nematode species was identified by both morphological and molecular means as Cactodera milleri (Graney and Bird, 1990). Addressing critical global health issues, such as antimicrobial resistance, infectious disease outbreaks, and natural disasters, requires strong coordination and management across sectors. The One Health approach is the integrative effort of multiple sectors working to attain optimal health for people, animals, and the environment, and is increasingly recognized by experts as a means to address complex challenges. However, practical application of the One Health approach has been challenging. The One Health Systems Mapping and Analysis Resource Toolkit (OH-SMART) introduced in this paper was designed using a multistage prototyping process to support systematic improvement in multi-sectoral coordination and collaboration to better address complex health concerns through an operational, stepwise, and practical One Health approach. To date, OH-SMART has been used to strengthen One Health systems in 17 countries and has been deployed to revise emergency response frameworks, improve antimicrobial resistance national action plans and create multi agency infectious disease collaboration protocols. The verification of remotely sensed estimates of surface variables is essential for any remote sensing study. The objective of this study was to compare leaf area index (LAI), surface temperature (Ts), and actual evapotranspiration (ETa), estimated using the remote sensing-based METRIC model and in situ measurements collected at the satellite overpass time. The study was carried out at a commercial corn field in eastern South Dakota. Six clear-sky images from Landsat 7 and Landsat 8 (Path 29, Row 29) were processed and used for the assessment. LAI and Ts were measured in situ, and ETa was estimated using an atmometer and independent crop coefficients. The results revealed good agreement between the variables measured in situ and estimated by the METRIC model. LAI showed r 2 = 0.76, and RMSE = 0.59 m 2 m -2 , the Ts comparison had an agreement of r 2 = 0.87 and RMSE 1.24 °C, and ETa presented r 2 = 0.89 and RMSE = 0.71 mm day -1 .

Foodborne non-typhoidal salmonellosis causes approximately 1 million illnesses annually in the USA. In April 2015, we investigated a multistate outbreak of 65 Salmonella Paratyphi B variant L(+) tartrate(+) infections associated with frozen raw tuna imported from Indonesia, which was consumed raw in sushi. Forty-six (92%) of 50 case-patients interviewed ate sushi during the week before illness onset, and 44 (98%) of 45 who specified ate sushi containing raw tuna. Two outbreak strains were isolated from the samples of frozen raw tuna. Traceback identified a single importer as a common source of tuna consumed by case-patients; this importer issued three voluntary recalls of tuna sourced from one Indonesian processor. Four Salmonella Weltevreden infections were also linked to this outbreak. Whole-genome sequencing was useful in establishing a link between Salmonella isolated from ill people and tuna. This outbreak highlights the continuing foodborne illness risk associated with raw seafood consumption, the importance of processing seafood in a manner that minimises contamination with pathogenic microorganisms and the continuing need to ensure imported foods are safe to eat. People at higher risk for foodborne illness should not consume undercooked animal products, such as raw seafood.

Whole apples have not been previously implicated in outbreaks of foodborne bacterial illness. We investigated a nationwide listeriosis outbreak associated with caramel apples. We defined an outbreak-associated case as an infection with one or both of two outbreak strains of Listeria monocytogenes highly related by whole-genome multilocus sequence typing (wgMLST) from 1 October 2014 to 1 February 2015. Single-interviewer open-ended interviews identified the source. Outbreak-associated cases were compared with non-outbreak-associated cases and traceback and environmental investigations were performed. We identified 35 outbreak-associated cases in 12 states; 34 (97%) were hospitalized and seven (20%) died. Outbreak-associated ill persons were more likely to have eaten commercially produced, prepackaged caramel apples (odds ratio 326·7, 95% confidence interval 32·2-3314). Environmental samples from the grower's packing facility and distribution-chain whole apples yielded isolates highly related to outbreak isolates by wgMLST. This outbreak highlights the importance of minimizing produce contamination with L. monocytogenes. Investigators should perform single-interviewer open-ended interviews when a food is not readily identified.

From a regulatory perspective, Asian gypsy moth is a species complex consisting of three species of Lymantria and two subspecies of Lymantria dispar (L.), differing from the European subspecies, L. dispar dispar (L.), by having consistently flight-capable females. As such, the invasion potential in North America is thought to exceed that of European gypsy moth. USDA-APHIS therefore has a monitoring program to detect Asian gypsy moth at high-risk introduction pathways. Molecular markers are used to improve the diagnosis of Asian gypsy moth. One such marker, which targets the FS1 locus, detects an allele, FS1-A, prevalent in Asian populations but occurring at low frequencies (3-6%) throughout the European gypsy moth's range in North America. However, some locales, such as Minnesota, exhibit elevated FS1-A frequencies. We studied the distribution of the FS1-A allele in northern Minnesota, 2013-2014, assessing spatial patterns in the distribution of the FS1-A allele using Moran's I and using spatial regression techniques to examine if the FS1-A allele was associated with putative movement pathways. We also used time series analysis to discern if temporal patterns in FS1-A or possible introduction events occurred. Our results indicated that FS1-A occurred randomly in space and time. We found no evidence that elevated FS1-A frequencies were associated with movement pathways or possible immigration events into this region over the two years. Elevated frequencies of the FS1-A allele within this region could be due to genetic drift and allelic surfing along the expanding population front, or to selection of physiological or behavioral traits., (© The Authors 2017. Selenium-enriched dietary supplements containing various selenium compounds are readily available to consumers. To ensure proper selenium intake and consumer confidence, these dietary supplements must be safe and have accurate label claims. Varying properties among selenium species requires information beyond total selenium concentration to fully evaluate health risk/benefits A LC-ICP-MS method was developed and multiple extraction methods were implemented for targeted analysis of common "seleno-amino acids" and related oxidation products, selenate, selenite, and other species relatable to the quality and/or accuracy of the labeled selenium ingredients. Ultimately, a heated water extraction was applied to recover selenium species from non-selenized yeast supplements in capsule, tablet, and liquid forms. For selenized yeast supplements, inorganic selenium was monitored as a means of assessing selenium yeast quality. There is a need for simple, fast, efficient and sensitive methods of analysis for glyphosate and its degradate aminomethylphosphonic acid (AMPA) in diverse matrices such as water, plant materials and soil to facilitate environmental research needed to address the continuing concerns related to increasing glyphosate use. A variety of water-based solutions have been used to extract the chemicals from different matrices. Many methods require extensive sample preparation, including derivatization and clean-up, prior to analysis by a variety of detection techniques. This review summarizes methods used during the past 15 years for analysis of glyphosate and AMPA in water, plant materials and soil. The simplest methods use aqueous extraction of glyphosate and AMPA from plant materials and soil, no derivatization, solid-phase extraction (SPE) columns for clean-up, guard columns for separation and confirmation of the analytes by mass spectrometry and quantitation using isotope-labeled internal standards. Escherichia coli is used to indicate fecal contamination in freshwater systems and is an indicator of the potential presence of human pathogens. However, naturalized E. coli strains that persist and grow in the environment confound the use of this bacterium as a fecal indicator. Here we examined the spatial and temporal distribution of E. coli in water and sediments of the Seven Mile Creek (SMC), a constructed, ephemeral watershed. E. coli concentrations showed variation by site and date, likely due to changes in temperature and rainfall. Horizontal fluorophore enhanced rep-PCR (HFERP) DNA fingerprint analyses indicated that E. coli populations were very diverse and consisted of transient and naturalized strains, which were especially prevalent in sediment. E. coli fingerprints from water and sediment collected in the same year clustered together with significant overlap, indicating exchange of strains between matrices. Isolates obtained during periods of flow, but not during non-flow conditions, clustered together regardless of sample site, indicating that transport between sites occurred. Naturalized E. coli strains were found in the SMC and strains become geographically isolated and distinct during non-flow conditions. Isolates collected during late spring to fall clustered together at each site, suggesting that temperature and growth of naturalized strains are likely factors affecting population dynamics. Results of this study show that newly introduced and naturalized E. coli strains are present in the SMC. In response to concerns of increasing significance of stink bugs (Hemiptera: Heteroptera: Pentatomidae) in northern states, a survey was conducted over 2 yr in Minnesota to characterize the Pentatomidae associated with field corn, Zea mays L. Halyomorpha halys (Stål), an exotic species, was not detected in this survey, despite continued detection of this species as an invader of human-made structures in Minnesota. Five species of Pentatomidae (four herbivorous; one predatory) were collected from corn. Across years, Euschistus variolarius (Palisot de Beauvois) and Euschistus servus euschistoides (Vollenhoven) had the greatest relative abundances and frequencies of detection. In 2012, the abundance of herbivorous species exceeded 25 nymphs and adults per 100 plants (i.e., an economic threshold) in 0.48% of fields. However, the abundance of herbivorous species did not reach economic levels in any fields sampled in 2013. The frequency of detection of herbivorous species and ratio of nymphs to adults was highest during reproductive growth stages of corn. The predator species, Podisus maculiventris (Say), was detected in 0 to 0.32% of fields. These results provide baseline information on the species composition and abundance of Pentatomidae in Minnesota field corn, which will be necessary for documentation of changes to this fauna as a result of the invasion of H. halys and to determine if some native species continue to increase in abundance in field crops., (© The Authors 2015. Background: There is a need for a simple, fast, efficient and sensitive method for analysis of glyphosate and its degradate aminomethylphosphonic acid (AMPA) in diverse matrices such as water, vegetation and soil., Results: Aqueous extracts from water, vegetation and soil were passed through reverse-phase and cation-exchange columns and directly injected into a tandem mass spectrometer using only a guard column for separation. Extraction efficiencies from the three matrices were >80% for both glyphosate and AMPA. The method reporting levels (MRLs) for glyphosate in water, vegetation and soil were 3.04 µg L(-1) , 0.05 mg kg(-1) and 0.37 mg kg(-1) respectively. AMPA MRLs were 5.06 µg L(-1) for water, 0.08 mg kg(-1) for vegetation and 0.61 mg kg(-1) for soil., Conclusions: A validated, simple and efficient liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for routine analysis of glyphosate and AMPA in water, vegetation and soil that uses minimal sample handling and clean-up will facilitate the additional environmental research needed to address the continuing concerns related to increasing glyphosate use., (Published 2013. In August 2010, the Minnesota Department of Agriculture and Minnesota Department of Health investigated an outbreak of six cases of Salmonella Newport infection occurring in northwestern Minnesota, which identified fresh blueberries as the cause. Initially, traditional traceback methods involving the review of invoices and bills of lading were used to attempt to identify the source of the outbreak. When these methods failed, novel traceback methods were used. Specifically, supplier-specific 12-digit Global Trade Item Numbers (GTINs) and shopper-card information were used to identify a single blueberry grower linked to cases, corroborating the results of a case-control study in which consuming fresh blueberries was statistically associated with illness (5 of 5 cases versus 8 of 19 controls, matched odds ratio [MOR] undefined, P = 0.02). Consuming fresh blueberries from retailer A was also statistically associated with illness (3 of 3 cases versus 3 of 18 controls, MOR undefined, P = 0.03). Based on initially incomplete evidence in this investigation, the invoices pointed to wholesaler A and grower A, based on first-in-first-out product rotation. However, when point-of-sale data were analyzed and linked to shopper-card information, a common GTIN was identified. This information led to an on-site record evaluation at retailer A, and the discovery of additional records at this location documented the supply chain from grower B to wholesaler C to retailer A, shifting the focus of the investigation from grower A to grower B. This investigation demonstrates the emerging concepts of Critical Tracking Events (CTEs) and Key Data Elements (KDE) related to food product tracing. The use of these shopper-cased data and the event data that were queried by investigators demonstrates the potential utility of consciously designed CTEs and KDEs at critical points in the supply chain to better facilitate product tracing.

Traceback methods by state regulatory agencies were used to complement traditional epidemiological cluster investigation methods and confirmed hazelnuts (also referred to as filberts) as the vehicle in a multistate outbreak of Escherichia coli O157:H7 infections. Bulk in-shell hazelnut and mixed-nut purchase locations were identified during the initial epidemiological interviews. Based on purchase dates and case onset dates, regulators in Minnesota, Michigan, and Wisconsin traced product back through the supply chain. Six (86%) retail locations received the suspect hazelnut or mixed-nut shipments from a Minnesota distributor, with one retailer (14%) receiving products from a Wisconsin distributor. Both distributors received 100% of their bulk in-shell hazelnuts and mixed nuts from a distributor in California. The California distributor received 99% of their hazelnuts from a packing company in Oregon. The California distributor received the hazelnuts in 50-lb (22.7-kg) bags and either resold them without opening the bags or used the in-shell hazelnuts in the manufacture of their in-shell mixed nuts. Records at the packing company in Oregon were incomplete or lacked sufficient detail needed to identify a suspect farm or group of suspect farms. Laboratory samples collected from human cases and subsequently recalled product matched the outbreak pulsed-field gel electrophoresis subtype of E. coli O157:H7. Hazelnut harvesting practices create a plausible route of contamination from fecal matter from domestic ruminants or wild deer. This outbreak investigation demonstrates the use of product traceback data to rapidly test an epidemiological hypothesis.

Sunflower rust caused by Puccinia helianthi (Schw.) is widespread in North America and occurs annually on cultivated sunflower (Helianthus annuus L.) and wild annual and perennial Helianthus spp., although severity on the U.S. sunflower crop has been increasing in recent years (2). P. helianthi is a autoecious, macrocyclic rust, but the aecial stage is rarely observed in the field (1,3,4). In most years, the earliest appearance of sunflower rust in North Dakota (ND) and surrounding states usually occurs in early August as the uredinial stage. Initial inoculum can result from urediniospores that overwinter in the Northern Great Plains, urediniospores blown in from areas south of North Dakota, or basidiospores completing the life cycle. However, aecia have been noted very infrequently and never widespread, indicating initial inoculum is usually urediniospores. Aecia of P. helianthi were first observed on 24 June 2008 in a commercial sunflower field (confection hybrid CHS 3126) near Mohall, ND. Aecia cups measuring 0.2 to 0.3 mm in diameter were observed in clusters that were 1 to 7 mm wide in diameter and containing as many as 150 cups. Aecia cups were bright orange but turned brown-black as they senesced. As many as 15 aecial clusters occurred on individual leaves or cotyledons. Aeciospores were ellipsoid, hyaline, and measured approximately 20 μm in diameter. On 4 July 2008, uredinia were first observed in the same Mohall, ND field. At that time, uredinia, aecia, and senesced aecia could all be found on the same leaves. In a non-fungicide-treated strip of the field, pustule coverage on the lower leaves was 10 to 20% by mid-July, pustule coverage on the upper four leaves of plants in the untreated strip was 56% by 27 August, and yield at harvest was less than 200 kg/ha. The rest of the field was sprayed twice with fungicides and yielded 1,571 kg/ha, which is similar to the statewide yield average of 1,573 kg/ha in 2008. To determine the prevalence of aecia in the primary growing regions of ND and Minnesota (MN), surveys were conducted in 75 sunflower fields in 18 counties between 22 and 24 July in ND and 34 fields in 8 counties between 17 and 31 July in MN. Incidence of aecia and uredinia were determined by visual observation of a minimum of 20 plants scouted in a 'W' pattern in the field. Rust was found in 31 and 53% of fields in ND and MN, respectively. In fields where rust was found, both aecia and uredinia were observed in 37% of the fields, aecia only in 29% of the fields, and uredinia only in 34% of the fields. Although it is uncertain why aecia were widespread in 2008, night temperatures in Mohall, ND, where aecia were first observed, reached the dew point temperature on 51 of 61 days in June and July, suggesting that dew or fog likely formed. Thus, favorable conditions for germination and infection early in the growing season indicate widespread occurrence of rust was likely a result of local inoculum sources. The early appearance of aecia is cause for concern for two reasons: significant yield loss can occur when rust appears early in the growing season; and the presence of aecia suggest that the pathogen completed its sexual cycle. Aster yellows (AY), a disease of small grain crops caused by aster yellows phytoplasma (AYp), produces disease symptoms similar to barley yellow dwarf (BYD). From 2003 to 2005, small grain production fields in Minnesota and North Dakota were surveyed to determine the incidences of AY and BYD. In-field spatial patterns of AY-infected plants also were investigated. Plants collected along a five-point transect line were tested for AYp using nested polymerase chain reaction (PCR) and quantitative real-time PCR assays, and extracted plant sap was tested for serotypes PAV and RPV of Barley yellow dwarf virus (BYDV) using enzyme-linked immunosorbent assays. During 2003, 2004, and 2005, AYp was detected in plants from 49, 15, and 7% of tested fields, respectively, whereas BYDV was found in plants from 2, 0, and 5% of fields, respectively. Average amplicon count number indicated an in-field spatial trend for greater incidence of AYp and increased populations of AYp in plants located near field edges, with comparably low copy numbers at transect point locations toward the direction of field center. AY is likely a common but largely undetected disease on small grain crops in the Upper Midwest.

Mice and rat populations are commonly controlled by two classes of rodenticide anticoagulants, coumarins and indandiones. However, poisoning of nontarget animals also often occurs. For cases such as these, a rapid, multiresidue method, which provides positive confirmation for both classes of anticoagulant rodenticides, is needed by diagnostic laboratories. A method was developed for the determination of seven anticoagulant rodenticides, coumafuryl, pindone, warfarin, diphacinone, chlorophacinone, bromadiolone, and brodifacoum, in diverse matrices, animal feed, cooked beef, and fruit-flavored beverages using high-performance liquid chromatography/electrospray/mass spectrometry. Detection was by MS/MS with electrospray ionization in negative mode. Confirmation was by retention time, m/z of molecular ion, and two parent-daughter transitions. Recoveries from selected the matrices ranged from 61 to 117%. Limits of quantitation were as low as 1.5-4.5 ng g-1. The developed method was rapid and provided the simultaneous confirmation and quantification of the seven anticoagulant rodenticides.

A federal and state program operated from 1918 until the 1980s to eradicate common barberry (Berberis vulgaris), the alternate host of Puccinia graminis, from the major areas of cereal production in the United States. Over 500 million bushes were destroyed nationally during the program, approximately 1 million in Minnesota. Some sites in Minnesota where barberry bushes were destroyed remained in the "active" class when eradication was phased out in the 1980s. Active sites were defined as those on which there was still a possibility of emergence of barberry seedlings or sprouts arising from the parent bush. In the present study, from 1998 to 2002, 72 of the approximately 1,200 active sites in Minnesota were surveyed. Areas within 90 m of mapped locations of previously destroyed bushes were searched carefully at each site. Reemerged barberry plants were found on 32 sites. The reproductive status and GPS coordinates were recorded for each reemerged bush. More than 90% of the barberry bushes were found in counties with less than 400 ha of wheat per county, mostly in southeastern Minnesota, but one bush was found in a major wheat-producing county in northwestern Minnesota. Reemergence of barberry may serve as a source of new wheat stem rust races in future epidemics.

The bark beetles Ips pini, I. perroti, and I. grandicollis are sympatric in pine forests of the north-central United States. They share the same limited phloem resource and often coexist within the same host trees. We tested whether phloem resources are partitioned in time and space by measuring spatial and seasonal colonization of logs. Differences among species in flight phenology, development time, voltinism, and spatial colonization patterns within logs reduce, but do not eliminate, species overlap. The bark beetle species share predation by Thanasimus dubius (Cleridae) and Platysoma cylindrica (Histeridae), which exploit pheromone signals for prey location. We employed pheromone traps to test for chemical communication among bark beetle species. Heterospecific signals tend to be deterrents when they are added to conspecific signals but attractants when they are alone, indicating that the communication system can both reduce and increase species overlap in resource use depending upon relative abundance of the species. Deterrence by heterospecific signals is probably a result of selection for minimizing interspecific competition. However, individuals may sometimes benefit from joining aggregations of other species because of (1) predator swamping, (2) improved success in attacking live trees, and (3) location of suitable, recently dead, trees. These benefits should be greatest for males (which locate and colonize host trees before signalling females) and indeed males tended to be more attracted than females by heterospecific signals. Shared resources, shared predators, and heterospecific pheromone communication all contribute to species interactions in this guild of bark beetles, but predicting whether the removal of one species will tend to increase or decrease the abundance of remaining species remains difficult. Species interactions are likely conditional and coexistence is probably promoted by benefits to rare species of multispecies associations.

Nine ice cream sample containers, representing three production lots involved in a 1994 outbreak of salmonellosis, were obtained from the manufacturer's distribution warehouse and from consumers. Single 100-g samples from each container were tested initially, with analyses beginning seven weeks after the ice cream was produced. Quantitative salmonella analysis of these samples was performed 14 weeks after the production date using a three-replicate, three-dilution most probable number (MPN) procedure using 100-, 10-, and 1-g samples. The MPN/g estimates ranged from 0.004 to 0.46 Salmonella enteritidis per g with six of nine samples showing an MPN value of 0.093 S. enteritidis per g. The 95% confidence interval for S. enteritidis among these samples was < 0.001 to 2.4 cells/g. The 95% upper limit of S. enteritidis per g was 0.38 for five of six consumer samples. Based on this, the number of S. enteritidis cells per serving (65 g) was 25. Based on the consumption of a single sundae cone (73 g, prepackaged), which caused severe illness in an eight-year-old boy and moderate to mild illness in the adult parents, the infective dose would appear to be no more than 28 cells.

The enforcement of dairy and food safety regulations has fallen most heavily on the states in the past, and this responsibility will no doubt continue, and even expand, in the future. States must be concerned not only about their own state laws but also about the impact of increasing federal regulations. The protection of sample integrity and chain of custody is an important function of a state regulatory program for food safety. Because of increasing demands imposed by new technology and food safety requirements, and because of very limited resources, states have difficulty maintaining adequate regulatory programs. The future challenge will be the ability of states to coordinate regulation, sample analysis, and enforcement between federal and state agencies. States must continue their roles of ensuring consumers that food is safe and wholesome while maintaining viable state food industries.

In an earlier study we documented attraction of the specialist parasitoidMacrocentrus grandii Goidanich to odors of com, potato, and snap bean. In the present study the chemical bases of the parasitoid's attraction to these food plants of its European corn borer host were compared. Volatile compounds from corn leaves were isolated using Tenax and identified by capillary gas chromatographic-mass spectrometry. Twenty-one compounds including aldehydes, ketones, alcohols, esters, and sesquiterpenes were present in corn. These were separated into fractions by column chromatography on Florisil. Wind-tunnel bioassays of the fractions indicated thatM. grandii was attracted to fractions containing nonpolar and slightly polar compounds including sesquiterpenes, aldehydes, a ketone, and esters. More polar compounds in corn, like alcohols, were not attractive. Attraction to potato odor was based on the presence of the same classes of compounds that were attractive in corn, but more polar compounds were involved in attraction to snap bean odor. This study indicated that only some compounds in each of the three odor complexes tested were attractive. It also documented that different compounds were involved in attraction ofM. grandii to different plant odors.
