Objectives: This study was conducted to measure financial toxicity (FT) among patients with cancer in Vietnam using the COmprehensive Score for financial Toxicity (COST) and to describe the cost management strategies employed by these patients. Methods: This comprehensive cross-sectional study enrolled 634 patients from 2 specialized oncology hospitals in Vietnam. Using COST cut-off scores, FT was classified as none/mild (≥26), moderate (14-25), or severe (≤13). Cost management strategies, or coping mechanisms, were classified into 4 groups: lifestyle changes, financial resource strategies, treatment modifications, and support seeking. Results: The prevalence of FT was 91.8%, with 51.7% of participants demonstrating severe and 40.1% exhibiting moderate FT. Severe FT was significantly associated with female, low education level, unstable employment, low household economic status, and advanced cancer stage. The most common coping strategies were as follows: among lifestyle changes, reducing spending on basic items and leisure activities (78.7%) and cutting back on essential household expenses (66.4%); among financial resource strategies, borrowing money from relatives or friends (49.1%) and withdrawing funds from retirement or savings accounts (34.1%); within treatment modifications, switching treatment facilities or doctors due to cost concerns (9.3%); and within support seeking, obtaining help from welfare or community organizations (18.8%). All strategies were significantly more likely to be used by patients with severe FT. Conclusions: FT was highly prevalent among patients with cancer. Most patients relied on lifestyle adjustments and coping strategies, underscoring the need for improved financial support systems to alleviate the economic burden associated with cancer care.

Objectives: This study validated the Vietnamese version of the Disaster Adaptation and Resilience Scale (DARS) for use in vulnerable communities in Vietnam. Methods: This was a cross-sectional study involving 595 adults from 2 identified communities. The original DARS assessment tool was translated, and the validity and reliability of the Vietnamese version of DARS (V-DARS) were assessed. The internal consistency of the overall scale and its subscales was evaluated using Cronbach’s alpha and McDonald’s omega reliability coefficients. Confirmatory factor analysis (CFA) was employed to evaluate its construct validity, building upon the factor structure identified in exploratory factor analysis (EFA). Construct validity was assessed based on convergent and discriminant validity. Results: Following the established criteria for EFA, 8 items were removed, resulting in a refined V-DARS structure comprising 35 items distributed across 5 distinct factors. Both alpha and omega reliability coefficients indicated strong internal consistency for the overall scale (α=0.963, ω=0.963) and for each of the 5 sub-scales (all>0.80). The compromised egg quality and leg abnormality during the end of the laying cycle (after 40 weeks) have been leading to poor animal welfare and substantial economic losses. Therefore, the effects of fermented calcium (Ca) butyrate, produced by fermentation by Clostridium butyricum, on production, eggshell quality, and tibial property of hens were explored. A total of 192 Hy-line brown laying hens at 50-week-old were assigned to a basal diet or the basal diet with 300 mg/kg of the fermented Ca butyrate from 50 to 58 weeks of age. Each treatment had 6 replicates with 16 hens each. The diet supplemented with 300 mg/kg fermented Ca butyrate notably increased egg weight, ovarian follicle number, and eggshell strength (P = 0.072) as compared to the basal diet, which were associated with cytokine secretion, toll-like receptor signaling pathways, and intestinal immunity based on the RNA-seq data from the granulosa. Dietary Ca butyrate inclusion decreased the expression of ileal tumor necrosis factor-alpha and serum pro-inflammatory cytokine concentration, as well as increased the content of serum immunoglobulin A when compared to the basal diet (both P 0.05). Cross-border e-commerce(CBEC) has created enormous economic benefits for consumers and vendors worldwide. Vietnamese Small and Medium-sized Enterprise (SMEs) can enhance their competitiveness and facilitate their integration into the global market via cross-border e-commerce adoption. This study assesses the critical determinants of cross-border e-commerce adoption intention in Vietnamese SMEs. This study integrates the factors of the technology–organisation–environment (TOE) framework and the theory of planned behaviour (TPB) to construct a holistic framework to assess the critical determinants of cross-border e-commerce adoption intentions among SMEs in Vietnam. The research method utilised Structural Equation Modelling Partial Least Squares (SEM-PLS) with SmartPLS (version 4.0) to evaluate the relationship between variables. The sample size consists of 400 respondents from Vietnamese SMEs. The results indicate that perceived behavioural control, subjective norms, and attitudes toward CBEC adoption are the critical determinants of cross-border e-commerce adoption intention among Vietnamese SMEs. For the technological dimensions of the TOE framework, perceived benefits, perceived compatibility and perceived security positively relate to attitude toward CBEC adoption; perceived complexity and perceived cost negatively relate to attitude toward CBEC adoption and perceived behavioural control. For the organisational dimensions of the TOE framework, government support, customer pressure, and competitive pressure positively relate to subjective norms. For the environmental dimensions of the TOE framework, organisational readiness, organisational innovativeness, and e-commerce knowledge positively relate to perceived behavioural control. By integrating the constructs of TPB and TOE into a single framework, this study adds to current knowledge by addressing the literature gap on the significant role of organisational and environmental factors in the adoption of SMEs' CBEC in developing countries like Vietnam. The managerial implications provided a holistic view of SMEs' perspectives and practices on promoting the CBEC adoption intention.

Objectives We assessed the prevalence of stress, anxiety, and depression among adolescents living in families with separated or divorced parents in Hue City, Vietnam and identified factors associated with these conditions. Methods This cross-sectional study enrolled 309 adolescents, aged 12 to 17 years, living in families with separated or divorced parents in Hue City, Vietnam. The depression anxiety stress scale-21 (DASS-21) was used to measure stress, anxiety, and depression. Predictors of overall and individual mental health problems were identified using ordered and binary logistic regression, respectively. Results The DASS-21 scale revealed a 49.2% prevalence of stress, while anxiety and depression had s prevalence rates of 61.5%. Among participants, 42.4% experienced all 3 mental health issues. Objectives The coronavirus disease 2019 (COVID-19) pandemic has increased the workload of healthcare workers (HCWs), impacting their health. This study aimed to assess sleep quality using the Pittsburgh Sleep Quality Index (PSQI) and identify factors associated with poor sleep among HCWs in Vietnam during the COVID-19 pandemic. Methods In this cross-sectional study, 1000 frontline HCWs were recruited from various healthcare facilities in Vietnam between October 2021 and November 2021. Data were collected using a 3-part self-administered questionnaire, which covered demographics, sleep quality, and factors related to poor sleep. Poor sleep quality was defined as a total PSQI score of 5 or higher. Results Participants’ mean age was 33.20±6.81 years (range, 20.0–61.0), and 63.0% were women. The median work experience was 8.54±6.30 years. Approximately 6.3% had chronic comorbidities, such as hypertension and diabetes mellitus. About 59.5% were directly responsible for patient care and treatment, while 7.1% worked in tracing and sampling. A total of 73.8% reported poor sleep quality. Multivariate logistic regression revealed significant associations between poor sleep quality and the presence of chronic comorbidities (odds ratio [OR], 2.34; 95% confidence interval [CI], 1.17 to 5.24), being a frontline HCW directly involved in patient care and treatment (OR, 1.59; 95% CI, 1.16 to 2.16), increased working hours (OR, 1.84; 95% CI,1.37 to 2.48), and a higher frequency of encountering critically ill and dying patients (OR, 1.42; 95% CI, 1.03 to 1.95). Conclusions The high prevalence of poor sleep among HCWs in Vietnam during the COVID-19 pandemic was similar to that in other countries. Working conditions should be adjusted to improve sleep quality among this population.

Abstract Water sources for irrigation systems in the Red River Delta are crucial to the socioeconomic growth of the region's communities. Human activities (discharge) have polluted the water source in recent years, and the water source from upstream is limited. Currently, the surface water quality index (WQI), which is calculated from numerous surface water quality parameters (physical, chemical, microbiological, heavy metals, etc.) is frequently used to evaluate the surface water quality of irrigation systems. However, the calculation of the WQI from water quality monitoring parameters remains constrained due to the need for a large number of monitoring parameters and the relative complexity of the calculation. To better serve the assessment of surface water quality in the study area, it is crucial and essential to conduct research to identify an efficient and accurate method of calculating the WQI. This study used machine learning and deep learning algorithms to calculate the WQI with minimal input data (water quality parameters) to reduce the cost of monitoring surface water quality. The study used the Bayes method (BMA) to select important parameters (BOD5, NH4 +, PO4 3−, turbidity, TSS, coliform, and DO). The results indicate that the machine learning model is more effective than the deep learning model, with the gradient boosting model having the most accurate prediction results because it has the highest coefficient of determination R2 (0.96). This is a solid scientific basis and an important result for the application of machine learning and deep learning algorithms to calculate WQI for the research area. The study also demonstrated the potential of artificial intelligence algorithms to improve water quality forecasting compared to traditional methods with minimal cost and time.

A spotted scat, Scatophagus argus, has a high nutritional value and is among Asia’s most widely consumed fish species. Thua Thien Hue’s consumption market considers this species to be of high economic value and requires protection and conservation of the population. However, the studies on the identification and genetic diversity of S. argus distributed in Vietnam are still lacking. Therefore, mitochondrial cytochrome c oxidase subunit I (COI) gene was utilized to distinguish different populations and investigate the genetic diversity of two populations of S. argus from Tam Giang lagoon, Thua Thien Hue province (n = 31) and Ca Mau province (n = 14). The sequencing results indicated 13 distinct haplotypes among 45 sequences. Five single nucleotide polymorphisms were observed to distinguish Hue spotted scat population. The S. argus population in Ca Mau province was higher haplotype diversity (Hd) and nucleotide diversity (π) than those of Thua Thien Hue province, which demonstrates that there are minor differences between haplotypes. There were genetic distances ranging from 0%–4% within the populations and 6.67% between the two populations. In addition to the sequencing, the comparison of morphology, biology, culture, and the growth rate was sufficient to distinguish the spotted scat S. argus in Thua Thien Hue from Ca Mau.

This study aims to assess the impacts of three components from Theory of Planned Behaviour (TPB) on Entrepreneurial intention and analyze the mediating effects of these three components in the relationship between Entrepreneurial education and Entrepreneurial intention and in the relationship between Self-Efficacy and Entrepreneurial intention among engineering students. Data were collected via a structured questionnaire from engineering students at Industrial University of Ho Chi Minh City in Vietnam. The methodology employed Structural Equation Modelling Partial Least Squares (SEM-PLS) using SmartPLS (version 3.3.3) to assess the relationship among variables. Findings confirm the directly positive relationship between Attitude toward entrepreneurship, Perceived behaviour control and Subjective norms on engineering student’s entrepreneurial intention. These three components of TPB mediate the relationship between Entrepreneurial education, Self-Efficay and engineering student’s entrepreneurial intention. This study contributes toward the understanding of engineering student’s entrepreneurial intention in Vietnam as an emerging market in Asia. The mediating effects of three components of TPB have been systematically investigated and the indirect roles of Entrepreneurial education and Self-Efficay have been validated by applying employed Structural Equation Modelling Partial Least Squares (SEM-PLS). Managerial implications are discussed to promote engineering student’s entrepreneurial intention in Vietnam. Higher education leaders and policymakers can utilise the findings to construct the initiatives of entrepreneurship development and their implementation for engineering students.

Purpose The current study aimed to use network analysis to investigate medical and health students’ readiness for online learning during the coronavirus disease 2019 (COVID-19) pandemic at the University of Medicine and Pharmacy, Hue University. Methods A questionnaire survey on the students’ readiness for online learning was performed using a Google Form from May 13 to June 22, 2021. In total, 1,377 completed responses were eligible for analysis out of 1,411 participants. The network structure was estimated for readiness scales with 6 factors: computer skills, internet skills, online communication, motivation, self-control, and self-learning. Data were fitted using a Gaussian graphical model with the extended Bayesian information criterion. Results In 1,377 students, a network structure was identified with 6 nodes and no isolated nodes. The top 3 partial correlations were similar in networks for the overall sample and subgroups of gender and grade levels. The self-control node was the strongest for the connection to others, with the highest nodal strength. The change of nodal strength was greatest in online communication for both gender and grade levels. The correlation stability coefficient for nodal strength was achieved for all networks. Conclusion These findings indicated that self-control was the most important factor in students’ readiness network structures for online learning. Therefore, self-control needs to be encouraged during online learning to improve the effectiveness of achieving online learning outcomes for students.

Droughts have a substantial impact on water supplies, agriculture, and ecosystems worldwide. Agricultural sustainability and production in the Mekong Delta of Vietnam are being jeopardized by droughts caused by climate change. Conventional forecasting methods frequently struggle to comprehend the intricate dynamics of meteorological occurrences connected to drought, necessitating the use of sophisticated prediction techniques. This study assesses the effectiveness of various statistical models (ARIMA), machine learning, and deep learning models (Gradient Boosting, XGBoost, RNN, and LSTM) in forecasting the SPEI over different time periods (1, 3, 6, and 12 months) across six prediction intervals. The models were developed and evaluated using data from 11 meteorological stations spanning from 1985 to 2022. These models incorporated various climatic variables, including precipitation, temperature, humidity, potential evapotranspiration (PET), Southern Oscillation Index (SOI) Anomaly, and sea surface temperature in the NINO4 region (SST_NINO4). The results demonstrate that XGBoost and LSTM models exhibit outstanding performance, showcasing lower error metrics and higher R² values compared to Gradient Boosting and RNN. The performance of the model fluctuated depending on the forecast step, with error metrics often increasing with longer prediction horizons. The use of climatic indices improved the accuracy of the model. These findings are consistent with earlier research on drought episodes in the Mekong Delta and support studies from other areas that show the effectiveness of advanced modeling tools for predicting droughts. The work emphasizes the capacity of machine learning and deep learning models to enhance the precision of drought forecasting, which is vital for efficient water resource management and agricultural planning in places prone to drought. Medical students experience extensive pressure during their undergraduate courses. Given the complex associations between psychological factors in association with academic pressure. We investigated the study with objectives: To examine psychological factors related to academic pressure by analysing interactions between ‘study motivation’, ‘study environment’, ‘study conditions’, ‘teacher quality’, ‘training programme’, ‘management system’, ‘evaluation’, and ‘extracurricular activities’ using a network analysis approach. A total of 878 medical students majoring in general medicine from the first, third, and fifth years of a six-year course at the largest medical university in central and highland regions of Vietnam were involved in this cross-sectional study. The approach used was convenient cluster sampling with a self-administered questionnaire by the participants. Network analysis for pairwise correlations between psychological factors was estimated . Important factors in the network analysis were calculated using centrality indices including node strength (S), closeness (C), and betweenness (B). The higher score of S, C, and B indicate the more importance of the node. The results obtained from the network analysis of eight psychological factors showed that ‘teaching quality’ was mostly connected with other factors overall, while the ‘training programme’ was seen in both genders and freshman students. ‘Study conditions’ and ‘training programme’ were mostly connected with other factors in junior and senior students, respectively. The strong pairwise correlation was confirmed: management system and evaluation activity, followed by study environment and study conditions, and teaching quality and training programme. Additionally, nodes with high centrality were shown to be ‘management system’ (S = 0.97, C = 0.019, B = 1), and ‘training programme’ (S = 0.96, C = 0.021, B = 4). Our study findings indicate that satisfaction with the training programme amongst eight psychological factors is the most important factor affecting academic pressure among medical students. The training programme is linked with teaching quality, whereas the management system is correlated with evaluation activity.

Prostate cancer metastasis is a significant cause of mortality in men. PKD3 facilitates tumor growth and metastasis, however, its regulation is largely unclear. The Hsp90 chaperone stabilizes an array of signaling client proteins, thus is an enabler of the malignant phenotype. Here, using different prostate cancer cell lines, we report that Hsp90 ensures PKD3 conformational stability and function to promote cancer cell migration. We found that pharmacological inhibition of either PKDs or Hsp90 dose-dependently abrogated the migration of DU145 and PC3 metastatic prostate cancer cells. Hsp90 inhibition by ganetespib caused a dose-dependent depletion of PKD2, PKD3, and Akt, which are all involved in metastasis formation. Proximity ligation assay and immunoprecipitation experiments demonstrated a physical interaction between Hsp90 and PKD3. Inhibition of the chaperone–client interaction induced misfolding and proteasomal degradation of PKD3. PKD3 siRNA combined with ganetespib treatment demonstrated a specific involvement of PKD3 in DU145 and PC3 cell migration, which was entirely dependent on Hsp90. Finally, ectopic expression of PKD3 enhanced migration of non-metastatic LNCaP cells in an Hsp90-dependent manner. Altogether, our findings identify PKD3 as an Hsp90 client and uncover a potential mechanism of Hsp90 in prostate cancer metastasis. The molecular interaction revealed here may regulate other biological and pathological functions.

Abstract Vietnam is exposed to different types of floods that cause severe economic losses, damage to infrastructure, and loss of life. Reliable information on the drivers, patterns and dynamics of flood risk is crucial for the identification, prioritization and planning of risk reduction and adaptation measures. Here, we present a systematic review of existing flood risk assessments in Vietnam. We evaluate the current status, persisting gaps, and challenges regarding the understanding and assessment of flood risk in the country. The literature review revealed that: (i) 65 % of the reviewed papers did not provide a clear definition of flood risk, (ii) assessments had a tendency to prioritize physical and environmental drivers of risk over social, economic or governance‐related drivers, (iii) future‐oriented assessments tended to focus on hazard and exposure trends, while vulnerability scenarios were often lacking, (iv) large and middle‐sized cities were assessed more frequently than others, (v) only few studies engaged with relevant local stakeholders for the assessment of risk and the development of potential solutions, and (vi) ecosystem‐based adaptation and flood risk insurance solutions were rarely considered. Based on these findings, we point out several directions for future research on flood risk in Vietnam.

OBJECTIVES The prevalence of diabetes mellitus is rapidly increasing in Vietnam, particularly among adults aged over 45 years. This study estimated trends in diabetes and prediabetes prevalence and determined risk factors in Vietnamese adults (over 45 years). METHODS A cross-sectional study was conducted based on data from an annual diabetes screening program among people aged 45-69 years in an urban city in central Vietnam (Da Nang). Joinpoint regression analyses were performed to calculate the annual percentage change and ptrend-values. Multinomial logistic regression analysis was used to determine risk factors. RESULTS In total, 3,765 men and 9,149 women were included in this analysis. The age-adjusted prevalence of diabetes and prediabetes in 2017 was 11.4% and 52.9%, respectively. The prevalence of diabetes was higher in men (15.1%) than in women (10.3%), but that of prediabetes was similar in both genders (53.4% vs. 52.8%). The prevalence of prediabetes significantly increased during the study period, whereas no upward or downward trend for diabetes was observed. The prevalence of obesity, abdominal obesity, hypertension, and dyslipidemia showed no obvious trend. Obesity, a high waist-to-hip ratio, hypertension, more severe abdominal obesity, and dyslipidemia were significantly associated with a higher risk of diabetes and prediabetes. CONCLUSIONS Diabetes and prediabetes were more prevalent among people aged over 45 years than in the general population. Da Nang has experienced a marked increase in the prevalence of prediabetes. These findings have significant implications regarding the need for nationwide public health interventions and management aiming at diabetes prevention and control.

Objective To identify the relationship of quit intention of smoking among daily smokers in Vietnam. Methods Data are subtracted from dataset of Global Adult Tobacco Survey (GATS) of 2010 and 2015 of the Global Tobacco Surveillance System (GTSS). Households were sampled using a stratified multistage design. All men who are currently smoking daily are selected for final analysis. Quit intention of smoking defines as dependent variable. Knowledge, attitude, practice toward smoking; Heaviness of Smoking Index; Demography define as independent variables. Multivariate logistics model was used. Results The proportion of smokers are interested in quitting smoking following: planning to quit within next month (6.2%); thinking about quitting within the next 12 months (15.5%); and will quit someday, but not the next 12 months (38.5%). High education, good attitude of harming of smoking on health were positively associated with intentions to quit smoking with adjusted odd ratio: 1.70 (1.04-2.75); 1.89 (1.19-2.98), respectively. Reversely, Heaviness of Smoking Index (HIS) with moderate smoking, less supported for taxes rise on tobacco products were negatively associated with intentions to quit smoking with adjusted odd ratio: 0.66 (0.51-0.83); 0.72 (0.56-0.92), respectively. Conclusions The finding implies that identifying the determinants of quit intentions provides practical evidence for shaping effective policies and programs for advancing quitting among smokers in Vietnam.

Fermentation has been used for centuries to produce food in South-East Asia and some foods of this region are famous in the whole world. However, in the twenty first century, issues like food safety and quality must be addressed in a world changing from local business to globalization. In Western countries, the answer to these questions has been made through hygienisation, generalization of the use of starters, specialization of agriculture and use of long-distance transportation. This may have resulted in a loss in the taste and typicity of the products, in an extensive use of antibiotics and other chemicals and eventually, in a loss in the confidence of consumers to the products. The challenges awaiting fermentation in South-East Asia are thus to improve safety and quality in a sustainable system producing tasty and typical fermented products and valorising by-products. At the end of the “AsiFood Erasmus+ project” (www.asifood.org), the goal of this paper is to present and discuss these challenges as addressed by the Tropical Fermentation Network, a group of researchers from universities, research centers and companies in Asia and Europe. This paper presents current actions and prospects on hygienic, environmental, sensorial and nutritional qualities of traditional fermented food including screening of functional bacteria and starters, food safety strategies, research for new antimicrobial compounds, development of more sustainable fermentations and valorisation of by-products. A specificity of this network is also the multidisciplinary approach dealing with microbiology, food, chemical, sensorial, and genetic analyses, biotechnology, food supply chain, consumers and ethnology.

The resilience concept has provided a new insight and approach to the conventional perspective of agricultural management by emphasizing the need to maintain a diversity of future options to adapt to inevitable and often unpredictable changes. The concept has been taken up by various academic disciplines and development sectors, yet ways to define and operationalize resilience as a measurable concept are still being developed. We contributed to this ongoing effort by implementing a subjective resilience assessment method based on farmers' perceptions of three resilience components: (1) the sensitivity of their agricultural systems to increased salinity intrusion, (2) the capacity to recover from salinity damage, and (3) the capacity to change to other systems if salinity increases in the future. We conducted 27 in-depth interviews with local and national authorities, 11 focus group discussions, and 118 semistructured and 219 structured interviews with farmers in case study villages located along salinity transects in the Mekong Delta and at different distances to sea dikes in the Red River Delta in Vietnam in 2015-2016. Results from the subjective resilience assessment reveal that none of the agricultural systems studied systematically scored higher than the other systems on all three resilience components, implying that an increase in one resilience component by switching agricultural systems would negatively affect others. Agricultural responses to this salinity problem will influence current and long-term adaptability of the systems to future changes in salinity intrusion and other social-ecological developments in the deltas. Improving resilience components, e.g., through policies and interventions, resource allocation, and farming system changes, to sustain agricultural production or facilitate transformation to alternative systems when necessary is critically important for agricultural systems facing stress. Complementing subjective resilience assessments with qualitative data is thus crucial for understanding the drivers of resilience to improve components of resilience for agricultural systems in the respective deltas.

Feeding rats with high-fat diet (HFD) with a single streptozotocin (STZ) injection induced obesity, slightly elevated fasting blood glucose and impaired glucose and insulin tolerance, and caused cardiac hypertrophy and mild diastolic dysfunction as published before by Koncsos et al. in 2016. Here we aimed to explore the renal consequences in the same groups of rats. Male Long-Evans rats were fed normal chow (CON; n = 9) or HFD containing 40% lard and were administered STZ at 20 mg/kg (i.p.) at week four (prediabetic rats, PRED, n = 9). At week 21 blood and urine samples were taken and kidney and liver samples were collected for histology, immunohistochemistry and for analysis of gene expression. HFD and STZ increased body weight and visceral adiposity and plasma leptin concentration. Despite hyperleptinemia, plasma C-reactive protein concentration decreased in PRED rats. Immunohistochemistry revealed elevated collagen IV protein expression in the glomeruli, and Lcn2 mRNA expression increased, while Il-1β mRNA expression decreased in both the renal cortex and medulla in PRED vs. CON rats. Kidney histology, urinary protein excretion, plasma creatinine, glomerular Feret diameter, desmin protein expression, and cortical and medullary mRNA expression of TGF-β1, Nrf2, and PPARγ were similar in CON and PRED rats. Reduced AMPKα phosphorylation of the autophagy regulator Akt was the first sign of liver damage, while plasma lipid and liver enzyme concentrations were similar. In conclusion, glomerular collagen deposition and increased lipocalin-2 expression were the early signs of kidney injury, while most biomarkers of inflammation, oxidative stress and fibrosis were negative in the kidneys of obese, prediabetic rats with mild heart and liver injury.

Sirtuin 1 (SIRT1) othologs are ubiquitous NAD+-dependent deacetylases that act as nutrient sensors and modulate metabolism and stress responses in diverse organisms. Both mammalian SIRT1 and Caenorhabditis elegans SIR-2.1 have been implicated in dietary restriction, longevity, and healthspan. Hsp90 is an evolutionarily conserved molecular chaperone that stabilizes a plethora of signaling ’client’ proteins and regulates fundamental biological processes. Here we report that Hsp90 is required for conformational stabilization of SIRT1 and SIR-2.1. We find that inhibition of Hsp90 by geldanamycin (GA) induces the depletion of mammalian SIRT1 protein in a concentration and time dependent manner in COS-7 and HepG2 cells. In contrast to SIRT1, SIRT2 level remains unchanged by GA treatment, reflecting a specific Hsp90 SIRT1 interaction. Hsp90 inhibition leads to the destabilization and proteasomal degradation of SIRT1. Moreover, we observe a GA-sensitive physical interaction between SIRT1 and Hsp90 by immunoprecipitation. We also demonstrate that hsp-90 gene silencing also induces SIR-2.1 protein depletion and proteasomal degradation in C. elegans. Our findings identify metazoan SIRT1 orthologs as Hsp90 clients and reveal a novel crosstalk between the proteostasis and nutrient signaling networks, which may have implications in various age related diseases.
