Abstract The surge in interest regarding the next generation of optical fiber transmission has stimulated the development of digital signal processing (DSP) schemes that are highly cost-effective with both high performance and low complexity. As benchmarks for nonlinear compensation methods, however, traditional DSP designed with block-by-block modules for linear compensations, could exhibit residual linear effects after compensation, limiting the nonlinear compensation performance. Here we propose a high-efficient design thought for DSP based on the learnable perspectivity, called learnable DSP (LDSP). LDSP reuses the traditional DSP modules, regarding the whole DSP as a deep learning framework and optimizing the DSP parameters adaptively based on backpropagation algorithm from a global scale. This method not only establishes new standards in linear DSP performance but also serves as a critical benchmark for nonlinear DSP designs. In comparison to traditional DSP with hyperparameter optimization, a notable enhancement of approximately 1.21 dB in the Q factor for 400 Gb/s signal after 1600 km fiber transmission is experimentally demonstrated by combining LDSP and perturbation-based nonlinear compensation algorithm. Benefiting from the learnable model, LDSP can learn the best configuration adaptively with low complexity, reducing dependence on initial parameters. The proposed approach implements a symbol-rate DSP with a small bit error rate (BER) cost in exchange for a 48% complexity reduction compared to the conventional 2 samples/symbol processing. We believe that LDSP represents a new and highly efficient paradigm for DSP design, which is poised to attract considerable attention across various domains of optical communications.

Abstract Background Despite being a prognostic predictor, cardiac autonomic dysfunction (AD) has not been well investigated in chronic obstructive pulmonary disease (COPD). We aimed to characterise computed tomography (CT), spirometry, and cardiopulmonary exercise test (CPET) features of COPD patients with cardiac AD and the association of AD with CT-derived vascular and CPET-derived ventilatory efficiency metrics. Methods This observational cohort study included stable, non-severe COPD patients. They underwent clinical evaluation, spirometry, CPET, and CT. Cardiac AD was determined based on abnormal heart rate responses to exercise, including chronotropic incompetence (CI) or delayed heart rate recovery (HRR) during CPET. Results We included 49 patients with FEV1 of 1.2–5.0 L (51.1–129.7%), 24 (49%) had CI, and 15 (31%) had delayed HRR. According to multivariate analyses, CI was independently related to reduced vascular volume (VV; VV ≤ median; OR [95% CI], 7.26 [1.56–33.91]) and low ventilatory efficiency (nadir VE/VCO2 ≥ median; OR [95% CI], 10.67 [2.23–51.05]). Similar results were observed for delayed HRR (VV ≤ median; OR [95% CI], 11.46 [2.03–64.89], nadir VE/VCO2 ≥ median; OR [95% CI], 6.36 [1.18–34.42]). Conclusions Cardiac AD is associated with impaired pulmonary vascular volume and ventilatory efficiency. This suggests that lung blood perfusion abnormalities may occur in these patients. Further confirmation is required in a large population-based cohort.

Rubber-foamed concrete demonstrates exceptional toughness, a low elastic modulus, and significant sensitivity to density. It is necessary to parameterize the density mix of rubber-foaming concrete to meet engineering design requirements. Density-mix design methods for foaming concrete rely mainly on empirical knowledge or trial-and-error approaches. In this paper, with numerous parametric tests and regression analysis based on general principles for density-mix designs applicable to both foamed and rubber-foamed concretes, the key design parameters, such as volume correction coefficient, rubber size effect coefficient, and water-reducing agent effect coefficient, have been proposed in order to optimize their respective densities more accurately. The tests demonstrated an optimal water-to-cement ratio of 0.45, corresponding to a volume correction factor of 1.027. Incorporating rubber particles and water-reducing agents has a more significant effect on the cement-paste volume. Controlling fluidity in the 200 to 300 mm range is crucial when designing foamed concrete with varying densities. The regression equation accurately predicts the paste’s measured volume and wetting density by incorporating volume corrections, size, and water reduction effect coefficients. By employing a foam excess coefficient of 1.1 and a mass coefficient of 1.25, the dry and wet density error of foam concrete is less than 5%. A comprehensive framework for optimizing mix design in terms of density is provided for applications in foamed concrete and rubber-foamed concrete, facilitating researchers in designing mix ratios for additional novel mixture-based foamed-concrete applications.

Despite considerable evidence for the benefit in chronic obstructive pulmonary disease (COPD), the implementation of pulmonary rehabilitation (PR) is insufficient. However, music therapy may help address this gap due to its unique benefits. Therefore, we aimed to develop a music-therapy facilitated pulmonary telerehabilitation program based on rhythm-guided walking, singing, and objective telemonitoring. A supervised, parallel-group, single-blinded, randomized controlled clinical trial will be conducted, including 75 patients with COPD anticipated to be randomized in a 1:1:1 ratio into three groups. The intervention groups will receive a 12-week remotely monitored rehabilitation program, while the usual care group will not receive any rehabilitation interventions. Of the two intervention groups, the multi-module music therapy group will contain rhythm-guided walking and singing training, while the rhythm-guided walking group will only include music tempo-guided walking. The primary outcome is the distance of the incremental shuttle walking test. Secondary outcomes include respiratory muscle function, spirometry, lower extremity function, symptoms, quality of life, anxiety and depression levels, physical activity level, training adherence, and safety measurements. The results of this study can contribute to develop and evaluate a home-based music-facilitated rehabilitation program, which has the potential to act as a supplement and/or substitute (according to the needs) for traditional center-based PR in patients with stable COPD.Clinical trial registration: https://classic.clinicaltrials.gov/, NCT05832814.

In this paper, the images of tunnel surface are obtained by tunnel lining rapid inspection system, and tunnel crack forest dataset (TCFD) is established. The disaster characteristics of tunnel cracks are analyzed and summarized. Solutions of tunnel crack segmentation (TCS) method are developed for the detection and recognition of cracks on tunnel lining. According to the image features of the tunnel lining and the optical principal of detection equipment, effective image pre-processing steps are carried out before crack extraction. The tunnel image of TCFD is divided into appropriate number of blocks to magnify the local features of tunnel cracks. Local threshold segmentation method is used to traverse the blocks successively, and the first target block with crack is obtained. The seed in the target block were obtained by adaptive localization method and mapped to the whole image. Region growing is performed through crack seed until complete tunnel crack is extracted. The results show that the precision, recall rate and F-measure of tunnel cracks under the TCS method can reach 92.58%, 93.07% and 92.82% without strong interference. According to the binary images processed by TCS method, the projection images of different types of tunnel cracks and their respective laws are obtained. Furthermore, the TCS method is implemented and deployed as a GUI software application.

Abstract Helmet mounted display systems (HMDs) are high-performance display devices for modern aircraft. We propose a novel method combining event-related potentials (ERPs) and BubbleView to measure cognitive load under different HMD interfaces. The distribution of the subjects’ attention resources is reflected by analyzing the BubbleView, and the input of the subjects’ attention resources on the interface is reflected by analyzing the ERP’s P3b and P2 components. The results showed that the HMD interface with more symmetry and a simple layout had less cognitive load, and subjects paid more attention to the upper portion of the interface. Combining the experimental data of ERP and BubbleView, we can obtain a more comprehensive, objective, and reliable HMD interface evaluation result. This approach has significant implications for the design of digital interfaces and can be utilized for the iterative evaluation of HMD interfaces.

Abstract. Background:. Exercise, as the cornerstone of pulmonary rehabilitation, is recommended to chronic obstructive pulmonary disease (COPD) patients. The underlying molecular basis and metabolic process were not fully elucidated. Methods:. Sprague-Dawley rats were classified into five groups: non-COPD/rest (n = 8), non-COPD/exercise (n = 7), COPD/rest (n = 7), COPD/medium exercise (n = 10), and COPD/intensive exercise (n = 10). COPD animals were exposed to cigarette smoke and lipopolysaccharide instillation for 90 days, while the non-COPD control animals were exposed to room air. Non-COPD/exercise and COPD/medium exercise animals were trained on a treadmill at a decline of 5° and a speed of 15 m/min while animals in the COPD/intensive exercise group were trained at a decline of 5° and a speed of 18 m/min. After eight weeks of exercise/rest, we used ultrasonography, immunohistochemistry, transmission electron microscopy, oxidative capacity of mitochondria, airflow-assisted desorption electrospray ionization-mass spectrometry imaging (AFADESI-MSI), and transcriptomics analyses to assess rectal femoris (RF). Abstract Background There is no universally accepted criterion for assessing exercise-induced desaturation (EID). The purpose of this study is to compare the two methods regularly used for determining EID in COPD patients, as well as to explore the risk factors and predictors related to EID. Methods The 6MWT was performed with continuous SpO2 monitoring on patients with stable COPD. Using two methods (method A: “SpO2rest–SpO2min ≥ 4% and/or SpO2min 0.05), but they were in excellent agreement (Kappa = 0.807, P = 0.001). Logistic regression analyses found that DLCO SB% pred, DLCO/VA% pred, CAT score, mean density, PD15, emphysema volume and %LAA were significant determinants of the EID. For predicting EID, the ROC analysis produced AUC and cutoffs of 0.689 and 50.45% (DLCO SB% pred), 0.707 and 75.0% (DLCO/VA% pred), 0.727 and 15 points (CAT score), 0.691 and − 955.00HU (PD15), 0.671 and − 856.46HU (mean density), 0.668 and 338.14 ml (emphysema volume) and 0.656 and 7.63% (%LAA), respectively. Conclusions Two methods evaluating EID in this research are in a good agreement, method A can find more EID patients by focusing on SpO2min. When conditions are constrained, it is also sufficient to assess EID in COPD patients by method B. In terms of the predictors of EID, DLCO SB% pred, DLCO/VA% pred, CAT score and CT-defined emphysema are all statistically significant test variables to determine EID.

In order to investigate the influence of different experimental conditions and additives on the performance of ultra-light aerated concrete (ULAC), more than 300 ULAC specimens with a dry density of around 160 kg/m3 were prepared in this study. The optimum conditions of the experimental process and the dosage range of three admixtures were obtained by two series of single-factor experiments. The results show that the unification of experimental conditions is necessary for the experimental process; the range of water-binder ratio (w/b) is 0.5 ∼ 0.52; the replacement ratio range of fly ash and silica fume are (Rf, 3∼6 %; Rsi, 3∼4 %); the amount of polypropylene fiber (Mpp) is 3∼5 %. Through orthogonal experimental design, the dependence of the compressive strength, flexural strength, dry density, thermal conductivity upon Rf, Rsi, w/b, Mpp, and conditioning agent (Ac, an inorganic material) of ULAC is here investigated. The impact of four factors and their interactions on all property indexes (PIs) was studied by analysis of variance. A modified multiple-index assessment method was proposed to optimize the parameter combination for four PIs of ULAC. The results show that the Rf is the significant influencing factor of all PIs, and the interactions among the three active additives significantly impact the performance of ULAC. Nevertheless, the interaction between Rf and w/b has little impact. The repeated tests show that the deviations between the property index and the target value obtained by the theoretical prediction model are 2.63 %, 2.59 %, 2 % and 4.79 %, respectively, and according to the comprehensive weighted scoring method, the deviations between them are 2.56 %, 1.44 %, 4 % and 4.6 %, respectively. The deviations obtained by the two methods are less than 5 %, demonstrating that this method could be widely used in the ULAC multi-index system parameter optimization.

In China, the population of wild musk deer, belonging to the family Moschidae, has drastically decreased in recent years owing to human activities and environmental changes. During the 1990s, artificial breeding of Alpine musk deer was conducted in Xinglong Mountain, Gansu Province, China, and their ex situ conservation was explored for over a decade. Ex situ protection is beneficial for expanding the population of animals and maintaining their genetic diversity; however, it can also induce metabolic diseases and parasitic infections and reduce reproductive capacity. The gut microbiota of animals has a considerable impact on host energy metabolism and immune regulation, thereby playing a crucial role in the overall health and reproductive success of the host. In this study, by comparing the differences in the intestinal microbiome of the musk deer according to their place of origin and migration, the changes in their gut microbiota and the influencing factors were explored to provide a theoretical basis for monitoring the health status of the musk deer. We used 16S rRNA high-throughput sequencing technology to analyze the structure and diversity of the gut microbiota of Alpine musk deer in Gansu (G, place of origin) and Sichuan (S, place of migration). The results showed that the dominant bacteria and genera in the intestinal microbiome of captive musk deer were similar in the places of origin and migration, but significant differences were observed in their relative abundance (p < 0.05). Regarding Firmicutes and Actinobacteria, which are related to plant cellulose digestion, the relative abundance in group G was higher than that in group S; regarding Proteobacteria and Verrucomicrobia, which are related to fat and starch intake, the relative abundance in group S was higher than that in group G; the relative abundance of Bacillus and Clostridium sensu stricto, which are related to fiber digestibility, was higher in group G than in group S; the relative abundance of conditional pathogens Acinetobacter and Escherichia–Shigella was higher in group S than in group G. The results of α and β diversity analysis also showed significant differences between the two groups (p < 0.05). The ACE and Shannon indices of musk deer in group G were considerably higher than those in group S, and the Simpson index of musk deer in group S was greater than that in group G, indicating that the abundance and diversity of intestinal microbiome were higher in musk deer of Gansu than those of Sichuan. Comparison of the changes in the intestinal microbiome of the musk deer according to the place of origin and migration showed that the plant cellulose content in the food of the musk deer, the fat content in the concentrated feed, and changes in the feeding environment have an impact on the intestinal microbiome. Effective monitoring of the health and immunity of the musk deer is crucial for ensuring their overall health, which in turn will aid in formulating a scientific and reasonable management plan for their conservation.

Abstract Background With the COVID-19 pandemic continuing, various treatments have become widely practiced. Stem cells have a wide range of applications in the treatment of lung diseases and have therefore been experimentally used to treat patients with COVID-19, but whether the expanded use of stem cells is safe and reliable still lacks enough evidence. To address this issue, we systematically reviewed the safety and efficiency of stem cell therapy in COVID-19 cases. Methods We searched PubMed, Embase, Web of Science, The Cochrane Library, CNKI, WanFang, VIP and SinoMed up to January 18, 2022. The included studies were assessed using the Risk-of-bias tool 1.0 and MINORS instrument. The adverse events, mortality, length of hospital day and laboratory parameters were analyzed by meta-analysis. We adhered to PRISMA reporting guideline. Results We have included 17 studies meeting the inclusion data. There were no significant differences in AEs (OR = 0·39, 95% CI = 0·12 to 1·33, P = 0·13, I2 = 58%) and SAEs (OR = 0·21, 95% CI = 0·04 to 1·03, P = 0·05, I2 = 0%) between stem cell therapy group and control group. Ferula sinkiangensis K. M. Shen (Apiaceae) is distributed in arid desert areas of Xinjiang, and its resin is a traditional Chinese medicine to treat gastrointestinal digestive diseases. To explore bioactive components from F. sinkiangensis, three new lignans and thirteen known components were isolated. The structural elucidation of the components was established utilizing spectroscopic analyses together with ECD calculations. Griess reaction results indicated new compounds 1 and 2 significantly decreased NO production in LPS-stimulated RAW 264.7 macrophages, and ELISA results indicated that they effectively attenuated LPS-induced inflammation by inhibiting TNF-α, IL-1β, and IL-6 expressions. The in silico approach confirmed that compound 1 docked into the receptors with strong binding energies of −5.84~−10.79 kcal/mol. In addition, compound 6 inhibited the proliferation of AGS gastric cancer cells with IC50 values of 15.2 μM by suppressing the cell migration and invasion. This study disclosed that F. sinkiangensis might be a promising potential resource for bioactive components.

After anthocyanin synthesis, a variety of anthocyanin compounds are produced through further methylation, glycosylation, and acylation. However, the effect of the potato methylase gene on anthocyanin biosynthesis has not been reported. Red and purple mutation types appear in tubers of the potato cultivar ‘Purple Viking’ with chimeric skin phenotypes. In this study, transcriptome and anthocyanin metabolome analyses were performed on skin of Purple Viking tubers and associated mutants. According to the metabolome analysis, the transformation of delphinidin into malvidin-3-O-glucoside and petunidin 3-O-glucoside and that of cyanidin into rosinidin O-hexoside and peonidin-3-O-glucoside were hindered in red tubers. Expression of methyltransferase gene OMT30376 was significantly lower in red tubers than in purple ones, whereas the methylation level of OMT30376 was significantly higher in red tubers. In addition, red skin appeared in tubers from purple tuber plants treated with S-adenosylmethionine (SAM), indicating the difference between purple and red was caused by the methylation degree of the gene OMT30376. Thus, the results of the study suggest that the OMT30376 gene is involved in the transformation of anthocyanins in potato tubers. The results also provide an important reference to reveal the regulatory mechanisms of anthocyanin biosynthesis and transformation.

Milk is one of the most common sources of protein in people’s daily lives, and it is also recognized by the World Health Organization (WHO) as one of the eight categories of food allergies to human beings. α-lactalbumin (α-La) is the main cause of milk allergy. In this study, a single-stranded DNA aptamer with high binding affinity to α-La were selected using systematic evolution of ligands by exponential enrichment (SELEX) method. Compared with the full-length sequence, the binding affinity of the truncated aptamer LA-1t for α-La was increased six times using fluorescence analysis. Circular dichroism (CD) indicated that the secondary structure of LA-1t contained a typical hairpin structure. Through the docking simulation of LA-1t and α-La, these experimental results were further explained theoretically, and the recognition mechanism was explained. Finally, the colorimetric and fluorescence signal of boron nitride quantum dots anchored to porous CeO2 nanorods (BNQDs/CeO2) were modulated by FAM-labeled LA-1t to achieve highly selective and sensitive determination of α-La. This dual-mode sensing strategy displayed sensitive recognition for α-La in a linear range of 5–4,000 ng/ml with the LOD was 3.32 ng/ml (colorimetry) and 0.71 ng/ml (fluorescence), respectively. Simultaneously, the colorimetry/fluorescence dual-mode sensing strategy was applied for detecting α-La in spiked real samples and demonstrated good stability and reliability.

Ex situ conservation is an important technique for protecting rare and endangered wildlife, and maintaining stable individual health is crucial to its success. Gut microbiota composition is a critical indicator of animal health and should therefore be closely monitored during ex situ conservation to track impacts on animal health. Forest musk deer (Moschus berezovskii) were historically distributed in Hebei Province, China, however, they are now extinct in the region. Thus, ex situ conservation efforts were conducted in 2016 whereby approximately 50 individuals were artificially migrated from Weinan, Shaanxi to Huailai, Hebei. To monitor gut health of these migrated individuals, we used 16S rRNA high-throughput sequencing technology to examine the microbiota differences between Huailai juvenile and Weinan juvenile groups, and between Huailai adult and Weinan adult groups. Alpha diversity analysis indicated that the richness of microbiota significantly decreased after migration to the Huailai area, and the beta diversity results also showed significant dissimilarity in gut microbial communities, demonstrating the distinct microbial structure differences in the forest musk deer population from the two areas, for both juvenile and adult groups, respectively. In addition, PICRUSt functional profile prediction indicated that the functions of gut digestion and absorption, and degradation of toxic substances were significantly weakened after ex situ conservation. Differences in diet composition between the individuals of the two sites were also observed and the impact of food on gut microbiota compositions within forest musk deer during ex situ conservation was investigated. This study provides a theoretical basis for developing ex situ conservation measures, especially for the protection of forest musk deer.

The effects of mother-infant separation (i.e., weaning) on the physiology, psychology and nutrition of mammalian infants have attracted much attention. Forest musk deer (FMD) is a first-class protected species in China and listed endangered in the IUCN Red List. The captive breeding population is not only an important source for restocking of wild resources, but also a necessary way to supply the market with legal musk. So far, there is no scientific basis for the appropriate separation time of FMD females and their infants. Therefore, we used metagenome sequencing and enzyme-linked immunosorbent assays to study changes in the fecal cortisol concentration, as well as the intestinal microbiome composition and function of females and fawns at three different separation times, i.e., after 80 days, 90 days and 100 days. The results showed that the increment of the cortisol concentration in female FMD increased with increasing lactation time. The increment of cortisol concentration in infant FMD was highest in the 80 days weaning group, but there was no significant difference between the 90 days and the 100 days separation time. Based on the annotation results of COG, KEGG and CAZy databases, the abundance of different functions annotated by the intestinal microbiome of mothers and fawns of the 90 days weaning group changed slightly after separation. Based on the above results, the separation of mother and infant FMD is recommended after 90 days, i.e., the separation time that triggered the lowest rate of weaning stress and that supported a relatively stable gastro-intestinal physiology.

Traditional visual-inertial simultaneous localization and mapping algorithms are usually designed based on CPUs, and they cannot effectively utilize the parallel computing function of GPUs if they are directly transplanted to an embedded board with a GPU module. However, the computing power of embedded devices is limited. It is unreasonable for the visual-inertial simultaneous localization and mapping algorithm to occupy most CPU computing resources while the GPU is idle. In this article, a parallelization scheme for the VINS-Mono algorithm based on GPU parallel computing technology is proposed. Based on the compute unified device architecture, the construction and solution of the incremental equation are parallelized in the nonlinear optimization process of the algorithm, and the parallelization methods provided by cuSOLVER and cuBLAS are used to carry out the marginalization of the algorithm. In addition, the program for the detection and matching of image feature points in the process of optical flow tracking is rewritten in the algorithm to realize the parallelization of optical flow tracking. After parallelization, the algorithm is found to run well on a heterogeneous computing model composed of a CPU and GPU and can fully exploit the parallel computing power of the GPU. The proposed method was tested on an NVIDIA’s Jetson TX2 module and compared with the VINS-Mono algorithm; the speeds of the construction and solution of the incremental equation were found to be the same, but the optical flow tracking and marginalization speed of the proposed scheme exhibited improvements of about 1.5–1.7 times and 1.9 times, respectively.

The influence of the single polycarboxylate superplasticizer (PCE) and blending it mixed hydroxypropyl methylcellulose (HPMC) on hardening moulding quality and surface pulverisation of foam concrete was investigated. An orthogonal experimental design was employed to determine the optimum combination of parameters for four property indexes (PIs) in this paper. A multi-index matrix analysis method was used to evaluate the parameter combinations and obtain the overall optimal performance for the PIs. The effect of calcium formate (CaF) on the early compressive strength of modified foam concrete with the most optimal combination in different density grades was also studied. The results indicate that the incomplete cement hydration reaction is the essential cause of pulverisation, which can be alleviated by adding a suitable PCE. Defoaming and precipitation occur when the PCE incorporated exceeds 0.1%, which can be mitigated by the addition of 0.02 to 0.06% HPMC. The orthogonal analysis indicates that the anti-cracking agent has a more substantial effect on the strength of the foam concrete than the thickening agent. The most significant factor of mechanical properties is PP fiber followed by dispersible latex powder (DLP). The optimal combination of foam concrete is 0.06% HPMC, 0.3% DLP, and 0.5% PP fiber. The early compressive strength of foam concrete can be significantly enhanced by increasing the CaF content. However, increasing the density level results in a decrease in the 28-day compressive strength of the foam concrete.

The shaft misalignment under mixed lubrication is an important factor affecting the running performance of the bearing, which can occur under heavy load and unsatisfactory assembly. This paper presents a misaligned journal mixed lubrication model coupling for the asperity contact effect, elastic deformation, viscosity–temperature, and viscosity–pressure effect. The finite difference method was employed to calculate the model, and an experimental apparatus designed in this paper was used to test the friction and temperature characteristics of the specimens. The results show that the pressure field, film thickness, and elastic deformation of the bearing conformed to asymmetric distribution along the axial direction under misalignment conditions and there was a notable end side effect. In addition, the frictional force and side leakage flow were evidently enhanced with the increase in the inclination angle in a certain range. The experimental results showed that there was a visible wear phenomenon on the end sides of the bush and shaft. The research results are beneficial for understanding the mixed lubrication mechanism of misaligned journal bearing.

In this work, the single-stranded DNA (ssDNA) aptamers specific to florfenicol (FF) and having a high binding affinity were prepared using the magnetic bead-based systematic evolution of ligands by the exponential enrichment technique (MB-SELEX). After 10 rounds of the MB-SELEX screening, aptamers that can simultaneously recognize FF and its metabolite florfenicol amine (FFA) were obtained. The aptamer with the lowest dissociation constant (Kd) was truncated and optimized based on a secondary structure analysis. The optimal aptamer selected was Apt-14t, with a length of 43 nt, and its dissociation constant was 4.66 ± 0.75 nM, which was about 7 times higher than that of the full-length sequence. The potential binding sites and interactions with FF were demonstrated by molecular docking simulations. In addition, a colorimetric strategy for nanogold aptamers was constructed. The linear detection range of this method was 0.00128–500 ng/mL and the actual detection limit was 0.00128 ng/mL. Using this strategy to detect florfenicol in actual milk and eggs samples, the spiked recoveries were 88.9–123.1% and 84.0–112.2%, respectively, and the relative standard deviation was less than 5.6%, showing high accuracy.

Aflatoxin M1 (AFM1), one of the most toxic mycotoxins, is a feed and food contaminant of global concern. To isolate the ssDNA aptamer of AFM1, synthesized magnetic graphene oxide nanomaterials, 12 rounds of subtractive systematic evolution of ligands by exponential enrichment (SELEX) selection were carried out. As a result, 24 candidate aptamers were selected, and their sequence similarity exceeded 97%. Their binding affinity and specificity were further examined by fluorescence and biofilm interferometry (BLI) methods. One aptamer (Apt-5) against AFM1 with a high affinity and specificity was isolated and demonstrated to be the optimal aptamer, whose dissociation constant reached the nanomolar level, Kd = 8.12 ± 1.51 nM. Additionally, molecular docking studies were used to predict the possible binding sites and mechanisms of the two. Based on Apt-5, an unlabeled aptamer-AuNPs colorimetric method was established to detect AFM1 in milk with a linear range of 0.078–10 ng/mL, and the actual detection limit was 0.078 ng/mL. These results demonstrated that this detection technique could be useful for the quantitative determination of AFM1 in milk and dairy products.

Agenesis of the corpus callosum (AgCC) is a congenital brain malformation characterized by the complete or partial failure to develop the corpus callosum. Despite missing the largest white matter bundle connecting the left and right hemispheres of the brain, studies have shown preserved inter-hemispheric communication in individuals with AgCC. It is likely that plasticity provides mechanisms for the brain to adjust in the context of AgCC, as the malformation disrupts programmed developmental brain processes very early on. A proposed candidate for neuroplastic response in individuals with AgCC is strengthening of intra-hemispheric structural connections. In the present study, we explore this hypothesis using a graph-based approach of the structural connectome, which enables intra- and inter-hemispheric analyses at multiple resolutions and quantification of structural characteristics through graph metrics. Structural graph metrics of 19 children with AgCC (13 with complete, 6 with partial AgCC) were compared to those of 29 typically developing controls (TDC). Associations between structural graph metrics and a wide range of neurobehavioral outcomes were examined using a multivariate data-driven approach (Partial Least Squares Correlation, PLSC). Our results provide new evidence suggesting structural strengthening of intra-hemispheric pathways as a neuroplastic response in the acallosal brain, and highlight regional variability in structural connectivity in children with AgCC compared to TDC. There was little evidence that structural graph properties in children with AgCC were associated with neurobehavioral outcomes. To our knowledge, this is the first report leveraging graph theory tools to explicitly characterize whole-brain intra- and inter-hemispheric structural connectivity in AgCC, opening avenues for future research on neuroplastic responses in AgCC.

Musk is a secretion of the forest musk deer (Moschus berezovskii). Normal musk is a brown solid secretion with a light fragrance. In this study, abnormal types of musk, namely, white and black musks, were discovered during the musk collection process. Researchers have long been concerned with the components of musk. Herein, GC-MS, headspace solid-phase microextraction (HS-SPME), and nonmetric multidimensional scaling (NMDS) were used to analyze the nonpolar organic components, volatile organic components, and sample similarities among different musks, respectively. Weaning is an important event for all mammals, including young forest musk deer. However, weaning stress may cause intestinal microbiota-related disorders. Therefore, high-throughput 16S rRNA gene sequencing was applied to study the dynamic changes in intestinal microbiota during pre-weaning (10 days before weaning) and post-weaning (10 days after weaning) in 15 young forest musk deer. We saw that intestinal microbiota diversity in the post-weaning period was significantly higher than that in the pre-weaning period. The most dominant bacterial phyla were similar in the two groups (Firmicutes, Bacteroidetes and Verrucomicrobia). Meanwhile, we applied Linear discriminant analysis effect size (LefSe) to identify the most differentially microbial taxa in the pre-weaning and post-weaning groups. In the post-weaning forest musk deer, the relative abundance of Actinobacteria, Spirochaetes, Ruminococcaceae_UCG-005, Treponema and Prevotella was higher than in the pre-weaning group. However, higher relative abundance of the phyla Bacteroidetes was found in the pre-weaning group compared with that in the post-weaning group. In summary, this research provides a theoretical foundation for the dynamics of young forest musk deer intestinal microbiota during the weaning transition, which may benefit in understanding the growth and health of forest musk deer.

SIRT1 is a stress sensor whose deacetylase activity is increased during cellular stress, but the molecular mechanism is unclear. This study used 16S rRNA high-throughput sequencing technology to examine the differences in gut microbiota between the Père David's deer populations in the Beijing and Shishou areas of China in order to understand the effects of ex situ conservation on the intestinal microflora in the Père David's deer.Results: On the phylum level, the main bacteria found in the Père David's deer populations from both areas were similar: Firmicutes and Bacteroidetes. However, the relative abundances of the two groups were significantly different. Alpha diversity results indicated that there was a difference in the evenness of the microflora between the two groups, and the beta diversity results further indicated that there was a significant difference in the microflora structure between the two groups.Conclusions: During the ex situ conservation process of the Père David's deer, their food sources may change, resulting in differences in the gut microbiota. The intestinal microflora in the Père David's deer from the same area are clustered. Therefore, the impact of changes in food on the gut microbiota of the Père David's deer should be taken into consideration during ex situ conservation.

Water undergoes structural change as it interfaces with hydrophilic surfaces, including the many hydrophilic surfaces within the cell. This interfacial water has become known as "Exclusion Zone (EZ) water" or "fourth-phase water" [1].We tested the hypothesis that anesthetics diminish the amount of EZ water, and that this change may correlate with functional changes in anesthesia. By using the local anesthetics Lidocaine and Bupivacaine as well as a general inhalational anesthetic, Isoflurane, we tracked the EZ size as these anesthetics were introduced.All three anesthetics diminished EZ size in a concentration-dependent manner at concentrations of 0.18 mM and greater for Bupivacaine, 0.85 mM and greater for Lidocaine, and 0.2% for Isoflurane. At extremely low (micromolar) concentrations, however, all three anesthetics increased EZ size.The sharp increase of EZ size associated with micromolar anesthetic concentrations follows a similar pattern to induction of general anesthesia, from the excitation stage (Stage II) to the depression and overdose stages of surgical anesthesia (Stages III and IV). The results are consistent with the hypothesis that anesthetics may act on water, a fundamental organizational component common to all cells.
