Abstract Background Breast cancer (BC) remains the most commonly malignancy among women worldwide. Although early-stage BC typically presents with curative possibilities, advanced-stage disease, especially with metastasis, is significantly limited in terms of effective therapeutic interventions, thereby establishing it as the second leading cause of cancer-related deaths in women. Antibody–Drug Conjugates (ADCs) establish a groundbreaking class of anti-neoplastic agents characterized by high specificity and targeting precision. These agents have been significant in reshaping the therapeutic approach to breast cancer, especially those subtypes with overexpression of the Human Epidermal Growth Factor Receptor 2 (HER2). Comprising monoclonal antibodies, cytotoxic payloads, and conjugative linkers, ADCs function by specifically targeting antigens on cancer cells, thereby facilitating the intracellular delivery of the toxic payload. The present investigation endeavors to synthesize existing primary research outcomes through rigorous bibliometric and data analytical approaches, thereby elucidating the current research landscape, delineating research foci, and identifying potential avenues for future innovation. Methods For bibliometric analysis, a comprehensive data set comprising 2181 entries related to ADCs in breast cancer was retrieved from the Web of Science Core Collection (WoSCC) spanning the years 1999 to 2023. This data was further filtered from the Science Citation Index Expanded (SCI-Expanded). Analysis software tools such as CiteSpace and VOSviewer were employed for multifaceted analyses such as trends of publications, contributions of countries, and burst analytics. In the dimension of clinical trials, we interrogated databases including ClinicalTrials.gov ( https://www.clinicaltrials.gov ) and the WHO International Clinical Trials Registry Platform (ICTRP) ( https://trialsearch.who.int ). A total of 239 clinical trials were initially sourced, among which, 175 were from ClinicalTrials.gov and 64 from ICTRP. After repetitive and correlation-based screening, 119 trials specifically addressing ADC therapeutic strategies in breast cancer were included. Analytical algorithms were executed using Microsoft-based software to evaluate treatment paradigms, emergent research themes, and progress. Results Our investigations signify a growing trend of research on ADCs, with consistent advancements in scientific achievements. The analysis revealed that variables such as economic stratification of nations, healthcare investment paradigms, and disease incidence rates serve as significant determinants in shaping research output. Geographically, the United States emerged as the predominant contributor to the research corpus (36.56%), closely followed by China (21.33%). The underpinning of research accomplishments was found to be significantly bolstered by advancements in molecular biology, immunology, and genetic research. Moreover, the advent of nuclear magnetic resonance diagnostic modalities has contributed saliently to the diagnostic and therapeutic management of breast cancer. Conclusion Our study provides a comprehensive overview of the ADC research landscape through rigorous bibliometric and clinical trial evaluations. At present, the ADC arena has witnessed the successful development and FDA approval of 14 distinct agents, substantially improving the clinical outcomes for a broad spectrum of oncological patients. Future research imperatives may include the exploration of ADCs targeting mutated oncoproteins, dual-specificity ADCs, combination payload strategies, peptide-drug conjugates (PDCs), and non-internalizing ADC modalities. With sustained academic and clinical focus, the ADC domain is poised for transformative advancements in targeted therapeutics across a variety of malignancies.

Abstract Background Thyroid cancer is a leading endocrine malignancy, with anaplastic and medullary subtypes posing treatment challenges. Existing therapies have limited efficacy, highlighting a need for innovative approaches. Methods We analyzed 658 articles and 87 eligible clinical trials using bibliometric tools and database searches, including annual publication and citation trends, were executed using Web of Science, CiteSpace, and VOS Viewer. Results Post-2018, there is a surge in thyroid cancer immunotherapy research, primarily from China and the University of Pisa. Of the 87 trials, 32 were Phase I and 55 were Phase II, mostly exploring combination therapies involving immune checkpoint inhibitors. Conclusion The study's dual approach verifies the swift advancement of thyroid cancer immunotherapy from diverse perspectives. Immune checkpoint inhibitors have become the preferred regimen for advanced MTC and ATC in late therapeutic lines. However, since ICB plays a pivotal role in ATC, current clinical trial data show that ATC patients account for more and the curative effect is more accurate. Anticipated future developments are inclined toward combination regimens integrating immunotherapy with chemotherapy or targeted therapies. Emerging approaches, such as bispecific antibodies, cytokine-based therapies, and adoptive cell therapies like CAR-T and TCR-T, are exhibiting considerable potential. Upcoming research is expected to concentrate on refining the tumor immune milieu and discovering novel biomarkers germane to immunotherapeutic interventions.

An extensive number of farmlands in the Poyang Lake region of China have been submerged due to the impact of flood disasters, resulting in significant agricultural economic losses. Therefore, it is of great importance to conduct the long-term temporal monitoring of flood-induced water body changes using remote sensing technology. However, the scarcity of optical images and the complex, fragmented terrain are pressing issues in the current water body extraction efforts in southern hilly regions, particularly due to difficulties in distinguishing shadows from numerous mountain and water bodies. For this purpose, this study employs Sentinel-1 synthetic aperture radar (SAR) data, complemented by water indices and terrain features, to conduct research in the Poyang Lake area. The results indicate that the proposed multi-source data water extraction method based on microwave remote sensing data can quickly and accurately extract a large range of water bodies and realize long-time monitoring, thus proving a new technical means for the accurate extraction of floodwater bodies in the Poyang Lake region. Moreover, the comparison of several methods reveals that CAU-Net, which utilizes multi-band imagery as the input and incorporates a channel attention mechanism, demonstrated the best extraction performance, achieving an impressive overall accuracy of 98.71%. This represents a 0.12% improvement compared to the original U-Net model. Moreover, compared to the thresholding, decision tree, and random forest methods, CAU-Net exhibited a significant enhancement in extracting flood-induced water bodies, making it more suitable for floodwater extraction in the hilly Poyang Lake region. During this flood monitoring period, the water extent in the Poyang Lake area rapidly expanded and subsequently declined gradually. The peak water area reached 4080 km2 at the height of the disaster. The severely affected areas were primarily concentrated in Yongxiu County, Poyang County, Xinjian District, and Yugan County.

BackgroundCervical cancer is the fourth most cancer and the fourth leading cause of cancer-related deaths in women worldwide. Current treatment for patients with advanced cervical cancer is limited. And in the urgent demand for novel effective therapies both as the first and the second line treatment for these patients, immunotherapy is developing fast and has made some achievements.MethodsThis study incorporated 1,255 topic-related articles and reviews from 1999 to 2022 in the Web of Science Core Collection (WoSCC). The WoS platform, Citespace, and VOS viewer provided the annual distribution of publications and citations, the analysis of researching countries and institutions, references, keywords (co-occurrence analysis, burst analysis, and timeline view analysis), and researching authors, respectively. For clinical trials, 720 trials and 114 trials from ClinicalTrials.gov and ICTRP were retrieved, respectively. And 296 trials were finally incorporated into the analysis.ResultsThe scientometric analysis showed that the study of immunotherapies in cervical cancer developed fast in recent years. Most publications were from the United States, followed by China. Seven of the top 10 co-cited references belong to clinical trials, and five of them were published in recent five years. There are lots of clinical trials us specific treatment patterns, some of which have represented excellent effects.ConclusionsBoth the scientometric analysis of the 1,255 publications and the analysis of clinical trials showed that the field of immunotherapies in cervical cancer developed so fast in recent years. It was found that a lot of clinical trials using various immunotherapies (mainly vaccine therapy, adoptive cell therapy, immune checkpoint blockade, and antibody-drug conjugate) for advanced cervical cancer are currently ongoing or have represented considerable effect. Centered in immunotherapies, immune checkpoint blockades have represented great efficacy and huge potential, especially combined with other therapies such as chemotherapy, targeted therapy, and other immunotherapies.

BackgroundEpithelial–mesenchymal transition (EMT) is a key factor in the invasion and migration of glioma cells, and the study of EMT in gliomas has become a hot topic over the past decade. Scientometric analysis is gaining more attention since it can obtain hot topics and emerging trends in a research field. This article analyzed the research related to EMT in gliomas for the first time, including descriptions of research situations, evaluations of research foci, and predictions of emerging trends.MethodsWe searched the topic-related original articles from January 2012 to December 2021 in the Web of Science Core Collection (WoSCC) by using a specific strategy, and a total of 1,217 publications were obtained. The WoS platform, VOS viewer, and CiteSpace were used to analyze the annual distribution of publications and citations, authors and density of keywords, and other analyses including countries, institutions, references, clustering, burst analysis, and the timeline view of keywords.ResultsScientometric analysis identified that the study of EMT in gliomas has developed fast and received continuous attention in the last decade. Based on the results of data analysis, most publications on the topic came from China, and the United States had the highest betweenness centrality. The top 10 co-cited references revealed the landmark documents that had greatly promoted the development of this field. The major focus is on the cellular and molecular mechanisms of EMT in gliomas, and the therapy related to EMT target and non-coding RNAs has been developing fast in recent years.ConclusionsThis study revealed the intimate connections between EMT and gliomas, and the complex mechanisms regulating EMT in gliomas had been studied widely in the last decade. Exploring the deep mechanisms of EMT in gliomas is the foundation of the targeted inhibitions, which can promote the development of therapies for gliomas.

Paraquat (PQ) is a ubiquitously applied herbicide. Long-term PQ exposure with low dose has been reported to induce abnormal expression of long non-coding RNAs (lncRNAs) in brain nerve cells, which could further lead to Parkinson's disease (PD). N6-methyladenosine (m6A) modification has recently been identified as having an important role in regulating the function of lncRNAs. However, how m6A modification regulates lncRNAs following PQ exposure remains largely unknown. Herein, this study reported m6A modification of lncRNAs in mouse neuroblastoma cells (Neuro-2a) following PQ induced reactive oxide species (ROS). M6A sequencing was performed to explore the m6A modificated pattern of lncRNAs in Neuro-2a cells which were treated with 200 μM PQ for 3 h. It was found that PQ hypermethylated total RNA and changed the expression of m6A methyltransferase and demethylase proteins, which leading to the alteration of m6A modification of lncRNAs. Furthermore, the functional analysis further revealed that N-acetyl-L-cysteine (NAC)，a ROS scavengers, partly reversed PQ-induced distinct m6A modificated pattern of lncRNAs. In addition, tow specific m6A modified lncRNAs were identified: cell division cycle 5-like (lncRNA CDC5L) and signal transducer and activator of transcription 3 (lncRNA STAT3), which could influence downstream autophagy related biological function. In summary, this work could potentially contribute to the new insight of lncRNAs m6A modification mechanism in the field of environmental toxicology.

Sharing economy (SE) platforms have become increasingly popular. However, some platform enterprises have engaged in problematic behavior, such as collecting and using personal information in serious violation of the relevant laws and regulations, making improving existing institutional mechanisms an urgent matter. This study develops a conceptual model based on institution-based trust to examine the effect of SE institutional mechanisms on consumer trust in sharing platforms and continuance intention toward the SE in China. In addition, this study investigates the moderating role of gender in the path relationship between the second-order construct “perceived effectiveness of sharing economy institutional mechanisms” (PESEIM) and consumer trust. SPSS23.0 and SmartPLS3.0 were used to empirically test the data collected from DiDi users in China. The results of the analysis indicate that PESEIM has significant positive impacts on consumer trust, which in turn enhances consumers’ continuance intention to use the DiDi platform, and that consumer trust can also mitigate perceived risk while promoting continuance intention. Importance-Performance Map Analysis (IPMA) results show that government regulation and privacy assurance are critical factors for sustaining continuance intention toward ride-sharing platforms. Furthermore, the results of multigroup analysis show that PESEIM has a stronger impact on male users than female consumers in building consumer trust.

This study presents a high-fidelity and high-efficiency digital class-D audio power amplifier (CDA), which consists of digital and analog modules. To realize a compatible digital input, a fully digital audio digital-to-analog converter (DAC) is implemented on MATLAB and Xilinx System Generator, which consists of a 16x interpolation filter, a fourth-order four-bit quantized delta-sigma (ΔΣ) modulator, and a uniform-sampling pulse width modulator. The CDA utilizes the closed-loop negative feedback and loop-filtering technologies to minimize distortion. The audio DAC, which is based on a field-programmable gate array, consumes 0.128 W and uses 7100 LUTs, which achieves 11.2% of the resource utilization rate. The analog module is fabricated in a 0.18 µm BCD technology. The postlayout simulation results show that the CDA delivers an output power of 1 W with 93.3% efficiency to a 4 Ω speaker and achieves 0.0138% of the total harmonic distortion (THD) with a transient noise for a 1 kHz input sinusoidal test tone and 3.6 V supply. The output power reaches up to 2.73 W for 1% THD (with transient noise). The proposed amplifier occupies an active area of 1 mm2.

High-speed rail (HSR) is often claimed to bring different regions and cities closer together by shortening travel times, which can reduce the costs and increase enterprises productivity to promote a sustainable economy. However, another view argues that HSR transfers economic activities from peripheral cities to core cities, resulting in unbalanced regional economic development and damaging the sustainability of the economy. Based on microdata from China, this paper empirically investigates the impact of HSR on the enterprises productivity in both core cities and peripheral cities and explores the impact mechanism from the perspective of allocation effect and distribution effect caused by HSR. The results show that the connection of HSR positively affects the enterprises productivity in core cities, while it negatively affects the enterprises productivity in peripheral cities, with effect values of 1.38% and -8.45%, respectively. The conclusion still holds after endogenous treatment and multiple robustness tests are conducted. Additionally, the allocation effect analysis shows that the market access caused by HSR has an optimization effect on the resource allocation efficiency of both core cities and peripheral cities. The distribution effect analysis reveals that the distribution of enterprise productivity in peripheral cities has market heterogeneity, regional heterogeneity, and location heterogeneity. The important policy significance of this paper is that, in order to promote the sustainable development of enterprises and the economy, it should reduce policy restrictions and promote the effective flow of capital and talents, carry out the dislocation development of industry for peripheral cities, and “build a nest to attract the phoenix.”

In duty-cycled wireless sensor networks, energy efficiency and packet latency are two most important metrics in the design of medium access control and routing algorithms. However, these two problems cannot be addressed well at the same time. In this article, we investigate the trade-off between energy consumption and latency and formulate them into a multi-objective optimization problem. By applying the single exponential smoothing method, we estimate the amount of data of next period and design two optimal sleep time controllers according to time scheduling model of network, so as to dynamically adjust the duty cycle of end node. Our controllers also consider the residual energy of end node. Finally, we propose two dynamic and adaptive medium access control algorithms for synchronous and asynchronous duty-cycled wireless sensor networks, respectively. We evaluate our protocols with different parameters and compare them with existing works. Performance results show that our proposed algorithms can balance power consumption among nodes and improve power efficiency while guaranteeing packet latency is minimized.

A series of new 2-phenyl-quinoline-4-carboxylic acid derivatives was synthesized starting from aniline, 2-nitrobenzaldehyde, pyruvic acid followed by Doebner reaction, amidation, reduction, acylation and amination. All of the newly-synthesized compounds were characterized by 1H-NMR, 13C-NMR and HRMS. The antibacterial activities of these compounds against Gram-negative (Escherichia coli, Pseudomonas aeruginosa) and Gram-positive bacteria (Staphylococcus aureus, Bacillus subtilis), as well as one strain of methicillin-resistant Staphylococcus aureus (MRSA) bacteria were evaluated by the agar diffusion method (zone of inhibition) and a broth dilution method (minimum inhibitory concentration (MIC)), and their structure-activity relationships were obtained and discussed. The results revealed that some compounds displayed good antibacterial activity against Staphylococcus aureus, and Compounds 5a4 and 5a7 showed the best inhibition with an MIC value of 64 μg/mL against Staphylococcus aureus and with an MIC value of 128 μg/mL against Escherichia coli, respectively. The results of the MTT assay illustrated the low cytotoxicity of Compound 5a4.
