Oxalate metal–organic framework derived atomic Ru3+-doped Co(OH)2 nanosheets are fabricated for highly efficient hydrogen evolution reaction, showing a very small overpotential of 36 mV at 30 mA cm−2 in 1.0 M KOH.

Osteoarthritis scaffold-based grafts fail because of poor integration with the surrounding soft tissue and inadequate tribological properties. To circumvent this, we propose electrospun poly(ε-caprolactone)/zein-based scaffolds owing to their biomimetic capabilities. The scaffold surfaces were characterized using Fourier-transform infrared spectroscopy, X-ray photoelectron spectroscopy, static water contact angles, and profilometry. Scaffold biocompatibility properties were assessed by measuring protein adsorption (Bicinchoninic Acid Assay), cell spreading (stained F-actin), and metabolic activity (PrestoBlue™ Cell Viability Reagent) of primary bovine chondrocytes. The data show that zein surface segregation in the membranes not only completely changed the hydrophobic behavior of the materials, but also increased the cell yield and metabolic activity on the scaffolds. The surface segregation is verified by the infrared peak at 1658 cm−1, along with the presence and increase in N1 content in the survey XPS. This observation could explain the decrease in the water contact angles from 125° to approximately 60° in zein-comprised materials and the decrease in the protein adsorption of both bovine serum albumin and synovial fluid by half. Surface nano roughness in the PCL/zein samples additionally benefited the radial spreading of bovine chondrocytes. This study showed that co-electrospun PCL/zein scaffolds have promising surface and biocompatibility properties for use in articular-tissue-engineering applications.

The assembly of nanomaterials into suprastructures offers the possibility to fabricate larger scale functional materials, whose inner structure strongly influences their functionality for a vast range of applications. In spite of the many current strategies, achieving multi-compartment structures in a targeted and versatile way remains highly challenging. Here, we describe a controllable and straightforward route to create uniform suprastructured materials with a multi-compartmentalized architecture by confining primary nanocapsules into droplets using a cross-junction microfluidic device. Following solvent evaporation from the droplets, the nanocapsules spontaneously assemble into precisely sized multi-compartment particles, which we term supracapsules. Thanks to the process, each spatially separated nanocapsule unit retains its cargo and functionalities within the resulting supracapsules. However, new collective properties emerge, and, particularly, programmable release profiles that are distinct from those of single-compartment capsules. Finally, the suprastructures can be disassembled into single-compartment units by applying ultra-sonication, switching their release to a burst-release mode. Hypothesis: The synthesis of compositionally heterogeneous particles is central to the development of complex colloidal units for self-assembly and self-propulsion. Yet, as the complexity of particles grows, synthesis becomes more prone to “errors”. We hypothesize that alternating-current dielectrophoretic forces can efficiently sort Janus particles, as a function of patch size and material, and colloidal dumbbells by size. Experiments: We prepared Janus particles with different patch size and material by physical vapor deposition and colloidal dumbbells via capillarity-assisted particle assembly. We then performed sorting experiments in a microfluidic chip comprising electrodes with asymmetric orifices, specifically exploiting the dielectric contrast between different portions of the particles or their size difference to steer them towards different outlets. Findings: We calculated that the DEP force for Janus particles may switch from positive to negative as a function of composition at a critical AC frequency, thus enabling sorting different particles crossing the electrodes’ region. The predictions are confirmed by optical microscopy experiments. We also show that intact and “broken” dumbbells can be simply separated as they experience different DEP forces. Diabetic peripheral neuropathy (DPN) is characterized by spontaneous pain in the extremities. Incidence of DPN continues to rise with the global diabetes epidemic. However, there remains a lack of safe, effective analgesics to control this chronic painful condition. Dorsal root ganglia (DRG) contain soma of sensory neurons and modulate sensory signal transduction into the central nervous system. In this study, we aimed to gain a deeper understanding of changes in molecular pathways in the DRG of DPN patients with chronic pain. We recently reported transcriptomic changes in the DRG with DPN. Here, we expand upon those results with integrated metabolomic, proteomic, and phospho-proteomic analyses to compare the molecular profiles of DRG from DPN donors and DRG from control donors without diabetes or chronic pain. Our analyses identified decreases of select amino acids and phospholipid metabolites in the DRG from DPN donors, which are important for cellular maintenance. Additionally, our analyses revealed changes suggestive of extracellular matrix (ECM) remodeling and altered mRNA processing. These results reveal new insights into changes in the molecular profiles associated with DPN.

SummaryThe mechanisms underlying facial and oral pain are still incompletely understood, posing major therapeutic challenges. Cyclin-dependent kinase 5 (Cdk5) is a key neuronal kinase involved in pain signaling. However, the regulatory roles of Cdk5 in orofacial pain signaling and the possibility of therapeutic intervention at the level of mouse trigeminal ganglion primary neurons remain elusive. In this study, we used optimized intravital imaging to directly compare trigeminal neuronal activities after mechanical, thermal, and chemical stimulation. We then tested whether facial inflammatory pain in mice could be alleviated by the Cdk5 inhibitor peptide TFP5. We demonstrated regulation of total Ca2+ intensities by Cdk5 activity using transgenic and knockout mouse models. In mice with orofacial inflammation, application of TFP5 specifically decreased total Ca2+ intensities in response to noxious stimuli. It also alleviated inflammation-induced allodynia by inhibiting activation of trigeminal peripheral sensory neurons. Cdk5 inhibitors may provide promising non-opioid candidates for pain treatment.

Control over the surface roughness of colloidal particles offers exciting opportunities to tailor the properties and the processing of a broad range of soft matter systems. Moreover, identifying surface roughness as a design parameter reveals the possibility to connect seemingly distinct phenomena and materials via the role played by roughness effects. In this feature article, we concisely review some approaches to synthesize and characterize rough colloidal particles, with a focus on model spherical colloids. We then discuss the impact that surface roughness has on both the high-shear rheology of dense suspensions and the stabilization of Pickering emulsions. Commenting on developments of our own research, we aim to offer an original perspective for a property-oriented development of colloidal particles that transcends classical divisions between materials and processes toward innovative solutions.

One of the dreams of nanotechnology is to create tiny objects, nanobots, that are able to perform difficult tasks in dimensions and locations that are not directly accessible. One basic function of these nanobots is motility. Movements created by self-propelled micro- and nanovehicles are usually dependent on the production of propellants from catalytic reactions of fuels present in the environment. Developing self-powered nanovehicles with internally stored fuels that display motion regulated by external stimuli represents an intriguing and challenging alternative. Herein, a one-step preparation of fuel-containing nanovehicles that feature a motion that can be regulated by external stimuli is reported. Nanovehicles are prepared via a sol-gel process confined at the oil/water interface of miniemulsions. The nanovehicles display shapes ranging from mushroom-like to truncated cones and a core-shell structure so that the silica shell acts as a hull for the nanovehicles while the core is used to store the fuel. Azo-based initiators are loaded in the nanovehicles, which are activated to release nitrogen gas upon increase of temperature or exposure to UV light. Enhanced diffusion of nanovehicles is achieved upon decomposition of the fuel.

Superparamagnetism exists only in nanocrystals, and to endow micro/macro-materials with superparamagnetism, superparamagnetic nanoparticles have to be assembled into complex materials. Most techniques currently used to produce such assemblies are inefficient in terms of time and material. Herein, we used evaporation-guided assembly to produce superparamagnetic supraparticles by drying ferrofluid droplets on a superamphiphobic substrate in the presence of an external magnetic field. By tuning the concentration of ferrofluid droplets and controlling the magnetic field, barrel-like, cone-like, and two-tower-like supraparticles were obtained. These assembled supraparticles preserved the superparamagnetism of the original nanoparticles. Moreover, other colloids can easily be integrated into the ferrofluid suspension to produce, by co-assembly, anisotropic binary supraparticles with additional functions. Additionally, the magnetic and anisotropic nature of the resulting supraparticles was harnessed to prepare magnetically actuable microswimmers.

We study the effect of entanglements on the glass transition of high molecular weight polymers, by the comparison of single-chain nanoparticles (SCNPs) and equilibrated melts of high-molecular weight polystyrene of identical molecular weight. SCNPs were prepared by electrospraying technique and characterized using scanning electron microscopy and atomic force microscopy techniques. Differential scanning calorimetry, Brillouin light spectroscopy, and rheological experiments around the glass transition were compared. In parallel, entangled and disentangled polymer melts were also compared under cooling from molecular dynamics simulations based on a bead-spring polymer model. While experiments suggest a small decrease in the glass transition temperature of films of nanoparticles in comparison to entangled melts, simulations do not observe any significant difference, despite rather different chain conformations.

Durable and biocompatible superhydrophobic surfaces are of significant potential use in biomedical applications. Here, a nonfluorinated, elastic, superhydrophobic film that can be used for medical wound dressings to enhance their hemostasis function is introduced. The film is formed by titanium dioxide nanoparticles, which are chemically crosslinked in a poly(dimethylsiloxane) (PDMS) matrix. The PDMS crosslinks result in large strain elasticity of the film, so that it conforms to deformations of the substrate. The photocatalytic activity of the titanium dioxide provides surfaces with both self-cleaning and antibacterial properties. Facile coating of conventional wound dressings is demonstrated with this composite film and then resulting improvement for hemostasis. High gas permeability and water repellency of the film will provide additional benefit for medical applications.

Visualization of dynamic cellular activity has greatly expanded our understanding of brain function. Recently, there has been an increasing number of studies imaging rodent brain activity in real time. However, traditional in vivo calcium imaging technology has been limited to superficial brain structures. Because the trigeminal ganglion (TG) is located deep within the cranial cavity of mice, few studies have been able to access to it. To circumvent this limitation, overlying brain tissue must be removed to expose the TG so that optical recording can access deep brain neural ensembles. This unit describes a procedure for conducting non-survival surgery to visualize the TG in live mice. Obtaining large ensembles of direct, real-time readouts of sensory neuron signaling, providing temporal and spatial information across the TG, will help to define the cellular basis of orofacial somatic sensing and pain perception. © 2019 by John Wiley & Sons, Inc.

Electrospun polymer mats are widely used in tissue engineering, wearable electronics, and water purification. However, in many environments, the polymer nanofibers prepared by electrospinning suffer from biofouling during long-term usage, resulting in persistent infections and device damage. Herein, we describe the fabrication of polymer mats with CeO2–x nanorods that can prevent biofouling in an aqueous environment. The embedded CeO2–x nanorods are functional mimics of natural haloperoxidases that catalyze the oxidative bromination of Br– and H2O2 to HOBr. The generated HOBr, a natural signaling molecule, disrupted the bacterial quorum sensing, a critical step in biofilm formation. The polymer fibers provide porous structures with high water wettability, and the embedded cerium oxide nanozymes act as a catalyst that can efficiently trigger oxidative bromination, as shown by a haloperoxidase assay. In this work, Gordonia amicalisa was used to treat two kinds of vulcanized synthetic rubbers, vulcanized isoprene rubber (v-IR) and vulcanized styrene butadiene rubber (v-SBR). The effects of microbial treatment on vulcanized rubbers were evaluated by changes of crosslinking density, sol fraction, morphology, element content and chemical group of treated rubber samples. The results showed that G. amicalisa can break C C bond mainly on v-IR and S S bonds mainly on v-SBR. After microbial treatment of 20 days, the crosslink densities of v-IR and v-SBR decreased by 13.7% and 22.1%, respectively. The carbon content on v-IR surface was decreased by 9.1%. FTIR analysis showed that C C bonds transferred into C O bonds, indicating the main chain scission. Regarding to the v-SBR, sulfur content on its surface was decreased by 22.9%, S S bonds were broken and S O producing, indicating the sulfur crosslink scission. Furthermore, the inducing mechanism of desulfurizing enzyme produce from G. amicalisa was investigated. v-SBR and v-IR were used as enzyme inducers, and dibenzothiophene was used as substrate reacting with desulfurizing-enzyme. The liquid chromatography and ultraviolet spectrum analysis showed that v-SBR was the specific substrates that can induce G. amicalisa to secret desulfurizing enzyme, making desulfurizing reaction happen.

Autonomous highlighting of damage in protective polymer coatings allows on-demand maintenance and enables prolongation of the lifetimes of the coated materials. To monitor the entire cycle of damage occurrence and successful healing, one must be able to visualize both processes and display the current health-state of the coating. Herein, we equipped coatings with nanocapsules that can self-indicate their mechanical micro-damage via color development. Hence, whenever the coating was damaged, the capsules broke and highlighted the damaged spot. As a second feature, the color development was reversed and discoloration occurred in the presence of (self-)healing compounds, allowing the user to monitor the healing process. Thus, in the first step, damages were highlighted via color “turn-on” and in the subsequent step a propagating healing reaction “turns-off” the damage indication system to trace the healing reaction and allow monitoring of the entire health cycle.

Kisspeptin plays a critical role in pubertal timing and reproductive function. In rodents, kisspeptin perikarya within the hypothalamic arcuate (ARC) and anteroventral periventricular (AVPV) nuclei are thought to be involved in LH pulse and surge generation, respectively. Using bilateral microinjections of recombinant adeno-associated virus encoding kisspeptin antisense into the ARC or AVPV of female rats at postnatal day 10, we investigated the relative importance of these two kisspeptin populations in the control of pubertal timing, estrous cyclicity, and LH surge and pulse generation. A 37% knockdown of kisspeptin in the AVPV resulted in a significant delay in vaginal opening and first vaginal estrous, abnormal estrous cyclicity, and reduction in the occurrence of spontaneous LH surges, although these retained normal amplitude. This AVPV knockdown had no effect on LH pulse frequency, measured after ovariectomy. A 32% reduction of kisspeptin in the ARC had no effect on the onset of puberty but resulted in abnormal estrous cyclicity and decreased LH pulse frequency. Additionally, the knockdown of kisspeptin in the ARC decreased the amplitude but not the incidence of LH surges. These results might suggest that the role of AVPV kisspeptin in the control of pubertal timing is particularly sensitive to perturbation. In accordance with our previous studies, ARC kisspeptin signaling was critical for normal pulsatile LH secretion in female rats. Despite the widely reported role of AVPV kisspeptin neurons in LH surge generation, this study suggests that both AVPV and ARC populations are essential for normal LH surges and estrous cyclicity.

Kisspeptin (encoded by KISS1) is a crucial activator of reproductive function. The role of kisspeptin has been studied extensively within the hypothalamus but little is known about its significance in other areas of the brain. KISS1 and its cognate receptor are expressed in the amygdala, a key limbic brain structure with inhibitory projections to hypothalamic centers involved in gonadotropin secretion. We therefore hypothesized that kisspeptin has effects on neuronal activation and reproductive pathways beyond the hypothalamus and particularly within the amygdala. To test this, we mapped brain neuronal activity (using manganese-enhanced MRI) associated with peripheral kisspeptin administration in rodents. We also investigated functional relevance by measuring the gonadotropin response to direct intra-medial amygdala (MeA) administration of kisspeptin and kisspeptin antagonist. Peripheral kisspeptin administration resulted in a marked decrease in signal intensity in the amygdala compared to vehicle alone. This was associated with an increase in luteinizing hormone (LH) secretion. In addition, intra-MeA administration of kisspeptin resulted in increased LH secretion, while blocking endogenous kisspeptin signaling within the amygdala by administering intra-MeA kisspeptin antagonist decreased both LH secretion and LH pulse frequency. We provide evidence for the first time that neuronal activity within the amygdala is decreased by peripheral kisspeptin administration and that kisspeptin signaling within the amygdala contributes to the modulation of gonadotropin release and pulsatility. Our data suggest that kisspeptin is a ‘master regulator’ of reproductive physiology, integrating limbic circuits with the regulation of gonadotropin-releasing hormone neurons and reproductive hormone secretion.

Cdk5 is a key neuronal kinase necessary for proper brain development, which has recently been implicated in modulating nociception. Conditional deletion of Cdk5 in pain-sensing neurons attenuates pain responses to heat in both the periphery and orofacial regions. Cdk5 activity is regulated by binding to the activators p35 and p39, both of which possess a cyclin box. Our previous examination of the nociceptive role of the well-characterized Cdk5 activator p35 using mice that either lack or overexpress this regulatory subunit demonstrated that Cdk5/p35 activity affects mechanical, chemical, and thermal nociception. In contrast, the nociceptive role of Cdk5’s other less-studied activator p39 is unknown. Here, we report that the knockout of p39 in mice did not affect orofacial and peripheral nociception. The lack of any algesic response to nociceptive stimuli in the p39 knockout mice contrasts with the hypoalgesic effects that result from the deletion of p35. Our data demonstrate different and nonoverlapping roles of Cdk5 activators in the regulation of orofacial as well as peripheral nociception with a crucial role for Cdk5/p35 in pain signaling.

In present work, three non-ionic surfactants (Tween 20, Tween 60 and Tween 80) were used to improve the affinity between the lipophilic ground tire rubber (GTR) and hydrophilic microbes. The growth characteristic of the Sphingomonas in the co-culture process was studied. The effects of different Tween surfactants on the biodesulfurization of GTR were investigated. Tween 20, among these three surfactants, showed best effect on enhancement of biodesulfurization. Results of SEM-EDS showed that the amount of sulfur in rubber surface layer of 0–4 μm was significantly decreased by 67%. XPS analysis data showed that the area of S–S bonds and S–C bonds were decreased while the S–O bonds were obviously increased. The mechanical properties of desulfurized-GTR/styrene butadiene rubber composite were improved. Also, the mechanism of the surfactants in improving the affinity between the GTR and Sphingomonas was proposed.

Kisspeptin plays a pivotal role in pubertal onset and reproductive function. In rodents, kisspeptin perikarya are located in 2 major populations: the anteroventral periventricular nucleus and the hypothalamic arcuate nucleus (ARC). These nuclei are believed to play functionally distinct roles in the control of reproduction. The anteroventral periventricular nucleus population is thought to be critical in the generation of the LH surge. However, the physiological role played by the ARC kisspeptin neurons remains to be fully elucidated. We used bilateral stereotactic injection of recombinant adeno-associated virus encoding kisspeptin antisense into the ARC of adult female rats to investigate the physiological role of kisspeptin neurons in this nucleus. Female rats with kisspeptin knockdown in the ARC displayed a significantly reduced number of both regular and complete oestrous cycles and significantly longer cycles over the 100-day period of the study. Further, kisspeptin knockdown in the ARC resulted in a decrease in LH pulse frequency. These data suggest that maintenance of ARC-kisspeptin levels is essential for normal pulsatile LH release and oestrous cyclicity.

Acute systemic stress disrupts reproductive function by inhibiting pulsatile gonadotropin secretion. The underlying mechanism involves stress-induced suppression of the GnRH pulse generator, the functional unit of which is considered to be the hypothalamic arcuate nucleus kisspeptin/neurokinin B/dynorphin A neurons. Agonists of the neurokinin B (NKB) receptor (NK3R) have been shown to suppress the GnRH pulse generator, in a dynorphin A (Dyn)-dependent fashion, under hypoestrogenic conditions, and Dyn has been well documented to mediate several stress-related central regulatory functions. We hypothesized that the NKB/Dyn signaling cascade is required for stress-induced suppression of the GnRH pulse generator. To investigate this ovariectomized rats, iv administered with Escherichia coli lipopolysaccharide (LPS) following intracerebroventricular pretreatment with NK3R or κ-opioid receptor (Dyn receptor) antagonists, were subjected to frequent blood sampling for hormone analysis. Antagonism of NK3R, but not κ-opioid receptor, blocked the suppressive effect of LPS challenge on LH pulse frequency. Neither antagonist affected LPS-induced corticosterone secretion. Hypothalamic arcuate nucleus NKB neurons project to the paraventricular nucleus, the major hypothalamic source of the stress-related neuropeptides CRH and arginine vasopressin (AVP), which have been implicated in the stress-induced suppression of the hypothalamic-pituitary-gonadal axis. A separate group of ovariectomized rats was, therefore, used to address the potential involvement of central CRH and/or AVP signaling in the suppression of LH pulsatility induced by intracerebroventricular administration of a selective NK3R agonist, senktide. Neither AVP nor CRH receptor antagonists affected the senktide-induced suppression of the LH pulse; however, antagonism of type 2 CRH receptors attenuated the accompanying elevation of corticosterone levels. These data indicate that the suppression of the GnRH pulse generator by acute systemic stress requires hypothalamic NKB/NK3R signaling and that any involvement of CRH therewith is functionally upstream of NKB.

A microbe with desulfurizing capability, Alicyclobacillus sp., was selected to recycle waste latex rubber (WLR). The growth characteristics of the microorganism and the technical conditions in the co-culture desulfurization process were studied. The desulfurization effect of Alicyclobacillus sp. on the WLR was characterized, and the mechanism for the microbial desulfurization of WLR was tentatively explored. The results showed that adding 5% (w/v) WLR into medium had little effect on the growth of Alicyclobacillus sp. The surfactant polysorbate 80 (Tween 80) had a toxic effect on Alicyclobacillus sp., but the growth of the microbe was vigorous if the proper technique was used: the mixing of WLR with Tween 80, followed by the addition of the mixture into the culture media. With the increase of desulfurization time, the swelling value of desulfurizated waste latex rubber (DWLR) increased, but the crosslink density decreased. After co-culture desulfurization for 8–10 days, a DWLR with good desulfurization effect was obtained. The mechanical properties of natural rubber (NR)/DWLR composite improved significantly over those of NR/WLR composite. XPS and FTIR results revealed that Alicyclobacillus sp. could break the crosslinked sulfur bonds and oxidize them to sulfones groups. The increase of O element content on the surface of DWLR was confirmed by water contact angle measurements. The relationship between the crosslink density and sol fraction of DWLR with different desulfurization times agreed with the Horikx equation, an indication that the microorganisms could break the crosslinked sulfur bonds on the surface of WLR, but leaving the main chains intact.

Stress exerts profound inhibitory effects on reproductive function by suppressing the pulsatile release of gonadotrophin-releasing hormone (GnRH) and therefore luteinising hormone (LH). This effect is mediated in part via the corticotrophin-releasing factor (CRF) system, although another potential mechanism is via GABAergic signalling within the medial preoptic area (mPOA) because this has known inhibitory influences on the GnRH pulse generator and shows increased activity during stress. In the present study, we investigated the role of the preoptic endogenous GABAergic system in stress-induced suppression of the GnRH pulse generator. Ovariectomised oestradiol-replaced rats were implanted with bilateral and unilateral cannulae targeting toward the mPOA and lateral cerebral ventricle, respectively; blood samples (25 μl) were taken via chronically implanted cardiac catheters every 5 min for 6 h for the measurement of LH pulses. Intra-mPOA administration of the specific GABA(A) receptor antagonist, bicuculline (0.2 pmol each side, three times at 20-min intervals) markedly attenuated the inhibitory effect of lipopolysaccharide (LPS; 25 μg/kg i.v.) but not restraint (1 h) stress on pulsatile LH secretion. By contrast, restraint but not LPS stress-induced suppression of LH pulse frequency was reversed by application of the selective GABA(B) receptor antagonist, CGP-35348, into the mPOA (1.5 nmol each side, three times at 20-min intervals). However, intra-mPOA application of either bicuculline or CGP-35348 attenuated the inhibitory effect of CRF (1 nmol i.c.v.) on the pulsatile LH secretion. These data indicate a pivotal and differential role of endogenous GABAergic signalling in the mPOA with respect to mediating psychological and immunological stress-induced suppression of the GnRH pulse generator.

“The NURBS Book” [L. Piegl, W. Tiller, The NURBS Book, second edn, Springer, 1997] is very popular in the fields of computer aided geometric design (CAGD) and geometric modeling. In Section 9.5.2 of the book, the well-known problem of the local cubic spline approximation is discussed. The key in local cubic spline approximation is cubic polynomial interpolation. In this short paper, we present the concept of single-side/double-side cubic curves and obtain the necessary and sufficient condition of a cubic curve being a single-side/double-side curve. Based on this result, for some cases of two end tangents being nearly parallel we present a new method for the problem of cubic polynomial interpolation. We also point out a flaw in Section 9.5.2 of the book and give the correction result.

Obesity is the major risk factor for early puberty, but emerging evidence indicates other factors including psychosocial stress. One key brain region notable for its role in controlling calorie intake, stress, and behavior is the amygdala. Early studies involving amygdala lesions that included the medial nucleus advanced puberty in rats. More recently it was shown that a critical site for lesion-induced hyperphagia and obesity is the posterodorsal subnucleus of the medial amygdala (MePD), which may explain the advancement of puberty. Glutamatergic activity also increases in the MePD during puberty without a corresponding γ-aminobutyric acid (GABA)ergic change, suggesting an overall activation of this brain region. In the present study, we report that neurotoxic lesioning of the MePD advances puberty and increases weight gain in female rats fed a normal diet. However, MePD lesioned rats fed a 25% nonnutritive bulk diet also showed the dramatic advancement of puberty but without the increase in body weight. In both dietary groups, MePD lesions resulted in an increase in socialization and a decrease in play fighting behavior. Chronic GABAA receptor antagonism in the MePD from postnatal day 21 for 14 days also advanced puberty, increased socialization, and decreased play fighting without altering body weight, whereas glutamate receptor antagonism delayed puberty and decreased socialization without affecting play fighting. In conclusion, our results suggest the MePD regulates the timing of puberty via a novel mechanism independent of change in body weight and caloric intake. MePD glutamatergic systems advance the timing of puberty whereas local GABAergic activation results in a delay.

Prolonged exposure to environmental stress activates the hypothalamic-pituitary-adrenal (HPA) axis and generally disrupts the hypothalamic-pituitary-gonadal axis. Because CRF expression in the central nucleus of the amygdala (CeA) is a key modulator in adaptation to chronic stress, and central administration of CRF inhibits the hypothalamic GnRH pulse generator, we tested the hypothesis that overexpression of CRF in the CeA of female rats alters anxiety behavior, dysregulates the HPA axis response to stress, changes pubertal timing, and disrupts reproduction. We used a lentiviral vector to increase CRF expression site specifically in the CeA of preweaning (postnatal day 12) female rats. Overexpression of CRF in the CeA increased anxiety-like behavior in peripubertal rats shown by a reduction in time spent in the open arms of the elevated plus maze and a decrease in social interaction. Paradoxically, puberty onset was advanced but followed by irregular estrous cyclicity and an absence of spontaneous preovulatory LH surges associated with proestrous vaginal cytology in rats overexpressing CRF. Despite the absence of change in basal corticosterone secretion or induced by stress (lipopolysaccharide or restraint), overexpression of CRF in the CeA significantly decreased lipopolysaccharide, but not restraint, stress-induced suppression of pulsatile LH secretion in postpubertal ovariectomized rats, indicating a differential stress responsivity of the GnRH pulse generator to immunological stress and a potential adaptation of the HPA axis to chronic activation of amygdaloid CRF. These data suggest that the expression profile of this key limbic brain CRF system might contribute to the complex neural mechanisms underlying the increasing incidence of early onset of puberty on the one hand and infertility on the other attributed to chronic stress in modern human society.
