Mass spectrometry (MS) has been developed for high-sensitivity structural determination of carbohydrates. Due to the similarity of oligosaccharide structures, there is still insufficient information about the correlation between carbohydrate chemical structure and mass spectra. A series of trisaccharides containing different glycosidic linkages (1→2, 1→3, 1→4, 1→6, 2→1 and 2→6), anomeric configuration (α and β), and monosaccharide (pentose, hexose and hexuronic acid), as well as two cyclodextrins (α- and β-cyclodextrin) were systematically investigated using matrix assisted laser desorption/ionization time-of-flight/time-of-flight mass spectrometry (MALDI TOF/TOF MS) in positive and negative ion modes using super-DHB (2,5-dihydroxybenzoic acid) and harmine hydrochloride as matrices, respectively. Cross-ring cleavage generated by A-types ions with neutral losses of 48, 60, 78, 90, or 120 Da were observed in the negative ion MS/MS spectra, and neutral losses of 60, 90, or 120 Da were observed in the positive ion MS/MS spectra. Glycosidic cleavages were observed in both modes. The linkage isomers were unambiguously identified by those diagnostic fragment ions with their relative intensities. Furthermore, the correlation between linkage pattern and characteristic ions successfully allowed the characterization of polysaccharides with different structural features.

Polysaccharides are important constituents in Dolichos lablab hull. Herein, pectin-glucuronoxylan complex from D. lablab hull designated as DLHP-3 (D. lablab hull polysaccharide,) was prepared by ion exchange and gel permeation chromatography, and further characterized by acid degradation and enzymatic hydrolysis, methylation combined with GC-MS, NMR and MALDI-TOF-MS analysis. Both of pectin and glucuronoxylan regions were found in DLHP-3. The glucuronoxylan region consisted of a →4)-β-Xylp-(1→ backbone with branches of α-GlcpA-(1→ substituted at O-2 site, and the ratio of xylose to glucuronic acid was about 5:1. Acetyl groups were mainly attached to O-3 site of →2,4)-β-Xylp-(1→ residues. The main chain of pectin region could be represented by →4)-α-GalpA-(1→4)-α-GalpA-(1→ and →2)-α-Rhap-(1→4)-α-GalpA-(1→ with partial methyl-esterification. The side chains were deduced to embrace arabinan and arabinogalactan linked to rhamnogalacturonan-I region. Pectin was probably covalently bound to glucuronoxylan. Our findings uncovered the molecular structure of pectin-glucuronoxylan complex from D. lablab hull.

The interleukin-6 (IL-6)/signal transducer and activator of transcription (STAT)-3 signaling pathway regulates proliferation and survival of intestinal epithelial cells and has profound impact on the tumorigenesis of colitis-associated cancer (CAC). Tea polysaccharides (TPS) are the major nutraceutical component isolated from tea-leaves and are known to possess antioxidant, anti-inflammatory, and antitumor bioactivities. Here, we investigated the antitumor activities of TPS on CAC using the azoxymethane/dextran sulfate sodium (AOM/DSS) mouse model and IL-6-induced colorectal cancer cell line (CT26) and determined whether TPS exerted its antitumor effects through the IL-6/STAT3 pathway. Results demonstrated that TPS significantly decreased the tumor incidence, tumor size, and markedly inhibited the infiltration of pro-inflammatory cells and the secretion of pro-inflammatory cytokines via balancing cellular microenvironment. Furthermore, we found that TPS suppressed the activation of STAT3 and transcriptionally regulated the expressions of downstream genes including MMP2, cyclin Dl, survivin, and VEGF both in vivo and in vitro. Thus, it was concluded that TPS attenuated the progress of CAC via suppressing IL-6/STAT3 pathway and downstream genes' expressions, which indicated that TPS may be a hopeful antitumor agent for the prevention and treatment of colon cancer.

This study aims at investigating physical–chemical properties of a water-soluble heteropolysaccharide from Plnatago asiatica L. leaves, and its immunomodulatory effects on macrophages RAW264.7 cells. Hot water-extracted crude polysaccharide from the plant leaves (PLLCP) was fractionated into four fractions by DEAE Sepharose Fast Flow eluted with 0.1–0.5 M NaCl solutions. The most abundant fraction (0.3 M NaCl elution), designated PLLP-2, was identified as a heteropolysaccharide with an average molecular weight (Mw) of 3.54 × 104 and composed mainly of Gal (34.4%), GalA (36.5%), Ara (10.1%) and Rha (8.4%). PLLP-2 was an acidic polysaccharide exhibiting inflaky curly aggregation with smooth surface. PLLCP and its main subfraction PLLP-2 displayed immunomodulatory activities by stimulating the production of pro-inflammatory cytokines, including TNF-α and IL-1β in macrophage RAW264.7 cells. These results indicate that the main polysaccharide fraction purified from P. asiatica L. leaves is probably pectin, and have potential immunomodulatory function.

There is increasing concern about possible adverse effects of soy based infant formulas (SBIF) due to their high amount of isoflavones (ISO). The aim of the present study was to investigate effects of neonatal exposure to ISO on reproductive system of female Wistar rats. Animals were exposed to an ISO depleted diet or a diet enriched with an ISO extract (IRD; 508mg ISO/kg) during embryogenesis and adolescence. Pups of each group were fed daily by pipette with ISO-suspension (ISO+; 32mg ISO/kg bw) or placebo from postnatal day (PND) 1 until PND23 resulting in plasma concentrations similar to levels reported in infants fed SBIF. The visceral fat mass was reduced by long-term IRD. Vaginal epithelial height was increased at PND23 and vaginal opening was precocious in ISO+ groups. Later in life, more often irregular estrus cycles were observed in rats of ISO+ groups. In addition, FSH levels and uterine epithelial heights were increased at PND80 in ISO+ groups. In summary, the results indicate that neonatal ISO intake, resulting in plasma concentrations achievable through SBIF, has an estrogenic effect on prepubertal rats and influences female reproductive tract later in life.

Konjac glucomannan (KGM) with a molecular weight (MW) of 823.4 kDa was partially degraded by endo-1,4-β-mannanase. Two hydrolyzed KGM fractions (KGM-M-1: 147.2 kDa and KGM-M-2: 21.5 kDa) were characterized and applied to the animal tests in comparison with the native KGM. After oral feeding to the mice, KGM-M-1 and KGM-M-2 significantly increased the levels of short chain fatty acids (SCFAs) in the colonic contents and the native KGM increased the SCFAs in the cecum. The more significant effect of the native KGM in the cecum may be attributable to its high viscosity, slowing down the movement of intestinal microflora through the cecum, while the lower MW KGM-M-1 and KGM-M-2 could move more easily through the colon to be fermented by colonic bacteria. This new finding may be useful for future research and development of low-MW KGM polysaccharides through enzyme hydrolysis for the desired gut health benefits.

Graphical abstract, Caenorhabditis elegans is a well-established laboratory animal model and has been widely used in biological research. However, it is still a challenge to obtain a good amount of quality RNA from a limited number of C. elegans for gene expression studies. To address this issue, the present study has compared different conditions to preserve C. elegans for RNA extraction after the failure of an initial effort to use RNAlater-preserved worms for RNA extraction. The effects of different concentrations of proteinase K, different worm life stages, and different worm numbers on RNA extraction were also investigated. The best results were achieved under the following conditions: 1) adult worms that were either freshly prepared or quickly frozen in liquid nitrogen followed by storage at −80 °C; 2) disruption of C. elegans with proteinase K (1 mg/mL) in a lysis buffer (65 °C for 10 min) prior to extraction with Trizol agent. This method can provide a stable, rapid, and effective means to extract RNA from C. elegans with variable worm numbers from 20 to 200. • RNAlater was inappropriate for preserving C. elegans for effective RNA extraction. • Proteinase K was verified for lysing a limited number of C. elegans for RNA extraction.

Isoflavones have been reported to stimulate muscle growth. The aim of this in vitro study was to examine anabolic activity and associated molecular mechanisms of a soy extract (SoyEx), isoflavone aglycones, and a mixture simulating the composition of SoyEx in C2C12 myotubes. C2C12 cells were differentiated into myotubes. The effects of SoyEx, genistein, daidzein, glycitein, and the mixture of genistein-daidzein-glycitein (Mix) on myotube diameter and number were determined. In addition, the expression of genes and proteins associated with anabolic activity was analyzed. Treatment with SoyEx, genistein, and Mix led to a significant increase of myotube diameter and an increase of the number of myotubes per area compared to the control cell. The increase of diameter by SoyEx was antagonized by an antiestrogen, not by an antiandrogen. Furthermore, gene expressions of insulin growth factor (IGF)-1 and its receptor (IGF-1R), as well as protein expression of myosin heavy chain (MHC), were significantly increased by SoyEx, genistein, and Mix. The effects induced by genistein and Mix were comparable to SoyEx. In conclusion, SoyEx displays an anabolic activity in C2C12 myotubes by binding to ER and modulating IGF-1 and MHC expression. Our studies with isoflavone aglycones and Mix indicate that the isoflavone aglycone with the highest anabolic bioactivity in SoyEx is genistein.

Many phytochemicals show promise in cancer prevention and treatment, but their low aqueous solubility, poor stability, unfavorable bioavailability, and low target specificity make administering them at therapeutic doses unrealistic. This is particularly true for (–)-epigallocatechin gallate, curcumin, quercetin, resveratrol, and genistein. There is an increasing interest in developing novel delivery strategies for these natural products. Liposomes, micelles, nanoemulsions, solid lipid nanoparticles, nanostructured lipid carriers and poly (lactide-co-glycolide) nanoparticles are biocompatible and biodegradable nanoparticles. Those nanoparticles can increase the stability and solubility of phytochemicals, exhibit a sustained release property, enhance their absorption and bioavailability, protect them from premature enzymatic degradation or metabolism, prolong their circulation time, improve their target specificity to cancer cells or tumors via passive or targeted delivery, lower toxicity or side-effects to normal cells or tissues through preventing them from prematurely interacting with the biological environment, and enhance anti-cancer activities. Nanotechnology opens a door for developing phytochemical-loaded nanoparticles for prevention and treatment of cancer.

The purpose of this study was to investigate the combinatory effects of an isoflavone (ISO)-rich diet and exercise on fat mass and lipid metabolism in ovariectomized (OVX) rats. Therefore the female Wistar rats were sedentary, performed an intense treadmill uphill running, received ISOs, or a combination of ISOs and running after ovariectomy. The exercise reduced visceral fat mass, adipocyte size and serum leptin in Sham animals and antagonized the increases of these parameters induced by OVX. ISOs reduced OVX induced increase of serum leptin. The combination of training and ISOs was most effective in reducing serum triglyceride levels. In OVX rats the training stimulated the expression of genes associated with fatty acid synthesis (SREBP-1c and FAS) in adipose tissue, soleus muscle, liver and genes associated with fatty acid oxidation (PPARδ and PGC-1α) in adipose tissue. ISOs stimulated the expression of SREBP-1c and FAS in soleus muscle and PGC-1α in adipose tissue, whereas suppressed hepatic SREBP-1c and FAS expression. Strong additive effects of ISOs combined with the training were observed for PPARδ and PGC-1α expressions in soleus muscle. In conclusion our results demonstrate that both the training and ISOs affect fat mass and fatty acid metabolism in OVX rats. The training seems to have a higher impact than ISO exposure in regulating gene expression in adipose tissue. However, the strongest effects for several of the addressed parameters could be observed in the combination group especially in the soleus muscle. Therefore a combination of training and an ISO-rich diet may have beneficial effects on fatty acid metabolism and could be a concept for the prevention of obesity in postmenopausal females.

Our previous study has demonstrated that Ganoderma atrum polysaccharide (PSG-1) has immunomodulatory activity on spleen lymphocytes. However, how PSG-1 exerts its effect on purified lymphocytes is still obscure. Thus, this study aimed to investigate the immunomodulatory activity of PSG-1 on purified T lymphocytes and further elucidate the underlying mechanism based on RNA sequencing (RNA-seq). Our results showed that PSG-1 promoted T lymphocytes proliferation and increased the production of IL-2, IFN-γ, and IL-12. Meanwhile, RNA-seq analysis found 394 differentially expressed genes. The potential influence of dietary phytoestrogen exposure on human health during different life phases including early childhood is a matter of scientific debate. In order to improve the risk-benefit assessment of exposure to dietary phytoestrogen, reliable and age-stratified exposure data are desirable. For contributing to the database on phytoestrogen exposure, in the present study plant-derived foods from the Chinese market were analysed by LC-MS/MS for their contents of phytoestrogens, including daidzein, genistein, secoisolariciresinol, glycitein and coumestrol. The analytical data showed the presence of phytoestrogens in a concentration range of less than 0.1 to about 50 μg g(-1). Dietary intake was assessed on the basis of average food intake data obtained from interviewing 1000 randomly selected people with the help of food frequency questionnaires. Based on the overall population sampled, the average total phytoestrogen intake was estimated at 232 μg kg(-1) day(-1). Genistein contributed to about 66%, secoisolariciresinol and glycitein to about 10% each, and daidzein to about 7% of the overall intake. Coumestrol was present only in trace amounts. Age-related exposure assessment indicated that pre-pubertal children (aged 0-14 years) were exposed at the highest level with an average total phytoestrogen intake of 621 μg kg(-1) day(-1). The substantially higher average exposure of children as compared with adults should trigger further research into the potential health effects of early life exposure to phytoestrogen.

Cyclocarya paliurus (Batal.) Iljinskaja, a well-known edible and medicinal plant, has been widely used in China as a traditional medicine for treating hypertension and diabetes. C. paliurus possesses various bioactivities, such as antihyperglycemic, antihyperlipidemic, antihypertensive, anticancer, antifatigue, antioxidation, antimicrobial, colon health-promoting, and immunological activities. Polysaccharides, as natural macromolecules with various biological activities, are considered to be the main effective components in C. paliurus. Here, we summarize studies of polysaccharides from C. paliurus over the past 20 years, including extraction and purification processes, structure, and bioactivities.

MRS culture medium was used as the basic medium for optimizing the fermented condition of Lactobacillus. rhamnosus. The four significant factors (beef extract, tween-80, peptone, diammonium citrate) were screened from these components by using a Plackeet-Burman design. Steepest ascent path was adopted to approach the optimal region, and then the Central Composite Design was used to determine the optimal levels of the main factors. Considering the fit model, cost and operation, the optimal conditions were determined as follows: beef extract 2.6%, tween-80 0.3%, peptone 2%, diammonium citrate 0.4%, with other components remaining the same, 3% inoculation, 18h culture at 37¡æ.Under such conditions, the experimental value of cell concentration were 3.54×109cfu/ml and 2.19-fold live bacteria counts in MRS-based culture medium, showing no difference with predicted value 3.68×109cfu/ml.

From Chloranthus multistachys, three sesquiterpene lactones, multislactone (1), codonolactone(2), lasianthuslactone A(3), and three diterpenes, 12R,15- dihydroxylabda-8(17), 13E- dien-19-oic acid(4), 12(S),15- dihydroxy labda-8(17), 13E-dien- 19-oic acid(5), 12-oxo-15- hydroxylabda-8(17), 13E-dien-19-oic acid(6) were isolated. The structures of the isolated compounds were established by means of NMR and MS analyses. Compound 1~6 were firstly isolated from this plant. Compound 1 is new natural products. Among them , compound 1 and 6 show anti- inflammatory activities.

Acetonitrile-water extracts of several Ganoderma species - a mushroom being used in Traditional Chinese Medicine - were analysed by liquid chromatography-UV detection in hydrophilic interaction chromatography (HILIC) and reversed-phase (RP) elution modes. A set of six polar stationary phases was used for HILIC runs. These columns had remarkably different separation properties under binary gradient conditions as evinced by hierarchical cluster analysis on retention patterns of seven test compounds. Complementary measurements of RP chromatograms were carried out on a C(18) packing. Injection precision (n=5) and intra-day precision (n=5) were each2.0% RSD (HILIC) and0.7% RSD (RP) for relative retention times of main characteristic peaks of a sample extract while for relative peak areas RSD values were max. 6.8%. Repetitive analysis (n=7) of a processed sample stored in the autosampler tray for 48h was used to confirm within-sequence sample stability. Eleven Ganoderma lucidum samples served as training set for the construction of column-specific simulated mean chromatograms. Validation with twelve samples comprising G. lucidum, Ganoderma sinense, Ganoderma atrum, and Ganoderma tsugae by correlation coefficient based similarity evaluation of peak patterns showed that a discrimination of G. lucidum from other Ganoderma species by means of chromatographic fingerprints is conceptually possible on all columns, except of a bare silica packing. The importance of the combined use of RP and HILIC fingerprints to improve the rate of correct sample classification was demonstrated by the fact that each one G. sinense specimen was wrongly assigned being G. lucidum by all HILIC fingerprints but not the RP fingerprint and vice versa. The present data revealed that (i) the analysis of complex biological materials by quasi orthogonal chromatographic modes such as HILIC and RP may deliver more discriminative information than single-mode approaches which strengthens the reliability of fingerprint-based sample classification and (ii) different retention and selectivity characteristics of polar bonded silica packings in the HILIC elution mode may only have a minor impact on chemometric sample discrimination capabilities in such kind of pattern-oriented metabolomics separation problems.

Ethnopharmacological relevance The seeds of Plantago asiatica L. were often used as a traditional Chinese medicine for some immunologically weak patients suffering from chronic illness. These uses could be related to immunomodulatory properties of the plant. Aim of the study In this study, effects of extract of the seeds of Plantago asiatica L. (ES-PL) were investigated on the maturation of dendritic cells (DCs), which play significant role in primary immune system. Materials and methods The phenotypes of DCs were analyzed by using flow cytometry while phagocytosis was assessed by the uptake of FITC-dextran. Antigen presenting ability to allogeneically naive or syngeneically primed T lymphocytes was examined by the lymphocyte proliferation of mixed lymphocyte reaction (MLR). In addition, the level of chemokine receptor CCR7 mRNA was determined by RT-PCR. Results DCs treated with ES-PL expressed higher levels of MHC class II molecules and major costimulatory molecules such as CD80 and CD86. Functional maturation of DCs treated with ES-PL was confirmed by decreased mannose receptor-mediated endocytosis and increased antigen presenting abilities to allogeneically naive or syngeneically primed T lymphocytes. The CCR7 mRNA expression in DCs treated with ES-PL was also enhanced. Conclusions These results indicated that ES-PL could induce the maturation of murine DCs.

In recent decades, the polysaccharides from the medicinal plants have attracted a lot of attention due to their significant bioactivities, such as anti-tumor activity, antioxidant activity, anticoagulant activity, antidiabetic activity, radioprotection effect, anti-viral activity, hypolipidemic and immunomodulatory activities, which make them suitable for medicinal applications. Previous studies have also shown that medicinal plant polysaccharides are non-toxic and show no side effects. Based on these encouraging observations, most researches have been focusing on the isolation and identification of polysaccharides, as well as their bioactivities. A large number of bioactive polysaccharides with different structural features and biological effects from medicinal plants have been purified and characterized. This review provides a comprehensive summary of the most recent developments in physiochemical, structural features and biological activities of bioactive polysaccharides from a number of important medicinal plants, such as polysaccharides from Astragalus membranaceus, Dendrobium plants, Bupleurum, Cactus fruits, Acanthopanax senticosus, Angelica sinensis (Oliv.) Diels, Aloe barbadensis Miller, and Dimocarpus longan Lour. Moreover, the paper has also been focused on the applications of bioactive polysaccharides for medicinal applications. Recent studies have provided evidence that polysaccharides from medicinal plants can play a vital role in bioactivities. The contents and data will serve as a useful reference material for further investigation, production, and application of these polysaccharides in functional foods and therapeutic agents.

cope : This study was to investigate anabolic adaptation of skeletal muscle in response to an isoflavone (ISO) enriched diet, training and their combinations in ovariectomized (OVX) rats. Methods and results : Female Wistar rats were sedentary, performed treadmill uphill running, received ISOs, or a combination of ISOs and running after ovariectomy. Body weight was increased by OVX. Both ISO and training treatment antagonized this increase. The weights of soleus and gastrocnemius muscles were increased only when training and ISOs were combined. In soleus muscle insulin-like growth factor (IGF)-1R, MyoD and Myogenin expressions were only up-regulated by training in Sham groups. However, a stimulation of IGF-1R and MyoD expression could be observed when ISOs and training were combined. In gastrocnemius muscle MyoD and Myogenin expressions were stimulated by either training or ISOs. Additive effects were detected when combining the two interventions. Conclusion : Our results indicate that the combination of ISOs and exercise is more efficient in increasing relative skeletal muscle mass and the expression of molecular markers related to anabolic adaptation in the skeletal muscle of female rats. Ganoderma (G.) is a famous medicinal fungus found in the regions of China, Japan, Korea, and Eastern Russia, and the polysaccharide from G. atrum (PSG) – one of the most important species of Ganodermataceae – has been shown to possess excellent bioactivity. Ultrasonic treatment has been widely used to improve the yield recovery, physicochemical properties and bioactivity of polysaccharides. In order to improve the yield recovery and modify the molecular structure of PSG, the authors employed the ultrasonic technique and assessed its effects on structure, physicochemical properties and antioxidant activity of PSG.

In sugar molecule of the title compound, C22H29Cl3O13·C7H8, the pyran­ose ring adopts the expected chair conformation. The acetoxy­methyl and two acet­oxy substituents are in the equatorial positions of this ring. In the crystal structure, there is a single weak C—H⋯O inter­molecular inter­action (H⋯O = 2.46 A), which links mol­ecules into chains in the a-axis dirextion.

Scope The phytoectysteroid ecdysterone (Ecdy) was reported to stimulate protein synthesis and enhance physical performance. The aim of this study was to investigate underlying molecular mechanisms particularly the role of ER beta (ERβ). Results In male rats, Ecdy treatment increased muscle fiber size, serum IGF-1 increased, and corticosteron and 17β-estradiol (E2) decreased. In differentiated C2C12 myoblastoma cells, treatment with Ecdy, dihydrotestosterone, IGF-1 but also E2 results in hypertrophy. Hypertrophy induced by E2 and Ecdy could be antagonized with an antiestrogen but not by an antiandrogen. In HEK293 cells transfected with ER alpha (ERα) or ERβ, Ecdy treatment transactivated a reporter gene. To elucidate the role of ERβ in Ecdy-mediated muscle hypertrophy, C2C12 myotubes were treated with ERα (ALPHA) and ERβ (BETA) selective ligands. Ecdy and BETA treatment but not ALPHA induced hypertrophy. The effect of Ecdy, E2, and BETA could be antagonized by an ERβ-selective antagonist (ANTIBETA). In summary, our results indicate that ERβ is involved in the mediation of the anabolic activity of the Ecdy. Conclusion These findings provide new therapeutic perspectives for the treatment of muscle injuries, sarcopenia, and cachectic disease, but also imply that such a substance could be abused for doping purposes.

The function of genistein (GEN) on tumor prevention and tumor promotion is discussed controversially. A possible interference of GEN with chemotherapy has been only rarely addressed so far. In this study, effects of GEN on the anti-tumor activity of cisplatin (CIS) were investigated in the presence and absence of estradiol (10(-10) M) in MCF-7 breast and HT-29 colon cancer cells. Cells were treated with graded concentrations of GEN (10(-4)-10(-6) M), E2, CIS and combinations. Cell growth, proliferation and apoptosis were determined as well as the expression level of PCNA, Ki67 and BCL-2 family members. CIS and GEN 10(-4) M inhibited cell growth and induced apoptosis in MCF-7 and HT-29 cells in the presence and absence of E2. Co-treatment with CIS and 10(-4)M GEN resulted in additive effects. In concentrations of 10(-5) and 10(-6) M, GEN stimulated cell growth in MCF-7 cells. It promoted proliferation, inhibited apoptosis and counteracted the anti-tumor activity of CIS in MCF-7 and HT-29 cells. Particularly the ability of CIS to induce apoptosis was antagonized. In ER alpha-positive MCF-7 cells, but not in ER alpha-negative HT-29 cells, E2 was able to neutralize the anti-CIS effects of GEN. Our data provide evidence that GEN in the absence of E2, a situation which occurs in postmenopausal women, directly affects the anti-tumor activity of cytostatic drugs like CIS. The exact molecular mechanism has to be investigated in future studies.

A rapid ultrasound-assisted extraction (UAE) procedure was developed for the determination of trace mercury (Hg) in tea leaves combined with atomic fluorescence spectrometry. Three variables including sonication time (St), ultrasonic bath temperature (T) and HNO3 : H2O2 (1 : 1, phi) volume (A2) showed the significant effect on extraction rate of total Hg evaluated by a Plackett-Burman design, and they were further optimized by a central composite design and response surface methodology. The results showed that the optimum extraction conditions were as follows: presonication time 6 min, St 8. 1 min, T 70. 5 degreesC, A2 4. 4 mL, and sample mass 300 mg. GBW10016 (tea leaves) was used as certified reference material; for comparative purposes, a microwave-assisted digestion (MAD) was used. The result obtained by optimized UAE method showed good agreement with the certified values. Under optimal conditions, recovery was evaluated to be 94.2% - 102.0% and the limit of detection 0.007 8 microg x L(-1). The relative standard deviation (RSD) of replicate measurements was generally less than 10%. The proposed UAE method was successfully applied to the determination of Hg in 63 samples of fresh tea leaves and 10 different branded tea samples. No significant differences were established between the analytical results of UAE method and MAD method. The Hg concentrations of them were found in the range of 4.6-17.3 microg x kg(-1) on a dried basis, which were within the permissible limit of the NY659-2003.

This study is to explore the effects of quercetin (QUE) on the 3 week-old mice ovarian development and relative hormone levels. The 3 week-old mice were exposed to QUE (45, 25, and 5 mg x kg(-1) x hd(-1)) by gavage for 50 days. The estrous cycle during 50 days and the changes of hormone level such as FSH, LH, etc were monitored. Moreover, the ovaries were removed after sacrifice. The organ index was measured, and the ratios of different stages of follicles were analyzed by HE staining. Furthermore, the proportion of PCNA positive cells during all stages was detected by immunohistochemistry. The results showed that QUE could increase body weight of mice and reduce the anogenital distance (AGD) to some extent, and was able to disrupt mice's estrous cycle, but it could not extend or reduce the cycle regularity. It increased ovarian organ index with a dose-dependent manner. The proportion of the primordial follicle and secondary follicles rose obviously, and that of mature follicles', atretic follicles' and corpus luteums' reduced, while primordial follicle had no change. Immunohistochemistry analysis showed that QUE could effectively increase the percentage of proliferating cells in all kinds of follicles. Serum hormone assay showed that there were significant changes of FSH and LH levels. In summary, QUE showed an estrogen-like effect on mice's ovarian development. The weight of ovary, the proportion of all kinds of follicles, the development of ovarian cells and the level of plasma hormone in mice were altered obviously by oral administration of QUE.

Herba Moslae is a well-known edible and medicinal plant in China, and the essential oil, which is assumed to contain the active components, was isolated by steam distillation method. The aim of this study was to investigate the antimicrobial activity and the antioxidant activity of the essential oil from Herba Moslae.The essential oil from Herba Moslae exhibited antimicrobial activity against all of the tested bacteria and fungi. The minimum inhibitory concentration (MIC) values for bacteria ranged from 118 to 472 microg mL(-1), and the minimum bactericidal concentration (MBC) values were from 236 to 944 microg mL(-1). The oil showed pronounced antibacterial and antifungal activity against Staphylococcus aureus and Aspergillus oryzae, but weak inhibition to Escherichia coli. The antioxidant activity of the essential oil from Herba Moslae was evaluated by using DPPH radical scavenging assay, beta-carotene/linoleic acid bleaching assay and metal chelating activity assay. In addition, the amount of total phenolic content in the oil (386 +/- 1 microg kg(-1)) was determined.This study suggests that the essential oil from Herba Moslae could be potentially used as a new potential source of natural antimicrobial, antioxidants and antiputrefactive in food industry.

Cyclocarya paliurus (Batal.) Iljinskaja is a well-known edible and medicinal plant in China and is commonly called the 'sweet tea tree'. It has been reported to have a broad range of therapeutic and nutritional values in lowering blood sugar, blood lipids and blood pressure. However, much of the annual C. paliurus production is wasted. Recent reports have suggested that polysaccharides are the main active ingredients responsible for the observed bioactivities of C. paliurus. In this study, response surface methodology was used to determine the optimal conditions for microwave-assisted extraction (MAE) of polysaccharides from C. paliurus.The optimal conditions for MAE of polysaccharides were a duration of microwave radiation of 20.8 min, a temperature of 100.9 degrees C and a water/material ratio of 22.1:1. Under these conditions the yield of polysaccharides was up to 5.04 +/- 0.09%. Compared with conventional heat reflux extraction and ultrasound-assisted extraction, MAE of polysaccharides from C. paliurus (Batal.) Iljinskaja saved a lot of time and gave the highest yield.The present study suggests that MAE is a good alternative method for the extraction of polysaccharides from C. paliurus (Batal.) Iljinskaja.

The present research investigated the effect of dynamic high pressure microfluidization (DHPM) in low pressure ranges (60-100 MPa) on the molecular structure of papain with the help of fluorescence spectra as the detection method. The result showed that after the treatment of DHPM at 60-100 MPa, the fluorescence intensity of papain, tryptophan (Trp) and tyrosine (Tyr) residues all decreased to different extents. Meanwhile, with the increase in the treatment pressure, their fluorescence intensity would gradually increase and the fluorescence emission peak would gradually red shift from 334 and 277. 5 nm before treatment to 335 and 278 nm after 100 MPa treatment for papain and Trp residue respectively; after the treatment, with the samples being placed at 0-4 degrees C for 24 h, the fluorescence spectra of papain, Tyr and Trp residues in various experiment groups basically maintained the same changing tendency compared to that of newly treated samples. Hence, it showed that after the treatment of DHPM in low pressure ranges, the Trp residue of papain was gradually brought to light and formed new and comparatively stable molecular conformation.

A water-soluble polysaccharide from the seeds of Plantago asiatica L. (P. asiatica L. polysaccharide, PLP) was extracted with hot water and purified by gel filtration chromatography. The chemical characteristics of PLP were determined by high-performance gel permeation chromatography (HPGPC) and Fourier transform infrared (FTIR) spectroscopy. In addition, the antioxidant activities of PLP in vitro were evaluated using various test systems, including scavenging of 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radicals, scavenging of superoxide radicals generated by 1,2,3-phentriol autoxidation, scavenging of hydroxyl radicals and inhibition of lipid peroxidation.The molecular weight of PLP was determined by HPGPC to be about 1894 kDa. PLP contained 29.2 g kg(-1) protein and 145.8 g kg(-1) uronic acid. The FTIR spectrum of PLP also revealed typical characteristics of a polysaccharide containing protein and uronic acid. Moreover, the results showed that PLP possessed antioxidant activities, but lower than those of ascorbic acid.PLP is an acid protein-bound polysaccharide of high molecular weight, but its structure needs further study. The present results suggest that PLP could potentially be used as a natural antioxidant.

In the present study, the contents of eighteen inorganic elements such as Ca, K, Mg, Cu, Cr, Zn and Sr in oleum camelliae during different stages of refining process were determined using ICP-AES technique. The recoveries obtained by the standard addition method ranged between 82.7% and 112.5%, and RSDs were lower than 6.66%. The detection limits of the method for the eighteen elements were in the range of 0.4-10 microg x L(-1). The experimental results proved that oleum camelliae is rich in inorganic elements. The results also showed that depending on the intensity of refining, the contents of elements will decrease accordingly. After the degumming process, the contents of elements P, Ca and Mg in oleum camelliae declined remarkably. Besides, the contents of some trace elements such as Fe, Cu, Mn and Ni that cause oxidation to occur decreased significantly after refining. In addition, Pb was not detected after the degumming and deacidification process while Co and Cd were not detected both in the crude oil and the refined oil. In conclusion, through refining many excessive metal elements in oleum camelliae were removed effectively and the levels of these elements were able to meet the national standards set by the government. Meanwhile, the experimental results indicated that the ICP-AES technique, which can be used to determine the inorganic elements in oleum camelliae, is easy to operate, rapid, accurate, and highly sensitive, and can also determine many elements simultaneously.
