Objective. The aim of this research was to create a new genetic signature of immune checkpoint-associated genes as a prognostic method for pediatric acute myeloid leukemia (AML). Methods. Transcriptome profiles and clinical follow-up details were obtained in Therapeutically Applicable Research to Generate Effective Treatments (TARGET), a database of pediatric tumors. Secondary data was collected from the Gene Expression Omnibus (GEO) to test the observations. In univariate Cox regression and multivariate Cox regression studies, the expression of immune checkpoint-related genes was studied. A three-mRNA signature was developed for predicting pediatric AML patient survival. Furthermore, the GEO cohort was used to confirm the reliability. A bioinformatics method was utilized to identify the diagnostic and prognostic value. Results. A three-gene (STAT1, BATF, EML4) signature was developed to identify patients into two danger categories depending on their OS. A multivariate regression study showed that the immune checkpoint-related signature (STAT1, BATF, EML4) was an independent indicator of pediatric AML. By immune cell subtypes analyses, the signature was correlated with multiple subtypes of immune cells. Conclusion. In summary, our three-gene signature can be a useful tool to predict the OS in AML patients.

In this study, the molecular mechanism of astragaloside Ⅳ(AS-Ⅳ) in the treatment of Parkinson's disease(PD) was explored based on network pharmacology, and the potential value of AS-Ⅳ in alleviating neuronal injury in PD by activating the PI3 K/AKT signaling pathway was verified through molecular docking and in vitro experiments. Such databases as SwissTargetPrediction, BTMAN-TAM, and GeneCards were used to predict the targets of AS-Ⅳ for the treatment of PD. The Search Tool for the Retrieval of Interacting Genes/Proteins(STRING) was employed to analyze protein-protein interaction(PPI) and construct a PPI network, and the Database for Annotation, Visualization and Integrated Discovery(DAVID) was used for Gene Ontology(GO) term enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway enrichment analysis. Based on the results of GO enrichment analysis and KEGG pathway analysis, the PI3 K/AKT signaling pathway was selected for further molecular docking and in vitro experiments in this study. The in vitro cell model of PD was established by MPP~+. The cell viability was measured by MTT assay and effect of AS-Ⅳ on the expression of the PI3 K/AKT signaling pathway-related genes and proteins by real-time polymerase chain reaction(RT-PCR) and Western blot. Network pharmacology revealed totally 122 targets of AS-Ⅳ for the treatment of PD, and GO enrichment analysis yielded 504 GO terms, most of which were biological processes and molecular functions. Totally 20 related signaling pathways were screened out by KEGG pathway analysis, including neuroactive ligand-receptor interaction, PI3 K/AKT signaling pathway, GABAergic synapse, and calcium signaling pathway. Molecular docking demonstrated high affinity of AS-Ⅳ to serine/threonine-protein kinases(AKT1, AKT2), phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit gamma(PIK3 CG), and phosphoinositide-3-kinase, catalytic, alpha polypeptide(PIK3 CA) on the PI3 K/AKT signaling pathway. In vitro experiments showed that AS-Ⅳ could effectively inhibit the decrease of the viability of PC12 induced by MPP~+ and up-regulate the mRNA expression levels of AKT1 and PI3 K as well as the phosphorylation levels of AKT and PI3 K. As an active component of Astragali Radix, AS-Ⅳ acts on PD through multiple targets and pathways. Furthermore, it inhibits neuronal apoptosis and protects neurons by activating the PI3 K/AKT signaling pathway, thereby providing reliable theoretical and experimental supports for the treatment of PD with AS-Ⅳ.

Myocardial infarction (MI) is the most prevalent cardiac disease with high mortality, leading to severe heart injury. Circular RNAs (circRNAs) are a new type of regulatory RNAs and participate in multiple pathological cardiac progressions. However, the role of circRNAs Postn (circPostn) in MI modulation remains unclear. Here, we aimed to explore the effect of circPostn on MI-induced myocardial injury and cardiac remodeling. We identified that the expression of circPostn was elevated in the plasma of MI patients, MI mouse model, and hypoxia and reoxygenation (H/R)-treated human cardiomyocytes. The depletion of circPostn significantly attenuated MI-related myocardium injury and reduced the infarct size in MI mouse model. The circPostn knockdown obviously enhanced left ventricular ejection fraction (LVEF) and left ventricular fraction shortening (LVFS) and inhibited left ventricular anterior wall thickness at diastole (LVAWd) and left ventricular posterior wall thickness at diastole (LVPWd). The depletion of circPostn was able to decrease MI-induced expression of collagen 1α1 and collagen 3α1 in the ventricular tissues of mice. The protein expression of collagen and α-smooth muscle actin (SMA) was up-regulated in MI mice and was inhibited by circPostn knockdown. Meanwhile, the expression of atrial natriuretic peptide (ANP) and brain natriuretic peptide (BNP) was repressed by circPostn depletion in the ventricular tissues of MI mice. Besides, the circPostn depletion attenuated cardiomyocyte apoptosis in mice. Mechanically, circPostn served as a miR-96-5p sponge and miR-96-5p-targeted BNIP3 in human cardiomyocytes, in which circPostn up-regulated BNIP3 expression by targeting miR-96-5p. circPostn promoted H/R-induced cardiomyocyte injury by modulating miR-96-5p/BNIP3 axis. Thus, we conclude that circPostn contributes to MI-induced myocardial injury and cardiac remodeling by regulating miR-96-5p/BNIP3 axis. Our finding provides new insight into the mechanism by which circPostn regulates MI-related cardiac dysfunction. circPostn, miR-96-5p, and BNIP3 are potential targets for the treatment of MI-caused heart injury.

Whether Chinese herbal formulas are effective in treatment of postmenopausal osteoporosis remains unclear. The aim of this study is to explore the experimental evidence of both in vitro and in vivo preclinical studies using Chinese herbal formulas in postmenopausal osteoporosis. Searches were applied to various databases with relevant keywords. Original in vivo and in vitro studies using Chinese herbal formulas to treat postmenopausal osteoporosis, and with full text available, were included. Er-Xian Decoction, Bu-Shen-Ning-Xin Decoction, Qing E Formula, Liuwei Dihuang Wan, and Xian-Ling-Gu-Bao Decoction, the most commonly studied formulas, were selected from the pool of Chinese medicine. The preclinical data indicated the potential use of Chinese herbal formulas in postmenopausal osteoporosis. The underlying mechanisms included bone morphogenetic protein (BMP), Wnt/[Formula: see text]-catenin, extracellular-signal-regulated kinase/c-Jun [Formula: see text] terminal kinase/mitogen-activated protein kinase (ERK/JNK/MAPK), estrogen receptor (ER), and osteoprotegerin/receptor activator of nuclear factor [Formula: see text]B ligand (OPG/RANKL) signaling pathways. This study demonstrated the anti-osteoporotic effect of Chinese herbal formulas targeting different pathways in bone metabolism. Further study with adequate sample size and follow-up time, appropriate controls, and optimal blinding is required.

The precise pathophysiological mechanisms of preeclampsia (PE) and preventative strategies remain unknown. Laboratory markers which can help in identifying PE patients from pregnant women and assessing the severity of PE during pregnancy are worthy to be explored. In this study, a retrospective case-control study was designed to assess whether the serum levels of albumin (ALB), total protein (TP), prealbumin (PA), alkaline phosphatase (ALP), lactic dehydrogenase (LDH), D-dimer, fibrinogen (Fbg), platelet (PLT) count, mean platelet volume (MPV), and platelet distribution width (PDW) can help in assessing PE and evaluate its severity. 256 pregnant women were enrolled and classified into 3 groups: mild preeclampsia (mPE,n=85), severe preeclampsia (sPE,n=78), and healthy normotensive controls (control,n=93). Our result showed that the serum levels of ALP, LDH, and D-dimer were significantly higher in mild or severe PE patients compared with the healthy controls (66 (52.5-76.5) vs. 168 (141.5-201.25) vs. 182.5 (120-191.5), 152 (139.75-166.25) vs. 183.5 (163.25-307) vs. 282 (215.25-306), 1.05 (0.65-1.57) vs. 3.05 (2.25-4.08) vs. 5.65 (2.29-7.71)), while ALB, TP, and PA are lower (38 (37-42) vs. 31.5 (25.5-34.5) vs. 28.5 (24-33), 65 (63-68.25) vs. 56.5 (52-61) vs. 51.5 (49-58),219.14±68.25vs.167.88±52.21vs.143.22±50.46). On the other hand, compared with the mPE group, the sPE group showed significantly lower PLT count but higher level of LDH, D-dimer, and Fbg. No significant differences in MPV or PDW were found between any of the two groups. In conclusion, the above markers except for the MPV and PDW may be correlated with PE severity in this patient cohort, indicating possible values of these potential biomarkers in auxiliary diagnosis and severity assessment of PE.

A novel ternary nanocomposite of CdSH/ZnO-GE was prepared for photoelectrochemical (PEC) selective sensing of Cu2+. ZnO nanorods and graphene (GE) nanosheets were first decorated on Ni foam (NF) simultaneously by a facile hydrothermal method and then CdS nanoparticles were loaded on ZnO-GE arrays through simple chemical bath deposition (CBD) method and following calcination process to form hexagonal CdS (CdSH). The obtained CdSH/ZnO-GE@NF sensor was characterized by scanning electron micrographs, X-ray diffraction and X-ray photoelectron spectroscopy. The CdSH/ZnO-GE@NF electrode exhibited enhanced photoelectrical activity with a photocurrent density of 6.8 mA·cm−2 under illumination at 0.5 V vs. SCE. For the PEC detection of Cu2+, the as-prepared sensor exhibited a low detection limit of 0.03 μM and wide linear concentration rang of 0.2 μM to 1 mM with high selectivity and stability. It was successfully applied to the detection of Cu2+ in real industrial waste water and sea water, suggesting that the CdSH/ZnO-GE@NF electrode is a promising sensor to detect copper pollution in natural environments.

Objective To summarize the experience with resurgery for recurrent valvular heart diseases. Methods From June 2004 to June 2015, 28 patients (15 males and 13 females) with ages ranging from 44 to 67 years (55.6±6.5 years) with recurrent heart valve disease underwent resurgery. The reasons for resurgery included perivalvular leakage (7 cases), bioprosthetic valve decline (6 cases in mitral valve and 3 in tricuspid valve), mechanical prostheses dysfunction (2cases), infective endocarditis after valve replacement (2 cases), restenosis of repaired native valve (1 case), and severe tricuspid insufficiency after left-side valve surgery (7 cases). Resurgery included mitral valve replacement in 18 patients and tricuspid valve replacement in 10. All the patients underwent third or fourth or even fifth cardiac surgery for valve replacement. Results There were 2 hospital deaths with a mortality of 7.1% (2/28). The main causes of early-stage deaths were low cardiac output syndrome. The main postoperative complications were respiratory failure in 3, low cardiac output syndrome in 2, reexploration for bleeding in 2 and serious infectious shock in 1. All the patients were found with the great improvement in heart function and the re-implanted prostheses worked well during follow-up. Conclusions Although resurgery for recurrent heart valve disease poses a continuing challenge to cardiac surgeon, it could be performed with the satisfactory results. Premature ovarian failure (POF) is a kind of gynecological disease that causes amenorrhea, infertility, menopause and urogenital symptoms. Currently hormone replacement therapy (HRT) is the most popular choice for women with POF to get rid of menopausal syndrome. However, as the popularization of Chinese herbs made Chinese medicine (CM) shine new lights, physicians are able to treat POF with both meno-herbs and integrated therapy. HRT has its own indications and contraindications. For example, unexplained vaginal bleeding, acute liver damage, liver dysfunction, vascular embolization, and breast cancer are all contraindications of HRT, and CM is taken by more physicians as an adjuvant therapy. This review, including a range of common Chinese herbs and formulations according to the existing literature, provides a general description of CM treating POF from the aspects of mechanisms and clinical application. It also highlights acupuncture as a unique physiotherapy for POF. Although the validity of CM has been supported by the evidence of many preclinical trials, clinical trials and meta-analysis, the adverse events with CM therapy still exist and no guarantee has been made for its safety. This review concludes the updated information for CM treating POF contributing to further studies.

Human papillomavirus (HPV) infections are common and generally harmless, but persistent infections can bring health problems like cancer and genital warts. For the uninfected group, HPV vaccines provide safe and effective protection, but they're type-restricted and expensive. For those infected, so far there have been a handful of treatments for HPV-associated benign or malignant diseases, traditional Chinese medicine being one of them. This systematic review focuses on the application of traditional Chinese medicine in HPV infection and related diseases on the basis of clinical findings. Moreover it covers compositions and mechanisms based on in vitro laboratory methods and animal models. Traditional Chinese medicine improves clinical index in the treatment of cervical cancer and genital warts; the mechanisms behind the effectiveness might be the regulation of cell apoptosis, viral gene transcription and translation, cell signal transduction pathways, and immune function.

To reuse rice straw and decrease the environmental pollution and resource waste that are caused by burning rice straw in China, this agricultural resource is increasingly being used to make pots with added formaldehyde and other chemicals for cultivating vegetables. A study was conducted to investigate the effects of rice straw-based pot powder on the growth and antioxidase activity of Chinese green cabbage seedlings and the soil microbial community using a molecular analysis. The results showed that there was no negative effect from the powder on cabbage growth, given that the cabbage plants that were treated with powder (1.2–3.2 g kg−1 rice straw pot powder) exhibited 1.8 to 2.5 times higher fresh biomass than the control (0 g kg−1 powder). Minor differences were found among key reactive oxygen system enzyme activities, but the superoxide dismutase activity decreased by 33–55% (1.2–3.2 g kg−1 powder). With the aging of the population and the upgrading of the consumption structure of national health demand in China, it has become a new trend for the public to actively seek health products and services on social networks. Based on the theory of reasoned behavior and the theory of expectancy confirmation, this study aims to analyze the cognitive factors and their effects on WeChat users&rsquo, purchase intention in the process of health product consumption. Considering that safety is a key feature of health products that distinguishes them from other consumer products, the &ldquo, satisfaction&rdquo, concept in the expectancy confirmation model is replaced by &ldquo, trust&rdquo, in this study. Two hundred and two (202) valid samples were collected by a questionnaire survey to analyze their intentions to buy health products on WeChat. Theoretical models and corresponding research hypotheses were verified by structural equation modeling. The research results show that emotional price and emotional experience are positively correlated with trust and purchase intention. There is an obvious negative correlation between privacy invasion and trust. Expectation confirmation is positively associated with trust. Moreover, the intermediary test shows that trust has completely mediated between emotional price and purchase intention, and trust also has a full intermediary effect on expectation confirmation and purchase intention.

The objective of this study is to evaluate the predictive value of sperm DNA fragmentation Index (DFI) in unexplained recurrent spontaneous abortion (RSA) and to investigate its correlation with conventional sperm parameters. Besides, we aimed to reveal the necessity of establishing a DFI clinical threshold of each laboratory for the prognostic diagnosis of RSA and establish our own DFI threshold. Semen samples were collected from male partners of RSA patients (n = 139) and healthy recent fathers (control, n = 200). DFI was tested using SCSA and conventional semen analysis was performed using an automatic semen analyzer. The DFI value and distribution were compared between the two groups using corresponding statistical software. The diagnostic threshold value was established by ROC curve. The correlation between DFI and the conventional semen parameters of the 139 cases was further analyzed using Student's t test and Mann-Whitney U test. Our result showed that DFI was significantly higher in RSA patients compared with normal donor controls. We established our own DFI threshold at 13.59%. There was only a weak partial correlation between DFI values and conventional sperm analysis parameters. Our present study suggested that DFI might be used as a valuable predictor for RSA independent of conventional sperm parameters. Additionally, we recommend that each laboratory should establish its own clinical DFI threshold for more precise prediction of RSA and we recommend that sperm DNA fragmentation test should be included in complete sperm quality assessment in addition to conventional semen analysis for RSA male partners.

Autophagy is an essential metabolic pathway mediated by lysosomal degradation, which is involved in scavenging and recycling senescent or damaged organelles and biological macromolecules in eukaryotic cells. The present study explored the association between the autophagic activity and chemotherapy resistance of leukaemia cells, and the possibility of using autophagy inhibitors to combat leukemic drug resistance. It was found that the levels of basic autophagy in multidrug‑resistant leukaemia cells (K562/ADM) were significantly higher compared with sensitive cells (K562), and that Adriamycin (ADM) was capable of inducing autophagic activity in K562 and K562/ADM cells. K562 and K562/ADM cells were treated with a series of hydroxychloroquine (HCQ) concentrations to inhibit cellular autophagy and detect cell sensitivity to ADM. The results demonstrated that the sensitivity of K562 cells to ADM was mildly enhanced by HCQ, and that the sensitivity of K562/ADM cells to ADM was markedly strengthened by HCQ. In addition, more typical morphological changes associated with apoptosis emerged, and the ratio of Bax/Bcl‑2 and activity of caspase‑3 were markedly increased in K562/ADM cells treated with HCQ. Notably, the expression of mdr1 mRNA and P‑glycoprotein (P‑gp) in drug‑resistant K562/ADM cells was upregulated along with increasing autophagic activity induced by ADM. Furthermore, HCQ significantly reduced the increase in P‑gp expression by inhibiting autophagic activity. Collectively, these findings indicated that the inhibition of autophagy significantly promoted the sensitivity of K562/ADM cells to ADM by facilitating apoptosis. Furthermore, inhibition of autophagy attenuated the expression of P‑gp; therefore, P‑gp may be involved in autophagic regulation in drug‑resistant cells.

Objective To observe surgical outcome of reoperation for paravalvular leakage and medium-and long-term survival. Methods The clinical data of 28 patients receiving reoperation for paravalvular leak (PVL) in General Hospital of PLA between October 1998 and October 2015. The patients included 22 males and 6 females, with a mean age of 55.3 years (ranging from 15-67 years). PVL was reported in 18 cases after mitral valve replacement, in 4 after aortic valve replacement, in 4 after multiple valve replacement (mitral PVL), in 1 case after multiple valve replacement (multiple PVL) and in 1 after radical surgery for trilogy of Fallot with tricuspid valve replacement (tricuspid PVL). There were 2 cases diagnosed as infective endocarditis contributing to PVL. The grades of heart function were NYHA Ⅱin 11 patients, NYHA Ⅲin 10, NYHA Ⅳin 7. Results Two patients died in hospital because of septic shock and MODS respectively. One patient developed hypoxic-ischemic encephalopathy after operation. There were 2 patients undergoing second thoracotomy on account of drainage post-operatively. One patient suffered recurrence of PVL. Twenty-two out of 26 patients were followed-up for 12 to 219 months (average 76.21 months) and 4 were lost to follow-up. One patient died of severe pancreatitis 31 months after hospital discharge and 1 had recurrence of PVL with heart function NYHA Ⅲ. Twenty-four survivals are in NYHA class Ⅱ. Conclusion Aggressive operation is effective treatment for moderate-severe PVL diagnosed by cardiac ultrasonography. DOI: 10.11855/j.issn.0577-7402.2016.06.13

In this paper, we report a facile method to successfully fabricate MnO2 nanoflowers loaded onto 3D RGO@nickel foam, showing enhanced biosensing activity due to the improved structural integration of different electrode materials components. When the as-prepared 3D hybrid electrodes were investigated as a binder-free biosensor, two well-defined and separate differential pulse voltammetric peaks for ractopamine (RAC) and salbutamol (SAL) were observed, indicating the simultaneous selective detection of both β-agonists possible. The MnO2/RGO@NF sensor also demonstrated a linear relationship over a wide concentration range of 17 nM to 962 nM (R=0.9997) for RAC and 42 nM to 1463 nM (R=0.9996) for SAL, with the detection limits of 11.6 nM for RAC and 23.0 nM for SAL. In addition, the developed MnO2/RGO@NF sensor was further investigated to detect RAC and SAL in pork samples, showing satisfied comparable results in comparison with analytic results from HPLC.

Co3O4 multilayered structures were successfully synthesized by a facile poly (ethylene glycol 20000) (PEG-20000) assisted hydrothermal technique in combination with calcination method. The final Co3O4 multilayered structures inherited perfectly the morphology of the preliminarily hydrothermal products. Experimental results obtained from the different growth stages demonstrate that the as-prepared precursor exhibit an interesting time-dependent evolution of building blocks, from urchin to multilayer. The possible formation mechanism for the hierarchical structures with various architectures is presented on account of the self-assembled growth induced by Ostwald ripening. Because of the unique structured composed of slices, the photocatalytic activity of the products was examined by measuring the photodecolourisation of methyl violet solution with ultraviolet radiation. The result shows that our products have a good photocatalytic activity.

This paper proposes a new method to predict the spindle deformation based on temperature data. The method introduces ANFIS (adaptive neuro-fuzzy inference system). For building the predictive model, we first extract temperature data from sensors in the spindle, and then they are used as the inputs to train ANFIS. To evaluate the performance of the prediction, an experiment is implemented. Three Pt-100 thermal resistances is used to monitor the spindle temperature, and an inductive current sensor is used to obtain the spindle deformation. The experimental results display that our prediction model can better predict the spindle deformation and improve the performance of the spindle.

Endometriosis (EMS) is a female hormone‑ dependent disease with controversial reports of its etiology and pathogenesis. Apoptosis is particularly important in the human endometrium due to the dynamic cycles of proliferation and shedding. Estrogen possessed antiapoptotic effects on endometrial stromal cells (ESCs), which appears to be exacerbated in women with EMS; however, the underlying mechanism of the antiapoptotic effects of estrogen on ESC remains unknown. The present study aimed to determine whether estrogen regulates the apoptosis of ESCs via thymic stromal lymphopoietin (TSLP) and the associated mechanism. An ELISA was conducted to detect TSLP content in the ESC culture medium treated with estrogen. Subsequently, the early apoptotic rate and expression of B‑cell lymphoma (Bcl‑2) of ESCs were analyzed by flow cytometry in the presence of recombinant human TSLP, anti‑human TSLP neutralizing antibody or estrogen. In the present study, it was reported that ESCs exhibited basal TSLP secretion in the absence of estrogen as reported in previous studies, and that estrogen promoted TSLP secretion of ESCs in a dose‑dependent manner. The results demonstrated that estrogen suppressed the apoptosis of ESCs associated with the promotion of Bcl‑2 expression, which may be partly reversed by inhibiting TSLP. Therefore, the findings of the present study revealed a novel mechanism of estrogen‑dependent apoptotic suppression of ESCs associated with TSLP secretion and Bcl‑2 regulation. Endogenous and estrogen‑induced endometrial TSLP may promote the initiation and development of EMS via the inhibition of apoptosis.

Patients with acute myeloid leukemia (AML) often have a poor prognosis due to drug resistance, which is regarded as a tough problem during the period of clinical therapeutics. It has been reported that autophagy, an important event in various cellular processes, plays a crucial role in mediating drug-resistance to cancer cells. Our study attempts to comparatively investigate the differences of basic and induced autophagic activity between drug-sensitive and multidrug-resistant AML cells. The level of basic autophagy in K562/ADM cells was higher than that in K562 cells, which could be characterized by more cytosolic contents-packaged autophagic vacuoles in K562/ADM cells when compared to that in K562 cells. The observation of MDC staining showed that the fluorescent intensity of autophagosomes in K562/ADM cells was stronger than that in K562 cells. The expression of Beclin1 and the ratio of LC3-II to LC3-I were distinctly higher in K562/ADM cells, however, P62 protein was relatively lower in K562/ADM cells. Furthermore, we found that nutrient depletion could induce autophagic activity of both cell lines. However, autophagic activity of K562/ADM cells was always maintained at a higher level in contrast with K562 cells. ADM (Adriamycin) was also capable of inducing autophagic activity of K562 and K562/ADM cells, but the autophagic alteration in K562 cells appeared earlier. Taken together, our findings suggest that autophagy exerts an important effect on formation and maintenance of drug-resistance in AML cells.

Iatrogenic aortic valve (AV) perforation during non-aortic cardiac operations is a rare complication. The suture-related inadvertent injury to an AV leaflet can produce leaflet perforation with aortic regurgitation after ventricular septal defect repair (VSDR). We report three consecutive patients who had iatrogenic aortic leaflet perforation during VSDR in other hospitals and referred to our hospital for reoperation. In all three cases, the perforated AV leaflets were preserved and repaired by autologous pericardial patch or direct local closure.

Thymic stromal lymphopoietin (TSLP), produced by cervical cancer (CC) cells, promotes angiogenesis, and the recruitment and functional regulation of eosinophils. It has been reported that microRNA (miR)-132 is aberrantly decreased in CC tissues. However, the function and mechanism of TSLP on the biological behaviors of CC cells is largely unknown. The aim of the present study was to investigate the effect of TSLP on the expression of miR-132 and the proliferation and invasion in vitro of CC cell lines, namely, HeLa and SiHa cells. The transcrpitional level of miR-132 was analyzed using reverse transcription-quantitative polymerase chaon reaction. The proliferation, invasion, and the expression of proliferation and invasion-related molecules in HeLa and SiHa cells in vitro were evaluated using bromodeoxyuridine cell proliferation, Matrigel invasion assays, flow cytometry and ELISA, respectively. Here, it was revealed that recombinant human TSLP (rhTSLP) downregulated the expression levels of miR-132 in HeLa and SiHa cells, and by contrast, the neutralizing antibodies for TSLP or TSLP receptor (TSLPR) upregulated miR-132 expression levels in HeLa and SiHa cells. The overexpression of miR-132 resulted in a lowered proliferation and invasiveness, decreased levels of proliferation-associated molecules marker of proliferation Ki-67 and proliferating cell nuclear antigen, and the decreased production of matrix metalloproteinase (MMP)2 and MMP9 in HeLa and SiHa cells. Compared with the control group, there was a higher level of proliferation and invasion in HeLa and SiHa cells following stimulation with rhTSLP. However, these effects induced by rhTSLP were significantly impaired in HeLa and SiHa cells with miR-132 overexpression. The results of the present study indicated that TSLP produced by CC cells downregulated miR-132 expression, and stimulated the proliferation and invasion of CC cells, thereby further promoting the development of CC.

Cervical cancer is one of the most frequently diagnosed cancers and is a major cause of death from gynecologic cancers worldwide; the cancer burden from cervical cancer is especially heavy in less developed countries. Most cases of cervical cancer are caused by persistent infection with carcinogenic human papillomavirus (HPV) genotypes 16 and 18. Non-resolving inflammation caused by HPV infection provides a microenvironment that facilitates cancer development. Molecular alterations during the process of HPV-induced carcinogenesis are characterized by DNA methylation within the HPV genome, promoter hypermethylation of tumor suppressor genes in the host genome, as well as genomic instability caused by viral DNA integrating into the host genome. Catalytic polypeptide-like apolipoprotein B mRNA editing enzymes (APOBECs) normally function as part of the innate immune system. APOBEC expression is stimulated upon viral infection and plays an important role in HPV-induced cervical cancer. APOBECs catalyze the deamination of cytosine bases in nucleic acids, which leads to a conversion of target cytosine (C) to uracil (U) and consequently a change in the single-stranded DNA/RNA sequence. APOBEC proteins mediate the complex interactions between HPV and the host genome and link immunity and viral infection during HPV-driven carcinogenesis. Understanding the effects of APOBECs in HPV-induced cervical carcinogenesis will enable the development of better tools for HPV infection control and personalized prevention and treatment strategies.

The dysfunction of NK cells in women with endometriosis (EMS) contributes to the immune escape of menstrual endometrial fragments refluxed into the peritoneal cavity. The reciprocal communications between endometrial stromal cells (ESCs) and lymphocytes facilitate the development of EMS. However, the mechanism of these communications on cytotoxicity of natural killer (NK) cells in endometriotic milieus is still largely unknown. To imitate the local immune microenvironment, the co-culture systems of ESCs from patients with EMS and monocyte-derived macrophages or of ESCs, macrophages and NK cells were constructed. The cytokine levels in the co-culture unit were evaluated by ELISA. The expression of functional molecules in NK cells was detected by flow cytometry (FCM). The NK cell behaviorsin vitrowere analyzed by cell counting kit-8 and cytotoxic activation assays. After incubation with ESCs and macrophages, the expression of CD16, NKG2D, perforin and IFN-γ, viability and cytotoxicity of NK cells were significantly downregulated. The secretion of interleukin (IL)-1β, IL-10 and transforming growth factor (TGF)-β in the co-culture system of ESCs and macrophages was increased. Exposure with anti-IL-10 receptor β neutralizing antibody (αhIL-10Rβ) or αTGF-β could partly reverse these effects of ESCs and macrophages on NK cellsin vitro. These results suggest that the interaction between macrophages and ESCs downregulates cytotoxicity of NK cells possibly by stimulating the secretion of IL-10 and TGF-β, and may further trigger the immune escape of ectopic fragments and promote the occurrence and the development of EMS.

In this paper, the current problems and demands of campus information system was analyzed and the desktop virtualization solutions were probed. Thus, under the cloud system structure, the user can simply login by the client in desktop systems of data center, as well as traditional computer operating. As a result, the flexibility and security in application of resources are enhanced. More over, schools can avoid the repeated investment and reduce the pressure on hardware and software maintenance, and improve the working efficiency.

To establish the relationships among wilt disease and pathogens and the chemical environment during the allelochemical interactions of plant–microbe–soil, extracts from decaying watermelon tissues were added to media to investigate the effect of allelochemical decaying watermelon tissues on Fusarium oxysporum f.sp. niveum (FON) in vitro. Twelve types of phenolic acids from various decaying watermelon cultivars (Zaojia 82-24 and Jingxing 3) with different levels of resistance to FON were analysed using HPLC. The results demonstrated that there were fewer types and quantities of chemical components released from decaying susceptible watermelon tissues (Zaojia 84-24) than there were from decaying resistant watermelon tissues (Jingxing 3). More antifungal phenolic acids in decaying resistant watermelon tissues (with a ratio of 29.2:1 of antifungal phenolic acids to fungus-stimulation phenolic acids) were observed than in decaying susceptible tissues (with a ratio of 8.4:1 of antifungal phenolic acids to fungus-stimulation phenolic acids). Decaying resistant watermelon tissue extracts inhibited the growth and conidia germination and formation of FON, while decaying susceptible extracts stimulated growth and conidia germination and formation. Extracts from decaying resistant watermelon tissues stimulated virulence factors considerably less than did those from decaying susceptible tissues. We conclude that the decaying plant extracts from susceptible watermelon cultivars stimulated FON because of their higher content of fungus-stimulation phenolic acids, while the decaying resistant watermelon plants inhibited FON because of their higher content of antifungal phenolic acids. These results imply that phenolic acids containing decaying plant residues in soil would induce or inhibit pathogens in plant–microbe interactions.

Morroniside is a water-soluble compound extracted from the fruit of Cornus officinalis and is used to protect lung activity against aging. In the present study, the manner in which morroniside regulates normal lung and cancer cells was examined. The human embryonic lung fibroblast (HELF) cell line and lung cancer A549 cell line, and their responses to morroniside treatment, were examined. Results showed that morroniside reverses the apoptotic effect of H2O2 on HELF cell growth, protecting cell proliferation and normal cell morphology and inhibiting apoptosis. However, these effects were not present in A549 cells. Western blotting showed that morroniside also markedly downregulated retinoblastoma protein in HELF cells. These results suggest that morroniside treatment exhibits different effects on apoptosis in HELF and A549 cells, making it a viable compound for decreasing the side effects of anticancer medicines in normal cells.

A new metal–organic framework, [Cd(TDC)(bix)(H2O)]n (H2TDC = thiophene-2,5-dicarboxylic acid; bix = 1,4-bis(imidazol-1-ylmethyl)benzene), has been synthesized under hydrothermal conditions and structurally characterized by elemental analysis, infrared spectroscopy, thermogravimetric analysis, UV–vis and single X-ray diffraction. Cd-MOF is a 2D infinite layer framework, which is further interconnected by hydrogen-bond interactions leading to a 3D supramolecular architecture. The photoluminescent properties of the Cd-MOF were investigated and this compound shows intense fluorescent emissions in the solid state. In addition, it exhibits good photocatalytic activities for the degradation of methyl organic under UV light irradiation.

In order to help to design the bogies for vehicle, the morphology, microhardness and SHCCT diagram of SMA490BW high strength weather resisting plate was carried out. The morphology were ferrite and little pearlite and granular bainite of samples at different cooling speed. With the cooling rate increased, the t8/5 decreased but the hardness increased. The phase content was composed of pearlite, ferrite and bainite at lower cooling rate, but the phase content was totally composed of bainite.

Ag nanoparticles (AgNP) with an average size of 12 nm are successfully decorated on the reduced graphene oxide (rGO) sheets through a simple one-pot hydrothermal method using gallic acid as the reducing agent. This AgNP/rGO hybrid has been successfully applied in the catalytic performance toward the reduction of H2O2. The nonenzymatic sensor demonstrates a linear relationship in a wide concentration range of 0.05–5 mM (R=0.999), and a high sensitivity of 255 μA cm−2 mM−1 to the detection of H2O2.

A new Cu metal–organic framework (Cu-MOF), [Cu(adp)(BIB)(H2O)]n (BIB = 1,4-bisimidazolebenzene; H2adp = adipic acid), is synthesised under hydrothermal conditions. Single-crystal X-ray analysis reveals a three-dimensional supramolecular architecture with channels originating from two-dimensional layers extended by hydrogen bonds. The cyclic voltammogram of a [Cu(adp)(BIB)(H2O)]n-modified electrode shows a pair of redox peaks at ca. 0.43 V in 0.1 M NaOH solution corresponding to CuIII(OH)-MOF/CuII(H2O)-MOF couple. The Cu-MOF-modified electrode shows high electrocatalytic activity towards H2O2 oxidation with a linear range from 0.1 μM to 2.75 μM and a detection limit of 0.068 μM. In this study, alfalfa protein modified by sodium hydroxide was used to prepare adhesive. The effect on the bonding property by adding different amount of sodium hydroxide was discussed. The upper limit using temperature of alfalfa protein-based adhesive was increased and its bonding property was improved after the protein modified by sodium hydroxide. The adhesive prepared by alfalfa protein modified by 6% sodium hydroxide had the best bonding strength of 2.5MPa.

Wood-plastic composites (WPC) were prepared by compression molding process using high density polyethylene and wood fiber as the main raw materials. The influence of nanozinc oxide, nanosilver antimicrobial powder and rosin amid treatment on WPC was investigated respectively by testing the mass loss, flexural strength, color change, surface tension, as well as surface images of the composites before and after the corrosion test. The results indicate that nanozinc oxide has little antimicrobial effect on WPC. While nanosilver antimicrobial powder and rosin amid both have effects on the anti-corrosion properties of WPC. It is also suggested that the WPC with the mass fraction of 6% rosin amid is the most antimicrobial, and its mass loss and flexural strength only decrease by 0.0788% and 2.24% respectively. These cost-efficient WPC can be practical to industrial application.

The remote sensing measurement of the surface SiO2content has important geological significance .An empirical formula for estimate SiO2content was established based on the correlation between ASTER thermal infrared data and tested Si element from NITON sites in Abra, Australia. The average error of estimated SiO2content is about 15%. ASTER silica alteration derived from SiO2content has good indicative function to find galena. Combined with other prospecting factors as bouger gravity, transient electromagnetic and so on, three vision prospecting targets were optimized, which need more exploration works.

A greenhouse experiment was conducted to assess the adverse impact of transgenic cotton on ecosystem and environment via effect of transgenic Bt+CpTI cotton root exudates on growth and antioxidant activity of conventional parental cotton. Results showed elevated reductive and oxidative species activities in the leaves of conventional parental cotton seedlings treated with varying concentrations of transgenic cotton root exudates. Compared to control, 14.9% to 39.9% increase in catalase, 8.8% to 114% increase in for peroxidase, 21.3% to 59.7% increase in phenylalanine ammonia-lyase and 5.8 to 19.5 fold in ascorbate specific peroxidase was observed. However, biomass and height of conventional cotton seedlings were not affected by any concentration of transgenic cotton root exudates. These results suggested that cultivation of transgenic Bt+CpTI cotton plants poses little risk to conventional parental cotton based on their root interactions.

Endometriosis (EMS) is associated with an abnormal immune response to endometrial cells, which can facilitate the implantation and proliferation of ectopic endometrial tissues. It has been reported that human endometrial stromal cells (ESCs) express interleukin (IL)15. The aim of our study was to elucidate whether or not IL15 regulates the cross talk between ESCs and natural killer (NK) cells in the endometriotic milieu and, if so, how this regulation occurs. The ESC behaviors in vitro were verified by Cell Counting Kit-8 (CCK-8), Annexin/PI, and Matrigel invasion assays, respectively. To imitate the local immune microenvironment, the co-culture system between ESCs and NK cells was constructed. The effect of IL15 on NK cells in the co-culture unit was investigated by flow cytometry (FCM). In this study, we found that ectopic endometrium from patients with EMS highly expressed IL15. Rapamycin, an autophagy inducer, decreased the level of IL15 receptors (i.e. IL15Rα and IL2Rβ). IL15 inhibits apoptosis and promotes the invasiveness, viability, and proliferation of ESCs. Meanwhile, a co-culture with ESCs led to a decrease in CD16 on NK cells. In the co-culture system, IL15 treatment downregulated the levels of Granzyme B and IFN-γ in CD16+NK cells, NKG2D in CD56dimCD16-NK cells, and NKP44 in CD56brightCD16-NK cells. On the one hand, these results indicated that IL15 derived from ESCs directly stimulates the growth and invasion of ESCs. On the other hand, IL15 may help the immune escape of ESCs by suppressing the cytotoxic activity of NK cells in the ectopic milieu, thereby facilitating the progression of EMS.

Based on a synthetic geological study of drilling, well logging and seismic data, core observations and geochemical analysis, it is recognized that Triassic sedimentary facies in the Tahe area of Tarim Basin include braided river deposits filling erosional valleys, and sublacustrine fan, canyon and delta facies. Braided river deposits filling erosional valleys are dominated by coarse-grained lithic quartz sandstone with oblique bedding, and represent the most important sedimentation type of sandstone in the study area. Sublacustrine fan and canyon facies are mainly distributed in the Middle Oil Member. Most delta sediments are deposited in highstand system tract (HST). Because of frequent changes in base level, delta sediments are commonly eroded and rarely preserved. Sedimentary cycles are clearly reflected by lithology, sedimentary structures and well logging data, and are closely related to the changes in lacustrine level. In accordance with the basic principle of sequence subdivision, seven type-I boundaries can be recognized in Triassic strata and six type-I sequences are subdivided correspondingly. In general, lowstand system tract (LST) is well developed within stratigraphic sequences and forms the main body of reservoir sandstone in this area; highstand system tract (HST) and transgressive system tract (TST) are often eroded by upper sequences or missed. Although various factors have different influences on terrestrial sequence stratigraphy, the classical sequence stratigraphy theory proposed by VAIL can be applied to terrestrial strata.

A new Cd(II) coordination polymer, [Cd(PMP)2] n (1) [PMP = 1-phenyl-3-methyl-5-pyrazolone], has been synthesized by the reaction of Cd(CH3COO)2·2H2O with PMP and characterized by elemental analysis, IR spectrum, fluorescence spectrum, UV–vis spectrum, TG–DTA, and electrochemical analysis. Single crystal X-ray diffraction reveals that the complex has a three-dimensional network. Each PMP acts as bidentate bridging ligand and each Cd(II) center is surrounded by two O atoms and two N atoms from four different PMP ligands to form distorted tetrahedral coordination geometry. The photoluminescence study shows the complex exhibited strong solid state blue fluorescent emission at room temperature, and the TG–DTA analysis demonstrated that it has highly thermal stability. Furthermore, the electrochemical property of the complex has been investigated for the first time in DMF by cyclic voltammetry, and the results showed that it has an irreversible process.

A new coordination polymer [Zn(PMP)2Cl] n (PMP = 1-phenyl-3-methyl-5-pyrazolone) has been prepared by the reaction of zinc chloride and PMP, and characterized by elemental analysis, IR, UV, and fluorescence spectra, TG/DTA, and electrochemical analysis. Single crystal X-ray diffraction revealed that the complex is monoclinic, space group P 2(1)/n, with a = 10.850(17), b = 17.578(2), c = 10.997(18) A, β = 104.999(3)°, V = 2025.8(5) A3, Z = 4, Dc = 1.470 mg m−3, F(000) = 920, goodness-of-fit = 0.905, and R 1 = 0.0441. The complex is a 1-D coordination polymer, and the coordination geometry of Zn(II) is a distorted tetrahedron.

Two new annonaceous acetogenins named as squamostanin-A and squamostanin-B were isolated from 95% EtOH extract of the seeds of Annona squamosa. Their structures were determined by spectroscopic method, and their cytotoxicities were evaluated using MTT method.

A new copper Schiff-base complex [Cu(C15H23N3O2)(NO3)]NO3 was synthesized and characterized by spectroscopic (IR), thermal (TG/DTA) and electrochemical methods. Single crystal X-ray diffraction revealed that the complex is monoclinic, space group P2(1)/c, with a = 23.549(2), b = 6.583(12), c = 12.765(15) A, β = 102.823(2)°, V = 1929.4(5) A3, Z = 4, D calcd = 1.601 Mg m−3, F(000) = 964, goodness-of-fit = 1.034 and R 1 = 0.0407. The Cu(II) is four-coordinate with distorted square planar geometry. The structure consists of isolated [Cu(C15H23N3O2)(NO3)]+ and a nitrate, which are further linked to a 3D network by significant N–H ··· O, N–H ··· N hydrogen bonds and weak O ··· Cu interaction.
