Humans have an immediate perception of shapes and surroundings through their senses and their cognitive capabilities. This innate ability enables the manual inspection of components in manufacturing environments. Trained inspectors combine their senses and handling skills with bespoke non-destructive testing instrumentation. However, manual inspections can be slow for large and/or complex geometries and prone to human factors. Automated non-destructive testing systems have emerged in recent years, to increase data acquisition speed, part coverage and inspection reliability. These tools work well when the robotic inspection takes place in a well-structured environment and an accurate part model is available. However, precisely registering the position of a part in the robot reference system makes the inspection setup very time-consuming. Furthermore, the geometry of a part may differ from its digital model, spoiling the inspection accuracy. This work introduces a new approach that mimics the human perception capability and gives full manipulation autonomy to robotic sensing applications. We use a single robotized sensor to introduce a fully autonomous single-pass geometric and volumetric inspection of complex parts. Our approach can be used to solve some key challenges in quality assurance for Industry 4.0 and can find applicability beyond robotic non-destructive testing.

Robotic sensing is used in many sectors to improve the inspection of large and/or complex parts, enhancing data acquisition speed, part coverage and inspection reliability. Several automated or semi-automated solutions have been proposed to enable the automated deployment of specific types of sensors. The trajectory to be followed by a robotic manipulator is typically obtained through the offline programmed tool paths for the inspection of a part. This method is acceptable for a part with known geometry in a well-structured and controlled environment. The part undergoing assessment needs to be precisely registered with respect to the robot reference system. It implies the need for a setup preparation phase for each new part, which can be very laborious and reliant on the human experience. This work combines real-time robot control and live sensor data to confer full autonomy to robotic sensing applications. It presents a novel framework that enables fully autonomous single-pass geometric and volumetric inspection of complex parts using one single robotised sensor. A practical and robust robot control sequence allows the autonomous correction of the sensor orientation and position to maximise the sensor signal amplitude. It is accompanied by an autonomous in-process path planning method, capable of keeping the inspection resolution uniform throughout the full extension of the free-form parts. Last but not least, a by-product of the framework is the progressive construction of the digital model of the part surface throughout the inspection process. The introduced framework is scalable and applicable to widely different fields.

This work presents computer-aided scan path generation for robotic non-destructive testing of complex shaped test-pieces. Off-line programmed scan path was used to robotically inspect an aluminium fixed leading edge skin panel of an aircraft wing by means of swept frequency eddy currents method. Eddy currents probe was deployed by means of a six-axis robotic arm KUKA KR5 arc. Reverse engineering of the test-piece was carried out to reconstruct CAD model of its surface. Positioning accuracy of the performed continuous scan was measured with a laser tracker in accordance with ISO 9283:1998 and is reported in the paper. The positional uncertainty of the NDT scan calculated as the standard deviation of the measured path coordinates from the command path coordinates does not exceed 0.5 mm which is rather moderate taking in account uncertainties associated with the off-line robot programming.

Type 2 diabetes mellitus (T2DM) is a common complication of obesity. Here, we have shown that activation of the IgG receptor FcγRIIB in endothelium by hyposialylated IgG plays an important role in obesity-induced insulin resistance. Despite becoming obese on a high-fat diet (HFD), mice lacking FcγRIIB globally or selectively in endothelium were protected from insulin resistance as a result of the preservation of insulin delivery to skeletal muscle and resulting maintenance of muscle glucose disposal. IgG transfer in IgG-deficient mice implicated IgG as the pathogenetic ligand for endothelial FcγRIIB in obesity-induced insulin resistance. Moreover, IgG transferred from patients with T2DM but not from metabolically healthy subjects caused insulin resistance in IgG-deficient mice via FcγRIIB, indicating that similar processes may be operative in T2DM in humans. Mechanistically, the activation of FcγRIIB by IgG from obese mice impaired endothelial cell insulin transcytosis in culture and in vivo. These effects were attributed to hyposialylation of the Fc glycan, and IgG from T2DM patients was also hyposialylated. In HFD-fed mice, supplementation with the sialic acid precursor N-acetyl-D-mannosamine restored IgG sialylation and preserved insulin sensitivity without affecting weight gain. Thus, IgG sialylation and endothelial FcγRIIB may represent promising therapeutic targets to sever the link between obesity and T2DM.

Item does not contain fulltext OBJECTIVE: Pregnancies in women with the antiphospholipid syndrome (APS) are frequently complicated by fetal loss and intrauterine growth restriction (IUGR). How circulating antiphospholipid antibodies (aPL) cause pregnancy complications in APS is poorly understood. We sought to determine whether the low-density lipoprotein receptor family member apolipoprotein E receptor 2 (ApoER2) mediates trophoblast dysfunction and pregnancy complications induced by aPL. METHODS: Placental and trophoblast ApoER2 expression was evaluated by immunohistochemistry and immunoblotting. Normal human IgG and aPL were purified from healthy individuals and APS patients, respectively. The role of ApoER2 in aPL-induced changes in trophoblast proliferation and migration and in kinase activation was assessed using RNA interference in HTR-8/SVneo cells. The participation of ApoER2 in aPL-induced pregnancy loss and IUGR was evaluated in pregnant ApoER2(+/+) and ApoER2(-/-) mice injected with aPL or normal human IgG. RESULTS: We found that ApoER2 is abundant in human and mouse placental trophoblasts and in multiple trophoblast-derived cell lines, including HTR-8/SVneo cells. ApoER2 and its interaction with the cell surface protein beta2 -glycoprotein I were required for aPL-induced inhibition of cultured trophoblast proliferation and migration. In parallel, aPL antagonism of Akt kinase activation by epidermal growth factor in trophoblasts was mediated by ApoER2. Furthermore, in a murine passive-transfer model of pregnancy complications of APS, ApoER2(-/-) mice were protected from both aPL-induced fetal loss and aPL-induced IUGR. CONCLUSION: ApoER2 plays a major role in the attenuation of trophoblast function by aPL, and the receptor mediates aPL-induced pregnancy complications in vivo in mice. ApoER2-directed interventions can now potentially be developed to combat the pregnancy complications associated with APS.

Introduzione In connessione con i cambiamenti socio-antropologici che caratterizzano la nostra epoca post-moderna, si è proposto il concetto di disidentità (Ferraro, Lo Verso, 2007, 2008; Ferraro, 2011; Ferraro, Giannone, Lo Verso, 2012) come chiave di lettura della condizione esistenziale espressa oggi da molti giovani-adulti e come cartina di tornasole rispetto all’affermarsi di nuovi quadri psicopatologici. Lo studio analizza i vissuti legati al tema della disidentità, per individuarne le forme d’espressione e valutare le capacità del gruppo d’intervenire su di essi. Metodo In 3 focus group (21 giovani adulti) a conduzione psicodinamica, sono stati somministrati prima e dopo l’esperienza di gruppo, test che esplorano alcuni livelli di organizzazione dell’identità: differenziazione del sé (DSI-R, Skowron, Fiedlander, 1998; Skowron, Schmitt, 2003), modalità relazionali (IIP, Horowitz et al., 1988), autostima (R-SES (Rosenberg, 1965; Prezza, Trombaccia, 1997), eventuali sofferenze psichiche, con particolare riguardo agli ambiti relazionali e lavorativi (OQ-45, Lambert et al., 1996; Lo Coco et al., 2006). Per i processi di gruppo (pre-post), sono stati utilizzati: GQ (Bormann, et al., 2011 ), CALPAS-group (Gaston, Marmar, 1993), GMLCS (Piper et al., 1983). Sono state condotte analisi descrittive e di associazione. Risultati Cambiamenti positivi significativi emergono in diversi aspetti indagati. Molti sono significativamente associati tra loro. Il processo dei gruppi rivela un crescendo di coesione e alleanza. Meno numerose ma interessanti sono le associazioni tra gli indici di processo e quelli di esito. Conclusioni Lo studio ha consentito d’individuare componenti empiriche utili alla prosecuzione della riflessione clinica sulla disidentità. I risultati consentono anche approfondimenti sul set(ting) gruppale, come esperienza sociale favorevolmente associata al miglioramento delle condizioni indagate.

Potocytosis represents a mechanism by which small and large molecules as well as macromolecular complexes are sequestered and transported by caveolae. Caveolae are flask-shaped plasma membrane specializations characterized by a filamentous coat consisting of caveolins that decorates the inside surface of each caveola membrane. They have endocytotic functions that differ from the clathrin-coated pit pathway. Ligands bound to receptors that are internalized by caveolae can be delivered to four different locations in the cell bypassing the lysosome and at least four different caveolae membrane traffic patterns during potocytosis can be distinguished. Hence, cells have two endocytic machines and each is designed to accomplish different tasks. This review provides a brief summary of the discovery of caveolae and of potocytosis, and focuses on recent discoveries of the unique endocytic capabilities of caveolae in a variety of different cells.

The high-normal blood pressure (also known as prehypertension) is a clinical condition characterized by an increased cardiovascular risk as well as by the presence of target organ damage. This include an increased left ventricular mass, an endothelial dysfunction and an early renal functional and structural damage. Whether this is the case also for alterations of retinal vessels network, which are frequently detectable in established hypertension, is still largey undefined. The present paper, after discussing the main characteristics of the high-normal blood pressure state, will review the different approaches used throughout the years for assessing retinal microcirculatory network. Data collected by our group in subjects with high normal blood pressure will be also discussed, showing that arterial venular ratio values are reduced in this individuals with high-normal blood pressure and more so in established hypertension. These data indicate that retinal microvascular alterations 1) are of early appearance in the clinical course of hypertension and 2) are of frequent detection in the high-normal blood pressure state. The possible hemodynamic and non-hemodynamic mechanisms resposible for these structural alteations of the retinal microcirculation will be also discussed. © 2013 Bentham Science Publishers.
