Alkaline suppress plant growth because Fe is insolubilized. Swiss chard ( Beta vulgaris L.) grown in alkaline condition with high Na levels such as aquaculture wastewater in dry lands take up Fe and grows well, but the mechanisms of Fe uptake and growth promotion by Na are still unknown. Here, we investigated the effects of alkaline pH, Fe deficiency, and high salt on Fe uptake, translocation, and distribution in salt tolerant epazote ( Dysphania ambrosioides L.), natrophilic Swiss chard, and euhalophyte romerito ( Suaeda edulis Flores Olv. & Noguez). These three species were grown for 6 days in 8 treatments at pH 5.0 or 8.2, 0.02 or 2.00 ppm Fe and 5 or 100 mM NaCl. We measured changes in medium pH, ferric-chelate reductase (FCR) activity in roots, Fe contents, and Na contents. Root ferric-chelate reductase activity was similar at pH 5.0 and pH 8.2 in all species. At high pH and salt, the medium pH in Swiss chard and romerito decreased markedly with time, and this acidification solubilized Fe and Fe uptake tended to increase. For adjusting the cation–anion balance, uptake of large amounts of Na could promote H release, resulting in the medium acidification. High salt increased the distribution of Fe to petioles in Swiss chard and to leaves in romerito, suggesting that Na contributes to the translocation of Fe from the root apoplast upward to the shoot. The facts that Na intensifies Fe uptake and translocation to the shoots could be a mechanism to promote the growth of Swiss chard and romerito at alkaline condition.

Aeration is considered beneficial for hydroponics. However, little information is available on the effects of aeration, and even less on solutions that use bubble flow and their agronomic effects. In this study, the effects of aeration intensity on plants were studied through cultivation experiments and flow field visualization. It was found that the growth of plants did not increase linearly with an increase in aeration intensity. From the results of this study, when the aeration intensity was within the low range (0.07–0.15 L·L−1 NS·min−1), increasing the aeration intensity increased the plant growth. However, after the aeration intensity reached a certain extent (0.15–1.18 L·L−1 NS·min−1), some indicators did not change significantly. When the aeration intensity continued to increase (1.18–2.35 L·L−1 NS·min−1), growth began to decrease. These results show that for increasing dissolved oxygen and promoting plant growth, the rule is not “the higher the aeration intensity, the better”. There is a reasonable range of aeration intensity within which crops grow normally and rapidly. In addition, increasing the aeration intensity means increasing energy utilization and operating costs. In actual hydroponics production, it is very important to find a reasonable aeration intensity range.

In hydroponics, the flow pattern of nutrient solution in a cultivation container affects the growth of plants. Even if the flow rate of nutrient solution is the same between containers, the flow pattern may differ based on the size and shape of the containers. Therefore, the flow pattern cannot be comprehensively described by flow rate alone. In order to identify the relationship between plant growth, root morphology, nutrient uptake, and flow pattern, a hydroponic cultivation of Swiss chard was carried out. In addition, in order to describe the flow pattern in a specific cultivation container, hydroponic flow patterns were observed via flow field visualization using particle image velocimetry. As a result, with the increase in flow rate, it was found that a specific flow rate can form an ideal flow pattern for plant growth. Under this flow pattern, nutrient absorption is promoted and roots are elongated, thereby absorbing more nutrients and further promoting plant growth. However, when the flow rate exceeds the ideal value, plant growth is hindered. In summary, identifying the ideal nutrient solution flow pattern in hydroponics can facilitate better crop production.

Genetic testing is needed for the treatment of colorectal cancer (CRC), especially molecular-targeted therapy. The effects of anti-EGFR therapy and prognosis are affected by the presence of KRAS mutations. However, whether primary CRC or metastatic tissues are appropriate in the analysis is still unclear. In the present study, we assessed the concordance of KRAS/BRAF mutation status and microsatellite instability (MSI) in primary CRC and corresponding metastases. This study enrolled 457 patients with surgically resected primary and corresponding metastatic CRC (499 synchronous metastases and 57 metachronous metastases) and seven local recurrences, and KRAS/BRAF mutation and MSI status were analysed for these tumours. The concordance rates of KRAS mutation, BRAF mutation, wild-type, MSI-H and MSS between primary CRC and corresponding metastases were 93.9% (214/228), 100% (30/30), 99.3% (304/306), 87.5% (21/24) and 100% (137/137), respectively. These high concordance rates were not different between synchronous and metachronous metastases. In conclusion, a high concordance of KRAS/BRAF mutation status and MSI status was observed between primary CRC and corresponding metastases in this study. Either primary CRC or metastatic tissues can be used for testing KRAS/BRAF mutation status and MSI status.

The amaranthaceous dwarf glasswort, Swiss chard, table beet, spinach and Mexican tea were grown in solutions containing 0, 20, 40, 80, 120, 160, 180 and 200 mol m−3 sodium chloride (NaCl). Maximum growth and increase of biomass production compared to that at 0 mol m−3 of dwarf glasswort was observed at 200 mol m−3 (245%), and Swiss chard (146%), table beet (128%) and spinach (138%) at 80 mol m−3. The growth of these species increased with increasing sodium (Na) concentration of shoot until it reached 4.18, 2.42, 1.60 and 1.58 mol kg−1, respectively. These indicate that the order of Na-loving character is dwarf glasswort >> Swiss chard > table beet = spinach. The water contents increased with increasing Na concentration until growth reached maximum in dwarf glasswort and Swiss chard. In these highly Na-loving species, Na is activity transported to shoots and utilized for producing osmotic pressure to absorb water.

Salinity is known to reduce NO 3 − uptake in many plants. Our previous study found that the increasing NaCl concentrations in media enhanced NO 3 − uptake in leaves. Here, we investigated whether nitrate ( NO 3 − ) uptake by Swiss chard (Beta vulgaris var. cicla L.) was affected more by Na+ than by K+, and elucidated which NO 3 − uptake processes were related to the application of these ions in the culture medium. Plants were grown hydroponically and subjected to 5 mM or 50 mM of either K or Na treatments for 7 days: K5 (K 5, Na 0), K50 (K 50, Na 0), Na5 (K 0, Na 5), and Na50 (K 0, Na 50). The leaf NO 3 − concentration was significantly enhanced in the Na treatments compared with the K treatments. The ratio of leaf to root NO 3 − concentrations in K5 and Na5 were 0.46 and 1.59, respectively, which indicates that NO 3 − is translocated to the leaves in the Na treatments. The NO 3 − concentration in the xylem sap, collected 6 h after the start of the treatments, was significantly higher in Na50 than in the other treatments. These are the opposite of the results for Arabidopsis, which accumulates NO 3 − in its roots to increase its salinity tolerance. Our results suggest that Na but not K enhances NO 3 − uptake and transport to the leaves in Swiss chard and that its function may relate to xylem translocation of NO 3 − .

Saline soil and saline groundwater reduce agricultural productivity on drylands. We are developing a new aquaponics system to improve food productivity on such lands while effectively utilizing saline groundwater. In this study, cultivation of Swiss chard (Beta vulgaris L. spp. cicla cv. Seiyou Shirokuki) was carried out using fish wastewater with a high salt concentration (1150 mg L−1 NaCl). The levels of microelements (e.g., Fe, Mn, Zn, and Cu) in the fish wastewater were very low, so we added microelements at 100% (W100), 50% (W50), 25% (W25), and 0% (W0) of the levels in the standard hydroponics solution to the fish wastewater and investigated the effects on growth of Swiss chard. At the first harvest, yields in all wastewater treatments were as high or higher than in the control. At the second harvest, yields in W100, W50, and W25 were not significantly different from the control, while in W0 the yield was significantly lower and chlorosis was evident. At the third harvest, the yield in all wastewater treatments was less than in the control, and chlorosis symptoms were observed in W25 and W0. Since leaf Mn and Zn concentrations in W25 and W0 had decreased to below the critical values for those microelements, Mn and Zn deficiency might have contributed to the observed chlorosis and yield loss. For the cultivation of Swiss chard with fish wastewater, sufficient yield (i.e., comparable to or better than the control) without chlorosis was obtained when microelements were added at 50% of the level of the control solution. In addition, since sufficient yield was obtained even in W0 at the first harvest, it is suggested that longer-term cultivation and higher yield could be achieved by applying 50% microelements after the first harvest.

Swiss chard (Beta vulgaris var. cicla L.) is a halophilic plant, which has increased growth under moderate salinity. The stimulation of total nitrogen (N) concentration was also observed with the increase of sodium chloride (NaCl) levels in previous studies. The purpose of this study was to investigate whether the stimulation of N uptake is the result of growth stimulation or the direct effect of NaCl. We compared the effects of NaCl on nitrate (NO3–) uptake and nitrate reductase (NR) activity in halophilic Swiss chard with those in salinity-tolerant barley (Hordeum vulgare L.). Both species were grown hydroponically at NaCl concentrations ranging from 0 to 100 mmol L−1. Leaf dry weight of both species was not significantly affected by the treatments. Microsatellite instability (MSI) is an important biomarker for screening for Lynch syndrome, and also of response to immune checkpoint inhibitors. The aim of this study is to create a predictive model to determine which elderly patients with colorectal cancer (CRC) should undergo MSI and/or immunohistochemistry testing on the basis of clinicopathological data. We analyzed a test cohort of CRC patients aged ≥50 years (n = 2219) by multivariate logistic regression analyses to identify predictors of high‐frequency MSI (MSI‐H). The created prediction model was validated in an external cohort (n = 992). The frequency of MSI‐H was 5.5% among CRC patients aged ≥ 50 years. The following five predictors of MSI‐H were identified in the test cohort: female (1 point), mucinous component (2 points), tumor size ≥ 60 mm (2 points), location in proximal colon (3 points), and BRAF mutation (6 points). The area under curve (AUC) in the receiver‐operating characteristic (ROC) analysis of this prediction model was 0.832 (95% confidence interval: 0.790–0.874). The sensitivity and specificity were 74.4% and 77.7%, respectively, for a cut‐off score of 4 points. The receiver‐operating characteristic curve of the validation cohort also showed an AUC of 0.856 (95% CI: 0.806–0.905). This prediction model is useful to select elderly CRC patients who should undergo MSI testing, and who may benefit from treatment with 5‐FU‐based adjuvant chemotherapy and cancer immunotherapy.

Background: A recent clinical trial on the immune check-point inhibitor pembrolizumab demonstrated that microsatellite instability (MSI) is a good biomarker for response to this inhibitor. However, clinicopathological features of advanced colorectal cancer (CRC) with high-frequency MSI (MSI-H) are unclear. Patients and Methods: A total of 2,439 surgically resected CRC tissues were analyzed for MSI status, and mutational status of V-Ki-Ras2 Kirsten rat sarcoma 2 viral oncogene homolog (KRAS), neuroblastoma RAS viral oncogene homolog (NRAS) and v-Raf murine sarcoma viral oncogene homolog B (BRAF). Stage IV cases were selected, and clinical and molecular features were evaluated. Results: There was no significant survival difference observed between MSI-H CRC and microsatellite-stable (MSS) CRC in patients with stage IV disease (3.92 vs. 2.50 years; p=0.766). However, hematogenous and lymphogenous metastasis-dominant CRC with MSI-H demonstrated poor prognosis, whereas peritoneal metastasis-dominant CRC with MSI-H demonstrated good prognosis, (1.33 vs. 5.2 years; p=0.006). Conclusion: Prognosis of stage IV CRC with MSI-H depended on the metastatic pattern. These findings provide useful information for the adaptation of CRC immunotherapy.

A toxic protein, dubbed molybdophyllysin, was isolated from the tropical toadstool Chlorophyllum molybdites by following its lethal effect in mice. Analysis of the protein using SDS-PAGE revealed a single 23-kDa band. Sequence analysis of molybdophyllysin tryptic fragments showed that this protein is highly homologous to metalloendopeptidases (MEPs) obtained from edible mushrooms, such as Grifola frondosa, Pleurotus ostreatus, and Armillaria mellea. These proteins include a HEXXH+D zinc-binding motif known as aspzincin. Accordingly, molybdophyllysin is a member of the deuterolysin family of zinc proteases. Molybdophyllysin retained its proteolytic activity at temperatures up to 60°C with an optimum pH of 7.0. The activity was inhibited by both 1,10-phenanthroline and N-bromosuccinimide, but molybdophyllysin exhibited strong resistance to SDS.

A typical filamentous bacterium, Sphaerotilus natans, secretes a thiolic glycoconjugate which is assembled into a microtube, so called sheath. The glycoconjugate is known to consist of a pentasaccharide-dipeptide repeating unit, but its chemical structure has not been completely elucidated. In order to determine its chemical structure, the sheath was broken down by performic acid oxidation. The released sulfonated derivative was water soluble which was suitable for detailed NMR analysis. The data exhibited the presence of two stoichiometric and one substoichiometric (relative abundance was about 0.5) acetylations, suggesting that the glycoconjugate is composed of two equimolar pentasaccharide-dipeptide repeating units each having either two or three acetyl groups. However, the position of substoichiometric acetylation could not be defined. To determine the position, the sheath was derivatized with a thiol selective fluorescent reagent followed by digestion with a specific polysaccharide lyase prepared from a sheath-degrading bacterium, Paenibacillus koleovorans. As expected, two fluorescent digests were recovered by reverse-phase HPLC and were subjected to NMR analysis. The data revealed that both digests are pentasaccharide-dipeptides which have unsaturated glucuronic acid and galactosamine residues at their reducing and non-reducing ends, respectively. It was also confirmed that one digest has 3-O-acetylated glucose residue while the other has non-derivatized glucose residue. The substoichiometric acetylation was thus identified with the 3-O-acetylation, and structural determination of the thiolic glycoconjugate was completed. By virtue of the clarification of the two digests' structures, the cleavage site was specified as (1→4)-α-galactosaminic bond to glucuronic acid. Based on the present and earlier findings, we propose a novel glycoconjugate category named thiopeptidoglycan and a novel polysaccharide lyase named thiopeptidoglycan lyase.

The present study aimed to investigate the effects of K-type and Ca-type artificial zeolites on the growth and water and element absorptions of kidney bean (Phaseolus vulgaris L.), tomato (Lycopersicon esculentum Mill), maize (Zea mays L.) and beet (Beta vulgaris L.) in high sodic soil. Tottori sand dune soil, which was used as a control, was converted to high sodic soil mixed with salts. Each type of zeolite was mixed into the high sodic soil at rates of 0, 1, 2 and 5%. The results showed that kidney bean, tomato and maize died in high sodic soil 25–27 days after transplanting (DAT), whereas beet survived, although its growth was extremely suppressed at 26 DAT. The addition of Ca-type zeolite improved growth in all of the tested plants. Even 4 DAT the growth of beet was improved by recovery of water absorption, and growth of tomato was improved by recovery of Ca and K absorptions and cation balance, and restriction of Na absorption. A toxic protein, called bolevenine, was isolated from the toxic mushroom Boletus venenatus based on its lethal effects on mice. On SDS-PAGE, in either the presence or absence of 2-mercaptoethanol, this protein showed a single band of approximately 12 kDa. In contrast, based on gel filtration and MALDI-TOFMS, its relative molecular mass was estimated to be approximately 30 kDa and approximately 33 kDa, respectively, indicating that the protein consists of three identical subunits. This toxin exhibited its lethal activity following injection at 10mg/kg into mice. The N-terminal amino acid sequence was determined up to 18, and found to be similar to the previously reported bolesatine, a toxic compound isolated from Boletus satanas.

Beets were grown on soils with various exchangeable sodium percentages (ESP). A saline non-sodic soil (SA, ESP = 3.2), a saline sodic soil (SO, ESP = 23), and a saline high sodic soil (HSO, ESP = 78) were prepared from Tottori sand dune soil (CO). K-type and Ca-type artificial zeolites (50 g kg−1) were applied to these soils in order to evaluate their effects on the chemical properties of saturation extracts of the soils, water deficit, cation uptake and transport, and cation balance of beet plants. In the zeolite-free treatments, beet growth was accelerated in SA and SO, but was suppressed in HSO compared with CO. The addition of both types of zeolites ameliorated plant growth in all the soils studied, especially HSO. The relative dry weight of the soils treated by the K-type zeolite to the zeolitefree soil was 189% for CO, 125% for SA, 130% for SO, and 222% for HSO. Type I crops (wheat and soybean), in which the potassium concentration (Kc) decreases when the nitrogen concentration (Nc) decreases, and type II crop (sunflower), in which Kc remains constant when Nc decreases, were grown in K-sufficient and K-deficient soils in pots. The CO2 exchange rate (Pnet; photosynthetic rate, PR; photorespiratory rate, and DR; dark respiratory rate), the distribution of photosynthesized 14C to organs, and the chemical compounds and free amino acids in fully expanded leaves were investigated. A greenhouse experiment was conducted to investigate the effect of exchangeable Na on the growth and absorption of metal elements in barley, rye, and maize. The plants were cultivated in soils whose exchangeable sodium percentages (ESP) were 6.6 (saline soil: Saline), 17.4 (saline-sodic soil: Sodic 1), and 39.6 (sodic soil: Sodic 2), which were prepared from Tottori sand dune soil (Control). The dry weight (DW) and concentrations of metal elements Ca, Mg, Mn, Zn, and Cu) in shoots were analyzed. The shoot DW was smaller with higher ESP, but in barley the difference between all the treatments was no longer observed with time. In Sodic soils, the growth of barley was vigorous, whereas rye growth was poor, and maize plants died by 5 weeks after planting. The reaction center-binding protein D1 of photosystem II (PS II) undergoes rapid turnover under light stress conditions. In the present study, we investigated the role of the extrinsic 33 kDa protein (OEC33) in the early stages of D1 turnover. D1 degradation was measured after strong illumination (1000-5000 microE m-2 S-1) of spinach manganese-depleted, PSII-enriched membrane and core samples in the presence and absence of the OEC33 under aerobic conditions at room temperature. PSII samples lacking the OEC33 were prepared by standard biochemical treatments with Tris or CaCl2/NH2OH while samples retaining the OEC33 were prepared with NH2OH or NaCl/NH2OH. The degradation of D1, monitored by SDS/urea-polyacrylamide gel electrophoresis and Western blotting using specific antibodies against D1, proceeds to a greater extent in NH2OH-treated samples than in Tris-treated samples over a 60 min illumination period. Under the same conditions, significantly more aggregation of D1 occurs in the Tris-treated samples than in the NH2OH-treated samples. The lower level of D1 degradation in Tris-treated samples is not due to secondary proteolysis, as judged from the time course for degradation at 25 degrees C or the degradation pattern at 4 degrees C. Similarly, for NaCl/NH2OH-treated samples, D1 degradation is greater and D1 aggregation less than in CaCl2/NH2OH-treated samples. The effect of the presence of the OEC33 on D1 degradation and aggregation is confirmed by reconstitution experiments in which the isolated OEC33 is restored back to Tris-treated samples. During very strong illumination, significant loss of CP43 also occurs in Tris-treated but not in NH2OH-treated samples. Structural analysis of PS II core complexes by Fourier transform infrared (FT-IR) spectroscopy revealed very little change in the protein secondary structure after 10 min illumination of NH2OH-treated samples while a large 10% decrease of alpha-helix content occurs in Tris-treated samples. On the basis of these results, we suggest that either (1) the OEC33 stabilizes the structural integrity of PS II such that it prevents the photodamaged D1 protein from aggregating with nearby polypeptides and thereby facilitating degradation or (2) the OEC33 specifically stabilizes CP43, a putative D1-specific protease, which normally promotes the efficient degradation of D1.

A sheath-forming sulfa oxidizer, Thiothrix nivea , was mixotrophically cultured in a medium supplemented with acetic acid and sodium disulfide. Its sheath, a microtube-like extracellular supermolecule, was prepared by selectively removing the cells with lysozyme, sodium dodecyl sulfate, and sodium hydroxide. The sheath was not visibly affected by hydrazine treatment, suggesting that it is not a proteinous supermolecule. From the acid hydrolysate of the sheath, glucose and glucosamine were detected in an approximate molar ratio of 1:1. Three other saccharic compounds were detected and recovered by HPLC as fluorescent derivatives prepared by reaction with 4-aminobenzoic acid ethyl ester. Nuclear magnetic resonance (NMR) analysis suggested that one of the derivatives was derived from an unidentified deoxypentose. NMR analysis for the other 2 derivatives showed that they were derived from β-1,4-linked disaccharides and tetrasaccharides, which were composed of glucose and glucosamine. The sheath was readily broken down by weak HCl treatment, releasing an unidentified deoxypentose and polymer. Chemical analysis showed the presence of β-1,4-linked d -Glc p and d -GlcN p in the polymer. NMR analysis revealed that the polymer had a repeating unit of →4)- d -Glc p -(β1→4)- d -GlcN p -(β1→. The solid-state 1D- 13 C NMR spectrum of the polymer in N -acetylated form supported this result. The molecular weight of the polymer was estimated to be 8.2 × 10 4 by size exclusion chromatography. Based on these results, the sheath of T. nivea is hypothesized to be assembled from alternately β-1,4-linked glucosaminoglucan grafted with unidentified deoxypentose.

Ingestion of the toxic mushroom Boletus venenatus causes a severe gastrointestinal syndrome, such as nausea, repetitive vomiting, diarrhea, and stomachache. A family of isolectins (B. venenatus lectins, BVLs) was isolated as the toxic principles from the mushroom by successive 80% ammonium sulfate-precipitation, Super Q anion-exchange chromatography, and TSK-gel G3000SW gel filtration. Although BVLs showed a single band on SDS-PAGE, they were further divided into eight isolectins (BVL-1 to -8) by BioAssist Q anion-exchange chromatography. All the isolectins showed lectin activity and had very similar molecular weights as detected by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS) analysis. Among them, BVL-1 and -3 were further characterized with their complete amino acid sequences of 99 amino acids determined and found to be identical to each other. In the hemagglutination inhibition assay, both proteins failed to bind to any mono- or oligo-saccharides tested and showed the same sugar-binding specificity to glycoproteins. Among the glycoproteins examined, asialo-fetuin was the strongest inhibitor. The sugar-binding specificity of each isolectin was also analyzed by using frontal affinity chromatography and surface plasmon resonance analysis, indicating that they recognized N-linked sugar chains, especially Galbeta1-->4GlcNAcbeta1-->4Manbeta1-->4GlcNAcbeta1-->4GlcNAc (Type II) residues in N-linked sugar chains. BVLs ingestion resulted in fatal toxicity in mice upon intraperitoneal administration and caused diarrhea upon oral administration in rats.

Finasteride is a type 2 5 alpha-reductase inhibitor that inhibits conversion of testosterone to dihydrotestosterone, a key mediator of male pattern hair loss (androgenetic alopecia). The objective of this study was to identify the optimal dosage of finasteride and to evaluate its efficacy and safety in the treatment of Japanese men with male pattern hair loss. In this double- blind randomized study, 414 Japanese men with male pattern hair loss received finasteride 1 mg (n = 139), finasteride 0.2 mg (n = 137), or placebo (n = 38) once daily for 48 weeks. Efficacy was evaluated by global photographic assessment, patient self-assessment, and investigator assessment. All efficacy endpoints showed significant improvement with finasteride therapy by 12 weeks (p0.05 versus placebo). At 48 weeks, 58%, 54%, and 6% of men in the finasteride 1 mg, finasteride 0.2 mg, and placebo groups, respectively, had improved based on assessments of global photographs. All efficacy endpoints were numerically superior for the 1 mg dose over the 0.2 mg dose at 48 weeks. Finasteride treatment was generally well tolerated. Finasteride 1 mg\day slows hair loss and improves hair growth in Japanese men with male pattern hair loss.

A case of pyoderma gangrenosum on the dorsal aspect of the right second and third fingers of a 50-year-old woman is reported. The patient had been wounded at the above-mentioned site by a cutter. This wound developed into a painful ulcer which enlarged rapidly. No bacterial infections were detected. Histologically, an inflammatory infiltrate of lymphocytes mixed with neutrophils and nucleodusts was seen throughout the entire dermis, but there was no evidence of vasculitis. Based on this data, the case was diagnosed as pyoderma gangrenosum. The patient responded to treatment with systemic corticosteroids. Pyoderma gangrenosum at this site of the fingers seems to be rare.
