To understand and confront the ongoing Asian/Chinese hate in the USA as another pandemic virus, this article digs into the root cause in history, as anti-Chinese sentiments are nothing new but invariably in a new context and with a new trigger. A close examination of how a racial hierarchy was constructed by the dominant group sets the stage for the study. The paradoxical relationship between the American ideal of equality and the racial hierarchy is discussed in depth. In doing so, we focus on the Chinese experience in the nineteenth century—the construction of the Transcontinental Rails and the Chinese Exclusion Act (1882–1943)—to argue that the roots of anti-Chinese sentiments rest in the racial hierarchy as well as its coexistence with the lofty ideal of freedom and equality.

The Machangqing Cu–Mo (Au) deposit is located in the central part of the Jinshajiang – Red River belt in the Sanjiang orogen, which lies across the Qiangtang terrane and western Yangtze craton, southwestern China. Zircon U–Pb dating constrains that the granite porphyry and porphyritic granite emplacements occurred at 35.92 ± 0.31 Ma and 34.92 ± 0.31 Ma, respectively. The Re–Os model ages of molybdenite are 34.94 ± 0.38 Ma. The new ages presented here, along with previously published data in the region, define a short duration of potassic magmatism and mineralization from 37 Ma to 34 Ma in the Jinshajiang – Red River belt. Zircon Ce4+/Ce3+ values of the porphyritic granite and granite porphyry vary from 50.32 to 1579.20 (averaging 481.01) and 33.18 to 1511.80 (averaging 452.98), respectively, and the log(fo2) values vary from –6.66 to −23.86 and −9.88 to −25.18, respectively, which plot within the range of the fayalite–magnetite–quartz buffer curve to the magnetite–hematite buffer curve, indicating an oxidized magma source, which may have facilitated the Cu–Au enrichment. Zircons from granitoids show εHf(t) values ranging from −0.75 to +2.33 and crustal model ages between 0.9 and 1.1 Ga. The features of Lu–Hf isotopes and wide range of Mg#, Cr, and Ni contents imply that the magmas of the Machangqing granitoids were probably derived from partial melting of juvenile lower crust and mixed with some mantle melts. Combined with the features of the Machangqing granitoids, the following evolution process are concluded. During the Cenozoic, the India–Asia continental collision triggered upwelling of hot asthenosphere and underplating of the thickened juvenile lower crust, which caused the formation of mafic and felsic magmas. Those magmas ascended, mixed, crystallized, and formed Machangqing ore-bearing granitoids in an intracontinental extension setting.

El americanismo vertical establece un orden jerarquizado que asegura el poder de una clase blanca dominante en una sociedad homogénea racial y culturalmente. La línea del color (en términos de Dubois) deviene en un límite cuya actual tensión máxima separa estos dos ejes, desfigurando culturalmente a la nación.

UU.) del siglo XXI, se plantean las mismas preguntas cuando el presidente Trump promete construir un muro «grande, grande», físico e impenetrable, en la frontera entre EE. UU. y México. El muro de Trump ha sido diseñado para detener a los inmigrantes ilegales mexicanos que ingresan a EE. UU., con el fin de proteger los intereses de los estadounidenses, que el idioma inglés no sea «contaminado» y sus valores no sean cuestionados. El muro de Trump se establece para delinear lo que son los EE. UU. y, por lo tanto, intenta «hacer que EE. UU. vuelva a ser grande». The Machangqing Cu-Mo-(Au) deposit in the central of Jinshajiang–Red River fold belt is a Cenozoic porphyry-skarn deposit in the western Yangtze Craton, southwestern China. Igneous rocks at Machangqing mainly consist of mafic and felsic rocks, including mafic dykes and barren syenite porphyry, as well as mineralized monzonite and granite porphyries with mafic microgranular enclaves (MMEs) in the ore-bearing porphyries. In this study, we compiled new and published geochemical and isotopic data for these intrusions, their MMEs and ore sulfides from the Machangqing deposit. Both mineralized and barren felsic porphyries, MMEs and mafic dyke were formed during the late Eocene (33–37 Ma), and have shoshonitic affinities (K2O + Na2O > 6 wt%, K2O/Na2O > 1 and Sr/Y > 40) ratios. They are enriched in LILE and LREE, but depleted in HFSE. Both mineralized and barren felsic porphyries are characterized by high SiO2 (64.67–72.81 wt%) and Al2O3 (13.43–16.10 wt%), but low Cr (average 37.63 ppm) and Ni (average 24.64 ppm) contents and low Mg# values (11–66). They have low whole–rock eNd(t) (–6.5 to –3.3) and relatively high (87Sr/86Sr)i (0.7061–0.7076), which fall inside the range of coeval mafic rocks in western Yunnan. The geochronological, geochemical and isotopic evidence suggest that the felsic magmas were formed by partial melting of the juvenile lower crust that had been injected by shoshonitic mafic magma as documented by the MMEs. The mafic dykes have high Mg# (71–76), Cr (average 492 ppm), Ni (average 195 ppm) contents, and were likely derived from partial melting of upper mantle sources metasomatized by paleo-subduction processes. The MMEs at Machangqing were produced by the mingling of metasomatized mantle-derived mafic melts with the juvenile Neoproterozoic thickened crust-derived felsic melts. The MMEs have spheroidal shapes and contain acicular apatite, indicating that the MMEs represent globules of coeval mafic melt injected into and partly assimilated by the felsic magmas. The ore sulfides have similar trace elemental and Pb isotopic features (206Pb/204Pb = 18.562–18.659; 207Pb/204Pb = 15.628–15.664; 208Pb/204Pb = 38.894–38.986) to the coeval mafic dykes, and their S-isotopic compositions (-2.24 to + 3.5‰) resemble typical mantle-source sulfides, suggesting a metallogenetic link with the coeval mafic magmas/melts and MMEs. Magmas of both the ore-bearing and barren porphyries are relatively oxidized in the FMQ to MH range (zircon Ce4+/Ce3+ = 87–950, log(fO2) = –22 to −5), although the ore-bearing ones are more oxidized. Petrological modelling suggests that the mineralized porphyries are derived from magma mixing dominated by partial melts of metasomatically-enriched mafic mantle, while the barren porphyries are products of magma mixing dominated by a juvenile lower crust component. We propose that magma mixing between lower crustal material with injected mafic mantle-derived magma has provided sufficient metals, sulfur and water to form the Machangqing Cu-Mo-(Au) deposit. The hydrous mafic magmas may have also remobilized the residual sulfides remaining in the continental lithospheric mantle (CLM), thus adding to the metal budget of these fertile magmas.

Ethnopharmacological relevance Cinnabar, a traditional Chinese mineral medicine with sedative and tranquilizing effects, is known to be toxic to the neural system, but its detailed pharmacological and toxicological mechanisms are still unclear. Aim of the study This study aimed to explore the potential neuropharmacological and neurotoxicological mechanisms of cinnabar by investigating the differentially expressed proteins in cerebral cortices of mice exposed to therapeutic and toxic doses of cinnabar. Materials and methods Label-free quantitative proteomics and bioinformatics analysis were used to characterize the proteins, pathways, and potential targets associated with therapeutic (50 mg/kg) and toxic (1000 mg/kg) doses of cinnabar in cerebral cortices of mice. Proteomic analysis was verified by parallel reaction monitoring. Results A total of 6370 and 6299 proteins were identified in the cerebral cortices of mice after exposure to therapeutic and toxic doses of cinnabar, among which 130 and 119 proteins were differentially expressed, respectively. Functional/pathway enrichment analysis showed that both exposure doses of cinnabar could affect transport processes in the cerebral cortex through different proteins. The changes induced by the therapeutic dose included pathways involved in translation and sphingolipid metabolism. Interestingly, for the toxic dose, differentially expressed proteins were enriched for functions and pathways related to RNA splicing, transcription, synaptic plasticity regulation and developmental processes, among which RNA splicing was the most significantly affected function. ATP6V1D and CX3CL1 were shown to be possible key proteins affected by cinnabar, leading to multiple functional changes in the cerebral cortex at the therapeutic and toxic doses, respectively. Furthermore, Connectivity Map (CMap) analysis predicted LRRK2 to be a potential therapeutic target and FTase to be a potential toxic target for cinnabar. Conclusion Our results suggest that the pathways and potential targets identified in the mouse cerebral cortex exposed to therapeutic and toxic doses of cinnabar are different, which provides novel insights into the potential molecular mechanisms underlying the pharmacological and toxicological effects of cinnabar.

The current cultural turmoil in the U.S. is much associated with Trumpism. This article asserts that Trumpism has its ideological root in nativism, a cultural tradition perpetuated by John Higham. To point out the disfigured Americanism in Trumpism, this paper theorizes the vertical America and the horizontal America, and dissects their complex interplay and symbiosis in an evolving American culture. The vertical Americanism sets hierarchies to secure power and control as well as maintain racial and cultural “purity;” the horizontal Americanism, epitomized by Martin Luther King and Barack Obama, equalizes and connects across races, creeds, and social classes. The color line (in Dubois’ term) thus becomes a converging and diverging boundary between the two Americas; it harbors and delineates Trumpism. It is argued that disfigurement of Americanism comes from a disruptive and destructive interplay of the vertical and the horizontal in Trump’s America.

Can a border wall really deter invaders and crossers alike, while defending, unifying, and maintaining the “purity” of the culture that it encircles? The first Chinese emperor Qin Shihuang (259-210 BC) and his successors in the following dynasties tried to achieve the purpose; the Great Wall of China testifies timelessly to such epic efforts. Have the Chinese rulers succeeded? The question has become increasingly daunting and unanswerable as history unfolds and evolves. Ironically, over more than two millennia, the piecemeal erection of the Great Wall has indeed sequenced a ‘wall-DNA’ in a cultural blueprint. Fast forwarding to the 21st century America, we find ourselves asking the same questions when President Trump vows to build a “great, great” wall, physical and impenetrable on the US-Mexico border. The Trump Wall is designed to stop Mexican illegal immigrants entering the US so that Americans’ interests will be protected, English language will not be “contaminated”, and American values will not be challenged. The Trump wall sets out to delineate what America is and thus attempts to “make America great again”. In an intercultural engagement, this article decodes a wall-DNA in American culture, argues that the Trump wll is more of a mental construct than a physical one, and calls for a redefinition of a 21st century American cultural identity, in front of the Trump Wall.

Alkaline phytase isolated from pollen grains of Lilium longiflorum (LlALP) possesses unique catalytic and thermal stability properties that suggest it has the potential to be used as a feed supplement. However, substantial amounts of active enzymes are needed for animal feed studies and endogenous levels of LlALP in lily pollen are too low to provide the required amounts. Active rLlALP2 (coded by LlAlp2 , one of two isoforms of alkaline phytase cDNA identified in lily pollen) has been successfully expressed in intracellular compartments of Pichia pastoris , however enzyme yields have been modest (25–30 mg/L) and purification of the enzyme has been challenging. Expression of foreign proteins to the extracellular medium of P. pastoris greatly simplifies protein purification because low levels of endogenous proteins are secreted by the yeast. In this paper, we first describe the generation of P. pastoris strains that will secrete rLlALP2 to the extracellular medium. Data presented here indicates that deletion of native signal peptides at the N- and C-termini of rLlALP2 enhanced α-mating factor (α-MF)-driven secretion by four-fold; chicken egg white lysozyme signal peptide was ineffective in the extracellular secretion of rLlALP2. Second, we describe our efforts to increase expression levels by employing a constitutive promoter from the glyceraldehyde-3-phosphate dehydrogenase gene (P GAP ) in place of the strong, tightly controlled promoter of alcohol oxidase 1 gene (P AOX1) . P GAP enhanced the extracellular expression levels of rLlALP2 compared to P AOX1 . Finally, we report on the optimization of the culture medium to enhance yields of rLlALP2. The strength of P GAP varies depending on the carbon source available for cell growth; secreted expression of rLlALP2 was highest when glycerol was the carbon source. The addition of histidine and Triton X-100 also enhanced extracellular expression. Taken together, the employment of P GAP under optimized culture conditions resulted in approximately eight-fold (75–80 mg/L) increase in extracellular activity compared to P AOXI (8–10 mg/L). The P. pastoris expression system can be employed as a source of active alkaline phytase for animal feed studies.

A low-cost, fully automated, electromagnetic field probing robot is described in this paper. The platform, called OpenEM Field Robot, is capable of mapping the spatial electromagnetic fields in free space. The system supports multiple antenna configurations and is scalable to large measurement domains. 3D printed antenna holders are also incorporated in order to allow for the near field measurements with minimal scattering and interference with the measurement. The goal of the system is to design a fully integrated platform for measuring electromagnetic fields that can be used with various other RF measurement devices and is configurable and easily modified for various applications in scientific research and education. Using the microindentation test, the stress-assisted-electrochemical corrosion of Cu46.25Zr45.25Al7.5Er1 bulk metallic glass (BMG) was studied in a 10 wt% NaCl electrolyte. The microindentation was performed in an indentation load range of 500 to 4000 mN to create shear bands over the deformation zone. Electric current of various densities was passed through the indented BMGs to evaluate the effect of shear bands and localized deformation on the electrochemical corrosion of the BMGs. Surface pits always initiated from the shear-banding zone and the contact edges between the indenter and the BMGs, and the size of the corroded zone grew with the increase in the polarization time, the indentation load, and the current density. Wormlike amorphous whiskers were formed over the corroded zone, and the density of the wormlike whiskers increased with the current density and polarization time.

As the mainstay in the treatment of various cancers, chemotherapy plays a vital role, but still faces many challenges, such as poor tumour selectivity and multidrug resistance (MDR). Targeted drug delivery using nanotechnology has provided a new strategy for addressing the limitations of the conventional chemotherapy. In the last decade, the volume of research published in this area has increased tremendously, especially with functional nano drug delivery systems (nanocarriers). Coupling a specific stimuli-triggered drug release mechanism with these delivery systems is one of the most prevalent approaches for improving therapeutic outcomes. Among the various stimuli, pH triggered delivery is regarded as the most general strategy, targeting the acidic extracellular microenvironment and intracellular organelles of solid tumours. In this review, we discuss recent advances in the development of pH-sensitive nanocarriers for tumour-targeted drug delivery. The review focuses on the chemical design of pH-sensitive biomaterials, which are used to fabricate nanocarriers for extracellular and/or intracellular tumour site-specific drug release. The pH-responsive biomaterials bring forth conformational changes in these nanocarriers through various mechanisms such as protonation, charge reversal or cleavage of a chemical bond, facilitating tumour specific cell uptake or drug release. A greater understanding of these mechanisms will help to design more efficient drug delivery systems to address the challenges encountered in conventional chemotherapy.

To investigate the arsenic levels in endemic arsenism in Datong City, Shanxi Province.A total of 85 inhabitants from one village in endemic arsenism area in Datong City, Shanxi Province were collected as research subjects. The People's Republic of China health industry standard for endemic arsenism was used to identify and diagnosis the patients. Daily drinking water and soil were collected and detected by atomic fluorescence spectrometry. The content of vegetables were detected by inductively coupled plasma mass spectrometry (ICP-MS).In the study, 85 samples were collected. Arsenic concentration in the daily drinking water were 14.41 - 90.34 μg/L, and the median value was 43.88 μg/L. The arsenic concentration of vegetables were 0.001 - 0.771 mg/kg, and 43.04% of samples, were higher than the maximal permissible limit of As in food. The results that the arsenic concentration of vegetables constant changes in the leaf vegetablestubersfruit vegetables. The health risk of intaking arsenic pollution in vegetables up to 71.77%. The arsenic levels in village of four directions were not exceeded the Chinese standards.Arsenic concentration in drinking water and vegetables are high in waterborn endemic arsenicosis area of Shanxi province. Arsenic in drinking water has been considered as a primary cause of arsenism, but direct intake of arsenic from vegetables can not be ignored.

There is increasing evidence that oxidative stress is an important risk factor for arsenic-related diseases. Peripheral blood leukocytes constitute an important defense against microorganisms or pathogens, while the research on the impact of chronic arsenic exposure on peripheral blood leukocytes is much more limited, especially at low level arsenic exposure. The purpose of the present study was to explore whether chronic arsenic exposure affects oxidative stress of peripheral blood leukocytes and possible linkages between oxidative stress and arsenic-induced skin lesions. 75 male inhabitants recruited from an As-endemic region of China were investigated in the present study. The classification of arsenicosis was based on the degree of skin lesions. Arsenic levels were measured in drinking water and urine by Atomic Fluorescence Spectroscopy. Urinary 8-hydroxy-2'-deoxyguanosine (8-OHdG) was tested by Enzyme-Linked Immunosorbent Assay. 8-OHdG of peripheral blood leukocytes was evaluated using immunocytochemical staining. 8-OHdG-positive reactions were only present in polymorphonuclear leukocytes (PMNs), but not in monocytes (MNs). The 8-OHdG staining of PMN cytoplasm was observed in all investigated populations, while the 8-OHdG staining of PMN nuclei was frequently found along with the elevated amounts of cell debris in individuals with skin lesion. Urinary arsenic levels were increased in the severe skin lesion group compared with the normal group. No relationship was observed between drinking water arsenic or urine 8-OHdG and the degree of skin lesions. These findings indicated that the target and persistent oxidative stress in peripheral blood PMNs may be employed as a sensitive biomarker directly to assess adverse health effects caused by chronic exposure to lower levels of arsenic.

Supplementation of animal feed with phytases has proven to be an effective strategy to alleviate phosphorous contamination of soil and water bodies. The inability of non-ruminant animals to digest phytates in corn and soybeans contributes to environmental contamination. Alkaline phytase from lily pollen (LlALP) exhibits unique catalytic and thermal stability properties that could be useful as a feed supplement. rLlALP2 was successfully expressed in Pichia pastoris ; however, enzyme yields were modest (8–10 mg/L). In this paper, we describe our efforts to enhance rLlALP2 yield by investigating the influence of the following potential limiting factors: transgene copy number, codon bias, sequence optimization, and temperature during expression. Data presented indicate that increasing rLlAlp2 copy number was detrimental to heterologous expression, clones with one copy of wt - rLlAlp2 produced the highest activity, clones with two, four and seven or more copies produced 70%, 25% and 10% respectively, of enzyme activity implying that gene dosage is not limiting rLlALP2 yield. Use of a sequence-optimized rLlAlp2 increased the yield of the active enzyme by 25–50% in one/two copy clones, suggesting that translational efficiency is not a major bottleneck for rLlALP2 expression. Reducing the temperature during heterologous expression led to increases of 1.2–20-fold suggesting that protein folding and post-translational processes may be the dominant factors limiting rLlALP2 expression. Early knowledge of the transgene copy number allowed us to develop a more rational strategy for yield enhancement. Cumulatively, sequence optimization and temperature reduction led to the doubling of rLlALP2 enzyme activity in P. pastoris .

Phytases catalyze the sequential hydrolysis of phytic acid (myo-insositol hexakisphosphate), the most abundant inositol phosphate in cells. Phytic acid constitutes 3-5% of the dry weight of cereal grains and legumes such as corn and soybean. The high concentration of phytates in animal feed and the inability of non-ruminant animals such as swine and poultry to digest phytates leads to phosphate contamination of soil and water bodies. The supplementation of animal feed with phytases results in increased bioavailability to animals and decreased environmental contamination. Therefore, phytases are of great commercial importance. Phytases with a range of properties are needed to address the specific digestive needs of different animals. Alkaline phytase (LlALP1 and LlALP2) which possess unique catalytic properties that have the potential to be useful as feed and food supplement has been identified in lily pollen. Substantial quantities of alkaline phytase are needed for animal feed studies. In this paper, we report the heterologous expression of LlALP2 from lily pollen in Pichia pastoris. The expression of recombinant LlALP2 (rLlALP2) was optimized by varying the cDNA coding for LlALP2, host strain and growth conditions. The catalytic properties of recombinant LlALP2 were investigated extensively (substrate specificity, pH- and temperature dependence, and the effect of Ca(2+), EDTA and inhibitors) and found to be very similar to that of the native LlALP2 indicating that rLlALP2 from P. pastoris can serve as a potential source for structural and animal feed studies.
