Background and Aim: To address the problems associated with the availability and prices of conventional feeds, researchers have started to include alternative feeds to reduce the cost of diets and increase profitability. This study examined the influences of olive cake (OC), either alone or in combination with Saccharomyces cerevisiae (SC), in the diet of black kids. Materials and Methods: Thirty kids were distributed into three treatments: A free OC diet (control [CON]), OC (20% OC), and OCSC (20% OC with 1 g SC head/d). While penned individually, the kids were fed daily and allowed to acclimate to their diets and pens for 7 days. After acclimation, the kids were fed the prescribed diets for 63 days. Five kids were chosen from each group on day 50 of the feeding period and moved to the metabolic cages to evaluate nutrient digestibility and N balance. At the end of the study, all kids were slaughtered to evaluate carcass characteristics and meat quality. Results: Feeding the OCSC diet increased (p = 0.035) DM intake for kids compared with the OC and CON. The CP intake was higher (p = 0.021) in the OCSC group than in the CON group. Growth performance parameters were higher (p ≤ 0.031) in the OCSC group than in the CON diet. Feed conversion efficiency was improved (p = 0.052) by incorporating OCSC compared with the CON diet. Consequently, economic return was enhanced (p = 0.003) by feeding the OC and OCSC groups. Kids fed the OCSC had a higher cold carcass weight (p = 0.054) than those fed the CON diet. The OCSC group also showed increased (p ≤ 0.027) loin cut and intermuscular fat weights compared with the OC and CON groups. All measured serum metabolites were similar in CON and groups incorporating OCs or OCSCs. Conclusion: The results showed that adding 20% OC combined with SC to the diet of growing kids improved some growth and carcass characteristic parameters without affecting their health status. Notably, using OC along with SC could be an option for feed cost reduction in kids’ diets.

Background and Aim: Feeding by-products, such as pomegranate seed pulp (PSP) and coconut meal (COC), to livestock may enhance production efficiency and increase profits. This study aimed to evaluate the effects of PSP and COC on milk production, body weight change, metabolic response (Exp. 1), digestibility, and N balance (Exp. 2). Materials and Methods: Twenty-four ewes nursing single lambs were randomly assigned to one of three diets: Control (CON) (n = 8), 7.5% PSP (n = 8), and 7.5% COC (n = 8) of dry matter (DM). Every sheep was born 3–4 days before the start of the experiment. The 1st week of the experiment was devoted to diet adaptation, while the data were collected during the following 8 weeks (Exp. 1). Nine lambs were randomly assigned to one of the three diets for intensive data collection to evaluate the diet digestibility and animal nitrogen (N) balance (Exp. 2). The data were analyzed using the MIXED SAS procedures. Results: The PSP group consumed more DM, followed by the COC and CON groups. For lambs, the final BW, total gain, and average daily gain in the PSP group were greater (p < 0.05) than those in the COC and CON groups. The CON group had lower milk output, total solids, protein, fat, and lactose levels than the PSP and COC groups (p < 0.05). The cost of milk production was lower (p < 0.05) in the PSP and COC diet groups than in the CON group. Blood serum parameters were similar among the dietary groups, except for total cholesterol and high-density lipoprotein, which were higher in the COC group than in the CON group. No differences were observed in nutrient digestibility and N balance. Conclusion: Feeding PSP and COC to nursing ewes appears beneficial because it increases milk production and pre-weaning lamb growth and reduces milk production cost.

Abstract Background Orchids are grown without soil in many regions of the world, but there is a lack of studies to define the balanced and adequate nutrient solution for their cultivation, mainly in the vegetative growth phase. Therefore, this paper aims to evaluate the optimal concentration of the nutrient solution based on the proposal by Hoagland and Arnon (1950) in the vegetative growth phase capable of increasing the nutrient contents, growth, and dry matter production of Dendrobium Tubtim Siam and Phalaenopsis Taisuco Swan. In addition, this paper aims to estimate a new nutrient solution from the optimal nutrient contents in the dry matter of these orchid species to be used in the vegetative growth phase. Results Nutrient contents, growth, and dry matter production increased as the nutrient solution concentration increased up to an average concentration of 62 and 77% for D. Tubtim Siam and P. Taisuco Swan, respectively. We found that the Hoagland and Arnon solution presented a group of nutrients with concentrations above the requirement for P. Taisuco Swan (nitrogen, phosphor, calcium, and sulfur) and D. Tubtim Siam (phosphor, calcium, magnesium, and sulfur), while other nutrients in the solution did not meet the nutritional demand of these orchid species, inducing nutritional imbalance in the vegetative growth phase. Conclusion We conclude that using a balanced nutrient solution created specifically for each orchid species in vegetative growth might favor their sustainable cultivation by optimizing the use of nutrients in the growing medium.

The importance of using agro-industrial by-products in feeding livestock is important in terms of preserve the environment and conversely, reducing the cost of feeding and production. Therefore, the purpose of this study was to assess how feeding jojoba meal (JOJ) affected growth performance, cost of production, and carcass features of growing lambs. Twenty-four Awassi lambs weighing 16.4 kg ± 0.17 kg were randomly selected and given two diets; a control diet without the inclusion of JOJ (CON), and JOJ100 with the presence of 100 g/kg dry matter (DM) of JOJ. The experiment lasted for 63 days preceded by 10 days as a period of adaptation. As scheduled on day 49 of the trial and, to assess nutritional digestibility and N balance, five lambs were randomly selected. During this period, lambs were housed in metabolic cages that allowed the collection of total fecal and urinary outputs. Lambs were slaughtered to investigate carcass features on day 64. Acid detergent fiber and EE intakes were greater (p ≤ 0.007) in the JOJ100 than in CON diet. Nutrient digestibility, N balance, growth rate, and carcass traits were not affected (p ≥ 0.136) by consumption of JOJ100. Cost of production declined (p = 0.046) by 17% with the JOJ100 group. High-density lipoprotein was increased (p = 0.012) with lambs consuming the JOJ100 diet. In summary, adding jojoba meal to the diet of lambs appeared to have no adverse effect on their performance, carcass quality, or health. Most importantly, this dietary supplement appeared to enhance the economic efficiency by reducing the cost of production.

Introduction: Targeted single nucleotide polymorphisms (SNPs) have been used in genomic prediction methodologies to enhance the accuracy of associated genetic transmitting abilities in Holstein cows. The objective of this study was to identify and validate SNPs associated with fertility traits impacting early embryo mortality.Methods: The mRNA sequencing data from day 16 normal (n = 9) and embryo mortality (n = 6) conceptuses from lactating multiparous Holstein cows were used to detect SNPs. The selection of specific genes with SNPs as preliminary candidates was based on associations with reproductive and fertility traits. Validation of candidate SNPs and genotype-to-phenotype analyses were conducted in a separate cohort of lactating primiparous Holstein cows (n = 500). After genotyping, candidate SNPs were filtered using a quality control pipeline via PLINK software. Continuous numeric and binary models from reproductive traits were evaluated using the mixed procedure for a generalized linear model-one way ANOVA or logistic regression, respectively.Results: Sixty-nine candidate SNPs were initially identified, but only 23 passed quality control procedures. Abstract Oligodendrocyte (OL) injury and subsequent loss is a pathologic hallmark of multiple sclerosis (MS). Stress granules (SGs) are membrane-less organelles containing mRNAs stalled in translation and considered as participants of the cellular response to stress. Here we show SGs in OLs in active and inactive areas of MS lesions as well as in normal-appearing white matter. In cultures of primary human adult brain derived OLs, metabolic stress conditions induce transient SG formation in these cells. Combining pro-inflammatory cytokines, which alone do not induce SG formation, with metabolic stress results in persistence of SGs. Unlike sodium arsenite, metabolic stress induced SG formation is not blocked by the integrated stress response inhibitor. Glycolytic inhibition also induces persistent SGs indicating the dependence of SG formation and disassembly on the energetic glycolytic properties of human OLs. We conclude that SG persistence in OLs in MS reflects their response to a combination of metabolic stress and pro-inflammatory conditions.

AbstractThe goal was to assess the influence of feeding olive leaves (OL) as a roughage source on nutrient intake, nutrient digestibility, N balance, growth performance and carcass characteristics of growing Awassi lambs. Twenty-four male lambs (initial body weight (BW) = 16.82 ± 0.998 kg; age = 70 ± 1.25 days) were randomly assigned to two isonitrogenous diets; OL at 0 (CON) or 150 (OL150) g/kg of dietary dry matter (DM). Diets were fed ad libitum for a period of 60 days after 7 days of adaptation. Feed intake and refusals were recorded daily. Body weights were recorded at the beginning of the study and at biweekly intervals thereafter. Intake of DM and crude protein was similar (P ≥ 0.05) between the two diets. Feeding OL decreased (P ≤ 0.01) intake of neutral detergent fiber and acid detergent fiber whereas it increased (P = 0.001) intake of ether extract compared with the CON diet. No differences (P ≥ 0.05) were observed in the final BW, average daily gain, and total gain between diets. However, the cost of gain was lower (P = 0.05) in lambs fed the OL diet versus the CON diet. None of the carcass characteristics nor meat quality parameters differed between the two diets. In summary, feeding olive leaves at 150 g/kg did not influence growth performance and carcass characteristics and meat quality but improved the economic value of using such products in diets of growing lambs. In addition, the use of this product has a positive impact on reducing environmental pollution.

Abstract Background In many regions of the world, K is being depleted from soils due to agricultural intensification a lack of accessibility, and the high cost of K. Thus, there is an urgent need for a sustainable strategy for crops in this environment. Si is an option for mitigating stress due to nutritional deficiency. However, the underlying effects of Si in mitigating K deficiency C:N:P homeostasis still remains unknown for bean plants. This is a species of great worldwide importance. Thus, this study aims to evaluate whether i) K deficiency modifies the homeostatic balance of C, N and P, and, if so, ii) Si supply can reduce damage caused to nutritional stoichiometry, nutrient use efficiency, and production of dry mass in bean plants. Results K deficiency caused a reduction in the stoichiometric ratios C:N, C:P, and P:Si in shoots and C:N, C:P, C:Si, N:Si, and P:Si in roots, resulting in a decrease in K content and use efficiency and reducing biomass production. The application of Si in K-deficient plants modified the ratios C:N, C:Si, N:P, N:Si, and P:Si in shoots and C:N, C:P, C:Si, N:Si, N:P, and P:Si in roots, increasing the K content and efficiency, reducing the loss of biomass. In bean plants with K sufficiency, Si also changed the stoichiometric ratios C:N, C:P, C:Si, N:P, N:Si, and P:Si in shoots and C:N, C:Si, N:Si, and P:Si in roots, increasing K content only in roots and the use efficiency of C and P in shoots and C, N, and P in roots, increasing the biomass production only in roots. Conclusion K deficiency causes damage to the C:N:P homeostatic balance, reducing the efficiency of nutrient use and biomass production. However, Si is a viable alternative to attenuate these nutritional damages, favoring bean growth. The future perspective is that the use of Si in agriculture in underdeveloped economies with restrictions on the use of K will constitute a sustainable strategy to increase food security.

Abstract Background C:N:P homeostasis in plants guarantees optimal levels of these nutrients in plant metabolism. H However, one of the causes to the effects of deficit irrigation is the loss of C:N:P homeostasis in leaves and stems that causes reduction in the growth of sugarcane. Being able to measure the impact of water deficit on C:N:P homeostasis in plants from the stoichiometric ratios of the concentrations of these nutrients in leaves and stems. This loss causes a decrease in nutritional efficiency, but can be mitigated with the use of silicon. Silicon favors the homeostasis of these nutrients and crop productivity. The magnitude of this benefit depends on the absorption of Si by the plant and Si availability in the soil, which varies with the type of soil used. Thus, this study aims to evaluate whether the application of Si via fertigation is efficient in increasing the absorption of Si and whether it is capable of modifying the homeostatic balance of C:N:P of the plant, causing an increase in nutritional efficiency and consequently in the production of biomass in leaves and stems of sugarcane ratoon cultivated with deficient and adequate irrigations in different tropical soils. Results Water deficit caused biological losses in concentrations and accumulation of C, N, and P, and reduced the nutrient use efficiency and biomass production of sugarcane plants cultivated in three tropical soils due to disturbances in the stoichiometric homeostasis of C:N:P. The application of Si increased the concentration and accumulation of Si, C, N, and P and their use efficiency and reduced the biological damage caused by water deficit due to the modification of homeostatic balance of C:N:P by ensuring sustainability of the production of sugarcane biomass in tropical soils. However, the intensity of attenuation of such deleterious effects stood out in plants cultivated in Eutrophic Red Oxisols. Si contributed biologically by improving the performance of sugarcane ratoon with an adequate irrigation due to the optimization of stoichiometric ratios of C:N:P; increased the accumulation and the use efficiency of C, N, and P, and promoted production gains in biomass of sugarcane in three tropical soils. Conclusion Our study shows that fertigation with Si can mitigate the deleterious effects of deficient irrigation or potentiate the beneficial effects using an adequate irrigation system due to the induction of a new stoichiometric homeostasis of C:N:P, which in turn improves the nutritional efficiency of sugarcane cultivated in tropical soils.

Anti-Müllerian hormone (AMH) is proposed as a biomarker for fertility in cattle, yet this associative relationship appears to be influenced by heat stress (HS). The objective was to test serum AMH and AMH-related single nucleotide polymorphisms (SNPs) as markers potentially predictive of reproductive traits in dairy cows experiencing HS. The study included 300 Holstein cows that were genotyped using BovineSNP50 (54,000 SNP). A genome-wide association study was then executed. Nine intragenic SNPs within the pathways that influence the AMH gene were found important with multiple comparisons adjustment tests (p < 1.09 × 10−6). A further validation study was performed in an independent Holstein cattle population, which was divided into moderate (MH; n = 152) and severe heat-stressed (SH; n = 128) groups and then subjected to a summer reproductive management program. Serum AMH was confirmed as a predictor of fertility measures (p < 0.05) in MH but not in the SH group. Cows were genotyped, which revealed four SNPs as predictive markers for serum AMH (p < 0.01), reproductive traits (p < 0.01), and additional physiological variables (p < 0.05). These SNPs were in the genes AMH, IGFBP1, LGR5, and TLR4. In conclusion, serum AMH concentrations and AMH polymorphisms are proposed as predictive markers that can be used in conjunction with genomic breeding value approaches to improve reproductive performance in Holstein cows exposed to summer HS conditions.

This study investigated the dietary effect of incorporating different levels of olive cake (OC) on the metabolic responses, growth performance, and carcass characteristics of black goat male kids. Thirty kids (body weight = 17.3 ± 0.40 kg) were randomly distributed into one of three equally sized dietary groups: a control diet (CON), OC at 75 g/kg (OC75), and OC at 150 g/kg (OC150) of dietary dry matter (DM). The results revealed that the intake of DM, crude protein (CP), neutral detergent fiber (NDF), and acid detergent fiber (ADF) was similar (p ≥ 0.11) among the three treatment groups. However, the OC150 group had the greatest (p < 0.0001) ether extract (EE) intake compared to the OC75 and CON groups. The growth measurements were similar (p ≥ 0.13) among the three groups. Feed efficiency was not affected by the inclusion of OC. In contrast to the CON diet, the cost of gain was, however, reduced (p = 0.004) in the OC diets. All three treatment groups’ digestibility of DM, CP, and ADF was similar. However, when compared to the OC75 and CON groups, the digestibility of NDF was better (p < 0.05) in the OC150 group. The N intake did not differ among the three experimental groups. Nitrogen retained as g/d was higher (p = 0.04) in the OC150 and OC75 groups compared to the CON group, while retention as a percentage (g/100 g) was similar among the three groups. Except for intermuscular fat, total fat, leg fat depth, and tissue depth, the inclusion of OC did not result in any discernible treatment effects on the carcass and meat quality parameters. Only alanine aminotransferase enzyme activity was lower (p < 0.0001) in OC-treated groups compared to the CON group. In summary, incorporating OC at 75 g/kg and 150 g/kg levels into the diets of black goat kids had positive comparable effects on some parameters related to growth performance, carcass attributes, and meat quality. Importantly, utilizing olive cake led to cost savings in production and may serve as a viable alternative feed source in goat nutrition.

Dairy production in Holstein cows in a semiarid environment is challenging due to heat stress. Under such conditions, genetic selection for heat tolerance appears to be a useful strategy. The objective was to validate molecular markers associated with milk production and thermotolerance traits in Holstein cows managed in a hot and humid environment. Lactating cows (n = 300) exposed to a heat stress environment were genotyped using a medium-density array including 53,218 SNPs. A genome-wide association study (GWAS) detected six SNPs associated with total milk yield (MY305) that surpassed multiple testing (p < 1.14 × 10−6). These SNPs were further validated in 216 Holstein cows from two independent populations that were genotyped using the TaqMan bi-allelic discrimination method and qPCR. In these cows, only the SNPs rs8193046, rs43410971, and rs382039214, within the genes TLR4, GRM8, and SMAD3, respectively, were associated (p < 0.05) with MY305, rectal temperature (RT), and respiratory rate. Interestingly, these variables improved as the number of favorable genotypes of the SNPs increased from 0 to 3. In addition, a regression analysis detected RT as a significant predictor (R2 = 0.362) for MY305 in cows with >1 favorable genotype, suggesting this close relationship was influenced by genetic markers. In conclusion, SNPs in the genes TLR4, GRM8, and SMAD3 appear to be involved in the molecular mechanism that regulates milk production in cows under heat-stressed conditions. These SNPs are proposed as thermotolerance genetic markers for a selection program to improve the milk performance of lactating Holstein cows managed in a semiarid environment.

The predictive abilities and accuracies of genomic best linear unbiased prediction (GBLUP) and the Bayesian (BayesA, BayesB, BayesC and Lasso) genomic selection (GS) methods for economically important growth (birth, weaning, and yearling weights) and carcass (depth of rib fat, apercent intramuscular fat and longissimus muscle area) traits were characterized by estimating the linkage disequilibrium (LD) structure in Brangus heifers using single nucleotide polymorphisms (SNP) markers. Sharp declines in LD were observed as distance among SNP markers increased. The application of the GBLUP and the Bayesian methods to obtain the GEBV for growth and carcass traits within k-means and random clusters showed that k-means and random clustering had quite similar heritability estimates, but the Bayesian methods resulted in the lower estimates of heritability between 0.06 and 0.21 for growth and carcass traits compared with those between 0.21 and 0.35 from the GBLUP methodologies. Although the prediction ability of the GBLUP and the Bayesian methods were quite similar for growth and carcass traits, the Bayesian methods overestimated the accuracies of GEBV because of the lower estimates of heritability of growth and carcass traits. However, GBLUP resulted in accuracy of GEBV for growth and carcass traits that parallels previous reports.

The porcine reproductive and respiratory syndrome (PRRS) is a viral disease that decreases the reproductive performance in breeding sows and leads to economic losses to the swine industry. The objective of the present study was to identify single nucleotide polymorphisms (SNP) associated to the number of live-born piglets in the first (LBP1) and second birth (LBP2) in breeding sows exposed to PRRS virus. The study included 100 pregnant females of the Landrace(¾)/ Yorkshire(¼) line, 75 of which were infected with the PRRS virus and 25 were free of PRRS. Individual blood samples (6-8 drops) were obtained and spotted onto FTA cards and subsequently processed for DNA extraction, which was genotyped using a 10,000 SNP chip for genomic profile. A simulation study was carried out to determine the genomic prediction performance of Artificial Neural Network model with 1 to 10 neurons (ANN-1-10) using 3361 SNP markers from BovineSNP50 (Infinium BeadChip, Illumina, San Diego, CA) on the first chromosome of Brangus beef cattle as a pilot study for two traits with heritabilities of 25% (h2T1= 0.25) and 50% (h2T2= 0.5) determined either by 50, 100, 250 or 500 QTL selected randomly from SNP markers. QTL effects were sampled from a multivariate normal distribution. Genomic predictions were carried out by Feed Forward Multi-Layer Perceptron ANN-1-10 with the back-propagation of errors algorithm employing the Levenberg-Marquardt algorithm to locate the optimal solution. Three sets of SNP panels were used for genomic prediction: only QTL (Panel-1), all SNP markers, including the QTL (Panel-2), and all SNP, excluding the QTL (Panel-3). Correlations between true genetic merits (breeding values) and predicted phenotypes from 10-fold disjoint cross-validation were used to assess predictive ability of ANN-1-10. Results indicated that an increase in heritability resulted in an increased predictive performance of ANN-1-10 for all scenarios. SNP Panels had a greater chance of including markers in LD with QTL, allowed the possibility of predicting the effect of each QTL from the collective action of several markers and performed better than the Panel including only QTL. In the other Panels, predictive performance of ANN-1-10 increased inconsistently with the number of neurons, which indicated that a few numbers of neurons were not be enough to learn specification of data and could cause the under fitting problem. Therefore, high number of neurons could be needed to learn relevant details of the data in the applications of ANN.

El presente artículo tiene como finalidad mostrar información completa, precisa y en tiempo real, sobre la movilidad de los usuarios en un campus, identificando tipos de usuarios y sus características, todo esto mediante una plataforma informática constituida por dos aplicaciones un API especializado en el almacenamiento de la data depurada hacia una base de datos relacional, y una aplicación web que presenta información al usuario final. Dicha plataforma se desarrolla mediante el uso de tecnologías y herramientas de código abierto. La información entregada procede de los Access Points instalados en la red de comunicaciones del campus, la cual, mediante tratamiento informático, es anonimizada, almacenada, depurada y clasificada para su posterior análisis. Como caso de estudio se ha implementado la aplicación en el contexto universitario específicamente en la Universidad Central del Ecuador.

Mechanisms implicated in disease progression in multiple sclerosis include continued oligodendrocyte (OL)/myelin injury and failure of myelin repair. Underlying causes include metabolic stress with resultant energy deficiency. Biotin is a cofactor for carboxylases involved in ATP production that impact myelin production by promoting fatty acid synthesis. Here, we investigate the effects of high dose Biotin (MD1003) on the functional properties of post-natal rat derived oligodendrocyte progenitor cells (OPCs). A2B5 positive OPCs were assessed using an in vitro injury assay, culturing cells in either DFM (DMEM/F12+N1) or "stress media" (no glucose (NG)-DMEM), with Biotin added over a range from 2.5 to 250 μg/ml, and cell viability determined after 24 hrs. Biotin reduced the increase in OPC cell death in the NG condition. In nanofiber myelination assays, biotin increased the percentage of ensheathing cells, the number of ensheathed segments per cell, and length of ensheathed segments. In dispersed cell culture, Biotin also significantly increased ATP production, assessed using a Seahorse bio-analyzer. For most assays, the positive effects of Biotin were observed at the higher end of the dose-response analysis. We conclude that Biotin, in vitro, protects OL lineage cells from metabolic injury, enhances myelin-like ensheathment, and is associated with increased ATP production.

The direction of individual B8 solar neutrinos has been reconstructed using the SNO+ liquid scintillator detector. Prompt, directional Cherenkov light was separated from the slower, isotropic scintillation light using time information, and a maximum likelihood method was used to reconstruct the direction of individual scattered electrons. A clear directional signal was observed, correlated with the solar angle. The observation was aided by a period of low primary fluor concentration that resulted in a slower scintillator decay time. This is the first time that event-by-event direction reconstruction in high light-yield liquid scintillator has been demonstrated in a large-scale detector.

The obesity epidemic in developed societies has led to increased cardiovascular diseases including pulmonary hypertension associated with left heart disease (PH-LHD), the largest and fastest-growing class of PH. Similar to obese humans, PH and heart failure (HF) are increasingly recognized in North American fattened beef cattle. We hypothesized that PH and HF in fattened beef cattle are novel, phenotypically distinct manifestations of bovine PH arising from left ventricular (LV) dysfunction similar to obesity-related PH-LHD in humans. We conducted a semi-quantitative histopathological assessment of cardiopulmonary tissues obtained from fattened beef cattle suffering end-stage HF compared to asymptomatic cattle of equivalent age undergoing the same fattening regimens. In HF animals we observed significant LV fibrosis, abundant cardiac adipose depots, coronary artery injury, and pulmonary venous remodeling recapitulating human obesity-related PH-LHD. Additionally, striking muscularization, medial hypertrophy, adventitial fibrosis, and vasa vasorum hyperplasia in the pulmonary arterial circulation were associated with sequela of pathologic right ventricular (RV) remodeling suggesting combined pulmonary venous and arterial hypertension. The association between obesity, pathologic cardiopulmonary remodeling, and HF in fattened beef cattle appears to recapitulate the complex pathophysiology of obesity-associated PH-LHD in humans. The identification of biological processes related to the regulation of complex traits is a difficult task. Commonly, complex traits are regulated through a multitude of genes contributing each to a small part of the total genetic variance. Additionally, some loci can simultaneously regulate several complex traits, a phenomenon defined as pleiotropy. The lack of understanding on the biological processes responsible for the regulation of these traits results in the decrease of selection efficiency and the selection of undesirable hitchhiking effects. The identification of pleiotropic key-regulator genes can assist in developing important tools for investigating biological processes underlying complex traits. A multi-breed and multi-OMICs approach was applied to study the pleiotropic effects of key-regulator genes using three independent beef cattle populations evaluated for fertility traits. A pleiotropic map for 32 traits related to growth, feed efficiency, carcass and meat quality, and reproduction was used to identify genes shared among the different populations and breeds in pleiotropic regions. Furthermore, data-mining analyses were performed using the Cattle QTL database (CattleQTLdb) to identify the QTL category annotated in the regions around the genes shared among breeds. This approach allowed the identification of a main gene network (composed of 38 genes) shared among breeds. This gene network was significantly associated with thyroid activity, among other biological processes, and displayed a high regulatory potential. In addition, it was possible to identify genes with pleiotropic effects related to crucial biological processes that regulate economically relevant traits associated with fertility, production and health, such as MYC, PPARG, GSK3B, TG and IYD genes. These genes will be further investigated to better understand the biological processes involved in the expression of complex traits and assist in the identification of functional variants associated with undesirable phenotypes, such as decreased fertility, poor feed efficiency and negative energetic balance.

Puberty is a complex physiological event by which animals mature into an adult capable of sexual reproduction. In order to enhance our understanding of the genes and regulatory pathways and networks involved in puberty, we characterized the transcriptome of five reproductive tissues (i.e. hypothalamus, pituitary gland, ovary, uterus, and endometrium) as well as tissues known to be relevant to growth and metabolism needed to achieve puberty (i.e., longissimus dorsi muscle, adipose, and liver). These tissues were collected from pre- and post-pubertal Brangus heifers (3/8 Brahman; Bos indicus x 5/8 Angus; Bos taurus) derived from a population of cattle used to identify quantitative trait loci associated with fertility traits (i.e., age of first observed corpus luteum (ACL), first service conception (FSC), and heifer pregnancy (HPG)). In order to exploit the power of complementary omics analyses, pre- and post-puberty co-expression gene networks were constructed by combining the results from genome-wide association studies (GWAS), RNA-Seq, and bovine transcription factors. Eight tissues among pre-pubertal and post-pubertal Brangus heifers revealed 1,515 differentially expressed and 943 tissue-specific genes within the 17,832 genes confirmed by RNA-Seq analysis. The hypothalamus experienced the most notable up-regulation of genes via puberty (i.e., 204 out of 275 genes). Combining the results of GWAS and RNA-Seq, we identified 25 loci containing a single nucleotide polymorphism (SNP) associated with ACL, FSC, and (or) HPG. Seventeen of these SNP were within a gene and 13 of the genes were expressed in uterus or endometrium. Multi-tissue omics analyses revealed 2,450 co-expressed genes relative to puberty. The pre-pubertal network had 372,861 connections whereas the post-pubertal network had 328,357 connections. A sub-network from this process revealed key transcriptional regulators (i.e., PITX2, FOXA1, DACH2, PROP1, SIX6, etc.). Results from these multi-tissue omics analyses improve understanding of the number of genes and their complex interactions for puberty in cattle.

Exosomes are membrane vesicles with a diameter of 40–100 nm that are secreted by many cell types into the extracellular milieu. Exosomes are found in cell culture supernatants and in different biological fluids and are known to be secreted by most cell types under normal and pathological conditions. Considerable research is focusing on the exploitation of exosomes in biological fluids for biomarkers in the diagnosis of disease. More recently, exosomes are being exploited for their therapeutic potential. Exosomes derived from dendritic cells, tumor cells, and malignant effusions demonstrate immunomodulatory functions and are able to present antigens to T-cells and stimulate antigen-specific T-cell responses. Exosomes have also been examined for their therapeutic potential in the treatment of infections such as toxoplasmosis, diphtheria, tuberculosis and atypical severe acute respiratory syndrome as well as autoimmune diseases. Attempts to find practical applications for exosomes continue to expand with the role of exosomes as a drug delivery system for the treatment of autoimmune/inflammatory diseases and cancers.

Sulfur mustard or mustard gas (HD) and its monofunctional analog, 2-chloroethyl ethyl sulfide (CEES), or “half-mustard gas,” are alkylating agents that induce DNA damage, oxidative stress, and inflammation. HD/CEES are rapidly absorbed in the skin causing extensive injury. We hypothesize that antioxidant liposomes that deliver both water-soluble and lipid-soluble antioxidants protect skin cells from immediate CEES-induced damage via attenuating oxidative stress. Liposomes containing water-soluble antioxidants and/or lipid-soluble antioxidants were evaluated using in vitro model systems. Initially, we found that liposomes containing encapsulated glutathione (GSH-liposomes) increased cell viability and attenuated production of reactive oxygen species (ROS) in HaCaT cells exposed to CEES. Next, GSH-liposomes were tested in a human epidermal model, EpiDerm. In the EpiDerm, GSH-liposomes administered simultaneously or 1 hour after CEES exposure (2.5 mM) increased cell viability, inhibited CEES-induced loss of ATP and attenuated changes in cellular morphology, but did not reduce caspase-3 activity. These findings paralleled the previously described in vivo protective effect of antioxidant liposomes in the rat lung and established the effectiveness of GSH-liposomes in a human epidermal model. This study provides a rationale for use of antioxidant liposomes against HD toxicity in the skin considering further verification in animal models exposed to HD.

"Agriscience" is an emerging field at the intersection of recently separate fields of "agriculture" and "science." For meaningful communication with and engagement of public audiences around agriscience, researchers, educators, and the public must have a consensus definition. We used personal meaning mapping to collect public audience understandings of the individual terms agriculture and science to find spontaneous overlap. We qualitatively coded them and compared them to each other. Very few participants explicitly included "agriculture" on science maps and vice versa. However, many maps included terms that related to the other topic; for example, on agriculture maps, many participants included "biology." Agriculture maps used more terms related to tangible products, while science maps contained more terms related to intangible results and specific disciplinary areas. We found some overlap of categories with both sets of standards but in differing amounts, reflecting the career emphasis of agricultural programs. The lack of consensus definitions of agriculture and science confound our efforts to support both public engagement with agriscience and literacy efforts in both science education and agricultural education. This research could form the basis for larger, broader quantitative public surveys and comparisons across geographic areas.

Equine maternal recognition of pregnancy (MRP) is a process whose signal remains unknown. During MRP the conceptus and endometrium communicate to attenuate prostaglandin F2α (PGF) secretion, sparing the corpus luteum and maintaining progesterone production. Recognition of a mobile conceptus by the endometrium is critical by days 14-16 post-ovulation (PO), when endometrium produces PGF, initiating luteolysis. The objective of this study was to evaluate endometrial gene expression changes based upon pregnancy status via RNA sequencing. This experiment utilized a cross-over design with each mare serving as both a pregnant and non-mated control on days nine, 11, and 13 PO (n = 3/status/day). Mares were randomly assigned to collection day and pregnancy confirmed by terminal uterine lavage at the time of endometrial biopsy. Total RNA was isolated and libraries prepared using Illumina TruSeq RNA sample preparation kit. Reads were mapped and annotated using HISAT2 and Stringtie. Expression values were evaluated with DESEQ2 (P ≤ 0.05 indicated significance). On day nine, 11, and 13 there were 1435, 1435 and 916 significant transcripts, respectively. Multiple genes with splice variants had different expression patterns within the same day. These are the first data to evaluate the endometrial transcriptome during MRP on days nine, 11, and 13.

This exploratory study examined the effects of naming animal ambassadors on participants' level of Empathy, Altruism, Connectedness to Nature, and Commitment to Engaging in Environmentally responsible Behaviors. Raptor educational presentations were used for this study. Two animal ambassadors were used in the experiment (one animal was given its common species name versus the other was named after its natural history). Observations, online questionnaires, and telephone interviews were used to assess the effects of implementing different named animal ambassadors in programing. Findings indicated no statistical significance between control and treatment groups, but found qualitative evidence signaling the effectiveness of naming animal ambassadors.
