Abstract

A second-generation dual purpose pulse forming network (DP-PFN) has been developed to power both the electro-thermal-chemical (ETC)/electro-thermal-ignition (ETI) lethality capability and the electro-magnetic armor (EMA) survivability capability improvements envisioned for future hybrid-electric vehicles. ETI decreases the ignition variability associated with the launching of conventional munitions by roughly a factor of ten which, in conjunction with the systems ballistic computer, greatly enhances hit probability of the round. ETC provides maximum performance in all conditions, including temperature compensation, resulting in increased lethality and range with higher average muzzle velocities. EMA uses stored electric energy to disrupt a shaped-charge jet and reduce it's depth of penetration. The second-generation DP-PFN for driving both ETI/ETC and EMA emulators is scheduled to be integrated into the power and energy (P&E) hardware-in-the-loop (HWIL) System Integration Laboratory (SIL) in Santa Clara, CA, in 2006. The DP-PFN is capable of providing either the short pulse lengths required by the EMA or the considerably longer pulse lengths required by the ETI/ETC gun as well as emulating various 'degraded functionality' states such as loss of individual capacitors. Integration of this DP-PFN marks another great milestone for the SIL, ensuring that it continues to be capable of emulating all the major hybrid-electric mobility functions of a ground combat vehicle as well as the major lethality and survivability electrical loads. This paper describes the DP-PFN components, the overall DP-PFN design philosophy and the planned integration and testing of the DP-PFN in the P&E HWIL SIL. Abstract

Marine algae perform approximately half of global carbon fixation, but their growth is often limited by the availability of phosphate or other nutrients 1,2 . As oceans warm, the area of phosphate-limited surface waters is predicted to increase, resulting in ocean desertification 3,4 . Understanding the responses of key eukaryotic phytoplankton to nutrient limitation is therefore critical 5,6 . We used advanced photo-bioreactors to investigate how the widespread marine green alga Micromonas commoda grows under transitions from replete nutrients to chronic phosphate limitation and subsequent relief, analysing photosystem changes and broad cellular responses using proteomics, transcriptomics and biophysical measurements. We find that physiological and protein expression responses previously attributed to stress are critical to supporting stable exponential growth when phosphate is limiting. Unexpectedly, the abundance of most proteins involved in light harvesting does not change, but an ancient light-harvesting-related protein, LHCSR, is induced and dissipates damaging excess absorbed light as heat throughout phosphate limitation. Concurrently, a suite of uncharacterized proteins with narrow phylogenetic distributions increase multifold. Notably, of the proteins that exhibit significant changes, 70 are not differentially expressed at the mRNA transcript level, highlighting the importance of post-transcriptional processes in microbial eukaryotes. Nevertheless, transcript-protein pairs with concordant changes were identified that will enable more robust interpretation of eukaryotic phytoplankton responses in the field from metatranscriptomic studies. Our results show that P-limited Micromonas responds quickly to a fresh pulse of phosphate by rapidly increasing replication, and that the protein network associated with this ability is composed of both conserved and phylogenetically recent proteome systems that promote dynamic phosphate homeostasis. Abstract

Some aspects, by a statistical approach, of the variation of the peak torque and mean cutting power encountered in gear hobbing are presented here. The cutting torque is measured by a purpose built strain gauge dynamometer and a wattmeter evaluates the power consumed. The problem is to determine whether a host of independent variables such as axial feed, module, material hardness, number of teeth in the gear blank and hobbing speed have a significant influence on the dependent variables, maximum cutting torque and average power. Mathematical models are constructed giving the relationship between these variables and the results indicate that the cutting speed has a significant influence on both the peak torque and average power. Abstract

The identification of poliovirus receptor-like 4 (PVRL4) as the second natural receptor for measles virus (MV) has closed a major gap in our understanding of measles pathogenesis, and explains how this predominantly lymphotropic virus breaks through epithelial barriers to transmit to a susceptible host. Advances in the development of wild-type, recombinant MVs which express fluorescent proteins making infected cells readily detectable in living tissues and animals, has also increased our understanding of this important and highly transmissible human disease. Thus, it is timely to review how these advances have provided new insights into MV infection of immune, epithelial and neural cells. This demands access to primate samples that help us understand the early and acute stages of the disease, which are challenging to dissect due to the mild/self-limiting nature of the infection. It also requires well-characterized and rather rare human tissue samples from patients who succumb to neurological sequelae to help study the consequences of the long-term persistence of this RNA virus in vivo. Collectively, these studies have provided unique insights into how the use of two cellular receptors, CD150 and PVRL4, governs the in vivo tissue-specific temporal patterns of virus spread and resulting pathological lesions. Analysis of tissue samples has also demonstrated the importance of differing mechanisms of virus cell-to-cell spread within lymphoid, epithelial and neural tissues in the dissemination of MV during acute and long-term persistent infections. Given the incentive to eradicate MV globally, and the inevitable question as to whether or not vaccination should cease in light of the existence of closely related morbilliviruses, a thorough understanding of measles pathological lesions is essential. Abstract

Background\ud \ud Malarial retinopathy (MR) has diagnostic and prognostic value in children with Plasmodium falciparum cerebral malaria (CM). A clinicopathological correlation between observed retinal changes during life and the degree of sequestration of parasitized red blood cells was investigated in ocular and cerebral vessels at autopsy.\ud \ud Methods\ud \ud In 18 Malawian children who died from clinically defined CM, we studied the intensity of sequestration and the maturity of sequestered parasites in the retina, in nonretinal ocular tissues, and in the brain.\ud \ud Results\ud \ud Five children with clinically defined CM during life had other causes of death identified at autopsy, no MR, and scanty intracerebral sequestration. Thirteen children had MR and died from CM. MR severity correlated with percentage of microvessels parasitized in the retina, brain, and nonretinal tissues with some neuroectodermal components (all P < .01). In moderate/severe MR cases (n = 8), vascular congestion was more intense (ρ = 0.841; P < .001), sequestered parasites were more mature, and the quantity of extraerythrocytic hemozoin was higher, compared with mild MR cases (n = 5).\ud \ud Conclusions\ud \ud These data provide a histopathological basis for the known correlation between degrees of retinopathy and cerebral dysfunction in CM. In addition to being a valuable tool for clinical diagnosis, retinal observations give important information about neurovascular pathophysiology in pediatric CM.
