Abstract

The organization of eukaryotic DNA is increasingly well understood at each extreme of chromatin organization. Advances in understanding the organization of DNA at the level of nucleosome and chromosome have outstripped knowledge of the intermediate states and function of DNA. Most would, however, agree on the following general hierarchy (1,2). The base-repeating unit of approx 146 bp, including a core histone group, is further organized into the 30-nm fiber and constrained by contact with the nuclear matrix to form loop and superloop structures capable of maintaining physical tension or supercoiling (3-5). These supercoiled units of DNA subsequently adopt repetitive patterns based around a protein core template, which together are observed as chromosomes. [ABSTRACT FROM AUTHOR]

Abstract

Apoptosis is a physiological, programmed mode of cell death, which is necessary for tissue modeling and organogenesis in embryonic development and in the control of homeostasis in a diversity of tissue types (1). The distinct morphological features of apoptosis clearly distinguish it from the passive mode of cell death, necrosis, which is an unprogrammed response to toxic stimuli. The diagnosis of apoptosis relies on detection of these morphological changes, i.e., condensation and fragmentation of chromatin, cell shrinkage associated with cytoplasmic condensation, and retention of cell membrane integrity. These changes may be accompanied by the fragmentation of the cell into membrane-bound apoptotic bodies containing cytoplasmic organelles, nuclear components, or both (2). Classical methods of diagnosing apoptosis include light and electron microscopy in which the above morphological changes can be visualized, and internucleosomal DNA fragmentation assays in which the fragmentation of DNA can be seen after gel electrophoresis as a ladder pattern representing the generation of multiple oligonucleosomal fragments. The widespread use of in vitro systems for studying cell death has led many researchers to turn to flow cytometry as an alternative to the classic methods of analyzing apoptosis, since it offers the advantages of rapid analysis of individual cells, low cell number requirement, and the opportunity for simultaneous measurement of several cellular parameters. Abstract

Tonsillar B lymphocytes of a particularly high buoyant density were prepared essentially free of contaminating monocytes and T cells. When exposed to anti-immunoglobulin, such cells initiated the hydrolysis of inositol phospholipids. This provides a postulated `dual signal' for growth through the liberation of intracellular calcium stores and the activation of protein kinase C. Nevertheless, neither anti-immunoglobulin nor direct agonists of this bifurcating pathway (respectively, calcium ionophore and the phorbol ester TPA) were capable, when used alone, of driving cells out of G0 and into RNA synthesis. All three agents did, however, induce two activation antigens at the surface of G0 B cells, which included CD23, p45 and a lineage-unrestricted antigen identified by the monoclonal antibody BK. 19.9. Cells that had been exposed to calcium ionophore, but not those activated with either TPA or anti-immunoglobulin, revealed further change indicated by an increased accessibility of their native DNA for the intercalating dye acridine orange. Cells receiving full mitogenic signals in the form of Staphylococcus aureus Cowan Strain I (SAC) or a combination of TPA and calcium ionophore showed the same initial sequelae but continued to enter the cell cycle and progress through to DNA synthesis. The observations identify two phases in the early activation of human B cells, both in terms of various temporal events, and the signals required to promote each activation state. Furthermore, cells receiving complete growth signals were required to transit these activation states before entering the proliferative cycle. 37. Prevention of hybridoma cell death by <TOGGLE>bcl-2</TOGGLE> during suboptimal culture conditions

Abstract

TB/C3 hybridoma cells were transected with either pEF-MClneopA or pEF bcl2-MClneopA vectors to produce a control cell line (TB/C3 pEF) and a cell line that overexpresses the “antiapoptotic” human bcl-2 protein (TB/C3 bcl2). Flow cytometry analysis of intracellular bcl-2 protein levels enabled near on-line monitoring of the stability of bcl-2 expression in the absence of drug selection. It was possible to maintain spontaneous selection of cells with the overexpression of bcl-2 protein during semicontinuous cultures at very low dilution rates, where cells were subjected to the selective conditions of nutrient limitation and high toxic metabolite concentrations. Interestingly, cells that overexpressed bcl-2 were adapted to suspension culture conditions significantly faster than control cells. Dual fluorescence staining with acridine orange and propidium iodide allowed for discrimination between viable, apoptotic, secondary necrotic, and necrotic cells, respectively. Compared with the usual trypan blue method of establishing culture viability, dual staining demonstrated that under stressful conditions a significant proportion of cells that excluded trypan blue were also undergoing cell death through apoptosis. In batch cultures the overexpression of bcl-2 more than doubled the membrane intact (MI) cell productive period (the integral of Ml cell density with respect to culture time) and increased the monoclonal antibody (mAb) production by approximately 40% when compared with the control cell line. The overexpression of bcl-2 protein also significantly extended the cell integrity and viability by the suppression of apoptosis in conditions of hypoxia, hyperoxia, glutamine deprivation, glucose deprivation, and serum limitation. The suppression of apoptosis in anaerobic conditions suggests that bcl-2 exerts its antiapoptotic activity by a mechanism that does not involve an oxidative reactive pathway. In conditions of excess thymidine, which suppressed cell proliferation, Ml cell density and specific mAb productivity were further enhanced by the overexpression of bcl-2, which suggests the possibility of accomplishing a controlled proliferation in immortalized cell lines without invoking cell death. Cell size and intracellular mAb were increased for TB/C3 bcl2 cells compared with TB/C3 pEF control cells when analyzed by flow cytometry. © 1997 John Wiley & Sons, Inc.
