Abstract

We report a method for preparing p-type silicon germanium bulk alloys directly from a boron-doped silica germania nanocomposite. This is the first successful attempt to produce and characterize the thermoelectric properties of SiGe-based thermoelectric materials prepared at temperatures below the alloy's melting point through a magnesiothermic reduction of the silica-germania nanocomposite. We observe a thermoelectric power factor that is competitive with the literature record obtained for high energy ball milled nanocomposites. The large grain size in our hot pressed samples limits the thermoelectric figure of merit to 0.5 at 800 C for an optimally doped Si80Ge20 alloy. © 2013 American Chemical Society.

Abstract

The function, structure, and mechanical properties of protein materials make them well-suited for a range of applications such as biosensors and biomaterials. Unlike in traditional polymer synthesis, their sequences are defined and, in the case of recombinant proteins, dictated by the chosen DNA sequence. As DNA synthesis has rapidly progressed over the past twenty years, the limiting bottleneck in protein materials development is the empirical optimization of protein expression. Herein, a low-cost, automated, high-throughput, combinatorial protein expression platform is developed to test permutations of DNA vectors and Escherichia coli (E. coli) strains in a 96-well plate format. Growth and expression are monitored with optical density at 600 nm (OD600) to measure growth, Bradford assays to establish the total protein concentration, and dot blot assays to determine the concentration of the protein of interest. With an eye toward accessibility for researchers without suites of biosynthetic equipment, automated camera-based assays are validated for the OD600 assay, via turbidimetry, and the Bradford assay, via colorimetry. High-yield expression conditions can be determined within a week. Notably, in several cases, previously un-expressible proteins are expressed successfully in viable yields. Collectively, an efficient approach to overcoming long-running synthesis challenges in protein materials development is established, which will expedite materials innovation.

Abstract

Motivation Identifying variant forms of gene clusters of interest in phylogenetically proximate and distant taxa can help to infer their evolutionary histories and functions. Conserved gene clusters may differ by only a few genes, but these small differences can in turn induce substantial phenotypes, such as by the formation of pseudogenes or insertions interrupting regulation. Particularly as microbial genomes and metagenomic assemblies become increasingly abundant, unsupervised grouping of similar, but not necessarily identical, gene clusters into consistent bins can provide a population-level understanding of their gene content variation and functional homology. Results We developed GeneGrouper, a command-line tool that uses a density-based clustering method to group gene clusters into bins. GeneGrouper demonstrated high recall and precision in benchmarks for the detection of the 23-gene Salmonella enterica LT2 Pdu gene cluster and four-gene Pseudomonas aeruginosa PAO1 Mex gene cluster among 435 genomes spanning mixed taxa. In a subsequent application investigating the diversity and impact of gene-complete and -incomplete LT2 Pdu gene clusters in 1130 S.enterica genomes, GeneGrouper identified a novel, frequently occurring pduN pseudogene. When investigated in vivo , introduction of the pduN pseudogene negatively impacted microcompartment formation. We next demonstrated the versatility of GeneGrouper by clustering distant homologous gene clusters and variable gene clusters found in integrative and conjugative elements. Pulmonary embolism (PE), which causes 10% of deaths in hospital in the UK and is a contributory factor in a further 10%, usually takes place because of the detachment of a vascular thrombus from the leg or pelvis. There are a number of ways of preventing PE, including the use of graduated compression stockings, drug treatment and the use of intermittent/sequential pneumatic calf compression. Symptoms of PE can vary considerably. In some patients, there may be pleuritic chest pain and dyspnoea, while in others there will be circulatory collapse.

Abstract

The periodical cicadas of North America (Magicicada spp.) are well-known for their long life cycles of 13 and 17 years and their mass synchronized emergences. Although periodical cicada life cycles are relatively strict, the biogeographic patterns of periodical cicada broods, or year-classes, indicate that they must undergo some degree of life cycle switching. We present a new map of periodical cicada Brood V, which emerged in 2016, and demonstrate that it consists of at least four distinct parts that span an area in the United States stretching from Ohio to Long Island. We discuss mtDNA haplotype variation in this brood in relation to other periodical cicada broods, noting that different parts of this brood appear to have different origins. We use this information to refine a hypothesis for the formation of periodical cicada broods by 1- and 4-year life cycle jumps.

A nursing development unit used a care delivery audit tool designed by Bowman, Webster and Thompson, to clarify, understand and measure nurses' work methods. It aimed to distinguish between primary nurisng, team nursing and task alocation. The pilot audit indicated a weakness in primary nursing although this was undertaken the ward team had been altered because of staff shortages. However the tool was easy to use in spite of its imitations.

Abstract

Nerve agents and pesticides represent a category of extremely toxic organophosphate compounds (OPs) that irreversibly inhibit the enzyme acetylcholinesterase, disturbing transmission in the synaptic clefts of muscles and nerves. Protection from these compounds necessitates the development of breathable barriers that can selectively block the passage of OPs. Hydrogels prepared from acrylamide, N,N′-methylene bis(acrylamide), N,N′- bis(acryloyl)cystamine, and hydrophilic pendant oximes are herein prepared, showing the ability to decontaminate and respond to the presence of OPs through a change in swelling. The oxime-based hydrogels show selective response only to malaoxon when tested against chemicals that are found in sweat as well as other reactive chemicals that are found in the environment. Pore sealing is demonstrated in perforated equilibrated gels within 3-4 h after the addition of malaoxon, showing actuation of the gel in response to organophosphates. This strategy demonstrates the ability to couple oxime-based decontamination and disulfide chemistry to produce hydrogels that can decontaminate organophosphate compounds, sense the decontamination product, and transduce this sensing response into actuation of the gel, which can be used to close pores in gel sheets or between fibers in a protective fabric coating.
