The majority of microbiome studies focused on understanding mechanistic relationships between the host and the microbiota have used mice and other rodents as the model of choice. However, the domestic pig is a relevant model that is currently underutilized for human microbiome investigations. In this study, we performed a direct comparison of the engraftment of fecal bacterial communities from human donors between human microbiotaassociated (HMA) piglet and mouse models under identical dietary conditions. Analysis of 16S rRNA genes using amplicon sequence variants (ASVs) revealed that with the exception of early microbiota from infants, the more mature microbiotas tested established better in the HMA piglets compared to HMA mice. Of interest was the greater transplantation success of members belonging to phylum Firmicutes in the HMA piglets compared to the HMA mice. Together, these results provide evidence for the HMA piglet model potentially being more broadly applicable for donors with more mature microbiotas while the HMA mouse model might be more relevant for developing microbiotas such as those of infants. This study also emphasizes the necessity to exercise caution in extrapolating findings from HMA animals to humans, since up to 28% of taxa from some donors failed to colonize either model. Traditional feed composition tables have been a useful tool in the field of animal nutrition throughout the last 70 yr. The objective of this paper is to discuss the challenges and opportunities associated with creating large feed ingredient composition tables. This manuscript will focus on three topics discussed during the National Animal Nutrition Program (NANP) Symposium in ruminant and nonruminant nutrition carried out at the American Society of Animal Science Annual Meeting in Austin, TX, on July 11, 2019, namely: 1) Using large datasets in feed composition tables and the importance of standard deviation in nutrient composition as well as different methods to obtain accurate standard deviation values, 2) Discussing the importance of fiber in animal nutrition and the evaluation of different methods to estimate fiber content of feeds, and 3) Description of novel feed sources, such as insects, algae, and single-cell protein, and challenges associated with the inclusion of such feeds in feed composition tables. Development of feed composition tables presents important challenges. For instance, large datasets provided by different sources tend to have errors and misclassifications. In addition, data are in different file formats, data structures, and feed classifications. Managing such large databases requires computers with high processing power and software that are also able to run automated procedures to consolidate files, to screen out outlying observations, and to detect misclassified records. Complex algorithms are necessary to identify misclassified samples and outliers aimed to obtain accurate nutrient composition values. Fiber is an important nutrient for both monogastrics and ruminants. Currently, there are several methods available to estimate the fiber content of feeds. Approximately 50% of sows are culled annually with more than one-third due to poor fertility. Our research demonstrated that age at puberty is an early pre-breeding indicator of reproductive longevity. Age at puberty can be measured early in life, has a moderate heritability, and is negatively correlated with lifetime number of parities. Detection of age at puberty is tedious and time consuming and is therefore not collected by the industry, which limits genetic progress. Genomic prediction is a viable approach to preselect gilts that will express puberty early and have superior reproductive longevity. The hypothesis that genetic variants explaining differences in age at puberty also explain differences in sow reproductive longevity was tested. Phenotypes, genotypes, and tissues from the UNL resource population (n > 1700) were used in genome-wide association analyses, genome, and RNA sequencing to uncover functional polymorphisms that could explain variation in puberty and reproductive longevity. A BeadArray including 56,424 SNP explained 25.2% of the phenotypic variation in age at puberty in a training set (n = 820). Evaluation of major windows and SNPs of subsequent batches of similar genetics (n = 412) showed that if all SNPs located in the major 1-Mb windows were tested, they explained a substantial amount of phenotypic variation (12.3 to 36.8%). Due to differences in linkage disequilibrium status, the most informative SNP from these windows explained a lower proportion of the variation (6.5 to 23.7%). To improve genomic predictive ability, the limited capability of BeadArray was enhanced by potential functional variants uncovered by genome sequencing of selected sires (n = 20; >20X). Genetic variants associated with traits such as age at puberty and litter size could provide insight into the underlying genetic sources of variation impacting sow reproductive longevity and productivity. Genomewide characterization and gene expression profiling were used using gilts from the University of Nebraska–Lincoln swine resource population (n = 1,644) to identify genetic variants associated with age at puberty and litter size traits. From all reproductive traits studied, the largest fraction of phenotypic variation explained by the Porcine SNP60 BeadArray was for age at puberty (27.3%). In an evaluation data set, the predictive ability of all SNP from highranked 1-Mb windows (1 to 50%), based on genetic variance explained in training, was greater (12.3 to 36.8%) compared with the most informative SNP from these windows (6.5 to 23.7%). In the integrated data set (n = 1,644), the top 1% of the 1-Mb windows explained 6.7% of the genetic variation of age at puberty. One of the high-ranked windows detected (SSC2, 12–12.9 Mb) showed pleiotropic features, affecting both age at puberty and litter size traits. The RNA sequencing of the hypothalami arcuate nucleus uncovered 17 differentially expressed genes (adjusted P < 0.05) between gilts that became pubertal early (180 d of age). Twelve of the differentially expressed genes are upregulated in the late pubertal gilts. One of these genes is involved in energy homeostasis (FFAR2), a function in which the arcuate nucleus plays an important contribution, linking nutrition with reproductive development. Energy restriction during the gilt development period delayed age at puberty by 7 d but increased the probability of a sow to produce up to 3 parities (P < 0.05). Identification of pleotropic functional polymorphisms may improve accuracy of genomic prediction while facilitating a reduction in sow replacement rates and addressing welfare concerns.

A total of 182 gilts were used to evaluate the effects of limiting energy intake during development compared to traditional corn-soybean meal based diets using the addition of soybean hulls to reduce caloric intake with ad libitum access to feed. The gilt developmental period consisted of 110 days, from 120 to 230 days of age. Data collected during development included body weight, backfat, longissimus muscle area, and feed intake, every 2 weeks. Estrus detection was conducted on a daily basis until breeding. Females were maintained in the breeding groups for 4 parities (785 days of age), at which time data collection ceased. Animals were only removed from the experiment for reproductive failure, ruptures, or severe lameness. Reproductive measurements included pre- and post-farrowing body weight and backfat, lactation length, lactation feed intake, number born alive, still-borns, mummified fetuses, weaned, litter birth and weaning body weights. Gilts fed the diets with 40% soybean hull inclusion consumed less feed compared to gilts fed the corn-soybean meal (control) diet (P < 0.05; 2.79 vs. 3.11 kg/d, respectively). There was no difference in daily feed intake during the overall developmental period when expressed on a % of body weight basis. Gilts consuming the soybean hulls consumed 33% less metabolizable energy during development. Reduced energy intake resulted in gilts that were 18.6 kg lighter, had 9 mm less backfat, and 5.5 cm2 less longissimus muscle area at breeding (P < 0.01). Gilts with lower energy intake had 30% less backfat, but only 12% less body weight and longissimus muscle area at breeding. Reduced energy intake delayed age at 1st detectable estrus by 11 days. The delay in puberty occurred earlier than the common age at which gilts are bred in commercial conditions. Feed costs during development were reduced by 17.26 U.S. dollars (USD)/hd and were 14.3% less for gilts fed the soybean hull diets compared to control diets. Gilts developed on the lowe

Traditional selection for sow reproductive longevity is ineffective due to low heritability and late expression of the trait. Incorporation of DNA markers into selection programs is potentially a more practical approach for improving sow lifetime productivity. Using a resource population of crossbred gilts, we explored pleiotropic sources of variation that influence age at puberty and reproductive longevity. Of the traits recorded before breeding, only age at puberty significantly affected the probability that females would produce a first parity litter. The genetic variance explained by 1-Mb windows of the sow genome, compared across traits, uncovered regions that influence both age at puberty and lifetime number of parities. Allelic variants of SNPs located on SSC5 (27–28 Mb), SSC8 (36–37 Mb) and SSC12 (1.2–2 Mb) exhibited additive effects and were associated with both early expression of puberty and a greater than average number of lifetime parities. Combined analysis of these SNPs showed that an increase in the number of favorable alleles had positive impact on reproductive longevity, increasing number of parities by up to 1.36. The region located on SSC5 harbors non-synonymous alleles in the arginine vasopressin receptor 1A (AVPR1A) gene, a G-protein-coupled receptor associated with social and reproductive behaviors in voles and humans and a candidate for the observed effects. This region is characterized by high levels of linkage disequilibrium in different lines and could be exploited in marker-assisted selection programs across populations to increase sow reproductive longevity. Individually fed heifers were used to determine the relationship of 3-methyl histidine, purine derivatives, and metabolizableprotein balance to feed efficiency. Heifers were fed finishing dietsthat were either deficient or sufficient in metabolizable protein. Urine samples were collected and analyzed for early, late, and entire feeding period concentrations of 3-methyl histidine, purine derivatives, and creatinine. Results from this study indicated a negative relationship between feed efficiency and metabolizable protein balance, and no relationship between 3-methyl histidine excretion and feed efficiency, suggesting that protein turnover and microbial protein synthesis are not related to feed efficiency

Two-hundred and forty pigs growing-finishing pigs were used to evaluate the feeding value of distillers dried grains with solubles (DDGS). Treatments consisted of 0, 5, 10 and 15% dietary DDGS inclusion. Treatments did not affect average daily gain (ADG), average daily feed intake (ADFI) or gain:feed (G:F) during the grower 1 period (P > 0.05). During the grower 2 period ADG and ADFI linearly decreased as DDGS increased (P < 0.05). No differences among treatments were detected throughout the feeding phase finisher 1 for ADG, ADFI, and G:F (P > 0.05). During the finisher 2 feeding phase, there was a linear reduction in ADG and ADFI in response to dietary DDGS inclusion (P = 0.01). Overall, linear reductions in ADG, ADFI and G:F were recorded as dietary DDGS increased (P < 0.05). Backfat and longissimus muscle area decreased as dietary DDGS concentration increased (P < 0.05). Overall, growth performance was reduced as dietary DDGS inclusion increased from 0 to 15%. The reduction in performance may have been partially explained or exacerbated by the elevated fiber concentration detected in the source of DDGS used in this study. Live weight and hot carcass weight decreased as dietary DDGS increased (P < 0.05). Dressing percentage did not differ among treatments (P = 0.72). After 10 d of retail display, no differences were observed among treatments for color or color change (P > 0.05). No differences in shear force were observed (P = 0.34). Total unsaturated fatty acids increased and total saturated fatty acids decreased (P < 0.05) as dietary DDGS increased. Treatments did not affect sensory characteristics (P > 0.05). The results of this investigation suggest that dietary DDGS inclusion altered fatty acid profile of the backfat of pigs by reducing total saturated fatty acid and increasing total unsaturated fatty acid concentration. Increasing concentration of dietary DDGS did not affect color, chemical composition or sensory characteristics of the LM.

Traditional corn (Zea mays L.) (TC), the primary grain used in swine (Sus scrofa) diets, stores a majority of its P as phytate, which is largely unavailable for digestion by nonruminant animals. Low- phytate corn (LPC) contains similar amounts of total P but a smaller percentage of P as phytate. When fed to swine, LPC increases P utilization and reduces P content of manure. While differences in P content between manure from animals fed TC and LPC diets have been documented, solubility and lability of manure P have not been compared. Manure P was characterized in manure from swine fed either LPC or TC diets in 2000 and 2001. Total P was lower (20 vs. 34 g kg-1) and N to P ratio was higher (4.5 vs. 3.3) in LPC manure than in TC manure. Manures were sequentially extracted with deionized water, 0.5 M NaHCO3, 0.1 M NaOH, and 1.0 M HCl. Extracts were analyzed for inorganic and total P. Most P (approximately 80%) in the extracts was in the inorganic form. Concentration of P in the water-extractable fraction was lower for LPC manure (10.2 g kg-1 in 2000 and 9.7 g kg-1 in 2001) than for TC manure (13.6 g kg-1 in 2000 and 17.0 g kg-1 in 2001). Percentage of total P in each extract was in the order of: H2O (60%), HCl (22%), NaHCO3 (12%), NaOH (8%), and residue (<1%). Total P and distribution of P in extracts indicates swine are able to utilize more P contained in LPC feed but the composition of P excreted in LPC manure is similar to TC manure. Solubility, crop availability, and lability of P in LPC manure should be similar to that of TC manure.

The P content of swine manure can be reduced through the addition of feed supplements or the use of selected corn hybrids. This study was conducted to compare interrill runoff losses of P and N from three soils following the application of swine manure obtained from selected diets. The soils used in this investigation included a Hersh sandy loam, Pierre silty clay, and Sharpsburg silt loam. Simulated rainfall was applied during both initial and wet runs to a soil pan on which swine manure produced from low phytate corn (LPC), phytase added to the diet (PHY), or a traditional corn diet (TCD) was added. Additional experimental treatments included inorganic fertilizer and an untreated check. For the initial rainfall simulation run, concentrations of dissolved P, bioavailable P, and Total P were greater for the fertilizer treatment than any of the manure treatments. Use of manure from a LPC diet generally did not result in a reduction in N and P concentrations in runoff when compared with the TCD. Concentrations and total amounts of nutrients transported in runoff were affected by soil type. Changing the TCD to LPC and PHY diets to reduce the P content of manure did not significantly affect the total amounts of DP, BAP, or Total P transported in runoff, when simulated rainfall was applied soon after manure application.

This publication is a revision of the previous swine nutrition publication prepared by the University of Nebraska and South Dakota State University. The focus of the publication continues to be on nutrient recommendations for swine. Specific factors (nutritional, environmental and managerial) that affect nutrient recommendations (Figure 1) have been considered and discussed. We believe that the identification and description of the factors in Figure 1 provide the framework for the nutrient recommendations presented in Tables 11 to 16. Industry advisors representing various facets of the pork industry were recruited to review and challenge the concepts incorporated in this publication. Also, in situations where “gray areas” existed, these industry representatives made specific proposals or recommendations. Therefore, our ultimate goal was to use the knowledge of respected swine nutritionists who represent a cross section of the feed industry to improve the application of this publication. In preparing this publication, a priority has been to discuss some of the controversial and experimental swine nutrition issues currently being explored. The discussion of these issues has been focused to emphasize results presented in the scientific literature. Forty-eight crossbred rabbits were used in three replications of a 2 x 4 factorial arrangement to investigate the short-term responses of tissue accretion, calpains and calpastatin activity, and nucleic acid and protein concentrations to β- adrenergic agonist (BAA) feeding. Rabbits were fed a 17% CP diet with or without 7 ppm of L644,969 and slaughtered after 1, 4, 8, or 16 d of treatment. Empty body dressing percentage and biceps femoris weight (as a percentage of empty body weight [EBWI) were significantly higher in the treated rabbits than in the controls after 16 d of treatment. Heart and liver weights (as a percentage of EBW) were higher ( P < .05) after 1 d and liver weight (as a percentage of EBW) was lower (P < .05) after 16 d in treated vs controls. Except for an elevation of skeletal muscle m-calpain after 16 d, BAA-supplementation did not affect the calpain-calpastatin system. Muscle RNA concentrations and RNA:DNA ratios were higher (P < .05) in treated rabbits after 1 d and remained higher thereafter. Protein:RNA ratios were lower ( P < .0 1) in treated than in control rabbits after 4 d and remained lower throughout the trial. Muscle DNA content was lower after 4 d and higher after 16 d; RNA content was higher after 4, 8, and 16 d; and protein content was higher after 16 d in treated vs control rabbits. Liver nucleic acid and protein concentrations were not affected by BAA treatment. Heart RNA:DNA ratios were higher ( P < .0 1) after 1 d and protein:RNA ratios were lower (P < .05) after 4 d in the treated rabbits than in the controls. Collectively, these data imply that BAA-induced muscle growth in rabbits occurs through hyperplasia and seems to be related to elevated protein synthetic capacity.
