1. Oil spills and sea turtles: documented effects and considerations for response and assessment efforts

Abstract: Hydrocarbon (i.e. oil) extraction, transport, consumption, and pollution occur daily in marine environments, threatening vulnerable natural resources, habitats, and biodiversity. However, threats of episodic oil spills to imperiled marine species are not as well-studied as more acute, readily apparent threats such as incidental capture in fisheries and habitat degradation. We performed a global review of spill incident databases to evaluate reported effects of oil spills on sea turtles, which are flagship species of the world’s oceans. Our goals were to (1) summarize available information about oil spills and their effects on sea turtles; (2) identify major knowledge gaps; and (3) provide recommendations related to oil spills and sea turtles for managers, researchers, and conservation groups around the world. Over 2000 oil spill incidents of variable magnitude (range: ~20 to >1 million barrels) have occurred worldwide in the past 60 yr in areas where sea turtles are found, but resulting effects on sea turtles have been reported in less than 2% of those incidents. Further, evidence indicates that most effects are related to heavy external oiling, while chemical effects of oil exposure have not been well defined. We recommend that, where available, resources be prioritized to document co-occurrence of turtles and oil, the degree and nature of oil exposure, and mortality and reproductive losses to individual sea turtles and their populations caused by spills and spill response activities. These data will best inform assessments of the extent and magnitude of adverse effects of oil spills on sea turtles.

2. Effects of incubation temperature on hatchling performance and phenotype in loggerhead sea turtle Caretta caretta

Abstract: Under natural conditions sea turtle eggs are subjected to a changing thermal environment, but little is known about the effect of these temperature fluctuations during incubation on the performance and phenotype of hatchlings. The aim of this study was to determine how incubation temperature pattern (increasing or stable) and incubation temperature regime (low or high) affect incubation and hatching duration, hatching and emergence success, hatchling phenotype (carapace length and weight) and self-righting interval at hatching. Loggerhead sea turtle Caretta caretta (Linnaeus, 1758) clutches were collected at different beaches on the Cape Verde archipelago and divided among incubators with different temperature regimes and patterns. Minimum straight carapace length and weight of all individuals were measured at hatching. In addition, the hatching duration and the time interval required for each hatchling to self-right were recorded. Results showed that incubation temperature regimes influenced all studied parameters more than the increasing temperature pattern. Low incubation temperature regimes, both in the increasing and stable pattern, increased the incubation time, produced bigger hatchlings and caused a slower righting response compared to the high temperature regimes. An optimal range of incubation temperatures was determined by assessing the most favorable values for hatchlings, although some differences were found in the higher temperatures of this optimal range between different rookeries. This means that turtle incubation in the laboratory should, as far as possible, follow the natural incubation temperature fluctuation of the studied rookery.

Abstract: Although sea turtles have received substantial focus worldwide, research on the immature life stages is still relatively limited. The latter is of particular importance, given that a large proportion of sea turtle populations comprises immature individuals. We set out to identify knowledge gaps and identify the main barriers hindering research in this field. We analyzed the perceptions of sea turtle experts through an online survey which gathered their opinions on the current state of affairs on immature sea turtle research, including species and regions in need of further study, priority research questions, and barriers that have interfered with the advancement of research. Our gap analysis indicates that studies on immature leatherback Dermochelys coriacea and hawksbill Eretmochelys imbricata turtles are lacking, as are studies on all species based in the Indian, South Pacific, and South Atlantic Oceans. Experts also perceived that studies in population ecology, namely on survivorship and demography, and habitat use/behavior, are needed to advance the state of knowledge on immature sea turtles. Our survey findings indicate the need for more inter-disciplinary research, collaborative efforts (e.g. data-sharing, joint field activities), and improved communication among researchers, funding bodies, stakeholders, and decision-makers.

4. Entanglement entropy at CFT junctions

Abstract: We consider entanglement through permeable junctions of N (1+1)-dimensional free boson and free fermion conformal field theories. In the folded picture we constrain the form of the general boundary state. We calculate replicated partition functions with interface operators inserted in the partially-folded picture, from which the entanglement entropy is calculated. The functional form of the universal and constant terms are the same as the N=2 case, depending only of the total transmission of the junction and the unit volume of the zero mode lattice. For N>2 we see a subleading divergent term which does not depend on the parameters of the junction. For N=3 we consider some specific geometries and discuss various limits.

Abstract: Abstract: In this paper we calculate the entanglement entropy for topological interfaces in rational conformal field theories for the case where the interface lies at the boundary of the entangling interval and for the case where it is located in the center of the entangling interval. We compare the results to each other and also to the recently calculated left/right entropy of a related BCFT. We also comment of the entanglement entropies for topological interfaces for a free compactified boson and Liouville theory.

Abstract: The signal recognition particle receptor (SR) is required for the cotranslational targeting of both secretory and membrane proteins to the endoplasmic reticulum (ER) membrane. During targeting, the SR interacts with the signal recognition particle (SRP) which is bound to the signal sequence of the nascent protein chain. This interaction catalyzes the GTP-dependent transfer of the nascent chain from SRP to the protein translocation apparatus in the ER membrane. The SR is a heterodimeric protein comprised of a 69-kD subunit (SR alpha) and a 30-kD subunit (SR beta) which are associated with the ER membrane in an unknown manner. SR alpha and the 54-kD subunits of SRP (SRP54) each contain related GTPase domains which are required for SR and SRP function. Molecular cloning and sequencing of a cDNA encoding SR beta revealed that SR beta is a transmembrane protein and, like SR alpha and SRP54, is a member of the GTPase superfamily. Although SR beta defines its own GTPase subfamily, it is distantly related to ARF and Sar1. Using UV cross-linking, we confirm that SR beta binds GTP specifically. Proteolytic digestion experiments show that SR alpha is required for the interaction of SRP with SR. SR alpha appears to be peripherally associated with the ER membrane, and we suggest that SR beta, as an integral membrane protein, mediates the membrane association of SR alpha. The discovery of its guanine nucleotide-binding domain, however, makes it likely that its role is more complex than that of a passive anchor for SR alpha. These findings suggest that a cascade of three directly interacting GTPases functions during protein targeting to the ER membrane.

10. Household determinants of biocontaminant exposures in Canadian homes

Abstract: Exposure to biocontaminants, such as dust mites, animal dander, bacteria, and mold, is associated with a range of health effects. This study identified household characteristics associated with indoor biocontaminant loadings in four Canadian cities. Floor dust was collected in 290 Canadian homes in Edmonton, Halifax, Montreal, and Windsor. The dust samples were analyzed for house dust mite allergens (Der f 1 and Der p 1), cat allergen (Fel d 1), cockroach allergen (Bla g 1), beta-(1,3)-D-glucan, and endotoxin. Household information was obtained through questionnaires and home inspections. We performed univariate and multivariate analyses to identify household determinants of biocontaminant loadings and mold odor presence. We observed large regional variations for all biocontaminants, except for cockroach allergen. The ranges of the contaminants measured in loadings and concentrations were similar to that of previous Canadian studies. Household characteristics including presence of carpeting, low floor cleaning frequency, older home age, presence of pets, and indoor relative humidity above 45% were positively associated with the presence of multiple indoor biocontaminants. High floor cleaning frequency and use of dehumidifiers were negatively associated with the presence of multiple indoor biocontaminants. Mold odor was positively associated with older home age, past water damage, and visible mold growth.

13. Synchronous Emplacement of the Anorthosite Xenolith-Bearing Beaver River Diabase and One of the Largest Lava Flows on Earth

Abstract: New geochronologic and paleomagnetic data from the North American Midcontinent Rift (MCR) reveal the synchronous emplacement of the Beaver River diabase, the anorthosite xenoliths within it, and the Greenstone Flow—one of the largest lava flows on Earth. A U-Pb zircon date of 1091.83 (Formula presented.) 0.21 Ma (2 (Formula presented.)) from one of the anorthosite xenoliths is consistent with the anorthosite cumulate forming as part of the MCR and provides a maximum age constraint for the Beaver River diabase. Paired with the minimum age constraint of a cross-cutting Silver Bay intrusion (1091.61 (Formula presented.) 0.14 Ma; 2 (Formula presented.)), these data tightly bracket the age of the Beaver River diabase to be 1091.7 (Formula presented.) 0.2 Ma (95% CI), coeval with the eruption of the Greenstone Flow (1091.59 (Formula presented.) 0.27 Ma; 2 (Formula presented.))—which is further supported by indistinguishable tilt-corrected paleomagnetic pole positions. Geochronological, paleomagnetic, mineralogical and geochemical data are consistent with a hypothesis that the Beaver River diabase was the feeder system for the Greenstone Flow. The large areal extent of the intrusives and large estimated volume of the volcanics suggest that they represent a rapid and voluminous ca. 1,092 Ma magmatic pulse near the end of the main stage of MCR magmatism.

Abstract: Shortcomings of approaches to classifying psychopathology based on expert consensus have given rise to contemporary efforts to classify psychopathology quantitatively. In this paper, we review progress in achieving a quantitative and empirical classification of psychopathology. A substantial empirical literature indicates that psychopathology is generally more dimensional than categorical. When the discreteness versus continuity of psychopathology is treated as a research question, as opposed to being decided as a matter of tradition, the evidence clearly supports the hypothesis of continuity. In addition, a related body of literature shows how psychopathology dimensions can be arranged in a hierarchy, ranging from very broad "spectrum level" dimensions, to specific and narrow clusters of symptoms. In this way, a quantitative approach solves the "problem of comorbidity" by explicitly modeling patterns of co-occurrence among signs and symptoms within a detailed and variegated hierarchy of dimensional concepts with direct clinical utility. Indeed, extensive evidence pertaining to the dimensional and hierarchical structure of psychopathology has led to the formation of the Hierarchical Taxonomy of Psychopathology (HiTOP) Consortium. This is a group of 70 investigators working together to study empirical classification of psychopathology. In this paper, we describe the aims and current foci of the HiTOP Consortium. These aims pertain to continued research on the empirical organization of psychopathology; the connection between personality and psychopathology; the utility of empirically based psychopathology constructs in both research and the clinic; and the development of novel and comprehensive models and corresponding assessment instruments for psychopathology constructs derived from an empirical approach.

The aim of this study was to assess the cost-effectiveness and budget impact of enzymatic debridement with clostridial collagenase ointment (CCO) compared with autolytic debridement with medicinal honey (MH) for PU treatment from a US payer/Medicare perspective in the hospital outpatient department setting.Methods: A cost-effectiveness analysis using a Markov model was developed using a 1-week cycle length across a 1-year time horizon. The three health states were inflammation/senescence, granulation/proliferation (ie, patients achieving 100% granulation), and epithelialization. Data sources included the US Wound Registry, Medicare fee schedules, and other published clinical and cost studies about PU treatment.Results: In the base case analysis over a 1-year time horizon, CCO was the economically dominant strategy (ie, simultaneously conferring greater benefit at less cost). Patients treated with CCO experienced 22.7 quality-adjusted life weeks (QALWs) at a cost of $6,161 over 1 year, whereas MH patients experienced 21.9 QALWs at a cost of $7,149. Patients treated with CCO achieved 11.5 granulation weeks and 6.0 epithelization weeks compared with 10.6 and 4.4 weeks for MH, respectively. The number of clinic visits was 40.1 for CCO vs 43.4 for MH, and the number of debridements was 12.3 for CCO compared with 17.6 for MH. Probabilistic sensitivity analyses determined CCO dominant in 72% of 10,000 iterations and cost-effective in 91%, assuming a benchmark willingness-to-pay threshold of $50,000/quality-adjusted life year ($962/QALW). The budget impact analysis showed that for every 1% of patients shifted from MH to CCO, a cost savings of $9,883 over 1 year for a cohort of 1,000 patients was observed by the payer. Conclusion: The results of these economic analyses suggest that CCO is a cost-effective, economically dominant alternative to MH in the treatment of patients with PUs in the hospital outpatient department setting. The alarmingly high rate of recurrence of ulcerations in diabetic foot requires a change in our approach to care and to the vernacular in the medical literature. With its high rates of morbidity and recurrence, care of the complex diabetic foot may be aptly comparable to many forms of cancer. Therefore, our efforts should be not only in rapid healing of open wounds but also in maximizing ulcer-free days for the patient in diabetic foot remission. One facet of the multidisciplinary approach in managing wounds is achieved by reducing peak plantar pressures by offloading the foot with various conservative and surgical techniques aimed at reducing areas of stress caused by ambulation and improper shoe gear. Evidence supports the use of total contact casts as the gold standard for offloading open wounds; however, other methods have gained popularity as well. Novel approaches in surgical techniques and advances in wearable technology appear to show promise in measuring and modulating dangerous pressure and inflammation to extend remission and improve quality of life for these most complex patients. Abstract: Preparing for public health emergencies is an ongoing process and involves a variety of approaches and tools. Tabletop exercises are one of the tools designed to simulate the emergence of a public health emergency and address some or all of the phases of emergency management: mitigation, preparedness, response, and recovery.1 They typically are designed to include participation of stakeholders from diverse and complementary backgrounds, including command, operations, logistics, planning, and finance.2 Effective tabletop exercises provide a plausible scenario that require cooperation and communication from these functional areas. Tabletops also require forward thinking and planning in a variety of scenarios. When a public health emergency occurs, decision makers may be overwhelmed with decisions that need their immediate attention. Tabletop exercises can provide a framework to help decision makers anticipate future challenges, which may provide the mental model encompassing knowledge and insights that inform both current and future decisions.

19. Metabolomics reveals chemical changes in Acer saccharum sap over a maple syrup production season

Abstract: Maple syrup, made by boiling the sap of Acer saccharum, is an important agriculture commodity in eastern Canada and New England. Although the collection season is relatively short, a rich progression in the sensory qualities of maple syrup can occur throughout the season. A risk associated with maple syrup production at the end of a season is the development of off-flavors that result in syrup with little to no commercial value. Maple syrup producers in Canada and the USA call this 'buddy syrup'. In this study, sugar maple (Acer saccharum) sap was collected in sequential samples through the harvest season from stands across Ontario. Metabolomics analysis of the sap samples was performed by high-resolution mass spectrometry. This revealed an evolution of the chemical composition, mainly occurring 30 days prior to leaf emergence. The major chemical constituent of maple syrup, sucrose, decreased sharply in late season sap, driven by microbial activity. The alditol mannitol increased in late season sap to concentrations ≥2 mg/mL and is likely an indicator of the start of photosynthesis. Amino acids, notably methionine and asparagine were present in higher amounts in late season sap. Non-targeted analysis revealed a series of related compounds that contained quaternary ammonium moieties including choline, hercynine, trigonelline, glycine betaine and carnitine increased in late season sap. These classes of compounds could act as methyl donors during the heating/evaporation of sap into syrup, affecting taste. Based on descriptions of the nature of buddy syrup and an extensive literature on flavors in foods, the amino acids methionine and asparagine were found as likely precursors to the compounds responsible for buddy syrup.

20. A foliar endophyte of white spruce reduces survival of the eastern spruce budworm and tree defoliation

Abstract: Wild eastern spruce budworm (Choristoneura fumiferana Clemens) were reared on white spruce (Picea glauca (Moench) Voss) trees, half of which had been previously inoculated with a native endophytic fungus, Phialocephala scopiformis DAOM 229536 Kowalski and Kehr (Helotiales, Ascomycota). Survival up to pupation and up to adult emergence was approximately 27% higher for budworm juveniles that developed on control trees compared to trees inoculated with the endophyte. The endophyte did not influence the size or sex of survivors but did reduce defoliation by approximately 30%. Reductions in defoliation on endophyte-inoculated versus control trees, due to reductions in survival of juvenile budworms, suggests that tree inoculations with P. scopiformis could play an important role in integrated management programs against the eastern spruce budworm.

Base-case comparisons were performed among CRT-Ds of three manufacturers – Boston Scientific Corporation, St. Jude Medical, and Medtronic – over a 6-year time horizon, as per the available clinical data. As a sensitivity analysis, we evaluated CRT-Ds as well as single-chamber implantable cardioverter defibrillator (ICD-VR) and dual-chamber implantable cardioverter defibrillator (ICD-DR) devices over a longer 10-year period. Abstract: A manipulative field study was carried out to determine whether the foliar endophyte fungus, Phialocephala scopiformisDAOM229536, decreased the performance of eastern spruce budworm, Choristoneura fumiferana larvae developing on white spruce trees. Overwintered second-instar budworm larvae from a laboratory colony or from a wild population were placed on endophyte positive or negative trees one or two weeks before budburst. The presence of the endophyte in the needles reduced the survival of C. fumiferana from both a wild population and a laboratory colony. Survival for budworm juveniles up to pupation and to adult emergence was 13% and 17% lower, respectively, on endophyte positive trees. Hysterectomy, the most common surgical treatment of AUB, has significant morbidity, low mortality, long recovery, and high associated health care costs. Global endometrial ablation (GEA) provides a surgical alternative with reduced morbidity, cost, and recovery time. The NovaSure® system utilizes unique radiofrequency impedance-based GEA technology. This study evaluated cost effectiveness of AUB treatment with NovaSure ablation versus other GEA modalities and versus hysterectomy from the US commercial and Medicaid payer perspectives. Methods: A health state transition (semi-Markov) model was developed using epidemiologic, clinical, and economic data from commercial and Medicaid claims database analyses, supplemented by published literature. Three hypothetical cohorts of women receiving AUB interventions were simulated over 1-, 3-, and 5-year horizons to evaluate clinical and economic outcomes for NovaSure, other GEA modalities, and hysterectomy. Results: Model analyses show lower costs for NovaSure-treated patients than for those treated with other GEA modalities or hysterectomy over all time frames under commercial payer and Medicaid perspectives. By Year 3, cost savings versus other GEA were $930 (commercial) and $3,000 (Medicaid); cost savings versus hysterectomy were $6,500 (commercial) and $8,900 (Medicaid). Coinciding with a 43%–71% reduction in need for re-ablation, there were 69%–88% fewer intervention/reintervention complications for NovaSure-treated patients versus other GEA modalities, and 82%–91% fewer versus hysterectomy. Furthermore, NovaSure-treated patients had fewer days of work absence and short-term disability. Cost-effectiveness metrics showed NovaSure treatment as economically dominant over other GEA modalities in all circumstances. With few exceptions, similar results were shown for NovaSure treatment versus hysterectomy. Conclusion: Model results demonstrate strong financial favorability for NovaSure ablation versus other GEA modalities and hysterectomy from commercial and Medicaid payer perspectives. Abstract: Mycotoxins are major food contaminants affecting global food security, especially in low and middle-income countries. The European Union (EU) funded project, MycoKey, focuses on “Integrated and innovative key actions for mycotoxin management in the food and feed chains” and the right to safe food through mycotoxin management strategies and regulation, which are fundamental to minimizing the unequal access to safe and sufficient food worldwide. As part of the MycoKey project, a Mycotoxin Charter (charter.mycokey.eu) was launched to share the need for global harmonization of mycotoxin legislation and policies and to minimize human and animal exposure worldwide, with particular attention to less developed countries that lack effective legislation. This document is in response to a demand that has built through previous European Framework Projects—MycoGlobe and MycoRed—in the previous decade to control and reduce mycotoxin contamination worldwide. All suppliers, participants and beneficiaries of the food supply chain, for example, farmers, consumers, stakeholders, researchers, members of civil society and government and so forth, are invited to sign this charter and to support this initiative.

Abstract: Surveys of foliar endophytes from the Acadian forest region over the past three decades have identified numerous phylogenetically diverse fungi producing natural products toxic to forest pests and diseases. The life histories of some conifer endophytes can be restricted to plant foliage or may include saprotrophic phases on other plants tissues or even alternate hosts. Considering the potentially broad host preferences of conifer endophytes we explored fungi isolated from understory species and their metabolites as part of an ongoing investigation of fungal biodiversity from the Acadian forest. We report a hitherto unidentified Xylariomycetidae species isolated from symptomatic Labrador tea (Rhododendron groenlandicum) leaves and mountain laurel (Kalmia latifolia) collected in coastal southern New Brunswick, Canada. Morphological and phylogenetic evidence demonstrated the unknown species was a novel Synnemapestaloides (Sporocadaceae) species, described here as Syn. ericacearum. A preliminary screening assay indicated that the culture filtrate extract of the new species was potently antifungal towards the biotrophic pathogen Microbotryum violaceum, warranting an investigation of its natural products. Abstract: The occurrence of P. verrucosum and ochratoxin A (OTA) were surveyed for 3 and 4 years, respectively. A total of 250 samples was collected from an average of 30 farms during the 2011, 2012, 2013 and 2014 winter seasons. Most storage bins surveyed were typically 11 m high round bins made of corrugated, galvanized steel, with flat-bottoms and conical roofs. Samples of clumped grain contained the most P. verrucosum (p<0.05, n = 10) followed by samples taken from the first load (n = 24, mean = 147±87 CFU/g) and last load (n = 17, mean = 101±77 CFU/g). Five grain samples (2.2%) tested positive for OTA, citrinin and OTB at concentrations of 14.7±7.9, 4.9±1.9 and 1.2±0.7 ng/g, with only three samples exceeding 5 ng/g. Grain samples positive for OTA were related to moisture resulting from either condensation or migrating moist warm air in the bin or areas where precipitation including snow entered the bin. Bins containing grain and clumps contaminated with OTA were studied in detail. A number of statistically-significant risk factors for OTA contamination were identified. These included 1) grain clumps accumulated around or directly under manhole openings, 2) debris and residue of old grain or grain clumps collected from the bin walls or left on storage floor and augers and 3) grain clumps accumulated around side doors. Even when grain enters storage below the 14.5% threshold of moisture, condensation and moisture migration occurs in hotspots in modern corrugated steel storage bins. Hot spots of OTA contamination were most often in areas affected by moisture migration due to inadequate aeration and exposure to moisture from precipitation or condensation. Further, we found that the nature of the condensation affects the nature and distribution of small and isolated areas with high incidence of toxin contamination and/or P. verrucosum prevalence in the grain bins examined.

50. Sampling for Personal Rapid Transit Empty Vehicle Redistribution

Abstract: A Personal Rapid Transit (PRT) system uses compact, computer-guided vehicles running on dedicated guideways to carry individuals or small groups directly between pairs of stations. PRT vehicles operate on demand, when a passenger requests service at his/her origin station. Because the number of trips requested from a station need not equal the number of trips ending there, some vehicles must move empty to balance the flows. The empty vehicle redistribution (EVR) problem is to decide which empty vehicles to move, either reactively, in response to known requests, or proactively, in anticipation of future requests. This paper develops a new algorithm for the EVR problem called Sampling and Voting (SV). SV chooses reactive movements using a simple nearest-neighbor rule, and it chooses proactive movements by generating an ensemble of possible sequences of future passenger requests, solving a deterministic optimization problem for each sequence individually, and then finding the empty vehicle movements that are common among the sequences. Moving vehicles proactively is essential for providing low passenger waiting times. The new SV algorithm is tested in simulation with several case study systems, and it produces significantly lower passenger waiting times than existing EVR algorithms. Variants of the SV method developed here for PRT are also applicable to conventional taxi systems and emergency response systems.
