ABSTRACTIntroductionOX40 is an immune checkpoint in cancer and its presence in cancer is a good prognosis, making it a highly relevant target for the development of new immunotherapies.Areas coveredThe patent literature reveals vital information on new trends in cancer therapies. The authors used the patent databases of the six major patent offices in the world: United States Patent and Trademark Office, European Patent Office, World Intellectual Property Organization, Japan Patent Office, State Office of Intellectual Property of China and Korean Intellectual Property Office, to generate a panorama of patents related to OX40 agonists. Specific patents have been grouped into innovative patents and adoption patents.Expert opinionAn increasing trend in the development of OX40 agonists in cancer, particularly in the years 2018 and 2019. United States was the leader in generating patents, followed by China and England. Major pharmaceutical companies have at least one anti-OX40 agonist, MEDI6469 and MEDI-0562 (AstraZeneca), PF-04518600 (Pfizer), GSK3174998 (GlaxoSmithKline), BMS-986,178 (Bristol-Myers Squibb) and MOXR0916 (Roche), which represent 68% of clinical trials conducted with OX40 agonists.

- Abstract
Schizosaccharomyces pombe (Sz. pombe), or fission yeast, is an ascomycete unicellular fungus that has been used as a model system for studying diverse biological processes of higher eukaryotic cells, such as the cell cycle and the maintenance of cell shape, apoptosis, and ageing. Sz. pombe is a rod-shaped cell that grows by apical extension; it divides along the long axis by medial fission and septation. The fission yeast has a doubling time of 2–4 hours, it is easy and inexpensive to grow in simple culture conditions, and can be maintained in the haploid or the diploid state. Sz. pombe can be genetically manipulated using methods such as mutagenesis or gene disruption by homologous recombination. Fission yeast was defined as a micro-mammal because it shares many molecular, genetic, and biochemical features with cells of higher eukaryotes in mRNA splicing, post-translational modifications as N-glycosylation protein, cell-cycle regulation, nutrient-sensing pathways as the target of rapamycin (TOR) network, cAMP-PKA pathway, and autophagy. This chapter uses Sz. pombe as a useful model for studying important cellular processes that support life such as autophagy, apoptosis, and the ageing process. Therefore, the molecular analysis of these processes in fission yeast has the potential to generate new knowledge that could be applied to higher eukaryotes.

“El cáncer de mama es la primera causa de muerte por neoplasia en mujeres a nivel mundial, es una enfermedad compleja y de etiología multifactorial, lo cual dificulta su tratamiento. Esta enfermedad se clasifica mediante inmunohistoquímica en tres subtipos: el subtipo positivo a receptores hormonales (ER+), el cual responde a la terapia hormonal, el subtipo positivo al receptor de crecimiento epidérmico humano tipo 2 (HER2+), que se trata con el anticuerpo monoclonal trastuzumab y el cáncer de mama triple negativo (TN) que carece de biomarcadores para su diagnóstico y para terapia dirigida. Existe una amplia información acerca de alteraciones presentes en las células cancerosas, por ejemplo, el incremento de especies reactivas de oxígeno (ERO), que está asociado al daño al ADN, la inestabilidad genómica, la activación de distintas vías de señalización relacionadas con la supervivencia y la proliferación celular y al incremento en la secreción de citocinas proinflamatorias, propiciando el origen y progresión del cáncer. En el presente trabajo, se determinaron los niveles y el origen de las ERO en diferentes líneas celulares de cáncer de mama.”

"El bloqueo farmacológico del RDA2 incrementa la activación de la proteína DARPP-32 en el animal hemicastrado y se observó un aumento a las 48 horas después de la microinyección. Así también, cuando se antagoniza el sistema dopaminérgico ovárico con sulpiride o SCH23390 en el animal entero, se incrementa la lipoperoxidación cuatro y doce horas después de la microinyección respectivamente, este efecto se mantuvo hasta por dieciséis horas. En este mismo modelo, se observó un aumento en la concentración de nitritos cuando se administró el sulpiride a las cuatro horas, en tanto que para el SCH23390 doce horas después. Sin embargo, en el animal hemicastrado ocurrió un incremento de la lipoperoxidación cuatro otras después sólo cuando el ovario derecho permanece in situ. Los resultados del presente estudio nos permiten inferir que bloqueo farmacológico de los receptores dopaminérgicos, aumenta el metabolismo de la dopamina que tiene como consecuencia el incremento del estrés oxidativo que deriva en daño a nivel de membranas celulares incrementando la lipoperoxidación, como resultado la alteración de las células productoras de hormonas esteroides sexuales en el ovario, además de que la ausencia de la unión de dopamina a su receptor interfiriere en la vía de señalización mediada por la DARPP-32."

"El factor inhibidor de la migración de macrófagos (MIF) es una proteína multifuncional que tiene un papel importante en el proceso inflamatorio. MIF es secretado principalmente por células inmunes y ciertos tipos de células que participan en la regulación de la respuesta inmune. En macrófagos, la secreción de MIF se ha relacionado a la inhibición de la autofagia y a otros procesos independientes de autofagia, pero todavía no se conoce con detalle su mecanismo de secreción. En cáncer, MIF participa en diversos estados de la progresión tumoral. En cáncer de mama en particular, se ha encontrado dependencia de autofagia de las células cancerosas de tipo triple negativo para sobrevivir. Nosotros medimos los niveles de MIF secretados por líneas celulares de cáncer de mama cuando se inhibe la autofagia con cloroquina, y se encontró por primera vez que la autofagia podría modular la secreción de MIF diferencialmente en líneas celulares de cáncer de mama de diferentes subtipos. Esta diferencia en la secreción de MIF puede tener impacto en la sobrevivencia, proliferación y en el tipo de respuesta inflamatoria en cada subtipo de cáncer."

"En este trabajo se propone caracterizar mediante mutagénesis de sitio la contribución de las estructuras: asa – P, asa intracelular S4 – S5 y segmento S6 del dominio DIII, sobre la inactivación lenta del canal de sodio de músculo esquelético de rata Nav1.4. Nosotros deducimos que las regiones de bisagra juegan un papel importante en el acoplamiento conformacional entre el dominio sensor de voltaje y el poro en canales iónicos dependientes de voltaje. Nuestros resultados determinan que, glicina 1148 en la asa intracelular S4–S5, lisina 1237 en la asa–P, glicina 1275 y asparagina 1281 en el segmento S6, situados en el dominio DIII están implicados en la inactivación lenta en el canal de sodio rNav1.4, consistente con una inactivación tipo C observada en canales de potasio, y la participación de estas estructuras en la inactivación lenta es independiente de la inactivación rápida."

29. Efecto de la infección con el virus de dengue sobre la expresión de genes supresores de la respuesta inmune antiviral

"En el presente trabajo se observo que la infección con el virus de dengue inhibe la fosforilación de la proteína STAT2 y la translocación nuclear de STAT1 fosforiladada y que estos efectos son revertidos cuando la infección ocurre posterior al pretratamiento con IFN-α. Asi mismo, se observó la disminución de la activación de respuesta inducida por la vía JAK-STAT del IFN-tipo I a través de la disminución de la expresión del ARNm de las proteínas de respuesta antiviral OAS y MXA durante la infección con el virus de denfue. Al igual que otros autores, no se observo el abatimiento total de replicación viral con el pretratamiento con IFN-α. No obstante, se observó que la infección con el virus de dengue indujo la transcripción y producción de las proteínas SOCS-1 y SOCS-3, principalmente SOCS-1 cuyos incrementos fueron más altos a los niveles mostrados por la proteína SOCS-3, la cual mostro incrementos a tiempos similares al incremento en el título viral."
