Abstract Attractivity and selectivity of two types of traps with synthetic, long-lasting, bisexual generic attractants were compared to conventional light traps to promote their wider use, as an easy-to-use standardised method for entomology. The targeted herbivorous Macroheterocera species playing important role in ecosystems as food source for higher trophic levels (e.g. predatory arthropods, birds and mammals), while other hand they can cause significant economic loss in agriculture. Data on their population dynamic and composition of their assemblages are necessary for both nature conservation and efficient pest management. Light- and semiochemical-baited traps with semisynthetic- (SBL = the acronym stands for semisynthetic bisexual lure) and synthetic lures (FLO = the acronym stands for floral lure of synthetic floral compounds) were used in species rich area of West Ukraine, and in all 10,926 lepidopterans trapped were identified. The attractivity of the light trap was highest with 252 species caught, while traps with semiochemicals captured 132 species including 28 exclusively caught only by them. The qualitative selectivity of light vs. semiochemical-baited traps differed considering both taxa and habitat preferences in such a way that they completed each-other. Differences in quantitative selectivity were also proved even in case of pest species. The parameters of methods varied depending on the phenological phases of the studied assemblages. Considering the revealed attractivity and selectivity, the parallel use of the two methods can offer improved reliable data for conservation biology and pest management.

Abstract Several Eurydema species (Hemiptera: Pentatomidae) are considered as pests, however, reports on their chemical ecology are scarce. In the current study we focused on Eurydema ornata (Linnaeus) a pentatomid pest of several brassicaceous crops. Since the species is known to feed preferably on generative parts of plants, a series of floral and green leaf volatiles were tested by electroantennography and compounds eliciting remarkable responses were also tested in the field. Three compounds elicited the most outstanding responses from antennae of E. ornata: allyl isothiocyanate, phenylacetaldehyde and ± linalool. Field experiments were conducted in Hungary between 2017 and 2021 to test the potential attractive effects of the compounds. Three Eurydema species were caught in the experiments E. ornata, E. oleracea (Linnaeus) and E. ventralis Kolenati. In the experiments combinations containing allyl isothiocyanate attracted both males and females of E. ornata. The compound was also attractive on its own, in a positive, dose-dependent manner. When presented alone, neither phenylacetaldehyde nor ± linalool was attractive to the species, furthermore, addition of these compounds to allyl isothiocyanate did not affect attraction considerably. To our knowledge this is the first demonstration of field attration of an Eurydema species to a semiochemical and one of the few reports on trapping of a pentatomid species with a synthetic plant volatile in the field. Perspectives regarding research and potential practical applications are discussed in the paper.

One of the most important pollinator taxa is Bombus (Hymenoptera, Apidae), the genus of bumble bees, since they are important, often specialized, pollinators of many plants. As a result of climate change, warming winters and changes in landscape structure, the distribution and frequency of Bombus species is constantly changing. To develop appropriate protection strategies, it is essential to monitor them and update the occurrence and threat status of the species. The last review of the distribution of Bombus species in Hungary was completed 20 years ago. Here we present updated distribution maps based on published data from the last 20 years together with unpublished data collected in 2018–2021. Based on the new data, we examine changes in the last two decades. In the case of 9 species further studies should be carried out to confirm the presence of stable populations, while 3 species are recommended for protection by law in Hungary. Seven species showed increasing frequency, B. argillaceus and B. haematurus.

Abstract The diurnal bees, lepidopterans, and other pollinators are among the most studied flower‐visiting insect taxa. They mostly play distinct functions in temperate grasslands and ecotones of grassland‐forest mosaics (such as in forest steppes). Although orthopterans are widely distributed in these habitats, however, their flower visitation is nearly unknown, especially in the temperate zone. During the development of traps with chemical lures to catch Lepidoptera pests, large numbers of Orthoptera were caught that provide a chance for studying the flower visitation and odor and indirectly the host plant preference of seven temperate zone Tettigoniidae species. Data on the attractivity of isoamyl alcohol‐based semisynthetic lures for Meconema thalassinum and efficiency of phenylacetaldehyde‐based lures on Leptophyes albovittata and Phaneroptera falcata were reported for the first time. Additionally, analysis of nature photos collected from internet sources, as part of a passive citizen science also supports the revealed preference of these species. Based on photos, the studied orthopterans mainly visit Asteraceae species including the most preferred Tanacetum vulgare, Pulicaria dysenterica, Achillea millefolium, Solidago canadensis, and Centaurea scabiosa. Based on catches of volatile traps, the first data were recorded on the attractivity of phenylacetaldehyde‐ and isoamyl alcohol‐based lures on three temperate zone Orthoptera species. Results of a passive citizen science study strengthen these results that may increase the knowledge on the host plant and habitat preference of Orthoptera species.

The colour preference of the plum psyllid, Cacopsylla pruni (Hemiptera, Psyllidae), is yet poorly studied. This species is the only known vector of the ‘Candidatus Phytoplasma prunorum’, the agent of European stone fruit yellows (ESFY), a devastating disease of several cultivated Prunus species in Europe. As ESFY is still uncurable, vector control, thus vector monitoring, is pivotal to protect these trees. Cacopsylla pruni is a univoltine, host-shelter-alternating species; overwintered adults migrate from conifer to wild or cultivated Prunus species (family Rosaceae) in late winter-early spring. To select the most effective colour indicating the arrivals of the immigrants, yellow, fluorescent yellow, white, red and transparent sticky traps were deployed in an apricot orchard in Hungary. The two most abundant species in sticky traps were C. pruni and C. melanoneura. Catches of white traps were significantly biased towards C. pruni as compared to C. melanoneura specimens. Moreover, white sticky traps were better at catching plum psyllids than the other colours. Attraction to white was strongest when immigrants from shelter plants kept arriving in the orchard, coinciding with the blooming principal phenophase of apricot trees. When the host flowering growth stage was over, catches of C. pruni in white traps declined sharply to the level of yellow traps that was highest during this post-blooming period. We recommended white sticky traps for promptly monitoring C. pruni in apricot orchards because it is more potent and more selective than yellow ones during the critically important early flowering interval.

BACKGROUND: Two jewel beetle species native to Europe, the cypress jewel beetle, Lamprodila (Palmar, Ovalisia) festiva L. (Buprestidae, Coleoptera), and the sinuate pear tree borer, Agrilus sinuatus Olivier (Buprestidae, Coleoptera), are key pests of ornamental thuja and junipers and of orchard and ornamental rosaceous trees, respectively. Although chemical control measures are available, due to the beetles’ small size, agility, and cryptic lifestyle at the larval stage, efficient tools for their detection and monitoring are missing. Consequently, by the time emerging jewel beetle adults are noticed, the trees are typically significantly damaged. METHODS: Thus, the aim of this study was to initiate the development of monitoring traps. Transparent, light green, and purple sticky sheets and multifunnel traps were compared in field experiments in Hungary. RESULTS: Light green and transparent sticky traps caught more L. festiva and A. sinuatus jewel beetles than non-sticky multifunnel traps, regardless of the larger size of the colored surface of the funnel traps. CONCLUSIONS: Although light green sticky sheets turned out to be optimal for both species, using transparent sheets can reduce catches of non-target insects. The key to the effectiveness of sticky traps, despite their reduced suitability for quantitative comparisons, may lie in the behavioral responses of the beetles to the optical features of the traps.

During field tests implemented in Transcarpathia (West Ukraine) in 2015, 6501 specimens belonging to 26 Brachyceran fly families were collected with traps baited with generic lures (originally developed for noctuid moths) based on fermenting liquid and floral compounds. Isoamyl alcohol-based baits generally attracted more flies than phenylacetaldehyde-based baits and unbaited controls; however, the phenylacetaldehyde-based traps were the most attractive to the Empididae and Milichiidae families. The isoamyl alcohol-based semisynthetic lure showed significant attractivity to the families of Muscidae, Ulidiidae, Sarcophagidae, Calliphoridae, Sciomyzidae, Heleomyzidae, Drosophilidae, Phoridae and Platystomatidae. Additionally, isoamyl alcohol-based semisynthetic lure is the first reported attractant of the Sciomyzidae family. Since our phenylacetaldehyde-based floral lure was also attractive to Heleomyzidae flies, both types of lures can be seen as the first known attractants of this family.

Lepidopteran assemblages were studied at 16 sampling sites in the Hajdúság Region between 2013 and 2020. Although studies targeted development of synthetic phenylacetaldehyde-based and semi-synthetic isoamyl alcohol-based baits for pest monitoring, traps caught 179 species belonging to the Sphingidae, Thyatiridae, Geometridae, Erebidae and Noctuidae families. Most species were pests or widely distributed generalists, but there were also many rare habitat specialists, for example, silvicol species, whose appearance was unexpected in the recently less forested region. The specificity of the two bait types tested differed notably both on family and subfamily levels. Semi-synthetic baits performed better and attracted a wide range of noctuids belonging mainly to the Xyleninae and Noctuinae subfamilies, while synthetic phenylacetaldehyde-based lures showed specificity to Plusiinae subfamilies with lower number of sampled species. Our data fill a gap of knowledge since the fauna studied formerly was nearly unknown and brings attention to the alternative use of volatile traps of agricultural pests in faunistical studies.

ABSTRACT Green lacewings (Chrysopidae) are important predators in agroecosystems. Frequently there are studies which treat these beneficial insects at family level as a whole. This approach, despite its practical advantages, may, however, include many species with markedly different life history traits. As green lacewings are also rather diverse in their chemical ecology, treating several different species within one unit may lead to confusion and confounding effects of natural phenomena in research. Besides interspecific differences, sex-specificity is another important factor to consider in respect of insect chemical ecology. This is especially important for green lacewings, in particular the attraction of females, since oviposition is crucial in their application as biological control agents. The aim of our paper is to provide a brief insight into the diversity of the chemical ecology of green lacewings with a special emphasis on the species- and sex-specific differences in their responses to semiochemicals.

ObjectivesThis study aimed to characterise the clinicopathological features and prognostic factors of a large cohort of Hungarian patients with adrenocortical cancer diagnosed between 2000-2021.Patients and methodsThis retrospective study included seventy-four patients (27 men and 47 women) with histologically confirmed adrenocortical cancer in a single tertiary referral endocrine centre. Descriptive statistics were performed, providing summaries of selected clinical and pathological parameters. Clinicopathological factors contributing to overall survival were analysed.ResultsThe median age of patients was 48,5 years (17-84 years) at diagnosis. The majority of cases were diagnosed at ENSAT stage II (39,2%) and stage IV (33,8%). At diagnosis, the median tumour size was 9,0 cm (4,5-20 cm). In 47 patients (71,6%), the tumour was hormonally active. The median overall survival and the 5-year survival rate were 23,5 months (95% CI, 17-30,5 months) and 18,3%, respectively. Primary tumour resection was performed in 68 patients (91,8%); R0 surgical resection was achieved in 30 patients. In univariate Cox regression model, tumours with stages III and IV, high proliferative activity (Ki67-index > 10%), R1-R2 surgical resection state and hormonal activity were associated with poorer survival. Cortisol excess, both isolated and combined with androgen production, was associated with poorer survival. Fifty-five patients were treated with mitotane. Low-cost monitoring tools are needed to implement IPM in arable crops. YATLORf (Yf) traps baited with respective synthetic pheromone lures have proven to be a reliable tool for monitoring Agriotes spp., Europe’s most harmful soil pests. To optimize Yf use, we studied the effect of lure position in the trap and crop density on trap performance. Yf management detail was studied between 2000–2003 and 2014–2016 in various countries, with the traps being arranged in blocks. Each block contained one trap per treatment (i.e., lure position) under study. It was ascertained that lure attractant capacity can vary greatly with the lure’s position in the trap and the extent of vegetation. Information for making practical decisions is given. The ‘low’ lure position is suitable for all species in all field conditions, and is the best choice for A. brevis. Lures for A. brevis and A. lineatus need to be placed in the low position when the field has no or sparse vegetation cover. The ‘high’ lure position is unsuitable for A. brevis and A. obscurus, and should be considered for some species only. There are no restrictions on position for catching A. sordidus, i.e., any position is suitable. Dense vegetation (e.g., wheat) reduced the Yf trap’s potential for catching A. sordidus. Placing the trap just outside the field, or in a nearby field with bare/sparse vegetation, maintained the maximum catching potential. Vegetation density also influenced beetle sex ratio, with A. brevis and A. sordidus females always found in traps placed in fields with bare or low-density vegetation. Our findings have made it possible to obtain consistent monitoring outputs and to begin studies on multi-baiting the same trap, which can significantly reduce monitoring costs.

Apelin, as a cardiokine/myokine, is emerging as an important regulator of cardiac and skeletal muscle homeostasis. Loss of apelin signaling results in premature cardiac aging and sarcopenia. However, the contribution of apelin to peak athletic performance remains largely elusive. In this paper, we assessed the impact of maximal cardiorespiratory exercise testing on the plasma apelin levels of 58 male professional soccer players. Circulating apelin-13 and apelin-36, on average, increased transiently after a single bout of treadmill exercise; however, apelin responses (Δapelin = peak − baseline values) showed a striking interindividual variability. Baseline apelin-13 levels were inversely correlated with those of Δapelin-13 and Δapelin-36. Δapelin-13 showed a positive correlation with the maximal metabolic equivalent, relative maximal O2 consumption, and peak circulatory power, whereas such an association in the case of Δapelin-36 could not be detected. In conclusion, we observed a pronounced individual-to-individual variation in exercise-induced changes in the plasma levels of apelin-13 and apelin-36. Since changes in plasma apelin-13 levels correlated with the indicators of physical performance, whole-body oxygen consumption and pumping capability of the heart, apelin, as a novel exerkine, may be a determinant of peak athletic performance.

Monitoring insect populations is essential to optimise pest control with the correct protection timing and the avoidance of unnecessary insecticide use. Modern real-time monitoring practices use automatic insect traps, which are expected to be able to estimate the population sizes of pest animals with high species specificity. There are many solutions to overcome this challenge; however, there are only a few data that consider their accuracy under field conditions. This study presents an opto-electronic device prototype (ZooLog VARL) developed by us. A pilot field study evaluated the precision and accuracy of the data filtering using an artificial neural network(ANN) and the detection accuracy of the new probes. The prototype comprises a funnel trap, sensor-ring, and data communication system. The main modification of the trap was a blow-off device that prevented the escape of flying insects from the funnel. These new prototypes were tested in the field during the summer and autumn of 2018, detecting the daily and monthly flight of six moth species (Agrotis segetum, Autographa gamma, Helicoverpa armigera, Cameraria ohridella, Grapholita funebrana, Grapholita molesta). The accuracy of ANN was always higher than 60%. In the case of species with larger body sizes, it reached 90%. The detection accuracy ranged from 84% to 92% on average. These probes detected the real-time catches of the moth species. Therefore, weekly and daily patterns of moth flight activity periods could be compared and displayed for the different species. This device solved the problem of multiple counting and gained a high detection accuracy in target species cases. ZooLog VARL probes provide the real-time, time-series data sets of each monitored pest species. Further evaluation of the catching efficiency of the probes is needed. However, the prototype allows us to follow and model pest dynamics and may make more precise forecasts of population outbreaks.

The purpose of this study was to explore the association of the MCT1 gene Glu490Asp polymorphism (rs1049434) with athletic status and performance of endurance athletes. A total of 1,208 Brazilians (318 endurance athletes and 890 non-athletes) and 867 Europeans (315 endurance athletes and 552 non-athletes) were evaluated in a case–control approach. Brazilian participants were classified based on self-declared ethnicity to test whether the polymorphism was different between Caucasians and Afro-descendants. Moreover, 66 Hungarian athletes underwent an incremental test until exhaustion to assess blood lactate levels, while 46 Russian athletes had their maximum oxygen uptake (O2max) compared between genotypes. In the Brazilian cohort, the major T-allele was more frequent in Caucasian top-level competitors compared to their counterparts of lower competitive level (P = 0.039), and in Afro-descendant athletes compared to non-athletes (P = 0.015). Similarly, the T-allele was more frequent in European athletes (P = 0.029). Meta-analysis of the Brazilian and European cohorts confirmed that the T-allele is over-represented in endurance athletes (OR: 1.48, P = 0.03), especially when Afro-descendant athletes were included in the meta-analysis (OR: 1.58, P = 0.005). Hemoconcentration during exercise is a well-known phenomenon, however, the extent to which dehydration is involved is unclear. In our study, the effect of dehydration on exercise-induced hemoconcentration was examined in 12 elite Hungarian kayak-canoe athletes. The changes of blood markers were examined during acute maximal workload in hydrated and dehydrated states. Dehydration was achieved by exercise, during a 120-minute extensive-aerobic preload. Our research is one of the first studies in which the changes in blood components were examined with a higher time resolution and a wider range of the measured parameters. Hydration status had no effect on the dynamics of hemoconcentration during both the hydrated (HS) and dehydrated (DHS) load, although lower maximal power output were measured after the 120-minute preload [HS Hemoglobin(Hgb)Max median 17.4 (q1 17.03; q3 17.9) g/dl vs. DHS HgbMax median 16.9 (q1 16.43; q3 17.6) g/dl (n.s); HS Hematocrit(Hct)Max 53.50 (q1 52.28; q3 54.8) % vs. DHS HctMax 51.90 (q1 50.35; q3 53.93) % (n.s)]. Thirty minutes after the maximal loading, complete hemodilution was confirmed in both exercises. Dehydration had no effect on hemoconcentration or hemodilution in the recovery period [HS HgbR30' 15.7 (q1 15.15; q3 16.05) g/dl (n.s.) vs. DHS HgbR30' 15.75 (q1 15.48; q3 16.13) g/dl (n.s.), HS HctR30' 48.15 (q1 46.5; q3 49.2) % vs. DHS HctR30' 48.25 (q1 47.48; q3 49.45) % (n.s.)], however, plasma osmolality did not follow a corresponding decrease in hemoglobin and hematocrit in the dehydrated group. Based on our data, metabolic products (glucose, lactate, sodium, potassium, chloride, bicarbonate ion, blood urea nitrogen) induced osmolality may not play a major role in the regulation of hemoconcentration and post-exercise hemodilution. From our results, we can conclude that hemoconcentration depends mainly on the intensity of the exercise.

PurposeThere is abundant evidence on the benefits of physical activity on cardiovascular health. However, there are only few data on the acute effects of physical exercise on the retina and choroid. Our aim was the in vivo examination of chorioretinal alterations following short intense physical activity by spectral domain optical coherence tomography (SD-OCT).MethodsTwenty-one eyes of 21 healthy, young subjects (mean age 22.5 ± 4.1 years, 15 males and 6 females) were recruited. Macular scanning with a SD-OCT was performed before and following a vita maxima-type physical strain exercise on a rowing ergometer until complete fatigue. Follow-up OCT scans were performed 1, 5, 15, 30 and 60 minutes following the exercise. The OCT images were exported and analyzed using our custom-built OCTRIMA 3D software and the thickness of 7 retinal layers was calculated, along with semi-automated measurement of the choroidal thickness. One-way ANOVA analysis was performed followed by Dunnett post hoc test for the thickness change compared to baseline and the correlation between performance and thickness change has also been calculated. The level of significance was set at 0.001.ResultsWe observed a significant thinning of the total retina 1 minute post-exercise (-7.3 ± 0.6 μm, p < 0.001) which was followed by a significant thickening by 5 and 15 minutes (+3.6 ± 0.6 μm and +4.0 ± 0.6 μm, respectively, both p ConclusionsOur study implies that in young adults, intense physical exercise has an acute effect on the granular layers of the retina, resulting in thinning followed by rebound thickening before normalization. We could not identify any clear correlation with either choroidal changes or performance that might explain our observations, and hence the exact mechanism warrants further clarification. We believe that a combination of vascular and mechanic changes is behind the observed trends.

The urine concentrating function of the kidney is essential to maintain the water homeostasis of the human body. It is mainly regulated by the arginine-vasopressin (AVP), which targets the type 2 vasopressin receptor (V2R) in the kidney. The inability of V2R to respond to AVP stimulation leads to decreased urine concentration and congenital nephrogenic diabetes insipidus (NDI). NDI is characterized by polyuria, polydipsia, and hyposthenuria. In this study, we identified a point mutation (S127F) in the AVPR2 gene of an NDI patient, and we characterized the impaired function of the V2R mutant in HEK293 cells. Based on our data, the S127F-V2R mutant is almost exclusively located intracellularly in the endoplasmic reticulum (ER), and very few receptors were detected at the cell surface, where the receptor can bind to AVP. The overexpressed S127F-V2R mutant receptor has negligible cAMP generation capability compared to the wild-type receptor in response to AVP stimulation. Since certain misfolded mutant proteins, that are retained in the ER, can be rescued by pharmacological chaperones, we examined the potential rescue effects of two pharmacochaperones on the S127F-V2R. We found that pretreatment with both tolvaptan (an established V2R inverse agonist) and MCF14 compound (a cell-permeable high-affinity agonist for the V2R) were capable of partially restoring the cAMP generating function of the receptor in response to vasopressin stimulation. According to our data, both cell permeant agonists and antagonists can function as pharmacochaperones, and serve as the starting compounds to develop medicines for patients carrying the S127F mutation.

Abstract The search for mates and food is mediated by volatile chemicals. Insects sense food odorants and sex pheromones through odorant receptors (ORs) and pheromone receptors (PRs), which are expressed in olfactory sensory neurons. Molecular phylogenetics of ORs, informed by behavioral and functional data, generates sound hypotheses for the identification of semiochemicals driving olfactory behavior. Studying orthologous receptors and their ligands across taxa affords insights into the role of chemical communication in reproductive isolation and phylogenetic divergence. The female sex pheromone of green budworm moth Hedya nubiferana (Lepidoptera, Totricidae) is a blend of two unsaturated acetates, only a blend of both elicits male attraction. Females produce in addition codlemone, which is the sex pheromone of another tortricid, codling moth Cydia pomonella. Codlemone also attracts green budworm moth males. Concomitantly, green budworm and codling moth males are attracted to the host plant volatile pear ester. A congruent behavioral response to the same pheromone and plant volatile in two tortricid species suggests co‐occurrence of dedicated olfactory channels. In codling moth, one PR is tuned to both compounds, the sex pheromone codlemone and the plant volatile pear ester. Our phylogenetic analysis finds that green budworm moth expresses an orthologous PR gene. Shared ancestry, and high levels of amino acid identity and sequence similarity, in codling and green budworm moth PRs offer an explanation for parallel attraction of both species to the same compounds. A conserved olfactory channel for a sex pheromone and a host plant volatile substantiates the alliance of social and habitat signals in insect chemical communication. Field attraction assays confirm that in silico investigations of ORs afford powerful predictions for an efficient identification of behavior‐modifying semiochemicals, for an improved understanding of the mechanisms of host plant attraction in insect herbivores and for the further development of sustainable insect control.

The addition of synthetic eugenol and benzyl acetate to the known floral chemical and moth attractant phenylacetaldehyde synergized the attraction of Amata phegea (Lepidoptera: Amatidae). Traps baited with the ternary blend caught ca. four times more A. phegea moths than traps baited with phenylacetaldehyde alone. Both female and male moths were attracted; in a preliminary test, the female numbers caught were almost double compared to the males. Most A. phegea were caught when the blend was formulated in a dispenser with medium release rates. Traps baited with the ternary lure in polyethylene bag dispensers detected a single well-pronounced peak in seasonal trapping, suggesting that this multicomponent bisexual lure could be efficient enough to be applied to the detection and monitoring of female and male A. phegea.

Mitotane has been used for the treatment of adrenocortical cancer (ACC) for over 50 years. Despite its widespread use both in monotherapy and in combination with chemotherapeutics, our knowledge of its mechanism of action and therapeutic efficacy is scarce. The number of patients with advanced ACC who have achieved complete remission documented by detailed clinical data is below ten. We report a case of a 64-year-old woman with a non-functional ACC. Histological examination showed vascular invasion, Ki67 of 10% and a mitotic count of 3/10 high-power field. Immunohistochemistry revealed p53 positivity. Pathological TNM grade was reported as T2N0M0, ENSAT stage 2. Nine months after the initial diagnosis, re-staging CT revealed multiple peritoneal nodules, lymph node and kidney metastases confirmed by histologic examination. Mitotane monotherapy was started with a maintenance dose between 2.0 and 2.5 grams/day. Partial remission was established at six months. Subsequently, for another 12 months, each of the three-monthly CT scans confirmed complete remission. Nineteen months after the initiation of mitotane, an unexpected sudden death occurred. A detailed autopsy work-up, performed in the full awareness of oncological history, confirmed complete remission. The authors review the molecular biomarkers and clinical features reported as predictors of response to mitotane monotherapy.

There is very limited experience regarding the interventional radiological treatment of adrenocortical cancer (ACC). We present the case of a 57-year-old female patient with a large, potentially unresectable left-sided ACC and two hepatic metastases. Both liver tumors were effectively treated by trans-arterial embolization (TAE), followed by TAE of the bulky primary tumor as a life-saving intervention necessitated by severe intratumoral bleeding. Surgical removal of the primary tumor revealed complete necrosis. The patient is considered tumor free after 3.5 years. To the best of our knowledge, this is the first report to show that even a primary ACC may be completely ablated by selective embolization, and the fourth to prove the curative potential of liver TAE for ACC metastases. This case highlights the potential of selective embolization in ACC treatment.

Abstract Green lacewings (Chrysopidae) are important predators of many soft-bodied pest insects, for instance aphids. Previous studies reported attraction of Chrysoperla carnea species-complex to a ternary floral bait. The larvae of these lacewings are important generalist predators in agroecosystems, however adults are non-predatory, they feed on pollen, nectar or honeydew. Squalene, a plant originated compound was previously reported to be attractive to the nearctic Chrysopa nigricornis. In the current study squalene was tested alone and in combination with the ternary bait in field experiments in Hungary. In our experiments, traps baited with squalene attracted predatory males of Chrysopa formosa. Traps baited with squalene and the ternary floral bait attracted adults of both C. formosa and C. carnea complex lacewings. To our knowledge this is the first report of a bait combination attractive to both Chrysoperla and Chrysopa species. This finding is of special interest considering the remarkably different feeding habits of the adults of these lacewings. Potential perspectives in biological control are discussed.

Background: In the rehabilitation of patients with cardiovascular disease (CVD) and after myocardial infarction (MI), lifestyle modifications—exercise and nutritional therapy, smoking cessation, stress management—are essential and have a major, long-term impact on the overall health of patients. Methods: After MI and acute primary care, a lifestyle medicine team (medical doctors, dietitian, exercise physiologist) supervised the therapy of a 40 year-old male patient for 12 months. The program included assessments, regular medical controls, personalized diet, and exercise therapy monitored by a heart rate watch. Results: Gradual and continuous weight loss, major reduction in medication and significant improvement in fitness level, blood glucose level, and cardiac function were measured after the program. Due to these positive changes, the patient’s overall health improved to an even better level than before his MI. Conclusion: The results of this program highlight the benefits and importance of the personalized therapy and the lifestyle medicine team approach in the rehabilitation of CVD patients. Individualized and supervised lifestyle therapy should be part of the primary hospital care of CVD patients lead by medical doctors and supported by other health care providers.

The histological analysis of adrenal tumors is difficult and requires great expertise. Tissue microRNA (miRNA) expression is distinct between benign and malignant tumors of several organs and can be useful for diagnostic purposes. MiRNAs are stable and their expression can be reliably reproduced from archived formalin-fixed, paraffin-embedded (FFPE) tissue blocks. Our purpose was to assess the potential applicability of combinations of literature-based miRNAs as markers of adrenocortical malignancy. Archived FFPE tissue samples from 10 adrenocortical carcinoma (ACC), 10 adrenocortical adenoma (ACA) and 10 normal adrenal cortex samples were analyzed in a discovery cohort, while 21 ACC and 22 ACA patients were studied in a blind manner in the validation cohort. The expression of miRNA was determined by RT-qPCR. Machine learning and neural network-based methods were used to find the best performing miRNA combination models. To evaluate diagnostic applicability, ROC-analysis was performed. We have identified three miRNA combinations (hsa-miR-195 + hsa-miR-210 + hsa-miR-503; hsa-miR-210 + hsa-miR-375 + hsa-miR-503 and hsa-miR-210 + hsa-miR-483-5p + hsa-miR-503) as unexpectedly good predictors to determine adrenocortical malignancy with sensitivity and specificity both of over 90%. These miRNA panels can supplement the histological examination of removed tumors and could even be performed from small volume adrenal biopsy samples preoperatively.

Sex pheromones are commonly used in traps to monitor populations and movements of male click beetles, but to date few attractants have been identified for females. Notable exceptions are plant-derived kairomones for female Agriotes brevis and A. ustulatus, allowing the monitoring of both males and females of these species with lures containing both pheromones and plant volatiles. The attractiveness of these plant volatiles for two congeners, A. obscurus and A. lineatus, which are agricultural pests in Europe and North America, was evaluated in the current study. Both the four-component MINIM plant-derived lure for A. brevis, and the blend of (E)-anethol and (E)-cinnamaldehyde for A. ustulatus, were not attractive to A. obscurus and A. lineatus, and instead appeared to reduce captures—both when compared to blank controls, and when blended with and compared to the sex pheromones of these species. This was most pronounced in A. obscurus, where (E)-anethol and (E)-cinnamaldehyde reduced male captures by 43 and 37%, respectively. Combining the pheromones of A. obscurus and A. lineatus reduced captures of these species by 77 and 19%, respectively, compared to these pheromones singly. This suggests that attractants for female click beetles can be highly species-specific, and that the blending of pheromones of congeneric species with each other, or with plant volatiles, can reduce captures. Further research into developing such attractants for economic species is urgently needed.

Athletes are often exposed to extreme physical stress during training or competitions. The consequent activation of the hypothalamus–hypophysis–adrenal (HPA) axis results in intensified steroid hormone production in the adrenal cortex. We determined the impact of an acute extreme physical stress on adrenal and gonadal steroidogenesis in healthy male professional athletes (n = 40). The subjects underwent an extreme physical load test until total voluntary fatigue between 14:00 and 18:00 when the hormone levels are relatively stable. Blood was taken before the start (baseline), at the peak load (peak), and 30 min following completion of the exercise (recovery). The vital parameters, lactate levels, and blood levels of the 14 steroid hormones were recorded. The multivariate statistical analysis of the results revealed that all monitored hormone levels increased upon stress. Significant changes in steroid concentrations were detected at peak versus baseline, peak versus recovery, and at baseline versus recovery. The mineralocorticoid (including aldosterone and corticosterone), glucocorticoid (11-deoxycortisol and cortisol), and androgen (androstenedione, dehydroepiandrosterone, and dehydroepiandrosterone sulfate) pathways, as well as gonadal testosterone synthesis are activated simultaneously under extreme physical load. The profiling of adrenal and gonadal steroid biosynthesis in athletes may help the characterization of their loading capacity.

Abstract Background Glucocorticoid resistance is a rare, sporadic or familial condition caused by mutation of the gene encoding the glucocorticoid receptor (GR). Clinically it is characterized by symptoms developed due to local, tissue-specific, or generalized partial insensitivity to glucocorticoids. Case presentation A 31-year-old woman was evaluated because of infertility at the Endocrine Unit of the 2nd Department of Medicine, Semmelweis University. During her laboratory investigations, elevated serum and salivary cortisol were observed which failed to be suppressed after administration of 1 mg dexamethasone. 24 h urinary cortisol was increased, but a normal midnight serum cortisol was detected suggesting a maintained circadian rhythm. Plasma dehydroepiandrosterone-sulfate and androstendione levels were also elevated. Repeated plasma ACTH measurements indicated slightly elevated or normal values. Bone mineral density was normal. All laboratory results confirmed the diagnosis of glucocorticoid resistance. Genetic counseling followed by Sanger sequencing of the coding region of the gene encoding human glucocorticoid receptor was performed and a missense mutation (Arg714Gln, R714Q) in a heterozygous form was detected. Following family screening, the same mutation was found in her clinically-healthy 35-year-old sister who had no fertility problems.This variant was not detected in more than 60 patients and controls tested either for glucocorticoid resistance or Cushing’s syndrome in our Laboratory and it was absent in Exome Variant Server, HumanGene Mutation Database and ExAC databases. Conclusions Our case fulfils the diagnostic criteria of glucocorticoid resistance, also named Chrousos syndrome. The glucocorticoid receptor gene mutation detected in our patient has been already reported in a 2-year-old child with hypoglycaemia, hypokalaemia, hypertension and premature puberty. These distinct phenotypes may suggest that other factors may modify the functional consequences of the R714Q variant of GR.

(1) Background: The principal aim of our work was to identify pheromone components for Plagionotus detritus (L.) (Coleoptera: Cerambycidae), which could be exploited for developing a pheromone-based monitoring system for the complementary purposes of plant protection in areas where it might become a pest, and natural conservation in areas where it is rare or endangered. (2) Methods: Collection and analysis of headspace volatiles were carried out with field-collected beetles. Bioactive volatile compounds identified [(R)-3-hydroxyhexan-2-one and (S)-2-hydroxyoctan-3-one] from extracts of males were purchased [(±)-3-hydroxyhexan-2-one], and synthesized [(S)-2-hydroxyoctan-3-one] and field-tested. Electroantennogram assays showed that antennae of the predatory beetle Clerus mutillarius F. (Coleoptera: Cleridae) also responded to the synthetic compounds. (3) Results: A two-component aggregation-sex pheromone consisting of (R)-3-hydroxyhexan-2-one and (S)-2-hydroxyoctan-3-one was identified for P. detritus. (±)-3-hydroxyhexan-2-one and (S)-2-hydroxyoctan-3-one attracted adults of P. detritus in field bioassays. Adults of the clerid C. mutillarius also were attracted to both compounds. The cerambycid Xylotrechus antilope Schönh was significantly attracted to traps baited with (S)-2-hydroxyoctan-3-one alone or the blend containing this compound. (4) Conclusions: Our data confirmed that 3-hydroxyhexan-2-one and 2-hydroxyoctan-3-one are male-produced pheromone components for P. detritus. These results show that both intraspecific and interspecific communication may play key roles in longhorn beetle life history and ecology, with closely and more distantly related species eavesdropping on each other’s signals.

Backround: Osteoporosis is a common disease and physical activity (PA) has a favourable influence on bone status. Aim: To establish normative data for calcaneal quantitative ultrasound (QUS) bone characteristics in children and to analyse the relationships between PA, anthropometric and bone parameters. Subjects and methods: Hungarian children aged 7–19 (n = 2674; 1325 girls, 1349 boys) provided PA, anthropometric and bone data. QUS parameters were registered with Sonost3000 densitometer (speed of sound: SOS, m/s; broadband ultrasound attenuation: BUA, dB/MHz; bone quantity index (BQI = αSOS + βBUA). Reference centiles of QUS parameters were constructed by LMS method. Multivariate linear regression models were used to analyse the relationships. Results: QUS bone parameters increased with age. There were no gender differences, except in 11- and 19-year-old children, where boys showed higher values. SOS (1497.15 ± 15.72 vs 1494.05 ± 14.81 m/s) and BQI (65.31 ± 16.71 vs 62.26 ± 15.78) were higher in athletic children. The regression model revealed significant relations between SOS and age, relative muscle mass and PA; BUA correlated with gender, morphological age, BMI, relative muscle mass and PA. Conclusions: Changes in the bone parameters among 7–19-year-old Hungarian children depended on age, anthropometric dimensions and the level of PA. The normative data could be used for monitoring QUS bone parameters in children, leading to more effective interventions for healthy bones.

The attraction range of YATLORf pheromone traps to adults of four species of Agriotes (A. brevis, A. sordidus, A. litigiosus, and A. ustulatus) was studied to provide additional information about the most harmful Agriotes species in Europe. Male click beetles were marked and released at different distances from a pheromone trap. The recapture rate was calculated and analyzed using analysis of variance. The recapture rate was significantly affected by distance, species, and wind direction. The recapture rate decreased as distance increased. The majority of beetles were caught from short distances (up to 10 m) within the first five days. A. brevis, a mainly crawling species, showed the lowest recapture rate. The wind direction affected the recovery rate, with a significantly lower number of beetles moving downwind from the release points. Maximum sampling ranges and effective sampling areas were calculated. The obtained estimations were low (53 to 86 m and 509 to 2602 m2, respectively) for all the considered Agriotes species, suggesting that they were unsuitable for use as mass trapping instruments to disrupt mating. However, it seems possible to use the traps not only as monitoring tools, but also as attract-and-kill strategies for most beetle populations.

Abstract There is no available blood marker for the preoperative diagnosis of adrenocortical malignancy. The objective of this study was to investigate the expression of extracellular vesicle-associated microRNAs and their diagnostic potential in plasma samples of patients suffering from adrenocortical tumors. Extracellular vesicles were isolated either by using Total Exosome Isolation Kit or by differential centrifugation/ultracentrifugation. Preoperative plasma extracellular vesicle samples of 6 adrenocortical adenomas (ACA) and 6 histologically verified adrenocortical cancer (ACC) were first screened by Taqman Human Microarray A-cards. Based on the results of screening, two miRNAs were selected and validated by targeted quantitative real-time PCR. The validation cohort included 18 ACAs and 16 ACCs. Beside RNA analysis, extracellular vesicle preparations were also assessed by transmission electron microscopy, flow cytometry and dynamic light scattering. Significant overexpression of hsa-miR-101 and hsa-miR-483-5p in ACC relative to ACA samples has been validated. Receiver operator characteristics of data revealed dCT hsa-miR-483-5p normalized to cel-miR-39 to have the highest diagnostic accuracy (area under curve 0.965), the sensitivity and the specifity were 87.5 and 94.44, respectively. Extracellular vesicle-associated hsa-miR-483-5p thus appears to be a promising minimally invasive biomarker in the preoperative diagnosis of ACC but needs further validation in larger cohorts of patients.

The Western corn rootworm (WCR), Diabrotica virgifera virgifera LeConte (Coleoptera: Chrysomelidae), is a significant invasive pest of maize plantations in Europe. Integrated pest management demands an adequate monitoring system which detects the activity of insects with high accuracy in real-time. In this study, we show and test a new electronic device (ZooLog KLP), which was developed to detect WCR in the field. The ZooLog KLP consists of a trapping element that attracts insects with its color and species-specific sex pheromone. The other part is an opto-electronic sensor-ring which detects the specimens when they fall into the trap. At detection, the time of catch is recorded and sent to a web interface. In this study, we followed WCR flight patterns for six weeks in two locations, using ZooLog KLP probes. We investigated sensor precision by comparing the number of catches to the number of detections. The tool reached high accuracy (95.84%) in recording WCR. We found a peak in flight activity in August and a bimodal daily pattern. This method may be beneficial in detecting the WCR during their activity, and this new device may serve as a prototype for real-time monitoring systems and improve the management of this pest.

Walnut husk fly (WHF), Rhagoletis completa Cresson 1929 is an invasive species spreading quickly and damaging walnuts in Croatia and neighbouring countries. We researched distribution of this pest in the continental part of Croatia, flight dynamics in Međimurje County and its influence on quality of walnut kernels. CSALOMON®PALz traps were used for monitoring the spread and flight dynamics of R. completa. Weight and the protein content of kernels and the presence of mycotoxin contamination were measured. Walnut husk fly was found in six counties (Istria County: pest reconfirmation, Zagreb County, The City of Zagreb, Varaždin County, Međimurje County and Koprivnica-Križevci County). The presence of the fly was not confirmed on one site in Koprivnica-Križevci County (locality Ferdinandovac) and in the eastern part of Croatia (Vukovar-Srijem County: Vinkovci locality). The flight dynamics showed rapid increase in number of adults only a year after the introduction into new area. The weight of infested kernels was 5.81% lower compared to not infested. Protein content was 14.04% in infested kernels and 17.31% in not infested kernels. There was no difference in mycotoxins levels. Additional researches on mycotoxin levels in stored nuts, ovipositional preferences of walnut husk fly and protection measures against this pest are suggested.

Background: Sensorineural hearing loss (SNHL) in newborns is estimated with an incidence around 1:10,000 per year and is divided into syndromic and non-syndromic forms. In case of present retrocochlear function‚ cochlear implantation allows speech and cognitive development in affected children, comparable to that of normal hearing children. Pathogenesis of SNHL remains unclear in many cases. Imaging of the temporal bone, such as computed tomography (CT) and magnetic resonance imaging (MRI), can reveal conspicuous findings, e.g. enlarged vestibular aqueduct (EVA) and Mondini malformation (MM) of the cochlea. These malformations can be a clinical sign for Pendred syndrome. Methods: We screened CT scans of 75 cochlear implant patients for EVA and MM. Results: Six patients were observed to have either EVA alone (n=3), or MM alone (n=2), or a combination of both (n=1). Further malformations of the temporal bone could be found within the whole group, as well. Conclusion: Our results confirm the general opinion on EVA and MM, being commonly found in patients with SNHL. A possible association with Pendred syndrome needs to be confirmed by genetic investigations with search for mutations in the SLC26A4 gene and further clinical tests, such as Perchlorate test for surveillance of thyroid function.

The prevalence and spectrum of sequence alterations in the SLC26A4 gene, which codes for the anion exchanger pendrin, are population-specific and account for at least 50% of cases of non-syndromic hearing loss associated with an enlarged vestibular aqueduct. A cohort of nineteen patients from Austria with hearing loss and a radiological alteration of the vestibular aqueduct underwent Sanger sequencing of SLC26A4 and GJB2, coding for connexin 26. The pathogenicity of sequence alterations detected was assessed by determining ion transport and molecular features of the corresponding SLC26A4 protein variants. In this group, four uncharacterized sequence alterations within the SLC26A4 coding region were found. Three of these lead to protein variants with abnormal functional and molecular features, while one should be considered with no pathogenic potential. Pathogenic SLC26A4 sequence alterations were only found in 12% of patients. SLC26A4 sequence alterations commonly found in other Caucasian populations were not detected. This survey represents the first study on the prevalence and spectrum of SLC26A4 sequence alterations in an Austrian cohort and further suggests that genetic testing should always be integrated with functional characterization and determination of the molecular features of protein variants in order to unequivocally identify or exclude a causal link between genotype and phenotype.

The influence of olfactory and visual stimuli on the mating behaviour of Theresimima ampellophaga males was investigated in the field. A female model attached to a pheromone source significantly increased the number of males contacting and showing copulatory responses compared to their reaction to a pheromone stimulus alone. The pheromone stimulus was the primary factor releasing male copulatory behaviour, independent of the spatial arrangement of olfactory and visual cues. The role of the visual cues decreased with increasing distance between visual and olfactory cues. The reaction of males to pairs of models with different characteristics of conspecific females, in combination with a pheromone source placed 2 cm upwind from the center of the line between each pair of models, was compared. The size and shape of the female seemed not to be important, while the colour, posture, sex and species belongings significantly influenced the male's mating reaction.

ObjectiveOur objective was to evaluate complex hormonal response in ball game and cyclic sport elite athletes through an incremental treadmill test, since, so far, variables in experimental procedures have often hampered comparisons of data.MethodsWe determined anthropometric data, heart rate, maximal oxygen uptake, workload, plasma levels of lactate, adrenaline, noradrenaline, dopamine, cortisol, angiontensinogen and endothelin in control (n = 6), soccer (n = 8), handball (n = 12), kayaking (n = 9) and triathlon (n = 9) groups based on a Bruce protocol through a maximal exercise type of spiroergometric test.ResultsWe obtained significant increases for adrenaline, 2.9- and 3.9-fold by comparing the normalized means for soccer players and kayakers and soccer players and triathletes after/before test, respectively. For noradrenaline, we observed an even stronger, three-time significant difference between each type of ball game and cyclic sport activity.ConclusionsExercise related adrenaline and noradrenaline changes were more pronounced than dopamine plasma level changes and revealed an opportunity to differentiate cyclic and ball game activities and control group upon these parameters. Normalization of concentration ratios of the monitored compounds by the corresponding maximal oxygen uptake reflected better the differences in the response level of adrenaline, noradrenaline, dopamine and cortisol.

Purpose. The interaction of hormones of the pituitary-adrenal axis and adrenal cortex-associated circulating microRNAs is mostly unknown. We have studied the effects of dexamethasone and adrenocorticotropin on the expression of five circulating microRNAs (hsa-miR-27a, hsa-miR-200b, hsa-miR-214, hsa-miR-483-5p, and hsa-miR-503) reported to be related to the adrenal cortex in plasma samples. Methods. Expression of microRNAs was studied in plasma samples of 10 individuals examined by 1 mg dexamethasone suppression test and another 10 individuals stimulated by 250 μg tetracosactide (adrenocorticotropin). Total RNA was isolated and microRNA expression was analyzed by real-time reverse transcription quantitative polymerase chain reaction normalized to cel-miR-39 as reference. Results. Only circulating hsa-miR-27a proved to be significantly modulated in vivo by hormonal treatments: its expression was upregulated by dexamethasone whereas it was suppressed by adrenocorticotropin. Secreted hsa-miR-27a was significantly induced by dexamethasone in vitro in NCI-H295R cells, as well. The expression of hsa-miR-483-5p proposed as diagnostic marker for adrenocortical malignancy was not affected by dexamethasone or tetracosactide administration. Conclusions. hsa-miR-27a expression is modulated by hormones of the hypothalamic-pituitary-adrenal axis both in vitro and in vivo. The biological relevance of hsa-miR-27a modulation by hormones is unclear, but the responsiveness of circulating microRNAs to hormones of the pituitary-adrenal axis is noteworthy.

Molecular imaging of the 18 kD Translocator protein (TSPO) with positron emission tomography (PET) is of great value for studying neuroinflammation in rodents longitudinally. Quantification of the TSPO in rodents is, however, quite challenging. There is no suitable reference region and the use of plasma-derived input is not an option for longitudinal studies. The aim of this study was therefore to evaluate the use of the standardized uptake value (SUV) as an outcome measure for TSPO imaging in rodent brain PET studies, using [11C]PBR28. In the first part of the study, healthy male Wistar rats (n = 4) were used to determine the correlation between the distribution volume (VT, calculated with Logan graphical analysis) and the SUV. In the second part, healthy male Wistar rats (n = 4) and healthy male C57BL/6J mice (n = 4), were used to determine the test-retest variability of the SUV, with a 7-day interval between measurements. Dynamic PET scans of 63 minutes were acquired with a nanoScan PET/MRI and nanoScan PET/CT. An MRI scan was made for anatomical reference with each measurement. The whole brain VT of [11C]PBR28 in rats was 42.9 ± 1.7. A statistically significant correlation (r2 = 0.96; p < 0.01) was found between the VT and the SUV. The test-retest variability in 8 brain region ranged from 8 to 20% in rats and from 7 to 23% in mice. The interclass correlation coefficient (ICC) was acceptable to excellent for rats, but poor to acceptable for mice.The SUV of [11C]PBR28 showed a high correlation with VT as well as good test-retest variability. For future longitudinal small animal PET studies the SUV can thus be used to describe [11C]PBR28 uptake in healthy brain tissue. Based on the present observations, further studies are needed to explore the applicability of this approach in small animal disease models, with special regard to neuroinflammatory models.

At five sites in Hungary and Italy, traps baited with phenylacetaldehyde caught significantly higher numbers (10 to 100 times more) of green lacewings than unbaited traps, which demonstrates that this compound is an attractant. Traps with three bait dispensers usually caught more than those with one dispenser, but the difference was significant only at two out of five test sites. There was no difference in the numbers caught by sticky delta and funnel traps baited with phenylacetaldehyde. However, funnel traps could be adapted to catch living green lacewings. The vast majority of the specimens belonged to the Chrysoperla carnea species complex. Ch. carnea sensu lato dominated the catches at all sites. At some sites 3-11% of the insects caught were Ch. lucasina Lacroix. Phenylacetaldehyde-baited traps were attractive to both sexes, but generally more females were caught than males. Funnel traps baited with three dispensers of phenylacetaldehyde caught green lacewing adults throughout the season in Hungary.

Systematic evaluation of the potential relationship between the common genetic variants of CYP21A2 and hormone levels.The relationships of CYP21A2 intron 2 polymorphisms and haplotypes with diverse baseline and stimulated blood hormone levels were studied in 106 subjects with non-functioning adrenal incidentaloma (NFAI). The rationale for using NFAI subjects is dual: i) their baseline hormone profiles do not differ from those of healthy subjects and ii) hormone levels after stimulation tests are available.The carriers (N = 27) of a well-defined CYP21A2 haplotype cluster (c5) had significantly elevated levels of cortisol (p = 0.0110), and 17-hydroxyprogesterone (p = 0.0001) after ACTH stimulation, and 11-deoxycortisol after metyrapone administration (p = 0.0017), but the hormone values were in normal ranges. In addition, the carriers (N = 33) of the C allele of the rs6462 polymorphism had a higher baseline aldosterone level (p = 0.0006). The prevalence of these genetic variants of CYP21A2 did not differ between NFAI and healthy subjects.The common CYP21A2 variants presumably exert the same effect on hormone levels in the healthy and disease-affected populations. Therefore, they may contribute to complex diseases such as some cardiovascular diseases, and may influence the genotype-phenotype correlation in patients with congenital adrenal hyperplasia (CAH) including the individual need for hormone substitution.

BackgroundSocial psychology research on doping and outcome based evaluation of primary anti-doping prevention and intervention programmes have been dominated by self-reports. Having confidence in the validity and reliability of such data is vital.Methodology/principal findingsThe sample of 82 athletes from 30 sports (52.4% female, mean age: 21.48±2.86 years) was split into quasi-experimental groups based on i) self-admitted previous experience with prohibited performance enhancing drugs (PED) and ii) the presence of at least one prohibited PED in hair covering up to 6 months prior to data collection. Participants responded to questionnaires assessing a range of social cognitive determinants of doping via self-reports; and completed a modified version of the Brief Implicit Association Test (BIAT) assessing implicit attitudes to doping relative to the acceptable nutritional supplements (NS). Social projection regarding NS was used as control. PEDs were detected in hair samples from 10 athletes (12% prevalence), none of whom admitted doping use. This group of 'deniers' was characterised by a dissociation between explicit (verbal declarations) and implicit (BIAT) responding, while convergence was observed in the 'clean' athlete group. This dissociation, if replicated, may act as a cognitive marker of the denier group, with promising applications of the combined explicit-implicit cognitive protocol as a proxy in lieu of biochemical detection methods in social science research. Overall, discrepancies in the relationship between declared doping-related opinion and implicit doping attitudes were observed between the groups, with control measures remaining unaffected. Questionnaire responses showed a pattern consistent with self-reported doping use.Conclusions/significanceFollowing our preliminary work, this study provides further evidence that both self-reports on behaviour and social cognitive measures could be affected by some form of response bias. This can question the validity of self-reports, with reliability remaining unaffected. Triangulation of various assessment methods is recommended.

BACKGROUND: Sudden cardiac death in competitive athletes is rare but it is significantly more frequent than in the normal population. The exact cause is seldom established and is mostly attributed to ventricular fibrillation. Myocardial hypertrophy and slow heart rate, both characteristic changes in top athletes in response to physical conditioning, could be associated with increased propensity for ventricular arrhythmias. We investigated conventional ECG parameters and temporal short-term beat-to-beat variability of repolarization (STV(QT)), a presumptive novel parameter for arrhythmia prediction, in professional soccer players. METHODS: Five-minute 12-lead electrocardiograms were recorded from professional soccer players (n = 76, all males, age 22.0±0.61 years) and age-matched healthy volunteers who do not participate in competitive sports (n = 76, all males, age 22.0±0.54 years). The ECGs were digitized and evaluated off-line. The temporal instability of beat-to-beat heart rate and repolarization were characterized by the calculation of short-term variability of the RR and QT intervals. BackgroundSocial science studies of doping practices in sport rely predominantly on self-reports. Studies of psychoactive drug use indicate that self-reporting is characterised by under-reporting. Likewise doping practice is likely to be equally under-reported, if not more so. This calls for more sophisticated methods for such reporting and for independent, objective validation of its results. The aims of this study were: i) to contrast self-reported doping use with objective results from chemical hair analysis and ii) to investigate the influence of the discrepancy on doping attitudes, social projection, descriptive norms and perceived pressure to use doping.Methodology/principal findingsA doping attitudes questionnaire was developed and combined with a response latency-based implicit association test and hair sample analysis for key doping substances in 14 athletes selected from a larger sample (N = 82) to form contrast comparison groups. Results indicate that patterns of group differences in social projection, explicit attitude about and perceived pressure to use doping, vary depending on whether the user and non-user groups are defined by self-report or objectively verified through hair analysis. Thus, self-confessed users scored higher on social projection, explicit attitude to doping and perceived pressure. However, when a doping substance was detected in the hair of an athlete who denied doping use, their self-report evidenced extreme social desirability (negative attitude, low projection and low perceived pressure) and contrasted sharply with a more positive estimate of their implicit doping attitude.Conclusions/significanceHair analysis for performance enhancing substances has shown considerable potential in validating athletes' doping attitude estimations and admissions of use. Results not only confirm the need for improved self-report methodology for future research in socially-sensitive domains but also indicate where the improvements are likely to come from: as chemical validation remains expensive, a more realistic promise for large scale studies and online data collection efforts is held by measures of implicit social cognition.
