This study investigated a method to evaluate the drought tolerance stability of a genotype in a single environmental trial by capturing the plant response to irrigation changes. Genotypes that exhibit stable phenotypes under various drought stress conditions are required for stable crop production. However, considerable time and money are required to evaluate the environmental stability of a genotype through multiple environmental trials. As an index of drought tolerance stability, we calculated the coefficient of variation (CV) of shoot fresh weight of 178 soybean (Glycine max(L.) Merr.) accessions in a total of nine types of drought treatments, including changing irrigation treatments (every five or ten days) over 3-year trials. To capture the plant responses to irrigation changes, time-series multispectral (MS) data were collected, including the timings of the irrigation/non-irrigation switch in the changing irrigation treatments. We built a random regression model (RRM) for each of the nine treatments using the time-series MS data. We built a genomic prediction model (MTRRMmodel) using the genetic random regression coefficients of RRM as secondary traits and evaluated the accuracy of each model for predicting CV. In two out of the three years, the prediction accuracy of MTRRMmodels built in the changing irrigation treatment was higher than that in the continuous drought treatment in the same year. When the CV was predicted using the MTRRMmodel across years in the changing irrigation treatment, the prediction accuracy was 61% higher than that of the simple genomic prediction model. These results suggest that drought tolerance stability can be evaluated in a single environmental trial, which may reduce the time and cost of selecting genotypes with high drought tolerance stability.

The root barrier to radial O2 loss (ROL) is a key root trait preventing O2 loss from roots to anoxic soils, thereby enabling root growth into anoxic, flooded soils.We hypothesized that the ROL barrier can also prevent intrusion of hydrogen sulphide (H2S), a potent phytotoxin in flooded soils. Using H2S- and O2-sensitive microsensors, we measured the apparent permeance to H2S of rice roots, tested whether restricted H2S intrusion reduced its adverse effects on root respiration, and whether H2S could induce the formation of a ROL barrier.The ROL barrier reduced apparent permeance to H2S by almost 99%, greatly restricting H2S intrusion. The ROL barrier acted as a shield towards H2S; O2 consumption in roots with a ROL barrier remained unaffected at high H2S concentration (500 μM), compared to a 67% decline in roots without a barrier. Importantly, low H2S concentrations induced the formation of a ROL barrier.In conclusion, the ROL barrier plays a key role in protecting against H2S intrusion, and H2S can act as an environmental signalling molecule for the induction of the barrier. This study demonstrates the multiple functions of the suberized/lignified outer part of the rice root beyond that of restricting ROL.

Aerenchymatous phellem (AP) is important for internal aeration and adaptation to waterlogging in plants. Herein, the extensive accumulation of triterpenoids such as lupeol and betulinic acid was identified in AP. However, the biological and physiological roles of these triterpenoids in plants are largely unknown. Lupeol is converted from 2,3-oxidosqualene by lupeol synthase (LUS) and oxidized to betulinic acid. Functional analysis of LUS genes in soybean revealed that GmLUS1 is crucial for triterpenoid biosynthesis in AP. Lupeol and betulinic acid were found to be the major components of epicuticular wax on the surface of AP cells, and they contributed to tissue hydrophobicity and oxygen transport to roots. Additionally, the lus1 mutant produced a shallow root system due to less oxygen transport via AP under waterlogged conditions. In conclusion, triterpenoid accumulation in AP aids internal aeration and root development for adaptation to waterlogging.

Lysigenous aerenchyma is a gas space created by cortical cell death that enables efficient oxygen diffusion within plants and reduces the energy costs associated with root cells. Recent studies have demonstrated the benefits of aerenchyma formation under flooding, drought, and nutrient deficiency, although further studies will be necessary to understand the mechanisms involved.

Flooding stress caused by excessive precipitation and poor drainage threatens upland crop production and food sustainability, so new upland crop cultivars are needed with greater tolerance to soil flooding (waterlogging). So far, however, there have been no reports of highly flooding-tolerant upland crop cultivars, including maize, because of the lack of flooding-tolerant germplasm and the presence of a large number of traits affecting flooding tolerance. To achieve the goal of breeding flooding-tolerant maize cultivars by overcoming these difficulties, we chose highly flooding-tolerant teosinte germplasm. These flooding-tolerance-related traits were separately assessed by establishing a method for the accurate evaluation of each one, followed by performing quantitative trait locus (QTL) analyses for each trait using maize × teosinte mapping populations, developing introgression lines (ILs) or near-isogenic lines (NILs) containing QTLs and pyramiding useful traits. We have identified QTLs for flooding-tolerance-related root traits, including the capacity to form aerenchyma, formation of radial oxygen loss barriers, tolerance to flooded reducing soil conditions, flooding-induced adventitious root formation and shallow root angle. In addition, we have developed several ILs and NILs with flooding-tolerance-related QTLs and are currently developing pyramided lines. These lines should be valuable for practical maize breeding programs focused on flooding tolerance.

Distance-to-time conversion by using the Gompertz model compensates for the differences in root elongation rates among different genotypes and conditions, and reveals age-dependent aerenchyma formation.

Plants require oxygen for the functioning of roots, and thus the establishment of a long-distance diffusion path from above-water tissues to the submerged roots is essential to survive flooding. Rice (Oryza sativa) constitutively forms aerenchyma (gas spaces) under aerobic conditions, and induces its formation in response to low-oxygen conditions. Constitutive aerenchyma formation in rice roots is regulated by the phytohormone auxin, whereas ethylene stimulates inducible aerenchyma formation. However, the net patterns of the ethylene-dependent and -independent (auxin-dependent) aerenchyma formation remain unclear. In the present study, we used a modeling approach to determine age-dependent aerenchyma formation in the wild-type rice and reduced culm number 1 mutant, in which ethylene production is reduced, to reveal the net patterns of ethylene-dependent and -independent aerenchyma formation. Subsequent comparison of age-dependent aerenchyma formation between rice and maize roots suggested that more rapid induction of ethylene-dependent aerenchyma formation and more aerenchyma in rice roots are essential to achieve efficient oxygen diffusion under low-oxygen conditions. Moreover, rice roots showed rapid increase in the expression levels of ethylene biosynthesis and responsive genes, suggesting that the local ethylene production at an early time point after root-cell emergence contributes to the rapid induction of the ethylene-dependent aerenchyma formation in rice. Significance In recent years, phenotypic plasticity has received attention for improving plant adaptability to variable environments. For more than half a century, it has been known that rice and cereal plants develop different types of lateral roots (LRs), unlike the dicot model plant Arabidopsis. Despite the importance of plastic LR development under variable water conditions, the molecular mechanisms regulating LR types are unknown. Here, we report the regulatory mechanism of LR primordium size in rice, an important determinant of LR type. We identified two WUSCHEL-related homeobox (WOX) transcription factors that opposingly regulate LR primordium size. Our findings form the basis for improving root phenotypic plasticity for sustainable crop production under variable environments., The development of a plastic root system is essential for stable crop production under variable environments. Rice plants have two types of lateral roots (LRs): S-type (short and thin) and L-type (long, thick, and capable of further branching). LR types are determined at the primordium stage, with a larger primordium size in L-types than S-types. Despite the importance of LR types for rice adaptability to variable water conditions, molecular mechanisms underlying the primordium size control of LRs are unknown. Here, we show that two WUSCHEL-related homeobox (WOX) genes have opposing roles in controlling LR primordium (LRP) size in rice. Root tip excision on seminal roots induced L-type LR formation with wider primordia formed from an early developmental stage. QHB/OsWOX5 was isolated as a causative gene of a mutant that is defective in S-type LR formation but produces more L-type LRs than wild-type (WT) plants following root tip excision. A transcriptome analysis revealed that OsWOX10 is highly up-regulated in L-type LRPs. OsWOX10 overexpression in LRPs increased the LR diameter in an expression-dependent manner. Conversely, the mutation in OsWOX10 decreased the L-type LR diameter under mild drought conditions. The qhb mutants had higher OsWOX10 expression than WT after root tip excision. A yeast one-hybrid assay revealed that the transcriptional repressive activity of QHB was lost in qhb mutants. An electrophoresis mobility shift assay revealed that OsWOX10 is a potential target of QHB. These data suggest that QHB represses LR diameter increase, repressing OsWOX10. Our findings could help improve root system plasticity under variable environments.

With the widespread use of high-throughput phenotyping systems, growth process data are expected to become more easily available. By applying genomic prediction to growth data, it will be possible to predict the growth of untested genotypes. Predicting the growth process will be useful for crop breeding, as variability in the growth process has a significant impact on the management of plant cultivation. However, the integration of growth modeling and genomic prediction has yet to be studied in depth. In this study, we implemented new prediction models to propose a novel growth prediction scheme. Phenotype data of 198 soybean germplasm genotypes were acquired for 3 years in experimental fields in Tottori, Japan. The longitudinal changes in the green fractions were measured using UAV remote sensing. Then, a dynamic model was fitted to the green fraction to extract the dynamic characteristics of the green fraction as five parameters. Using the estimated growth parameters, we developed models for genomic prediction of the growth process and tested whether the inclusion of the dynamic model contributed to better prediction of growth. Our proposed models consist of two steps: first, predicting the parameters of the dynamics model with genomic prediction, and then substituting the predicted values for the parameters of the dynamics model. By evaluating the heritability of the growth parameters, the dynamic model was able to effectively extract genetic diversity in the growth characteristics of the green fraction. In addition, the proposed prediction model showed higher prediction accuracy than conventional genomic prediction models, especially when the future growth of the test population is a prediction target given the observed values in the first half of growth as training data. This indicates that our model was able to successfully combine information from the early growth period with phenotypic data from the training population for prediction. This prediction method could be applied to selection at an early growth stage in crop breeding, and could reduce the cost and time of field trials.

Multi-spectral (MS) imaging enables the measurement of characteristics important for increasing the prediction accuracy of genotypic and phenotypic values for yield-related traits. In this study, we evaluated the potential application of temporal MS imaging for the prediction of above-ground biomass (AGB) and determined which developmental stages should be used for accurate prediction in soybean. Field experiments with 198 accessions of soybean were conducted with four different irrigation levels. Five vegetation indices (VIs) were calculated using MS images from soybean canopies from early to late growth stages. To predict the genotypic values of AGB, VIs at the different growth stages were used as secondary traits in a multi-trait genomic prediction. The accuracy of the prediction model increased starting at an early stage of growth (31 days after sowing). To predict phenotypic values of AGB, we employed multi-kernel genomic prediction. Consequently, the prediction accuracy of phenotypic values reached a maximum at a relatively early growth stage (38 days after sowing). Hence, the optimal timing for MS imaging may depend on the irrigation levels.

A maize introgression line is developed harboring QTLs from Zea nicaraguensis for constitutive aerenchyma, which allows oxygen to diffuse along roots immediately after waterlogging and contributes to oxygen deficiency tolerance., Zea nicaraguensis is a wild relative of Zea mays subsp. mays (maize) that has high waterlogging tolerance. One of its traits is constitutive aerenchyma formation (CAF) in roots and this may be one of the reasons for the tolerance, but it has not yet been proven by comparing plants that differ only in CAF in the same genetic background. We therefore produced an introgression line AE24-50-44-91 (IL-AE91) possessing four quantitative trait loci for CAF from Z. nicaraguensis in the background of maize (inbred line Mi29). The degree of root CAF in IL-AE91 was intermediate between that of Mi29 and Z. nicaraguensis. Seedlings of IL-AE91 grown aerobically were more tolerant to transfer to oxygen-deficient conditions than were Mi29 seedlings. On day 2 of oxygen deficiency, the root extension rate and viability of root-tip cells in IL-AE91 were ~2.7 and ~1.3 times greater, respectively, than they were in Mi29. On day 4, the area of aerenchyma at 80 mm from the root tips was ~1.5 times greater in IL-AE91 and radial oxygen loss from the apical parts of roots was ~3.4 times higher than in Mi29. These results demonstrate that CAF reduces the stress from low external oxygen levels caused by soil waterlogging.

Waterlogged soils contain monocarboxylic acids produced by anaerobic microorganisms. These “organic acids” can accumulate to phytotoxic levels and promote development of a barrier to radial O2 loss (ROL) in roots of some wetland species. Environmental cues triggering root ROL barrier induction, a feature that together with tissue gas-filled porosity facilitates internal aeration, are important to elucidate for knowledge of plant stress physiology. We tested the hypothesis that comparatively low, non-toxic, concentrations of acetic, propionic, butyric, and/or hexanoic acids might induce root ROL barrier formation in rice. Each organic acid, individually, triggered the ROL barrier in roots but with no effect (acetic or butyric acids) or with only slight effects (propionic or hexanoic acids) on root extension. Transcripts of four genes related to suberin biosynthesis were increased by some of the organic acid treatments. Respiration in root tissues was not, or moderately, inhibited. Beyond a narrow concentration range, however, respiration declined exponentially and the order (least to greatest) for EC50 (effective concentration for 50% inhibition) was butyric, propionic, acetic, then hexanoic acid. An understanding of the environmental cue for root ROL barrier induction should enhance future work to elucidate the molecular regulation of this root trait contributing to plant flooding tolerance.

In flooded soils, an efficient internal aeration system is essential for root growth and plant survival. Roots of many wetland species form barriers to restrict radial O2 loss (ROL) to the rhizosphere. The formation of such barriers greatly enhances longitudinal O2 diffusion from basal parts towards the root tip, and the barrier also impedes the entry of phytotoxic compounds produced in flooded soils into the root. Nevertheless, ROL from roots is an important source of O2 for rhizosphere oxygenation and the oxidation of toxic compounds. In this paper, we review the methodological aspects for the most widely used techniques for the qualitative visualization and quantitative determination of ROL from roots. Detailed methodological approaches, practical set-ups and examples of ROL from roots with or without barriers to ROL are included. This paper provides practical knowledge relevant to several disciplines, including plant–soil interactions, biogeochemistry and eco-physiological aspects of roots and soil biota.

(1) Drought and flooding are contrasting abiotic stressors for plants. Evidence is accumulating for root anatomical traits being essential for the adaptation to drought or flooding. However, an integrated approach to comprehensively understand root anatomical traits has not yet been established. (2) Here we analysed the root anatomical traits of 18 wild Poaceae species differing in adaptation to a range of soil water content. Regression model analyses revealed the optimal anatomical traits that were required by the plants to adapt to low or high soil water content. (3) While the area and number of each root tissue (e.g. stele, cortex, xylem or aerenchyma) were not strongly correlated to the soil water content, the ratio of the root tissue areas [cortex to stele ratio (CSR), xylem to stele ratio (XSR) and aerenchyma to cortex ratio (ACR)] could fully explain the adaptations of the wild Poaceae species to the soil water gradients. (4) Our results demonstrate that the optimal anatomical traits for the adaptations to soil water content can be determined by three indices (i.e. CSR, XSR and ACR), and thus we propose that these root anatomical indices can be used to improve the tolerance of crops to drought and flooding stresses.

By applying genomic prediction to growth data, it will be possible to predict the growth of untested genotypes. Predicting the growth process will be useful for crop breeding, as variability in the growth process has a significant impact on the management of plant cultivation. Laser microdissection (LM) is a rapid, easy, one-step, and efficient method of conserving and isolating single cell or cell clusters from fixed tissue sections under direct microscopic visualization. LM is currently the only method that can be used to isolate homogeneous cells from heterogeneous tissue. This method was first developed for tumor analysis. Multiple generations of LM instruments are available on the market. For instance, the Veritas™ microdissection combines the LM system based on the IR with UV laser cutting. The desired cells can be harvested specific and wanted cells can be harvested directly by cutting target cells abroad from unwanted cells. The extracted cells are then utilized in various disciplines of experimental and clinical biology, such as genomic analysis and proteomics. LM is a an efficient and powerful tool that facilitates the analysis of small amounts of molecules isolated from a complex tissue, thus offering new opportunities to understand physiological and fundamental processes. Here, we describe a method of preparing paraffin sections of maize roots for laser microdissection to three parts (stele, cortex and outer layers) by LM using a microwave embedding method. This approach allows RNA to be extracted from each type of tissue separately. The RNA integrity of our samples following LM ranged between 7 and 7.2, indicating that this RNA can be reliably used for further analysis. The work reported in this paper highlights how advances in protocols and methods have made LM a powerful and sometimes essential tool for genomic and proteomic analyses of tiny amounts of biomolecules extracted from a few cells isolated from a complex tissue in their physiological context, thus offering a new pathway to understand fundamental, physiological and/or patho-physiological processes.

Background and Aims Dutch tomato cultivars tend to have a greater yield than Japanese cultivars even if they are grown under the same conditions. Factors contributing to the increased yield of the Dutch cultivars were a greater light use efficiency and greater leaf photosynthetic rate. On the other hand, the relationship between tomato yields and anatomical traits is still unclear. The aim of this study is to identify the anatomical traits related to the difference in yield between Dutch and Japanese cultivars. Methods Anatomical properties were compared during different growth stages of Dutch and Japanese tomatoes. Hormone profiles and related gene expression in hypocotyls of Dutch and Japanese cultivars were compared in the hypocotyls of 3- and 4-week-old plants. Key results Dutch cultivars have a more developed secondary xylem than Japanese cultivars, which would allow for greater transport of water, mineral nutrients and phytohormones to the shoots. The areas and ratios of the xylem in the hypocotyls of 3- to 6-week-old plants were larger in the Dutch cultivars. In reciprocal grafts of the Japanese and Dutch cultivars, xylem development at the scion and rootstock depended on the scion cultivar, suggesting that some factors in the scion are responsible for the difference in xylem development. The cytokinin content, especially the level of N6-(Δ 2-isopentenyl) adenine (iP)-type cytokinin, was higher in the Dutch cultivars. This result was supported by the greater expression of Sl-IPT3 (a cytokinin biosynthesis gene) and Sl-RR16/17 (a cytokinin-responsive gene) in the Dutch cultivars. Conclusions These results suggest that iP-type cytokinins, which are locally synthesized in the hypocotyl, contribute to xylem development. The greater xylem development in Dutch cultivars might contribute to the high yield of the tomato.

Internal oxygen diffusion from shoot to root tips is enhanced by the formation of aerenchyma (gas space) in waterlogged soils. Lysigenous aerenchyma is created by programmed cell death and subsequent lysis of the root cortical cells. Rice (Oryza sativa) forms aerenchyma constitutively under aerobic conditions and increases its formation under oxygen-deficient conditions. Recently, we have demonstrated that constitutive aerenchyma formation is regulated by auxin signaling mediated by Auxin/indole-3-acetic acid protein (AUX/IAA, IAA). While ethylene is involved in inducible aerenchyma formation, the relationship of auxin and ethylene during aerenchyma formation remains unclear. Here, we examined the effects of oxygen deficiency and ethylene on aerenchyma formation in the roots of a rice mutant (iaa13) in which auxin signaling is suppressed by a mutation in the degradation domain of IAA13 protein. The results showed that AUX/IAA-mediated auxin signaling contributes to ethylene-dependent inducible aerenchyma formation in rice roots. An auxin transport inhibitor abolished aerenchyma formation under oxygen-deficient conditions and reduced the expression of genes encoding ethylene biosynthesis enzymes, further supporting the idea that auxin is involved in ethylene-dependent inducible aerenchyma formation. Based on these studies, we propose a mechanism that underlies the relationship between auxin and ethylene during inducible aerenchyma formation in rice roots.

Previously, we cloned the cDNAs of FLOWERING LOCUS T (FT) and TERMINAL FLOWER 1 (TFL1) homologs FaFT1 and FaTFL1-1, respectively, from domesticated octoploid strawberries (Fragaria × ananassa Duch.). We produced Arabidopsis transgenic lines containing FaFT1 or FaTFL1-1 to understand their function. We placed the introduced genes under the control of the Cauliflower mosaic virus (CaMV) 35S promoter, and found that FaFT1 over-expression caused no early flowering except for one line at short day conditions, but aerial rosette phenotype instead. From the results and its expression pattern previously investigated, it was speculated that the FaFT1 acts at the vegetative propagation stage including runner formation, but not at the flower inductive stage. In contrast, the over-expression of FaTFL1-1 rescued the tfl1-2 mutant, suggesting that it possesses flower-suppressing ability. From its spatial expression pattern in crowns (stem tissue including meristem), FaTFL1-1 seemed to be related to flower inducing at apical portion of crowns by its down regulation, and to flower suppressing at the basal portion of crowns by its up regulation.

Premise of the study The aboveground part of one-leaf plants (Gesneriaceae) consists of a unique shoot-like unit called a phyllomorph, which is composed of a growing lamina and one petiolode (a stem- and petiole-like organ). The phyllomorph has three meristems: a basal meristem (BM; involved in lamina growth), a petiolode meristem (PM; required for midrib and petiolode growth), and a groove meristem (GM; contributes to inflorescence and new phyllomorph development). Although the GM has a tunica-corpus structure like a conventional shoot apical meristem (SAM), the vegetative GM of a one-leaf plant does not form any organ primordia and has been considered a defective or suppressed SAM. In this study, we evaluated the nature of the three meristems to reconsider their roles in phyllomorph development in the one-leaf species Monophyllaea glabra. Methods Mitotic activities of meristem cells were monitored by incorporation of a thymidine analog (EdU) into DNA. Orthologs of SHOOTMERISTEMLESS (STM) and PHANTASTICA, ROUGH SHEATH2, ASYMMETRIC LEAVES1 (PHAN/RS2/AS1) were isolated from M. glabra, and their expression patterns were investigated by quantitative reverse transcription polymerase chain reaction in combination with a laser microdissection technique. Key results Mitotic activities were detected in all three phyllomorph meristems. Strong expression of the STM and PHAN/RS2/AS1 orthologs was detected in the vegetative GM and BM, respectively. Conclusions The vegetative GM is an active meristem that expresses the STM ortholog. We postulate that the GM is required for growth of a phyllomorph by providing undifferentiated cells and/or growth regulators to the BM and/or PM.

Plants typically respond to waterlogging by producing new adventitious roots with aerenchyma and many wetland plants form a root barrier to radial O2 loss (ROL), but it was not known if this was also the case for lateral roots. We tested the hypothesis that lateral roots arising from adventitious roots can form a ROL barrier, using root-sleeving electrodes and O2 microsensors to assess ROL of Zea nicaraguensis, the maize (Zea mays ssp. mays) introgression line with a locus for ROL barrier formation (introgression line (IL) #468) from Z. nicaraguensis and a maize inbred line (Mi29). Lateral roots of Z. nicaraguensis and IL #468 both formed a ROL barrier under stagnant, deoxygenated conditions, whereas Mi29 did not. Lateral roots of Z. nicaraguensis had higher tissue O2 status than for IL #468 and Mi29. The ROL barrier was visible as suberin in the root hypodermis/exodermis. Modelling showed that laterals roots can grow to a maximum length of 74 mm with a ROL barrier, but only to 33 mm without a barrier. Presence of a ROL barrier in lateral roots requires reconsideration of the role of these roots as sites of O2 loss, which for some species now appears to be less than hitherto thought.

Flooding causes oxygen deprivation in soils. Plants adapt to low soil oxygen availability by changes in root morphology, anatomy, and architecture to maintain root system functioning. Essential traits include aerenchyma formation, a barrier to radial oxygen loss, and outgrowth of adventitious roots into the soil or the floodwater. We highlight recent findings of mechanisms of constitutive aerenchyma formation and of changes in root architecture. Moreover, we use modelling of internal aeration to demonstrate the beneficial effect of increasing cortex-to-stele ratio on sustaining root growth in waterlogged soils. We know the genes for some of the beneficial traits, and the next step is to manipulate these genes in breeding in order to enhance the flood tolerance of our crops.

Self-incompatibility (SI) is a sophisticated system for pollen selectivity to prevent pollination by genetically identical pollen. In Brassica, it is genetically controlled by a single, highly polymorphic S-locus, and the male and female S-determinant factors have been identified as S-locus protein 11 (SP11)/S-locus cysteine-rich protein (SCR) and S-locus receptor kinase (SRK), respectively. However, the overall molecular system and identity of factors in the downstream cascade of the SI reaction remain unclear. Previously, we identified a self-compatible B. rapa mutant line, TSC28, which has a disruption in an unidentified novel factor of the SI signaling cascade. Here, in a genetic analysis of TSC28, using an F2 population from a cross with the reference B. rapa SI line Chiifu-401, the causal gene was mapped to a genetic region of DNA containing markers BrSA64 and ACMP297 in B. rapa chromosome A1. By fine mapping using an F2 population of 1,034 plants, it was narrowed down to a genetic region between DNA markers ACMP297 and BrgMS4028, with physical length approximately 1.01 Mbp. In this genomic region, 113 genes are known to be located and, among these, we identified 55 genes that were expressed in the papilla cells. These are candidates for the gene responsible for the disruption of SI in TSC28. This list of candidate genes will contribute to the discovery of a novel downstream factor in the SP11-SRK signaling cascade in the Brassica SI system.

Asymmetric cell division is a key step in cellular differentiation in multicellular organisms. In plants, asymmetric zygotic division produces the apical and basal cells. Mitogen-activated protein kinase (MPK) cascade in Arabidopsis act in asymmetric divisions such as zygotic division and stomatal development, but whether the effect on cellular differentiation by this cascade is direct or indirect following asymmetric division is not clear. Here, we report the analysis of a rice mutant, globular embryo 4 (gle4). In two- and four-cell-stage embryos, asymmetric zygotic division and subsequent cell division patterns were indistinguishable between the wild type and gle4 mutants. However, marker gene expression and transcriptome analyses showed that specification of the basal region was compromised in gle4. We found that GLE4 encodes MPK6 and GLE4/ MPK6 is essential in cellular differentiation rather than in asymmetric zygotic division. Our findings provide a new insight into the role of MPK in plant development. We propose that the regulation of asymmetric zygotic division is separate from the regulation of cellular differentiation that leads to apical-basal polarity.

In roots of gramineous plants, lysigenous aerenchyma is created by the death and lysis of cortical cells. Rice (Oryza sativa) constitutively forms aerenchyma under aerobic conditions, and its formation is further induced under oxygen-deficient conditions. However, maize (Zea mays) develops aerenchyma only under oxygen-deficient conditions. Ethylene is involved in lysigenous aerenchyma formation. Here, we investigated how ethylene-dependent aerenchyma formation is differently regulated between rice and maize. For this purpose, in rice, we used the reduced culm number1 (rcn1) mutant, in which ethylene biosynthesis is suppressed. Ethylene is converted from 1-aminocyclopropane-1-carboxylic acid (ACC) by the action of ACC oxidase (ACO). We found that OsACO5 was highly expressed in the wild type, but not in rcn1, under aerobic conditions, suggesting that OsACO5 contributes to aerenchyma formation in aerated rice roots. By contrast, the ACO genes in maize roots were weakly expressed under aerobic conditions, and thus ACC treatment did not effectively induce ethylene production or aerenchyma formation, unlike in rice. Aerenchyma formation in rice roots after the initiation of oxygen-deficient conditions was faster and greater than that in maize. These results suggest that the difference in aerenchyma formation in rice and maize is due to their different mechanisms for regulating ethylene biosynthesis.

Heat stress occurrence during seed filling leads to the formation of a chalky portion in the limited zone of the starchy endosperm of rice grains. In this study, isolation of aleurone, dorsal, central and lateral tissues of developing endosperm by laser-microdissection (LM) coupled with gene expression analysis of a 44 K microarray was performed to identify key regulatory genes involved in the formation of milky-white (MW) and white-back (WB) grains during heat stress. Gene regulatory network analysis classified the genes changed under heat stress into five modules. The most distinct expression pattern was observed in modules where most of the small heat shock proteins and cellular organization genes were changed under heat stress in dorsal aleurone cells and dorsal starchy endosperm zones. The histological observation supported the significant increase in cell number and size of dorsal aleurone cells in WB grains. With regard to the central starchy endosperm zone, preferential down-regulation of high molecular weight heat shock proteins (HMW HSPs), including a prominent member encoding endoplasmic reticulum (ER) chaperones, by heat stress was observed, while changes in expression of starch biosynthesis genes were minimal. Characterization of transgenic plants suppressing endosperm lumenal binding protein gene (BiP1), an ER chaperone preferentially down-regulated at the MW zone under heat stress, showed evidence of forming the chalky grains without disturbing the expression of starch biosynthesis genes. The present LM-based comprehensive expression analysis provides novel inferences that HMW HSPs play an important role in controlling redox, nitrogen and amino acid metabolism in endosperm leading to the formation of MW and WB chalky grains under heat stress.

BACKGROUND AND AIMS: Soil waterlogging often causes oxygen deficiency in the root systems of plants and severely inhibits plant growth. Formation of aerenchyma – interconnected spaces that facilitate the movement of gases between and within the aerial and submerged parts of plants – is an adaptive trait for coping with waterlogged conditions. Soybean (Glycine max) forms porous secondary tissues known as aerenchymatous phellem (AP), which are derived from the outermost cell layer of phellogen. To understand what factors other than waterlogging are involved in phellogen and AP formation, we examined how their formation in soybean seedlings was affected by darkness, CO(2) deficiency and blockage of phloem transport. METHODS: Aerenchymatous phellem and phellogen formation were expressed as area ratios in cross-sections of hypocotyl. CO(2) was depleted by use of calcium oxide and sodium hydroxide. Phloem transport was blocked by heat-girdling of hypocotyls. Sucrose levels were measured by spectrophotometry. KEY RESULTS: Under light conditions, waterlogging induced the accumulation of high concentrations of sucrose in hypocotyls, followed by phellogen and AP formation in hypocotyls. Phellogen formation and AP formation were inhibited by darkness, CO(2) deficiency and blockage of phloem transport. Phellogen formation and AP formation were also inhibited by excision of shoots above the epicotyl, but they recovered following application of sucrose (but not glucose or fructose application) to the cut surface. CONCLUSIONS: The results demonstrate that sucrose derived from leaves is essential for AP and phellogen formation in soybean hypocotyls under waterlogged soil conditions. Maintenance of a high sucrose concentration is thus essential for the development of phellogen and AP and the differentiation of phellogen to AP.

Knowledge of the genetic regulation of adventitious roots, aerenchyma, and radial oxygen loss barrier formation, and the signaling for acclimation, will assist the development of waterlogging-tolerant crops.

Seed germination rates and plant development and growth under abiotic stress are important aspects of crop productivity. Here, our characterization of the rice (Oryza sativa L.) mutant reduced culm number11 (rcn11) showed that RCN11 controls growth of plants exposed to abnormal temperature, salinity and drought conditions. RCN11 also mediates root aerenchyma formation under oxygen-deficient conditions and ABA sensitivity during seed germination. Molecular studies showed that the rcn11 mutation resulted from a 966-bp deletion that caused loss of function of β1,2-xylosyltransferase (OsXylT). This enzyme is located in the Golgi apparatus where it catalyzes the transfer of xylose from UDP-xylose to the core β-linked mannose of N-glycans. RCN11/OsXylT promoter activity was observed in the basal part of the shoot containing the shoot and axillary meristems and in the base of crown roots. The level of RCN11/OsXylT expression was regulated by multiple phytohormones and various abiotic stresses suggesting that plant specific N-glycosylation is regulated by multiple signals in rice plants. The present study is the first to demonstrate that rice β1,2-linked xylose residues on N-glycans are critical for seed germination and plant development and growth under conditions of abiotic stress.
