In the present article, we describe a mechanistic study of a novel derivative of N-amidino-substituted benzimidazo[1,2-α]quinoline in two human colorectal cancer cell lines differing in p53 gene status. We used a proteomic approach based on two-dimensional gel electrophoresis coupled with mass spectrometry to complement the results obtained by common molecular biology methods for analyzing cell proliferation, cell cycle, and apoptosis. Tested quinoline derivative inhibited colon cancer cell growth, whereby p53 gene status seemed to be critical for its differential response patterns. DNA damage and oxidative stress are likely to be the common triggers of molecular events underlying its antiproliferative effects. In HCT 116 (wild-type p53), this compound induced a p53-dependent response resulting in accumulation of the G1- and S-phase cells and induction of apoptosis via both caspase-3-dependent and caspase-independent pathways. Quinoline derivative triggered transient, p53-independent G2-M arrest in mutant p53 cells (SW620) and succeeding mitotic transition, whereby these cells underwent cell death probably due to aberrant mitosis (mitotic catastrophe). Proteomic approach used in this study proved to be a valuable tool for investigating cancer cell response to newly synthesized compound, as it specifically unraveled some molecular changes that would not have been otherwise detected (e.g., up-regulation of the p53-dependent chemotherapeutic response marker maspin in HCT 116 and impairment in ribosome biogenesis in SW620). Finally, antiproliferative effects of tested quinoline derivative on SW620 cells strongly support its possible role as an antimetastatic agent and encourage further in vivo studies on the chemotherapeutic potential of this compound against colorectal carcinoma. Delphi consensus consultation methods and community-based participatory research (CBPR) are distinct approaches that have traditionally been employed separately. This paper explores the integration of Delphi methods with CBPR in a research project that sought to identify effective self-management strategies for bipolar disorder (BD). We introduce our Canadian-based network which specializes in CBPR in BD, and outline the key principles of CBPR approaches. Delphi consensus consultation methods are described and we present the five phases of our Delphi consensus consultation project, conducted within a CBPR framework. Examples of how each project phase incorporated the principles of CBPR are provided, as are personal reflections of community members involved in the project, and broader reflections on challenges commonly encountered in CBPR projects.

Sustainable transport is an increasing focus for planning authorities, and New Zealand's local authorities have been developing initiatives to encourage more sustainable forms of mobility. This study focuses on a cycling initiative in a provincial local authority that extensively relied on a monitoring and evaluation (M&E) programme. We argue that M&E programmes are often black boxed and there is little understanding of how they work within organisations. Drawing on work from the sociology of quantification, this study examines the multiple translations that the M&E underwent and the actors involved in it, in order for it to become useful and accessible.

Desmitis of the collateral ligament of the distal interphalangeal joint is a cause of lameness in performance horses. The objective of this prospective, experimental, ex vivo feasibility study was to evaluate the success of ultrasound-guided injection of the collateral ligaments of the distal interphalangeal joint in the equine forelimb. Seventy-six ultrasound-guided dye injections of the collateral ligament of the distal interphalangeal joint were performed on horses' cadaver limbs. The hooves were sectioned transversely to verify the location of the dye relative to the collateral ligaments and surrounding structures. Evaluations of transverse sections were performed independently by two experienced observers. A scoring system was used to assess injection of the collateral ligament of the distal interphalangeal joint at the proximal, middle, and distal aspect over the length of the ligament. The collateral ligament was injected at any point in 97.4% of cases. The ligament was injected over the entire scored length in 43.2% of cases (32/74), over two scored length areas in 45.9% of cases (34/74), and in one area in 10.8% of cases (8/74). The distal interphalangeal joint and the common digital extensor tendon were also injected in 81.6% (62/76) and 43.4% (33/76) of the cases, respectively. Use of the ultrasound had a positive and negative predictive value of 98% and 9%, respectively. In this study, ultrasound guidance was useful for confirming injection of the collateral ligament of the distal interphalangeal joint but did not prevent injecting the distal interphalangeal joint and the common digital extensor tendon.

Summary Understanding the mechanisms for cellular aging is a fundamental question in biology. Normal red blood cells (RBCs) survive for approximately 100 days, and their survival is likely limited by functional decline secondary to cumulative damage to cell constituents, which may be reflected in altered metabolic capabilities. To investigate metabolic changes during in vivo RBC aging, labeled cell populations were purified at intervals and assessed for abundance of metabolic intermediates using mass spectrometry. A total of 167 metabolites were profiled and quantified from cell populations of defined ages. Older RBCs maintained ATP and redox charge states at the cost of altered activity of enzymatic pathways. Time-dependent changes were identified in metabolites related to maintenance of the redox state and membrane structure. Enhanced recovery after surgery (ERAS) programs provide a format for multidisciplinary care and has been shown to predictably improve short term outcomes associated with surgical procedures. Esophagectomy has historically been associated with significant levels of morbidity and mortality and as a result routine application and audit of ERAS guidelines specifically designed for esophageal resection has significant potential to improve outcomes associated with this complex procedure.A team of international experts in the surgical management of esophageal cancer was assembled and the existing literature was identified and reviewed prior to the production of the guidelines. Well established procedure specific components of ERAS were reviewed and updated with changes relevant to esophagectomy. Procedure specific, operative and technical sections were produced utilizing the best current level of evidence. All sections were rated regarding the level of evidence and overall recommendation according to the evaluation (GRADE) system.Thirty-nine sections were ultimately produced and assessed for quality of evidence and recommendations. Some sections were completely new to ERAS programs due to the fact that esophagectomy is the first guideline with a thoracic component to the procedure.The current ERAS society guidelines should be reviewed and applied in all centers looking to improve outcomes and quality associated with esophageal resection.

Cnk1 (connector enhancer of kinase suppressor of Ras 1) is a pleckstrin homology (PH) domain–containing scaffold protein that increases the efficiency of Ras signaling pathways, imparting efficiency and specificity to the response of cell proliferation, survival, and migration. Mutated KRAS (mut-KRAS) is the most common proto-oncogenic event, occurring in approximately 25% of human cancers and has no effective treatment. In this study, we show that selective inhibition of Cnk1 blocks growth and Raf/Mek/Erk, Rho and RalA/B signaling in mut-KRAS lung and colon cancer cells with little effect on wild-type (wt)-KRAS cells. Cnk1 inhibition decreased anchorage-independent mut-KRas cell growth more so than growth on plastic, without the partial "addiction" to mut-KRAS seen on plastic. The PH domain of Cnk1 bound with greater affinity to PtdIns(4,5)P2 than PtdIns(3,4,5)P3, and Cnk1 localized to areas of the plasma membranes rich in PtdIns, suggesting a role for the PH domain in the biological activity of Cnk1. Through molecular modeling and structural modification, we identified a compound PHT-7.3 that bound selectively to the PH domain of Cnk1, preventing plasma membrane colocalization with mut-KRas. PHT-7.3 inhibited mut-KRas, but not wild-type KRas cancer cell and tumor growth and signaling. Thus, the PH domain of Cnk1 is a druggable target whose inhibition selectively blocks mutant KRas activation, making Cnk1 an attractive therapeutic target in patients with mut-KRAS–driven cancer. Significance: These findings identify a therapeutic strategy to selectively block oncogenic KRas activity through the PH domain of Cnk1, which reduces its cell membrane binding, decreasing the efficiency of Ras signaling and tumor growth.

VC-01 is a novel cell replacement therapy being developed as a potential long-term diabetes therapy to more effectively control glycemia, reduce hypoglycemia risk and mitigate diabetes-related complications. VC-01 is derived from directed differentiation of human embryonic stem cells to pancreatic progenitor cells, which are loaded into a durable, retrievable delivery device and then implanted subcutaneously. Once implanted and matured, the cells secrete insulin and other islet hormones in response to blood glucose levels. The delivery device is immunoprotective to implanted cells. VC-01 is currently being evaluated in a 24-month open-label, dose-escalating Phase 1/2 study in T1D patients with minimal insulin-producing β-cell function. Cohort 1 has evaluated product at a subtherapeutic dose to assess initial safety and tolerability and establish implantation techniques. To date, 19 subjects have been implanted and evaluated throughout implantation duration until final product explant. Product has been safe and well tolerated; adverse events have been relatively minor and generally related to the surgical procedure. The delivery device has provided protection against the host adaptive immune system with no evidence of allogeneic or autoimmune rejection or sensitization based upon a sensitive panel of immune function tests. No off-target growth of donor cells has been detected. The potential for prolonged cell survival has been demonstrated, as long as 24 months, but has been inconsistent among subjects and primarily limited by a foreign body response to the device component. Positive immunohistochemical staining for pancreatic islet cell markers NKX6-1, insulin, and glucagon has been observed. These preliminary results demonstrate the immune-protective nature of the encapsulation device and safety of this human stem cell-derived product, but also indicate the ongoing requirement for optimization of the host response to device materials. This paper presents the Download Parser, a tool for handling text downloads from large online databases. Many universities have access to full-text databases which allow the user to search their holdings and then view and ideally download the full text of relevant articles, but there are important problems in practice in managing such downloads, because of factors such as duplication, unevenness of formatting standards, lack of documentation. The tool under discussion was devised to parse downloads, clean them up and standardise them, identify headlines and insert suitably marked-up headers for corpus analysis. RESUMO Este artigo apresenta o Download Parser, uma ferramenta para gerenciar downloads de texto de grandes bancos de dados online. Muitas universidades têm acesso a bases de dados com textos completos que permitem ao usuário pesquisar e, em seguida, visualizar e, idealmente, baixar o texto completo de artigos relevantes. Todavia há problemas importantes na prática do gerenciamento de tais downloads, por causa de fatores como duplicação, falta de padrão de formatação e falta de documentação. A ferramenta em discussão foi concebida para analisar downloads, limpá-los e padronizá-los, identificar títulos e inserir cabeçalhos adequadamente etiquetados para análise de corpus.

U.S. regulations have historically focused on control of liquid polychlorinated biphenyls (PCBs). Nonliquid PCBs present as plasticizers in some construction materials were generally viewed as immobile and not of public health or environmental concern. By the 2000s, nonliquid PCBs began receiving significant regulatory attention, particularly in schools. More recently, PCB-containing construction materials at commercial and industrial facilities are also receiving increased attention. This article highlights the drivers that have led to the increased focus on nonliquid PCBs in construction materials and why this trend is likely to continue, and highlights some of the complexities that can arise for stakeholders.

In this paper we analyse the discourse of austerity in British broadsheets. Theoretically, we combine insights from discourse analysis and political science. Methodologically, we present a novel procedure to build and analyse a robust corpus derived from LexisNexis. Our analysis of this corpus shows the powerful actors in the discourse, and how they were able to exercise this power through ideas and language. Their visibility in the press, and their discursive performance are crucial elements in our analysis. Our analysis of the discourse shows that austerity has negative connotations; that no independent economic expertise was visible; that George Osborne was the most influential actor, providing clever catchphrases; that the oppositional Labour Party failed to develop an alternative narrative; that journalists took up elements of Osborne’s narrative rather than developing their own; and that right-wing think tanks had little visibility.

Analysis of newspaper reporting on the topic of energy security in eight countries – four from the global North (France, Germany, the UK, and the United States) and four from the global South (China, India, Brazil, and South Africa) – produces no support for the thesis that news is disseminated from core countries to the periphery and semi-periphery. There is an important difference between China and the other three fast-developing countries and a highly asymmetric flow of news not aligned to old core-periphery boundaries. In general, energy security is mainly covered in trade and business outlets and less in newspapers with mass circulation, indicating that the topic is still an elite concern. In some instances, attention has surged at the same time in different countries. But very few of these instances show a homogenous coverage across countries. Despite increasingly globalised media, news is created and consumed at a national level.

A series of tip-over and off-the-dock impact tests were performed with stand-up forklifts to investigate the potential for injury to the operator of a forklift in these types of accidents, when the forklift is equipped with an operator’s compartment door. One Crown Equipment Company 35RRTT Model and one 35RCTT Model stand-up forklifts were used in the impact tests. The only modification to the forklifts for the tests was the placement of a door on the entrance to the operator’s compartment. A Hybrid III anthropomorphic test device (ATD) was placed in the operator’s compartment as a human surrogate. During each test, head accelerations, chest accelerations, neck loads, and lumbar loads were measured on the ATD. The motion of the forklift and the ATD were filmed with real-time video and high-speed cameras. Results from the impact tests indicate that there is a high risk of head injury in a right-side tip-over accident and a high risk of head injury and neck injury in a left-side tip-over accident. There is a high risk of a head injury, neck injury, and thoracic injury in off-the-dock forks-trailing accidents. In an off-the-dock forks-leading accident, there is a high risk of arm/shoulder injury, head injury, and neck injury. In both tip-over and off-the-dock forks-trailing accidents, there is a high probability of an entrapment injury under the overhead guard on the forklift.

The notion of ‘textual colligation’ predicts that certain lexical items have a tendency to occur at particular points in a text, i.e. the beginning or end of texts, paragraphs or sentences. This paper describes new corpus-based methods developed to identify the profile of words, clusters (n-grams) and concgrams (non-contiguous patterns in variant order) in terms of their most common textual locations. Groups of co-occurring text-initial items are then analyzed in terms of their discourse function in relation to theories of newspaper structure. This analysis illustrates how methods from corpus linguistics, when targeted to specific textual positions, can complement text-linguistic analyses.

Background Self-management represents an important complement to psychosocial treatments for bipolar disorder (BD), but research is limited. Specifically, little is known about self-management approaches for elevated mood states; this study investigated self-management strategies for: (1) maintaining balance in mood, and (2) stopping progression into hypomania/mania. Methods To identify the common components of BD self-management, Delphi Consensus Consultation methods were combined with a Community-Based Participatory Research (CBPR) approach across five study phases: (1) Qualitative dataset content analysis; (2) Academic/grey literature reviews; (3) Content analysis; (4) Two Delphi rounds (rating strategies on a 5-point Likert scale, Very Unhelpful-Very Helpful), and; (5) Quantitative analysis and interpretation. Participants were people with BD and healthcare providers. Results Phases 1 and 2 identified 262 and 3940 candidate strategies, respectively; 3709 were discarded as duplicates/unintelligible. The remaining 493 were assessed via Delphi methods in Phase 4: 101 people with BD and 52 healthcare providers participated in Round 1; 83 of the BD panel (82%) and 43 of the healthcare provider panel (83%) participated in Round 2—exploratory factor analysis (EFA) was conducted on Round 2 results. Limitations EFA was underpowered and sample was not ethnically diverse, limiting generalizability. Discussion High concordance was observed in ratings of strategy effectiveness between the two panels. Future research could usefully investigate the provisional discovery here of underlying factors which link individual strategies. For example, ‘maintaining hope’ underpinned strategies for maintaining balance, and ‘decreasing use of stimulants’ underpinned strategies to interrupt hypo/manic ascent. There is merit in combining CBPR and Delphi methods.

Rapidly expanding data collections from ocean observing systems create a need for new tools to process, to explore, and to analyze data. This article presents capabilities of the EventViewer in the context of exploring ocean observing system data. The EventViewer is a graphical user interface for visualizing and exploring patterns in events where events constitute changes of interest in a time series. The EventViewer allows users to make queries on events stored in an events database and explore various spatial, temporal, and thematic patterns in events. Once events are selected, the user can assign event spatial, temporal, and thematic categories to graphic display elements in the interface called bands, stacks, and panels. Selected events are displayed according to their associations with the user-specified categories. The spatial, temporal, and thematic categories can be directly manipulated and rearranged among the bands, stacks, and panels to change the view of events and highlight different patterns. The EventViewer supports exploration of periodic patterns, spatial and temporal trends, and event‐event relationships. The EventViewer functionality is illustrated with oceanographic events extracted from ocean observing system sensor data collected in the Gulf of Maine. Events were extracted from multiple time series variables collected at a number of locations and depths.

BackgroundWe developed a simple non-invasive Cardiac Output Response to Stress (CORS) Test to improve diagnosis and monitoring of heart failure in primary care.AimThe aim of the present study was to assess test-retest reproducibility of the CORS test.MethodThirty-two healthy volunteers (age, 64±10, female n=18) were recruited. Cardiac output was measured continuously using bioreactance method in supine and standing position, and during a two 3-min stages of a step-exercise protocol (10 and 15 steps per minute) using a 15-cm height bench. The CORS test was performed twice i.e. Test 1 and Test 2.ResultsCardiac output and stroke volume were not significantly different between the two tests at supine (6.2±1.4 versus 6.3±1.7 L/min, P = 0.84; 102±24 versus 108±32 ml/beat, P = 0.36), standing (5.7±2.1 versus 5.7±1.9 L/min, P = 0.99; 82±32 versus 83±29 ml/beat, P = 0.93), stage one step-exercise (8.5± 1.8 versus 8.2±1.9 L/min, P = 0.56; 104±26 versus 104±27 ml/beat, P = 0.99) and stage two step-exercise (9.9±1.7 versus 9.6±2.0 L/min, P = 0.51; 109±29 versus 111±26 ml/beat, P = 0.76). There was a significant positive relationship between Test 1 and Test 2 cardiac outputs (r=0.92, PConclusionThe CORS test demonstrates acceptable reproducibility and can potentially be used in primary care to identify and monitor heart failure.

Following the lead of human athletic training, equine massage therapy is becoming a more common part of the management of equine athletes and pleasure horses alike. The basic science rationale for massage is supported by research indicating that massage may affect a number of physiologic systems as well as cellular and fascialcomponentsofthemuscularsystem.Equinetherapeuticmassage,orsportsmassage,employsanumberof techniques first developed in humans and has been reported to increase range of motion and stride length, reduce activity of nociceptive pain receptors, and reduce physiologic stress responses. Additional preliminary research indicates that massage therapy also may improve some aspects of exercise recovery. Although important evidence has begun to document the potential benefits of massage therapy for equine athletes, the current reviewmaysaylessaboutthetrueclinicaleffectsofmassage therapythanitdoesaboutthecurrentstateofresearchin this field. Additional prospective study of massage therapy using sufficient scientific rigor will be necessary to provide veterinarians, trainers, and owners with definitive data and scientifically based confidence in the use of equine massage. In the meantime, the preliminary research, anecdotal positive effects, and case studies indicating potential benefit are not to be ignored; equine massage therapy already plays a valuable practical role in the care and training of many equine athletes.

The distal interphalangeal joint and the common digital extensor tendon were also injected in 81.6% (62/76) and 43.4% (33/76) of the cases, respectively. Use of the ultrasound had a positive and negative predictive value of 98% and 9%, respectively. In this study, ultrasound guidance was useful for confirming injection of the collateral ligament of the distal interphalangeal joint but did not prevent injecting the distal interphalangeal joint and the common digital extensor tendon.
