Invasive pulmonary aspergillosis (IPA) is an opportunistic infection that usually threatens immunocompromised patients. However, there are some reports of IPA in immunocompetent patients without the obvious classic risk factors. We present the case of an 82-year-old woman with a prior medical history of chronic obstructive pulmonary disease (COPD) and a recent short-term corticosteroid regimen for an acute exacerbation. She was admitted with dyspnoea, cough, and pleuritic pain and was diagnosed with pneumonia. Clinical deterioration occurred, and a diagnosis of IPA was made. She received treatment with voriconazole but died 14 days after admission. This case highlights the importance of considering IPA among the possible causes of infection in this population. Prompt institution of appropriate antifungal therapy is paramount for the management of this condition.

Human DNA polymerase delta is essential for DNA replication and acts in conjunction with the processivity factor proliferating cell nuclear antigen (PCNA). In addition to its catalytic subunit (p125), pol delta comprises three regulatory subunits (p50, p68, and p12). PCNA interacts with all of these subunits, but only the interaction with p68 has been structurally characterized. Here, we report solution NMR–, isothermal calorimetry–, and X-ray crystallography–based analyses of the p12–PCNA interaction, which takes part in the modulation of the rate and fidelity of DNAsynthesis by pol delta.We show that p12 binds with micromolar affinity to the classical PIP-binding pocket of PCNA via a highly atypical PIP box located at the p12Nterminus. Unlike the canonical PIP box of p68, the PIP box of p12 lacks the conserved glutamine; binds through a 2-fork plug made of an isoleucine and a tyrosine residue at 3 and 8 positions, respectively; and is stabilized by an aspartate at 6 position, which creates a network of intramolecular hydrogen bonds. These findings add to growing evidence that PCNAcan bind a diverse range of protein sequences that may be broadly grouped as PIP-like motifs as has been previously suggested.

International audience; Information about small-scale fisheries (SSF) is often scarce and scattered. This is partly due to insufficient attention on this sector, whose contribution to society is often assumed to be negligible. It also results from SSF being highly diverse, with complex patterns of harvest and post-harvest activities taking place in a wide range of aquatic environments and often in remote areas. Existing fisheries information systems fail to fully capture the characteristics and essence of SSF, resulting in a lack of integrated and up-to-date data, which further marginalizes the sector in policymaking and governance. To help rectify the situation, the Too Big To Ignore project developed the Information System on Small-scale Fisheries (ISSF), a Web-based, open data portal to collect and disseminate knowledge on various aspects of SSF. This paper describes the conception of ISSF and its key features, and presents some results extracted from the analysis of ISSF data, illustrating the importance of such a global database on SSF.

Light-chain deposition disease is a rare paraproteinaemia characterized by deposition of monoclonal light-chain immunoglobulins with a non-amyloid structure, most frequently affecting the kidney. The authors present the case of a 58-year-old man admitted due to haemorrhagic shock caused by spontaneous splenic rupture. Investigation showed coagulopathy, homogeneous hepatomegaly with elevated cholestasis enzymes but normal bilirubinaemia, and a nephrotic syndrome with monoclonal kappa light chains. Liver and bone marrow biopsies revealed light-chain deposition disease. Splenic rupture as the presentation of a paraproteinaemia is rare. The diagnosis of light-chain deposition disease should be considered in case of a monoclonal plasma cell disorder with Congo-red negative deposits on histology. LEARNING POINTS Spontaneous splenic rupture is a rare presentation of a paraproteinaemia. Light-chain deposition disease is a rare disease with non-fibrillar deposition of monoclonal light-chain immunoglobulins most frequently affecting the kidney. p15PAF is an oncogenic intrinsically disordered protein that regulates DNA replication and lesion bypass by interacting with the human sliding clamp PCNA. In the absence of DNA, p15PAF traverses the PCNA ring via an extended PIP-box that contacts the sliding surface. Here, we probed the atomic-scale structure of p15PAF-PCNA-DNA ternary complexes. Crystallography and MD simulations show that, when p15PAF occupies two subunits of the PCNA homotrimer, DNA within the ring channel binds the unoccupied subunit. The structure of PCNA-bound p15PAF in the absence and presence of DNA is invariant, and solution NMR confirms that DNA does not displace p15PAF from the ring wall. Thus, p15PAF reduces the available sliding surfaces of PCNA, and may function as a belt that fastens the DNA to the clamp during synthesis by the replicative polymerase (pol δ). This constraint, however, may need to be released for efficient DNA lesion bypass by the translesion synthesis polymerase (pol η). Accordingly, our biochemical data show that p15PAF impairs primer synthesis by pol η-PCNA holoenzyme against both damaged and normal DNA templates. Este trabalho busca estimar a eficiência da economia de água potável no Campus Santa Mônica, Universidade Federal de Uberlândia, utilizando reservatórios de água pluvial para a substituição de água tratada por água de chuva para fins não potáveis. Por meio de informações obtidas junto a Prefeitura Universitária foi possível estimar a demanda diária de água de abastecimento para os Blocos desse Campus, em especial para aqueles com predominância de salas de aulas. A fim de verificar a capacidade de obtenção de água de chuva nas áreas impermeáveis buscou-se no site Hidroweb da Agência Nacional de Águas (ANA) os dados de precipitação diários no período de 1980 a 2015. Com as informações de oferta e demanda de água das principais edificações da área de estudo, construiu-se em Planilhas Eletrônicas, o balanço hídrico diário e a estimativa da eficiência de reservatórios na faixa de volume de 10 a 80m . O período médio de retorno do capital investido variou de 4,1 a 41,3 anos em função das características de cada bloco, tais como: área do telhado, volume do reservatório e demanda. Verificou-se que a escolha do reservatório com base apenas na porcentagem da demanda atendida é ineficaz, o bloco 3Q, por exemplo, apresentou para a demanda 10L/discente.dia uma porcentagem de demanda atendida 32% e 65% superior respectivamente as demandas de 15 e 20L/discente.dia, porém a economia média anual de água potável para a demanda de 20L/discente.dia foi, em média, 9% e 24% maior que as economias médias das demandas de 15 e 10L/discente.dia respectivamente. Além disso, através de uma regressão linear múltipla foram propostas fórmulas para o cálculo da economia anual nos campi da UFU em função do tipo de telhado da cobertura, da população do bloco, do volume do reservatório de armazenamento instalado, da demanda diária por água e da área total da cobertura. Para todos os campi da UFU o coeficiente de correlação R foi superior a 0,83. By means of information obtained from the University City Hall, it was possible to estimate the daily supply water demand for the Blocks of this Campus Especially those with a predominance of classrooms . In order to verify the capacity to obtain rainwater in the impermeable areas, the daily rainfall data from 1980 to 2015 was searched on the National Water Agency's website (Hidroweb). Significance Deciphering microbial virulence mechanisms is of fundamental importance for the treatment of infectious diseases. Legionella pneumophila, the causative agent of Legionnaires’ pneumonia, hijacks a variety of host cell factors during intracellular growth. Herein, we uncovered the molecular mechanism by which the L. pneumophila effector RidL targets the host VPS29, a scaffolding protein of endosome-associated sorting machineries. Using X-ray crystallography, we determined the structure of RidL, both alone and in complex with retromer. We found that RidL uses a hairpin loop similar to that present in cellular ligands to interact with retromer. This sophisticated molecular mimicry allows RidL to outcompete cellular ligands for retromer binding, explaining how L. pneumophila utilizes the endosomal sorting machinery to facilitate targeting of effector proteins., Microbial pathogens employ sophisticated virulence strategies to cause infections in humans. The intracellular pathogen Legionella pneumophila encodes RidL to hijack the host scaffold protein VPS29, a component of retromer and retriever complexes critical for endosomal cargo recycling. Here, we determined the crystal structure of L. pneumophila RidL in complex with the human VPS29–VPS35 retromer subcomplex. A hairpin loop protruding from RidL inserts into a conserved pocket on VPS29 that is also used by cellular ligands, such as Tre-2/Bub2/Cdc16 domain family member 5 (TBC1D5) and VPS9-ankyrin repeat protein for VPS29 binding. Consistent with the idea of molecular mimicry in protein interactions, RidL outcompeted TBC1D5 for binding to VPS29. Furthermore, the interaction of RidL with retromer did not interfere with retromer dimerization but was essential for association of RidL with retromer-coated vacuolar and tubular endosomes. Our work thus provides structural and mechanistic evidence into how RidL is targeted to endosomal membranes.
