MiBe-hankkeen päätavoitteena oli selvittää voiko mikrobiaktiivisuus vaikuttaa bentoniitin toimintakykyyn ns. worst case-olosuhteissa. Tulokset osoittivat, että mikrobit pystyvät niille suotuisissa olosuhteissa (slurry), jotka simuloivat suomalaisia loppusijoitusolosuhteita muuttamaan bentoniitin koostumusta (bentoniitin rakenteellinen rauta pelkistyi) siten, että sen paisuntakyky heikkeni. Tämä havaittiin riippumatta kokeen lähtöolosuhteista (hapellinen/hapeton) ja siitä, mistä aktiivinen mikrobiyhteisö oli peräisin (bentoniitti, pohjavesi, pintavesi). Tulokset pätevät olosuhteissa, joissa mikrobeille on tarjolla energialähde, eikä niiden aktiivisuutta rajoita paine, joka estäisi energian ja aineenvaihduntatuotteiden siirtymistä. On kuitenkin mahdollista, että tällaiset olosuhteet toteutuvat paikallisesti loppusijoituksen raj apinnoilla. Tämä aiheuttaa huomioon otettavan riskin mikrobiaktiivisuuden leviämisestä bentoniitissa sen paikallisen toimintakyvyn muutoksen seurauksena.MoToPro-hankeen yhteistyönä toteutettiin moniestevuorovaikutuskoe, jonka reaktoreihin oli lisätty Olkiluodon hapetonta pohjavettä, bentoniittia, pieni määrä energialähteitä sekä kuparipaloja. Kahden vuoden jälkeen todettiin sulfaatinpelkistyksen olleen voimakasta reaktoreissa, joihin oli lisätty energialähteitä. Lisäksi todettiin aktiivinen raudan pelkistyminen, jonka seurauksena 40-50% bentoniitin rakenteellisesta raudasta oli pelkistynyt bentonitti-slurryssä, mutta myös kompaktoidun bentoniitin pintakerroksessa. Odotetusti, huomioiden lyhyt tutkimusaika, mikrobien monimuotoisuus oli lisääntynyt syvemmällä kompaktoidussa bentoniitissa, mutta mikrobimäärät olivat matalampia kuin pintakerroksessa.

MiBe-hankkeen päätutkimuskohde kolmantena tutkimusvuonna oli vuonna 2016 aloitetun bentoniitin mikrobiologisen säilytyskokeen tulosten analysointi ja tieteellisen artikkelin kirjoitus. Kokeesta tutkittiin myös ylimääräiset näytteet, joilla selvitettiin mikrobiaktiivisuuden vaikutuksia bentoniitin rakenteellisen raudan hapetusasteeseen sekä mineralogiaan. Lähes viiden vuoden kokeen päätulokset osoittivat, että bentoniitista liukeneva orgaaninen hiili ylläpiti mikrobiatiivisuutta vain vajaa kaksi vuotta. Hapellisissa olosuhteissa aloitetussa kokeessa happi kulutettiin lähes kokonaan muutamassa kuukaudessa. Bakteeritoiminta aktivoitui nopeasti uudelleen pienellä määrällä lisättyjä yksinkertaisia orgaanisia hiiliyhdisteitä (Na-formiaatti ja - asetaatti). Neljän vuoden jälkeen todettiin mikrobiaktiivisuuden seurauksena bentoniitin rakenteellisen raudan pelkistyneen kokonaan XANES-mittauksessa (X-ray absorption near edge structure) lukuunottamatta abioottisia konrollinäyteitä. Mikrobi yhteisöt poikkesivat merkittävästi toisistaan hapettomasti ja hapellisesti aloitetuissa kokeissa, mutta molemmissa kokeissa todettiin bentoniitin raudan pelkistyminen. Erityisesti hapettomissa bentoniittikokeissa todettiin bentoniitin mineralogiassa illiitin osuuden kasvaneen suhteessa smektiittiin XRD (X-ray diffraction) -mittauksessa. Tämä voi olla osoitus bentoniitin paisuntakyvyn pienenemisestä.MoToPro-hankeen yhteistyönä kesällä 2020 aloitetusta moniestevuorovaikutuskokeesta tutkittiin lähtötilanteen ja kesän 2021 näytteitä. Analysoitujen bentoniitinäytteiden perusteella mikrobiyhteisöihin vaikutti voimakkaasti sijainti bentoniitin pinnassa tai syvemmällä, sekä pieni määrä lisättyjä energialähteitä. Sulfaatinpelkistäjäbakteerien osuus yhteisöstä oli selvästi suurempi bentoniitin pintakerroksessa sekä näytteissä, joihin oli lisätty energialähteitä. Vuoden säilytyksen jälkeen sulfatinpelkistäjien osuus oli edelleen kasvanut, vaikka yhteisön valtalajien osuudet olivat muuttuneet (Desulfomicrobium->Desulfurivibio + Desulfocapsaceae).

MiBe-hankeen toisena vuonna kesällä 2020 aloitettiin MoToPro-hankkeen yhteinen moniestevuorovaikutuskoe. Kokeessa tutkitaan mikrobiyhteisöjen ja abioottisten reaktioiden kokonaisuutta lasi- ja POM- (polyoksimetyleeni) reaktoreissa, joissa on mukana kupari, bentoniitti, kallioperä, loppusijoitustilan vesi, seoskaasu sekä ravinteita kiihdyttämään mikrobiologista aktiivisuutta. Pääosa kokeista toteutetaan bentoniitti-slurrynä, mutta mukana on myös kaksi kompaktoitua bentoniittikoetta. Moniestekokeesta otettiin lähtötilannetta kuvaavat näytteet kaksi viikkoa kokeen pystytyksen jälkeen ja niiden analysointi jatkuu seuraavaan vuoteen.Projektin toinen tutkimuskohde on vuonna 2016 aloitettu bentoniitin mikrobiologinen säilytyskoe. Kokeesta tutkittiin neljännen vuoden näytteet sen jälkeen, kun mikrobeja oli pyritty aktivoimaan ravinnelisäyksillä reilun vuoden ajan. Tulokset osoittivat, että formiaatti- ja asetaattilisäykset kasvattivat bakteerien Ja sulfaatinpelkistäj ien määriä huomattavasti. Hapettomissa näytteissä mm. rikki/rauta yhdisteitä pelkistävien Desul fomicrobium, Trichlomonas, Thermincola -lajien, jotka pystyvät hyödyntämään asetaattia ja/tai formiaattia suhteelliset osuudet mikrobiyhteisössä kasvoivat. Arkeonienja sienten rooli jäi pieneksi, eivätkä ne hyötyneet ravinnelisäyksistä. Tulokset osoittivat näytteissä olevan aktiivisia sulfaatin- ja raudanpelkistäjiä. Mikrobitoiminnan vaikutusta bentoniitin rakenteelle selvitettiin bentoniitin rakenteellisen raudan hapetusasteen määrityksellä. Käytettävissä ollut määritysmenetelmä ei ollut sopiva kivimurskaa sisältäville näytebentoniiteille, joten mikrobiaktiivisuuden vaikutukset bentoniitin rakenteelle jäivät epäselviksi. Tulevana vuonna raudan hapetusastetta bentoniitissa selvitetään yhteistyössä Helsingin Yliopiston kanssa XANES-mittauksilla.

Estimation of archaeal numbers by use of fluorescent DNA binding dyes is challenging, because primers targeting the archaeal 16SrRNA genes readily also bind to bacterial 16S rRNA gene sequences, especially when the relative abundance of bacteria is greater than that of archaea. In order to increase specificity, we optimized a fluorescent probe-based assay using previously published archaeal primers and probe. The assay was tested on genomic DNA of pure bacterial and archaeal cultures and optimized using PCR amplicons of the archaeal pure cultures. The used bacterial strains showed slight amplification using the fluorescent dye assay, whereas all archaeal strains could be amplified with the archaea primers used. Due to differences in genome size and number of 16S rRNA gene copies between the tested archaeal strains, the amplification level varied greatly between the strains. Therefore, we also tested the amplification using PCR amplified fragments of the archaeal 16S rRNA genes. The tests with the archaeal 16S rRNA gene amplicons showed good amplification, although the amplification efficiency still varied between archaeal strains. The qPCR assay was used to estimate the archaeal numbers in process water of a multi-metal mine's metallurgical plant [1] and will be used in similar future microbiological analysis included in the H2020 ITERAMS project (Grant agreement# 730480).

Spent nuclear fuel will be deposited in deep geological repositories in the crystalline bedrock in Finland and Sweden. The repositories rely on the multi-barrier KBS-3 concept, where the first barriers consist of copper-sheeted cast-iron canisters that contain the spent nuclear fuel rods. However, the repositories will be influenced by both abiotic and biotic factors. The repositories, when closed, will be flooded with a mixture of both the local groundwater and surface water and eventually return to an anoxic deep subsurface environment. This environment will contain microbial communities that utilize chemical components that have been produced both abiotically and biologically, which will determine the composition and size of the microbial communities in the repositories. Overtime, integrity of the storage canisters may become compromised due to, for example, microbially influenced corrosion, which may result from the corrosive nature of hydrogen sulfide produced in biological sulfate reduction. Radionuclides originating from the decay of the spent nuclear fuel may escape into the repository environment, from where they may diffuse further into the surface environment. In this chapter, we aim to shed light on some of the most important aspects of the Fennoscandian Shield deep crystalline bedrock biosphere identified to date, with examples of putative biogeochemical cycles that prevail in these environments.

openaire: EC/H2020/730480/EU//ITERAMS The changes in water quality owing to recirculation of water in mineral processing plants can compromise the plant performance as well as maintenance needs. Therefore, mining process water quality assessment is becoming critical. Nevertheless, very few studies have investigated the suitability of the current analysis methodology practiced in certified laboratories for evaluating mining process water quality. This article presents two case studies to highlight the major issues encountered when performing sampling for physicochemical and chemical parameters in process water at two European mine sites using procedures from two certified laboratories. In addition, microorganisms were shown to be abundant in process waters and likely affect the mining water chemistries. However, the protocols used for microbial studies are not optimal for mining process samples, and need to be improved. The results showed difficulties in providing satisfactory results when analyzing control samples. Additionally, the analysis results presented a strong imbalance in TDS and sulfur compounds. Several potential causes associated with the poor quality of the analysis results have been outlined with a specific focus on the preservation methods. A literature review on the degradation of thiosalts suggested that the current preservation procedures are not suitable for preserving sulfur compounds. Moreover, the results indicated that the water matrix strongly influenced the validity of the chosen analysis method. In conclusion, the analysis methods should be customized for the different mining water matrix types in order to ensure the accuracy and reproducibility of the results.

Microorganisms enter the flotation process mainly from intake water and ore material. The diversity and number of microorganisms can vary significantly from mine to mine. In flotation, the conditions including oxygen levels, temperature, and nutrients from ore, intake water, and reagents are often favorable for the microbial growth. The mining industry aims to close the water loops, which is expected to result in the accumulation of microorganisms in the process waters with potential effects on flotation performance. Bioflotation, bioleaching, and bio-oxidation have been studied for decades as tools for concentrating and dissolving minerals. In contrast, there is limited scientific literature or industrial knowledge about microorganisms that naturally inhabit and prevail in minerals processing applications over a wide pH range. Microorganisms affect minerals when they selectively attach to the surfaces, produce extracellular polymeric substances (EPS) and polysaccharides, oxidize or reduce the minerals, change the pH and Eh of the process solution, and degrade organic flotation chemicals. Microorganisms contain different structural components that affect their surface chemistry, charge, and behavior in flotation, but these properties may also change via adaptation and solution conditions. Almost all studies on flotation have focused on chemical and physical parameters, and the role of naturally occurring microorganisms has remained underexplored. Advances in genomics and proteomics offer possibilities to describe not only which microorganisms are present, but also which physiological functions are being exercised. This article reviews the current knowledge of microorganisms in various mineral processes, identifies potential microbe–mineral interactions in flotation, describes the gaps in current knowledge, and concludes with the potential effects of microorganisms on flotation, especially in closed water loops.

MiBe-hankeen ensimmäisenä vuonna on suunniteltu aktiivisesti MoToPro-hankkeen monieste-vuorovaikutuskoetta. Monitieteellinen koesuunnitelma on valmistunut MoToPro- hankkeiden yhteistyönä ja koe aloitetaan keväällä 2020, kun tarvittavat valmistelut on saatu loppuun. Uusi mikrobiologinen bentoniittikoe toteutetaan yhteisen vuorovaikutuskokeen osana, näin varmistetaan kokeiden luotettavuus ja monipuolinen analysointi. Kokeessa tutkitaan mikrobiyhteisöjen ja abioottisten reaktioiden kokonaisuutta reaktoreissa, joissa on mukana kupari, bentoniitti, kallioperä, loppusijoitustilan vesi, seoskaasu sekä ravinteita.Koska uusi mikrobiologinen bentoniittikoe on järkevintä aloittaa yhteisen moniestekokeen osana, MiBe-hankkeessa voitiin tutkia tarkemmin jo käynnissä olevan bentoniittikokeen näytteitä. Kokeen sekvensointitulokset osoittivat bentoniittien m ikrobiyhteisöjen olevan monimuotoisia ja muuttuvan erilaiseksi kuin näytteisiin lisättyjen vesien mikrobiomit olivat alun perin. Hapettomissa olosuhteissa aloitettujen bentoniittien mikrobiomeja hallitsivat rikkiyhdisteiden pelkistykseen kykenevät bakteerit. ltiölliset bakteerit olivat yleisiä bentoniiteissa, joiden vesi oli alun perin steriilisuodatettu, eli ne olivat todennäköisesti peräisin kuivasta bentoniitista. Hapellisissa olosuhteissa aloitettujen bentoniittikokeiden mikrobiomit olivat monimuotoisemmat kuin hapettomien kokeiden mikrobiomit. Hapellisissa kokeissa ei noussut esille selviä hallitsevia ryhmiä ja rikkiyhdisteiden pelkistykseen liittyviä bakteereja löydettiin vain satunnaisesti. Kahden vuoden koeaikana bentoniittien mikrobiomit eivät olleet muuttuneet oleellisesti missään näytteistä. Mikrobiyhteisöt, mukaan lukien arkeonit ja sienet, vaikuttivat olevan melko stabiilissa tilassa ja on todennäköistä, että aktiivisuutta on rajoittanut energialähteiden loppuminen. Kokeeseen alettiin lisäämään reilun kahden ja puolen vuoden jälkeen ravinteita, mikrobitoiminnan aktivoimiseksi ja jo viikon sisällä hapettomissa kokeissa havaittiin bentoniitin mustumista, joka indikoi mikrobitoiminnan voimistumista. Tarkoitus on jatkaa ravinteiden lisäämistä näytteisiin 3-4 kk välein, jotta mikrobitoiminta pysyy aktiivisena sekä tutkita kokeista mikrobitoiminnan vaikutuksia bentoniitin rakenteeseen, kemiaan ja mikrobiomeihin MiBe-hankkeen seuraavan tutkimusvuoden aikana.

Two long-term potentially oil exposed Baltic Sea coastal sites near old oil refineries and harbours were compared to nearby less exposed sites in terms of bacterial, archaeal and fungal microbiomes and oil degradation potential. The bacterial, archaeal and fungal diversities were similar in oil exposed and less exposed sampling sites based on bacterial and archaeal 16S rRNA gene and fungal 5.8S rRNA gene amplicon sequencing from both DNA and RNA fractions. The number of genes participating in alkane degradation (alkB) or PAH-ring hydroxylation (PAH–RHDα) were detected by qPCR in all water and sediment samples. These numbers correlated with the number of bacterial 16S rRNA gene copies in sediment samples but not with the concentration of petroleum hydrocarbons or PAHs. This indicates that both the clean and the more polluted sites at the Baltic Sea coastal areas have a potential for petroleum hydrocarbon degradation. The active community (based on RNA) of the coastal Baltic Sea water differed largely from the total community (based on DNA). The most noticeable difference was seen in the bacterial community in the water samples were the active community was dominated by Cyanobacteria and Proteobacteria whereas in total bacterial community Actinobacteria was the most abundant phylum. The abundance, richness and diversity of Fungi present in water and sediment samples was in general lower than that of Bacteria and Archaea. Furthermore, the sampling location influenced the fungal community composition, whereas the bacterial and archaeal communities were not influenced. This may indicate that the fungal species that are adapted to the Baltic Sea environments are few and that Fungi are potentially more vulnerable to or affected by the Baltic Sea conditions than Bacteria and Archaea.

Bentonite is an important buffer material between the copper canister and bedrock in geological disposal of spent nuclear fuel in Finland. The long-term experiment with compacted Na-bentonite with low saline ground-water simulant and copper canister was initiated in 1997. The aim of the experiment was to evaluate changes in chemical, mineralogical and microstructural parameters in bentonite in oxic and anoxic conditions. The experiment was planned to be finished after ten months, but two samples were stored and dismantled in 2012. Although the experimental setup was not initially designed for microbiological studies, they were performed the end of the experiment. Changes in the bentonite mineralogy included the observation of secondary copper minerals formation in the middle part of the bentonite matrix in copper cylinder in oxic conditions. Copper content in pyrite increased when moving from the middle of the bentonite towards the copper cylinder surface in both oxic and anoxic experiments. Microstructural studies on bentonite did not show any significant differences in bentonite structure between samples taken from anoxic and oxic experiments. Chemical changes were typical dissolution of gypsum and calcite, which releases sodium, sulphate and carbonates into external water, while calcium exchanges sodium in the interlamellar spaces of bentonite. The presence of living microbes on bentonite and on copper surface could not be demonstrated in this study but microscopical studies revealed living microbial cells in the external water surrounding copper cylinders. According to the sequencing, sulphate (SRB) and iron reducing bacteria (IRB) were detected in bentonite, water and copper surface. SRBs can produce corrosive sulphide and IRBs can be related to processes that could be linked to the loss of swelling properties in bentonite. Fungal conidia and hyphae were detected by SEM in water and several groups of Ascomycetes and Basidiomycetes were identified from bentonite samples.

Gender in early childhood education is currently a discussed topic in the news and the subject also draws a lot of attention in the media in general. Different approaches to gender in preschools have brought both criticism and praise among custodians preschool teachers and within the community. Several kindergartens in Sweden have an explicit focus on gender work and some of them are also LBTQ-certified. The aim of this study is to investigate teachers’ attitudes to current gender work in preschools and to empirically examine how different perspectives on gender work in Sweden and Finland interact in preschools. The aim is also to discover alternative ways to work with gender and equality within a preschool framework. The study is based on semi-structured interviews and questionnaires. I have compared the Swedish preschool curriculum with the Finnish equivalent to get the views of the political governing documents that differ regarding gender and gender equality work in these two countries. (”Varhaiskasvatuksen suunnitelman perusteet” translated as "The fundamental features/basics of the plan for early childhood education", Finlands national curriculum). The theoretical framework used in the study is a gender perspective approach. The study points out that preschool teachers’ tensions differ to some degree, depending on the preschool and depending on preschool teachers' education, own values ​​and enthusiasm for the subject. The study lifts the reflection about it as important. The study shows that preschool teachers in Sweden are aware of work with gender, but interprets the way to implement these goals through the curriculum objectives in a multifaceted way. Both the Finnish national curriculum and the Helsinki City applied version of the national curriculum ("Foundations of early childhood education") lift up many important learning objectives, but do not specifically raise transgender work or work against traditional gender patterns. The qualitative interviews conducted in Finland present highlights that a discussion debate about gender work is ongoing in the country where their curriculum is to be renewed by the year 2017. The majority of the preschool teachers in both my interviews and questionnaires, in Sweden and Finland, pointed out that gender work has changed through the past decade. The majority of the preschool teachers also pointed seeing the individual, rather than the gender, as the most important in their work. A special feature of my study gives indication that a general discourse shift from compensatory pedagogic to norm critical pedagogy has taken place. Technique in activities was raised as a non-gender-coded material for children and digital tools in preschool as an entrance for more men in the teacher profession. What also has been found in my study is that gender is defined mainly depending on the context and that perverted approaches take time to change. My conclusion is that preschool teachers’ intention in my study, is not to limit children based on their gender. Their main intention and challenge is to continue offer children a multi-faceted activity based on children's interests and according to curriculum.

Anaerobic, oligotrophic deep subsurface groundwater is an extremely demanding environment for microorganisms. This literature review presents an overview of microbial metabolism connected with dominant electron acceptors in such environments, including geological nuclear waste disposal repositories. The microbial activity is considerably lower in deep subsurface groundwater than in nutrient rich environments. The most common applicable terminal electron acceptors in this environment are nitrate, Mn(IV), Fe(III), sulfate, and CO2 in the order of the decreasing energy yields. In deep geological repositories sulfate reduction to sulfide is considered as a major risk for safe disposal of high radioactive waste copper and steel capsules due to capsule biocorrosion. The literature review also outlines 35S-label technique applicable to measure the rate of microbiologically produced sulfide. The general features of radioactive isotopes, their decay and detection are reviewed. Different kind of labelled sulfur distillation methods have been developed since 1950's. The applications of 35 SO4-tracer methods in environmental microbiology have started from ocean sediment sulfate reduction rate studies. At present distillation methods used are able to fractionate acid volatile sulfur compounds (FeS, HS) and chromium reducible sulfur compounds (FeS2, S0). 35S-tracer methods are applicable also to study environments with low sulfate reduction rates, these studies are reviewed. Finally, problems associated with so called cryptic sulfur cycle in environment are summarized.
