4. RCE Rhine-Meuse: Towards Learning for Sustainable Development

Abstract

In the context of the UN Decade of Education for Sustainable Development (DESD), United Nations University (UNU) initiated the creation of a network of Regional Centres of Expertise (RCEs) for Education for Sustainable Development. This paper describes the philosophy behind the activities performed by one of those RCEs, RCE Rhine-Meuse, an RCE initiative in the triangle formed by the cities of Eindhoven (the Netherlands), Cologne (Germany) and Leuven (Belgium). A small and flexible structure was created that allows the organisation to operate at the lowest possible level of costs. Safeguards in relation to both academic quality and stakeholder participation were instituted to guarantee a lasting commitment on the part of regional actors; and a strategic advisory board made up of regional stakeholders and academic professionals was formed. Some examples of RCE Rhine-Meuse projects are given. (Contains 1 figure and 2 notes.)

Abstract

Introducción: La afectación ósea de la enfermedad de Gaucher (EG) tipo 1 es invalidante y silente, para su diagnóstico debe realizarse resonancia magnética (RM) sistemáticamente. El objetivo del estudio fue saber si el tipo de hospital implica diferencias en el uso de medios diagnósticos en la EG. Material y métodos: Se analizan los recursos diagnósticos disponibles y su empleo en la afectación ósea según el tipo de hospital. Resultados: Treinta y un pacientes de hospitales de referencia y 16 de comarcales. El 70% de los casos presentaban afectación ósea (formas graves el 60%). La RM se empleó en el diagnóstico inicial (65%), y el seguimiento (93%) especialmente en hospitales de referencia. La determinación de quitotriosidasa se emplea el 60% de los casos en seguimiento. Conclusiones: La red de hospitales públicos españoles está suficientemente dotada para la atención de pacientes con EG en todos sus niveles asistenciales, aunque se infrautilizan los recursos disponibles. Background: bone involvement in type 1 Gaucher’s disease can be devastating and is oftenly silent. Bone MR is generally recomended. The aim of the study was to elucidate whether the size and location of hospitals in Spain implies any difference in management of GD patients. Material and methods: we surveyed the type of facilities in the hospital (namely MRI) as well as for the presence, type, severity and methodology of follow-up of bone involvement associated to GD, according to the category of hospital (local or reference). Results: 31 patients were followed in reference hospitals whereas 16 other were in local hospitals. 70% of cases had some type of bone involvement, 60% had severe bone disease. MRI was the first choice for diagnosis in 65% and for follow-up in 93% of cases. MRI is less indicated among patients from local hospitals. Chitotriosidase is measured in a high, but insufficient, proportion of the followed patients (60%). Conclusions: the Spanish hospital network, either reference or local hospitals, have an adequate infrastructure for the management of GD patients. However, main diagnostic resources are being currently underused.

44. Membrane related effects in endothelial cells induced by human cytomegalovirus.

Abstract

Previously, we have reported on the increase in procoagulant activity of human umbilical vein endothelial cells (HUVEC) after infection with human cytomegalovirus (HCMV). When using microvascular endothelial cells from foreskin (MVEC), we also observe a significant increase in membrane perturbation and a concomittant increase in procoagulant activity. This effect is both observed with a laboratory HCMV strain (AD169) with low pathogenicity for endothelium and a HUVEC adapted strain (VHL-E) that readily infects endothelial cells. We compared the membrane perturbation of two types of endothelial cells, HUVEC and MVEC with human embryonal fibroblasts (HEF), being fully permissive for both strains. A membrane effect was only found in endothelial cells. Our results suggest that HCMV induces in MVEC more merocyanine-540 incorporation in the membrane as in HUVEC. The increase in the procoagulant activity induced by HCMV was more pronounced in MVEC than in HUVEC. Inactivated virus, as well as virus pre-incubated with heparin was unable to evoke membrane perturbation. It therefore appears that HCMV induces a rapid membrane response in vascular endothelium and that physical interaction of the virion and the endothelial cell is required to elicit this response. Abstract

Fibrin deposits in tumor beds are an intriguing phenomenon. It has been suggested that fibrin plays a role as a provisional matrix in which the tumor grows and induces development of a vascular network. On the other hand fibrin possibly protects the tumor nodule from host defense mechanisms. We therefore investigate whether tumor cells can induce a procoagulant response in endothelial cells leading to fibrin formation. For our studies we employed a modification of the matrix model of Montesano in which sprouting of endothelial cell aggregates can be followed. This system allows us to study in vitro the involvement of coagulation in tumor growth and angiogenesis. Cocultures of endothelial cell aggregates and avascular tumor spheroids in collagen type I gels results in the appearance of extracellular vesicle-like structures on the endothelial sprouts. The vesicles formed on endothelial cell sprouts upon coculturing with tumor cells exhibit an increased amidolytic activity, suggestive of factor X/Xa activity, not dependent on tissue factor exposure. Experiments using HgCl2 and Iodoacetamide point to the importance of SH groups in the factor X/Xa activity on endothelial cell sprouts.

50. Local anesthetic inhibition of pancreatic phospholipase A2action on lecithin monolayers

Abstract

Using quantitative data previously reported for the penetration of local anesthetics into lecithin monolayers, the effects of surface and subphase concentrations of anesthetics on the inhibition of pancreatic phospholipase A2action on didecanoyl phosphatidylcholine monolayers was investigated. Inhibition as a function of subphase concentration of anesthetic was in the order: dibucaine > tetracaine > butacaine > lidocaine = procaine. Inhibition as a function of surface concentration showed no obvious correlation; procaine inhibited at a very low surface concentration, followed by lidocaine at a somewhat higher concentration, and tetracaine, butacaine and dibucaine only at rather high concentrations. Ultraviolet difference spectroscopy indicated an interaction between lidocaine and enzyme in the subphase. Fluorescence studies showed that lidocaine is a competitive inhibitor of enzyme-lipid interface interaction. It is proposed that the more surface-active anesthetics inhibit by surface effects while the less surface-active anesthetics (lidocaine and procaine) inhibit by interaction with the enzyme in the subphase, which prevents enzyme penetration at the monolayer interface.
