Candida albicans is often implicated in nosocomial infections with fatal consequences. Its virulence is contributed to hydrolytic enzymes and biofilm formation. Previous research focused on studying these virulence factors individually. Therefore, this study aimed to investigate the impact of biofilm formation on the hydrolytic activity using an adapted low-cost method. Eleven strains of C. albicans were used. The biofilms were formed on pre-treated silicone discs using 24-well plates and then deposited on the appropriate agar to test each enzyme, while the planktonic cells were conventionally seeded. Biofilms were analysed using Raman spectroscopy, fluorescent and scanning electron microscopy. The adapted method provided an evaluation of hydrolytic enzymes activity in C. albicans biofilm and showed that sessile cells had a higher phospholipase and proteinase activities compared with planktonic cells. These findings were supported by spectroscopic and microscopic analyses, which provided valuable insights into the virulence mechanisms of C. albicans during biofilm formation. This study employs scanning electron microscopy (SEM) to delve into the intricate pollen morphology of Cucurbitaceae (Gourd Family) species, unraveling the nuanced details of their structural features. Concurrently, the research investigates the antimicrobial potentials encoded within these pollen grains, shedding light on their possible applications in the realm of natural antimicrobial agents. Cucurbitaceae plants showcases significant antibacterial and antifungal potentials, underscoring its potential as a source for novel antimicrobial compounds. This research endeavors to provide a comprehensive analysis of pollen morphometry in 12 Cucurbitaceous species, with the primary goal of identifying pollen morpho‐types to enrich the taxonomic understanding of the Cucurbitaceae. Following the Erdtman protocol, the study involved processing, measuring, and capturing pollen grains using SEM techniques. Qualitative data were analyzed to evaluate the variations in pollen types, size, and exine stratified sculptured layering. The pollen grains exhibit characteristics of being monads, ranging in size from medium to very large, with the prevailing shape being prolate‐spheroidal in six species. Notably, the maximum polar axis diameter was recorded for Cucurbita pepo (106.3 μm), while the minimum was observed in Zehneria anomala (32.6 μm). The grains of Cucurbitaceae exhibit diverse surface patterns, including reticulate columellae, reticulate, verrucate‐scabrate, verrucate‐gemmate, echinate baculate, reticulate‐perforate, and micro‐reticulate. This SEM investigation illuminates the intricacies of Cucurbitaceae pollen morphology while concurrently highlighting their noteworthy antimicrobial potentials. Optical monitoring technologies can be used to monitor membrane fouling in real-time and online with the in-situ, fast and nondestructive advantages, which are of great significance for the in-depth understanding of membrane fouling and the optimization of fouling control strategies. In this paper, several kinds of optical real-time monitoring technologies for membrane fouling that had been studied and developed rapidly in recent years were reviewed, including microscopic imaging techniques such as direct observation through membrane, optical coherence tomography and laser confocal microscopy, molecular spectroscopies such as excitation-emission matrix and Raman spectroscopy, and particle image velocimetry. The principles, the typical applications in fouling characterization of these techniques and the derivative techniques were described comprehensively, and the advantages and disadvantages of each technique were analyzed from the aspects of monitoring area and foulant information provided. Finally, the future development direction of the optical monitoring technologies was put forward, which would promote the application of these technologies in the membrane water treatment industries.

Knee orthoses assist patients with impaired gait through the amendment of knee abnormalities, restoration of mobility, alleviation of pain, shielding, and immobilization. The inevitable issues with laborious traditional plaster molding procedures for orthoses can be resolved with 3D printing. However, a number of challenges have limited the adoption of 3D printing, the most significant of which is the proper material selection for orthoses. This is so because the material used to make an orthosis affects its strength, adaptability, longevity, weight, moisture response, etc. This study intends to examine the mechanical, physical, and dimensional characteristics of three-dimensional (3D) printing materials (PLA, ABS, PETG, TPU, and PP). The aim of this investigation is to gain knowledge about these materials' potential for usage as knee orthosis materials. Tensile testing, Olympus microscope imaging, water absorption studies, and coordinate measuring machine-based dimension analysis are used to characterize the various 3D printing materials. Based on the investigation, PLA outperforms all other materials in terms of yield strength (25.98 MPa), tensile strength (30.89 MPa), and shrinkage (0.46%). PP is the least water absorbent (0.15%) and most flexible (407.99%); however, it is the most difficult to fabricate using 3D printing. When producing knee orthoses with 3D printing, PLA can be used for the orthosis frame and other structural elements, PLA or ABS for moving parts like hinges, PP for padding, and TPU or PP for the straps. This study provides useful information for scientists and medical professionals who are intrigued about various polymer materials for 3D printing and their effective utilization to fabricate knee orthoses. The dichroic mirror (DM) is a key component in microscope. We found a ghost in the reflection channel of a dual‐channel fluorescence microscope and studied the relationship between the ghost and the incidence angle θ into the DM. The DM emission surface reflection generated ghost if the θ is not 45°. We analyzed the distance and intensity relationship between the ghost and the primary image, which is θ‐dependent and was demonstrated by imaging live cells and a stage micrometer. The ghost can be eliminated by placing the DM between objective and tube lens, but not between tube lens and detector, ensuring that the incident light into the DM is approximately parallel. Furthermore, the transmitted light of the DM is shifted towards a longer wavelength with increasing θ. IntroductionOral Squamous Cell Carcinoma (OSCC) poses a significant challenge in oncology due to the absence of precise diagnostic tools, leading to delays in identifying the condition. Current diagnostic methods for OSCC have limitations in accuracy and efficiency, highlighting the need for more reliable approaches. This study aims to explore the discriminative potential of histopathological images of oral epithelium and OSCC. By utilizing a database containing 1224 images from 230 patients, captured at varying magnifications and publicly available, a customized deep learning model based on EfficientNetB3 was developed. The model’s objective was to differentiate between normal epithelium and OSCC tissues by employing advanced techniques such as data augmentation, regularization, and optimization.MethodsThe research utilized a histopathological imaging database for Oral Cancer analysis, incorporating 1224 images from 230 patients. These images, taken at various magnifications, formed the basis for training a specialized deep learning model built upon the EfficientNetB3 architecture. The model underwent training to distinguish between normal epithelium and OSCC tissues, employing sophisticated methodologies including data augmentation, regularization techniques, and optimization strategies.ResultsThe customized deep learning model achieved significant success, showcasing a remarkable 99% accuracy when tested on the dataset. This high accuracy underscores the model’s efficacy in effectively discerning between normal epithelium and OSCC tissues. Furthermore, the model exhibited impressive precision, recall, and F1-score metrics, reinforcing its potential as a robust diagnostic tool for OSCC.DiscussionThis research demonstrates the promising potential of employing deep learning models to address the diagnostic challenges associated with OSCC. The model’s ability to achieve a 99% accuracy rate on the test dataset signifies a considerable leap forward in earlier and more accurate detection of OSCC. Opsariichthys bidens (O. bidens) is a fish species native to China. It has become a popular ornamental fish because of its interesting colour dimorphism. Its colour pattern is unique; however, with the exception of model species, such as zebrafish, the molecular mechanisms of most fish colour patterns remain unclear. In the present study, we compared and analysed the transcriptomic data of the skin and anal fins of male and female O. bidens. A total of 9028 differentially expressed genes (DEGs) were identified, of which 5259 were upregulated and 3769 were downregulated in males. Compared with known pigmentation genes, Mitf, Tyr, Tyrp1, Dct, Pnp4a, Alx4a, Gch1, and Csf1r were identified as potential genes related to colour dimorphism. Second, we tracked the process of colour development in male O. bidens and found that there were three stages in the formation of body colour. We explored the distribution, migration, and expression patterns of genes related to pigment cells at each stage using microscopic imaging, transmission electron microscopy, enzyme-linked immunosorbent assay, and high-performance liquid chromatography. Melanophores, xanthophores, and iridophores were found to be distributed on the skin or anal fins of O. bidens. It was inferred that this unique body colour pattern is caused by the differentiation and migration of pigment cells at specific stages. This discovery provides a basis for studying the molecular mechanisms of pigmentation in ornamental fish and will contribute to the genetic selection and breeding of O. bidens in ornamental fish cultures in the future.

In this study, two different types of amorphous carbonaceous Precambrian rock, classified as noble elite shungite and black raw shungite, were tested as possible electrode materials of natural origin. Both types were machined into cylindrical shapes to form the corresponding solid electrodes and their physicochemical and electrochemical properties were compared with the standard glassy carbon electrode (GCE). The raw stones were first subjected to microscopic imaging by using scanning electron microscopy and energy-dispersive X-ray spectroscopy, both of which indicated significant differences in their morphology and in the content of impurities. An electrode prototype manufactured from noble elite shungite (EShE) with a carbon content of about 94% (w/w) has offered a very satisfactory electrochemical performance with a nearly identical heterogeneous electron-transfer rate constant of 7.8 × 10−3 cm s−1 for ferro/ferricyanide redox couple, a slightly narrower potential range (~2.1 V) and a relatively low double-layer capacitance (of ca. 50 μF), resulting in low background currents comparable to those at the GCE. In contrast, the second electrode based on black raw shungite (BShE) with a carbon content of ca. 63% (w/w) exhibited markedly worse electrochemical properties and more than four times higher double-layer capacitance, both of which were probably due to the presence of poorly conductive impurities. The whole study has been completed with three different examples of electroanalytical applications, revealing that the first type, EShE, is a more suitable material for the preparation of electrodes and may represent a cheap alternative to commercially marketed products. Marine microalgae are widespread in the ocean and play a crucial role in the ecosystem. Automatic identification and location of marine microalgae in microscopy images would help establish marine ecological environment monitoring and water quality evaluation system. We proposed a new dataset for the detection of marine microalgae and a range of detection methods, the dataset including images of different genus of algae and the same genus in different states. We set the number of unbalanced classes in the data set and added images of mixed water samples in the test set to simulate the actual situation in the field. Then we trained, validated and tested the, TOOD, YOLOv5, YOLOv8 and variants of RCNN algorithms on this dataset. The results showed both one-stage and two-stage object detection models can achieve high mean average precision, which proves the ability of computer vision in multi-object detection of microalgae, and provides basic data and models for real-time detection of microalgal cells.

The purification of microplastic samples is a crucial step in environmental research, aiming to remove contaminants and ensure accurate analysis. This study explores various methods for purifying microplastics, focusing on the removal of organic contaminants, specifically fulvic acids. Comparative analysis using microscopic imaging techniques was conducted to evaluate the effectiveness of these methods. The results highlight the complexities involved in microplastic purification and the challenges associated with different approaches. While some methods showed promising outcomes, such as the sonification method, it was also observed that they could potentially alter the microplastic structure. Additionally, the limitations and potential interferences of other purification methods, such as Fenton's solution, were identified. This study underscores the importance of selecting purification methods that minimize negative impacts on microplastics and ensure reliable analysis. Further research is needed to optimize purification techniques and fully understand their effects on microplastic integrity. • We developed a new assay for chitin content analysis in yeasts. • Our assay classified 12 yeast species based on chitin patterns and morphology. • These species were classified into the phylogenetic clades identified by Tsui. • The imaging-cytometry assay exhibited 85 % accuracy. Pathogenic fungi are a major threat to public health, and fungal infections are becoming increasingly common and treatment resistant. Chitin, a component of the fungal cell wall, modifies host immunity and contributes to antifungal resistance. Moreover, chitin content is regulated by chitin synthases and chitinases. However, the specific roles and mechanisms remain unclear. In this study, we developed a cytometric imaging assay to quantify chitin content and identify the distribution of chitin in the yeast cell wall. The Candida albicans SC5314 and Nakaseomyces glabratus (ex. C. glabrata) ATCC2001 reference strains, as well as 106 clinical isolates, were used. Chitin content, distribution, and morphological parameters were analysed in 12 yeast species. Moreover, machine learning statistical software was used to evaluate the ability of the cytometric imaging assay to predict yeast species using the values obtained for these parameters. Our imaging-cytometry assay was repeatable, reproducible, and sensitive to variations in chitin content in C. albicans mutants or after antifungal stimulation. The evaluated parameters classified the yeast species into the correct clade with an accuracy of 85 %. Beneficial plant-microbe interactions offer a sustainable biological solution with the potential to boost low-input food and bioenergy production. A better mechanistic understanding of these complex plant-microbe interactions will be crucial to improving plant production as well as performing basic ecological studies investigating plant-soil-microbe interactions. Here, a detailed description for ecosystem fabrication is presented, using widely available 3D printing technologies, to create controlled laboratory habitats (EcoFABs) for mechanistic studies of plant-microbe interactions within specific environmental conditions. Two sizes of EcoFABs are described that are suited for the investigation of microbial interactions with various plant species, including Arabidopsis thaliana, Brachypodium distachyon, and Panicum virgatum. These flow-through devices allow for controlled manipulation and sampling of root microbiomes, root chemistry as well as imaging of root morphology and microbial localization. This protocol includes the details for maintaining sterile conditions inside EcoFABs and mounting independent LED light systems onto EcoFABs. Detailed methods for addition of different forms of media, including soils, sand, and liquid growth media coupled to the characterization of these systems using imaging and metabolomics are described. Together, these systems enable dynamic and detailed investigation of plant and plant-microbial consortia including the manipulation of microbiome composition (including mutants), the monitoring of plant growth, root morphology, exudate composition, and microbial localization under controlled environmental conditions. We anticipate that these detailed protocols will serve as an important starting point for other researchers, ideally helping create standardized experimental systems for investigating plant-microbe interactions.

“Trim-Away” technology enables rapid degradation of endogenous proteins without prior modification of protein-coding genes or mRNAs through delivery of antibodies that target proteins of interest. Although this approach can be readily applied to almost any cytosolic protein, strategies for cytosolic antibody delivery have been limited to microinjection or electroporation, which require skill-dependent operation or specialized equipment. Thus, the development of antibody delivery methods that are convenient, scalable, and preferably do not require detachment of adherent cells is required to extend the versatility of the Trim-Away method. Here, we developed a cell resealing technique optimized for Trim-Away degradation, which uses the pore-forming toxin streptolysin O (SLO) to permeabilize the cell membrane and delivered the antibodies of interest into HEK293T, HeLa, and HK-2 cell lines. We demonstrated the ability of Trim-Away protein degradation using IKKα and mTOR as targets, and we showed the availability of the developed system in antibody screening for the Trim-Away method. Furthermore, we effectively coupled Trim-Away with cyclic immunofluorescence and microscopic image-based analysis, which enables single-cell multiplexed imaging analysis. Taking advantage of this new analysis strategy, we were able to compensate for low signal-to-noise due to cell-to-cell variation, which occurs in the Trim-Away method because of the heterogenous contents of the introduced antibody, target protein, and TRIM21 in individual cells. Therefore, the reported cell resealing technique coupled with microscopic image analysis enables Trim-Away users to elucidate target protein function and the effects of target protein degradation on various cellular functions in a more quantitative and precise manner.

The decline in the performance of spiral-wound reverse osmosis (SWRO) membranes is frequently due to biofouling. This study focus on qualitative and quantitative diagnosis of SWRO membrane biofouling. Bacterial counts on the different surfaces of the fouled membranes were carried out. Surface enhanced Raman spectroscopy (SERS) was performed to highlight clogging materials as well as their natures and identity. The topography of the fouled membranes and the structures of biofilms were visualized by fluorescence microscopy (FM) and scanning electron microscopy (SEM). The results indicated the presence of bacteria in the different SWRO membrane areas. Those strongly adhered were significantly higher than those weakly. It varied between 26 × 105 and 262 × 105 CFU m−2. However, SERS mapping showed different fouling levels and the thickness of the fouling layer was 5 µm. Periodontal diseases, ranging from gingivitis to periodontitis, are prevalent oral diseases affecting over 50% of the global population. These diseases arise from infections and inflammation of the gums and supporting bones, significantly impacting oral health. The established link between periodontal diseases and systemic diseases, such as cardiovascular diseases, underscores their importance as a public health concern. Consequently, the early detection and prevention of periodontal diseases have become critical objectives in healthcare, particularly through the integration of advanced artificial intelligence (AI) technologies. This paper aims to bridge the gap between clinical practices and cutting-edge technologies by providing a comprehensive review of current research. We examine the identification of causative factors, disease progression, and the role of AI in enhancing early detection and treatment. Our goal is to underscore the importance of early intervention in improving patient outcomes and to stimulate further interest among researchers, bioengineers, and AI specialists in the ongoing exploration of AI applications in periodontal disease diagnosis.

Extracellular polymeric substances (EPS) are an important part of microbial aggregates in wastewater biotreatment. EPS maintain the spatial arrangement of microbial aggregates and greatly affect the structure and properties. The production and distribution of EPS can reflect the attachment and aggregation of microorganisms. Microscopic imaging technique (MIT) can visually show the apparent morphology, content, spatial distribution, adhesion, and adsorption of EPS of microbial aggregates in wastewater biotreatment. First, in the review, the characteristics of EPS are summarized. Then we describe the principles, applications, and features of scanning electron microscopy, transmission electron microscopy, confocal laser scanning microscopy, atomic force microscopy, and their joint techniques in the characterization of the appearance, content, distribution, and properties of EPS in microbial aggregates, to provide reference for MIT application. Although MITs are widely used to characterize EPS properties, there are still problems such as complicated sample preparation, inability to analyze EPS quantitatively, and inability to perform dynamic characterization. To better characterize the EPS in microbial aggregate, we summarized and prospected the MIT. The current trend shows that in the future, real-time observation of EPS changes in microbial aggregates and multitechnique combination to characterize EPS will be developed and applied. This study highlighted the taxonomic utilization of palynological metaphors for selected members (53) of family Poaceae. Multiple microscopic technique light and scanning electrons had been employed for detailed analysis. Results reported monad pollen type in all studied 53 members, which showed its limited taxonomic value up to family level. In relation to shape of pollen both polar and equatorial views strikingly differed from each other. Like semi angular pollen observed in Dactyloctenium aegyptium (L.) Wild. whereas round‐minutely irregular pollen in Lolium temulentum L. Polar and equatorial diameter also showed variation, that is, Desmostachya bipinnata (L.) Stapf. can be differentiated from Sorghum bicolor (L.) Moench on diameter variation basis. A pore characteristic does not show much qualitative variation; however pore sizes differ species to species. The most frequently scarbate sculpturing was observed in 28 species followed by verrucate. Hence it can be said that pollen shape, polar, and equatorial diameters, pore size, P/E ratio, pore sculpturing are of good taxonomic value and holds a significant position in identification and delimitation of Poaceae taxa. Using LAURDAN spectral imaging and spectral phasor analysis we concurrently studied the growth and hydration state of subcellular organelles (lamellar body-like, LB-like) from live A549 lung cancer cells at different post-confluence days. Our results reveal a time dependent two-step process governing the size and hydration of these intracellular LB-like structures. Specifically, a first step (days 1 to 7) is characterized by an increase in their size, followed by a second one (days 7 to 14) where the organelles display a decrease in their global hydration properties. Interestingly, our results also show that their hydration properties significantly differ from those observed in well-characterized artificial lamellar model membranes, challenging the notion that a pure lamellar membrane organization is present in these organelles at intracellular conditions. Finally, these LB-like structures show a significant increase in their hydration state upon secretion, suggesting a relevant role of entropy during this process.

The research on non-invasive ophthalmic imaging has become a hot topic in diagnosing ophthalmic diseases, especially assessment of corneal biomechanics has been an unmet clinical need in ophthalmology for many years. The Brillouin microscopy technique based on spontaneous Brillouin scattering has emerged as a unique elastography method due to its advantages of non-contact, label-free, and high-resolution biomechanical imaging of biological tissues. Here, we developed a three-dimensional (3D) optical microscope system for ex-vivo porcine cornea Brillouin imaging. To do this, we designed and experimentally demonstrated a setup with double-stage VIPA etalons and CMOS camera that integrate to a fully parallel dispersive Brillouin spectrometer. The designed spectrometer exhibits a smaller dispersion rate to enhance the spectral sampling rate and downsizes the size of the double-stage-based system. Using the optical microscope system, we measured and mapped the Brillouin frequency shifts of the cornea and crystalline lens in ex-vivo porcine eyes at micron-scale spatial resolution, and a spectral accuracy of < 50 MHz can be achieved without interpolating the spectra. The sudden outbreak of COVID-19 rapidly developed into a global pandemic, which caused tens of millions of infections and millions of deaths. Although SARS-CoV-2 is known to cause COVID-19, effective approaches to detect SARS-CoV-2 using a convenient, rapid, accurate, and low-cost method are lacking. To date, most of the diagnostic methods for patients with early infections are limited to the detection of viral nucleic acids via polymerase chain reaction (PCR), or antigens, using an enzyme-linked immunosorbent assay or a chemiluminescence immunoassay. This study developed a novel method that uses localized surface plasmon resonance (LSPR) sensors, optical imaging, and artificial intelligence methods to directly detect the SARS-CoV-2 virus particles without any sample preparation. The virus concentration can be qualitatively and quantitatively detected in the range of 125.28 to 106 vp/mL through a few steps within 12 min with a limit of detection (LOD) of 100 vp/mL. The accuracy of the SARS-CoV-2 positive or negative assessment was found to be greater than 97%, and this was demonstrated by establishing a regression machine learning model for the virus concentration prediction (R2 > 0.95). Fluorogens with aggregation-induced emission characteristics (AIEgens) have been developed in an unprecedented manner in the biomedical field over the past decade, which is ascribable to their superior fluorescence quantum yields, photosensitivities, as well as their “turn-on” characteristics in their aggregate states. By deliberately engineering their molecular structures and nanocarriers, the input energies of AIEgens can be channeled into specific forms, thereby enabling bioimaging, phototherapy, and even synergetic therapy. Moreover, multiple targeting strategies have been tailored for nidus and pathogen localization that subsequently guide therapeutic processes. This review integrates application-oriented design strategies for AIEgens, demonstrating how microscopic imaging, bioimaging, and theranostics can be developed to combat tumors, pathogens, and other diseases, and critically discusses challenges and perspectives for clinical translation.

The objective of the study was to evaluate the effect of trichome (fuzz) removal on the efficacy of ultraviolet-C in inactivating Escherichia coli O157:H7 on peach fruit, and quality of peach [Prunus persica (L.) Batsch, cv. PF25] fruit as affected by fuzz removal and ultraviolet-C. Peach (cultivar PF25) fruit, with and without fuzz removal, were inoculated with a five-strain cocktail of E. coli O157:H7 and treated with ultraviolet-C at doses of 0, 221, and 442 mJ/cm2. Fuzz was rubbed off using damped cloths. Survival of E. coli populations was determined at days 1, 4, and 7 at 20 °C. To study fruit quality, noninoculated fruit with and without fuzz removal were treated with ultraviolet-C at the same doses. Results demonstrated that ultraviolet-C at 442 mJ/cm2 reduced the population of E. coli by 1.2 to 1.4 log colony-forming units (CFU)/fruit on peach with fuzz, and 0.9 to 1.1 log CFU/fruit on fruit without fuzz 1 day after ultraviolet-C treatment. However, E. coli populations of all samples were similar with additional storage time, resulting in no significant difference among the treatments after 7 days of storage at 20 °C. Ultraviolet-C at doses up to 442 mJ/cm2 did not have any significant effect on the surface color of peaches during 7 days of storage, although fruit with fuzz removal increased L*, hue, and chroma values. In addition, fuzz removal promoted the loss of firmness during storage. Furthermore, ultraviolet-C at 442 mJ/cm2 increased antioxidant capacity of peach skin with fuzz. Overall, our results suggested that fuzz removal had marginal effects on the efficacy of ultraviolet-C, and ultraviolet-C did not negatively affect the quality of peaches.

Abstract Background Dormant chemotherapy-resistant leukemia cells can survive for an extended period before relapse. Nevertheless, the mechanisms underlying the development of chemoresistance in vivo remain unclear. Methods Using intravital bone imaging, we characterized the behavior of murine acute myeloid leukemia (AML) cells (C1498) in the bone marrow before and after chemotherapy with cytarabine. Results Proliferative C1498 cells exhibited high motility in the bone marrow. Cytarabine treatment impaired the motility of residual C1498 cells. However, C1498 cells regained their migration potential after relapse. RNA sequencing revealed that cytarabine treatment promoted MRTF-SRF pathway activation. MRTF inhibition using CCG-203971 augmented the anti-tumor effects of chemotherapy in our AML mouse model, as well as suppressed the migration of chemoresistant C1498 cells. Conclusions These results provide novel insight into the role of cell migration arrest on the development of chemoresistance in AML, as well as provide a strong rationale for the modulation of cellular motility as a therapeutic target for refractory AML.

In this study, two different types of amorphous carbonaceous Precambrian rock, classified as noble elite shungite and black raw shungite, were tested as possible electrode materials of natural origin. Both types were machined into cylindrical shapes to form the corresponding solid electrodes and their physicochemical and electrochemical properties were compared with the standard glassy carbon electrode (GCE). The raw stones were first subjected to microscopic imaging by using scanning electron microscopy and energy-dispersive X-ray spectroscopy, both of which indicated significant differences in their morphology and in the content of impurities. An electrode prototype manufactured from noble elite shungite (EShE) with a carbon content of about 94% (w/w) has offered a very satisfactory electrochemical performance with a nearly identical heterogeneous electron-transfer rate constant of 7.8 × 10−3 cm s−1 for ferro/ferricyanide redox couple, a slightly narrower potential range (~2.1 V) and a relatively low double-layer capacitance (of ca. 50 μF), resulting in low background currents comparable to those at the GCE. In contrast, the second electrode based on black raw shungite (BShE) with a carbon content of ca. 63% (w/w) exhibited markedly worse electrochemical properties and more than four times higher double-layer capacitance, both of which were probably due to the presence of poorly conductive impurities. The whole study has been completed with three different examples of electroanalytical applications, revealing that the first type, EShE, is a more suitable material for the preparation of electrodes and may represent a cheap alternative to commercially marketed products.

While many clinical laboratory tests are now highly automated, body fluid cell counting, particularly in low-cellularity samples such as cerebral spinal fluid (CSF), is often performed manually. Here, we report a simple, cost-effective method to obtain white and red blood cell counts from human body fluids such as CSF. The method consists of a compact, automated, and low-cost fluorescence microscope system, coupled to a sample chamber containing all of the necessary reagents in dry form to stain and prepare the sample. Sample focus and scanning are handled automatically, and the acquired multimodal images are automatically analyzed to extract cell counts. Comparison with manual counting on over 200 clinical samples shows excellent agreement. As the system counts a substantially larger image region than a standard manual cell count, we find our sensitivity to extremely low cellularity samples to potentially be higher than the manual gold standard, evidenced by our system recording images of cells in samples whose cell count was registered as "0" by a trained user. Thus, our system holds promise for routine, automated, and sensitive analysis of body fluids whose cellularity extends across a wide dynamic range. [ABSTRACT FROM AUTHOR]

For improving the detection accuracy of wear debris visual information for online wear monitoring, a full field-of-view Online Visual Ferrograph (OLVF) debris detector is presented. In current OLVF debris detector, a semi-transparent optical diffusion film is clamped into the gap between the bottom optical quartz window and two magnetic poles as the background of full field-of-view OLVF microscopic imaging, by which the emitted light of circular ring LED array is diffusely reflected. A reflected light microscopic imaging system is developed to form the full field-of-view imaging of OLVF, and a mathematical relationship between its depth of field and the oil-pumping and oil-free axial deviations of CMOS image plane is established, by which the oil chamber height can be determined. An optical focusing mechanism is applied to adjusting the optical magnification and the optical resolution of about $7.2~\mu \text{m}$ is derived under $2.0\times $ optical magnification. Moreover, a microscopic imaging model is proposed to evaluate the characteristics of full field-of-view OLVF microscopic imaging. The simulation results show that the high-quality microscopic imaging can be obtained near the optical axis of CMOS image plane under $2.0\times $ optical magnification and oil attenuation coefficient of less than 2.0. Two experiments were conducted to validate the microscopic imaging performance and effectiveness of the current detector. The results show that the detector can be applied for capturing the full field-of-view reflected ferrogram of wear debris from different in-use lube oils, and it can also detect debris concentration more accurately for monitoring the fast-changing wear of the machines. To tackle the challenges faced by human-operated microscopy, deep-learning-based methods have been proposed for microscopic image analysis of a wide range of microorganisms, including viruses, bacteria, fungi, and parasites. Electrochemical and mechanical properties of lithium-ion battery materials are heavily dependent on their 3D microstructure characteristics. A quantitative understanding of the role played by stochastic microstructures is critical for the prediction of material properties and for guiding synthesis processes. Furthermore, tailoring microstructure morphology is also a viable way of achieving optimal electrochemical and mechanical performances of lithium-ion cells. To facilitate the establishment of microstructure-resolved modeling and design methods, a review covering spatially and temporally resolved imaging of microstructure and electrochemical phenomena, microstructure statistical characterization and stochastic reconstruction, microstructure-resolved modeling for property prediction, and machine learning for microstructure design is presented here. The perspectives on the unresolved challenges and opportunities in applying experimental data, modeling, and machine learning to improve the understanding of materials and identify paths toward enhanced performance of lithium-ion cells are presented. We present a wide-field interferometric imaging module for biomedical and metrological measurements, employing shearing interferometry with constant off-axis angle (SICA) that can work, for the first time, with a low-coherence light source. In the SICA module, the shearing distance between the interfering beams can be fully controlled without a direct relation with the off-axis angle. In contrast to our previous SICA module, here we use a low-coherence illumination source, providing quantitative phase profiles with significantly lower spatial coherent noise. Although a low-coherence source is used, we obtain off-axis interference on the entire camera sensor, where the optical path difference between the two beams is compensated by using a glass window positioned in the confocal plane. This highly stable, common-path, low-coherence, single-shot interferometric module can be used as an add-on unit to a conventional bright-field microscope illuminated by a low-coherence source. We demonstrate the advantages of using the module by quantitative phase imaging of a polymer bead, fluctuations in a human white blood cell, and dynamics of human sperm cells.

Automating the classification of camera-obtained microscopic images of White Blood Cells (WBCs) and related cell subtypes has assumed importance since it aids the laborious manual process of review and diagnosis. Several State-Of-The-Art (SOTA) methods developed using Deep Convolutional Neural Networks suffer from the problem of domain shift - severe performance degradation when they are tested on data (target) obtained in a setting different from that of the training (source). The change in the target data might be caused by factors such as differences in camera/microscope types, lenses, lighting-conditions etc. This problem can potentially be solved using Unsupervised Domain Adaptation (UDA) techniques albeit standard algorithms presuppose the existence of a sufficient amount of unlabelled target data which is not always the case with medical images. In this paper, we propose a method for UDA that is devoid of the need for target data. Given a test image from the target data, we obtain its ‘closest-clone’ from the source data that is used as a proxy in the classifier. We prove the existence of such a clone given that infinite number of data points can be sampled from the source distribution. We propose a method in which a latent-variable generative model based on variational inference is used to simultaneously sample and find the ‘closest-clone’ from the source distribution through an optimization procedure in the latent space. We demonstrate the efficacy of the proposed method over several SOTA UDA methods for WBC classification on datasets captured using different imaging modalities under multiple settings. Nonlinear optics, which is a subject for studying the interaction between intense light and materials, has great impact on various research fields. Since many structures in biological tissues exhibit strong nonlinear optical effects, nonlinear optics has been widely applied in biomedical studies. Especially in the aspect of bio-imaging, nonlinear optical techniques can provide rapid, label-free and chemically specific imaging of biological samples, which enable the investigation of biological processes and analysis of samples beyond other microscopy techniques. In this review, we focus on the introduction of nonlinear optical processes and their applications in bio-imaging as well as the recent advances in this filed. Our perspective of this field is also presented.

Natural products (NPs) are an important source for new drug discovery over the past decades, which have been demonstrated to be effectively used in cancer prevention, treatment, and adjuvant therapy. Many methods, such as the genomic and metabolomic approaches, immunochemistry, mass spectrometry, and chromatography, have been used to study the effects of NPs on cancer as well as themselves. Because of the advantages in specificity, sensitivity, high throughput, and cost-effectiveness, optical imaging (OI) approaches, including optical microscopic imaging and macroscopic imaging techniques have also been applied in the studies of NPs. Optical microscopic imaging can observe NPs as cancer therapeutics at the cellular level and analyze its cytotoxicity and mechanism of action. Optical macroscopic imaging observes the distribution, metabolic pathway, and target lesions of NPs in vivo, and evaluates NPs as cancer therapeutics at the whole-body level in small living animals. This review focuses on the recent advances in NPs as cancer therapeutics, with particular emphasis on the powerful use of optical microscopic and macroscopic imaging techniques, including the studies of observation of ingestion by cells, anticancer mechanism, and in vivo delivery. Finally, we prospect the wider application and future potential of OI approaches in NPs as cancer therapeutics.

The structural modeling and representation of cells is a complex task as different microscopic, spectroscopic and other information resources have to be combined to achieve a three-dimensional representation with high accuracy. Moreover, to provide an appropriate spatial representation of the cell, a stereoscopic 3D (S3D) visualization is favorable. In this work, a structural cell model is created by combining information from various light microscopic and electron microscopic images as well as from publication-related data. At the mesoscopic level each cell component is presented with special structural and visual properties; at the molecular level a cell membrane composition and the underlying modeling method are discussed; and structural information is correlated with those at the functional level (represented by simplified energy-producing metabolic pathways).

The mechanical performance of bamboo is highly dependent on its structural arrangement and the properties of biomacromolecules within the cell wall. The relationship between carbohydrates topochemistry and gradient micromechanics of multilayered fiber along the diametric direction was visualized by combined microscopic techniques. Along the radius of bamboo culms, the concentration of xylan within the fiber sheath increased, while that of cellulose and lignin decreased gradually. At cellular level, although the consecutive broad layer (Bl) of fiber revealed a relatively uniform cellulose orientation and concentration, the outer Bl with higher lignification level has higher elastic modulus (19.59–20.31 GPa) than that of the inner Bl close to the lumen area (17.07–19.99 GPa). Comparatively, the cell corner displayed the highest lignification level, while its hardness and modulus were lower than that of fiber Bl, indicating the cellulose skeleton is the prerequisite of cell wall mechanics. The obtained cytological information is helpful to understand the origin of the anisotropic mechanical properties of bamboo.
