Botryosphaeriaceae species are the major causal agents of walnut dieback worldwide, along with Diaporthe species. Botryosphaeria dothidea and Neofusicoccum parvum are the only two Botryosphaeriaceae species associated with this recently emergent disease in France, and little is known about their diversity, structure, origin and dispersion in French walnut orchards. A total of 381 isolates of both species were genetically typed using a sequence-based microsatellite genotyping (SSR-seq) method. This analysis revealed a low genetic diversity and a high clonality of these populations, in agreement with their clonal mode of reproduction. The genetic similarity among populations, regardless of the tissue type and the presence of symptoms, supports the hypothesis that these pathogens can move between fruits and twigs and display latent pathogen lifestyles. Contrasting genetic patterns between N. parvum populations from Californian and Spanish walnut orchards and the French ones suggested no conclusive evidence for pathogen transmission from infected materials. The high genetic similarity with French vineyards populations suggested instead putative transmission between these hosts, which was also observed with B. dothidea populations. Overall, this study provides critical insight into the epidemiology of two important pathogens involved in the emerging dieback of French walnut orchards, including their distribution, potential to mate, putative origin and disease pathways.

Tandem repeats are frequent across the human genome, and variation in repeat length has been linked to a variety of traits. Recent improvements in long read sequencing technologies have the potential to greatly improve tandem repeat analysis, especially for long or complex repeats. Here, we introduce LongTR, which accurately genotypes tandem repeats from high-fidelity long reads available from both PacBio and Oxford Nanopore Technologies. Habitat fragmentation reduces gene flow, causing genetic differentiation and diversity loss in endangered species through genetic drift and inbreeding. However, the impact of habitat fragmentation on ectomycorrhizal (ECM) fungi remains unexplored, despite their critical roles in forest ecosystems. Here, we investigated the population genetic structure and the demographic history of Rhizopogon togasawarius, the ECM fungus specifically colonizing the host tree Pseudotsuga japonica, across its entire distribution range (>200 km). These two species are designated as endangered species on the IUCN Red List since they are found only in small, fragmented forests in Japan. We analysed 236 R. togasawarius individuals from five remaining populations across the Kii Peninsula and the Shikoku Island, separated by a sea channel. Simple sequence repeat analyses using 20 loci revealed strong genetic differentiation among populations (FST = 0.255), even significant in the nearest population pair separated by a distance of only 8 km (FST = 0.075), indicating extremely limited gene flow between populations. DIYABC‐RF analyses implied that population divergence occurred approximately 6000 generations ago between the two regions, and nearly 1500 generations ago between the nearest populations within Shikoku Island, related to past climate events. Because of prolonged genetic isolation, significant inbreeding was confirmed in four of five populations, where effective population sizes became very small (Ne = 9.0–58.0). Although evaluation of extinction risks for microorganisms is challenging, our conservation genetic results indicated that habitat fragmentation increases extinction risk through population genetic mechanisms, and therefore should not be overlooked in biodiversity conservation efforts. Introduction: The first small scale cultivation of potatoes in the Nordic countries began roughly 300 years ago, and later became an important staple food in the region. Organized conservation efforts began in the 1980s, and today, potato landraces, improved varieties, and breeding lines are conserved in genebanks at the Nordic Genetic Resource Center (NordGen), Sweden, and the Norwegian Genetic Resource Centre (NGS), Norway, as well as at potato breeding companies across Nordic countries. All these collections house a diverse array of genotypes with local names and local growing histories from the whole region. However, the presence of duplicates, and inconsistent naming has led to confusion. Methods: In this study, 198 accessions of cultivated potato (Solanum tuberosum L.) have been genotyped with 62 microsatellite (SSR) markers. The analyzed accessions came from three collections: 43 accessions from the Danish Potato Breeding Foundation in Vandel (LKF-Vandel), 90 from NordGen and 65 from NGS. Results and discussion: The genetic analysis revealed 140 unique potato genotypes and 31 groups/clusters of duplicates, most of which contained duplicate pairs and the others three to ten accessions. Several accessions with distinct names were genetically identical or very similar, suggesting historical sharing, and regional distribution of seed potatoes, leading to the emergence of diverse local names. Moreover, many improved varieties from early potato breeding were revealed to have duplicates that have been considered Nordic landraces. Furthermore, potato accessions with identical names but originating from different collections were confirmed to be duplicates. These findings have already influenced management decisions and will further improve management practices for Nordic potato collections. The importance of monitoring and management of yellow rust, caused by Puccinia striiformis f. sp. tritici (Pst), has markedly increased in Russia over recent years because of intensifying negative effects of this destructive disease on wheat production. The Pst virulence survey was conducted in 2019–2021 in the European (North‐West, Volga, North Caucasus) and Asian (Siberia) parts of Russia, and the same 109 isolates have been genotyped here with 20 SSR markers. Fifty‐three multilocus genotypes (MLGs) were detected, seven of which occurred in two or more regions. The most frequent MLG was found in three European Pst collections (except Dagestan) and Siberia. SSR‐based variation within the regional collections was low because many MLGs were closely related. Nevertheless, each collection contained relatively different small genotype groups. No clear geographic structure of the Russian Pst population was observed because the five studied regional collections shared groups of similar genotypes. The most singular SSR genotype was identified in the North‐West collection, but genotypes of four invasive isolates belonging to the PstS2 temperature‐adapted aggressive strain did not considerably differ from others. The observed heterozygosity at all SSR loci exceeded the expected one in all regional Pst collections, which could imply asexual reproduction of the pathogen. Both the SSR and virulence analyses demonstrated similar patterns of relationships between the regional Pst collections, although the magnitude of differences between the collections was larger for virulence pathotypes. However, no significant association was established between the virulence pathotypes and SSR genotypes because multiple pathotypes had the same genotype and vice versa. [ABSTRACT FROM AUTHOR]

Arboreal ants are ecologically important in tropical forests, but there are few studies using DNA markers to examine their population and colony structure. Colonies of the arboreal turtle ant Cephalotes goniodontus create trail networks through the canopy of the tropical forest, in dense vegetation where it is difficult to determine how long a nest is used and how neighboring colonies partition space. We monitored 53 nest sites for up to six years and, using seven microsatellite markers, genotyped samples of workers collected at or near 41 nests over 1–4 years. We calculated average relatedness within samples collected at a given location, and between samples collected at the same location in successive years, and performed pedigree analysis to predict the number of queens that produced each sample of workers. Fifteen samples were highly related (r ≥ 0.6) from single colonies, of which 11 were monogynous and the remaining four had two queens; 19 were of intermediate relatedness (0.1 ≤ r < 0.6) with 1–6 queens, and 7 were groups of unrelated workers (r < 0.1) from at least 4 queens. Colonies persisted at the same nest site for 2–6 years. The smallest distance we found separating nests of different colonies was 16.2 m. It appears that different colonies may share foraging trails. Our study demonstrates the feasibility of using a cost-efficient genotyping method to provide information on colony structure and life history of ant species. [ABSTRACT FROM AUTHOR]

Population genetic analysis of invasive populations can provide valuable insights into the source of introductions, pathways for expansion, and their demographic histories. Flathead catfish (Pylodictis olivaris) are a prolific invasive species with high fecundity, long‐distance dispersal, and piscivorous feeding habits that can lead to declines in native fish populations. In this study, we analyse the genetics of invasive P. olivaris in the Mid‐Atlantic region to assess their connectivity and attempt to reconstruct the history of introduced populations. Based on an assessment across 13 microsatellite loci, P. olivaris from the Susquehanna River system (N = 537), Schuylkill River (N = 33), and Delaware River (N = 1) have low genetic diversity (global Hobs = 0.504), although we detected no evidence of substantial inbreeding (FIS = −0.083 to 0.022). P. olivaris from these different river systems were genetically distinct, suggesting separate introductions. However, population structure was much weaker within each river system and exhibited a pattern of high connectivity, with some evidence of isolation by distance. P. olivaris from the Susquehanna and Schuylkill rivers showed evidence for recent genetic bottlenecks, and demographic models were consistent with historical records, which suggest that populations were established by recent founder events consisting of a small number of individuals. Our results show the risk posed by small introductions of P. olivaris, which can spread widely once a population is established, and highlight the importance of prevention and sensitive early detection methods to prevent the spread of P. olivaris in the future. Simple Summary: This study genetically characterizes the Cape Verde palm tree, Phoenix atlantica, exploring its genetic differentiation and recent divergence from its relative, Phoenix dactylifera, while also examining its significance for conservation efforts and enhancing our understanding of the historical trajectories of African palm trees. The Cape Verde palm tree, Phoenix atlantica, holds significant ecological and cultural importance within the Cape Verde archipelago. However, its genetic distinctiveness has been questioned due to its close relationship and morphological similarity to the date palm (Phoenix dactylifera). In this study, we used an expanded sample set, 18 simple sequence repeat (SSR) markers, and a plastid minisatellite to characterize P. atlantica in Cape Verde and investigate its relationship with other Phoenix species. Our findings identify genetic markers that differentiate the P. atlantica genetic pool, including a unique fixed allele. We also provide evidence of the recent divergence of P. atlantica from Northern African date palm populations, suggesting a relatively recent colonization of Cape Verde by palm trees. Additionally, we characterized the genetic composition of palm tree populations across three Cape Verde islands, concluding that wild samples from certain populations in Boavista and Sal are best suited for establishing a seed and/or germplasm bank for replantation efforts, representing a crucial step for the conservation of Cape Verde's natural heritage. Overall, our results enhance the understanding of the historical trajectories and genetic characterization of palm trees in Africa, offering valuable insights for conservation strategies. Key message: Populations of the European white elm (Ulmus laevis Pall.) in Switzerland can be considered natural. They show no evidence of genetic differentiation from other European populations. In the past, the U. laevis populations were probably more widespread and continuous in Switzerland with a larger gene flow. Context: In Switzerland, at the margin of its distribution range, U. laevis is rare and considered endangered. Whether the species is native to Switzerland has been disputed, and it is often surmised to be solely cultivated, without any natural population in the country. Aims: The structure of genetic diversity among Swiss populations of U. laevis and comparison to European populations are expected to shed light on the origin of local populations and support their management. Methods: We analyzed 19 populations (194 individuals) in Switzerland and 15 populations (158 individuals) from other European countries, using a set of five microsatellite loci. Results: (1) 90% of the genetic variation in European and Swiss populations occurs within populations. (2) We did not detect isolation by distance at the regional or continental scale. (3) Clustering analysis did not reveal any spatial pattern in the level of admixture of individuals within Swiss or other European populations. Conclusion: Moderate levels of genetic diversity and evidence for recent gene flow between populations indicate that habitat deterioration, loss, and fragmentation are the main threats to the persistence of U. laevis populations in Switzerland. Winter flounder Pseudopleuronectes americanus (Walbaum 1792) are a coastal flatfish species of economic and cultural importance that have dwindled to <15, % of their historic abundance in the southern New England/Mid‐Atlantic region of the United States, with evidence indicating near‐extirpation of certain local populations. This species exhibits intricate behaviors in spawning and migration that contribute to population complexity and resilience. These behaviors encompass full or partial philopatry to natal estuaries, the generation of multiple pulses of larval delivery, and partial migration. The patterns of genetic diversity within and among estuaries and cohorts presented here carry important implications in understanding the susceptibility to demographic shocks, even if the full extent of genetic diversity within and among winter flounder stocks on the US East Coast remains unresolved. Our findings reveal connectivity between estuaries in Long Island, New York, suggesting the potential for genetic rescue of depleted subpopulations. Family reconstruction and relatedness analysis indicate that split cohorts and migration contingents are not the result of genetically distinct lineages. We found no evidence for genetic structure separating these groups, and in some instances, we were able to detect closely related individuals that belonged to different migratory contingents or cohorts. Characterizing the spatial and behavioral organization of this species at the population level is crucial for comprehending its potential for recovery, not only in terms of biomass but also in reinstating the complex population structure that supports resilience. The search for generality in winter flounder spawning and migration behavior remains elusive, but perhaps the lack of generalities within this species is what has allowed it to persist in the face of decades of environmental and anthropogenic stressors. There is a general expectation that urban populations will be fragmented and the movement of individuals will be restricted leading to low effective population sizes, low genetic diversity, higher inbreeding, and higher differentiation than populations living in more continuous habitat. In this study, we compare the genetic diversity and differentiation of Texas horned lizards that are found in four small towns (Kenedy, Karnes City, Rockdale, and Smithville) in Texas and at Tinker Air Force Base, Oklahoma to populations that occur in 16 natural areas and to an introduced population in South Carolina. We also present more detailed spatial genetic data and home range data for several of the towns. Texas horned lizards (Phrynosoma cornutum) living in small towns have lower genetic diversity, higher differentiation, and smaller effective population sizes than populations located in more natural areas. There was evidence for human‐mediated movement of lizards into town; however, it has not been enough to counteract the effects of drift. Home range size is smaller in town than in more natural areas. Genetic patterns suggest dispersal occurs over short distances and is inhibited across areas with a high percent of impervious surface and major roads. These data suggest that effective planning to maintain suitable habitat and corridors to facilitate movement is critical to maintaining small terrestrial species like the Texas horned lizard and must be integrated into the early stages of urban development. The conservation biology field underscores the importance of understanding genetic diversity and gene flow within plant populations and the factors that influence them. This study employs Simple Sequence Repeat (SSR) molecular markers to investigate the genetic diversity of the endangered plant species Saussurea involucrata, offering a theoretical foundation for its conservation efforts. Utilizing sequencing results to screen SSR loci, we designed and scrutinized 18 polymorphic microsatellite primers across 112 samples from 11 populations in the Bayinbuluke region. Our findings reveal high genetic diversity (I = 0.837, He = 0.470) and substantial gene flow (Nm = 1.390) among S. involucrata populations (China, Xinjiang), potentially attributed to efficient pollen and seed dispersal mechanisms. Principal Coordinate Analysis (PCoA) indicates a lack of distinct genetic structuring within the Bayinbuluke populations. The cluster analysis using STRUCTURE reflected the genetic structure of S. involucrata to a certain extent compared with PCoA. The results showed that all samples were divided into four groups. To safeguard this species, we advocate for the in situ conservation of all S. involucrata populations in the area. Many mammals have a promiscuous mating system with multiple sired litters. Promiscuity can increase the genetic variability, reduce the risk of inbreeding, and increase the effective population size, and is therefore crucial for preventing genetic loss and maintaining adaptability. This is particularly true for small and threatened populations. The garden dormouse (Eliomys quercinus) is a threatened species, which exhibited a drastic decline over the last 20–30 years. The aim of this study was therefore to investigate the mating system of the garden dormouse in mountain forest habitat by parentage analyses using 5 polymorphic microsatellite markers combined with morphometric data and information about the nesting behavior. Genetic parentage analyses revealed that 64% (9 of 14) of the litters were sired by at least two males, suggesting that garden dormice have a promiscuous mating system. The genetic findings were further supported by indirect indicators of promiscuity, such as testes, that were nearly four times larger than predicted for a rodent of its body mass and only slight male biased sexual size dimorphism. The finding of a promiscuous mating system in garden dormice should be taken into account in future conservation efforts. Due to its habitat preferences and limited dispersal potential garden dormice are vulnerable to forest fragmentation. The connection of suitable habitats facilitates dispersal and promotes access to potential mating partners, which could be especially important for populations colonizing new habitats. The pink river dolphin, or bufeo, is one of the dolphins which lives in the rivers of the Orinoco and Amazon basins in South America. The Bolivian bufeo population is considered a differentiated species (Inia boliviensis) from the Amazon and Orinoco species (Inia geoffrensis). Until now, no study has completed an extensive population genetics analysis of the bufeo in Bolivian rivers. We analyzed 82 bufeos from different rivers from the Mamoré and Iténez (Guaporé) river basins for the mt control region (CR), nuclear microsatellites, and DQB-1 gene sequences to determine if the inner rapids of these Bolivian river basins have some influence on the genetic structure of this species. The first relevant result was that the genetic diversity for CR, and the microsatellites were substantially lower in the Bolivian bufeos than in the dolphins studied in other areas of the Amazon and Orinoco. However, the DQB-1 gene sequences yielded similar genetic diversity to those found in other areas. The second relevant result is the existence of some significant genetic heterogeneity among the bufeo populations within Bolivia, although in a small degree, but this differentiation is independent of the inner rapids of the Bolivian rivers we sampled. The third relevant result was the existence of significant isolation by distance for the CR, but not for microsatellites and DQB-1 gene sequences. This was related to differential gene flow capacity of females (philopatric) and males (less philopatric and more migrants) and, possibly, to different selective patterns affecting the molecular markers studied. The fourth relevant result was related to diverse demographic changes of these bufeos. At least two or three bottleneck events and one or two population expansions have occurred in the Bolivian bufeo population. The major part of these events occurred during the Pleistocene. Abstract—In this study we present the results of a comparative genetic analysis of bees of the Apismelliferacaucasica subspecies with the subspecies A.m.carnica and A.m.mellifera. We performed polymorphism analysis of nine microsatellite loci (Ap243, 4a110, A24, A8, A113, A88, Ap049, A28, and A43) and determined the haplotypes of the tRNAleu-COII locus. Analysis of the genetic structure of representatives of three subspecies of honey bees that are widespread in Russia showed a significant level of their differentiation even when using a small set of microsatellite loci. An assessment of the prevalence of tRNAleu-COII haplotypes in the three studied samples showed that for A.m.caucasica the predominant haplotype was C2j. [ABSTRACT FROM AUTHOR]

Wild species living in captivity are subject to loss of genetic diversity, inbreeding depression, and differentiation among populations. Only very few species have been under human care for centuries but have not been selectively bred, have free-ranging movements most of the time, and retain porous barriers to gene flow between wild and captive populations. Such captive populations are expected to retain high levels of genetic diversity and anthropogenic factors should result in a limited genetic differentiation from wild populations. Asian elephants have been trained and used by humans for at least 4000 years as war animals, mounts of kings and draught animals. In Myanmar and Laos, elephants are still being used for hauling timber in the forest while retaining traditional management practices including seasonal release, free mating and movement. However, habitat fragmentation, isolation and reduced gene flows are threatening both semi-captive and wild pools. We genotyped 167 semi-captive elephants from Laos and Myanmar using a panel of 11 microsatellite loci to estimate the genetic diversity and population structure. We found that elephants of both countries presented high levels of genetic diversity and a low degree of inbreeding, if any. This agrees with the expected high level of genetic diversity in semi-captive populations. We found a weak differentiation along a geographical gradient from southern Laos to northern Myanmar but no differentiation between wild-caught and captive-born pools. The potential value for conservation of a large population of semi-captive elephants has been recognized but the conservation community has yet to fully explore the potential role semi-captive elephants could play in maintaining gene flows. Better knowledge of genetic relationships between the Fortymile caribou herd and its neighbors is needed for conservation decision-making in Canada. Here, we contribute the first fine-scale analysis of genetic population structure in nine contiguous caribou herds at the geographic boundaries between Barren-ground and Northern Mountain caribou, and at the Alaska-Yukon border. Using pairwise differentiation metrics, STRUCTURE, and discriminant analysis of principal components (DAPC) to analyze 15 microsatellite loci in 379 caribou, we found complex patterns of genetic differentiation. The Fortymile was the only herd assigned to more than one genetic cluster, indicative of its history as a larger herd whose range expansions and gene flow to other herds were likely important to maintaining diversity across a functioning genetic metapopulation. Some small herds (Chisana, Klaza, and White Mountains) were genetically distinct, while others (Hart River, Clear Creek, Mentasta) exhibited little differentiation from herds they occasionally overlap, including herds assigned to different conservation units (DUs). This genetic connectivity does not result from demographic connectivity, as episodic contact during rut, rather than herd switching, is the likely mechanism. Unusually, one small herd (White Mountains) maintained genetic differentiation despite rut overlap with Fortymile. Our data reveal that some herds with different ecological and behavioral attributes are demographically independent but nonetheless genetically connected. Thus, we suggest that managing caribou for an appropriate level of genetic connectivity, while also supporting herd persistence, will be essential to conserve caribou genetic diversity in the region. Maâmora is a cork oak forest in Morocco known for its production of sweet acorns. Genetic diversity and relationships were evaluated in 240 individuals from five Cantons. The nine inter simple sequence repeat (ISSR) primers used in this study generated 368 polymorphic fragments and showed 100% polymorphism. A moderate level of genetic diversity (I = 0.28, He = 0.173) was observed. The analysis of molecular variance (AMOVA) showed that the highest level of diversity occurred within the populations (75%). A genetic structure was obtained using unweighted pair group method with arithmetic mean (UPGMA) and principal coordinate analysis (PCoA) analyses. 39 alleles were generated with a mean of 557 alleles per locus using seven simple sequence repeats (SSR) markers to establish DNA-fingerprinting for genotyping of 47 "Plus" tree systematically characterized. The analysis of diversity revealed significant polymorphism (94%) and allelic richness with high observed heterozygosity, which was confirmed by the negative value of Fis (− 0319). Nm is equal to 10.23, and Gst and Fst are equal to 0.022 and 0.052, respectively, revealing low inter-population variability. Genetic typing of the entire collection of "Plus" trees has identify 47 different genotypes. Eight unique alleles and 16 genotypes with specific markers were identified in this study. Humans and wildlife experience complex interactions in urban ecosystems, favoring the presence of commensal species, among which invasive species are particularly successful. Rodents are the main vertebrate group introduced to oceanic islands, where the invasion process and dispersal patterns strongly influence their evolutionary and genetic patterns. We evaluated the house mouse Mus musculus and the black rat Rattus rattus on Cozumel Island, Mexico. We assessed genetic diversity and structure, connectivity, gene flow, relatedness and bottleneck signals based on microsatellite loci. Our genetic findings suggest that introduction of individuals of different geographic sources to the island promotes high allelic diversity and the effective establishment of migrants. We identified a clear genetic structure and low connectivity for the two species, tightly linked with anthropogenic and urban features. Notably, we found that the genetic structure of the house mouse sampled within the city of San Miguel Cozumel is associated with the historical human population growth pulses accompanying the urbanization of the city. At the fine-scale genetic level, the main urban drivers of connectivity of the house mouse were both the impervious land surfaces, i.e. the urban landscape, and the informal commerce across the city (a proxy of resources availability). Chances of a secondary invasion to natural environments have been relatively low, which is crucial for the endemic taxa of the island. The Doradidae fishes constitute one of the most diverse groups of Neotropical freshwater environments. Acanthodoradinae is the oldest lineage and the sister group to all other thorny catfishes, and it includes only the genus Acanthodoras. The diversity of Acanthodoras remains underestimated, and the use of complementary approaches, including genetic studies, is an important step to better characterize this diversity and the relationships among the species within the genus. Therefore, we conducted a comprehensive analysis using conventional cytogenetic techniques and physical mapping of three multigene families (18S and 5S ribosomal DNA [rDNA], U2 small nuclear DNA [snDNA]) and four microsatellite motifs, namely (AC)n, (AT)n, (GA)n, and (GATA)n, in two sympatric species from the Negro River: Acanthodoras cataphractus and Acanthodoras cf. polygrammus. We found significant differences in constitutive heterochromatin (CH) content, distribution of the microsatellite (AT)n, and the number of 5S rDNA and U2 snDNA sites. These differences may result from chromosome rearrangements and repetitive DNA dispersal mechanisms. Furthermore, the characterization of the diploid number (2n) of these Acanthodoras species enables us to propose 2n = 58 chromosomes as the plesiomorphic 2n state in Doradidae based on ancestral state reconstruction. Acanthodoradinae is the oldest lineage of the thorny catfishes, and knowledge about its cytogenetic patterns is crucial for disentangling the karyotype evolution of the whole group. Thus, this study contributes to the understanding of the mechanisms behind chromosome diversification of Doradidae and highlights the importance of Acanthodoradinae in the evolutionary history of thorny catfishes. Within the whitefly Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) complex, two cryptic species, namely Middle East-Asia Minor 1 (MEAM1) and Mediterranean (MED), are important invasive pests affecting global agriculture and horticulture. They were introduced into China sequentially in the mid-1990s and around 2003, respectively. Subsequently, the latter invader MED has outcompeted the earlier invader MEAM1, becoming the dominant population in the field. Although extensive studies have explored the underlying mechanisms driving this shift, the contribution of population genetics remains notably underexplored. In this study, we analyzed the genetic diversity and structure of 22 MED and 8 MEAM1 populations from various regions of China using mitochondrial DNA sequencing and microsatellite genotyping. Our results indicate low and moderate levels of genetic differentiation among geographically separate populations of MED and MEAM1, respectively. Median-joining network analysis of mtCOI gene haplotypes revealed no clear geographic structuring for either, with common haplotypes observed across provinces, although MED had more haplotypes. Comparative analyses revealed that MED presented greater genetic diversity than MEAM1 on the basis of two markers. Furthermore, analysis of molecular variance supported these findings, suggesting that while some genetic variation exists between populations, a significant amount is also present within populations. These findings reveal the population genetics of the two invasive cryptic species of the B. tabaci complex in China and suggest that the disparities in genetic diversity drive the displacement of their populations in the field. Background: The Aedes albopictus mosquito is of medical concern due to its ability to transmit viral diseases, such as dengue and chikungunya. Aedes albopictus originated in Asia and is now present on all continents, with the exception of Antarctica. In Mozambique, Ae. albopictus was first reported in 2015 within the capital city of Maputo, and by 2019, it had become established in the surrounding area. It was suspected that the mosquito population originated in Madagascar or islands of the Western Indian Ocean (IWIO). The aim of this study was to determine its origin. Given the risk of spreading insecticide resistance, we also examined relevant mutations in the voltage-sensitive sodium channel (VSSC). Methods: Eggs of Ae. albopictus were collected in Matola-Rio, a municipality adjacent to Maputo, and reared to adults in the laboratory. Cytochrome c oxidase subunit I (COI) sequences and microsatellite loci were analyzed to estimate origins. The presence of knockdown resistance (kdr) mutations within domain II and III of the VSSC were examined using Sanger sequencing. Results: The COI network analysis denied the hypothesis that the Ae. albopictus population originated in Madagascar or IWIO; rather both the COI network and microsatellites analyses showed that the population was genetically similar to those in continental Southeast Asia and Hangzhou, China. Sanger sequencing determined the presence of the F1534C knockdown mutation, which is widely distributed among Asian populations, with a high allele frequency (46%). Conclusions: These results do not support the hypothesis that the Mozambique Ae. albopictus population originated in Madagascar or IWIO. Examining patterns of genetic diversity are crucial for conservation planning on endangered species, while inferring the underlying process of recent anthropogenic habitat modifications in the context potential long‐term demographic changes remains challenging. The globally endangered scaly‐sided merganser (SSME), Mergus squamatus, is endemic to a narrow range in Northeast Asia, and its population has recently been contracted into two main breeding areas. Although low genetic diversity has been suggested in the Russian population, the genetic status and demographic history of these individuals have not been fully elucidated. We therefore examined the genetic diversity and structure of the breeding populations of the SSME and investigated the relative importance of historical and recent demographic changes to the present‐day pattern of genetic diversity. Using 10 nuclear microsatellite (SSR) markers and mitochondrial DNA (mtDNA) control region sequences, we found limited female‐inherited genetic diversity and a high level of nuclear genetic diversity. In addition, analysis of both markers consistently revealed significant but weak divergence between the breeding populations. Inconsistent demographic history parameters calculated from mtDNA and bottleneck analysis results based on SSR suggested a stable historical effective population size. By applying approximate Bayesian computation, it was estimated that populations started to genetically diverge from each other due to recent fragmentation events caused by anthropogenic effects rather than isolation during Last Glacial Maximum (LGM) and post‐LGM recolonization. These results suggest that limited historical population size and shallow evolutionary history may be potential factors contributing to the contemporary genetic diversity pattern of breeding SSME populations. Conservation efforts should focus on protecting the current breeding habitats from further destruction, with priority given to both the Russian and Chinese population, as well as restoring the connected suitable breeding grounds. Montane biodiversity is particularly vulnerable to rapid oscillations in environmental conditions. Recent modelling showed that only three of the 19 butterfly species of Erebia (Nymphalidae, Satyrinae) currently present in Iberia would persist in a worst‐case scenario for 2070. One of them is Erebia palarica, endemic to Northwest Spain. We combined genetics, morphometrics and ecological niche modelling to reconstruct its evolutionary history.First, sequences of the cytochrome oxidase I (COI) mitochondrial gene obtained from 305 specimens revealed a double star‐like network that suggests a demographic expansion from two ancestral populations. This finding is congruent with the larger and disjunct putative distribution revealed by climatic modelling during the Last Glacial Maximum.Second, genotyping of 262 samples with seven microsatellite markers produced a large cluster at the centre‐East of the Cantabrian range, mostly affected by isolation by distance, and revealed different levels of structure in the western localities.Lastly, we reviewed the intraspecific taxonomy of the species. The westernmost site (Queixa) stands as a clearly separated unit according to genetic and morphometric analyses, which further supports its vague former description as subspecies E. p. castroviejoi.Altogether, the case of E. palarica exemplifies the complexity that narrow endemics can show and highlights that understanding the evolutionary history of species is crucial for designing adequate conservation strategies. This study characterizes the chromosomal organization of DNA repetitive sequences and the karyotypic evolution in four representatives of the solitary wasp genus Trypoxylon using conventional and molecular cytogenetic techniques. Our findings present the first cytogenetic data for Trypoxylon rogenhoferi (2n = 30) and Trypoxylon albonigrum (2n = 32), while the karyotypes of Trypoxylon nitidum (2n = 30) and Trypoxylon lactitarse (2n = 30) were similar to those previously described. Fluorochrome staining and microsatellite distribution data revealed differences in the constitutive heterochromatin composition among species. Trypoxylon nitidum and T. albonigrum exhibited one major rDNA cluster, potentially representing an ancestral pattern for aculeate Hymenoptera, while T. rogenhoferi and T. lactitarse showed two pericentromeric rRNA gene sites, suggesting amplification events in their ancestral clade. The (TCAGG)n motif hybridized in the terminal regions of the chromosomes in all four Trypoxylon species, which may suggest that this sequence represents DNA telomeric repeat. Notably, the presence of this repetitive sequence in the centromeric regions of certain chromosome pairs in two species supports the hypothesis of chromosomal fusions or inversions in the ancestral karyotype of Trypoxylon. The hoary fox (Lycalopex vetulus) is the only species of the Canidae (Mammalia: Carnivora) endemic to Brazil, and so far has been the target of few genetic studies. Using microsatellites and mtDNA markers, we investigated its present genetic diversity and population structure. We also tested the hypothesis that this species currently hybridizes with the pampas fox (L. gymnocercus), as suggested by previous mtDNA data from two individuals. We collected tissue and blood samples from animals representing most of the two species' distributions in Brazil (n  = 87), including their recently discovered geographic contact zone in São Paulo state. We observed that the hoary fox exhibits high levels of genetic diversity and low levels of population structure. We identified six individuals from São Paulo state with clear evidence of hybridization based on introgressed pampas fox mitochondrial DNA and/or admixed microsatellite genotypes (three individuals bore both types of evidence). These results demonstrate the existence of admixed individuals between hoary and pampas foxes in southeastern Brazil, representing the first identified case of interspecies admixture between native South American canids. We discuss our findings in the context of the evolutionary history of these foxes and address potential conservation implications of this interspecies hybridization process. [ABSTRACT FROM AUTHOR]

Establishing genetic diversity in the population is an essential first step for the sustainable conservation of valuable genetic resources under threat of extinction. Two local goat breeds, Kalofer long-haired (KLH) and Bulgarian screw-horned long-haired (BSHL), were genotyped using a panel of 14 microsatellite markers to determine their population structure and differentiation. The overall mean number of alleles was 8.82, higher in BSHL (9.71) as compared with KLH (7.92). The Ho and He values were almost equal for BSHL (0.75; 0.76, respectively) and KLH goats (0.74; 0.76, respectively). The value of the coefficient of population differentiation (FST) was very low (0.024), indicating only 2% genetic differentiation between the two goat breeds. The structure and PCoA analyses revealed considerable overlap between the two breeds, although some flocks of each breed differentiated from the common genetic pool. The results obtained in the present study can serve to differentiate purebred animals that are the basis of a selection process and a breeding strategy with a view to the conservation of these two local goat breeds. Based on 20 autosomal microsatellite markers, the results of analysis of the genetic structure and spatial autocorrelation in the populations of grey wolf (Canis lupus Linnaeus, 1758) on the territory of the Russian Federation and the Republic of Kazakhstan are described. Using molecular markers, hidden genealogical patterns are revealed, which have the most pronounced effect on distances up to 150 km but may be traced over much wider spaces, up to 700–1600 km. Our research has shown that identification and exclusion of closely related genotypes has limited effect on the results of the analysis of intrapopulation genetic diversity. Meanwhile, this procedure is recommended for a more adequate assessment of the population structure since it allows for streamlining some statistical approaches. Thus, the obtained data demonstrate the integrating effect of natal migration, which works against the differential effect of philopatry. Goat willow (Salix caprea L.) is an ecologically important, cold-tolerant pioneer species that spreads from central Anatolia to Black Sea regions in Türkiye. Completed and ongoing construction of hydroelectric plants, especially in northeastern Türkiye, threaten the genetic resources of goat willow in the region due to habitat loss and fragmentation. Therefore, it is important to assess genetic diversity structure and magnitude of goat willow populations in the northeastern Black Sea Region for effective conservation and utilization of genetic resources of the species. To characterize the genetic diversity of goat willow, 180 trees were sampled from 5 river systems and screened by using ten microsatellite markers for genotyping, consequently analysis were carried out to estimate population genetic diversity parameters of populations. The results revealed that both observed (Ho = 0.50) and expected (He = 0.53) genetic diversities were low, but observed genetic diversity was less than expected due fragmentation and reduction in population sizes. Genetic structure analysis of populations suggested the presence of three major groups. This structuring appeared to be caused by the restricted gene flow (pollen, seed or vegetative material) through insects, wind and water due to geographical barrier (the east Black Sea mountain ranges) acting as geographic isolation mechanism between the populations of the Çoruh river and the river systems located in the west of Çoruh river. To prevent further deterioration of genetic diversity due to habitat loss and fragmentation in the studied river systems in the region, we suggest that one population with high genetic diversity, unique alleles and low inbreeding from each genetic structure cluster should be designated as in situ conservation sites. The genetic resources from such in situ designated populations will facilitate future efficient restoration, genetic resource management and utilization works with the species. Linum suffruticosum s.l. is a taxonomic complex widespread in the Western Mediterranean basin. The complex is characterized by a high phenotypic and cytogenetic diversity, and by a unique three-dimensional heterostyly system that makes it an obligate outcrosser. We studied the patterns of genetic diversity and structure of populations throughout the entire distribution of L. suffruticosum s.l. with microsatellite markers. We analysed their relationships with various biological and ecological variables, including the morph ratio and sex organ reciprocity of populations measured with a novel multi-dimensional method. Populations consistently showed an approximate 1:1 morph ratio with high sex organ reciprocity and high genetic diversity. We found high genetic differentiation of populations, showing a pattern of isolation by distance. The Rif mountains in NW Africa were the most important genetic barrier. The taxonomic treatment within the group was not related to the genetic differentiation of populations, but to their environmental differentiation. Genetic diversity was unrelated to latitude, elevation, population size, niche suitability or breeding system. However, there was a clear influence of ploidy level on the genetic diversity of populations, and a seeming centre–periphery pattern in its distribution. Our results suggest that polyploidization events, high outcrossing rates, isolation by distance and important geographical barriers to gene flow have played major roles in the microevolutionary history of this species complex. DOCKS is a smart docking system developed within the framework of the Space Rider Observer Cube mission (SROC) but suitable for use in more general scenarios. This paper presents the mechanical design of the system and the kinematic tests performed on it. The system merges a classical probe drogue configuration with a gripper-like design, to manage the connection between the parts, and it is equipped with a suite of sensors to estimate the relative pose of the target. The system is also equipped with a dedicated computer, making it a smart standalone system. A series of kinematic tests have been conducted to validate the capability of the system to passively manage misalignment during the docking manoeuvre. Short tandem repeats (STRs) are genomic regions consisting of repeated sequences of 1-6 bp in succession. Single-nucleotide polymorphism (SNP)-based genome-wide association studies (GWASs) do not fully capture STR effects. To study these effects, we imputed 445,720 STRs into genotype arrays from 408,153 White British UK Biobank participants and tested for association with 44 blood phenotypes. Using two fine-mapping methods, we identify 119 candidate causal STR-trait associations and estimate that STRs account for 5.2%-7.6% of causal variants identifiable from GWASs for these traits. These are among the strongest associations for multiple phenotypes, including a coding CTG repeat associated with apolipoprotein B levels, a promoter CGG repeat with platelet traits, and an intronic poly(A) repeat with mean platelet volume. Our study suggests that STRs make widespread contributions to complex traits, provides stringently selected candidate causal STRs, and demonstrates the need to consider a more complete view of genetic variation in GWASs.

Abstract Botryosphaeriaceae species are the major causal agents of walnut dieback worldwide, along with Diaporthe species. Botryosphaeria dothidea and Neofusicoccum parvum are the only two Botryosphaeriaceae species associated with this recently emergent disease in France, and little is known about their diversity, structure, origin and dispersion in French walnut orchards. A total of 381 isolates of both species were genetically typed using a sequence-based microsatellite genotyping (SSR-seq) method. This analysis revealed a low genetic diversity and a high clonality of these populations, in agreement with their clonal mode of reproduction. The genetic similarity among populations, regardless of the tissue type and the presence of symptoms, supports the hypothesis that these pathogens can move between fruits and twigs and display latent pathogen lifestyles. Contrasting genetic patterns between N. parvum populations from Californian and Spanish walnut orchards and the French ones suggested no conclusive evidence for pathogen transmission from infected materials. The high genetic similarity with French vineyards populations suggested instead putative transmission between these hosts, which was also observed with B. dothidea populations. Overall, this study provides critical insight into the epidemiology of two important pathogens involved in the emerging dieback of French walnut orchards, including their distribution, potential to mate, putative origin and disease pathways.

Abstract Environmental gradients in the sea may coincide with phenotypic or genetic gradients resulting from an evolutionary balance between selection and dispersal. The population differentiation of the swimming crab, Liocarcinus depurator, an important by-catch species in the Mediterranean Sea and North-East Atlantic, was assessed using both genetic and morphometric approaches. A total of 472 specimens were collected along its distribution area, and 17 morphometric landmarks, one mitochondrial gene (COI) and 11 polymorphic microsatellite markers were scored in 350, 287 and 280 individuals, respectively. Morphometric data lacked significant differences, but genetic analyses showed significant genetic differentiation between Atlantic and Mediterranean populations, with a steeper gradient in COI compared to microsatellite markers. Interestingly, nuclear differentiation was due to an outlier locus with a gradient in the Atlantic-Mediterranean transition area overlapping with the mtDNA gradient. Such overlapping clines are likely to be maintained by natural selection. Our results suggest a scenario of past isolation with local adaptation and secondary contact between the two basins. Local adaptation during the process of vicariance may reinforce genetic differentiation at loci maintained by environmental selection even after secondary contact.

Abstract Key message Populations of the European white elm (Ulmus laevis Pall.) in Switzerland can be considered natural. They show no evidence of genetic differentiation from other European populations. In the past, the U. laevis populations were probably more widespread and continuous in Switzerland with a larger gene flow. Context In Switzerland, at the margin of its distribution range, U. laevis is rare and considered endangered. Whether the species is native to Switzerland has been disputed, and it is often surmised to be solely cultivated, without any natural population in the country. Aims The structure of genetic diversity among Swiss populations of U. laevis and comparison to European populations are expected to shed light on the origin of local populations and support their management. Methods We analyzed 19 populations (194 individuals) in Switzerland and 15 populations (158 individuals) from other European countries, using a set of five microsatellite loci. Results (1) 90% of the genetic variation in European and Swiss populations occurs within populations. (2) We did not detect isolation by distance at the regional or continental scale. (3) Clustering analysis did not reveal any spatial pattern in the level of admixture of individuals within Swiss or other European populations. Conclusion Moderate levels of genetic diversity and evidence for recent gene flow between populations indicate that habitat deterioration, loss, and fragmentation are the main threats to the persistence of U. laevis populations in Switzerland.

The Canary Islands have historically been a crossway among at least three continents; therefore, the genetic influences on their local animal breeds have been extremely diverse. In the Canarian chicken population, genetic diversity is evident but it has never been studied. The aim of this study was to assess the population structure and genetic diversity within the Canarian chicken population, as well as between the Canarian chicken population, the Spanish local populations, and the commercial chicken populations with microsatellites, with a view to reinforce the official recognition of the breed, for the design and development of a conservation program. Blood samples were collected at random from 198 animals of the Canarian and compared with Spanish local and commercial strains, in order to determine differentiation and genetic relatedness. The five phenotypic varieties of Canarian chicken, the population structure and genetic diversity within the Canarian chicken population had a higher unbiased expected heterozygosity than the observed heterozygosity. Comparing the Canarian vs other local Spanish breeds and commercial strains, FST was relatively high (0.179) (0.164 – 0.195), the neighborhood network showed that the Canarian varieties did not cluster with the other Spanish breeds. The STRUCTURE, confirmed that the Rubilana variety differed from the other four Canary Islands. We conclude that the Canary Islands chicken population shows a differentiated genetic profile, versus other Spanish and cosmopolitan breeds. The theory that the existence of genetic varieties is based on the color of feathers was definitively discarded, except in the Rubilana population, which could be admitted as a genetically different variety.

Satellites in very low Earth orbits can leverage the aerodynamics forces for control purposes. Three-axis attitude control can be achieved by adjusting the orientation of aerodynamic surfaces, benefiting from both drag and lift forces. However, complete control over the orbital motion of a satellite is unattainable using aerodynamic forces alone, as positive thrust cannot be generated. Nonetheless, this limitation does not apply when focusing on the control of a satellite formation. By properly modulating the orientation of panels, positive and negative relative forces can be generated. This paper shows that even in a formation of simple microsatellites, it is possible to achieve formation and attitude simultaneous control by using no other actuators than the aerodynamics surfaces. • Very Low Earth orbits are appealing but the large atmospheric density can be a problem. • The atmospheric forces can be used for control purposes. • Formation control and attitude control can be individually realized by using aerodynamic forces. • A GNC architecture is implemented to realized both goals with the same aerodynamic actuators. • Performance in keeping or acquiring a desired state (attitude and relative position) is promising. Abstract The white-bellied pangolin is subject to intense trafficking, feeding both local and international trade networks. In order to assess its population genetics and trace its domestic trade, we genotyped 562 pangolins from local to large bushmeat markets in western central Africa. We show that the two lineages described from the study region (WCA and Gab) were overlapping in ranges, with limited introgression in southern Cameroon. There was a lack of genetic differentiation across WCA and a significant signature of isolation-by-distance possibly due to unsuspected dispersal capacities involving a Wahlund effect. We detected a c. 74.1–82.5% decline in the effective population size of WCA during the Middle Holocene. Private allele frequency tracing approach indicated up to 600 km sourcing distance by large urban markets from Cameroon, including Equatorial Guinea. The 20 species-specific microsatellite loci provided individual-level genotyping resolution and should be considered as valuable resources for future forensic applications. Because admixture was detected between lineages, we recommend a multi-locus approach for tracing the pangolin trade. The Yaoundé market was the main hub of the trade in the region, and thus should receive specific monitoring to mitigate pangolins’ domestic trafficking. Our study also highlighted the weak implementation of CITES regulations at European borders.

Abstract Polyandry, the practice of females mating with multiple males, is a strategy found in many insect groups. Whether it increases the likelihood of receiving beneficial genes from male partners and other potential benefits for females is controversial. Strepsiptera are generally considered monandrous, but in a few species females have been observed copulating serially with multiple males. Here we show that the offspring of a single female can have multiple fathers in two Strepsiptera species: Stylops ovinae (Stylopidae) and Xenos vesparum (Xenidae). We studied female polyandry in natural populations of these two species by analysis of polymorphic microsatellite loci. Our results showed that several fathers can be involved in both species, in some cases up to four. Mating experiments with S. ovinae have shown that the first male to mates with a given female contributes to a higher percentage of the offspring than subsequent males. In X. vesparum, however, we found no significant correlation between mating duration and offspring contribution. The prolonged copulation observed in S. ovinae may have the advantage of reducing competition with sperm from other males. Our results show that monandry may not be the general pattern of reproduction in the insect order Strepsiptera.

The European pond turtle (Emys orbicularis) is an endangered Old World species. The phylogeographic history of E. orbicularis has been extensively studied throughout its range. While local genetic variation holds valuable information for conservation efforts, specific regional details have yet to be thoroughly examined everywhere. The Valencian region on the Iberian Peninsula is one such area. Here, different evolutionary lineages of E. orbicularis meet, forming a part of the western Mediterranean hybrid zone. In this study, we comprehensively sampled all Valencian localities where E. orbicularis occurs. Using mitochondrial DNA and nuclear microsatellites, we determined the genetic structure of E. orbicularis populations. Our data reveals that the mitochondrial haplotype originating from the glacial refugium in the Apennine Peninsula is primarily restricted to the northern part of the Valencian region. Additionally, human-mediated dispersal processes may have contributed to the complex relationships between the evolutionary lineages of E. orbicularis in the natural hybrid zone. On a finer scale, E. orbicularis in the Valencian region can be classified into five distinct genetic populations. Morphometric analyses revealed sexual dimorphism in the plastron shape and slight variation among genetic populations. Notably, female, but not male, plastron shape correlates with individual heterozygosity. Given the ongoing threat posed by exotic turtles, we propose implementing targeted management strategies to mitigate the presence of alien turtles. These strategies should be directed towards localities that represent each genetic population within the region, reflecting the extensive population structure observed in our study. Biodiversity patterns are shaped by the interplay between geodiversity and organismal characteristics. Superimposing genetic structure onto landscape heterogeneity (i.e., landscape genetics) can help to disentangle their interactions and better understand population dynamics. Previous studies on the sub‐Antarctic Prince Edward Islands (located midway between Antarctica and Africa) have highlighted the importance of landscape and climatic barriers in shaping spatial genetic patterns and have drawn attention to the value of these islands as natural laboratories for studying fundamental concepts in biology. Here, we assessed the fine‐scale spatial genetic structure of the springtail, Cryptopygus antarcticus travei, which is endemic to Marion Island, in tandem with high‐resolution geological data. Using a species‐specific suite of microsatellite markers, a fine‐scale sampling design incorporating landscape complexity and generalised linear models (GLMs), we examined genetic patterns overlaid onto high‐resolution digital surface models and surface geology data across two 1‐km sampling transects. The GLMs revealed that genetic patterns across the landscape closely track landscape resistance data in concert with landscape discontinuities and barriers to gene flow identified at a scale of a few metres. These results show that the island's geodiversity plays an important role in shaping biodiversity patterns and intraspecific genetic diversity. This study illustrates that fine‐scale genetic patterns in soil arthropods are markedly more structured than anticipated, given that previous studies have reported high levels of genetic diversity and evidence of genetic structing linked to landscape changes for springtail species and considering the homogeneity of the vegetation complexes characteristic of the island at the scale of tens to hundreds of metres. By incorporating fine‐scale and high‐resolution landscape features into our study, we were able to explain much of the observed spatial genetic patterns. Our study highlights geodiversity as a driver of spatial complexity. More widely, it holds important implications for the conservation and management of the sub‐Antarctic islands.
