Microplastics (MPs) are emerging pollutants, causing potential threats to aquatic ecosystems and serious concern in aggregating with microalgae (critical primary producers). When entering water bodies, MPs are expected to sink below the water surface and disperse into varying water compartments with different light intensities. However, how light influences the aggregation processes of algal cells onto MPs and the associated molecular coupling mechanisms and derivative risks remain poorly understood. Herein, we investigated the aggregation behavior between polystyrene microplastics (mPS, 10 µm) and Chlorella pyrenoidosa under low (LL, 15 μmol·m -2 ·s -1 ), normal (NL, 55 μmol·m -2 ·s -1 ), and high light (HL, 150 μmol·m -2 ·s -1 ) conditions from integrated in vivo and in silico assays. The results indicated that under LL, the mPS particles primarily existed independently, whereas under NL and HL, C. pyrenoidosa tightly bounded to mPS by secreting more protein-rich extracellular polymeric substances. Infrared spectroscopy analysis and density functional theory calculation revealed that the aggregation formation was driven by non-covalent interaction involving van der Waals force and hydrogen bond. These processes subsequently enhanced the deposition and adherence capacity of mPS and relieved its phytotoxicity. Microalgae are considered to be more useful and effective to use in biomass production than other photosynthesis organisms. However, microalgae need to be altered to acquire more desirable traits for the relevant purpose. Although neutron radiation is known to induce DNA mutations, there have been few studies on its application to microalgae, and the optimal relationship between irradiation intensity and mutation occurrence has not been established. In this study, using the unicellular red alga Cyanidioschyzon merolae as a model, we analyzed the relationship between the absorbed dose of two types of neutrons, high-energy (above 1 MeV) and thermal (around 25 meV) neutrons, and mutation occurrence while monitoring mutations in URA5.3 gene encoding UMP synthase. As a result, the highest mutational occurrence was observed when the cells were irradiated with 20 Gy of high-energy neutrons and 13 Gy of thermal neutrons. Using these optimal neutron irradiation conditions, we next attempted to improve the lipid accumulation of Euglena gracilis, which is a candidate strain for biofuel feedstock production. As a result, we obtained several strains with a maximum 1.3-fold increase in lipid accumulation compared with the wild-type. Light management strategy is crucial for improving microalgal production in terms of higher biomass and economically valuable bioactive molecules. However, green light has received less attention in developing light managements for algae and higher plant due to its low absorption rate by chlorophyll. In this study, the effects of green light supplementation, in the combination with red and blue light were investigated in Haematococcus pluvialis. 10% and 20% of green light supplementations were applied in 3:2 ratios of red and blue LED light combinations as an expense of red-light. Growth rates, chlorophyll concentration, and dry weight were measured to assess the growth kinetics of H. pluvialis along with the relative transcript accumulations of four mRNAs: Rubisco, PTOX 2 , PsaB, and PsbS. Growth rates, chlorophyll concentrations and dry weight were found significantly higher in presence of 10% green light supplementation compared to red and blue light combinations. The relative transcript accumulations of Rubisco and PsbS genes showed significant upregulation at the end of the experiments (with the fold change of 42.91 ± 12.08 and 98.57 ± 27.38, respectively, relative to the beginning of the experiments) compared to combinations of red and blue light (fold change of 19.09 ± 3.0 and 47.77 ± 14.21, respectively, relative to beginning of the experiments). PsaB and PTOX 2 transcripts did not show significant accumulation differences between treatments. It seems that green light has a dose dependent additive effect on the growth rate of H. pluvialis. Rapid light curves are one of the most widely used methods for assessing the physiological state of photosynthetic organisms. While the method has been applied in a range of physiological studies over the last 20 years, little progress has been made in adapting it for the new age of multi-parametric phenotyping. In order to advance research that is aimed at evaluating the physiological impact of multiple factors, the Phenoplate was developed: a simultaneous assessment of temperature and light gradients. It was used to measure rapid light curves of three marine microalgae across a temperature gradient and altered phosphate availability. The results revealed that activation of photoprotective mechanisms occurred with high efficiency at lower temperatures, and relaxation of photoprotection was negatively impacted above a certain temperature threshold in Tetraselmis sp. It was observed that Thalassiosira pseudonana and Nannochloropsis oceanica exhibited two unique delayed non-photochemical quenching signatures: in combinations of low light with low temperature, and darkness with high temperature, respectively. Small-scale photobioreactors (PBRs) in the inoculum stage were designed with internal (red or green) and external white LED light as an initial step of a larger-scale installation aimed at fulfilling the integral biorefinery concept for maximum utilization of microalgal biomass in a multifunctional laboratory. The specific growth rate of Scenedesmus obliquus (Turpin) Kützing biomass for given cultural conditions was analyzed by using MAPLE software. For the determination of total polyphenols, flavonoids, chlorophyll "a" and "b", carotenoids and lipids, UHPLC-HRMS, ISO-20776/1, ISO-10993-5 and CUPRAC tests were carried out. Under red light growing, a higher content of polyphenols was found, while the green light favoured the flavonoid accumulation in the biomass. Chlorophylls, carotenoids and lipids were in the same order of magnitude in both samples. The dichloromethane extracts obtained from the biomass of each PBR synergistically potentiated at low concentrations (0.01-0.05 mg/mL) the antibacterial activity of penicillin, fluoroquinolones or oregano essential oil against the selected food-borne pathogens ( Staphylococcus aureus , Escherichia coli and Salmonella typhimurium ) without showing any in vitro cytotoxicity. Both extracts exhibited good cupric ion-reducing antioxidant capacity at concentrations above 0.042-0.08 mg/mL. The UHPLC-HRMS analysis revealed that both extracts contained long chain fatty acids and carotenoids thus explaining their antibacterial and antioxidant potential. The applied engineering approach showed a great potential to modify microalgae metabolism for the synthesis of target compounds by S. obliquus with capacity for the development of health-promoting nutraceuticals for poultry farming.

The demand for natural antioxidants to be used in food industry is increasing, as synthetic antioxidants are toxic and have high production costs. Specifically, food processing and preservation require antioxidants resistant to thermal sterilization processes. In this study, twenty-five strains among microalgae and cyanobacteria were screened as antioxidants producers. The species Enallax sp., Synechococcus bigranulatus and Galdieria sulphuraria showed the highest content of chlorophyll a and total carotenoids. In vitro stability and antioxidant activity of the ethanolic extracts were performed. The results revealed that pigments present in the extracts, obtained from the previously mentioned species, were stable at room temperature and exhibited in vitro free radical scavenging potential with IC 50 values of 0.099 ± 0.001, 0.048 ± 0.001 and 0.13 ± 0.02 mg mL -1 , respectively. Biocompatibility assay showed that the extracts were not toxic on immortalized cell lines. The antioxidant activity was also tested on a cell-based model by measuring intracellular ROS levels after sodium arsenite treatment. Noteworthy, extracts were able to exert the same protective effect, before and after the pasteurization process. Results clearly indicate the feasibility of obtaining biologically active and thermostable antioxidants from microalgae. Green solvents can be used to obtain thermo-resistant antioxidants from cyanobacteria and microalgae which can be used in the food industry. Eukaryotic phytoplankton have a small global biomass but play major roles in primary production and climate. Despite improved understanding of phytoplankton diversity and evolution, we largely ignore the cellular bases of their environmental plasticity. By comparative 3D morphometric analysis across seven distant phytoplankton taxa, we observe constant volume occupancy by the main organelles and preserved volumetric ratios between plastids and mitochondria. We hypothesise that phytoplankton subcellular topology is modulated by energy-management constraints. Consistent with this, shifting the diatom Phaeodactylum from low to high light enhances photosynthesis and respiration, increases cell-volume occupancy by mitochondria and the plastid CO 2 -fixing pyrenoid, and boosts plastid-mitochondria contacts. Changes in organelle architectures and interactions also accompany Nannochloropsis acclimation to different trophic lifestyles, along with respiratory and photosynthetic responses. By revealing evolutionarily-conserved topologies of energy-managing organelles, and their role in phytoplankton acclimation, this work deciphers phytoplankton responses at subcellular scales.

Cyanobacteria and microalgae present in the aquatic or terrestrial environment may be emitted into the air and transported along with air masses over long distances. As a result of staying in the atmosphere, these organisms may develop a greater tolerance to stressful factors, but this topic is still relatively unknown. The main aim was to show an autecological characteristic of some airborne microalgae and cyanobacteria strains by a factorial laboratory experiment approach, including changes in irradiance, temperature, and salinity conditions. The additional purpose of this work was also to present part of the Culture Collection of Baltic Algae (CCBA) collection, which consists of airborne algae (AA) isolated from the atmospheric air of the southern Baltic Sea region. Altogether, 61 strains of airborne cyanobacteria and microalgae from the southern Baltic Sea region were isolated from May 2018 to August 2020. Selected microorganisms were tested in controlled laboratory conditions to identify their response to different irradiance (10-190 µmol photons m -2 s -1 ), temperature (13-23 °C), and salinity conditions (0-36 PSU). The highest numbers of cells (above 30 × 10 5 cell mL -1 ) were recorded for cyanobacterium Nostoc sp., and for diatoms Nitzschia sp., Amphora sp., and Halamphora sp. We found that for cyanobacterium Nostoc sp. as well as for green alga Coccomyxa sp. the maximum cell concentrations were recorded at the salinity of 0 PSU. Moreover, cyanobacteria Planktolyngbya contorta , Pseudanabaena catenata , Leptolyngbya foveolarum , Gloeocapsa sp., and Rivularia sp. were able to grow only at a salinity of 0 PSU. On the other hand, in the range of 16-24 PSU, the highest cell numbers of examined diatoms have been identified. Our research provided that deposited airborne microalgae and cyanobacteria showed full colonization potential. The present experiment suggests that the adaptive abilities of microorganisms, in particular those producing toxins, may contribute to the spread in the future. Thus, it may increase human exposure to their negative health effects. Any distinctive adaptations of the genera give them an additional competitive advantage and a greater chance for territorial expansion.

Algal biofuels are a promising alternative to fossil fuels, but their widespread use is hindered by problems with mass production. Light-emitting diodes (LEDs) with specific light wavelengths could be used as an energy source for algal growth and lipid synthesis. In this study, the effects of light source on the biomass and lipid production of the aerial microalga Coccomyxa sp. KGU-D001 were evaluated using LEDs. The integration of two-phase cultures, including growth and lipid production under the stress of nitrate depletion, was assessed for efficient lipid production under liquid- or aerial-phase conditions. Different wavelengths of light (blue, green, and red) were tested under liquid- and aerial-phase conditions. Under aerial-phase culture, the fatty acid contents in biofilm reached 320 mg g DWC -1 with the red LEDs. In view of these findings, we describe a one-step culture method for growth and lipid accumulation in algal biofilm under aerial-phase culture with red LED irradiation. When Coccomyxa biofilm was cultured on wet cotton wool with BBM in a petri dish under the red LED, it was able to grow and accumulate lipids under the aerial-phase condition. The presence, biosynthesis and functional role of sterols in the green microalga Haematococcus pluvialis remain poorly understood. In this work we studied the effect of high-light (HL) stress on sterol synthesis in H. pluvialis UTEX 2505 cells. HL stress induced the synthesis of sterols in parallel with that of triacylglycerides (TAG), giving rise to the synthesis of cholesterol over that of phytosterols. Blockage of the carotenogenic 1-deoxy-D-xylulose 5-phosphate (MEP) pathway is shown to be involved in HL-induced sterol synthesis. In addition, high irradiance exposure induced MEP- and fatty acid (FA)-biosynthetic transcripts. The pharmacological inhibition of these pathways suggests a possible feedback regulation of sterol and FA homeostasis. Finally, both lipid classes proved crucial to the adequate photosynthetic performance of H. pluvialis grown under HL intensity stress. Economically feasible photosynthetic cultivation of microalgal and cyanobacterial strains is crucial for the biological conversion of CO 2 and potential CO 2 mitigation to challenge global warming. To overcome the economic barriers, the production of value-added chemicals was desired by compensating for the overall processing cost. Here, we engineered cyanobacteria for photosynthetic squalene production and cultivated them in a scalable photobioreactor using industrial flue gas. First, an inducer-free gene expression system was developed for the cyanobacteria to lower production const. Then, the recombinant cyanobacteria were cultivated in a closed photobioreactor (100 L) using flue gas (5% CO 2 ) as the sole carbon source under natural sunlight as the only energy source. Seasonal light intensities and temperatures were analyzed along with cyanobacterial cell growth and squalene production in August and October 2019. As a result, the effective irradiation hours were the most critical factor for the large-scale cultivation of cyanobacteria. Thus, an automated photobioprocess system will be developed based on the regional light sources.

A novel method for quantitative detection of the viability of a single microalgae cell by two-level photoexcitation is proposed in this paper. This method overcomes the difficulty of traditional methods in determining the cell viability by a fixed standard under a single photoexcitation. It is experimentally confirmed that this method is not limited by the species, morphology, size and structure of microalgae cells. An evaluation criterion of universal applicability is presented for the assessment of cell viability based on the large amount of experimental data. To the best of our knowledge, this is the first time that the relative fluorescence yield ratio F r has been used to characterize the viability of single microalgae cells during cell migration. By using the relative fluorescence yield ratio, this method does not require the intensity of the excitation light to be very low for the assessment of the fluorescence yield of a dark-adapted microalgae cell, nor to be very strong to reach the saturated light level to assess the maximum fluorescence yield. Therefore, this method greatly reduces the technical difficulties of developing a sensor device. Well balanced portability, accuracy and universal applicability make it suitable for on-site real-time detection.

In this article, we investigate the question of the impact of the diel vertical migration (DVM) and the light attenuation by the cells on both spatial pattern and population dynamics of phytoflagellates. For these purposes, we performed a simulation study by using a spatially explicit individual-based model (IBM). The designed IBM includes 2 global mechanisms: cellular motion which is modelled through a stochastic differential equation and demographic process that is density and light intensity dependent. We showed that under no-DVM, for competitive environment, increasing the light absorption helps the creation of a strong oscillatory behaviour, that breeds aggregation-break up rhythm, which is beneficial. However, when we pass to DVM regime, it will be more advantageous for the cells to reduce their light absorption. In regards of DVM effect, we showed that it has other benefits, apart from the acquisition of resources, such as enhancing the attraction mechanism that promotes cooperation and also helping the creation of spatial voids that permit the penetration of the light. Also, we showed that the DVM reduces the predation rate, so we prove quantitatively that the DVM reduces, in general, the grazing losses. Also, we found that the DVM strategy depends on the impact of the competition and cooperation between the cells on the division and death rates. So, probably for the motile species, the local competition is not high in order to let the cells gain advantages from the DVM regarding their survivability that is explained by an annual cyclic behaviour under seasonal environment., (Copyright © 2020. Accumulation of high-value products in microalgae is not conducive with rapid cell growth, which is the potential conflict in microalgal production. Overcoming such conflict faces numerous challenges in comprehensively understanding cell behavior and metabolism. Here, we show a fully integrated interaction between cell behavior, carbon partitioning, carbon availability and path rate of central carbon metabolism, and have practically overcome the production conflict of Chromochloris zofingiensis. We demonstrate that elevated carbon availability and active path rate of precursors are determinants for product biosynthesis, and the former exhibits a superior potential. As protein content reaches a threshold value to confer survival advantages, carbon availability becomes the major limiting factor for product biosynthesis and cell reproduction. Based on integrated interaction, regulating the C/N balance by feeding carbon source under excess light increases content of high-value products without inhibiting cell growth. Our findings provide a new orientation to achieve great productivity improvements in microalgal production.

Corals have evolved as optimized photon augmentation systems, leading to space-efficient microalgal growth and outstanding photosynthetic quantum efficiencies. Light attenuation due to algal self-shading is a key limiting factor for the upscaling of microalgal cultivation. Coral-inspired light management systems could overcome this limitation and facilitate scalable bioenergy and bioproduct generation. Here, we develop 3D printed bionic corals capable of growing microalgae with high spatial cell densities of up to 10 9  cells mL -1 . The hybrid photosynthetic biomaterials are produced with a 3D bioprinting platform which mimics morphological features of living coral tissue and the underlying skeleton with micron resolution, including their optical and mechanical properties. The programmable synthetic microenvironment thus allows for replicating both structural and functional traits of the coral-algal symbiosis. Our work defines a class of bionic materials that is capable of interacting with living organisms and can be exploited for applied coral reef research and photobioreactor design.

The Chlorella vulgaris has been generally recognized as a promising microalgal model to study stress-related responses due to its ability to withstand against ionizing and non-ionizing radiation. The objective of the present study was to investigate the effect of CaCl 2 pre-treatment at different concentrations on the responses of microalga C. vulgaris under gamma radiation toxicity. Changes in growth, physiological parameters and biochemical compositions of the algae pretreated with 0.17 (normal), 5, and 10 mM CaCl 2 were analyzed under 300 Gy gamma irradiation and compared to those of gamma-free control. The results showed that parameters including specific growth rate, cell size, chlorophyll and protein contents, ascorbate peroxidase (APX), and superoxide dismutase (SOD) activity, Ferric Reducing Antioxidant Power (FRAP), and the ratios of nucleic acid to protein negatively affected by gamma irradiation. All these parameters, except for the ratios of nucleic acid to protein significantly increased in the algae when pretreated with a CaCl 2 content higher than normal concentration. The analysis also showed that parameters including catalase activity, proline, and carotenoid content, the level of lipid peroxidation, and electrolyte leakage (EL) significantly increased under gamma irradiation but not affected significantly under different CaCl 2 pre-treatments. Additionally, specific growth rate, chlorophyll a and protein content, APX and SOD activity, FRAP, lipid peroxidation, electrolyte leakage, and the ratios of nucleic acid to protein were the only parameters that significantly affected by the interaction of gamma toxicity and CaCl 2 pretreatment. Overall, the results suggested that regardless of the CaCl 2 effect, the algal cells responded to gamma radiation more efficiently by increasing proline, carotenoids content, and CAT activity. More important, it was concluded that calcium had an essential role in modifying the detrimental effect of gamma toxicity on the algae mainly by increasing the activity of ascorbate peroxidase and superoxide dismutase and maintaining the reducing antioxidant power (FRAP) of the cells at a high level., Competing Interests: Declaration of competing interest The authors declare no competing financial interests., (Copyright © 2020 Elsevier Inc. Microalgae accumulate bioavailable selenium-containing amino acids (Se-AAs), and these are useful as a food supplement. While this accumulation has been studied in phototrophic algal cultures, little data exists for heterotrophic cultures. We have determined the Se-AAs content, selenium/sulfur (Se/S) substitution rates, and overall Se accumulation balance in photo- and heterotrophic Chlorella cultures. Laboratory trials revealed that heterotrophic cultures tolerate Se doses ∼8-fold higher compared to phototrophic cultures, resulting in a ∼2-3-fold higher Se-AAs content. In large-scale experiments, both cultivation regimes provided comparable Se-AAs content. Outdoor phototrophic cultures accumulated up to 400 μg g -1 of total Se-AAs and exhibited a high level of Se/S substitution (5-10%) with 30-60% organic/total Se embedded in the biomass. A slightly higher content of Se-AAs and ratio of Se/S substitution was obtained for a heterotrophic culture in pilot-scale fermentors. The data presented here shows that heterotrophic Chlorella cultures provide an alternative for Se-enriched biomass production and provides information on Se-AAs content and speciation in different cultivation regimes.

The pelagophyte Aureococcus anophagefferens has caused recurrent brown tide blooms along the northeast coast of the United States since the mid-1980's, and more recently spread to other regions of the globe. These blooms, due to the high cell densities, are associated with severe light attenuation that destroys the sea grass beds which provide the basis for many fisheries. Data collected by transmission electron microscopy, PCR, and metatranscriptomic studies of the blooms, support the hypothesis that large dsDNA viruses play a role in bloom dynamics. While a large (~140 nm) icosahedral virus, with a 371 kbp genome, was first isolated more than a decade ago, the constraints imposed by environmental parameters on bloom infection dynamics by Aureococcus anophagefferens Virus, (AaV) remain unknown. To investigate the role light plays in infection by this virus, we acclimated A. anophagefferens to light intensities of 30 (low), 60 (medium) or 90 μmol photons m-2 s-1 (high) and infected cultures at these irradiance levels. Moreover, we completed light shift experiments where acclimated cultures were exposed to even lower light intensities (0, 5, and 15 μmol photons m-2 s-1) consistent with irradiance found during the peak of the bloom when cell concentrations are highest. The abundance of viruses produced per lytic event (burst size) was lower in the low irradiance acclimated cultures compared to the medium and high acclimated cultures. Transferring infected cultures to more-limiting light availabilities further decreased burst size and increased the length of time it took for cultures to lyse, regardless of acclimation irradiance level. A hypothetical mechanism for the reduced efficiency of the infection cycle in low light due to ribosome biogenesis was predicted from pre-existing transcriptomes. Background: The green microalga Dunaliella salina accumulates a high proportion of β-carotene during abiotic stress conditions. To better understand the intracellular flux distribution leading to carotenoid accumulation, this work aimed at reconstructing a carbon core metabolic network for D. salina CCAP 19/18 based on the recently published nuclear genome and its validation with experimental observations and literature data., Results: The reconstruction resulted in a network model with 221 reactions and 212 metabolites within three compartments: cytosol, chloroplast and mitochondrion. The network was implemented in the MATLAB toolbox CellNetAnalyzer and checked for feasibility. Furthermore, a flux balance analysis was carried out for different light and nutrient uptake rates. The comparison of the experimental knowledge with the model prediction revealed that the results of the stoichiometric network analysis are plausible and in good agreement with the observed behavior. Accordingly, our model provides an excellent tool for investigating the carbon core metabolism of D. salina., Conclusions: The reconstructed metabolic network of D. salina presented in this work is able to predict the biological behavior under light and nutrient stress and will lead to an improved process understanding for the optimized production of high-value products in microalgae.

Tailoring spectral quality during microalgal cultivation can provide a means to increase productivity and enhance biomass composition for downstream biorefinery. Five microalgae strains from three distinct lineages were cultivated under varying spectral intensities and qualities to establish their effects on pigments and carbon allocation. Light intensity significantly impacted pigment yields and carbon allocation in all strains, while the effects of spectral quality were mostly species-specific. High light conditions induced chlorophyll photoacclimation and resulted in an increase in xanthophyll cycle pigments in three of the five strains. High-intensity blue LEDs increased zeaxanthin tenfold in Rhodella sp. APOT_15 relative to medium or low light conditions. White light however was optimal for phycobiliprotein content (11.2 mg mL -1 ) for all tested light intensities in this strain. The highest xanthophyll pigment yields for the Chlorophyceae were associated with medium-intensity blue and green lights for Brachiomonas submarina APSW_11 (5.6 mg g -1 lutein and 2.0 mg g -1 zeaxanthin) and Kirchneriella aperta DMGFW_21 (1.5 mg g -1 lutein and 1 mg g -1 zeaxanthin), respectively. The highest fucoxanthin content in both Heterokontophyceae strains (2.0 mg g -1 ) was associated with medium and high white light for Stauroneis sp. LACW_24 and Phaeothamnion sp. LACW_34, respectively. This research provides insights into the application of LEDs to influence microalgal physiology, highlighting the roles of low light on lipid metabolism in Rhodella sp. The versatile use of biopigments in food, feed, cosmetic, pharmaceutical and analytical industries emphasized to find different and renewable sources of biopigments. Microalgae, including cyanobacteria, are becoming a potential candidate for pigment production as these have fast-growing ability, high pigment content, highly variable and also have "Generally recognized as safe" status. These algal groups are known to produce different metabolites that include hormones, vitamins, biopolythene and biochemicals. We discuss here the potential use of microalgal biopigments in our daily life as well as in food and cosmetic industries. Pigment like carotenoids has many health benefits such as antioxidant, anti-inflammatory properties and also provide photo-protection against UV radiation. This review details the effect of various abiotic and biotic factors such as temperature, light, nutrition on maximizing the pigment content in the microalgal cell. This review also highlights the potential of microalgae, whether in present native or engineered strain including the many metabolic strategies which are used or can be used to produce a higher amount of these valuable biopigments. Additionally, future challenges in the context of pigment production have also been discussed.

The present study investigated the effect of light intensity and mixing on microalgae growth in a raceway by comparing the performance of a paddlewheel to a combination of paddlewheel and CO 2 spargers in a 20 L raceway. The increase of light intensity was known to be able to increase the microalgal growth rate. Increasing paddlewheel rotation speed from 13 to 30 rpm enhanced C. vulgaris growth by enhancing culture mixing. Simulation results using computational fluid dynamics (CFD) indicated that both the turnaround areas of the raceway and the area opposite the paddlewheel experienced very low flow velocities (dead zones) of less than 0.1 m/min, which could cause cell settling and slow down growth. The simulated CFD velocity distribution in the raceway was validated by actual velocity measurements. The installation of CO 2 spargers in the dead zones greatly increased flow velocity. The increase of paddlewheel rotation speed reduced the dead zones and hence increased algal biomass production. By complementing the raceway paddlewheel with spargers providing CO 2 at 30 mL/min, we achieved a dry cell weight of 5.2 ± 0.2 g/L, which was about 2.6 times that obtained without CO 2 sparging., (Copyright © 2019 The Society for Biotechnology, Japan. The unicellular green microalga Haematococcus pluvialis accumulates large amounts of the red ketocarotenoid astaxanthin. Aiming to cultivate these microalgae with high astaxanthin efficiency, cultivations were scaled-up from 1000 mL bottle to 2 L and 8 L airlift photobioreactor using volumetric power consumption rate (W/m 3 ) as scale up strategy. After cultivations, computational fluid dynamics (CFD) simulation was used to investigate the flow patterns, mixing efficiency and gas holdup profile within the 2 L photobioreactor. At the end, astaxanthin content was enhanced with increasing the cultivation volume and highest astaxanthin amount of 49.39 ± 1.64 mg/g cell was obtained in 8 L photobioreactor. Hydrodynamic characteristics of photobioreactor was simulated and gas holdup showed difference between the riser and the downcomer regions. Velocity profiles of air and medium had higher values inside the draft tube than obtained in downcomer region. However liquid circulation was achieved from draft tube to the downcomer, mixing was not provided effectively considering the turbulence kinetic energy. In this work, the key moments of the development of the so-called thin-layer cascades (TLC) for microalgae production are described. Development started at the end of the 1950s when the first generation of TLCs was set-up in former Czechoslovakia. Since, similar units for microalgae culturing, which are relatively simple, low-cost and highly productive, have been installed in a number of other countries worldwide. The TLCs are characterized by microalgae growth at a low depth (< 50 mm) and fast flow (0.4-0.5 m/s) of culture compared to mixed ponds or raceways. It guarantees a high ratio of exposed surface to total culture volume (> 100 1/m) and rapid light/dark cycling frequencies of cells which result in high biomass productivity (> 30 g/m 2 /day) and operating at high biomass density, > 10 g/L of dry mass (DW). In TLCs, microalgae culture is grown in the system of inclined platforms that combine the advantages of open systems-direct sun irradiance, easy heat derivation, simple cleaning and maintenance, and efficient degassing-with positive features of closed systems-operation at high biomass densities achieving high volumetric productivity. Among significant advantages of thin layer cascades compared to raceway ponds are the operation at much higher cell densities, very high daylight productivities, and the possibility to store the culture in retention tanks at night, or in unfavourable weather conditions. Concerning the limitations of TLCs, one has to consider contaminations by other microalgae that limit cultivation to robust, fast-growing strains, or those cultured in selective environments.

We have worked out a rapid 1-day test based on photosynthesis measurements to estimate suitable growth temperature of microalgae cultures. To verify the proposed procedure, several microalgae-Chlorella, Nostoc, Synechocystis, Scenedesmus, and Cylindrospermum-were cultured under controlled laboratory conditions (irradiance, temperature, mixing, CO 2 , and nutrient supply) to find the optima of photosynthetic activity using the range between 15 and 35 °C. These activities were recorded at each temperature step after 2 h of acclimation which should be sufficient as oxygen production and the PQ cycle are regulated by fast processes. Photosynthetic activity was measured using three techniques-oxygen production/respiration, saturating pulse analysis of fluorescence quenching, and fast fluorescence induction kinetics-to estimate the temperature optima which should correspond to high growth rate. We measured all variables that might have been directly related to growth-photosynthetic oxygen evolution, maximum photochemical yield of PSII, F v /F m , relative electron transport rate rETR max , and the transients V j and V i determined by fast fluorescence induction curves. When the temperature optima for photosynthetic activity were verified in growth tests, we found good correlation. For most of tested microalgae strains, temperature around 30 °C was found to be the most suitable at this setting. We concluded that the developed test can be used as a rapid 1-day pre-screening to estimate a suitable growth temperature of microalgae strains before they are cultured in a pilot scale.

Sustainability of irrigation systems is crucially important for both economical and efficient usage of natural water sources. Irrigation system efficiency could be negatively affected by some natural flora such as micro (Arthrospira maxima) and macro (Cladophora sp.) algae. In this study, efficiency of UV-C radiation, which has been proven effectiveness in demolishing of bacteria and microbes in medicine, for micro and macro algae. Our study was performed under laboratory conditions and was carried out in two main phases. In the first phase, spectral absorbance curves were established for micro and macro algae, the greatest absorbance value was determined in UV region (300 nm to 450 nm). In the second phase, to analyze the growth of algae species depending on UV-C illumination, micro algae within three different basins with the same cross-sectional area and different water depths (D1:12.5 cm, D2:25 cm, D3:45cm) were exposed to UV-C radiation for different durations (2, 4, 6, 8 and 16 s). The maximum inhibition on the growth rate of A. maxima was observed in 16 s UV-C radiation treatment groups of D1 and D2 basins. Since Cladophora sp. macro algae are aquatic weeds floating over the water surface, they were exposed to UV-C radiation similar to micro algae only in D1 basin. The greatest reduction rate in biomass values of Cladophora sp. was observed in 16 s UV-C exposed groups. The experimental findings showed that UV-C radiation could be used to reduce growth of micro and macro algae. Outdoor high cell-density microalgae cultivation is highly challenging due to unavailability of appropriate CO 2 feeding strategy under diurnal sunlight intensities. Hence, a novel real time light based CO 2 feeding strategy was firstly developed under diurnal simulated sunlight (LED) to test on Chlorella sp. in a 10 L scale bubble column photobioreactor. The strategy yielded a biomass titer of 5.12 g L -1 under simulated sunlight, far higher than existing biomass-density and pH-control based CO 2 feeding strategies. In outdoor culturing, the proposed feeding strategy yielded high biomass titers of 6.8 and 9.0 g L -1 in growth-phase of two-stage and single-stage lipid induction studies respectively with same biomass productivity of 0.8 g L -1  day -1 . Subsequently, two-stage lipid induction strategy of 6.8 g L -1 titer yielded biodiesel productivity of 120 g L -1  day -1 , whereas single-stage strategy of 9.0 g L -1 titer was unable to induce lipid. Interpreting and predicting the combined effects of toxicants in the environment is an important challenge in ecotoxicology. How such effects are connected across different levels of biological organisation is an additional matter of uncertainty. Such knowledge gaps are particularly prominent with regards to how ionising radiation interacts with contaminants. We assessed the response of twelve endpoints at the subcellular, individual and population level in a green microalga when exposed singly and jointly to gamma radiation and cadmium (Cd). We used a fully factorial experimental design where observed effects were compared to those predicted by the Independent Action (IA) model for mixture toxicity to determine whether they deviated from additivity. Subcellular endpoints (e.g., catalase, thiamine diphosphate, xanthophyll cycle pigments) showed an increased antioxidant and/or photoprotective response. However, our results indicate that this protection was not sufficient to prevent lipid peroxidation, which also increased with dose. At ecologically relevant doses, most interactions between gamma radiation and Cd regarding subcellular-, individual- and population-level endpoints were additive as predicted by the IA model. However, exposure to binary mixtures displayed antagonistic interactions between gamma radiation and Cd at the higher end of the tested dose spectrum. No correlations were observed between subcellular endpoints and higher-level endpoints, but there were linkages between individual and population endpoints. Our results suggest that antagonistic interactions between gamma radiation and Cd can occur at higher doses and that these interactions seem to disseminate from subcellular and individual to population level. Current research aimed to increase mixotrophic biomass from various organic carbon sources by exploring best light conditions. Three substrates glucose, acetic acid and glycerol were studied for their effects on mixotrophic microalgae cultivation under four light conditions. Light irradiance exhibited variability in growth response and photosynthetic efficiency based on type of substrates used in mixotrophic growth. Each substrate showed variability in light requirements for their effective assimilations. From growth responses, glucose and acetic acid respectively exhibited heterotrophic and mixotrophic (better growth in light) natures. Continuous light-deficient condition was adequate for effective mixotrophic growth as well as energy saving for glucose. However, light-sufficient condition required for effective acetic acid supported mixotrophic growth. Mixotrophic benefits from glycerol and its uptake by Chlorella protothecoides was negligible in all light conditions. Scaling of phototrophic bioprocesses can be extremely challenging especially when reactor types in the considered scales differ. In this study, the mean integral photon flux density was used to transfer light-dependent growth kinetics of Nannochloropsis salina 40.85 and Nannochloropsis gaditana 2.99 grown with constant LED irradiation from flat-plate gas-lift photobioreactors (0.09 m 2 ) to thin-layer cascade photobioreactors (8 m 2 ). Even though completely different reactors were used, comparable growth rates were achieved on both scales with both strains by application of comparable mean light availabilities in the microalgae suspensions. In contrast, the light-dependent growth kinetics change significantly when irradiation varies dynamically (day-night cycles). The maximum intra-day growth rate of N. salina with dynamic climate simulation was doubled to 0.07 h -1 compared to constant irradiation, but tolerance of the microalgae against excessive irradiation was drastically reduced compared to constant irradiation. Microalgae are an ubiquitous and powerful driver of geochemical cycles which have formed Earth's biosphere since early in the evolution. Lately, microalgal research has been strongly stimulated by economic potential expected in biofuels, wastewater treatment, and high-value products. Similar to bacteria and other microorganisms, most work so far has been performed on the level of suspensions which typically contain millions of algal cells per millilitre. The thus obtained macroscopic parameters average cells, which may be in various phases of their cell cycle or even, in the case of microbial consortia, cells of different species. This averaging may obscure essential features which may be needed for the correct understanding and interpretation of investigated processes. In contrast to these conventional macroscopic cultivation and measuring tools, microfluidic single-cell cultivation systems represent an excellent alternative to study individual cells or a small number of mutually interacting cells in a well-defined environment. A novel microfluidic photobioreactor was developed and successfully tested by the photoautotrophic cultivation of Chlorella sorokiniana. The reported microbioreactor facilitates automated long-term cultivation of algae with controlled temperature and with an illumination adjustable over a wide range of photon flux densities. Chemical composition of the medium in the microbioreactor can be stabilised or modulated rapidly to study the response of individual cells. Furthermore, the algae are cultivated in one focal plane and separate chambers, enabling single-cell level investigation of over 100 microcolonies in parallel. NMR spectroscopy coupled with multivariate data analysis techniques was applied to obtain meaningful information about nontargeted metabolic changes on  Isochrysis galbana upon acclimation to different environmental conditions at indoor lab-scale. The effects of temperature (from 15 to 30 °C) and incident irradiance (from 250 to 1600 μmol m -2 s -1 ) at a constant dilution rate of 0.3 h -1 were evaluated. High irradiances stimulated a decrease of chlorophyll a, fucoxanthin and amino acids content, and the conversion of polar fatty acids (PLs, GLs, DGDGs, SGDGs) to neutral fatty acids (saturated and unsaturated). High temperatures together with high irradiances decreased PUFAs concentration, including omega-3 fatty acids. Under low irradiance and temperature organic osmolytes (homarine, DMSP, GBT, and glycerol), and sugars (glucose, trehalose, and galactose) were also reduced.

The effects of light intensity on the development of algal-bacterial granular sludge (ABGS) were investigated over a period of 12 weeks. The ABGS developed at low light intensity (142 ± 10 μmol m -2 ·s -1 ) exhibited excellent settling ability (SVI 30 of 30.9 mL/g), COD and TN removal efficiencies (97.6% and 60.4%, respectively). High light intensity (316 ± 12 μmol m -2 ·s -1 ) accelerated granular biomass growth (5.3 g/L) and enhanced the TP removal efficiency (83.7%). Extracellular polymeric substance (EPS) analysis revealed that low light intensity induced more large weight distribution protein production (9-12 kDa and 50-150 kDa), predominantly tryptophan and aromatic proteins. Furthermore, N-acyl-homoserine lactones (AHLs) with a side chain ≤ C 10 were commonly shared in the ABGS, and the ABGS developed at low light intensity had a higher C 6 - and 3OC 8 -HSL content, which effectively promoted the biofilm formation. The add-back studies showed that the AHLs facilitated the regulation of EPS synthesis. Statistical analysis indicated that the AHLs content had a close correlation with the EPS production, the 50th percentile of the particle size distribution and microbial community assembly, suggesting that AHLs-mediated quorum sensing have an important ecological role in EPS expression and algal-bacterial granulation. Cell disruption and protein solubilization protocols for the relative quantification of individual subunits in photosystems were developed for photosynthetic organisms including cyanobacterium Synechocystis sp. PCC 6803, green-algae Chlamydomonas reinhardtii, and seed plant Arabidopsis thaliana. The optimal methods for the disruption of Chlamydomonas, Synechocystis, and Arabidopsis cells were sonication, microbeads (Φ approximately 0.1 mm), and large beads (Φ = 5.0 mm), respectively. Extraction of the total proteins exceeded 90% using each optimal cell disruption method. Solubilization efficiency of membrane proteins was improved by the phase transfer surfactant (PTS) method. Ninety seven and 114 proteins from Chlamydomonas and Synechocystis, respectively, including membrane proteins such as photosystem proteins, ATP synthase, and NADH dehydrogenase, were successfully analyzed by nano-liquid chromatography tandem mass spectrometry. These results also indicated the improved efficiency of solubilization and trypsin digestion using PTS buffer. The results of the relative quantitative evaluation of photosystem subunits in Chlamydomonas and Synechocystis grown under high-light conditions were consistent with those of previous studies. Thus, the optimal cell disruption and PTS methods allow for comprehensive relative quantitative proteome analysis of photosynthetic organisms. Additionally, NdhD1 and NdhF1, which are NDH-1 subunit homologs, were increased under high-light conditions, suggesting that the NDH-1L complex, including NdhD1 and NdhF1, is increased under high-light conditions. This study is the first to explore the influence of incident light intensity on the photosynthetic responses under mixotrophic growth of microalga Asteracys sp. When grown mixotrophically, there was an enhanced regulation of non-photochemical quenching (NPQ) of the excited state of chlorophyll (Chl) a within the cells in response to white cool fluorescent high light (HL; 600 µmol photons m -2  s -1 ). Simultaneous measurement of reactive oxygen species (ROS) production as malondialdehyde (MDA) and ascorbate peroxidase (APX), an ROS scavenger, showed improved management of stress within mixotrophic cells under HL. Despite the observed decrease in quantum yield of photosynthesis measured through the Chl a fluorescence transient, no reduction in biomass accumulation was observed under HL for mixotrophy. However, biomass loss owing to photoinhibition was observed in cells grown phototrophically under the same irradiance. The measurements of dark recovery of NPQ suggested that "state transitions" may be partly responsible for regulating overall photosynthesis in Asteracys sp. The partitioning of photochemical and non-photochemical processes to sustain HL stress was analysed. Collectively, this study proposes that mixotrophy using glucose leads to a change in the photosynthetic abilities of Asteracys sp. while enhancing the adaptability of the alga to high irradiances.

As major primary producers in marine environments, diatoms are considered a valuable feedstock of biologically active compounds for application in several biotechnological fields. Due to their metabolic plasticity, especially for light perception and use and in order to make microalgal production more environmentally sustainable, marine diatoms are considered good candidates for the large-scale cultivation. Among physical parameters, light plays a primary role. Even if sunlight is cost-effective, the employment of artificial light becomes a winning strategy if a high-value microalgal biomass is produced. Several researches on marine diatoms are designed to study the influence of different light regimens to increase biomass production enriched in biotechnologically high-value compounds (lipids, carotenoids, proteins, polysaccharides), or with emphasised photonic properties of the frustule.

In the present study, to improve the photosynthetic betacarotene productivity of Dunaliella salina, a blue-red LED wavelength-shifting system (B-R system) was investigated. Dunaliella salina under the B-R system showed enhanced density and beta-carotene productivity compared to D. salina cultivated under single light-emitting diode light wavelengths (blue, white, and red light-emitting diode). Additionally, we developed blue light-adapted D. salina (ALE-D. salina) using an adaptive laboratory evolution (ALE) approach. In combination with the B-R system applied to ALE-D. salina (ALE B-R system), the beta-carotene concentration (33.94 ± 0.52 μM) was enhanced by 19.7% compared to that observed for the non-ALE-treated wild-type of D. salina (intact D. salina) under the B-R system (28.34 ± 0.24 μM).

Microalgae have been considered as alternative sustainable resources for high-value bioproducts such as lipids (especially triacylglycerides [TAGs]), polyunsaturated fatty acids (PUFAs), and carotenoids, due to their relatively high photosynthetic efficiency, no arable land requirement, and ease of scale-up. It is of great significance to exploit microalgae for the production of high-value bioproducts. How to improve the content or productivity of specific bioproducts has become one of the most urgent challenges. In this review, we will describe high-value bioproducts from microalgae and their biosynthetic pathways (mainly for lipids, PUFAs, and carotenoids). Recent progress and strategies for the enhanced production of bioproducts from microalgae are also described in detail, and these strategies take advantages of optimized cultivation conditions with abiotic stress, chemical stress (addition of metabolic precursors, phytohormones, chemical inhibitors, and chemicals inducing oxidative stress response), and molecular approaches such as metabolic engineering, transcriptional engineering, and gene disruption strategies (mainly RNAi, antisense RNA, miRNA-based knockdown, and CRISPR/Cas9).

The mutagenesis is an emerging strategy for screening microalgal candidates for CO 2 biofixation and biomass production. In this study, by 96-well microplates-UV mutagenesis, a mutant stemmed from Scenedesmus obliquus was screened and named as SDEC-1M. To characterize SDEC-1M, it was cultivated under air and high level CO 2 (15% v/v), and its parental strain (PS) was considered as control. Growth characterizations showed that SDEC-1M grew best in high level CO 2 . It indicated that the mutant had high CO 2 tolerance (HCT) and growth potential under high level CO 2 . Richer total carbohydrate content (37.26%) and lipid content (24.80%) demonstrated that, compared to its parental strain, SDEC-1M was apt to synthesize energy storage materials, especially under high CO 2 level. Meanwhile, the highest light conversion efficiency (approximately 18 %) was also obtained. Thus, the highest overall biomass productivities were achieved in SDEC-1M under high level CO 2 , largely attributed to that the highest productivities of total lipid, total carbohydrate, and crude protein were also achieved in the meantime. By modified UV, therefore, mutagenized SDEC-1M was the better candidate for CO 2 biofixation and biofuel production than its parental strain.

A photobioreactor (PBR) contains microalgae which under illumination consume carbon dioxide and substrate dissolved in water, and produce oxygen. The process is used in water recovery resource facilities with a continuous flow of wastewaster through the PBR. With several PBRs in series the reduction of substrate can be improved. This paper contains a thorough analysis of a model of PBRs in series, where each PBR is modelled with a system of three ordinary differential equations for the concentrations of dissolved substrate and biomass (algae), and the internal cell quota of substrate to biomass. Each PBR has a certain volume and irradiation. The absorption rate of substrate into the cells is modelled with Monod kinetics, whereas the biomass growth rate is modelled with Droop kinetics, in which both a minimum and a maximum internal cell quota are assumed. The main result is that the model has a unique stable steady-state solution with algae in all PBRs. Another stable steady-state solution is the wash-out solution with no algae in the system. Other steady-state solutions are combinations of these two with no algae in some of the first PBRs and algae in the rest of the PBRs in the series. Conditions on the illumination, volumetric flow and volumes of the PBRs are given for the respective solution. Background: Photosynthetic microalgae have been in the spotlight of biotechnological production (biofuels, lipids, etc), however, current barriers in mass cultivation of microalgae are limiting its successful industrialization. Therefore, a mathematical model integrating both the biological and hydrodynamical parts of the cultivation process may improve our understanding of relevant phenomena, leading to further optimization of the microalgae cultivation., Results: We introduce a unified multidisciplinary simulation tool for microalgae culture systems, particularly the photobioreactors. Our approach describes changes of cell growth determined by dynamics of heterogeneous environmental conditions such as irradiation and mixing of the culture. Presented framework consists of (i) a simplified model of microalgae growth in a culture system (the advection-diffusion-reaction system within a phenomenological model of photosynthesis and photoinhibition), (ii) the fluid dynamics (Navier-Stokes equations), and (iii) the irradiance field description (Beer-Lambert law). To validate the method, a simple case study leading to hydrodynamically induced fluctuating light conditions was chosen. The integration of computational fluid dynamics (ANSYS Fluent) revealed the inner property of the system, the flashing light enhancement phenomenon, known from experiments., Conclusion: Our physically accurate model of microalgae culture naturally exhibits features of real system, can be applied to any geometry of microalgae mass cultivation and thus is suitable for biotechnological applications.

Two marine microalgae, Nitzschia closterium and Isochrysis zhangjiangensis, treated for 9 days with photosynthetically active radiation (PAR), were subjected to 3 days of ultraviolet light band A at 365 nm (UVA365) followed by a 3 day post-UVA PAR treatment. Compared with the control that only had PAR treatment, UVA365 treatment significantly ( p < 0.05) increased the amounts of total xanthophylls in N. closterium and I. zhangjiangensis by up to 3.53- and 1.23-fold, respectively, after 3 days of UVA365 irradiation. In particular, UVA365 treatment markedly ( p < 0.05) increased two major bioactive xanthophyll pigments, fucoxanthin in N. closterium and diadinoxanthin in I. zhangjiangensis, by up to 1.97- and 1.52-fold, respectively. UVA365 treatment significantly ( p < 0.05) increased the accumulation of another bioactive microalgal metabolite, total mycosporine-like amino acids, in N. closterium and I. zhangjiangensis by up to 1.40- and 1.30-fold, respectively. UVA365 irradiation has potential as an efficient approach for enhancing the production of valuable microalgal metabolites.

Mixotrophy is a metabolic strategy in which an organism is autotrophic and heterotrophic simultaneously. Considering that the aquatic environment provides several organic sources of carbon, it is probably common for microalgae to perform mixotrophy and not only photoautotrophy, but little is known about microalgae mixotrophy. The present work aimed at investigating the growth, photosynthetic activity, morphology, and biochemical composition of the microalga Chlorella sorokiniana in mixotrophic and photo-mixotrophic conditions, comparing it with photoautotrophy. The results showed pH changes after glucose addition, reaching pH 11.62 in mixotrophic and 10.47 in sequential photo-mixotrophic cultures, which limited the microalgal growth. Highest biomass was obtained in the mixotrophic culture in comparison with the sequential photo-mixotrophic one. Rapid light saturation curves showed that α (photosynthetic efficiency, 1.69) and relative electron transport rate (rETR; 565.61) were higher in the mixotrophic cultures, whereas the highest I k (irradiance saturation, 386.68) was obtained in the photoautotrophic ones. In the sequential photo-mixotrophic cultures, photosynthetic activity varied during glucose consumption, decreasing the maximum quantum yield F v /F m after glucose addition, indicating change in metabolism, from photoautotrophy to mixotrophy by the microalga. The results showed that the mixotrophic cultures had higher production of chlorophyll a (6.26 mg mL -1 ), cell density (6.62 × 10 7 cell mL -1 ), and lipids (0.06 pg μm -3 ). Sequential photo-mixotrophic cultures showed the highest biovolume (360.5 μm 3  cell -1 ) and total carbohydrates (0.026 pg μm -3 ). The protein concentration was 3.2 and 2.4 times higher in photoautotrophy and photo-mixotrophic growth, respectively, than in mixotrophy, but lipids were three times higher under mixotrophy. The biochemical changes we observed indicate that the microalga's plasticity in face of new environmental characteristics, such as the presence of organic carbon, can change the flow of energy through natural ecosystems.

This paper is focused on the assessment of the production of algae in batch bioreactors. Hydraulic retention time, carbon loading rate and light color were the inputs of the study and algae production the main output. Bioreactors were operated in semi-continuous mode and tests lasted two months, more than two times the period required to meet a steady-state response. This steady-state was verified with plateau responses in both, soluble parameters and suspended solids. Results points out the great relevance of temperature. Likewise, they show that green light improves the production of algae, as well as long HRT and high CLR. Maximum production rates attained were in the range 4-14 mg d -1  L -1 . The ratio COD /TSS for this biofuel was almost constant (3.13 mg COD mg -1 TSS) but the quality of the product obtained in terms of the Mean Oxidation State of Carbon is completely different. Microalgae biotechnologies are rapidly developing into new commercial settings. Several high value products already exist on the market, and systems development is focused on cost reduction to open up future economic opportunities for food, fuel and freshwater production. Light is a key environmental driver for photosynthesis and optimising light capture is therefore critical for low cost, high efficiency systems. Here a novel high-throughput screen that simulates fluctuating light regimes in mass cultures is presented. The data was used to model photosynthetic efficiency (PE µ , mol photon -1 m 2 ) and chlorophyll fluorescence of two green algae, Chlamydomonas reinhardtii and Chlorella sp. Response surface methodology defined the effect of three key variables: density factor (D f , 'culture density'), cycle time (t c , 'mixing rate'), and maximum incident irradiance (I max ). Both species exhibited a large rise in PE µ with decreasing I max and a minimal effect of t c (between 3-20 s). However, the optimal D f of 0.4 for Chlamydomonas and 0.8 for Chlorella suggested strong preferences for dilute and dense cultures respectively. Chlorella had a two-fold higher optimised PE µ than Chlamydomonas, despite its higher light sensitivity. These results demonstrate species-specific light preferences within the green algae clade. Our high-throughput screen enables rapid strain selection and process optimisation.

Main Conclusion: Microalgae treated with blue light have potential for production of human nutrition supplement and biofuel due to their higher biomass productivity and favorable fatty acid composition. Chlorella vulgaris, Chlorella pyrenoidosa, Scenedesmus quadricauda and Scenedesmus obliquus are representative green microalgae which are widely reported for algal production. In this study, we provide a systematic investigation of the biomass productivity, photosynthetic pigments, chlorophyll fluorescence and fatty acid content of the four green microalgae. The strains were grown in two primary monochromatic light wavelengths [red and blue LEDs (light emitting diode)], and in white LED conditions, respectively. Among them, blue LED light was determined as the best light for growth rate, followed by red LED and white LED. The chlorophyll generation was more sensitive to the monochromatic blue light. The polyunsaturated fatty acids (PUFAs) such as α-linolenic acid (18:3), which were perfect for human nutrition supplementation, showed high concentrations in these algae strains under blue LED. Collectively, the results indicate that the blue LED is suitable for various food, feed, and algal biofuel productions due to both biomass and fatty acid productivity.

Melatonin (MLT), a ubiquitously distributed small molecule, functions in plant responses to various biotic and abiotic stresses. However, the interactions between melatonin and other important molecules in Haematococcus pluvialis response stresses are largely unknown. In the present study, exogenous melatonin improved H. pluvialis resistance to nitrogen starvation and high light. We concluded that exogenous melatonin treatment prevented the reactive oxygen species (ROS) burst and limited cell damage induced by abiotic stress through activation of antioxidant enzymes and antioxidants. Astaxanthin, a major antioxidant in H. pluvialis cells, exhibited a 2.25-fold increase in content after treatment with melatonin. The maximal astaxanthin content was 32.4 mg g -1 . The functional roles of the nitric oxide (NO)-mediated mitogen activated protein kinase (MAPK) signaling pathway and cyclic adenosine monophosphate (cAMP) signaling pathway induced by melatonin were also evaluated. The results clearly indicate that cAMP signaling pathways are positively associated with microalgal astaxanthin biosynthesis. Additionally, the NO-dependent MAPK signaling cascade is activated in response to astaxanthin accumulation induced by melatonin, confirming that MAPK is a target of NO action in physiological processes. This work is the first to use H. pluvialis as in vivo model and documents the influence of melatonin on the physiological response to abiotic stress in this microalgae.

Dinoflagellates of the genus Symbiodinium live in symbiosis with many invertebrates, including reef-building corals. Hosts maintain this symbiosis through continuous regulation of Symbiodinium cell density via expulsion and degradation (postmitotic) and/or constraining cell growth and division through manipulation of the symbiont cell cycle (premitotic). Importance of premitotic regulation is unknown since little data exists on cell cycles for the immense genetic diversity of Symbiodinium. We therefore examined cell cycle progression for several distinct SymbiodiniumITS2-types (B1, C1, D1a). All types exhibited typical microalgal cell cycle progression, G 1 phase through to S phase during the light period, and S phase to G 2 /M phase during the dark period. However, the proportion of cells in these phases differed between strains and reflected differences in growth rates. Undivided larger cells with 3n DNA content were observed especially in type D1a, which exhibited a distinct cell cycle pattern. We further compared cell cycle patterns under different growth light intensities and thermal regimes. Whilst light intensity did not affect cell cycle patterns, heat stress inhibited cell cycle progression and arrested all strains in G 1 phase. Violaxanthin is a major xanthophyll pigment in the microalga Eustigmatos cf. polyphem , but the amount produced after propagation can vary depending upon culture conditions. In this study, the effects of cultivation time, nitrogen concentration, light intensity, and culture mode on violaxanthin production were investigated. The results showed that this microalga vigorously grew and maintained a high level of violaxanthin in the fed-batch culture, and the highest violaxanthin productivity of 1.10 ± 0.03 mg L -1 d -1 was obtained under low light illumination with 18 mM of initial nitrogen supply for ten days. Additionally, violaxanthin was purified from E. cf. polyphem by silica gel chromatography and preparative high-performance liquid chromatography (PHPLC), and identified with high-resolution mass spectrometry (HRMS). The antioxidant activity of the purified violaxanthin was evaluated by three tests in vitro: reducing power assay, 2,2-diphenyl-1-picrylhydrazyl (DPPH), and 2,2-azobis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) radical assays. The strongest inhibition of purified violaxanthin occurred during the scavenging of ABTS⁺ radicals, with EC 50 of 15.25 μg mL -1 . In conclusion, this is the first report to investigate the effects of different culture conditions on violaxanthin accumulation in E. cf. polyphem and provide a novel source for the production of violaxanthin that can be used for food and pharmaceutical applications.

In this study, the effect of abiotic stress on the acidophilic eukaryotic microalga, Coccomyxa onubensis, was analyzed for the production of lutein and PUFAs (polyunsaturated fatty acids). It grows autotrophically at a pH of 2.5. It showed a growth rate of 0.30 d -1 , and produced approximately 122.50 mg·L -1 ·d -1 biomass, containing lipids (300.39 mg g -1 dw), lutein (5.30 mg g -1 dw), and β-carotene (1.20 mg g -1 dw). The fatty acid methyl ester (FAME) fraction was 89.70 mg g -1 dw with abundant palmitic acid (28.70%) and linoleic acid (37.80%). The addition of 100 mM NaCl improved the growth rate (0.54 d -1 ), biomass productivity (243.75 mg·L -1 ·d -1 ), and lipids accumulation (416.16 mg g -1 dw). The microalga showed a lutein content of 6.70 mg g -1 dw and FAME fraction of 118.90 mg g -1 dw; 68% of the FAMEs were PUFAs. However, when 200-500 mM salt was added, its growth was inhibited but there was a significant induction of lutein (up to 7.80 mg g -1 dw). Under continuous illumination with PAR (photosynthetically active radiations) +UVA (ultraviolet A, 8.7 W m -2 ), C. onubensis showed a growth rate of 0.40 d -1 , and produced 226.3 mg·L -1 ·d -1 biomass, containing lipids, (487.26 mg g -1 dw), lutein (7.07 mg g -1 dw), and FAMEs (232.9 mg g -1 dw); 48.4% of the FAME were PUFAs. The illumination with PAR + UVB (ultraviolet B, 0.16 W m -2 ) was toxic for cells.
