Abstract

Future cosmic microwave background (CMB) experiments are primarily targeting a detection of the primordial $B$-mode polarisation. The faintness of this signal requires exquisite control of systematic effects which may bias the measurements. In this work, we derive requirements on the relative calibration accuracy of the overall polarisation gain ($\Delta g_\nu$) for LiteBIRD experiment, through the application of the blind Needlet Internal Linear Combination (NILC) foreground-cleaning method. We find that minimum variance techniques, as NILC, are less affected by gain calibration uncertainties than a parametric approach, which requires a proper modelling of these instrumental effects. The tightest constraints are obtained for frequency channels where the CMB signal is relatively brighter (166 GHz channel, $\Delta {g}_\nu \approx 0.16 \%$), while, with a parametric approach, the strictest requirements were on foreground-dominated channels. We then propagate gain calibration uncertainties, corresponding to the derived requirements, into all frequency channels simultaneously. We find that the overall impact on the estimated $r$ is lower than the required budget for LiteBIRD by almost a factor $5$. The adopted procedure to derive requirements assumes a simple Galactic model. We therefore assess the robustness of obtained results against more realistic scenarios by injecting the gain calibration uncertainties, according to the requirements, into LiteBIRD simulated maps and assuming intermediate- and high-complexity sky models. In this case, we employ the so-called Multi-Clustering NILC (MC-NILC) foreground-cleaning pipeline and obtain that the impact of gain calibration uncertainties on $r$ is lower than the LiteBIRD gain systematics budget for the intermediate-complexity sky model. Abstract

Large angular scale surveys in the absence of atmosphere are essential for measuring the primordial $B$-mode power spectrum of the Cosmic Microwave Background (CMB). Since this proposed measurement is about three to four orders of magnitude fainter than the temperature anisotropies of the CMB, in-flight calibration of the instruments and active suppression of systematic effects are crucial. We investigate the effect of changing the parameters of the scanning strategy on the in-flight calibration effectiveness, the suppression of the systematic effects themselves, and the ability to distinguish systematic effects by null-tests. Next-generation missions such as LiteBIRD, modulated by a Half-Wave Plate (HWP), will be able to observe polarisation using a single detector, eliminating the need to combine several detectors to measure polarisation, as done in many previous experiments and hence avoiding the consequent systematic effects. While the HWP is expected to suppress many systematic effects, some of them will remain. We use an analytical approach to comprehensively address the mitigation of these systematic effects and identify the characteristics of scanning strategies that are the most effective for implementing a variety of calibration strategies in the multi-dimensional space of common spacecraft scan parameters. We also present Falcons, a fast spacecraft scanning simulator that we developed to investigate this scanning parameter space.

Abstract

We assess the capabilities of the LiteBIRD mission to map the hot gas distribution in the Universe through the thermal Sunyaev-Zeldovich (SZ) effect. Our analysis relies on comprehensive simulations incorporating various sources of Galactic and extragalactic foreground emission, while accounting for specific instrumental characteristics of LiteBIRD, such as detector sensitivities, frequency-dependent beam convolution, inhomogeneous sky scanning, and $1/f$ noise. We implement a tailored component-separation pipeline to map the thermal SZ Compton $y$-parameter over 98% of the sky. Despite lower angular resolution for galaxy cluster science, LiteBIRD provides full-sky coverage and, compared to the Planck satellite, enhanced sensitivity, as well as more frequency bands to enable the construction of an all-sky $y$-map, with reduced foreground contamination at large and intermediate angular scales. By combining LiteBIRD and Planck channels in the component-separation pipeline, we obtain an optimal $y$-map that leverages the advantages of both experiments, with the higher angular resolution of the Planck channels enabling the recovery of compact clusters beyond the LiteBIRD beam limitations, and the numerous sensitive LiteBIRD channels further mitigating foregrounds. The added value of LiteBIRD is highlighted through the examination of maps, power spectra, and one-point statistics of the various sky components. After component separation, the $1/f$ noise from LiteBIRD is effectively mitigated below the thermal SZ signal at all multipoles. Cosmological constraints on $S_8=\sigma_8\left(\Omega_{\rm m}/0.3\right)^{0.5}$ obtained from the LiteBIRD-Planck combined $y$-map power spectrum exhibits a 15% reduction in uncertainty compared to constraints from Planck alone. Abstract

LiteBIRD, the next-generation cosmic microwave background (CMB) experiment, aims for a launch in Japan's fiscal year 2032, marking a major advancement in the exploration of primordial cosmology and fundamental physics. Orbiting the Sun-Earth Lagrangian point L2, this JAXA-led strategic L-class mission will conduct a comprehensive mapping of the CMB polarization across the entire sky. During its 3-year mission, LiteBIRD will employ three telescopes within 15 unique frequency bands (ranging from 34 through 448 GHz), targeting a sensitivity of 2.2\,$\mu$K-arcmin and a resolution of 0.5$^\circ$ at 100\,GHz. Its primary goal is to measure the tensor-to-scalar ratio $r$ with an uncertainty $\delta r = 0.001$, including systematic errors and margin. If $r \geq 0.01$, LiteBIRD expects to achieve a $>5\sigma$ detection in the $\ell=$2-10 and $\ell=$11-200 ranges separately, providing crucial insight into the early Universe. We describe LiteBIRD's scientific objectives, the application of systems engineering to mission requirements, the anticipated scientific impact, and the operations and scanning strategies vital to minimizing systematic effects. Abstract

Abstract Abstract: Trichiurus lepturus (Actinopterygii, Perciformes) is a commercially and economically important fish. A total of 60 specimens of this cutlassfish were collected of the coast the municipalities of Niterói and Cabo Frio, state of Rio de Janeiro, Brazil. The fish were measured, necropsied, filleted and had their organs investigated for digenetic trematodes. Taxonomic identification was based on morphological and morphometric characters. The specimens of T. lepturus were parasitized with adult specimens of Lecithochirium monticellii. Parasite indices of prevalence, intensity, mean intensity, abundance, mean abundance, range of infection, and site of infection of parasitic species were evaluated. Notes on the taxonomy of the parasite were also included. This is the first report of L. monticellii parasitizing T. lepturus in Brazil.

Abstract

We present detailed forecasts for the constraints on primordial magnetic fields (PMFs) that will be obtained with the LiteBIRD satellite. The constraints are driven by the effects of PMFs on the CMB anisotropies: the gravitational effects of magnetically-induced perturbations; the effects on the thermal and ionization history of the Universe; the Faraday rotation imprint on the CMB polarization; and the non-Gaussianities induced in polarization anisotropies. LiteBIRD represents a sensitive probe for PMFs and by exploiting all the physical effects, it will be able to improve the current limit coming from Planck. In particular, thanks to its accurate $B$-mode polarization measurement, LiteBIRD will improve the constraints on infrared configurations for the gravitational effect, giving $B_{\rm 1\,Mpc}^{n_{\rm B} =-2.9} < 0.8$ nG at 95% C.L., potentially opening the possibility to detect nanogauss fields with high significance. We also observe a significant improvement in the limits when marginalized over the spectral index, $B_{1\,{\rm Mpc}}^{\rm marg}< 2.2$ nG at 95% C.L. From the thermal history effect, which relies mainly on $E$-mode polarization data, we obtain a significant improvement for all PMF configurations, with the marginalized case, $\sqrt{\langle B^2\rangle}^{\rm marg}<0.50$ nG at 95% C.L. Faraday rotation constraints will take advantage of the wide frequency coverage of LiteBIRD and the high sensitivity in $B$ modes, improving the limits by orders of magnitude with respect to current results, $B_{1\,{\rm Mpc}}^{n_{\rm B} =-2.9} < 3.2$ nG at 95% C.L. Finally, non-Gaussianities of the $B$-mode polarization can probe PMFs at the level of 1 nG, again significantly improving the current bounds from Planck. Altogether our forecasts represent a broad collection of complementary probes, providing conservative limits on PMF characteristics that will be achieved with LiteBIRD., Comment: 51 pages, 24 figures, abstract shortened

8. Tumor-Derived Exosomes Induce the Formation of Neutrophil Extracellular Traps: Implications For The Establishment of Cancer-Associated Thrombosis

Abstract

Abstract Cancer patients are at an increased risk of developing thromboembolic complications. Several mechanisms have been proposed to explain cancer-associated thrombosis including the release of tumor-derived extracellular vesicles and the activation of host vascular cells. It was proposed that neutrophil extracellular traps (NETs) contribute to the prothrombotic phenotype in cancer. In this study, we evaluated the possible cooperation between tumor-derived exosomes and NETs in cancer-associated thrombosis. Female BALB/c mice were orthotopically injected with 4T1 breast cancer cells. The tumor-bearing animals exhibited increased levels of plasma DNA and myeloperoxidase in addition to significantly increased numbers of circulating neutrophils. Mice were subjected to either Rose Bengal/laser-induced venous thrombosis or ferric chloride-induced arterial thrombosis models. The tumor-bearing mice exhibited accelerated thrombus formation in both models compared to tumor-free animals. Treatment with recombinant human DNase 1 reversed the prothrombotic phenotype of tumor-bearing mice in both models. Remarkably, 4T1-derived exosomes induced NET formation in neutrophils from mice treated with granulocyte colony-stimulating factor (G-CSF). In addition, tumor-derived exosomes interacted with NETs under static conditions. Accordingly, the intravenous administration of 4T1-derived exosomes into G-CSF-treated mice significantly accelerated venous thrombosis in vivo. Taken together, our observations suggest that tumor-derived exosomes and neutrophils may act cooperatively in the establishment of cancer-associated thrombosis.

Abstract

We present a study of the impact of an uncertainty in the beam far side-lobe knowledge on the measurement of the Cosmic Microwave Background $B$-mode signal at large scale. It is expected to be one of the main source of systematic effects in future CMB observations. Because it is crucial for all-sky survey missions to take into account the interplays between beam systematic effects and all the data analysis steps, the primary goal of this paper is to provide the methodology to carry out the end-to-end study of their effect for a space-borne CMB polarization experiment, up to the cosmological results in the form of a bias $\delta r$ on the tensor-to-scalar ratio $r$. LiteBIRD is dedicated to target the measurement of CMB primordial $B$ modes by reaching a sensitivity of $\sigma \left( r \right) \leq 10^{-3}$ assuming $r=0$. As a demonstration of our framework, we derive the relationship between the knowledge of the beam far side-lobes and the tentatively allocated error budget under given assumptions on design, simulation and component separation method. We assume no mitigation of the far side-lobes effect at any stage of the analysis pipeline. We show that $\delta r$ is mostly due to the integrated fractional power difference between the estimated beams and the true beams in the far side-lobes region, with little dependence on the actual shape of the beams, for low enough $\delta r$. Under our set of assumptions, in particular considering the specific foreground cleaning method we used, we find that the integrated fractional power in the far side-lobes should be known at a level as tight as $\sim 10^{-4}$, to achieve the required limit on the bias $\delta r < 1.9 \times 10^{-5}$. The framework and tools developed for this study can be easily adapted to provide requirements under different design, data analysis frameworks and for other future space-borne experiments beyond LiteBIRD.

Abstract

We estimate the efficiency of mitigating the lensing $B$-mode polarization, the so-called delensing, for the $LiteBIRD$ experiment with multiple external data sets of lensing-mass tracers. The current best bound on the tensor-to-scalar ratio, $r$, is limited by lensing rather than Galactic foregrounds. Delensing will be a critical step to improve sensitivity to $r$ as measurements of $r$ become more and more limited by lensing. In this paper, we extend the analysis of the recent $LiteBIRD$ forecast paper to include multiple mass tracers, i.e., the CMB lensing maps from $LiteBIRD$ and CMB-S4-like experiment, cosmic infrared background, and galaxy number density from $Euclid$- and LSST-like survey. We find that multi-tracer delensing will further improve the constraint on $r$ by about $20\%$. In $LiteBIRD$, the residual Galactic foregrounds also significantly contribute to uncertainties of the $B$-modes, and delensing becomes more important if the residual foregrounds are further reduced by an improved component separation method., Comment: 21 pages, 7 figures

Abstract

We explore the capability of measuring lensing signals in $LiteBIRD$ full-sky polarization maps. With a $30$ arcmin beam width and an impressively low polarization noise of $2.16\,\mu$K-arcmin, $LiteBIRD$ will be able to measure the full-sky polarization of the cosmic microwave background (CMB) very precisely. This unique sensitivity also enables the reconstruction of a nearly full-sky lensing map using only polarization data, even considering its limited capability to capture small-scale CMB anisotropies. In this paper, we investigate the ability to construct a full-sky lensing measurement in the presence of Galactic foregrounds, finding that several possible biases from Galactic foregrounds should be negligible after component separation by harmonic-space internal linear combination. We find that the signal-to-noise ratio of the lensing is approximately $40$ using only polarization data measured over $90\%$ of the sky. This achievement is comparable to $Planck$'s recent lensing measurement with both temperature and polarization and represents a four-fold improvement over $Planck$'s polarization-only lensing measurement. The $LiteBIRD$ lensing map will complement the $Planck$ lensing map and provide several opportunities for cross-correlation science, especially in the northern hemisphere.

Abstract

We study the possibility of using the $LiteBIRD$ satellite $B$-mode survey to constrain models of inflation producing specific features in CMB angular power spectra. We explore a particular model example, i.e. spectator axion-SU(2) gauge field inflation. This model can source parity-violating gravitational waves from the amplification of gauge field fluctuations driven by a pseudoscalar "axionlike" field, rolling for a few e-folds during inflation. The sourced gravitational waves can exceed the vacuum contribution at reionization bump scales by about an order of magnitude and can be comparable to the vacuum contribution at recombination bump scales. We argue that a satellite mission with full sky coverage and access to the reionization bump scales is necessary to understand the origin of the primordial gravitational wave signal and distinguish among two production mechanisms: quantum vacuum fluctuations of spacetime and matter sources during inflation. We present the expected constraints on model parameters from $LiteBIRD$ satellite simulations, which complement and expand previous studies in the literature. We find that $LiteBIRD$ will be able to exclude with high significance standard single-field slow-roll models, such as the Starobinsky model, if the true model is the axion-SU(2) model with a feature at CMB scales. We further investigate the possibility of using the parity-violating signature of the model, such as the $TB$ and $EB$ angular power spectra, to disentangle it from the standard single-field slow-roll scenario. We find that most of the discriminating power of $LiteBIRD$ will reside in $BB$ angular power spectra rather than in $TB$ and $EB$ correlations., Comment: 22 pages, 13 figures. Submitted to JCAP

Abstract

We estimate the efficiency of mitigating the lensing B-mode polarization, the so-called delensing, for the LiteBIRD experiment with multiple external data sets of lensing-mass tracers. The current best bound on the tensor-to-scalar ratio, r, is limited by lensing rather than Galactic foregrounds. Delensing will be a critical step to improve sensitivity to r as measurements of r become more and more limited by lensing. In this paper, we extend the analysis of the recent LiteBIRD forecast paper to include multiple mass tracers, i.e., the CMB lensing maps from LiteBIRD and CMB-S4-like experiment, cosmic infrared background, and galaxy number density from Euclid- and LSST-like survey. We find that multi-tracer delensing will further improve the constraint on r by about 20%. In LiteBIRD, the residual Galactic foregrounds also significantly contribute to uncertainties of the B-modes, and delensing becomes more important if the residual foregrounds are further reduced by an improved component separation method.

Abstract

We explore the capability of measuring lensing signals in LiteBIRD full-sky polarization maps. With a 30 arcmin beam width and an impressively low polarization noise of 2.16 ìK-arcmin, LiteBIRD will be able to measure the full-sky polarization of the cosmic microwave background (CMB) very precisely. This unique sensitivity also enables the reconstruction of a nearly full-sky lensing map using only polarization data, even considering its limited capability to capture small-scale CMB anisotropies. In this paper, we investigate the ability to construct a full-sky lensing measurement in the presence of Galactic foregrounds, finding that several possible biases from Galactic foregrounds should be negligible after component separation by harmonic-space internal linear combination. We find that the signal-to-noise ratio of the lensing is approximately 40 using only polarization data measured over 80% of the sky. This achievement is comparable to Planck's recent lensing measurement with both temperature and polarization and represents a four-fold improvement over Planck's polarization-only lensing measurement. The LiteBIRD lensing map will complement the Planck lensing map and provide several opportunities for cross-correlation science, especially in the northern hemisphere.

Abstract

We study the possibility of using the LiteBIRD satellite B-mode survey to constrain models of inflation producing specific features in CMB angular power spectra. We explore a particular model example, i.e. spectator axion-SU(2) gauge field inflation. This model can source parity-violating gravitational waves from the amplification of gauge field fluctuations driven by a pseudoscalar "axionlike"field, rolling for a few e-folds during inflation. The sourced gravitational waves can exceed the vacuum contribution at reionization bump scales by about an order of magnitude and can be comparable to the vacuum contribution at recombination bump scales. We argue that a satellite mission with full sky coverage and access to the reionization bump scales is necessary to understand the origin of the primordial gravitational wave signal and distinguish among two production mechanisms: quantum vacuum fluctuations of spacetime and matter sources during inflation. We present the expected constraints on model parameters from LiteBIRD satellite simulations, which complement and expand previous studies in the literature. We find that LiteBIRD will be able to exclude with high significance standard single-field slow-roll models, such as the Starobinsky model, if the true model is the axion-SU(2) model with a feature at CMB scales. We further investigate the possibility of using the parity-violating signature of the model, such as the TB and EB angular power spectra, to disentangle it from the standard single-field slow-roll scenario. We find that most of the discriminating power of LiteBIRD will reside in BB angular power spectra rather than in TB and EB correlations.

26. The Changi-Marina Bay Corridor: green strategies for Singapore’s soft power

Abstract

Singapore’s city-making strategies are seen by many Asia Pacific cities as a model to achieve today’s desired ‘world-class city’ status. They represent a source of innovation to design alternative configurations and rethink established norms of urban and social standards. This article looks at the urban landscape of the city of Singapore through infrastructural systems instead of individual design masterpieces, focusing on the urban corridor defined by the connection of Changi Airport, East Coast Parkway (ECP), and Marina Bay as a single urban entity, unified by a visual and experiential choreography. At a time of intense global competition in the aviation industry, Changi has deployed new strategies to sustain its reputation as an innovative transportation hub. The leg between Changi Airport and Marina Bay is an integral part of Singapore, experienced by the majority of its annual visitors arriving in the city-state. The 19-kilometre route, a major infrastructural project begun in the 1970s and still in progress, is a highly landscaped and scenic artery of the island, through which the airport has injected its hyper-urbanism into the city. While conveying the history of the developments of the Corridor, we consider the soft power strategies used by the city state to develop a form of transnational elite urbanism based on leisure and recreation, where urban planning, design, and green infrastructure play a key role. We reflect on the global and local dimension of Singapore, discussing how the ECP corridor reveals only one side of the city’s double character.On the one hand, this article critically analyses the rhetorical use of landscaping ingrained in the Corridor to attract foreign investment and tourism, where the artificialisation of green interventions has overwritten the presence of the native vegetation and morphology. Abstract

International audience; Introduction: We aimed to describe patients with coexisting infective endocarditis (IE) and bacterial meningitis (BM).Methods: We merged two large prospective cohorts, an IE cohort and a BM cohort, with only cases of definite IE and community-acquired meningitis. We compared patients who had IE and BM concurrently to patients with IE only and BM only.Results: Among the 1030 included patients, we identified 42 patients with IE-BM (4.1%). Baseline characteristics of patients with IE-BM were mostly similar to those of patients with IE, but meningitis was the predominant presentation at admission (39/42, 92.3%). Causative pathogens were predominantly Streptococcus pneumoniae (18/42, 42.9%) and Staphylococcus aureus (14/42, 33.3%). All pneumococcal IE were associated with BM (18/18). BM due to oral and group D streptococci, Streptococcus agalactiae, and S. aureus were frequently associated with IE (14/30, 46.7%). Three-month mortality was 28.6% in patients with IE-BM, 20.5% in patients with IE, and 16.6% in patients with BM.Conclusions: Patients with pneumococcal IE or altered mental status during IE must be investigated for BM. Patients with S. aureus, oral and group D streptococcal or enterococcal BM, or unfavorable outcome in pneumococcal meningitis would benefit from an echocardiography. Patients with the dual infection have the worst prognosis. Their identification is mandatory to initiate appropriate treatment.

Abstract

The influence of bleeding on color, texture profile, and chemical quality of frozen pirarucu loins were investigated. The loins were obtained from eighteen (n=18) pirarucu carcasses from animals slaughtered by bleeding (BLE; n = 9) or without bleeding (NON-BLE; n = 9). The loins were sliced and assigned randomly for 1 and 12 months at -20ºC to analyze pH, water holding capacity, instrumental color, and texture profile BLE samples exhibited higher L* values (P < 0.05), whereas BLE and NON-BLE exhibited similar (P > 0.05) pH, a* values, b* values, and color stability at months 1 and 12. NON-BLE samples demonstrated greater (P < 0.05) hardness and chewiness at month 1, whereas similar (P > 0.05) springiness and cohesiveness were observed in BLE and NON-BLE samples at months 1 and 12. During storage, Both samples (BLE and NON-BLE) demonstrated a decrease (P < 0.05) in pH and water holding capacity; and an increase (P < 0.05) in lightness, hardness, and chewiness. Also, BLE and NON-BLE exhibited stable (P > 0.05) redness, yellowness, color stability, and cohesiveness. NON-BLE samples exhibited a decrease (P < 0.05) in springiness from the 1st to the 12th month of frozen storage. Bleeding positively influenced the quality of frozen pirarucu.

Abstract

Human induced pluripotent stem cells (hiPSCs) were differentiated into a specific mesoderm subset characterized by KDR + CD56 + APLNR + (KNA + ) expression. KNA + cells had high clonal proliferative potential and specification into endothelial colony-forming cell (ECFCs) phenotype. KNA + cells differentiated into perfused blood vessels when implanted subcutaneously into the flank of nonobese diabetic/severe combined immunodeficient mice and when injected into the vitreous of type 2 diabetic mice ( db/db mice). Transcriptomic analysis showed that differentiation of hiPSCs derived from diabetics into KNA + cells was sufficient to change baseline differences in gene expression caused by the diabetic status and reprogram diabetic cells to a pattern similar to KNA + cells derived from nondiabetic hiPSCs. Proteomic array studies performed on retinas of db/db mice injected with either control or diabetic donor–derived KNA + cells showed correction of aberrant signaling in db/db retinas toward normal healthy retina. These data provide “proof of principle” that KNA + cells restore perfusion and correct vascular dysfunction in db/db mice.

Abstract

AIMS/HYPOTHESIS: Homo Sapiens evolved under conditions of intermittent food availability and prolonged fasting between meals. Periods of fasting are important for recovery from meal-induced oxidative and metabolic stress, and tissue repair. Constant high energy-density food availability in present-day society contributes to the pathogenesis of chronic diseases, including diabetes and its complications, with intermittent fasting (IF) and energy restriction shown to improve metabolic health. We have previously demonstrated that IF prevents the development of diabetic retinopathy in a mouse model of type 2 diabetes (db/db); however the mechanisms of fasting-induced health benefits and fasting-induced risks for individuals with diabetes remain largely unknown. Sirtuin 1 (SIRT1), a nutrient-sensing deacetylase, is downregulated in diabetes. In this study, the effect of SIRT1 stimulation by IF, fasting mimicking cell culture conditions (FMC) or pharmacological treatment using SRT1720 was evaluated on systemic and retinal metabolism, systemic and retinal inflammation and vascular and bone marrow damage. METHODS: The effects of IF were modelled in vivo using db/db mice and in vitro using bovine retinal endothelial cells or rat retinal neuroglial/precursor R28 cell line serum starved for 24 h. mRNA expression was analysed by quantitative PCR (qPCR). SIRT1 activity was measured via histone deacetylase activity assay. NR1H3 (also known as Liver X receptor alpha (LXRα)) acetylation was measured via western blot analysis. RESULTS: IF increased Sirt1 mRNA expression in mouse liver and retina when compared with non-fasted animals. IF also increased SIRT1 activity eightfold in mouse retina while FMC increased SIRT1 activity and expression in retinal endothelial cells when compared with control. Sirt1 expression was also increased twofold in neuronal retina progenitor cells (R28) after FMC treatment. Moreover, FMC led to SIRT1-mediated LXRα deacetylation and subsequent 2.4-fold increase in activity, as measured by increased mRNA expression of the genes encoding ATP-binding cassette transporter (Abca1 and Abcg1). These changes were reduced when retinal endothelial cells expressing a constitutively acetylated LXRα mutant were tested. Increased SIRT1/LXR/ABC-mediated cholesterol export resulted in decreased retinal endothelial cell cholesterol levels. Direct activation of SIRT1 by SRT1720 in db/db mice led to a twofold reduction of diabetes-induced inflammation in the retina and improved diabetes-induced visual function impairment, as measured by electroretinogram and optokinetic response. In the bone marrow, there was prevention of diabetes-induced myeloidosis and decreased inflammatory cytokine expression. CONCLUSIONS/INTERPRETATION: Taken together, activation of SIRT1 signalling by IF or through pharmacological activation represents an effective therapeutic strategy that provides a mechanistic link between the advantageous effects associated with fasting regimens and prevention of microvascular and bone marrow dysfunction in diabetes.

Abstract

Background: DLK1-DIO3 genomic region comprises one of the largest microRNA (miRNAs) clusters in human genome. In previous studies we showed the downregulation of several miRNAs from the genomic region in papillary thyroid carcinoma (PTC). Due to the large number of miRNAs within this region the individual contribution of these molecules to PTC development and progression remains unclear. Methods: We used different computational resources to clarify the contribution of DLK1-DIO3-derived miRNAs to PTC. Results: Our analysis suggests that 12 miRNAs from this region cooperate to modulate distinct cancer-relevant biological processes, potentially responding for most of the impact of DLK1-DIO3-derived miRNAs to PTC development and progression. The overexpression of miR-485-5p in two PTC cell lines decreased proliferation and migration, confirming the biological relevance of in silico data. Conclusion: Our results shed light on the role of DLK1-DIO3 region, harboring several tumor suppressor miRNAs in thyroid cancer and open perspectives for the functional exploration of these miRNAs as therapeutic targets for PTC.

Abstract

Hyperbaric oxygen is a clinical treatment that contributes to wound healing by increasing fibroblasts proliferation, collagen synthesis, and production of growth factors, inducing angiogenesis and inhibiting antimicrobial activity. It also has been shown that hyperbaric oxygen treatment (HBO), through the activation of nitric oxide synthase promotes an increase in the nitric oxide levels that may improve endothelial progenitor cells (EPC) mobilization from bone marrow to the peripheral blood and stimulates the vessel healing process. However, cellular mechanisms involved in cell proliferation and activation of EPC after HBO treatment remain unknown. Therefore, the present work aimed to analyze the effect of HBO on the proliferation of pre-treated bone marrow-derived EPC with TNF-alpha. Also, we investigated the expression of ICAM and eNOS by immunochemistry, the production of reactive species of oxygen and performed an in vitro wound healing. Although 1h of HBO treatment did not alter the rate of in vitro wound closure or cell proliferation, it increased eNOS expression and decreased ICAM expression and reactive oxygen species production in cells pre-treated with TNF-alpha. These results indicate that HBO can decrease the inflammatory response in endothelial cells mediated by TNF-alpha, and thus, promote vascular recovery after injury.

Abstract

Cancer patients are at an increased risk of developing thromboembolic complications. Several mechanisms have been proposed to explain cancer-associated thrombosis including the release of tumor-derived extracellular vesicles and the activation of host vascular cells. It was proposed that neutrophil extracellular traps (NETs) contribute to the prothrombotic phenotype in cancer. In this study, we evaluated the possible cooperation between tumor-derived exosomes and NETs in cancer-associated thrombosis. Female BALB/c mice were orthotopically injected with 4T1 breast cancer cells. The tumor-bearing animals exhibited increased levels of plasma DNA and myeloperoxidase in addition to significantly increased numbers of circulating neutrophils. Mice were subjected to either Rose Bengal/laser-induced venous thrombosis or ferric chloride-induced arterial thrombosis models. The tumor-bearing mice exhibited accelerated thrombus formation in both models compared to tumor-free animals. Treatment with recombinant human DNase 1 reversed the prothrombotic phenotype of tumor-bearing mice in both models. Remarkably, 4T1-derived exosomes induced NET formation in neutrophils from mice treated with granulocyte colony-stimulating factor (G-CSF). In addition, tumor-derived exosomes interacted with NETs under static conditions. Accordingly, the intravenous administration of 4T1-derived exosomes into G-CSF-treated mice significantly accelerated venous thrombosis in vivo. Taken together, our observations suggest that tumor-derived exosomes and neutrophils may act cooperatively in the establishment of cancer-associated thrombosis.

Abstract

In diabetic dyslipidemia, cholesterol accumulates in the plasma membrane, decreasing fluidity and thereby suppressing the ability of cells to transduce ligand-activated signaling pathways. Liver X receptors (LXRs) are the main cellular mechanism by which intracellular cholesterol is regulated and play important roles in inflammation and disease pathogenesis. N,N-dimethyl-3β-hydroxy-cholenamide (DMHCA), a selective LXR agonist, specifically activates the cholesterol efflux arm of the LXR pathway without stimulating triglyceride synthesis. Thus, DMHCA possesses superior clinical potential as a cholesterol lowering agent than current LXR pan-agonist. In this study, we use a multi-systems approach to understand the effects and molecular mechanisms of DMHCA treatment in type 2 diabetic db/db mice and human -derived circulating angiogenic cells (CACs), which are vascular reparative cells. We find that DMHCA is sufficient to correct the retina-bone marrow (BM) axis in diabetes, thereby restoring retinal structure, function, and cholesterol homeostasis, rejuvenating membrane fluidity in circulating vascular reparative cells, hampering systemic inflammation, and correcting BM dysfunction. Using single-cell RNA-seq on lineage-sca1+cKit+(LSK) hematopoietic stem cells (HSCs) from untreated and DMHCA-treated diabetic mice, we provide novel insights into hematopoiesis and reveal DMHCA’s mechanism of action in correcting diabetic HSCs by reducing myeloidosis and increasing CACs and erythrocyte progenitors. Taken together, these findings demonstrate the broad and pleiotropic effects of DMHCA treatment, which has exciting potential to correct the retina-BM axis in diabetic subjects.

Abstract

Human induced pluripotent stem cells (hiPSC) differentiated into a specific mesoderm subset expressing vascular endothelial growth factor receptor 2, neural cell adhesion molecule 1, and apelin G protein-coupled receptor APJ (called KNA+), possessed all of the phenotypic and functional endothelial colony forming cell potential that resides in differentiating hiPSC. Thus, we postulated KNA+ mesoderm treatment would correct diabetic retinal capillary vasodegeneration. KNA+ cells derived from diabetic (D) and non-diabetic (N) patient-derived hiPSC displayed comparable phenotypic and functional properties. N-KNA+ or D-KNA+ intravitreal injections into type II diabetic ( db/db ) murine eyes led to retinal vascular engraftment, showed no toxicity, visual acuity was significantly improved, and diabetes-induced scotopic and photopic ERG defects were corrected. Proteomic arrays revealed that hiPSC-derived KNA+ cell administration restored several aberrant cell signaling pathways in the neural retina. These results support the efficacy of both hiPSC-derived D-KNA+ and N-KNA+ cells in correcting vasodegeneration in the diabetic murine retina.

Abstract

SummaryIn diabetes, the retina, a tissue with unique metabolic needs, demonstrates dysregulation of the intricate balance between nutrient availability and utilization. This results in cholesterol accumulation, pro-inflammatory and pro-apoptotic changes, and consequently neurovascular damage. Sirtuin 1 (SIRT1), a nutrient sensing deacetylase, is downregulated in the diabetic retina. In this study, the effect of SIRT1 stimulation by fasting or by pharmacological activation using SRT1720, was evaluated on retinal cholesterol metabolism, inflammation and neurovascular damage. SIRT1 activation, in retinal endothelial cells (REC) and neuronal retinal progenitor cells (R28), led to Liver X Receptor alpha (LXRα) deacetylation and subsequent increased activity, as measured by increased ATP-binding cassette transporter (ABC) A1 and G1 mRNA expression. In turn, increased cholesterol export resulted in decreased REC cholesterol levels. SIRT1 activation also led to decreased inflammation. SIRT1 activation, in vivo, prevented diabetes-induced inflammation and vascular and neural degeneration. Diabetes-induced visual function impairment, as measured by electroretinogram and optokinetic response, was significantly improved as a result of SIRT1 activation. Taken together, activation of SIRT1 signaling is an effective therapeutic strategy that provides a mechanistic link between the advantageous effects associated with fasting regimes and prevention of diabetic retinopathy (DR).
