Isothiocyanates (ITCs) are a class of natural or synthetic compounds characterized by the general formula R–NCS. ITCs show antihyperalgesic effects in models of central and peripheral nervous tissue injury and anti-inflammatory properties. The pharmacodynamics are strictly related to the release of the gasotransmitter hydrogen sulfide (H2S) from their moiety. In particular, phenyl ITC (PITC) and 3-carboxyphenyl ITC (3C-PITC) exhibit interesting slow H2S-release properties suitable for treating painful pathology. The aim of the present work was to evaluate the efficacy of PITC and 3C-PITC against mechanical hyperalgesia and spontaneous pain induced by nerve injury and osteoarthritis.Methods: Nerve injury and osteoarthritis were induced in rats by ligation of the sciatic nerve (chronic constriction injury) and intra-articular injection of monoiodoacetate, respectively. Behavioral tests were performed 14 days after damage induction.Results: Single subcutaneous administrations of PITC, 3C-PITC (4.43 and 13.31 µmol kg-1, respectively) were able to completely reverse hypersensitivity to noxious stimuli in both models of neuropathic and osteoarticular pain. The effect of ITCs was compared with that of NaHS, the prototypical H2S donor, showing similar efficacy and higher potency. ITCs and NaHS also reduced spontaneous pain.Conclusion: ITCs offer a promising novel approach to counteract persistent, drug-resistant painful pathology. 2. F1 Domain of the Leishmania (Leishmania) donovani Nucleoside Hydrolase Promotes a Th1 Response in Leishmania (Leishmania) infantum Cured Patients and in Asymptomatic Individuals Living in an Endemic Area of Leishmaniasis

Abstract

The Leishmania (Leishmania) donovani nucleoside hydrolase NH36 is the main antigen of the Leishmune® vaccine and one of the promising candidates for vaccination against visceral leishmaniasis. The antigenicity of the N-terminal (F1), the central (F2), or the C-terminal recombinant domain (F3) of NH36 was evaluated using peripheral blood mononuclear cells (PBMC) from individuals infected with L. (L.) infantum from an endemic area of visceral leishmaniasis of Spain. Both NH36 and F1 domains significantly increased the PBMC proliferation stimulation index of cured patients and infected asymptomatic individuals compared to healthy controls. Moreover, F1 induced a 19% higher proliferative response than NH36 in asymptomatic exposed subjects. In addition, in patients cured from visceral leishmaniasis, proliferation in response to NH36 and F1 was accompanied by a significant increase of IFN-γ and TNF-α secretion, which was 42–43% higher, in response to F1 than to NH36. The interleukin 17 (IL-17) secretion was stronger in asymptomatic subjects, in response to F1, as well as in cured cutaneous leishmaniasis after NH36 stimulation. While no IL-10 secretion was determined by F1, a granzyme B increase was detected in supernatants from cured patients after stimulation with either NH36 or F1. These data demonstrate that F1 is the domain of NH36 that induces a recall cellular response in individuals with acquired resistance to the infection by L. (L.) infantum. In addition, F1 and NH36 discriminated the IgG3 humoral response in patients with active visceral leishmaniasis due to L. (L.) donovani (Ethiopia) and L. (L.) infantum (Spain) from that of endemic and non-endemic area controls. NH36 showed higher reactivity with sera from L. (L.) donovani-infected individuals, indicating species specificity. We conclude that the F1 domain, previously characterized as an inducer of the Th1 and Th17 responses in cured/exposed patients infected with L. (L.) infantum chagasi, may also be involved in the generation of a protective response against L. (L.) infantum and represents a potential vaccine candidate for the control of human leishmaniasis alone, or in combination with other HLA epitopes/antigens.

3. Compatibilization efficiency of carboxylated nitrile rubber and epoxy pre-polymer in nitrile/acrylic rubber blends

Abstract

An investigation has been made of the effects from a compatibilizer, viz. carboxylated nitrile rubber (XNBR), on several properties of nitrile rubber (NBR) and acrylic rubber (ACM) blends, including curing characteristics, mechanical, dynamic mechanical and dielectric properties. The presence of XNBR until 10 phr resulted in an improvement of the ultimate tensile properties, especially elongation at break. The mechanical properties associated to the volume fraction of rubber in the network (Vr) and torque values suggest the co-vulcanization phenomenon imparted by the compatibilization. The oil resistance of NBR/ACM (50:50 wt. (%)) blends (compatibilized and non compatibilized) was similar to that observed for pure ACM and significantly higher than NBR. The addition of small amounts of epoxy pre-polymer in combination with XNBR resulted in an additional improvement of the tensile properties. The dynamic mechanical and dielectric properties of the blends were also investigated. The loss modulus values of the compatibilized blends were significantly lower indicating an increase of the elastic characteristics. All blends presented two dielectric relaxation peaks confirming the heterogeneity of the compatibilized blends

4. Concentrados de ácidos graxos insaturados obtidos a partir de óleo de Carpa (Cyprinus carpio) utilizando o método da complexação com uréia

Abstract

O trabalho teve como objetivo estabelecer as melhores condições para obter concentrados de ácidos graxos mono e poliinsaturados a partir do óleo branqueado de carpa (Cyprinus carpio), utilizando o método de complexação com uréia. Os fatores de estudo foram: relação de uréia-ácido graxo (2:1 e 6:1), temperatura de cristalização (4ºC e -12ºC) e tempo de cristalização (14 e 24 h). As respostas para análise estatística foram o somatório de ácidos graxos saturados (Σ(AGS)); somatório de ácidos graxos monoinsaturados e poliinsaturados (Σ(AGMI+AGPI)); somatório de ácido graxo eicosapentaenoico e docosahexaenóico (Σ(EPA+DHA)). A relação uréia/AG se mostrou muito significativa, de forma diretamente proporcional na obtenção dos concentrados. A relação entre temperatura e rendimento dos concentrados foi de forma inversamente proporcional, e o tempo foi significativo, porém com menor influência. As melhores condições para a obtenção de concentrados foram: maior relação de uréia/AG (6:1), menor temperatura (-12ºC) e menor tempo (14 h). Sendo que nestas condições, os AGMI+AGPI constituíram 85,2% do concentrado, e entre estes os EPA+DHA foram de 9,4%. As frações líquidas apresentaram rendimento em massa de até 65,4%, e seu percentual de ácidos graxos livres ficou em média 35,8 g/100g.

Abstract

Significance\ud \ud Communicating in ways that motivate engagement in social distancing remains a critical global public health priority during the COVID-19 pandemic. This study tested motivational qualities of messages about social distancing (those that promoted choice and agency vs. those that were forceful and shaming) in 25,718 people in 89 countries. The autonomy-supportive message decreased feelings of defying social distancing recommendations relative to the controlling message, and the controlling message increased controlled motivation, a less effective form of motivation, relative to no message. Message type did not impact intentions to socially distance, but people’s existing motivations were related to intentions. Findings were generalizable across a geographically diverse sample and may inform public health communication strategies in this and future global health emergencies.\ud \ud \ud \ud Abstract\ud \ud Finding communication strategies that effectively motivate social distancing continues to be a global public health priority during the COVID-19 pandemic. This cross-country, preregistered experiment (n = 25,718 from 89 countries) tested hypotheses concerning generalizable positive and negative outcomes of social distancing messages that promoted personal agency and reflective choices (i.e., an autonomy-supportive message) or were restrictive and shaming (i.e., a controlling message) compared with no message at all. Results partially supported experimental hypotheses in that the controlling message increased controlled motivation (a poorly internalized form of motivation relying on shame, guilt, and fear of social consequences) relative to no message. On the other hand, the autonomy-supportive message lowered feelings of defiance compared with the controlling message, but the controlling message did not differ from receiving no message at all. Unexpectedly, messages did not influence autonomous motivation (a highly internalized form of motivation relying on one’s core values) or behavioral intentions. Results supported hypothesized associations between people’s existing autonomous and controlled motivations and self-reported behavioral intentions to engage in social distancing. Controlled motivation was associated with more defiance and less long-term behavioral intention to engage in social distancing, whereas autonomous motivation was associated with less defiance and more short- and long-term intentions to social distance. Overall, this work highlights the potential harm of using shaming and pressuring language in public health communication, with implications for the current and future global health challenges.

Contains fulltext : 284232.pdf (Publisher’s version ) (Closed access) The COVID-19 pandemic (and its aftermath) highlights a critical need to communicate health information effectively to the global public. Given that subtle differences in information framing can have meaningful effects on behavior, behavioral science research highlights a pressing question: Is it more effective to frame COVID-19 health messages in terms of potential losses (e.g., "If you do not practice these steps, you can endanger yourself and others") or potential gains (e.g., "If you practice these steps, you can protect yourself and others")? Collecting data in 48 languages from 15,929 participants in 84 countries, we experimentally tested the effects of message framing on COVID-19-related judgments, intentions, and feelings. Loss- (vs. gain-) framed messages increased self-reported anxiety among participants cross-nationally with little-to-no impact on policy attitudes, behavioral intentions, or information seeking relevant to pandemic risks. These results were consistent across 84 countries, three variations of the message framing wording, and 560 data processing and analytic choices. Thus, results provide an empirical answer to a global communication question and highlight the emotional toll of loss-framed messages. Critically, this work demonstrates the importance of considering unintended affective consequences when evaluating nudge-style interventions. 26 p.

Contains fulltext : 282577.pdf (Publisher’s version ) (Open Access) The UK House of Commons Science and Technology Committee has called for evidence on the roles that different stakeholders play in reproducibility and research integrity. Of central priority are proposals for improving research integrity and quality, as well as guidance and support for researchers. In response to this, we argue that there is one important component of research integrity that is often absent from discussion: the pedagogical consequences of how we teach, mentor, and supervise students through open scholarship. We justify the need to integrate open scholarship principles into research training within higher education and argue that pedagogical communities play a key role in fostering an inclusive culture of open scholarship. We illustrate these benefits by presenting the Framework for Open and Reproducible Research Training (FORRT), an international grassroots community whose goal is to provide support, resources, visibility, and advocacy for the adoption of principled, open teaching and mentoring practices, whilst generating conversations about the ethics and social impact of higher-education pedagogy. Representing a diverse group of early-career researchers and students across specialisms, we advocate for greater recognition of and support for pedagogical communities, and encourage all research stakeholders to engage with these communities to enable long-term, sustainable change.

Abstract

Unsaturated fatty acid concentrates (UFAC) were nanoencapsulated by emulsification/lyophilization using chitosan:gelatin matrices as wall material. Nanoemulsions presented droplets size between 40 and 280 nm. There was a decrease in droplets size due to gelatin addition, resulting in a decrease of encapsulation efficiency and an increase in porosity and pores volume. UFAC oxidation in chitosan capsules was slower than in free oil. Sorption isotherms were performed at 4 and 25 °C, and GAB model showed the best fit to experimental data. Differential enthalpy and entropy increased with the moisture content decrease, and the adsorption process was spontaneous. Pores radius increased with the increase of gelatin in the microstructures. The critical storage conditions of the microstructures were calculated. The range of water activity, which represents the maximum stability zone, was of 0.65–0.75 at 4 °C and of 0.73–0.85 at 25 °C. This zone hinders the oxygen diffusion through the pores, retarding oxidation.

Abstract

The COVID-19 pandemic has increased negative emotions and decreased positive emotions globally. Left unchecked, these emotional changes might have a wide array of adverse impacts. To reduce negative emotions and increase positive emotions, we tested the effectiveness of reappraisal, an emotion-regulation strategy that modifies how one thinks about a situation. Participants from 87 countries and regions (n = 21,644) were randomly assigned to one of two brief reappraisal interventions (reconstrual or repurposing) or one of two control conditions (active or passive). Results revealed that both reappraisal interventions (vesus both control conditions) consistently reduced negative emotions and increased positive emotions across different measures. Reconstrual and repurposing interventions had similar effects. Importantly, planned exploratory analyses indicated that reappraisal interventions did not reduce intentions to practice preventive health behaviours. The findings demonstrate the viability of creating scalable, low-cost interventions for use around the world. Abstract

With a view to sustainable production and circular economy, the recovery of biomass otherwise destined for disposal will be a strategic objective in the near future. Yearly, the tomato industry produces hundreds of thousands of tons of waste, which could be usefully applied in various sectors. This work proposes the use of the skins of a hyperpigmented (black-skinned) tomato line for the stabilization of silver nanoparticles. Aqueous extracts produced from this residue have proven to be suitable for producing silver nanoparticles with an average diameter of 22.4 nm. Subsequently AgNPs were tested for in vitro cowpea weevil (Callosobruchus maculatus) control. At a concentration of 20% they proved capable of inducing 52% mortality on adults of the insect. Abstract

The sorption isotherms of chitosan-glycerol films and of pure chitosan films using the static gravimetric method were performed. The experimental data were adjusted by several sorption models, and the GAB equation was the one that best represented the experimental data, being chosen to predict the adsorption and desorption behaviors of the films for the ranges studied of water activity (0.043–0.888) and of temperatures (20–50 °C). The chitosan-glycerol films presented higher values of monolayer moisture and lower heat involved in the monolayer compared to the pure chitosan films. The values of net isosteric heat and of differential entropy for the films increased exponentially with the moisture content decrease. Dry films with moisture content (Xe) lower than 0.10 kg kg−1 (dry basis) presented morphological changes according to the isosteric heat and entropy curves, indicating water sorption more easily and the more amorphous material (crystallinity of 9%). While dry films at Xe = 0.40 kg kg−1 (dry basis) maintained the characteristic of semicrystalline material (crystallinity of 33%). The enthalpy–entropy compensation showed that the process was controlled by the enthalpy and, it was only spontaneous for the samples in adsorption. The films were classified as a micro and mesoporous materials, and the average pores sizes were from 0.6 to 17 nm for desorption and, from 0.7 to 21 nm for adsorption.
