Aim of study: The objective of this study was to evaluate the effects of spineless cactus genotypes (Nopalea cochenillifera or Opuntia stricta) on the carcass characteristics and meat quality of feedlot lambs. Abstract Scientometrics enables us to comprehend the interests and trends in scientific knowledge production and dissemination. In this study, we evaluate the effects of gender, academic experience, location of residence, and graduate program score on the quantity of published articles, the number of citations and the H-index of researchers belonging to Brazilian graduate programs in Biodiversity. Variables related to the researchers were measured, and the relevance in explaining scientific production was examined using hierarchical models. In graduate programs, there were more men than females. The number of articles as first author and the H-index increase progressively through the researchers’ career, while the number of citations increases at the beginning of their careers, stabilizing between 10 and 20 years, and increasing again after 30 years of career. We concluded that gender, academic experience, and graduate program score were the most important variables in explaining the scientific production of graduate programs in Biodiversity in Brazil.

Hypertrophic cardiomyopathy is a common genetic heart disease characterized predominantly by non-dilated hypertrophy of the left ventricle, in the absence of other cardiac, systemic, or metabolic diseases that can cause the observed degree of hypertrophy. The main features of left ventricular outflow tract obstruction include anatomical - basal septal hypertrophy in combination with a relatively small left ventricular cavity, leading to a change in the geometry of the outflow tract from the left ventricle, pushing the mitral valve apparatus forward, and functional - systolic anterior motion of elongated mitral valve leaflets with subsequent mitral-septal contact. Primary anatomical changes of the mitral valve make the valve more susceptible to abnormal flow vectors generated in the left ventricle cavity, creating the conditions for systolic anterior motion and mitral regurgitation. Septal myectomy, performed by experienced surgeons in specialized centers, eliminates obstruction at all levels of the left ventricular outflow tract, with a clinical success rate of 90-95% and a mortality rate of less than 1%. Some surgeons complement septal myectomy with intervention on the mitral valve, taking into account anatomical features when isolated septal myectomy may be ineffective. The choice of the most optimal concurrent intervention on the mitral valve during septal myectomy is undefined. Techniques for valve-sparing mitral valve surgery have been developed, including resection of secondary chords, edge-to-edge repair, anterior leaflet plication, and others. This review presents the hemodynamic outcomes of concomitant interventions on the mitral valve in addition to septal myectomy in patients with obstructive hypertrophic cardiomyopathy.

Polymerization of axin proteins is essential to suppress Wnt/β-catenin signaling. Here, the authors identify an aggregation site in the conductin/axin2 RGS domain that prevents its polymerization and show that a short peptide masking this aggregon promotes polymerization of conductin/axin2, downregulation of Wnt pathway activity and growth inhibition of colorectal cancer cells.

BackgroundChronic kidney disease is commonly found in patients with aortic stenosis (AS) undergoing transcatheter aortic valve replacement (TAVR) and has marked impact in their prognosis. It has been shown however that TAVR may improve renal function by alleviating the hemodynamic barrier imposed by AS. Nevertheless, the predictors of and clinical consequences of renal function improvement are not well established. Our aim was to assess the predictors of improvement of renal function after TAVR.MethodsThe present work is an analysis of the Brazilian Registry of TAVR, a national non-randomized prospective study with 22 Brazilian centers. Patients with baseline renal dysfunction (estimated glomerular filtration rate [eGFR] < 60mL/min/1.73m2) were stratified according to renal function after TAVR: increase >10% in eGFR were classified as TAVR induced renal function improvement (TIRFI); decrease > 10% in eGFR were classified as acute kidney injury (AKI) and stable renal function (neither criteria).ResultsA total of 819 consecutive patients with symptomatic severe AS were included. Of these, baseline renal dysfunction (estimated glomerular filtration rate [eGFR] < 60mL/min/1.73m2) was present in 577 (70%) patients. Considering variance in renal function between baseline and at discharge after TAVR procedure, TIRFI was seen in 197 (34.1%) patients, AKI in 203 (35.2%), and stable renal function in 177 (30.7%). The independent predictors of TIRFI were: absence of coronary artery disease (OR: 0.69; 95% CI 0.48-0.98; P = 0.039) and lower baseline eGFR (OR: 0.98; 95% CI 0.97-1.00; P = 0.039). There was no significant difference in 30-day and 1-year all-cause mortality between patients with stable renal function or TIRFI. Nonetheless, individuals that had AKI after TAVR presented higher mortality compared with TIRFI and stable renal function groups (29.3% vs. 15.4% vs. 9.5%, respectively; p < 0.001).ConclusionsTIRFI was frequently found among baseline impaired renal function individuals but was not associated with improved 1-year outcomes.

Despite the medical importance of G protein-coupled receptors (GPCRs), in vivo cellular heterogeneity of GPCR signaling and downstream transcriptional responses are not understood. We report the comprehensive characterization of transcriptomes (bulk and single-cell) and chromatin domains regulated by sphingosine 1-phosphate receptor-1 (S1PR1) in adult mouse aortic endothelial cells. First, S1PR1 regulates NFκB and nuclear glucocorticoid receptor pathways to suppress inflammation-related mRNAs. Second, S1PR1 signaling in the heterogenous endothelial cell (EC) subtypes occurs at spatially-distinct areas of the aorta. For example, a transcriptomically distinct arterial EC population at vascular branch points (aEC1) exhibits ligand-independent S1PR1/ß-arrestin coupling. In contrast, circulatory S1P-dependent S1PR1/ß-arrestin coupling was observed in non-branch point aEC2 cells that exhibit an inflammatory gene expression signature. Moreover, S1P/S1PR1 signaling regulates the expression of lymphangiogenic and inflammation-related transcripts in an adventitial lymphatic EC (LEC) population in a ligand-dependent manner. These insights add resolution to existing concepts of endothelial heterogeneity, GPCR signaling and S1P biology.

Resumo: Objetivando-se avaliar o efeito da adição de palma forrageira associado à maniçoba na dieta de ovinos sobre o perfil de indicadores bioquímicos do metabolismo energético e proteico, foi realizado um delineamento em blocos casualizados onde foram utilizados 24 ovinos machos, sem padrão racial definido, com peso corporal médio de 19,77±1,95 kg e idade média de seis meses, divididos igualmente em três tratamentos: concentrado + feno Tifton 85, concentrado + feno de maniçoba e concentrado + silagem de maniçoba, e semelhantes partes de palma forrageira. Realizaram-se quatro coletas de sangue, que constituíram as repetições, com intervalos de 15 dias (0d, 15d, 3d0 e 45d). BACKGROUND:The emergence of the Chikungunya virus (CHIKV) is currently expanding. In 2015, 38,332 cases of Chikungunya were reported to the Brazilian epidemiological surveillance system. Eighteen months after notification of the first case in the city of Feira de Santana, we conducted the first serosurvey to define the magnitude of transmission in a rural community in Brazil. METHODOLOGY/MAIN FINDINGS:The serosurvey was conducted in a random sample of 450 residences in the Chapada district, located 100 kilometers from Feira de Santana. We administered questionnaires and tested 120 sera from Chapada district residents for CHIKV IgM- and IgG-specific antibodies. An individual with CHIKV infection was defined as any person with CHIKV IgM or IgG antibodies detected in the serum. One Hundred cases of Chikungunya were reported after prolonged rainfall, which reinforced the relationship between the rainfall index and CHIKV transmission. Eighteen months after the start of the outbreak, we identified a seroprevalence of 20% (95% CI, 15.4-35%). CHIKV IgG- and IgM-specific antibodies were detected in 22/120 (18.3%) and 6/120 (5.0%) individuals, respectively. Because of their strong dependence on the environment, the spatial distribution of pond-breeding amphibians can be greatly influenced by anthropogenic habitat alteration. In some agricultural landscapes in Brazil, the anuran Pseudopaludicola mystacalis appears to be highly influenced by land use. Because adult males and tadpoles of this species are usually found in marshy areas with cattle hoof prints, we hypothesized that P. mystacalis preferentially occupies aquatic habitats with marshy areas that are trampled by cattle. To test our hypothesis, we assessed whether the occurrence of P. mystacalis is associated with the presence of cattle and trampled marshy areas, and which environmental features best explain the spatial distribution and abundance of P. mystacalis. To do so, we sampled 38 aquatic habitats in an area intensely used for livestock in southeastern Brazil. We found that the presence of cattle and trampled marshy areas in aquatic habitats are positively associated to P. mystacalis occurrence. Additionally, the abundance of calling males is better predicted by variables of landscape and local habitat structure. Specifically, the size of trampled marshy areas and the proportion of herbaceous vegetation within the aquatic habitat are positively associated with abundance, while distance to nearest aquatic habitat are negatively associated with abundance of calling males. All three of these variables can be directly or indirectly linked to the presence of cattle or grazing management. Therefore, this work shows evidence that Pseudopaludicola mystacalis is positively influenced by grazing management with cattle, and draws attention to other unknown potential consequences of different land use to fresh water diversity.

In Madagascar, little information is available on drug residues in animal products. Official veterinary services have been recently informed about the misuse of human injectable contraceptives in pig farms. Farmers and local animal health workers have been suspected of treating pigs for growth promotion and of using human progestins, especially medroxyprogesterone acetate (MPA), as a chemical alternative method for castration of sows that are then fattened before culling. Indeed, MPA may arrest the onset of farrowing and induce post-lactational anestrus in sows. Because the use of synthetic hormones in pig husbandry is consid­ered a fraud by the Malagasy legislation, an exploratory study was carried out to confirm these suspicions and investigate the main molecules concerned. We investigated 80 pigs slaughtered in seven Malagasy abattoirs and raised in eight (out of 22) Malagasy regions (i) to confirm the contamination of carcasses by anabolic hormones by liquid chromatography coupled to mass spectrometry (LC-MS/MS), (ii) to identify the concerned molecules, and (iii) to explore consumers’ exposure to hormone residues. Only adult sows were sampled con­sidering that chemical castration with progestogens was the most common hypothesis of misuse of synthetic hormones in the field. The screening of the 80 kidney-fat samples did not reveal residues of progestogens other than MPA. MPA-positive samples were detected in 10 out of 15 districts (66.7%), and in all eight surveyed regions except one (87.5%), Diana Region in Northern Madagascar. Our results revealed heavy contamination of pork products by synthetic hormone residues; its prevalence was high considering that such residues are not supposed to occur accord­ing to the Malagasy legislation. Investigations by Malagasy vet­erinary services revealed that farmers and animal health workers purchased syringes of progestogens (Confiance™, Pfizer). These low-price progestins are easily available in private local dispen­saries and basic health centers (public sector). Without any control, farmers can easily administer MPA to pigs. It has thus become urgent to launch public-awareness campaigns and improve control within the sectors of animal production and public health over the entire country.

Antibiotics are used in animal feeds as growth promoters or for disease prevention and treatment. Drugs and their metabolites accumulate into body cells and constitute the so-called drug residues. Residual antimicrobials in food are an increasing risk for human health. Antibiotic residues in meat and other foods are suspected to be responsible for drug allergies and emergence of antimicrobial-resistant bacteria. In Madagascar, bacterial resistance against several antibiotics has been regularly observed in pathogens isolated from humans and pigs. Suspicion of drug misuse in farms has been frequently recorded by animal health professionals but no guidelines are available on the good usage of antibiotics in livestock. Since scarce information is available on antimicrobial residues in ani­mal products in Madagascar, this study aimed at determining the prevalence of pork meat contaminated by such residues and sold in Antananarivo markets. A total of 967 meat samples (diaphragm muscle) were collected during 2010 and 2011 in the four main abattoirs of Antananarivo, and in three additional regional abattoirs, i.e. Antsirabe, Mahitsy (Central Madagascar) and Tsiroamandidy (Western Madagascar). Analysis for antimicrobial detection in meat was performed with the commercial Premi®-test kit (DSM, Urmond, Netherlands) at the National Veterinary Diagnostic Laboratory in Antananarivo. Premi®-test is a broad spectrum microbial screening test for the detection of antibiotic and sulfonamide residues in meat or meat products at, or below maximum residue limit (MRL) levels. MRL is the maximum concentration of residues following administration of a veterinary medicine which is legally permit­ted or acceptable in food under European Union legislation. The Premi®-test kit contains a standardized number of spores of Bacillus stearothermophilus, very sensitive to many antibiotics and sulfa compounds, in agar medium with selected nutrients together with an indicator. Premi®-test is based on the inhibi­tion of the growth of B. stearothermophilus. When meat juice is added to the Premi®-test ampoule and heated at 64°C, spores germinate. The germinated spores multiply and form an acid when no inhibitory substances are present, leading to an indica­tor color change from purple to yellow. When antimicrobial mol­ecules are present above the detection level, no growth occurs and the color remains purple. Table I presents the results of the percentage of positive samples in the various regions of Madagascar. On average 37.2% sam­ples were contaminated by antimicrobial residues. A significant increase from 32 to 39% was observed between 2010 and 2011, respectively. No significant differences were found between samples according to sex, breed or age class in individual ani­mals. No differences between pig farm origins were found either (Figure 1). However, Amoron’i Mania Region, and particularly suburban Ambositra, was the most contaminated area in 2010 (67%; n = 9) and Melaky region (Western Madagascar) in 2011. Pork meat samples originating from the same production area were less contaminated by drug residues when the animals were slaughtered in urban abattoirs compared to provincial abat­toirs. In this first step toward a national surveillance system, we confirm that drug residues in animal products are a serious public health concern for Madagascar.

Periodontitis is an inflammatory condition that affects the supporting tissues surrounding teeth. The occurrence of periodontitis is associated with shifts in the structure of the communities that inhabit the gingival sulcus. Although great inter-subject variability in the subgingival microbiome has been observed in subjects with periodontitis, it is unclear whether distinct community types exist and if differences in microbial signatures correlate with host characteristics or with the variable clinical presentations of periodontitis. Therefore, in this study we explored the existence of different community types in periodontitis and their relationship with host demographic, medical and disease-related clinical characteristics. Clustering analyses of microbial abundance profiles suggested two types of communities (A and B) existed in the 34 subjects with periodontitis evaluated. Type B communities harbored greater proportions of certain periodontitis-associated taxa, including species historically associated with the disease, such as Porphyromonas gingivalis, Tannerella forsythia and Treponema denticola, and taxa recently linked to periodontitis. In contrast, subjects with type A communities had increased proportions of different periodontitis-associated species, and were also enriched for health-associated species and core taxa (those equally prevalent in health and periodontitis). Periodontitis subgingival clusters were not associated with demographic, medical or disease-specific clinical parameters other than periodontitis extent (proportion of sites affected), which positively correlated with the total proportion of cluster B signature taxa. In conclusion, two types of microbial communities were detected in subjects with periodontitis. Host demographics and underlying medical conditions did not correlate with these profiles, which instead appeared to be related to periodontitis extent, with type B communities present in more widespread disease cases. The two identified periodontitis profiles may represent distinct dysbiotic processes potentially requiring community-tailored therapeutic interventions.

This study aimed to evaluate intake, digestibility, and performance in sheep fed increasing levels of annatto byproduct (AB). A total of 32 male sheep without defined breed were used. Their initial weight was 23.17±1.45 kg. The animals were housed in individual pens and offered feed twice a day. Nutrient intake was quantified by the difference between the fractions present in the offered feed and the remains. The apparent digestibility was estimated with the aid of the external marker LIPE®. Weight gain was measured by the difference between the initial and final weight of the animals. The intake of dry matter (DM; g/day; g/kg BW; g/kg0.75), organic matter (g/day), crude protein (CP; g/day), and neutral detergent fibre (g/day) was not affected (P>0.05) by addition of AB. The intake of ether extract (EE; g/day) and non-fibre carbohydrates (g/day) was influenced by the inclusion of AB. The apparent digestibility of DM, organic matter, CP, and EE was not affected (P>0.05) by the addition of AB. The AB can be included in the diet at levels up 300 g/kg of total DM without affecting consumption, digestibility, and weight gain in sheep.

Antisense-based molecules targeting HIV-1 RNA have the potential to be used as part of gene or drug therapy to treat HIV-1 infection. In this study, HIV-1 RNA was screened to identify more conserved and accessible target sites for ribozymes based on the hepatitis delta virus motif. Using a quantitative screen for effects on HIV-1 production, we identified a ribozyme targeting a highly conserved site in the Gag coding sequence with improved inhibitory potential compared to our previously described candidates targeting the overlapping Tat/Rev coding sequence. We also demonstrate that this target site is highly accessible to short hairpin directed RNA interference, suggesting that it may be available for the binding of antisense RNAs with different modes of action. We provide evidence that this target site is structurally conserved in diverse viral strains and that it is sufficiently different from the human transcriptome to limit off-target effects from antisense therapies. We also show that the modified hepatitis delta virus ribozyme is more sensitive to a mismatch in its target site compared to the short hairpin RNA. Overall, our results validate the potential of a new target site in HIV-1 RNA to be used for the development of antisense therapies.

With the objective of studying the role of wild fauna in the epidemiology of fasciolosis disease, a definitive wild-host inventory was carried out in a french farm where infected domestic hosts (cows) cohabit with wild potential ones. Liver flukes, faecal eggs and antibodies were looked for in lagomorphs (Oryctolagus cuniculus) and rodents (Myocastor coypus, Ondatra zybethicus, Rattus norvegicus, Arvicola sapidus and micromammal species) trapped in the study area. Presence of Fasciola hepatica was detected in two species: O. cuniculus and M. coypus. Infection rates were respectively 34 % (42/124) and 55 % (106/193). Liver flukes were found in 78 M. coypus (n = 192) and 11 O. cuniculus (n = 35). No other species was infected by F. hepatica. The number of animals shedding fluke eggs was higher in M. coypus (49 out of 1 27 sampled; 38.6 %) than in O. cuniculus (two out of 17 sampled; 11.7 %).The results indicate that M. coypus may play a role in the maintenance and the dissemination of F. hepatica in various environments and open a discussion on the role of other natural wild hosts.

For a long time nucleic acid-based approaches directed towards controlling the propagation of Hepatitis C Virus (HCV) have been considered to possess high potential. Towards this end, ribozymes (i.e. RNA enzymes) that specifically recognize and subsequently catalyze the cleavage of their RNA substrate present an attractive molecular tool. Here, the unique properties of a new generation of ribozymes are taken advantage of in order to develop an efficient and durable ribozyme-based technology with which to target HCV (+) RNA strands. These ribozymes resulted from the coupling of a specific on/off adaptor (SOFA) to the ribozyme domain derived from the Hepatitis Delta Virus (HDV). The former switches cleavage activity "on" solely in the presence of the desired RNA substrate, while the latter was the first catalytic RNA reported to function naturally in human cells, specifically in hepatocytes. In order to maximize the chances for success, a step-by-step approach was used for both the design and the selection of the ribozymes. This approach included the use of both bioinformatics and biochemical methods for the identification of the sites possessing the greatest potential for targeting, and the subsequent in vitro testing of the cleavage activities of the corresponding SOFA-HDV ribozymes. These efforts led to a significant improvement in the ribozymes' designs. The ability of the resulting SOFA-HDV ribozymes to inhibit HCV replication was further examined using a luciferase-based replicon. Although some of the ribozymes exhibited high levels of cleavage activity in vitro, none appears to be a potential long term inhibitor in cellulo. Analysis of recent discoveries in the cellular biology of HCV might explain this failure, as well as provide some ideas on the potential limits of using nucleic acid-based drugs to control the propagation of HCV. Finally, the above conclusions received support from experiments performed using a collection of SOFA-HDV ribozymes directed against HCV (-) strands.

SUMMARY: Seismic traveltime tomography is a geophysical imaging method to infer the 3-D interior structure of the solid Earth. Most commonly formulated as a linearized inverse problem, it maps differences between observed and expected wave traveltimes to interior regions where waves propagate faster or slower than the expected average. The Earth's interior is typically parametrized by a single kind of localized basis function. Here we present an alternative approach that uses matching pursuits on large dictionaries of basis functions.Within the past decade the (Learning) Inverse Problem Matching Pursuits [(L)IPMPs] have been developed. They combine global and local trial functions. An approximation is built in a so-called best basis, chosen iteratively from an intentionally overcomplete set or dictionary. In each iteration, the choice for the next best basis element reduces the Tikhonov–Phillips functional. This is in contrast to classical methods that use either global or local basis functions. The LIPMPs have proven their applicability in inverse problems like the downward continuation of the gravitational potential as well as the MEG-/EEG-problem from medical imaging. Here, we remodel the Learning Regularized Functional Matching Pursuit (LRFMP), which is one of the LIPMPs, for traveltime tomography in a ray theoretical setting. In particular, we introduce the operator, some possible trial functions and the regularization.
