3. Fast and pure phase-shifting off-axis holographic microscopy with a digital micromirror device

Abstract

We present a phase-shifting digital holographic microscopy technique, where a digital micromirror device enables to perform a precise phase-only shift of the reference wave. By coupling the beam into a monomode fiber, we obtain a laser mode with a constant phase shift, equally acting on all pixels of the hologram. This method has the advantage of being relatively simple and compatible with high frame rate cameras, which makes it of great interest for the observation of fast phenomena. We demonstrate the validity of the technique in an off-axis configuration by imaging living paramecia caudata.

Gleichzeitig entstehen auf den frei werdenden Bergbauflächen neue Nutzungsmöglichkeiten in Form von Stadtquartieren und Gewerbeflächen. An diesen Standorten besteht die Chance, die geothermische Energie des Grubenwassers zur Beheizung und zur Kühlung von Gebäuden zu nutzen.

Abstract

Ultra-low district heating (ULTDH) networks are operated at temperatures below 30 °C. However, the temperatures supplied are still too low for direct heating of the consumers. Heat pumps use the heating network as a source and can supply the consumers with space heating and domestic hot water as needed. Although individual manufacturers provide performance figures for the source temperature of 20 °C, these are usually derived from extrapolation from standard conditions with source temperatures of 0 °C and 10 °C. Up to now practical measurements are missing how efficiently heat pumps work in an ULTDH network environment and how the network temperature spread affects the performance. In this paper test rigs are used to investigate heat pumps under the conditions in ULTDH networks at a network supply temperature of 20 °C. It is shown that heat pumps can operate up to twice as efficient as with a geothermal probe. Furthermore, the network temperature spread has a significant influence on the performance of the heat pump. Heat pumps operate significantly better with a low source temperature spread.

Abstract

This article investigates possibilities for including prosumer structures in heating networks. Therefore, the performance of three different heating system scenarios for a new housing district is investigated. Low (60 °C) and ultra-low (20 °C) network temperatures are investigated. The case study includes about 100 different single-family and multi-family houses. Two scenarios contain networks with temperatures of 20 and 60 °C. The third scenario is extended with solar collectors and a supermarket as decentralised prosumers who supply heat to the network in times of surplus. The network can simulate bidirectional flows. Based on the principles of graph theory, a combined hydraulic and thermal model is developed and used for this study. The results show a heat loss reduction in ultra-low temperature networks due to lower network working temperatures (20 °C) of about 80% compared to a low-temperature network. The implementation of prosumers considerably reduces the demand from a central heat source. In this study, 20% of the total heating demand can be covered by excess heat of the prosumer supply.

Abstract

International audience; We show that compressive sensing (CS) calculations are very ecient to reconstruct in 3D sparse objects whose 2D hologram has been recorded by digital holographic microscopy. The method is well adapted to image small scattering objects moving within a larger motionless object. This situation corresponds to red blood cells (RBCs) circulating in the vascular system of a zebrash (Danio rerio) larva. RBCs positions are imaged in 3D from a single hologram, while the RBCs trajectories, i.e. the perfused blood vessels, are imaged from a sequence of holograms. With respect to previous work (Donnarumma et al., Opt. express, 24, 26887, 2016), we get a gain of ∼ 500 in calculation speed.

Abstract

International audience; Open channels are peculiar incidents modes of the optical field that are transmitted by a highly diffusing medium with high efficiency (100 % in the ideal case). The existence of these modes has been predicted by Dorokhov et al. [1] more than 25 years ago. Although many experiments were made to prepare these modes [2,3], the maxima of transmission obtained in experiments, are ever much lower than 100 %. To test the theory, we have considered here another Dorokhov's prediction: the average transmission of the modes that sustain the transmitted field must be TT=66%, where TT is defined by N2 TT= N1 T (where T is the averaged transmission of energy, and N1 and N2 the number of incident and transmitted modes i.e. the size of the incident and transmitted mode basis). International audience; From the session Tomography and Computational Imaging (JSII_1) (invited) 4D Digital Holographic Microscopy (4D-DHM) is a new technique capable of performing 3D imaging of moving objects. 3D reconstruction is made from a single hologram, but is restricted to sparse objects that occupy only small part of the volume. For a sequence of holograms, 4D-DHM therefore performs 4D (x; y; z and t) imaging. 4D-DHM is well suited to image micro circulation, and has been validated imaging red blood cells circulating in a zebrafish embryo in vivo [1-3]. The displayed 2D images correspond to the projections along z of the 3D images rotated along the y direction by-30° (b), 0° (c) and +30° (d). Bar is 50 m, The principle of the 4D DHM is illustrated by Fig. 1 (a). The setup is an upright microscope, which has been modified to perform Digital Holographic Microscopy (DHM) in transmission. The white light illumination is replaced by a laser, which is split in two beams of different directions. To perform holographic detection a reference arm (ELO field) is added on the detection side. The field E scattered by the object interferes with ELO , and a camera records sequences of frames. These frames are used to calculate the holograms, and to reconstruct in 3D the field scattered by the sample. By a proper analysis of this information, 3D images of the blood vessels, and 4D images of the moving red blood cells (RBCs) can be obtained see Fig,1 (b) to (d). To get these images, we have proceeded by the following way [2,3]. First, we calculate the holograms which differences of frames, and select by the way the signal from the moving components of the object (i.e.) the RBCs signal. We have then extract from the data, the holograms corresponding to the two illumination. For each RBC, and each illumination, the field that can be reconstructed by holography as a narrow cone of light (narrow because RCBs are much larger than the wavelength), which is angularly tilted like the illumination. The location of RBCs is then obtained by considering the intersection of the two cones. To prevent the signals of the different RBCs to mix together, the positions of the RBCs that give the largest signals are calculated first. The contributions of these RBCs are then removed from the hologram, and the positions of the following RBCs are calculated from the hologram thus cleaned. The reconstruction by cleaning is highly non-linear (unlike conventional holographic reconstructions), since it is the algorithm that decides to place a RBC in a plane and to remove the field diffused by this RBC from the other planes. In order to converge successively it requires to image sparses objects, that occupy only small part of the holographic reconstructed volume. 4D-DHM is thus capable of performing 3D reconstruction is made from a single hologram, but is restricted to sparse objects that occupy only small part of the volume.

Abstract

Ultrasound-modulated optical tomography (UOT) is a technique that images optical contrast deep inside scattering media. Heterodyne holography is a promising tool able to detect the UOT tagged photons with high efficiency. In this work, we describe theoretically the detection of the tagged photon in heterodyne holography based UOT, show how to filter the untagged photon, and discuss the effect of shot noise. The discussion considers also speckle decorrelation. International audience; A set of 55 clarinet reeds is observed by holography, collecting 2 series of measurements made under 2 different moisture contents, from which the resonance frequencies of the 15 first modes are deduced. A statistical analysis of the results reveals good correlations, but also significant differences between both series. Within a given series, flexural modes are not strongly correlated. A Principal Component Analysis (PCA) shows that the measurements of each series can be described with 3 factors capturing more than $90\%$ of the variance: the first is linked with transverse modes, the second with flexural modes of high order and the third with the first flexural mode. A forth factor is necessary to take into account the individual sensitivity to moisture content. Numerical 3D simulations are conducted by Finite Element Method, based on a given reed shape and an orthotropic model. A sensitivity analysis revels that, besides the density, the theoretical frequencies depend mainly on 2 parameters: $E_L$ and $G_{LT}$. An approximate analytical formula is proposed to calculate the resonance frequencies as a function of these 2 parameters. The discrepancy between the observed frequencies and those calculated with the analytical formula suggests that the elastic moduli of the measured reeds are frequency dependent. A viscoelastic model is then developed, whose parameters are computed as a linear combination from 4 orthogonal components, using a standard least squares fitting procedure and leading to an objective characterization of the material properties of the cane \textit{Arundo donax}.

International audience; We study the dynamics of individual polystyrene ellipsoids of different aspect ratios trapped at the air-water interface. Using particle tracking and in situ vertical scanning interferometry techniques we are able to measure translational drags and the protrusion in air of the ellipsoids. We report that translational drags on the ellipsoid are unexpectedly enhanced: despite the fact that a noticeable part of the ellipsoid is in air, drags are found larger than the bulk one in water.

Abstract

International audience; Ultrasound-modulated optical tomography (UOT) is a technique that combines light and ultrasound able to image deep inside scattering media. A pulsed version of technique involving holography is proposed and discussed. Ultrasound-modulated optical tomography (UOT) [1] also called acousto-optic imaging [2] is a technique that combines light and ultrasound able to image deep inside scattering media. The ultimate goal of the technique is breast cancer imaging. Heterodyne holography combined with UOT is a powerful tool to detect the UOT tagged photons. Previous experiments are made with phantom samples. Since the sample does not move, the phase of the field remains correlated. Holographic detection is then efficient and shot noise sensitivity is reached [3]. To perform imaging with breast, a new setup with light and ultrasound pulses is proposed. Numerical simulations, that fits with the results of previous work, are made to extrapolate the phantom results to breast imaging with the new setup. They show that breast tumor imaging is possible. Abstract

International audience; An holographic microscopy reconstruction method compatible with high numerical aperture microscope objective (MO) up to NA=1.4 is proposed. After off axis and reference field curvature corrections, and after selection of the +1 grating order holographic image, a phase mask that transforms the optical elements of the holographic setup into an afocal device is applied in the camera plane. The reconstruction is then made by the angular spectrum method. The field is first propagated in the image half space from the camera to the afocal image of the MO optimal plane (plane for which MO has been designed) by using a quadratic kernel. The field is then propagated from the MO optimal plane to the object with the exact kernel. Calibration of the reconstruction is made by imaging a calibrated object like an USAF resolution target for different positions along z. Once the calibration is done, the reconstruction can be made with an object located in any plane z. The reconstruction method has been validated experimentally with an USAF target imaged with a NA=1.4 microscope objective. Near-optimal resolution is obtained over an extended range (±50 µm) of z locations.

Abstract

International audience; The dynamics of colloidal particles at interfaces between two fluids plays a central role in microrheology, encapsulation, emulsification, biofilm formation, water remediation and the interface-driven assembly of materials. Common intuition corroborated by hydrodynamic theories, suggests that such dynamics is governed by a viscous force lower than that observed in the more viscous fluid. Here, we show experimentally that a particle straddling an air/water interface feels a large viscous drag that is unexpectedly larger than that measured in the bulk. We suggest that such a result arises from thermally activated fluctuations of the interface at the solid/air/liquid triple line and their coupling to the particle drag through the fluctuation–dissipation theorem. Our findings should inform approaches for improved control of the kinetically driven assembly of anisotropic particles with a large triple-line-length/particle-size ratio, and help to understand the formation and structure of such arrested materials.

Abstract

We propose an holographic microscopy reconstruction method, which propagates the hologram, in the object half space, in the vicinity of the object. The calibration yields reconstructions with an undistorted reconstruction grid i.e. with orthogonal $x$, $y$ and $z$ axis and constant pixels pitch. The method is validated with an USAF target imaged by a $\times$60 microscope objective, whose holograms are recorded and reconstructed for different USAF locations along the longitudinal axis: -75 to +75 $\mu$m. Since the reconstruction numerical phase mask, the reference phase curvature and MO form an afocal device, the reconstruction can be interpreted as occurring equivalently in the object or in image half space.

International audience; This chapter introduces the principles of digital holography, as well as the associated reconstruction methods. It presents the basic properties related to a digital hologram. In order to discuss the digital reconstruction of the object wave at the object plane, the chapter presents the basics of the scalar diffraction of light. Algorithms used to back-propagate the object field estimated at the sensor plane are based on this approach. The chapter discusses the use of the theoretical basics of wave propagation to numerically reconstruct the object wave at the object plane. It also discusses the different experimental configurations to record a digital hologram. The chapter provides basic experimental arrangements and gives few explications on how it works. Digital holographic interferometry exploits not only the amplitude of the object, but also its phase. Finally, the chapter describes the basic principle of digital holographic interferometry, and quantitative phase tomography.

Abstract

Due to the existence of open channels, light could theoretically be transmitted through disorderedmedia with 100% efficiency. However, because of the large number of channels to be controlledor measured, this has been never observed experimentally. To avoid this difficulty we considerobservables easier to handle experimentally. By measuring the correlations of the field transmittedthrough a disordered medium, we were able to count the number of channels involved in transmissionand to compare this number with theory. For a sample of transmission 1=25, we get a number ofmodes about 15 times smaller than for free space propagation. This figure is in good agreementwith theoretical prediction.

Abstract

International audience; We present a wide-field microscopy technique for the 3D mapping of optical intensity distributions using Brownian gold nanopar-ticles as local probes, which are localized by off-axis holography. Fast computation methods allow us to localize hundreds of particles per minute with accuracies as good as 3 × 3 × 10nm3 for immobilized particles. Factors limiting this accuracy are discussed and the possibilities of the technique are illustrated through the 3D optical mapping of an evanescent and a propagative wave. Our results pave the way for a new stochastic imaging technique, well adapted to subwavelength optical characterization in water-based systems.

Abstract

We describe a novel resonant cavity system which allows the bulk magneto-conductivity of single crystals of anisotropic metals to be measured at GHz frequencies. The cavity can be made to rotate in a static magnetic field, permitting detailed studies of the dependence on the magnetic field orientation of the high-frequency magnetoconductivity. In this paper, the apparatus is used to measure the Fermi-surface topology of two organic superconductors; the details revealed are inaccessible to conventional fermiological techniques such as the de Haas-van Alphen effect. In κ-(BEDT-TTF)2Cu(NCS)2, Fermi-surface traversal resonances (FTRs) are observed. The angle dependence of the FTRs shows that the quasi-one-dimensional (Q1D) Fermi sheets of this material possess two distinct corrugations, with corrugation axes making angles of 17.3° and -19.4° with the ka-axis. Such data form important input parameters for current models of superconductivity in the organics, which invoke spin-density-wave-like fluctuations caused by partial nesting of the Q1D Fermi sheets. In β''-(BEDT-TTF)2SF5CH2CF2SO3, cyclotron resonance is observed, along with its second and third harmonics. The detailed angle dependence of the intensities of the various cyclotron harmonics allows the elongation and orientation of the closed section of the Fermi surface to be deduced, and strongly suggests that the interplane transport is coherent in this material. The effective mass deduced from the cyclotron resonance measurements is greater than that determined from magnetic quantum oscillations, in agreement with recent theoretical predictions.

International audience; An original technique that combines digital holography, dual illumination of the sample and cleaning algorithm 3D reconstruction is proposed. It uses a standard transmission microscopy setup coupled with a digital holography detection. The technique is 4D, since it allows to determine, at each time step, the 3D locations (x, y, z) of many moving objects that scatter the dual illumination beam. The technique has been validated by imaging the microcirculation of blood in a fish larvae sample (the moving objects are thus red blood cells RBCs). Videos showing in 4D the moving RBCs superimposed with the perfused blood vessels are obtained.

Abstract

Electron-spin-resonance measurements at high frequencies have been made on the antiferromagnet ${\mathrm{FeI}}_{2}$ with strong uniaxial anisotropy. We have found signals whose g value is 16.5 together with the one with $g=7.0$ reported before [A. R. Fert et al., Solid State Commun. 26, 693 (1978)]. We interpret these electron-spin resonance signals as originating from single-ion four magnon and two magnon bound states, respectively. The energies for creating the magnon bound states are calculated using an Ising approximation. One of the four magnon bound states is assigned to the one due to the exchange interaction either the third nearest-neighbor one in the c plane ${(J}_{3})$ or the between planes one ${(J}_{0})$ and its value is determined to be ${J}_{3}{/k}_{\mathrm{B}}$ (or ${J}_{0}{/k}_{\mathrm{B}})=\ensuremath{-}0.45$ K.

We describe instrumentation for conducting high sensitivity millimeter-wave cavity perturbation measurements over a broad frequency range (40-200 GHz) and in the presence of strong magnetic fields (up to 33 tesla). A Millimeter-wave Vector Network Analyzer (MVNA) acts as a continuously tunable microwave source and phase sensitive detector (8-350 GHz), enabling simultaneous measurements of the complex cavity parameters (resonance frequency and Q-value) at a rapid repetition rate (approx. 10 kHz). We discuss the principal of operation of the MVNA and the construction of a probe for coupling the MVNA to various cylindrical resonator configurations which can easily be inserted into a high field magnet cryostat. Abstract

International audience; We report theoretical predictions and measurements of the capillary force acting on a spherical colloid smaller than the capillary length that is placed on a curved uid interface of arbitrary shape. By coupling direct imaging and interferometry, we are able to measure the in situ colloid contact angle and to correlate its position with respect to the interface curvature. Extremely tiny capillary forces down to femto-Newton can be measured with this method. Measurements agree well with a theory relating the capillary force to the gradient of Gaussian curvature and to the mean curvature of the interface prior to colloidal deposition. Numerical calculations corroborate these results.

Abstract

International audience; Acousto-Optical Coherence Tomography (AOCT) is a variant of Acousto Optic Imaging (also called Ultrasound modulated Optical Tomography) that makes possible to get resolution along the ultrasound propagation axis $z$. We present here new AOCT experimental results, and we study how the $z$ resolution depends on time step between phase jumps $T_\phi$, or on the correlation length $\Delta z$. By working at low resolution, we perform a quantitative comparison of the $z$ measurements with the theoretical Point Spread Function (PSF). We present also images recorded with different $z$ resolution, and we qualitatively show how the image quality varies with $T_\phi$, or $\Delta z$.

Abstract

We report a demonstration of phase-resolved vibrometry, in which out-of-plane sinusoidal motion is assessed by heterodyne holography. In heterodyne holography, the beam in the reference channel is an optical local oscillator (LO). It is frequency-shifted with respect to the illumination beam to enable frequency conversion within the sensor bandwidth. The proposed scheme introduces a strobe LO, where the reference beam is frequency-shifted and modulated in amplitude, to alleviate the issue of phase retrieval. The strobe LO is both tuned around the first optical modulation side band at the vibration frequency, and modulated in amplitude to freeze selected mechanical vibration states sequentially. The phase map of the vibration can then be derived from the demodulation of successive vibration states.

Abstract

We propose here to combine sideband holography with stroboscopic illumination synchronized with the vibration of an object. By sweeping the optical frequency of the reference beam such a way the holographic detection is tuned on the successive sideband harmonic ranks, we are able to image the instantaneous velocities of the object. Since the stroboscopic illumination is made with an electronic device, the method is compatible with fast (up to several MHz) vibration motions. The method is demonstrated with a vibrating clarinet reed excited sinusoidally at 2 kHz, and a stroboscopic illumination with cyclic ratio 0.15. Harmonic rank up to n = $\pm$100 are detected, and a movie of the instantaneous velocities is reported.

International audience; By using the dark field holographic microscopy technique described in Opt. Express, 19 pp 26044-26055 (2011) we have track gold particles in brownian motion in living cells. First results are presented. The particles remain confined to a region a few microns in size. We interpret this as a cage effect related to the cell structure.

Abstract

International audience; We have performed off axis heterodyne holography with very weak illumination by recording holograms of the object with and without object illumination in the same acquisition run. We have experimentally studied , how the reconstructed image signal (with illumination) and noise background (without) scale with the holographic acquisition and reconstruction parameters that are the number of frames, and the number of pixels of the reconstruction spatial filter. The first parameter is related to the frequency bandwidth of detection in time, the second one to the bandwidth in space. The signal to background ratio varies roughly like the inverse of the bandwidth in time and space. We have also compared the noise background with the theoretical shot noise background calculated by Monte Carlo simulation. The experimental and Monte Carlo noise background agree very well together.

Abstract

BACKGROUND: We designed an excimer laser keratomileusis delivery system to increase the regularity of the refractive cut surface and allow greater precision in the level and shape of the ablated zone. METHODS: A parallel faced corneal disc was produced by microkeratectomy from six human eyes and surgical keratectomy in 12 beagle corneas. A 193-nanometer excimer laser that was used to project an oval beam onto the corneal disc was rotated on a flat surface to ensure overlapping of the ovally ablated areas between pulses. RESULTS: Electron microscopy of eye bank lenticules demonstrated a circular smooth regularly concave ablation zone. Histological examination of nine clear corneas confirmed thinning of the stroma without fibroblastic reaction and no epithelial hypertrophy. Mean preoperative corneal power of 43.15 ± 2.18 decreased postoperatively to 33.61 ±2.34. CONCLUSIONS: The new technique of excimer laser keratomileusis has the advantage of a cut surface smoother and the clear zone is devoid of the stepwise concavity and irregularity seen in diaphragm based photoablation delivery systems. Abstract

International audience; We present a new technique that combines off-axis Digital Holography and Dark Field Microscopy to track 100nm gold particles diffusing in water. We show that a single hologram is sufficient to localize several particles in a thick sample with a localization accuracy independent of the particle position. From our measurements we reconstruct the trajectories of the particles and derive their 3D diffusion coefficient. Our results pave the way for quantitative studies of the motion of single nanoparticle in complex media.

Abstract

Acousto-optical coherence tomography (AOCT) is a variant of acousto-optic imaging (also called ultrasonic modulation imaging) that makes it possible to get the z resolution with acoustic and optic continuous wave beams. We describe here theoretically the AOCT effect, and we show that the acousto-optic “tagged photons” remain coherent if they are generated within a specific z region of the sample. We quantify the z selectivity for both the “tagged photon” field and for the Lesaffre et al. [Opt. Express17, 18211 (2009)] photorefractive signal.

The expected signal and the chosen method of reconstruction are described. We also discuss about how to implement the numerical calculation to reach real-time 3D tracking.

International audience; In this paper, we explore the 3D structure of light scattering from dark-field illuminated live 3T3 cells in the presence of 40 nm gold nanomarkers. For this purpose, we use a high resolution holographic microscope combining the off-axis heterodyne geometry and the phase-shifting acquisition of the digital holograms. A comparative study of the 3D reconstructions of the scattered fields allows us to locate the gold markers which yield, contrarily to the cell structures, well defined bright scattering patterns that are not angularly titled and clearly located along the optical axis (z). This characterization is an unambiguous signature of the presence of gold biological nanomarkers, and validates the capability of digital holographic microscopy to discriminate them from background signals in live ce
