Paediatric heart transplant is an established treatment for end stage heart failure in children, however patients have to commit to lifelong medical surveillance and adhere to daily immunosuppressants to minimise the risk of rejection. Compliance with immunosuppressants can be burdensome with their toxic side effects and need for frequent blood monitoring especially in children. Though the incidence of early rejection episodes has significantly improved overtime, the long-term allograft health and survival is determined by Cardiac Allograft Vasculopathy (CAV) which affects a vast number of post-transplant patients. Once CAV has set in, there is no medical or surgical treatment to reverse it and graft survival is significantly compromised across all age groups. Current treatment strategies include novel immunosuppressant agents and drugs to lower blood lipid levels to address the underlying immunological pathophysiology and to manage traditional cardiac risk factors. Translational researchers are seeking novel immunological approaches that can lead to permanent acceptance of the allograft such as using regulatory T cell (Tregs) immunotherapy. Clinical trials in the setting of graft versus host disease, autoimmunity and kidney and liver transplantation using Tregs have shown the feasibility and safety of this strategy. This review will summarise current knowledge of the latest clinical therapies for CAV and pre-clinical evidence in support of Treg therapy for CAV. We will also discuss the different Treg sources and the considerations of translating this into a feasible immunotherapy in clinical practice in the paediatric population.

Abstract The visualization of dynamic graphs is a challenging task owing to the various properties of the underlying relational data and the additional time-varying property. For sparse and small graphs, the most efficient approach to such visualization is node-link diagrams, whereas for dense graphs with attached data, adjacency matrices might be the better choice. Because graphs can contain both properties, being globally sparse and locally dense, a combination of several visual metaphors as well as static and dynamic visualizations is beneficial. In this paper, a visually and algorithmically scalable approach that provides views and perspectives on graphs as interactively linked node-link and adjacency matrix visualizations is described. As the novelty of this technique, insights such as clusters or anomalies from one or several combined views can be used to influence the layout or reordering of the other views. Moreover, the importance of nodes and node groups can be detected, computed, and visualized by considering several layout and reordering properties in combination as well as different edge properties for the same set of nodes. As an additional feature set, an automatic identification of groups, clusters, and outliers is provided over time, and based on the visual outcome of the node-link and matrix visualizations, the repertoire of the supported layout and matrix reordering techniques is extended, and more interaction techniques are provided when considering the dynamics of the graph data. Finally, a small user experiment was conducted to investigate the usability of the proposed approach. The usefulness of the proposed tool is illustrated by applying it to a graph dataset, such as e co-authorships, co-citations, and a Comprehensible Perl Archive Network distribution.

While graph drawing focuses more on the aesthetic representation of node-link diagrams, graph visualization takes into account other visual metaphors making them useful for graph exploration tasks in information visualization and visual analytics. Although there are aesthetic graph drawing criteria that describe how a graph should be presented to make it faster and more reliably explorable, many controlled and uncontrolled empirical user studies flourished over the past years. The goal of them is to uncover how well the human user performs graph-specific tasks, in many cases compared to previously designed graph visualizations. Due to the fact that many parameters in a graph dataset as well as the visual representation of them might be varied and many user studies have been conducted in this space, a state-of-the-art survey is needed to understand evaluation results and findings to inform the future design, research, and application of graph visualizations. In this article, we classify the present literature on the topmost level into graph interpretation, graph memorability, and graph creation where the users with their tasks stand in focus of the evaluation, not the computational aspects. As another outcome of this work, we identify the white spots in this field and sketch ideas for future research directions.

Algal blooms consisting of potentially toxic cyanobacteria are a growing source water management challenge faced by water utilities globally. Commercially available sonication devices are designed to mitigate this challenge by targeting cyanobacteria-specific cellular features and aim to inhibit cyanobacterial growth within water bodies. There is limited available literature evaluating this technology; therefore, a sonication trial was conducted in a drinking water reservoir within regional Victoria, Australia across an 18-month period using one device. The trial reservoir, referred to as Reservoir C, is the final reservoir in a local network of reservoirs managed by a regional water utility. Sonicator efficacy was evaluated through qualitative and quantitative analysis of algal and cyanobacterial trends within Reservoir C and surrounding reservoirs using field data collected across three years preceding the trial and during the 18-month duration of the trial. Qualitative assessment revealed a slight increase in eukaryotic algal growth within Reservoir C following device installation, which is likely due to local environmental factors such as rainfall-driven nutrient influx. Post-sonication quantities of cyanobacteria remained relatively consistent, which may indicate that the device was able to counteract favorable phytoplankton growth conditions. Qualitative assessments also revealed minimal prevalence variations of the dominant cyanobacterial species within the reservoir following trial initiation. Since the dominant species were potential toxin producers, there is no strong evidence that sonication altered Reservoir C’s water risk profiles during this trial. Statistical analysis of samples collected within the reservoir and from the intake pipe to the associated treatment plant supported qualitative observations and revealed a significant elevation in eukaryotic algal cell counts during bloom and non-bloom periods post-installation. Corresponding cyanobacteria biovolumes and cell counts revealed that no significant changes occurred, excluding a significant decrease in bloom season cell counts measured within the treatment plant intake pipe and a significant increase in non-bloom season biovolumes and cell counts as measured within the reservoir. One technical disruption occurred during the trial; however, this had no notable impacts on cyanobacterial prevalence. Acknowledging the limitations of the experimental conditions, data and observations from this trial indicate there is no strong evidence that sonication significantly reduced cyanobacteria occurrence within Reservoir C.

Diastolic dysfunction is an important determinant for prognosis and survival in several paediatric heart diseases. We aimed to explore its possible impact on outcome in children with dilated cardiomyopathy. From 2006 to 2016, children less than 18 years old with dilated cardiomyopathy were retrospectively enrolled. Echocardiographic diastolic function parameters and child outcomes were analysed. Of 43 children aged 0.2 to 16.1 years old referred with dilated cardiomyopathy, 8 patients required cardiac transplant or mechanical assist devices (18%), 24 had persistently abnormal left ventricular function and/or dilatation (56%) and 11 patients recovered (26%). There was no significant difference in mitral velocities on Tissue Doppler Imaging, mitral valve inflow velocities, isovolumic relaxation time, left atrial area z-score and mitral lateral E/e’ ratios between patients with recovery and patients with disease progression or persistently abnormal ventricular function and/or dilation. This is the first study on childhood dilated cardiomyopathy to address individual echocardiographic diastolic function parameters and their association to recovery. In this study, echocardiographic parameters for diastolic function did not predict recovery.

There is a growing interest in eye tracking technologies applied to support traditional visualization techniques like diagrams, charts, maps, or plots, either static, animated, or interactive ones. More complex data analyses are required to derive knowledge and meaning from the data. Eye tracking systems serve that purpose in combination with biological and computer vision, cognition, perception, visualization, human-computer-interaction, as well as usability and user experience research. The 10 articles collected in this thematic special issue provide interesting examples how sophisticated methods of data analysis and representation enable researchers to discover and describe fundamental spatio-temporal regularities in the data. The human visual system, supported by appropriate visualization tools, enables the human operator to solve complex tasks, like understanding and interpreting three-dimensional medical images, controlling air traffic by radar displays, supporting instrument flight tasks, or interacting with virtual realities. The development and application of new visualization techniques is of major importance for future technological progress.

We present an algorithmic and visual grouping of participants and eye-tracking metrics derived from recorded eye-tracking data. Our method utilizes two well-established visualization concepts. First, parallel coordinates are used to provide an overview of the used metrics, their interactions, and similarities, which helps select suitable metrics that describe characteristics of the eye-tracking data. Furthermore, parallel coordinates plots enable an analyst to test the effects of creating a combination of a subset of metrics resulting in a newly derived eye-tracking metric. Second, a similarity matrix visualization is used to visually represent the affine combination of metrics utilizing an algorithmic grouping of subjects that leads to distinct visual groups of similar behavior. To keep the diagrams of the matrix visualization simple and understandable, we visually encode our eye- tracking data into the cells of a similarity matrix of participants. The algorithmic grouping is performed with a clustering based on the affine combination of metrics, which is also the basis for the similarity value computation of the similarity matrix. To illustrate the usefulness of our visualization, we applied it to an eye-tracking data set involving the reading behavior of metro maps of up to 40 participants. Finally, we discuss limitations and scalability issues of the approach focusing on visual and perceptual issues.
