Abstract Gestational hypertension (PIH), especially pre-eclampsia (PE), is a common complication of pregnancy. This condition poses significant risks to the health of both the mother and the fetus. Emerging evidence suggests that epigenetic modifications, particularly DNA methylation, may play a role in initiating the earliest pathophysiology of PIH. This article describes the relationship between DNA methylation and placental trophoblast function, genes associated with the placental microenvironment, the placental vascular system, and maternal blood and vascular function, abnormalities of umbilical cord blood and vascular function in the onset and progression of PIH, as well as changes in DNA methylation in the progeny of PIH, in terms of maternal, fetal, and offspring. We also explore the latest research on DNA methylation-based early detection, diagnosis and potential therapeutic strategies for PIH. This will enable the field of DNA methylation research to continue to enhance our understanding of the epigenetic regulation of PIH genes and identify potential therapeutic targets.

Mentougou District, the core region of Xishan in Beijing, is located at the junction of Yanshan and Taihang mountains, where mountainous areas account for 98.5%. Over the long geological history, it has remained rich geoheritages. The lack of systematic analysis on the distribution characteristics and evolution of geoheritages constraint their protection and utilization of geoheritages in this area. This study has systematically reviewed the types of geoheritages in this area through remote sensing, ground investigation, and rock and mineral identification. The results show that: (1) The rich geoheritages in this area are diverse; 100 types of geoheritages have been identified, including geological profiles, geological structures, paleontological fossils, and landscape geology. (2) The distribution of geological profile types such as the Qingbaikou Series and Malan loess is closely related to the exposure area of the Mesoproterozoic-Quaternary strata. Folds and faults are mainly distributed in the middle and high mountain areas from Miaofengshan Town to Zhaitang Town; the acidic rock mass of Yanshan period are mainly distributed along large faults; loess terraces, river terraces, gorges, and water landscape are mainly distributed along the main channel of the Yongdinghe River and its major tributaries. (3) Multiple marine intrusions in this area created lots of stratigraphic profile relics; the Qin Yu Movement, Jixian Movement, and Yanshan Movement led to many structural features, faults, and folds, which dominated the formation of multiple mineral deposits. This study can provide basic information for further in-depth research on geoheritages and their protection and utilization in this area.

Abstract The adverse use of alcohol is a serious global public health problem. Maternal alcohol consumption during pregnancy usually causes prenatal alcohol exposure (PAE) in the developing fetus, leading to a spectrum of disorders known as fetal alcohol spectrum disorders (FASD) and even fetal alcohol syndrome (FAS) throughout the lifelong sufferers. The prevalence of FASD is approximately 7.7 per 1,000 worldwide, and is even higher in developed regions. Generally, Ethanol in alcoholic beverages can impair embryonic neurological development through multiple pathways leading to FASD. Among them, the leading mechanism of FASDs is attributed to ethanol-induced neuroinflammatory damage to the central nervous system (CNS). Although the underlying molecular mechanisms remain unclear, the remaining multiple pathological mechanisms is likely due to the neurotoxic damage of ethanol and the resultant neuronal loss. Regardless of the molecular pathway, the ultimate outcome of the developing CNS exposed to ethanol is almost always the destruction and apoptosis of neurons, which leads to the reduction of neurons and further the development of FASD. In this review, we systematically summarize the current research progress on the pathogenesis of FASD, which hopefully provides new insights into differential early diagnosis, treatment and prevention for patents with FASD.

Abstract Background Postoperative delirium (POD) is more prevalent among elderly patients with type 2 diabetes mellitus (T2DM). Insulin resistance (IR) can be assessed using the triglyceride-glucose (TyG) index, a novel biomarker. This study aims to investigate the predictive potential of the TyG index for POD in elderly patients with T2DM. Materials and methods Elderly patients (≥ 65) with T2DM who underwent non-neurosurgery and non-cardiac surgery were enrolled. Univariate and multivariate logistic regression analyses were conducted to assess the association between the TyG index and POD. Additionally, subgroup analyses were performed to compare the sex-specific differences in the predictive ability of the TyG index for POD. Results A total of 4566 patients were included in this retrospective cohort. The receiver operating characteristic (ROC) curve analysis determined the optimal cut-off value for the TyG index to be 8.678. In the univariate model, a TyG index > 8.678 exhibited an odds ratio (OR) of 1.668 (95% CI: 1.210–2.324, P = 0.002) for predicting POD. Abstract Background Lymphoma is the most common secondary cause of hemophagocytic lymphohistiocytosis (HLH) in adults. Lymphoma‐associated HLH (LA‐HLH) in the elderly population is not rare, however, little has been reported regarding clinicopathological characteristics, prognostic factors, and outcomes of LA‐HLH in the elderly population. Methods We retrospectively analyzed a multicenter cohort of elderly patients with LA‐HLH. Clinicopathological features and treatment information were collected. The impacts of baseline characteristics and treatments on survival outcomes were analyzed. Results A total of 173 elderly patients with LA‐HLH were included. Compared with young patients, elderly patients showed different clinical and laboratory features. Regarding lymphoma subtypes, B‐cell lymphoma was more common in elderly patients (elderly 61.3% vs. young 32.3%, p 1407 U/L), and a higher creatinine level (>96.8 μmol/L) were independent predictors of decreased overall survival and 60‐day survival. A prognostic index was established and demonstrated to be robust in predicting the overall survival and 60‐day survival of elderly patients with LA‐HLH. Conclusions LA‐HLH in elderly patients displayed heterogeneous clinicopathological features and survival outcomes. Treatments need to be optimized to improve the outcomes of elderly patients with LA‐HLH.

Summary: The Par3 polarity protein is critical for subcellular compartmentalization in different developmental processes. Variants of PARD3, encoding PAR3, are associated with intelligence and neurodevelopmental disorders. However, the role of Par3 in glutamatergic synapse formation and cognitive functions in vivo remains unknown. Here, we show that forebrain-specific Par3 conditional knockout leads to increased long, thin dendritic spines in vivo. In addition, we observed a decrease in the amplitude of miniature excitatory postsynaptic currents. Surprisingly, loss of Par3 enhances hippocampal-dependent spatial learning and memory and repetitive behavior. Phosphoproteomic analysis revealed proteins regulating cytoskeletal dynamics are significantly dysregulated downstream of Par3. Mechanistically, we found Par3 deletion causes increased Rac1 activation and dysregulated microtubule dynamics through CAMSAP2. Together, our data reveal an unexpected role for Par3 as a molecular gatekeeper in regulating the pool of immature dendritic spines, a rate-limiting step of learning and memory, through modulating Rac1 activation and microtubule dynamics in vivo.

Abstract Peste des petits ruminants (PPR) is an acute, contact infectious disease caused by the small ruminant morbillivirus (SRMV), and its morbidity in goats and sheep can be up to 100% with significant mortality. Nanobody generated from camelid animals such as alpaca has attracted wide attention because of its unique advantages compared with conventional antibodies. The main objective of this study was to produce specific nanobodies against SRMV and identify its characteristics. To obtain the coding gene of SRMV-specific nanobodies, we first constructed an immune phage-displayed library from the VHH repertoire of alpaca that was immunized with SRMV-F and -H proteins. By using phage display technology, the target antigen-specific VHHs can be obtained after four consecutive rounds of biopanning. Results showed that the size of this VHH library was 2.26 × 1010 CFU/mL and the SRMV-F and -H specific phage particles were greatly enriched after four rounds of biopanning. The positive phage clones were selected and sequenced, and total of five independent different sequences of SRMV-specific nanobodies were identified. Subsequently, the DNA fragments of the five nanobodies were cloned into E. coli BL21(DE3), respectively, and three of them were successfully expressed and purified. Specificity and affinity towards inactivated SRMV of these purified nanobodies were then evaluated using the ELISA method. Results demonstrated that NbSRMV-1-1, NbSRMV-2-10, and NbSRMV-1-21 showed no cross-reactivity with other antigens, such as inactivated BTV, inactivated FMDV, His-tag labeled protein, and BSA. The ELISA titer of these three nanobodies against inactivated SRMV was up to 1:1000. However, only NbSRMV-1-21 displayed SRMV neutralizing activity at a maximum dilution of 1:4. The results indicate that the nanobodies against SRMV generated in this study could be useful in future applications. This study provided a novel antibody tool and laid a foundation for the treatment and detection of SRMV.

Satellite remote sensing technology significantly contributes to intelligent transportation by optimizing traffic planning via global perspectives and rich data, enhancing traffic efficiency and reducing environmental impact. However, current target detection models frequently exhibit low accuracy in vehicle detection tasks due to complex background interference in satellite imageries and a need for critical semantic information. To improve vehicle target detection accuracy, this study introduces SatDetX-YOLO, a vehicle detection model for satellite remote sensing images based on YOLOv8. The model involves reconstructing the backbone network with FasterNet for enhanced feature extraction, a redesigned decoupled head for improved computational efficiency and complex data processing, and incorporating the Deformable Attention Module (DAM) to increase sensitivity to small targets and feature correlation capture. Employing the Maximum Probabilistic Distance IoU (MPDIoU) loss function enhances adaptability and generalization to diverse vehicle targets. Experimental results demonstrate that under comparable FPS, SatDetX-YOLO’s Precision (P), Recall (R), and Mean Average Precision (mAP) improved by 3.5%, 3.3%, and 3.2%, respectively. Despite a minor reduction in FPS, the model significantly enhances detection accuracy, striking a balance between accuracy and speed.

Abstract Sequencing strategies have continually advanced, with high-throughput sequencing (HTS) technologies emerging as pivotal tools in plant phylogenomics. As a standard form of target capture sequencing, hybridization target enrichment sequencing (Hyb-seq) is innovative and flexible, and then HTS strategy is widely adopted in phylogenomics. The universal Angiosperms353 probe sets (Angiosperms353) are instrumental in Hyb-seq and have been witnessed increased application in recent years. This review integrates numerous publications and empirical datasets to comprehensively assess the potential applications of Angiosperms353. Through evaluations using 18-taxa and 343-taxa genomic datasets, this review explores potential factors influencing Angiosperms353 gene capture. The RNA-seq strategy yielded the highest number of Angiosperms353 genes, followed by whole-genome sequencing and genome skimming. Increased sequencing depth enhanced gene capture yields, and no evident bias was observed across all tested angiosperm groups. In the genomics era with extensive HTS data, this review provides comprehensive guidelines for the practical application of Angiosperms353, promoting resource recycling and maximizing genomic data sharing and utilization.

Abstract Rolling bearings working in oscillating applications often suffer from false brinelling and fretting corrosion that limit their service life. At present, the study of oscillation-induced failures is primarily dominated by a large number of sliding and bench tests. However, most of the previous work are not able to fully describe the two closely related but different failure mechanisms. In this study, the origin and evolution of the two failures are thoroughly investigated under strictly controlled oscillating rolling conditions. The results show that failure evolves from false brinelling to fretting corrosion under dry friction conditions regardless of the oscillation amplitude. This process is accompanied by a large area peeling and serious oxidation, and the oxygen content increases from 2.50 wt% to 21.22 wt%. Oxidation is a crucial factor in the evolution of false brinelling into fretting corrosion. However, the distribution of microwear and oxygen content depend on the oscillation amplitude. Under lubrication conditions, the friction coefficient (COF) is smaller in the initial stage and led to local slip or transient instability of rolling elements. Therefore, the size of the wear marks and COF increased and appeared to be larger than those under a dry friction condition. However, grease can separate the contact surfaces, only surface deformation owing to false brinelling and slight damage at the roughness level occurred. The evolution of false brinelling and fretting corrosion is also closely related to residual stress. False brinelling often occurrs with residual stress accomulation, meanwhile, the residual stress increases from 120 to 300 MPa. When peeling occurs, residual stress is released. The failure pattern transformed from false brinelling to fretting corrosion and is accompanied by oxidation. So far, none of investigation is able to show satisfactory evolution of oscillating-induced failures, this study may contribute to more scientific understanding of rolling bearings against long-run reciprocating oscillating wear.

Abstract Reconstruction of irregular oral-maxillofacial bone defects with an inflammatory microenvironment remains a challenge, as chronic local inflammation can largely impair bone healing. Here, we used magnesium silicate nanospheres (MSNs) to load microRNA-146a-5p (miR-146a) to fabricate a nanobiomaterial, MSN+miR-146a, which showed synergistic promoting effects on the osteogenic differentiation of human dental pulp stem cells (hDPSCs). In addition, miR-146a exhibited an anti-inflammatory effect on mouse bone marrow-derived macrophages (BMMs) under lipopolysaccharide (LPS) stimulation by inhibiting the NF-κB pathway via targeting tumor necrosis factor receptor-associated factor 6 (TRAF6), and MSNs could simultaneously promote M2 polarization of BMMs. MiR-146a was also found to inhibit osteoclast formation. Finally, the dual osteogenic-promoting and immunoregulatory effects of MSN+miR-146a were further validated in a stimulated infected mouse mandibular bone defect model via delivery by a photocuring hydrogel. Collectively, the MSN+miR-146a complex revealed good potential in treating inflammatory irregular oral-maxillofacial bone defects.

The forest experience is good for people’s physical and mental health. However, few studies on the effects of pure forest based on the duration and way of experience on people’s physical and mental recovery. In this study, we took 180 first-year college students as research objects and conducted experiments in Pinus sylvestris and Betula platyphylla and the control group of grass plot. The changes of physiological and psychological indexes of the subjects were compared by two perception methods (onsite perception, video perception) and three perception duration (10 min, 20 min, 30 min). The results indicated that: (1) Differences between the two pure forests were mainly reflected in short-term recovery of diastolic blood pressure (DBP) and long-term recovery of total mood disorder (TMD). (2) Video perception was more conducive to short-term recovery of systolic blood pressure (SBP) and diastolic blood pressure (DBP). (3) Viewing the Pinus sylvestris for 20 min in different ways was the best way to relieve stress. It is suggested that, Pinus sylvestris can be used as the rehabilitation perception material, and reasonable path length or perception time can be selected for landscape construction in future. These results can provide scientific reference for landscape design based on forest health and environmental perception.

Abstract Across the globe, tree species are under high anthropogenic pressure. Risks of extinction are notably more severe for species with restricted ranges and distinct evolutionary histories. Here, we use a global dataset covering 41,835 species (65.1% of known tree species) to assess the spatial pattern of tree species’ phylogenetic endemism, its macroecological drivers, and how future pressures may affect the conservation status of the identified hotspots. We found that low-to-mid latitudes host most endemism hotspots, with current climate being the strongest driver, and climatic stability across thousands to millions of years back in time as a major co-determinant. These hotspots are mostly located outside of protected areas and face relatively high land-use change and future climate change pressure. Our study highlights the risk from climate change for tree diversity and the necessity to strengthen conservation and restoration actions in global hotspots of phylogenetic endemism for trees to avoid major future losses of tree diversity.

The present research examined the impact of L-glutamic acid (Glu) supplementation on the growth performance, muscle composition, gene expression correlated with muscle growth, and intestinal health of largemouth bass. There were 525 fish in total, which were distributed randomly into five groups. Each group had three replicates, and each replicate consisted of 35 fish. Groups with control and experimental diets were assigned glutamic acid amounts of 0.2%, 0.4%, 0.6%, and 0.8%. The findings demonstrated that glutamic acid supplementation enhanced growth performance, feed intake (FI), and condition factor (CF), with the best value being attained at 0.4% Glu. The mean muscle fiber area was increased and the muscle fiber density was decreased in the 0.6% Glu group. The levels of total amino acids and specific amino acids, such as glutamic acid, aspartic acid, leucine, valine, alanine, and glycine, were shown to be higher in the 0.6% Glu group. In the 0.6% Glu group, the mRNA expression levels of atrogin-1, murf-1, foxo3a, and 4e-bp1 were decreased compared to the control group. Conversely, the mRNA expression levels of myf5, myog, myod, s6k1, tor, akt, and pi3k were increased in the 0.6% Glu group compared to the control group. The 0.4% Glu group had higher intestinal amylase, lipase, and protease activities and greater villus height, villus width, and muscle thickness. In summary, Glu can support largemouth bass growth, muscular development, intestinal digestion, and absorption.

Callose, found in the cell walls of higher plants such as β-1,3-glucan with β-1,6 branches, is pivotal for both plant development and responses to biotic and abiotic stressors. Plasmodesmata (PD), membranous channels linking the cytoplasm, plasma membrane, and endoplasmic reticulum of adjacent cells, facilitate molecular transport, crucial for developmental and physiological processes. The regulation of both the structural and transport functions of PD is intricate. The accumulation of callose in the PD neck is particularly significant for the regulation of PD permeability. This callose deposition, occurring at a specific site of pathogenic incursion, decelerates the invasion and proliferation of pathogens by reducing the PD pore size. Scholarly investigations over the past two decades have illuminated pathogen-induced callose deposition and the ensuing PD regulation. This gradual understanding reveals the complex regulatory interactions governing defense-related callose accumulation and protein-mediated PD regulation, underscoring its role in plant defense. This review systematically outlines callose accumulation mechanisms and enzymatic regulation in plant defense and discusses PD’s varied participation against viral, fungal, and bacterial infestations. It scrutinizes callose-induced structural changes in PD, highlighting their implications for plant immunity. This review emphasizes dynamic callose calibration in PD constrictions and elucidates the implications and potential challenges of this intricate defense mechanism, integral to the plant’s immune system.

Cellulose aerogels are considered as ideal thermal insulation materials owing to their excellent properties such as a low density, high porosity, and low thermal conductivity. However, they still suffer from poor mechanical properties and low flame retardancy. In this study, mullite-fibers-reinforced bagasse cellulose (Mubce) aerogels are designed using bagasse cellulose as the raw material, mullite fibers as the reinforcing agent, glutaraldehyde as the cross-linking agent, and chitosan as the additive. The resulted Mubce aerogels exhibit a low density of 0.085 g/cm3, a high porosity of 93.2%, a low thermal conductivity of 0.0276 W/(m∙K), superior mechanical performances, and an enhanced flame retardancy. The present work offers a novel and straightforward strategy for creating high-performance aerogels, aiming to broaden the application of cellulose aerogels in thermal insulation.

The derivation of the particulate-backscattering coefficient (bbp) from Lidar signals is highly influenced by the parameter χp(π), which is defined by χp(π) = bbp/(2πβp(π)). This parameter facilitates the correlation of the particulate-volume-scattering function at 180°, denoted βp(π), with bbp. However, studies exploring the global and seasonal fluctuations of χp(π) remain sparse, largely due to measurement difficulties of βp(π) in the field conditions. This study pioneers the global data collection for χp(π), integrating bbp observations from Biogeochemical Argo (BGC-Argo) floats and βp(π) data from the Cloud-Aerosol Lidar with Orthogonal Polarization (CALIOP) spaceborne lidar. Our findings indicate that χp(π) experiences significant seasonal differences globally, peaking during summer and nadiring in winter. The global average χp(π) was calculated as 0.40, 0.48, 0.43, and 0.35 during spring, summer, autumn, and winter, respectively. The daytime values of χp(π) slightly exceeded those registered at night. To illuminate the seasonal variations in χp(π) in 26 sea regions worldwide, we deployed passive ocean color data MODIS bbp and active remote sensing data CALIOP βp(π), distinguishing three primary seasonal change patterns—the “summer peak”, the “decline”, and the “autumn pole”—with the “summer peak” typology being the most common. Post recalibration of the CALIOP bbp product considering seasonal χp(π) variations, we observed substantial statistical improvements. Specifically, the coefficient of determination (R2) markedly improved from 0.84 to 0.89, while the root mean square error (RMSE) declined from 4.0 × 10−4 m−1 to 3.0 × 10−4 m−1. Concurrently, the mean absolute percentage error (MAPE) also dropped significantly, from 31.48% to 25.27%.

The objective of this study was to explore the dynamic changes in the gut microbiota of Simmental calves before weaning and to compare the microbial composition and functionality between healthy calves and those with diarrhea. Fourteen neonatal Simmental calves were divided into a healthy group (n = 8) and a diarrhea group (n = 6). Rectal stool samples were collected from each calf on days 1, 3, 5, 7, 9, 12, 15, 18, 22, 26, 30, 35, and 40. High-throughput sequencing of the 16S rRNA gene V1–V9 region was conducted to examine changes in the gut microbiota over time in both groups and to assess the influence of diarrhea on microbiota structure and function. Escherichia coli, Bacteroides fragilis, and B. vulgatus were the top three bacterial species in preweaning Simmental calves. Meanwhile, the major functions of the fecal microbiota included “metabolic pathways”, “biosynthesis of secondary metabolites”, “biosynthesis of antibiotics”, “microbial metabolism in diverse environments”, and “biosynthesis of amino acids”. For calves in the healthy group, PCoA revealed that the bacterial profiles on days 1, 3, 5, 7, and 9 differed from those on days 15, 18, 22, 26, 30, 35, and 40. The profiles on day 12 clustered with both groups, indicating that microbial structure changes increased with age. When comparing the relative abundance of bacteria between healthy and diarrheic calves, the beneficial Lactobacillus johnsonii, Faecalibacterium prausnitzii, and Limosilactobacillus were significantly more abundant in the healthy group than those in the diarrhea group (p < 0.05). This study provides fundamental insights into the gut microbiota composition of Simmental calves before weaning, potentially facilitating early interventions for calf diarrhea and probiotic development.

The quality of raw camel milk is affected by its bacterial composition and diversity. However, few studies have investigated the bacterial composition and diversity of raw camel milk. In this study, we obtained 20 samples of camel milk during spring and summer in Urumqi and Hami, Xinjiang, China. Single-molecule real-time sequencing technology was used to analyze the bacterial community composition. The results revealed that there were significant seasonal differences in the bacterial composition and diversity of camel milk. Overall, Epilithonimonas was the most abundant bacterial genus in our samples. Through the annotated genes inferred by PICRUSt2 were mapped against KEGG database. Non-parametric analysis of the bacterial community prediction function revealed a strong bacterial interdependence with metabolic pathways (81.83%). There were clear regional and seasonal differences in level 3 metabolic pathways such as fat, vitamins, and amino acids in camel milk. In addition, we identified lactic acid bacteria in camel milk with antibacterial and anti-tumor activities. Our findings revealed that camel milk from Xinjiang had serious risk of contamination by psychrophilic and pathogenic bacteria. Our research established a crucial theoretical foundation for ensuring the quality and safety of camel milk, thereby contributing significantly to the robust growth of China’s camel milk industry.

The Antarctic krill is a pivotal species in the Southern Ocean ecosystem, primarily due to its extraordinary nutritional content and plentiful resources. Studying the distribution of these resources and their environmental impact factors is crucial for the successful development of Antarctic krill fisheries. Traditional methodologies such as acoustic measurements, however, often face limitations in their capacity to provide a comprehensive and uninterrupted assessment. Moreover, the six-month duration of polar nights in polar regions presents significant challenges for traditional satellite observations. In this context, LiDAR, an active remote sensing observation method, offers a promising alternative. Known for their high resolution, flexibility, and efficiency, LiDAR systems can obtain detailed information on diurnal ocean parameters in polar regions on a vast scale and in a systematic way. Our study utilizes the spaceborne LiDAR system, CALIPSO, to successfully attain continuous Antarctic krill CPUE over the past decade, using various models such as the generalized linear model (GLM), artificial neural network (ANN), and support vector machine (SVM). A comparative analysis of the prediction results reveals that while both ANN and SVM models outperform the GLM, the SVM’s prediction capabilities are somewhat unstable. Our findings reveal CALIPSO’s potential in overcoming challenges associated with traditional satellite observations during polar winters. In addition, we found no obvious pattern of interannual variation in krill CPUE, with high values predominantly occurring from February to May. This suggests that krill is mainly located around the South Shetland Islands during January-April, before moving offshore towards South Georgia in May-June. A substantial krill aggregation community is found in the South Atlantic waters, indicating high potential for krill fishing. The optimum mix layer depth range for high krill CPUE is 270-390 m, with a chlorophyll concentration of approximately 0.1 mg m-3. The optimum sea surface temperature range is between -1.4-5.5°C, and the sea ice coverage range is approximately 0-0.1×106 km2. The predicted Antarctic krill bioresource has risen from 2.4×108 tons in 2011 to 2.8×108 tons in 2020. This increase in krill biomass aligns with the biomass of krill assessed by CCAMLR.

Background Nitric oxide (NO) is the most important vasodilator secreted by vascular endothelial cells, and its abnormal synthesis is involved in the development of cardiovascular disease. The prenatal period is a critical time for development and largely determines lifelong vascular health in offspring. Given the high incidence and severity of gestational hypoxia in mid‐late pregnancy, it is urgent to further explore whether it affects the long‐term synthesis of NO in offspring vascular endothelial cells. Methods and Results Pregnant Sprague–Dawley rats were housed in a normoxic or hypoxic (10.5% O2) chamber from gestation days 10 to 20. The thoracic aortas of fetal and adult male offspring were isolated for experiments. Gestational hypoxia significantly reduces the NO‐dependent vasodilation mediated by acetylcholine in both the fetal and adult offspring thoracic aorta rings. Meanwhile, acetylcholine‐induced NO synthesis is impaired in vascular endothelial cells from hypoxic offspring thoracic aortas. We demonstrate that gestational hypoxic offspring exhibit a reduced endothelial NO synthesis capacity, primarily due to increased expression of NADPH oxidase 2 and enhanced reactive oxygen species. Additionally, gestational hypoxic offspring show elevated levels of miR‐155‐5p in vascular endothelial cells, which is associated with increased expression of NADPH oxidase 2 and reactive oxygen species generation, as well as impaired NO synthesis. Conclusions The present study is the first to demonstrate that gestational hypoxia impairs endothelial NO synthesis via the miR‐155‐5p/NADPH oxidase 2/reactive oxygen species axis in offspring vessels. These novel findings indicate that the detrimental effects of gestational hypoxia on fetal vascular function can persist into adulthood, providing new insights into the development of vascular diseases.

Summary: The “Fetal Origins of Adult Disease (FOAD)” hypothesis holds that adverse factors during pregnancy can increase the risk of chronic diseases in offspring. Here, we investigated the effects of prenatal hypoxia (PH) on brain structure and function in adult offspring and explored the role of the N6-methyladenosine (m6A) pathway. The results suggest that abnormal cognition in PH offspring may be related to the dysregulation of the m6A pathway, specifically increased levels of YTHDF3 in the hippocampus. YTHDF3 interacts with BTG2 and is involved in the decay of Cbln1 mRNA, leading to the down-regulation of Cbln1 expression. Deficiency of Cbln1 may contribute to abnormal synaptic function, which in turn causes cognitive impairment in PH offspring. This study provides a scientific clues for understanding the mechanisms of impaired cognition in PH offspring and provides a theoretical basis for the treatment of cognitive impairment in offspring exposed to PH.

Abstract Depression has become one of the most common mental illnesses in the world. For better prediction and diagnosis, methods of automatic depression recognition based on speech signal are constantly proposed and updated, with a transition from the early traditional methods based on hand‐crafted features to the application of architectures of deep learning. This paper systematically and precisely outlines the most prominent and up‐to‐date research of automatic depression recognition by intelligent speech signal processing so far. Furthermore, methods for acoustic feature extraction, algorithms for classification and regression, as well as end to end deep models are investigated and analysed. Finally, general trends are summarised and key unresolved issues are identified to be considered in future studies of automatic speech depression recognition.

Geoheritage represents typical geological phenomena formed through internal and external geological processes during the Earth's long evolution. It serves as a carrier of crucial information regarding the Earth's evolution, the origin of life, and changes in the geographic environment. Additionally, it is a valuable geological resource for human utilization. In order to provide essential information for the scientific protection and utilization of geoheritage resources, this paper summarizes and further categorizes the types, grades, and distribution characteristics of geoheritage in China based on extensive firsthand data obtained from recent investigations. The results can be summaried in four aspects: (1) As of the end of 2018, a total of 7,267 important geoheritage sites (including global, national, and provincial levels) have been discovered and surveyed in China. Among them, there are 3,154 geoheritage sites of basic geology, 3,935 geoheritage sites of geomorphic landscape, and 178 geoheritage sites of geological disaster. They can be further categorized into 13 classes and 46 sub-classes, such as stratigraphic sections, rock profiles, rock and soil landforms, and earthquake relics. (2) Through expert evaluations and comparative studies, there are 218 Grade I (global level) geoheritage sites, 1,919 Grade II (national level) geoheritage sites, 4,732 Grade III (provincial level) geoheritage sites, and 398 Grade IV (below provincial level) geoheritage sites in China. (3) Geoheritage in China are mainly distributed in five regions: Southwest China, East China, Central China, Northwest, and North China. The geoheritage sites of basic geology are mainly distributed in the Southwest China, Central China, East China, Northwest China, and North China, with the Southwest China having the highest number, accounting for approximately 27%. The geoheritage sites of geomorphic landscape are mainly distributed in the Southwest China, Northwest China, East China, Central China, and North China, with the Southwest China having the highest number, accounting for approximately 29%. The geoheritage sites of geological disaster are mainly distributed in the Southwest China, Northwest China, and Central China, with the Southwest China having the highest number, accounting for approximately 47%.

Abstract Water inflow is one of the main geohazards that threaten the safety of tunnels and other underground engineering projects. Faulted zone is one of the important geological triggers for such events. Numerical investigations on the evolution of flow behavior in tunnels across fault zones are of significance to the predication and prevention of this type of geohazards. In this work, a numerical investigation model with two overlapped parallel faults is established at a steady stage according to the "Three Zones" fault structure theory. The rapid turbulent flow in the fault zone is simulated by using the improved Darcy-Brinkman seepage model, while the slow laminar flow in ordinary rock zone is described by Darcy equation. The effect of relative position and distance between the tunnel excavation face and overlapped parallel faults to the groundwater pore pressure and flow velocity is studied through several scenarios, and the water inflow rate into the tunnel is calculated. The numerical investigation results reveal that while the tunnel face is excavated into the fault center core, the fractured zone, the ordinary rock zone, and the center of the overlapped faults, the pore pressure value ahead of the excavation face increases while the flow velocity decreases sequentially. The inflow rate is the largest while the tunnel face is excavated to center of the fault center core, which is closely related to the range of the overlapped area. The investigation results offer a practical reference for predicting early warning of water inflow geohazard when a tunnel cross two overlapped parallel faults.

In recent years, caffeic acid and its derivatives have received increasing attention due to their obvious physiological activities and wide distribution in nature. In this paper, to clarify the status of research on plant-derived caffeic acid and its derivatives, nuclear magnetic resonance spectroscopy data and possible biosynthetic pathways of these compounds were collected from scientific databases (SciFinder, PubMed and China Knowledge). According to different types of substituents, 17 caffeic acid and its derivatives can be divided into the following classes: caffeoyl ester derivatives, caffeyltartaric acid, caffeic acid amide derivatives, caffeoyl shikimic acid, caffeoyl quinic acid, caffeoyl danshens and caffeoyl glycoside. Generalization of their 13C-NMR and 1H-NMR data revealed that acylation with caffeic acid to form esters involves acylation shifts, which increase the chemical shift values of the corresponding carbons and decrease the chemical shift values of the corresponding carbons of caffeoyl. Once the hydroxyl group is ester, the hydrogen signal connected to the same carbon shifts to the low field (1.1~1.6). The biosynthetic pathways were summarized, and it was found that caffeic acid and its derivatives are first synthesized in plants through the shikimic acid pathway, in which phenylalanine is deaminated to cinnamic acid and then transformed into caffeic acid and its derivatives. The purpose of this review is to provide a reference for further research on the rapid structural identification and biofabrication of caffeic acid and its derivatives.

A novel functional polycarbonate (PAGC), characterized by the presence of double bonds within its side chain, was successfully synthesized through a ternary copolymerization of propylene oxide (PO), allyl glycidyl ether (AGE), and carbon dioxide (CO2). Polyhedral oligomeric silsesquioxanes octamercaptopropyl (POSS-SH) was employed as a crosslinking agent, contributing to the formation of organic–inorganic hybrid materials. This incorporation was facilitated through thiol-ene click reactions, enabling effective interactions between the POSS molecules and the double bonds in the side chains of the polycarbonate. Scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS) confirmed a homogeneous distribution of silicon (Si) and sulfur (S) in the polycarbonate matrix. The thiol-ene click reaction between POSS-SH and the polycarbonate led to a micro-crosslinked structure. This enhancement significantly increased the tensile strength of the polycarbonate to 42 MPa, a notable improvement over traditional poly (propylene carbonate) (PPC). Moreover, the cross-linked structure exhibited enhanced solvent resistance, expanding the potential applications of these polycarbonates in various plastic materials.

Abstract Background Potassium (K) is an essential nutrient for plant growth and development. However, plant fertilization ignoring the soil K level is very likely to cause excessive fertilizer use, and further arouse a series of side effects. This study investigated the response of cotton growth to different soil K levels and the uptake of major nutrients, aiming to evaluate the appropriate K supply level for cotton growth. Using a random block design with 6 soil K levels, we conducted 18 micro-zones field experiments over two continuous years. The soil available K concentration of each treatment was K1 (99.77–100.90 mg·kg−1), K2 (110.90–111.26 mg·kg−1), K3 (123.48–128.88 mg·kg−1), K4 (140.13–145.10 mg·kg−1), K5 (154.43–155.38 mg·kg−1), and K6 (165.77–168.75 mg·kg−1). Cotton nutrient contents, soil nutrient contents, accumulation and distribution of dry matter in cotton were determined, and the relationships between K content in soil and plants and dry matter accumulation were analyzed. Results The soil K content had a significantly positive relationship with dry matter and K accumulation in cotton plants. There were significant differences in dry matter accumulation, single-plant seed cotton yield, mineral nutrient uptake and the proportion of K accumulation in reproductive organs among different soil K levels. The results showed that there was significant difference between K4 and lower K level treatments (K1 and K2), but no significant difference between K4 and higher K level treatments (K5 and K6) in dry matter, single-plant seed cotton yield, or accumulation, distribution and seed cotton production efficiency of N, P and K. Conclusion The soil K level of K4 was able to provide sufficient K for cotton growth in our experiment. Therefore, when the soil K level reached 140.13 mg·kg−1, further increasing the soil K concentration no longer had a significant positive effect on cotton growth.

Abstract Background Myocardial injury post-stroke is a common sequela of acute stroke. Triglyceride-glucose index (TyG index), a valuable surrogate indicator of insulin resistance, has been suggested to be closely related to cardiovascular outcomes. However, it is unknown whether the TyG index is independently associated with a higher risk of myocardial injury post-stroke. We therefore investigated the longitudinal association between TyG index and risk of myocardial injury post-stroke in older patients with first-ever ischemic stroke and no prior cardiovascular comorbidities. Methods We included older patients with first-ever ischemic stroke and no prior cardiovascular comorbidities between January 2021 to December 2021. The individuals were stratified into low and high TyG index groups according to the optimal cutoff value with TyG index. We performed logistic regression analysis, propensity score matching (PSM) analysis, restricted cubic spline analysis, and subgroup analyses to explore the longitudinal association between TyG index and risk of myocardial injury post-stroke. Results We included 386 individuals with a median age of 69.8 years (interquartile range: 66.6, 75.3). The optimal TyG index cut-off for predicting myocardial injury post-stroke was 8.9 (sensitivity 67.8%; specificity 75.5%; area under curve 0.701). Multivariate logistic regression analysis revealed that the risk of genesis of myocardial injury post-stroke increased with elevated TyG index (odds ratio [OR], 2.333; 95% confidence interval [CI], 1.201–4.585; P = 0.013). BackgroundLate-onset cobalamin C (cblC) deficiency is associated with a wide range of neurological and psychiatric symptoms, hematological manifestations, anorexia, renal failure, ocular abnormalities, dermatitis, and pancreatitis. However, the neuroimaging characteristics of late-onset cblC deficiency remain insufficiently documented. Common findings include diffuse white matter swelling, varying degrees of severe leukoaraiosis, hydrocephalus, corpus callosum atrophy, and symmetric bilateral basal ganglia lesions. In this report, we present a case of late-onset cblC deficiency in adults presenting with cerebellar ataxia as the primary symptom. The MRI findings revealed bilateral lateral cerebellar hemispheres exhibiting symmetric hyperintensity, primarily observed in diffusion-weighted imaging (DWI), which is a rarely reported imaging change in this context.Case presentationOur patient was a male who experienced symptoms starting at the age of 30 years, including unsteady walking, apparent cerebellar ataxia, and cognitive impairment upon nervous system examination. Brain magnetic resonance imaging (MRI) exhibited symmetric hyperintensity in the bilateral lateral cerebellar hemispheres, predominantly manifested in DWI, without any enhancement. Subsequently, significantly elevated blood total homocysteine and urinary methylmalonic acid levels were observed. Genetic analysis confirmed the presence of MMACHC compound heterozygous mutants c.482G > A and c.609G > A, thus confirming the diagnosis of cblC deficiency. These variants were classified as likely pathogenic following the guidelines of the American College of Medical Genetics and Genomics (ACMG) and were verified using Sanger sequencing. Following treatment, the patient experienced improvements in walking ability and cognition, a significant decrease in blood total homocysteine levels, and reversal of the imaging lesions.In conclusionLate-onset cblC deficiency presents with diverse clinical and imaging manifestations. Early diagnosis and treatment are crucial in achieving a favorable prognosis. This case serves as a reminder to clinicians not to overlook genetic metabolic disorders, particularly those causing multisite damage, in adult patients with undiagnosed neurological disorders, especially those affecting the cerebellum. Notably, methylmalonic acidemia should be considered within the spectrum of bilateral cerebellar lesions.

Abstract Introduction Ferroptosis has recently been recognized as a new cause of ischemia reperfusion injury due to blood–brain barrier (BBB) disruption followed by secondary iron‐loaded transferrin (TF) influx. As a novel and independent cell death pathway, ferroptosis was characterized by iron‐dependent lipid peroxidation, decline of GSH, GPX4, and shrinking mitochondria. Cottonseed oil (CSO), a liposoluble solvent, can alleviate ischemia stroke injuries and oxidative stress. However, the effect of CSO on ischemic stroke–induced ferroptosis has not been explored. In this study, we investigated the effect of CSO on ferroptosis caused by cerebral ischemic injury in rats. Methods We conducted the subcutaneous injection of 1.3 mL/kg CSO every other day for 3 weeks on rats with middle cerebral artery occlusion–reperfusion (MCAO‐R) injury. We used Garcia Test, TTC staining, HE, Nissl and NeuN staining, Evans blue test, 68Ga‐citrate PET, Western blot, immunofluorescence staining, Elisa kits, and transmission electron microscopy to detect the infarct volume, neural injuries, and ferroptosis‐related indexes. Results CSO treatment could significantly ameliorate MCAO‐R‐induced neurological dysfunction in a male rat model. Furthermore, it reduced infarct volume and neuronal injuries; protected BBB integrity; reduced the influx of iron ion, TF, and TF receptors; up‐regulated anti‐ferroptosis proteins (GPX4, xCT, HO1, FTH1), while down‐regulating ferroptosis‐related protein ACSL4; increased the activity of GSH and SOD; and decreased MDA and LPO levels. Mitochondrial destruction induced by ischemic stroke was also alleviated by CSO treatment. Conclusion CSO treatment can alleviate ischemic stroke injury via ferroptosis inhibition, which provides a new potential therapeutic mechanism for CSO neuroprotection against ischemic stroke.

Abstract Background Water shortage caused by global warming seriously affects the yield and quality of vegetable crops. β-carotene, the lipid-soluble natural product with important pharmacological value, is abundant in celery. Transcription factor MYB family extensively disperses in plants and plays regulatory roles in carotenoid metabolism and water scarcity response. Results Here, the AgMYB5 gene encoding 196 amino acids was amplified from celery cv. ‘Jinnanshiqin’. In celery, the expression of AgMYB5 exhibited transactivation activity, tissue specificity, and drought-condition responsiveness. Further analysis proved that ectopic expression of AgMYB5 increased β-carotene content and promoted drought tolerance in transgenic Arabidopsis thaliana. Moreover, AgMYB5 expression promoted β-carotene biosynthesis by triggering the expression of AtCRTISO and AtLCYB, which in turn increased antioxidant enzyme activities, and led to the decreased contents of H2O2 and MDA, and the inhibition of O2 − generation. Meanwhile, β-carotene accumulation promoted endogenous ABA biosynthesis of transgenic Arabidopsis, which resulted in ABA-induced stomatal closing and delayed water loss. In addition, ectopic expression of AgMYB5 increased expression levels of AtERD1, AtP5CS1, AtRD22, and AtRD29. Conclusions The findings indicated that AgMYB5 up-regulated β-carotene biosynthesis and drought tolerance of Arabidopsis.

To improve the accuracy of short term traffic flow prediction and to solve the problems of nonlinearity of short term traffic flow, more noise in the data, and more difficult to determine the parametes of long short term memory networks, a combined traffic flow prediction model based on variational modal decomposition (VMD) and improved dung beetle optimization-long short term memory network (IDBO-LSTM) is proposed. First, to extract various modal components, the historical traffic flow data are smoothed using variational modal decomposition (VMD). Second, the LSTM prediction model is built for each individual subsequence, and the parameters of the LSTM are optimized using the IDBO algorithm which combines Singer chaos mapping, variable spiral search strategy, and Levy flight strategy. Finally, to acquire the final prediction results, the predicted values of various subsequences are added up and reassembled. Experiments were conducted using data collected from eight sensors along an interstate highway in California, and taking the straight road morning peak (S-M) data as an example, compared with LSTM and VMD-LSTM, the MAE of VMD-IDBO-LSTM is reduced by 26.69 and 7.5108, MAPE is reduced by 8.08059% and 2.27569%, and RMSE is reduced by 33.6912 and 8.7657. According to the findings, the VMD-IDBO-LSTM model that was proposed is capable of significantly improving the accuracy of short-term traffic flow prediction while also effectively addressing nonlinearity, data noise, and the difficulty of identifying the LSTM parameters.

With the development of modern medicine, the research methods of occurrence, development and treatment of orthopedic diseases are developing rapidly. The microenvironment provided by traditional orthopedic research methods differ considerably from the human body, resulting in poor or inconsistent conclusions in previous studies. Microfluidic technology has shown its advantages in the field of orthopedic research, especially in providing bionic mechanical stimulation environment. The microfluidic device can simulate the complex internal environment through the fine and complex structure and perfusion control system, and provide a stable, controllable and efficient culture system. Moreover, it can serve as a manufacturing device, which can produce bone grafts or bone like organs for tissue engineering with bionic structure. It can also simultaneously act as a detection device, which can realize high-throughput detection of small samples at low cost. In addition, we can establish in vitro physiological or pathological models on microfluidic systems to assist in the diagnosis and treatment of orthopedic diseases. This paper reviews the medical application of microfluidic devices in orthopedics.

The underlying causes of biodiversity disparities among geographic regions have long been a fundamental theme in ecology and evolution. However, the patterns of phylogenetic diversity (PD) and phylogenetic beta diversity (PBD) of congeners that are disjunctly distributed between eastern Asia–eastern North America (EA−ENA disjuncts) and their associated factors remain unknown. Here we investigated the standardized effect size of PD (SES-PD), PBD, and potentially associated factors in 11 natural mixed forest sites (five in EA and six in ENA) where abundant EA−ENA disjuncts occur. We found that the disjuncts in ENA possessed higher SES-PD than those in EA at the continental scale (1.96 vs −1.12), even though the number of disjunct species in ENA is much lower than in EA (128 vs 263). SES-PD of the EA−ENA disjuncts tended to decrease with increasing latitude in 11 sites. The latitudinal diversity gradient of SES-PD was stronger in EA sites than in ENA sites. Based on the unweighted unique fraction metric (UniFrac) distance and the phylogenetic community dissimilarity, PBD showed that the two northern sites in EA were more similar to the six-site ENA group than to the remaining southern EA sites. Based on the standardized effect size of mean pairwise distances (SES-MPD), nine of eleven studied sites showed a neutral community structure (−1.96 ≤ SES-MPD ≤ 1.96). Both Pearson's r and structural equation modeling suggested that SES-PD of the EA–ENA disjuncts was mostly associated with mean divergence time. Moreover, SES-PD of the EA–ENA disjuncts was positively correlated with temperature-related climatic factors, although negatively correlated with mean diversification rate and community structure. By applying approaches from phylogenetics and community ecology, our work sheds light on historical patterns of the EA−ENA disjunction and paves the way for further research.

Abstract Background Previous studies have shown that depression is often accompanied by an increase in mtDNA copy number and a decrease in ATP levels; however, the exact regulatory mechanisms remain unclear. Methods In the present study, Western blot, cell knockdown, immunofluorescence, immunoprecipitation and ChIP-qPCR assays were used to detect changes in the Ahi1/GR-TFAM-mtDNA pathway in the brains of neuronal Abelson helper integration site-1 (Ahi1) KO mice and dexamethasone (Dex)-induced mice to elucidate the pathogenesis of depression. In addition, a rescue experiment was performed to determine the effects of regular exercise on the Ahi1/GR-TFAM-mtDNA-ATP pathway and depression-like behavior in Dex-induced mice and Ahi1 KO mice under stress. Results In this study, we found that ATP levels decreased and mitochondrial DNA (mtDNA) copy numbers increased in depression-related brain regions in Dex-induced depressive mice and Ahi1 knockout (KO) mice. In addition, Ahi1 and glucocorticoid receptor (GR), two important proteins related to stress and depressive behaviors, were significantly decreased in the mitochondria under stress. Intriguingly, GR can bind to the D-loop control region of mitochondria and regulate mitochondrial replication and transcription. Importantly, regular exercise significantly increased mitochondrial Ahi1/GR levels and ATP levels and thus improved depression-like behaviors in Dex-induced depressive mice but not in Ahi1 KO mice under stress. Conclusions In summary, our findings demonstrated that the mitochondrial Ahi1/GR complex and TFAM coordinately regulate mtDNA copy numbers and brain ATP levels by binding to the D-loop region of mtDNA Regular exercise increases the levels of the mitochondrial Ahi1/GR complex and improves depressive behaviors. The aim of this study is to better understand diffusive convection (DC) and its role in the upper ocean dynamic environment and sea ice melting in the Canada Basin. Based on a moored dataset with 6737 profiles collected from August 2003 to August 2011 in the upper layer of the Canada Basin, DC between the warm and salty Atlantic Water (AW) and the colder and less salty Lower Halocline Water (LHW) were investigated. The moorings were designated at four stations: A, B, C, and D, located at the southwestern, southeastern, northeastern, and northwestern parts of the basin, respectively. During the observation period, the temperature, salinity, and depth of the AW and LHW exhibited unique temporal variations. The temperature and salinity of the AW varied among stations, with a decreasing trend from northwest to southeast, consistent with the propagation path of the AW in the Canada Basin. The temperature and salinity of the LHW were similar at all stations. The AW and LHW cores were located between depths of 320–500 m and 160–300 m, respectively, and both gradually deepened over time. Distinct DC staircase structures were observed between the AW and LHW, more pronounced at stations C and D than at stations A and B, which is speculated to be related to eddies at stations A and B during the observation period. The vertical heat fluxes through the DC staircase layer at stations C and D (FHc_C and FHc_D) were estimated using an empirical formula. FHc_C ranged from 0.05 to 0.94 W/m2, and FHc_D ranged from 0.05 to 0.6 W/m2, with the maximum probability value for both at approximately 0.2 W/m2. The effective diffusivities at these two stations (KT_C and KT_D) are similar, ranging from 2 × 10−6 to 3 × 10−5 m2/s, with the highest probability occurring at 6 × 10−6 m2/s. Both the probability density function of the heat flux and the effective diffusivity skewed towards larger values and obey a lognormal distribution, indicating turbulence intermittency of the DC staircase in the Canada Basin. These finding offers new insights into the heat transport and turbulence in the DC staircase, and then bring a deeper understanding of sea ice melting in the Canada Basin.

The influence of sintering parameters on the microstructure, phase composition and mechanical property of the Ti _3 SiC _2 /Cu composites sintered by spark plasma sintering technique was investigated and the related sintering mechanism was clarified in detail. Results indicated that during the heating process, one part of the high energy adsorbed by the composites let Cu melt and fill the gaps inside the composites. Meanwhile, there’s different molten condition about Cu duel to different heating temperature that cause Cu fill in the most space between Ti _3 SiC _2 particles. The other part of the energy caused to the local high temperature, contributing for the chemical reaction and the formation of TiC _x and Cu _3 Si. Therefore, at the same heating rate, the composites showed better mechanical property and higher density with a longer heating time. The heating stage played an important role in the change of the mechanical property, microstructure and volume of the composites. During the holding stage, because the amount of the reactants significantly decreased, the related chemical reaction got slow and the energy needed during this stage was lower than that during the heating stage. And the particle of Ti _3 SiC _2 need more time for moving to the space of Cu. Therefore, the higher the holding temperature, the more significant was the diffusion of the phases. The more uniform the phases, the higher was the density. When heating temperature (higher than holding temperature) cause Cu complete melt and holding temperature keep the Cu in semi-melt, there will be a more effective sintering method.

The monotypic genus Pseudomarrubium is perhaps the most poorly understood taxon within the subfamily Lamioideae (Lamiaceae). While advances in molecular phylogenetics have clarified the phylogeny and taxonomy of Lamioideae, the systematic placement of Pseudomarrubium remains unclear, as it has never been tested using molecular phylogenetic analyses due to an inaccessibility of plant material. In this study, we used high-throughput next-generation sequencing to infer the systematic placement of Pseudomarrubium based on old herbarium collections as well as silica gel-dried leaves. Phylogenomic analyses demonstrate that Pseudomarrubium is a member of tribe Phlomideae and nested within Phlomoides, and a new combination, Phlomoides eremostachydioides, is proposed. The study highlights that herbarium-preserved samples, even very old ones, are invaluable resources that can be used to generate reliable genome-scale data and be used for molecular phylogenetic inquiries.

SARS‐CoV‐2 aptamer is a favorable candidate for the recognition and detection of SARS‐CoV‐2, owing to its small size and easy synthesis. However, the issue of compromised binding affinities in real samples and targeting mutant SARS‐CoV‐2 hinder wide applications of the aptamer. In this study, it is discovered that molecular crowding could increase binding affinity of CoV2‐6C3 aptamer against RBD (Receptor Binding Domain) of SARS‐CoV‐2 via increasing the absolute value of the enthalpy change. The values of the equilibrium dissociation constant in molecular crowding decrease by 70% and 150%, respectively, against wild‐type and mutant RBD compared with those in buffer without crowding. Moreover, the detection limit of SARS‐CoV‐2 pseudovirus is up to 5 times lower under molecular crowding compared to dilute conditions. The discovery deepens the understanding of aptamer‐target interaction mechanisms in crowding conditions and provides an effective way to apply SARS‐CoV‐2 aptamer for virus recognition and detection.

IntroductionPhosphorus (P) deficiency hinders cotton (Gossypium hirustum L.) growth and development, seriously affecting lint yield and fiber quality. However, it is still unclear how P fertilizer affects fiber length.MethodsTherefore, a two-year (2019-2020) pool-culture experiment was conducted using the split-plot design, with two cotton cultivars (CCRI-79; low-P tolerant and SCRC-28; low-P sensitive) as the main plot. Three soil available phosphorus (AP) contents (P0: 3 ± 0.5, P1: 6 ± 0.5, and P2 (control) with 15 ± 0.5 mg kg−1) were applied to the plots, as the subplot, to investigate the impact of soil AP content on cotton fiber elongation and length. ResultsLow soil AP (P0 and P1) decreased the contents of the osmotically active solutes in the cotton fibers, including potassium ions (K+), malate, soluble sugar, and sucrose, by 2.2–10.2%, 14.4–47.3%, 8.7–24.5%, and 10.1–23.4%, respectively, inhibiting the vacuoles from facilitating fiber elongation through osmoregulation. Moreover, soil AP deficiency also reduced the activities of enzymes participated in fiber elongation (plasma membrane H+-ATPase (PM-H+-ATPase), vacuole membrane H+-ATPase (V-H+-ATPase), vacuole membrane H+-translocating inorganic pyrophosphatase (V-H+-PPase), and phosphoenolpyruvate carboxylase (PEPC)). The PM-H+-ATPase, V-H+-ATPase, V-H+-PPase, and PEPC were reduced by 8.4–33.0%, 7.0–33.8%, 14.1–38.4%, and 16.9–40.2%, respectively, inhibiting the transmembrane transport of the osmotically active solutes and acidified conditions for fiber cell wall, thus limiting the fiber elongation. Similarly, soil AP deficiency reduced the fiber length by 0.6–3.0 mm, mainly due to the 3.8–16.3% reduction of the maximum velocity of fiber elongation (VLmax). Additionally, the upper fruiting branch positions (FB10–11) had higher VLmax and longer fiber lengths under low soil AP. DiscussionCotton fibers with higher malate content and V-H+-ATPase and V-H+-PPase activities yielded longer fibers. And the malate and soluble sugar contents and V-H+-ATPase and PEPC activities in the SCRC-28's fiber were more sensitive to soil AP deficiency in contrast to those of CCRI-79, possibly explaining the SCRC-28 fiber length sensitivity to low soil AP.

As the primary energy source for a plant host and microbe to sustain life, sugar is generally exported by Sugars Will Eventually be Exported Transporters (SWEETs) to the host extracellular spaces or the apoplast. There, the host and microbes compete for hexose, sucrose, and other important nutrients. The host and microbial monosaccharide transporters (MSTs) and sucrose transporters (SUTs) play a key role in the “evolutionary arms race”. The result of this competition hinges on the proportion of sugar distribution between the host and microbes. In some plants (such as Arabidopsis, corn, and rice) and their interacting pathogens, the key transporters responsible for sugar competition have been identified. However, the regulatory mechanisms of sugar transporters, especially in the microbes require further investigation. Here, the key transporters that are responsible for the sugar competition in the host and pathogen have been identified and the regulatory mechanisms of the sugar transport have been briefly analyzed. These data are of great significance to the increase of the sugar distribution in plants for improvement in the yield.

The satellite lidar-derived ocean particulate backscattering coefficient (bbp) has rarely been validated globally with in situ observations, and we need to understand how well the satellite CALIPSO lidar bbp approach performs. Whether lidar bbp performs better in terms of observation accuracy compared to passive ocean color remote sensing has yet to be evaluated for detailed validation. With the continued deployment of the BGC-Argo float array in the global open ocean in recent years, data have accumulated with a total of 42,932 particulate backscattering coefficients (bbp) from 2010 to 2017, allowing for a finer spatial and temporal scale evaluation of the performance of the CALIPSO lidar-observed bbp. We evaluated the performance of CALIPSO-retrieved bbp products using the data detected by the BGC-Argo floats at 12 spatiotemporal matchup scales and discussed the differences in product performance at various interannual, seasonal, and spatial scales. We compare lidar, float, and ocean color bbp at the same locations and times and find that lidar bbp outperforms ocean color data. We also analyzed the key conversion factor β(π)/bbp at different spatial and temporal scales and found that there was a seasonal difference in the optimal conversion factor.

Parkinson’s disease (PD) is one of the most common degenerative diseases. It is most typically characterized by neuronal death following the accumulation of Lewis inclusions in dopaminergic neurons in the substantia nigra region, with clinical symptoms such as motor retardation, autonomic dysfunction, and dystonia spasms. The exact molecular mechanism of its pathogenesis has not been revealed up to now. And there is a lack of effective treatments for PD, which places a burden on patients, families, and society. CRISPR Cas9 is a powerful technology to modify target genomic sequence with rapid development. More and more scientists utilized this technique to perform research associated neurodegenerative disease including PD. However, the complexity involved makes it urgent to organize and summarize the existing findings to facilitate a clearer understanding. In this review, we described the development of CRISPR Cas9 technology and the latest spin-off gene editing systems. Then we focused on the application of CRISPR Cas9 technology in PD research, summarizing the construction of the novel PD-related medical models including cellular models, small animal models, large mammal models. We also discussed new directions and target molecules related to the use of CRISPR Cas9 for PD treatment from the above models. Finally, we proposed the view about the directions for the development and optimization of the CRISPR Cas9 technology system, and its application to PD and gene therapy in the future. All these results provided a valuable reference and enhanced in understanding for studying PD.

Amyotrophic lateral sclerosis (ALS) is a deadly neurological disease with a complicated and variable pathophysiology yet to be fully understood. There is currently no effective treatment available to either slow or terminate it. However, recent advances in ALS genomics have linked genes to phenotypes, encouraging the creation of novel therapeutic approaches and giving researchers more tools to create efficient animal models. Genetically engineered rodent models replicating ALS disease pathology have a high predictive value for translational research. This review addresses the history of the evolution of gene editing tools, the most recent ALS disease models, and the application of CRISPR/Cas9 against ALS disease.
