To investigate the correlation between plasma glutathione peroxidase 4 (GPX4) and N-acetyl-neuraminic acid (Neu5Ac) with clinical risk stratification and outcomes of acute coronary syndrome (ACS) patients.Between October 2018 and July 2019, 413 patients that were scheduled for coronary angiography were enrolled in this prospective study at the First Affiliated Hospital of Bengbu Medical College, Bengbu, China. Patients were divided into control and ACS groups. Patients with ACS were divided into 3 risk levels based on their thrombolysis in myocardial infarction risk score. After discharge, ACS patients were followed for the incidence of major adverse cardiac events (MACEs). For the analysis of cumulative endpoint event occurrences, the Kaplan-Meier method was applied.The ACS group had lower plasma GPX4 but higher Neu5Ac levels than the control group. There was a greater increase in plasma Neu5Ac in the high-risk group when compared with the medium-risk and low-risk groups, while GPX4 levels were higher in the low-risk group. The MACEs group had higher plasma Neu5Ac but lower GPX4 levels than the non-MACEs group. The plasma Neu5Ac was an independent risk factor but GPX4 was a protective factor for MACEs.Glutathione peroxidase 4 and Neu5Ac levels in plasma can be used to diagnose, stratify risks, and predict long-term outcomes in patients with ACS.

Objective: To analyze the clinical characteristics of patients with type 2 diabetes mellitus (T2DM) with acute coronary syndrome (ACS), the global registry of acute coronary events (GRACE)score,the thrombolysis in myocardial infarction (TIMI) score and clinical prognosis. Method: The study was a retrospective one-center observational study, continuous inclusion of 600 ACS patients diagnosed by coronary angiography in our hospital from October 2018 to July 2019.Collect general clinical data, laboratory examination results, imaging data and interventional treatment data of all patients. Were divided into: T2DM with ACS group (group DA) and non-T2DM with ACS (group NDA) according to whether or not they were associated with T2DM. According to the GRACE、TIMI score, the two groups were divided into high risk group, middle risk group and low risk group.All patients underwent coronary angiography to calculate the number of vascular lesions and Gensini scores. Design questionnaire, after discharge to 2 groups of patients by telephone or outpatient follow-up average of 10 months, statistics of the occurrence of MACE events. Result: Among the 600 patients included in the study ,362 were male (60.3%) and 238 were female (39.7%) with mean age (64.7±10.3) years. The baseline data showed that the G、TG、UA、CR levels were higher in the DA group than in the NDA group; the proportion of men was lower than in the NDA group. The results of coronary angiography showed that the Gensini score of DA group was higher than that of NDA group, and the proportion of single lesion was lower than that of NDA group. The binary Logistic regression analysis suggested that age and CRP were independent risk factors for MACE events in patients with T2DM. GRACE risk stratification showed that the proportion of high risk group in DA group was significantly higher than that in NDA group, and there was no significant difference between low and middle risk group. TIMI risk stratification showed that the proportion of high risk group in DA group was significantly higher than that in NDA group, while the proportion of low and middle risk group was lower than that in NDA group. The ROC curve shows that the area (AUC) below the ROC curve that GRACE、TIMI score predicted the occurrence of MACE events in patients with T2DM and ACS was 0.707 and 0.586. Conclusion: Patients with T2DM and ACS had higher clinical risk stratification than without T2DM. GRACE score compared with the TIMI score had better predictive value for the occurrence of MACE events after discharge of T2DM with ACS patients.

Two new polycyclic polyprenylated acylphloroglucinols (PPAPs) possessing a rare benzoyl substituted bicyclo[3.2.1]octane core, hyperxylones A (1) and B (2), along with three new dearomatized isoprenylated acylphloroglucinols (DIAPs), hyperxylones C - E (3-5), were isolated from the roots of Hypericum beanii. The structures of 1-5 were determined by high-resolution electrospray ionization mass spectroscopy (HRESIMS) and 1D/2D nuclear magnetic resonance (NMR) spectroscopic analyses, gauge-independent atomic orbital (GIAO) NMR calculations, and electronic circular dichroism (ECD) calculations. Compounds 1 and 2 were biomimetically semi-synthesized starting from 5 and 4, respectively, enabling the correct stereochemical assignment of 5 and 4. Moreover, compounds 1 and 2 showed anti-nonalcoholic steatohepatitis (NASH) activity by inhibiting lipid deposition in L02 cells; compounds 3 and 5 exhibited nitric oxide (NO) inhibitory activity in lipopolysaccharides (LPS)-induced RAW264.7 cells.

With the emerging of smart metering around the world, there is a growing demand to analyse the residential energy usage. In this paper, we propose a Deep Neural Network (DNN)-based approach for non-intrusive load monitoring (NILM), which can achieve effective and efficient estimation of individual appliance usage according to a single main meter reading in a non-intrusive manner. Considering practical situations, two training methods are provided. The first training approach is fully supervised learning, which requires a ground truth of label, indicating the state of the appliance (ON/OFF), to build a prediction model. The second training approach is semi-supervised learning, leading to better performance by F-Measure metric while only requiring some more unlabelled training data. Experimental results on the low-sample rate REDD dataset demonstrate the superior performance of our proposed DNN-based method compared with Hidden Markov Model (HMM)based and Graph Signal Processing (GSP)-based approaches.

Subjected to external lateral excitations, large-amplitude sloshing may take place in propellant tanks, especially for spacecraft in low-gravity conditions, such as landers in the process of hover and obstacle avoidance during lunar soft landing. Due to lateral force of the order of gravity in magnitude, the amplitude of liquid sloshing becomes too big for the traditional equivalent model to be accurate. Therefore, a new equivalent mechanical model, denominated the “composite model”, that can address large-amplitude lateral sloshing in partially filled spherical tanks is established in this paper, with both translational and rotational excitations considered. The hypothesis of liquid equilibrium position following equivalent gravity is first proposed. By decomposing the large-amplitude motion of a liquid into bulk motion following the equivalent gravity and additional small-amplitude sloshing, a better simulation of large-amplitude liquid sloshing is presented. The effectiveness and accuracy of the model are verified by comparing the slosh forces and moments to results of the traditional model and CFD software.

Liquid sloshing of a large amplitude is prone to take place under excitation, especially when the gravitational intensity becomes small. A composite equivalent mechanical model is established to efficiently and accurately predict the slosh forces and moments exerted on a spherical propellant tank in a spacecraft. The modeling is based on an assumption of the dynamic equilibrium position of the liquid, which changes with time-dependent tank motion. Equivalent gravity is defined to describe the dynamic equilibrium position and act as a restoring force during sloshing. Sloshing of the liquid is assumed to take place around the dynamic equilibrium position. The validity of the modeling method is verified through numerical examples. Good stability and the ability to accurately predict the dynamic response under the violent sloshing of the composite model are revealed.

OBJECTIVE: To explore the effects of simvastatin on vascular endothelial cell apoptosis and Bcl-2 protein expression in the aorta in a rat model of atherosclerosis. METHODS: Thirty-six rats were randomized into control group (n=10), atherosclerosis model group (n=13) and simvastatin intervention group (n=13). In the latter two groups, rat models of atherosclerosis were established by intraperitoneal injection of vitamin D3 combined with high-fat feeding for 6 weeks, and the control rats were fed with regular diet. In the intervention group, the rats were further fed with high-fat diet with daily simvastatin treatment for 4 weeks. After the treatments, the pathological changes and plaque in the thoracic aorta were observed, and the expression of Bcl-2 protein was detected with immunohistochemistry. TUNEL assay was used to determine the apoptosis index (AI) of the vascular endothelial cells. RESULTS: Compared with that in the control group, Bcl-2 protein expression in the aorta of atherosclerotic rats was significantly decreased (P < 0.05); simvastatin treatment obviously increased the expression of Bcl-2 protein in atherosclerotic rats (P < 0.05) to a level similar to that in the control group. The AI was the highest in the model group (P < 0.05) and comparable between the control and simvastatin treatment group. CONCLUSION: The therapeutic effect of simvastatin against atherosclerosis is probably mediated by up-regulation of Bcl-2 protein, which inhibits vascular endothelial cell apoptosis in rats with aortic atherosclerosis.
