Background: Toxocara infection is one of the most neglected tropical diseases that has remained undiagnosed in developing countries. The disease leads to many signs and symptoms such as fever, coughing or wheezing, belly pain, hepatomegaly, loss of appetite, rash, lymphadenopathy, red and painful eyes, strabismus, blurred or cloudy vision, and the like. Objective: The present study aimed at investigating the seroprevalence and other demographic parameters of toxocariasis among hospitalized patients diagnosed with coronavirus disease 19 (COVID-19) in Karaj district, Alborz province, Iran. Materials and Methods: This study was undertaken between March and September 2021. Ninety-two sera samples obtained from COVID-19 patients and 56 seemingly healthy individuals were tested for anti-Toxocara immunoglobulin G via an immunological assay (enzyme-linked immunosorbent assay, ELISA). The demographic characteristics of the participants were collected through a questionnaire. Results: The findings indicated that during the study period, the seroprevalence of toxocariasis in the case group was significantly higher than in the control group (15.2% vs. 3.6%). Among the 92 seropositive COVID-19 patients, 35.7% (5/14) were males and 64.3% (9/14) were females. No deaths were observed in the Toxocara-positive individuals with COVID-19. Conclusion: Toxocariasis is a worldwide zoonotic parasitic infection in developing countries. The findings of the present study revealed the importance of Toxocara spp. infection screening in COVID-19 patients, and simple empirical treatment can be life-saving in such positive cases.

Background: Echinococcus granulosus is one of the most important pathogens in medicine. This parasite is able to cause a hydatid cyst in humans, which causes various human and financial losses annually. Objective: This study focused on determining the polymorphism of antigen B (AgB) of E. granulosus in the human and animal samples of Alborz province. Materials and Methods: In this study, 50 samples of the hydatid cyst fluid (HCF), 10 human samples, and 40 sheep samples were collected, then the DNA was extracted from the samples. During the polymerase chain reaction (PCR) process, the corresponding fragment was replicated in the AgB gene. Subsequently, PCR-single-strand conformation polymorphism (SSCP) was performed to investigate genetic diversity. Results: All 50 isolates used in this study showed the AgB in 350 bp after conducting PCR. In addition, all isolates demonstrated similar band patterns after the PCR-SSCP assay. Conclusion: In 50 isolates, one banding pattern was observed after PCR-SSCP, while no polymorphism was detected in any of the isolates.

Background: Toxocariasis is a parasitic disease with global distribution. Many paratenic or transport hosts have been identified as the sources of infection, including vertebrates such as birds and humans. The definitive hosts of Toxocara are dogs and cats whose parasites live in their guts. Hence, the diagnosis of the disease in these animals is important. Objective: The present study was performed using the loop-mediated isothermal amplification (LAMP) method to diagnose toxocariasis. Materials and Methods: In the present study, 60 samples of pet feces were collected and subsequently evaluated by direct smear, formalin-ether, and LAMP. Results: Toxocara infection in dogs by microscopic and formal ether methods and LAMP method was 10% and 13.3%, respectively, while feline infection with this parasite was not observed either microscopically or molecularly. Conclusion: The present findings revealed that pet dogs can be considered as a source of infection for humans. Therefore, it is recommended that animal owners receive the necessary training to prevent the risks of transmitting zoonotic diseases.

Background: Giardia and Cryptosporidium are common parasitic protozoa that cause acute intestinal infections in children. These two parasites are mostly found in aquatic environments, including raw water, wastewater, and even treated water. Objective: The present study aimed to examine parasitic contamination of drinking water resources by cysts and the oocyst of Giardia and Cryptosporidium in Alborz province, Iran. Materials and Methods: Water samples from three rivers and seven randomly-selected wells of Alborz province were examined using Sheather, formol-ether, and immuno-fluorescence assay (IFA) techniques. The prepared slides were examined with optical and fluorescence microscopes. Results: IFA technique revealed that 28% of the wells were contaminated with both parasites. It was also shown that all rivers’ drainage basins were contaminated with Cryptosporidium parasite, while 66% of rivers’ drainage basins were contaminated with Giardia parasite. Conclusion: The results showed that water resources of Alborz province contained Giardia cysts and Cryptosporidium oocysts, which required health care officials to pay serious attention to treating drinking water.

Toxocariasis is a zoonotic parasitic infection with important public health considerations. The aim of the study was to assess the prevalence of anti-A cross-sectional study involving 540 owners of dogs and cats was conducted between July - December 2020. A questionnaire administered by direct interviews was used to collect socio-demographic information and data on associated risk factors. Blood samples were collected and tested by indirect enzyme-linked immunosorbent assay (ELISA).The overall sero-prevalence of toxocariasis among the 540 participants was 16.7% (90 of 540). When participants included in the sample were classified by age, those aged 10-29 years demonstrated higherGiven the findings and the progressive impact of toxocariasis in public health and its high prevalence in developing countries, including Iran, measures should be taken to inform the public about zoonoses and eliminate their putative transmission.

Introduction and objective Toxocariasis, predominantly caused by Toxocara canis , is a common zoonotic parasitosis worldwide. Toxocara infection is a cause of vision impairment and blindness. The presented study investigates the frequency of antibodies against Toxocara among uveitis patients and the epidemiological factors associated with disease. Material and methods Fifty-four patients with uveitis and 59 healthy subjects were studied. Anti- Toxocara antibodies status was determined in all serum samples using enzyme linked immunosorbent assay (ELISA), and seropositive samples analyzed by Western blot (WB) technique. Results The frequency of Toxocara canis infection was found to be significantly higher in uveitis patients, compared to healthy controls by the use of ELISA test, being 14.8% and 1.7%, respectively. From 8 seropositive samples, 5 (62.5%) patients exhibited Toxocara immunoglobulin G (IgG) antibodies in response to Western blot, whereas in the control group, none were detected positive by Western blot. No significant difference was found between pet owners, nor between different places of residence. The seroprevalence to Toxocara among uveitis patients was significantly related to gender, age and medical diagnosis. The highest prevalence was found in patients with posterior uveitis (27.8%). Conclusions Anti- Toxocara antibody titers are associated with the risk of vision impairment -uveitis. The risk factor associated with Toxocara exposure identified in this study warrants further investigation.

Hydatidosis is one of the most important zoonotic parasitic diseases caused by the larval stage of Echinococcus granulosus which causes great health and economic losses. The aim of this study was to use the sequencing method to evaluate genotypes of E. granulosus isolated from humans and bovines using mitochondrial cytochrome c oxidase subunit 1 (cox1) gene. The samples were taken in the East Azerbaijan Province, Northwest Iran. Overall, 26 hydatid cyst samples (10 human and 16 cattle isolates) were collected. DNA extraction was taken from the protoscoleces of human and germinal layer of bovine samples. PCR was performed using the mitochondrial cytochrome c oxidase subunit 1(cox1) gene, and then it was sequenced. Sequences were analyzed for identification of their genotypes. All 16 bovine isolates were recognized as G1 genotypes (sheep strain) and G1B subtypes. Out of ten human host samples, seven isolates were G1B subtypes, and three samples were identified as G3 genotypes. The results of this study showed that G1 and especially G1B are the predominant genotype and subtype in humans and cattle in Northwest Iran.

Background As the daily number of coronavirus infection disease 19 (COVID19) patients increases, the necessity of early diagnosis becomes more obvious. In this respect, we aimed to develop a serological test for specifically detecting anti-SARS-CoV2 antibodies. Methods We collected serum and saliva samples from 609 individuals who work at TBZMED affiliated hospitals in Tabriz, Iran, from April to June of 2020. Real-time PCR technique was used to detect SARS-CoV-2 genome using specific primers. An enzyme linked immunosorbent assay (ELISA) test was designed based on virus nucleocapsid (N), spike (S) and its receptor binding domain (RBD) protein, and the collected sera were subjected to IgM and/or IgG analysis. Result Real-time PCR results showed that 66 people were infected with the SARS-CoV-2. Our designed ELISA kit showed 93.75% and 98% of sensitivity and specificity, respectively. In this study, 5.74% of participants had specific IgG against RBD, whereas the percentage for IgM positive individuals was 5.58%. Approximately the same results were observed for S protein. The number of positive participants for NP increased further, and the results of this antigen showed 7.38% for IgG and 7.06% for IgM. Conclusion The ELISA test beside real-time PCR could provide a reliable serologic profile for the status of the disease progress and early detection of individuals. More importantly, it possesses the potential to identify the best candidates for plasma donation according to the antibody titers.

Background: Cystic echinococcosis is a main zoonotic infection. It can cause serious clinical problems for human health around the world. Genotypic specification of Echinococcus granulosus in human is important due to control and prevention programs. Objective: In this investigation, genetic characteristics of human isolates of E. granulosus in Karaj, Iran, were studied. Materials and Methods: In this review, 3 isolates of surgically removed hydatid cysts were obtained from patients in Shahid Madani hospital, Karaj, Iran in 2014. DNA was extracted from the protoscolex of the cyst, and polymerase chain reaction (PCR) assay was done on the COX1 gene. Results: DNA fragments were sequenced and the results were aligned and analyzed. Among the isolates, 3 (100%) were E. granulosus (G1) strain. Conclusion: The G1 genotype was the most superior strain from human isolates of hydatid cyst in Karaj.

Leishmaniasis is a major public infectious disease caused by the genus Leishmania. No effective drug or vaccination strategy for leishmaniasis has been designed yet. Several intracellular Leishmania antigens have been recognized to serve in vaccination, ensuring long-lasting protection against Leishmania infection. Lipophosphoglican 3 (LPG3) as a member of the heat shock protein 90 family involves in the synthesis of lipophosphoglycan (LPG) and implicates in parasite virulence. Regarding the immunological properties of LPG3 particularly its N-terminal fragment, it would be considered as a favourable adjuvant in Leishmania vaccination.

Background: Toxoplasmosis is a worldwide neglected tropical and sub-tropical infection caused by Toxoplasma gondii (T. gondii). Most of the previous studies on the seroprevalence of T. gondii in Iran have been done at provincial level and on specific populations, including pregnant females. Socioeconomic parameters are associated with the prevalence of this disease. In this study, the authors evaluated the presence of anti-T. gondii antibodies and the related risk factors in Iranian adolescents. Methods: This was a cross-sectional study on serum samples from the third Iranian national school-based survey (the CASPIAN III study), which included 10- to 18-year-old students. Participants were selected by multistage sampling from 27 Iranian provinces. In this study, serum samples of 882 adolescents from 16 provinces were randomly selected and examined for IgG and IgM antibodies against T. gondiiby theEnzymeLinked Fluorescent Assay (ELFA).Demographicandsocioeconomic factors related to T. gondii infection were gathered using the global school-based health survey (GSHS) questionnaire. Results: The overall T. gondii IgGandIgMseropositivitywas56.3 (95 CI: 53.4 to 59.2)and3.7 (95CI: 2.7 to 4.9), respectively. In multivariate logistic regression model, family size was statistically associated with seroprevalence of anti T. gondii IgG. Living in crowded households (households with more than 4 people vs. households with less than 4 people) increased the risk of seropositivity of T. gondii (OR: 1.40, 95 CI: 1.10 to 1.99). Conclusions: The results of this study indicate that about 40 of Iranian adolescents have not had contact with the T. gondii, thus the risk of congenital toxoplasmosis might be high in young females. Also, household size was the main factor associated with T. gondii infection. Preventive strategies and health education in Iranian adolescents are recommended. © 2017, Pediartric Infections Research Center.

Echinococcus granulosus as an etiologic agent of hydatid cyst is one of the most important zoonotic helminthes in the world that causing enormous economic and health losses. The aim of this study was to evaluate genotype of E. granulosus isolated from sheep using mitochondrial cytochrome c oxidase subunit 1 (cox1) gene and sequencing method in East Azerbaijan province, northwest of Iran. Nineteen sheep hydatid cyst samples were collected. Genomic DNA was extracted from protoscoleces using commercial DNA extraction kit. Mitochondrial cox1 region was amplified by polymerase chain reaction (PCR) and all isolates were sequenced. Afterward, sequences were analyzed for determination of genotypes by related software. G1 (94.73 %) and G3 (5.27 %) genotypes were identified from the isolates which out of 19 hydatid cysts, 17 samples were G1B, 1 sample G1D and the other one had G3 genotype. Results of this study indicate that common sheep strain (G1); especially G1B is the dominant subtype of E. granulosus in East Azerbaijan province.

BACKGROUND Treatment of vivax malaria with primaquine prevents the risk of relapse. This study was designed to assess the efficacy of 8 weeks of primaquine treatment in prevention of relapse in patients with vivax malaria in south and south-east Iran by SSCP-PCR and sequencing. METHODS A total of 163 symptomatic vivax malaria cases were followed up in Hormozgan and Sistan, Baluchestan provinces in south and south-east Iran between December 2008 and December 2011. DNA was extracted from primary and secondary positive samples. A variation region of PvMSP-1 gene was selected and amplified by PCR. The obtained fragments were processed in polyacrylamide gel for single-strand conformational polymorphism (SSCP) and then sequenced. RESULTS Among 145 patients treated with chloroquine plus primaquine who completed the study period, two patients (1.4%) experienced a secondary infection after the initial episode of Plasmodium vivax. The comparison between primary and secondary isolates by SSCP indicated different banding patterns and electrophoretic mobility. Alignment of nucleotide sequences between pair primary and secondary isolates revealed dissimilar homology. Secondary isolates of both patients were considered as reinfection. Five of the 18 cases (28%) treated with chloroquine only revealed secondary infection. Analysis of nucleotide sequences and SSCP patterns indicated the relapse in all of them. CONCLUSION This survey indicates that intake of primaquine, 0.75 mg/kg, weekly for 8 consecutive weeks, is effective for the prevention of relapse in vivax cases in Iran.

Leishmania major is the main causal agent of cutaneous leishmaniasis (CL) that remains a serious public health concern in many tropical and subtropical countries. A long-lasting protective vaccine against leishmaniasis remains as a medical unmet need. Lipophosphoglycan 3 (LPG3) is one of the class II LPG genes from HSP90 family involved in the host immune responses. The aim of the present study is to investigate the capability of recombinant LPG3 (rLPG3) to induce Th1, Th2, Th17 responses. The results showed that rLPG3 in moderate and high concentrations significantly induced expression of Th1 lineage-specific transcription factor (T-bet) and cytokine (IFN-γ)(P0.05). Moreover, the Th1-stimulating effect of rLPG3 was confirmed by significant induction of IFN-γ secretion from treated T cells (P0.01). However, no significant effect of rLPG3 on Th2 and Th17 lineage cells was observed even in high concentration. Our findings demonstrate that rLPG3 induces Th1, but not Th2 and Th17, lineage responses. Further studies are needed to investigate adjuvant properties of rLPG3 for leishmania therapy.

High-mobility group protein two (HMGA2), a nonhistone nuclear-binding protein and its downregulators; vimentin, matrix metallopeptidase-9 (MMP-9), and E-cadherin are shown to contribute to tumor progression and metastasis. Thus, in this study, we checked simultaneous delivery of HMGA-2 siRNA and the anticancer drug doxorubicin to enhance the anticancer treatment effects. For this purpose, we used MTT assay and real-time polymerase chain reaction (RT-PCR). Our results showed that dual delivery of Dox and HMGA-2 siRNA by trimethyl chitosan (TMC) significantly inhibited breast cancer cells growth. Additionally, the delivery of siRNA significantly silenced HMGA-2, vimentin, and MMP9 mRNAs, but led to overexpression of E-cadherin mRNA.

A protective response against tetanus toxin and toxoid demands efficient specific T cell and B cell responses. Tetanus neurotoxin (TeNT), a 150 kDa polypeptide, is the main cause of tetanus disease. TeNT consists of two structurally distinct chains, a 50 kDa N-terminal light (L) and a 100 kDa C-terminal heavy (H) chain. C-terminal heavy (H) chain (fragment C) has two sub-domains named as proximal HCN and carboxy sub-domain or HCC. Beside neural binding property, HCC has been recently found as an immunodominant module of TeNT. In the present study, we investigated the effects of recombinant HCC (rHCC) on the expression of lineage specific transcription factors and secretion of a panel of functional cytokines including IFN-γ, IL-4, and IL-17 from purified human T cells. Our results revealed that T-bet transcript level, as TH1 specific transcription factor, was significantly increased in the cells treated with 10 and 20 µg/ml of rHCC following 48 h treatment(p0.05). Treated purified human T cells with rHCC showed significant increase in IFN-γ mRNA level and cytokine secretion, but not IL-4 and IL-17, following 48 h treatment. In conclusion, our results showed that treatment of T cells with r HCC resulted in development of Th1 lineage phenotype, which might lead to a specific and protective antibody mediated response against tetanus toxin.
