Fire blight, caused by the bacterial pathogen Erwinia amylovora, is a highly destructive disease of apple and pear. Because the apple tree gets systemically infected with E. amylovora and eventually dies, E. amylovora is a considerably important pathogen in the orchard that requires long-term management. In addition, it is crucial to prevent the spread of the pathogen by expeditious diagnosis. In this study, via comparative approaches to the genome sequences of the strains of various Erwinia spp., we designed specific primers targeting a hypothetical gene that is single copy and located in the chromosomal DNA of E. amylovora. This primer set specifically amplified the DNA of E. amylovora but no other bacteria, including E. pyrifoliae, Pectobacterium spp., Pantoea spp., and Dickeya chrysanthemi. Furthermore, the SYBR Green-based real-time PCR using the primer set allowed accurate estimation of the population of E. amylovora. Developing a rapid and accurate diagnostic method using the novel primer set enables effective defense against pathogen spread through continuous monitoring and quick response.

The present study describes a SYBR Green real-time quantitative (q) PCR assay to detect Erwinia pyrifoliae in plants. E. pyrifoliae, first described in South Korea, is a phytopathogenic bacterial species in the genus Erwinia. In particular, specific detection, quantitation, and identification of E. pyrifoliae is still challenging, as symptoms resulting from its colonization of Asian pear blossoms are very similar to those caused by E. amylovora. E. pyrifoliae has biochemical, phenotypic, and genetic properties similar to those of E. amylovora. Moreover, other Erwinia species, including E. tasmaniensis and E. billingiae, are also detected by currently available molecular methods and with traditional methods as well. Therefore, in this study, previously published genome sequences of the genera Erwinia and Pantoea were compared to exploit species-specific genes for use as improved qPCR targets to detect E. pyrifoliae. In silico analyses of the selected gene and designed primer sequences, in conjunction with bio-SYBR Green real-time qPCR, confirmed the robustness of this newly developed assay. Consequently, the bio-SYBR Green real-time qPCR-based protocols developed here can be used for rapid and specific detection of E. pyrifoliae. During the nationwide survey of fire blight, the typical shoot blight symptoms were found on Korean mountain ash (Sorbus alnifolia) which was located near an orchard that produced fire blight on pear trees in Eumseong, Korea, May 2021. To identify the causal agent, we progressed isolation from the symptomatic leaves and shoots. Two white and mucoid colonies were isolated into the pure culture. Two isolates were identified as Erwinia amylovora according to the colony–polymerase chain reaction (PCR) with amsB primers and the phylogenetic tree using 16S rRNA sequences. To test of pathogenicity of two isolates, we inoculated immature pear fruits and understock of apple. We observed necrosis and oozes on immature pear fruits and shoot blight resulting in necrosis on apple shoots six days after inoculation. Colonies were recovered from the inoculated pears and apples, and identity was confirmed through colony PCR for amsB genes. To our knowledge, E. amylovora was first reported on Korean mountain ash native to South Korea.

Penthorum chinense (Penthoraceae) is a threatened South Korean medicinal plant used to treat alcohol-related diseases and prepare cosmetics in traditional Chinese medicine. Wetlands, the native habitat of this plant, have been decreasing owing to damage caused by landisation and urban development in recent years. In this study, we considered the effect of temperature and gibberellic acid treatment on P. chinense seed germination to improve conservation outcomes of the species. The optimum germination temperature was investigated using constant and alternating temperature treatments (60 conditions). In addition, gibberellic acid (GA3 ) treatment was performed to overcome physiological dormancy. We determined the germination (GP, %) and the number of days to reach 50% maximum germination (T50). The optimum temperature regime was 7°C / 23°C: GP was 95%, and T50 was 15 days. The GA3 treatment increased the GP of P. chinense seeds from 28 to 94% and decreased the T50 value from 14.6 to 6.3 days at concentrations above 40 mg L–1. After the GA3 treatment, the growth of radicles was delayed or inhibited. For the germination of P. chinense seeds, a diurnal temperature difference of ≥ 15°C is required.

Polysomnography (PSG) is the gold-standard for sleep apnea and hypopnea syndrome (SAHS) diagnosis. Because the PSG system is not suitable for long-term continuous use owing to the high cost and discomfort caused by attached multi-channel sensors, alternative methods using a non-contact sensor have been investigated. However, the existing methods have limitations in that the radar-person distance is fixed, and the detected apnea hypopnea (AH) event cannot be provided in real-time. In this paper, therefore, we propose a novel approach for real-time AH event detection with impulse-radio ultra-wideband (IR-UWB) radar using a deep learning model. 36 PSG recordings and simultaneously measured IR-UWB radar data were used in the experiments. After the clutter was removed, IR-UWB radar images were segmented by sliding a 20-s window at 1-s shift, and categorized into two classes: AH and N. A hybrid model combining the convolutional neural networks and long short-term memory networks was trained with the data, which consisted of class-balanced segments. Time sequenced classified outputs were then fed to an event detector to identify valid AH events. Therefore, the proposed method showed a Cohen’s kappa coefficient of 0.728, sensitivity of 0.781, specificity of 0.956, and an accuracy of 0.930. According to the apnea-hypopnea index (AHI) estimation analysis, the Pearson’s correlation coefficient between the estimated AHI and reference AHI was 0.97. In addition, the average accuracy and kappa of SAHS diagnosis was 0.98 and 0.96, respectively, for AHI cutoffs of ≥ 5, 15, and 30 events/h. The proposed method achieved the state-of-the-art performance for classifying SAHS severity without any hand-engineered feature regardless of the user’s location. Our approach can be utilized for a cost-effective and reliable SAHS monitoring system in a home environment.

Introduction Shift work is known to be associated with sleep disturbance and circadian misalignment and may contribute to alterations in the brain implicated in emotion dysregulation. In this study, we examined 1) neural correlates of emotion dysregulation, including emotional reactivity and regulation in shift workers and 2) relationships between neural correlates of emotion dysregulation, sleep disturbance, and circadian misalignment. Methods Fifty-six rotating shift workers (SW, 42 females, 30.80 ± 6.94 years old) who had non-standard shift schedules and 52 healthy controls (HC, 36 females, 31.69 ± 7.86 years old) who did not work as shift schedules participated in this study. Both SW and HC completed self-reported measures of sleep and mood problems, and wrote sleep diary and wore actigraphy to assess sleep quality and circadian rhythms. They also underwent one night polysomnography and were scanned while they were performing the emotion regulation task. We analyzed group differences in mood, sleep, circadian rhythms, and neural bases of emotional reactivity and regulation, and examined relationships between these variables. Results SW showed significant differences in actigraphy-estimated sleep (i.e., lower sleep efficiency) and circadian rhythms (e.g., greater motor activity [M10] and lower interdaily stability) compared to HC. SW also exhibited increased anterior insula (AI) activation in response to negative pictures, but reduced activation in the dorsomedial prefrontal cortex (dMPFC) and AI-dMPFC functional connectivity during emotion regulation compared to HC. AI activation and AI-dMPFC functional connectivity were significantly associated with sleep disturbance (i.e., sleep diary-estimated sleep onset latency) and circadian misalignment (e.g., M10 and interdaily stability). Furthermore, a mediation analysis revealed that shift work was associated with increased motor activity during the most active 10-hour period, which then contributed to increased AI activation in response to negative pictures. Conclusion Our findings suggest that shift work may contribute to sleep disturbance and circadian misalignment, and neural alterations associated with emotional reactivity and regulation. Furthermore, the mediation result suggests that increased motor activity along with misaligned circadian rhythm may serve as a pathway, through shift work contributes to heightened emotional reactivity to negative information. Study Objectives This study investigated the altered neural function involved in emotional interference and its role in linking sleep disturbance and depressive/anxiety symptoms in rotating shift workers. Methods Sixty rotating shift workers and 61 controls performed the emotional Stroop task in three blocks (emotional-related, sleep-related, and neutral words) during functional magnetic resonance imaging (fMRI) assessments. Sleep disturbance and depressive/anxiety symptoms were assessed using self-report measures and sleep diaries. Actigraphy was used to assess the sleep and circadian variables. fMRI scans were performed to compare brain activation during the emotional Stroop task. The proposed moderating models were tested using the PROCESS macro in SPSS software. Results A significant condition effect on reaction time was detected. Regardless of the group, reaction times were longer in the negative emotional word and sleep-related conditions than in the neutral word condition. Whole-brain analysis revealed that rotating shift workers show greater neural activation in the left dorsolateral prefrontal cortex (DLPFC) compared with controls while performing the emotional Stroop task with negative emotional words. Sleep disturbance was more strongly associated with depressive symptoms when activation of the left DLPFC was higher during the emotional Stroop task with negative words. Conclusions The left DLPFC may play important roles in increased sensitivity to emotional information, possibly due to circadian misalignment, and has moderating effects on the association between sleep disturbance and depressive symptoms in rotating shift workers. These findings will help to identify possible brain regions where interventions can be performed to correct sleep and mood problems in rotating shift workers.

As platform owners interact with end users and complementors, their demand-side characteristics and performance affect the overall value creation of ecosystems. This research investigated how the emergence of popular complements on a mobile communication platform impacts the usage of other complements by the platform’s end users and how platform owners can benefit from such demand spillovers. Using individual user-level app usage data, we empirically demonstrated how the presence of a popular app alters the demand structure of a platform through changes in the usage of other apps operating within it. The findings reveal that popular app adoption by users increases the app usage, excluding the usage of popular apps, only within the platform offering a popular app, supporting the existence of positive spillovers from popular complement adoption on a platform. Such positive within-platform spillovers occur through the increased usage of both existing and new apps, with stronger positive spillovers for new apps. These findings imply that platform owners can reap benefits by coordinating the launch of new complements and the promotion of less-known counterparts to end users with the emergence of a popular app. All these results shed light on how platform owners can manage their complements and create value from popular complements.

STUDY OBJECTIVES: In the present study, factors associated with sleep perception were identified by comparing clinical characteristics and polysomnographic variables between insomnia patients with negative and positive sleep state misperception (NSSM and PSSM, respectively). METHODS: Self-reported and objective sleep measures were retrospectively collected, including the Pittsburgh Sleep Quality Index (PSQI), Beck Depression Inventory, and a questionnaire on “morning feeling” after nocturnal polysomnography in 150 patients with insomnia. Based on the misperception index (MI), participants were classified into NSSM (MI > 0, n = 115) and PSSM (MI < 0, n = 35) groups. RESULTS: The PSSM group had more N3 sleep on nocturnal polysomnography than the NSSM group (P = .002). The NSSM group showed a higher PSQI score (P < .001), longer self-reported sleep-onset latency (SOL) (P = .001), and a greater SOL discrepancy (P = .001). Self-reported feelings of tiredness and morning awakenings in the morning were higher in the NSSM group (P = .029 and P = .038). The MI negatively correlated with a proportion of N3 sleep (P = .005) and positively correlated with PSQI (P < .001), morning awakenings (P = .01), self-reported SOL (P < .001), and SOL discrepancy (P < .001) in patients with insomnia. Multiple regression analysis showed that N3 sleep, PSQI, and morning awakenings were significantly associated with MI in patients with insomnia. CONCLUSIONS: The proportion of slow-wave sleep and self-reported measures may be associated with perception of sleep in patients with insomnia. Objective and self-reported characteristics of patients with insomnia should be carefully evaluated and managed because they may influence the perception of sleep. Penthorum chinense Pursh is a plant belonging to the Crassulaceae family and is a representative perennial herb that grows nationwide in watersides and wetlands. This study was conducted to enhance the competitiveness of wild plant seeds and contribute to the conservation of species diversity through the evaluation of antioxidant and antidiabetic activities of P. chinense seed extracts based on extraction solvents. The total phenolics content was 19.83 mg GAE/g seeds in the 75% ethanol extract, whereas the flavonoid content was the highest (10.89 mg QE/g seeds) in the 75% methanol extract. Total phenolicss content was higher in the organic solvent extract than in the water extract. The DPPH radical scavenging activity was the highest (IC50 27.60 μg/mL) in the 75% methanol extract, and the ABTS radical scavenging activity was the highest (IC50 53.54 μg/mL) in the 75% methanol extract. Total antioxidant capacity was the highest (30.87 μmol AAE/g) in the 75% ethanol extract. The α-glucosidase-inhibitory activity was the highest (IC50 46.47 μg/mL) in the 75% ethanol extract. From the results of this study, it was confirmed that P. chinense seed extracts, which possess high phenolics content, exhibit antioxidant and antidiabetic effects, and could be used as an effective health functional food material and natural medicine.

STUDY OBJECTIVES: To assess the seasonality of restless legs syndrome (RLS) using data from the Korean national health insurance database. METHODS: We retrospectively reviewed a randomly selected sample representing 3% of the national health insurance claims database in South Korea. From this sample, we obtained the monthly numbers of patients with RLS and diagnoses from 2009 to 2016, along with prescriptions for monthly dopamine agonists and clonazepam for patients with RLS from 2009 to 2013. Total dopamine agonist and clonazepam doses were converted to levodopa-equivalent doses, and the monthly cumulative prescription dose was calculated. Cosinor analysis was used to evaluate the seasonal pattern of each variable. RESULTS: This study included 11,466 patients with RLS and their diagnoses and 4,887 prescriptions for dopamine agonists and clonazepam. There were significant seasonal patterns in the numbers of patients with RLS (P < .001) and diagnoses (P < .001), both of which peaked in August. The magnitude of the greatest difference in the number of patients with RLS between August (highest) and February (lowest) was 29.96% (95% confidence interval, 24.03–100.80), and that of the number of RLS diagnoses was 39.56% (95% confidence interval, 31.24–47.89). The cumulative prescription dose of medication showed no significant seasonality. CONCLUSIONS: Our findings suggest that the prevalence of RLS is seasonally affected, with an increase during summer. Erwinia amylovora, a causal bacterium of fire blight disease, is registered as a prohibited quarantine pathogen in Korea. To control the disease, the government should diagnose the disease, dig and bury the host trees when fire blight occurs. Fire blight was the first reported in 43 orchards of Anseong, Cheonan, and Jecheon in 2015, and 42.9 ha of host trees were eradicated. However, the disease spread to eleven cities, so that 348 orchards and 260.4 ha of host trees were eradicated until 2019. Fire blight of Asian pear occurred mainly in the southern part of Gyeonggi, and Chungnam province, on average of 29±9.2 orchards per year. And the age of the infected trees were mostly 20-30 years old. In apple trees, the disease occurred mainly in the northern part of Gyeonggi, Gangwon, and Chungbuk province, on average of 41±57.6 orchards per year, increased highly in 2018 and 2019. The age of infected apple trees were under 20 years old. Therefore, because the disease spread rapidly in young apple trees, spraying control agents to the trees in a timely manner and removing infected trees quickly are important to prevent the spread of fire blight in the orchard of immature trees.

Background: Reactor pressure vessel (RV) with internals (RVI) are activated structures by neutron irradiation and volume contaminated wastes. Thus, to develop safe and optimized disposal plan for them at a disposal site, it is important to perform exact activation calculation and evaluate the dose rate on the surface of casks which contain cut-off pieces. Materials and Methods: RV and RVI are subjected to neutron activation calculation via Monte Carlo methodology with MCNP6 and ORIGEN-S program—neutron flux, isotopic specific activity, and gamma spectrum calculation on each component of RV and RVI, and dose rate evaluation with MCNP6. Results and Discussion: Through neutron activation analysis, dose rate is evaluated for the casks containing cut-off pieces produced from decommissioned RV and RVI. For RV cut-off ones, the highest value of dose rate on the surface of cask is 6.97 × 10-1 mSv/hr and 2 m from it is 3.03 × 10-2 mSv/hr. For RVI cut-off ones, on the surface of it is 0.166 × 10-1 mSv/hr and 2 m from it is 1.04 × 10-1 mSv/hr. Dose rates for various RV and RVI cut-off pieces distributed lower than the limit except the one of 2 m from the cask surface of RVI. It needs to adjust contents in cask which carries highly radioactive components in order to decrease thickness of cask. Conclusion: Two types of casks are considered in this paper: box type for very-low-level waste (VLLW) as well as low-level waste (LLW) and cylinder type for intermediate-level waste (ILW). The results will contribute to the development of optimal loading plans for RV and RVI cut-off pieces during the decommissioning of nuclear power plant that can be used to prepare radioactive waste disposal plans for the different types of wastes—ILW, LLW, and VLLW.

We have generated a system concept for a non-destructive assay (NDA) system to certify identified isotopes and their activity concentrations for radioactive waste in a large container. Various optimization studies have evaluated the system performance in terms of the MDA (minimum detectable activity) results using the Monte Carlo simulation in conjunction with experimental studies. The proposed system consists of a total of eight HPGe (high-pressure germanium) detectors: four detectors on the top and the other four detectors on the bottom. The top and the bottom detector arrays are aligned and face each other. This detector arrangement has advantages in employing the attenuation correction as proposed by T. Chang [IEEE Trans. Nucl. Sci. NS-25, 638 (1978)]. We also found that the UFOV (useful field of view) of the detector highly influences the system efficiency, which leads to an improvement in the MDA performance. However, while the wider FOV of the detector improves the detection efficiency by allowing incoming radiation from other segmented volumes, it may suffer from non-uniform performance or increased errors in estimating an activity concentration for each segmented volume. To prevent such potential errors in employing the wider FOV, we have proposed an activity estimation algorithm, a so-called ‘fine volume reconstruction’, based on a back-projection method that estimates the activity concentration of each segmented volume. We demonstrate the feasibility of the conceptual system for use as a free release assay system for ISO containers up to a size of 2.4 × 6.0 × 1.3 m3 (W x L x H). The detector efficiency was about four times higher than those of typical commercial systems. Future studies include fine-tuning of the activity reconstruction algorithm and a validation study on various materials and non-uniform activity concentrations.

The Kori-Unit 1 nuclear power plant, which is scheduled to be decommissioned after permanent shutdown, is expected to generate large amounts of various types of radioactive waste during the decommissioning process. Among these, nuclear reactors and internal structures have high levels of radioactivity and the dismantled structure must have the proper size and weight on the primary side. During decommissioning, it is important to prepare an appropriate and efficient disposal method through analysis of the disposal status and the legal restrictions on wastes generated from the reactors and internal structures. Nuclear reactors and internal structures generate radioactive wastes of various levels, such as medium, very low, and clearance. A radiation evaluation indicates that wastes in the clearance level are generated in the reactor head and upper head insulation. In this study, a clearance waste safety evaluation was conducted using the RESRAD-RECYCLE code, which is a safety evaluation code, based on the activation evaluation results for the clearance level wastes. The clearance scenario for the target radioactive waste was selected and the maximum individual and collective exposure doses at the time of clearance were calculated to determine whether the clearance criteria limit prescribed by the Nuclear Safety Act was satisfied. The evaluation results indicated that the doses were significantly low, and the clearance criteria were satisfied. Based on the safety assessment results, an appropriate metal recycle and disposal method were suggested for clearance, which are the subject of the deregulation of internal structures of nuclear power plant.

Erwinia amylovora and Erwinia pyrifoliae cause fire blight and black-shoot blight, respectively, in apples and pears. E. pyrifoliae is less pathogenic and has a narrower host range than that of E. amylovora. Fire blight and black-shoot blight exhibit similar symptoms, making it difficult to distinguish one bacterial disease from the other. Molecular tools that differentiate fire blight from black-shoot blight could guide in the implementation of appropriate management strategies to control both diseases. In this study, a primer set was developed to detect and distinguish E. amylovora from E. pyrifoliae by conventional polymerase chain reaction (PCR). The primers produced amplicons of different sizes that were specific to each bacterial species. PCR products from E. amylovora and E. pyrifoliae cells at concentrations of 104 cfu/ml and 107 cfu/ml, respectively, were amplified, which demonstrated sufficient primer detection sensitivity. This primer set provides a simple molecular tool to distinguish between two types of bacterial diseases with similar symptoms.

Unnecessary and inappropriate laboratory testing accounts for a significant portion of waste in health care utilization. The aim of this study was to examine the diagnostic value of the anti-nuclear antibody (ANA) test by examining the rate of ANA associated rheumatic disease (AARD) diagnosis among ANA tested and ANA positive subjects and positive predictive value (PPV) of ANA test leading to AARD diagnosis in different ANA titers and different subsets of patients in 5 hospitals affiliated with a university.We retrospectively extracted data from all subjects who were tested for ANA from year 2010 to 2019. Those who were first evaluated at or referred to rheumatology were further evaluated with extraction of data including ANA titer and ultimate diagnosis. PPVs for ANA test were evaluated after stratification according to clinically relevant key parameters, such as patient age (younger65 years vs. older), sex, and requesting department.From 2010 to 2019, A total of 94,153 patients were tested for ANA, of which 13,600 (14.4% of the total) were positive. AARD was diagnosed in only 0.69% among all ANA tested patients and 4.74% among ANA positive patients. The AARD diagnosis rate of ANA positive patients varied widely from 0.1% to 8.7% by requesting department. Using cutoff values above 1:320 yielded PPVs of 15.6 and 7.8% for all AARs and systemic lupus erythematosus. The PPV was significantly higher in young age (65 years) and in women, and when it was requested from internal medicine vs other departments.AARD was diagnosed in less than 1% of all ANA tested patients in university-affiliated hospitals. This result shows that careful consideration before ordering the screening ANA is needed to improve the utility of the test for providers and patients and to reduce health costs spurred by unnecessary testing and its consequences.
