Introduction: Integrating genetic data into conservation management decisions is a challenging task that requires strong partnerships between researchers and managers. Conservation in Latin America is of crucial relevance worldwide given the high biodiversity levels and the presence of hotspots in this region. Methods: We conducted a survey across Latin America to identify gaps and opportunities between genetic researchers and conservation managers. We aimed to better understand conservation managers' points of view and how genetic research could help conservation practitioners to achieve their goals, by implementing genetic assessments that could effectively inform conservation practices. We distributed an online survey via four regional collaborating organizations and 32 focal points based in 20 Latin American countries. The target respondents were conservation managers of species or areas in Latin America. Results: We collected a total of 468 answered questionnaires from 21 Latin American countries. Most respondents (44%) were from an academic or research institution while non-academics were mainly from non-governmental institutions (30%) and government agencies (25%). Most respondents (65%) have performed or used genetic assessments in their managed area or species, either alone, in partnership, contracting someone else or using published results. For the majority of this group, the genetic results were relevant to their conservation management goals, helping to inform management decisions. Respondents that had not performed genetic assessments (35%) were mainly from the non-academic group, and their main barriers were limited access to funds, genetic lab facilities, and trained personnel to design studies and conduct lab work. Discussion: From the findings, we describe the current situation and provide a general diagnosis of the conservation-genetics gap in Latin America. We describe the gender gap, academic-practitioner co-development of conservation questions and projects, and the nationality and residency of Latin American conservation managers in relation to the countries where they work. We discuss opportunities to co-create research questions and co-develop studies based on conservation practitioners' needs. We offer recommendations for overcoming barriers to integrate genetic information into conservation actions, and advance agendas that fit the needs and realities of the highly heterogeneous, biodiverse and challenging Latin American region. Abstract Background DNA-based studies have demonstrated that avian genetic mating systems vary widely, with many species deviating from long-assumed monogamy by practicing extra-pair paternity and conspecific brood parasitism. Colonially breeding waterbirds provide interesting models in which to investigate this question because they show nesting habits proposed to promote alternative reproductive strategies. However, little is known about the genetic mating systems of this group of birds, mainly due to difficulties in obtaining genetic data from incubating adults at nests that are necessary for conducting conventional parentage studies. Here, we inferred kinship patterns among offspring in broods of three co-distributed waterbird species, Wood Stork (Mycteria americana), Roseate Spoonbill (Platalea ajaja) and Great Egret (Ardea alba egretta), to investigate genetic mating system in the absence of parental data. Results Multi-step analyses combining estimates of relatedness coefficients, formulation of relationship-hypotheses, significance testing of alternative hypotheses, and maximum-likelihood sibship reconstruction techniques revealed evidence that alternative reproductive strategies may be present in natural populations of Wood Storks and Roseate Spoonbills, whereas relatedness of co-nestlings diagnosed in the Great Egrets did not deviate from a hypothesis of genetic monogamy. Specifically, under this analytical framework, inferred kinship relationships revealed that Great Egret nests contained full-sibling nestlings (100%), with the Roseate Spoonbill (RS) and Wood Stork (WS) exhibiting proportions of half-siblings (RS: 5%) and/or unrelated nestlings (RS: 24%; WS: 70%), patterns consistent with extra-pair paternity and conspecific brood parasitism, respectively. Conclusions We provide evidence that genetic monogamy occurs in Brazilian natural breeding colonies of the Great Egret, but is not the sole reproductive strategy employed by the Wood Stork and the Roseate Spoonbill. In fact, extra-pair paternity and conspecific brood parasitism were common in the latter two species, with a combined frequency of 7.5% and 11.3% in Roseate Spoonbill and Wood Stork, respectively. Although geographically co-distributed, differences among these species may be due to variation in their life histories. From a methodological standpoint, the approach implemented here, although not free from limitations, can have broad application for analyzing systems with limited genealogical information and/or in studying similarly challenging organisms in which obtaining genetic data on complete families is problematic.

Abstract: Detecting trends in population size fluctuations is a major focus in ecology, evolution, and conservation biology. Populations of colonial waterbirds have been monitored using demographic approaches to determine annual census size (Na). We propose the addition of genetic estimates of the effective number of breeders (Nb) as indirect measures of the risk of loss of genetic diversity to improve the evaluation of demographics and increase the accuracy of trend estimates in breeding colonies. Here, we investigated which methods of the estimation of Nb are more precise under conditions of moderate genetic diversity, limited sample sizes and few microsatellite loci, as often occurs with natural populations. We used the wood stork as a model species and we offered a workflow that researchers can follow for monitoring bird breeding colonies. Our approach started with simulations using five estimators of Nb and the theoretical results were validated with empirical data collected from breeding colonies settled in the Brazilian Pantanal wetland. In parallel, we estimated census size using a corrected method based on counting active nests. Both in simulations and in natural populations, the approximate Bayesian computation (ABC) and sibship assignment (SA) methods yielded more precise estimates than the linkage disequilibrium, heterozygosity excess, and molecular coancestry methods. In particular, the ABC method performed best with few loci and small sample sizes, while the other estimators required larger sample sizes and at least 13 loci to not underestimate Nb. Moreover, according to our Nb/Na estimates (values were often ≤0.1), the wood stork colonies evaluated could be facing the loss of genetic diversity. We demonstrate that the combination of genetic and census estimates is a useful approach for monitoring natural breeding bird populations. The Western Yellow-breasted Chat ( (Deppe, 1830)) is a Neotropical migrant, with a Canadian distribution restricted to breeding populations in southern British Columbia. Given its small population size and diminishing breeding habitat, Yellow-breasted Chats are federally endangered in Canada. We used genotypic data at eight microsatellite loci to assess genetic diversity, reconstruct population structure and demographic history, and characterize genetic mating system of Yellow-breasted Chats sampled across 60 nesting sites at five locations in the Okanagan Valley ( n = 148). Microsatellite-based analyses indicated lack of significant genetic differentiation among breeding sites and no genetic evidence of population decline. Parentage assignments indicated moderate levels of extra-pair paternity, with 30.7% offspring not sired by attending males. Patterns of sibship among nestlings revealed 49.1% of the clutches were composed entirely of full-siblings, with half-siblings and unrelated nestlings present in some broods. These findings suggest that extra-pair paternity is common in Yellow-breasted Chats, similar to other avian species, and present the first evidence of conspecific brood parasitism in warblers. Our findings add to a growing body of research informing the need to establish a national park in the south Okanagan to preserve critical habitat and connect populations of species at risk.
