The understanding of cryptic species complexes with their often highly interesting biogeographical patterns is still a crucial aspect in evolutionary biology and related disciplines. Trichoptera are a group of insects particularly rich in unresolved groups. One example is the Wormaldia occipitalis species complex in which morphological studies suggest remarkable patterns of differentiation. In order to determine genetic differentiation and phylogenetic structure, one mitochondrial (COI) and two nuclear markers (CAD, wingless) were analysed for the W. occipitalis species complex around the Alps and northwards to Germany. The morphology-defined differentiation pattern was also observed at the genetic level. The morphologically well distinguishable groups W. occipitalis and W. subterranea were identified as two genetically distant monophyletic groups with about 10 % genetic divergence of the mitochondrial marker. These two taxa likely split during the Mio-Pliocene transition. Genetic analyses revealed four subgroups within W. occipitalis and three within W. subterranea. Several possible postglacial dispersal and differentiation processes are proposed. Thereby, W. occipitalis from the western Alps and individuals of W. subterranea from the eastern Alps spread towards Central Europe after the Last Glacial Maximum. Today, both species groups are sympatric and partly syntopic in the recolonised area in western Germany but apparently allopatric in their centres of origin around the Alps. The high genetic differentiation, lack of detectable genetic evidence for hybridisation, their syntopic distribution and the morphological distinctness indicate that W. occipitalis and W. subterranea are two distinct species. The genetically determined subgroups might represent subspecies.

The raising economic importance of cannabis arouses interest in positively influencing the secondary plant constituents through external stimuli. One potential possibility to enhance the secondary metabolite profile is the use of UV light. In this study, the influence of spectral UV quality at different intensity levels on photomorphogenesis, growth, inflorescence yield, and secondary metabolite composition was investigated. Three UV spectra with five different intensities were considered: L1 (UVA:B = 67:33, 4.2 W/m2), L2 (UVA:B = 94:6, 4.99 W/m2), L3_1 (UVA:B = 99:1, 1.81 W/m2), L3_2 (UVA:B = 99:1, 4.12 W/m2) and L3_3 (UVA:B = 99:1, 8.36 W/m2). None of the investigated UV treatments altered the cannabinoid profile. Regarding the terpenes investigated, light variant L3_1 was able to positively influence the terpene profile. Especially linalool (+29%), limonene (+25%) and myrcene (+22%) showed an increase, compared to the control group without UV treatment. Growth and leaf morphology also showed significant changes compared to the control. While a high UVA share increased the leaf area, a higher UVB share led to a smaller leaf area. Of the UV sources examined, only L3_1 with 1.81 W/m2 and a radiation dose of 117.3 kJ m2 d-1 is suitable for practical use in commercial cannabis cultivation. The terpene concentration for this group was in part significantly increased with constant yield and cannabinoid concentration.

IntroductionThe circular bioeconomy concept revolves around biological production cycles that reintroduce products or waste from one production system to another, aiming to maximize resource utilization while minimising environmental impact. The purpose of this study was to evaluate and compare the element flow when integrating black soldier fly larvae (BSF) production with Nile tilapia production using varying experimental fish feed.MethodsTilapia (42.5 ± 11.2 g) were reared in recirculating aquaculture systems (RAS) at 25.5°C for 10 weeks and fed equal daily rations of four experimental diets containing fishmeal (FM), poultry blood meal (PBM), black soldier fly meal (BSF) and poultry by-product meal (PM) as the single main protein source, respectively. Faeces was collected daily from settling columns installed in the RAS and subsequently fed to BSF larvae.Results and discussionThe fish exhibited the highest biomass gain when fed with FM (1,001 g) or PM (901 g). The growth was lowest for those fed with PBM (406 g). The fish fed with PBM also produced the highest amount of faeces (234 g). When the fish faeces were utilized as a substrate for rearing black soldier fly (BSF) larvae and although the biomass gain did not differ significantly, the feed conversion ratio (FCR) varied among larvae fed with different fish faeces, ranging from 8.36 to 25.04. Furthermore, the concentration of analysed elements (Al, B, Ca, Co, Cu, Fe, K, Mg, Mn, Mo, Na, P, S, and Zn) varied based on the type of fish faeces provided. Results illustrate that a modulation of BSFL elemental composition is possible through manipulating the feed provided to the fish and emphasize the importance of fish feed composition for fish performance as well as the compositional quality of the larvae reared on the resulting fish faeces. Hence, if BSF larvae production is to be harnessed for the recycling of aquaculture sludge, ingredient choice and subsequent elemental composition of fish feeds are pivotal to larvae composition.

During their time as Teaching Assistants (TAs), graduate students develop a variety of skills, knowledge, and attitudes (SKAs) based on teaching and related facilitation experiences. As TAs move on to future opportunities, their prior experiences form a foundation upon which additional teaching experience builds. Presently, there are few tools to gauge pedagogical growth during graduate student involvement as TAs in a specific post-secondary course, or as a consequence of their participation in a specialized TA training or teaching program. We identified key factors that underlie TA development and designed a new survey based on these for TAs to self-assess SKAs that they bring with them from prior experience, and those that they develop or enhance during their time as a TA. We designed a survey to test how TA experience with non-traditional roles in a First-year Seminar in Science course, SCIE113, affects SKAs that can be used in future teaching endeavours. We administered the survey to 18 current and past SCIE 113 TAs as of December 2015 (representing the complete population). The results showed that TAs with similar levels of experience shared similar skills, knowledge, and attitudes. Those TAs with the most experience had greater abilities in roles previously identified as non-traditional TA roles. Others working with graduate students can use or adapt the survey questions to investigate and stimulate the growth and development of TAs in their course or program.

The genus Erebia comprises numerous species in Europe. Due to preference of cold environments, most species have disjunct distributions in the European mountain systems. However, their biogeographical patterns may differ significantly. The Bright-eyed ringlet Erebia oeme is widespread in high-altitude grasslands of the European high mountains, hence showing a disjunct distribution pattern. Over its distribution, E. oeme shows high morphological variability indicating pronounced intraspecific differentiation. We analysed two mitochondrial (COI, Cytb) and eight nuclear markers as well as the Wolbachia surface protein-coding gene (WSP). A total of four lineages were identified: two Balkan lineages, one Slovenian lineage as well as one lineage containing all other individuals (Alps, Massif Central, Pyrenees). COI data only indicate a fifth lineage in the southern Carpathians. The region of origin of E. oeme is most likely the western Balkans. From here, E. oeme spread to the eastern Balkan area and further to the southern Carpathians as well as to Slovenia, from where it rapidly crossed the Alps to Massif Central and Pyrenees. Wolbachia was found to be highly prevalent with multiple strains, partly within one population. However, recent Wolbachia impact on the mitochondrial DNA and consequently influence on the intraspecific genetic structure was not detected. [ABSTRACT FROM AUTHOR]

Brønsted acids, such as phosphoric acids derived from chiral 1,1′‐bi‐2‐naphthol (BINOL), are important catalysts in the formation of carbon–carbon and carbon–heteroatom bonds, for example. The catalytic activity of these Brønsted acids is strongly linked to their acidity, and as such, the evaluation of compounds to determine pKa values provides insight into their catalytic activity. Herein, a 19F{1H} NMR methodology is detailed to determine the pKa of a fluorinated binaphthyl‐derived phosphinic acid, rac‐1, in acetonitrile and in the presence of a fluorinated sulfonamide reference compound (2–4). The approach was tested initially using 2 and 3, with the ΔpKa (0.08) in strong agreement with previously reported values (6.6 for 2 and 6.68/6.73 for 3). Sigmoidal curves of normalised chemical shift change (Δδ) against equivalents of the base phosphazene P1‐tBu added overlapped for 2 and 3, but in the case of rac‐1 and either 2, 3 or 4, there was significant separation. A variety of different approaches for determining the ΔpKa were compared. Values of pKa determined when the normalised Δδ was 90% were optimal for 2 and 3, whereas a normalised Δδ of 75% was optimal for 4, resulting in the pKa of rac‐1 being determined to be 8.47–8.71. Determining plant responses to hazardous air pollutants is critical in predicting food security programs and challenges in the future. This study aimed to determine the effects of various ozone levels on plant growth responses (leaf area, dry matter, and number of leaves) and biochemical quality (photopigments and glucosinolates) on Brassica campestris L. ssp. chinensis (Pak-Choi). The experiment was conducted within test chambers under different ozone concentrations (60, 150, and 240 ppb for 2 h/day). Leaf area and dry matter were negatively correlated with increasing ozone concentrations, but the number of leaves was not affected by ozone treatment. Lycopene and chlorophylls also showed the same tendency. Even if the ambient ozone concentration was only elevated for a short time, various glucosnilates (GLS) have been diversely affected. The total aliphatic GLS content was reduced. In contrast, the total indole GLS increased at the highest ozone concentration, and the aromatic GLS significantly increased and then decreased as the ozone concentration level increased. These results provide evidence of the strong effect of ozone stress on the plant quality of Pak-Choi with respect to certain secondary plant metabolites. These findings provide an understanding of elevated ozone effects in urban horticulture sites on the growth and metabolite profiling of Brassica plants.

Microbial communities drive biogeochemical cycles through networks of metabolite exchange that are structured along energetic gradients. As energy yields become limiting, these networks favor co-metabolic interactions to maximize energy disequilibria. Here we apply single-cell genomics, metagenomics, and metatranscriptomics to study bacterial populations of the abundant "microbial dark matter" phylum Marinimicrobia along defined energy gradients. We show that evolutionary diversification of major Marinimicrobia clades appears to be closely related to energy yields, with increased co-metabolic interactions in more deeply branching clades. Several of these clades appear to participate in the biogeochemical cycling of sulfur and nitrogen, filling previously unassigned niches in the ocean. Notably, two Marinimicrobia clades, occupying different energetic niches, express nitrous oxide reductase, potentially acting as a global sink for the greenhouse gas nitrous oxide.

For the first time, the recently proposed “talkative power” paradigm combining power electronics and communication engineering is extended by channel coding and advanced receiver design aspects. In this concept, the pulses that control the duty cycle of switched-mode power converters are simultaneously used for data transmission by modifying the pulse phase, pulse frequency, pulse position or pulse width. The data sequence is embedded into the switching signal, similar to random modulation. The data can be used for example in point-to-point links, inside multi-port power converters, and for communication in networked power grids. As an application example a buck converter is studied. Its output voltage is numerically calculated for arbitrary two-level switching signals. The corresponding system model serves as a common framework for switching waveform design, receiver design, and performance analysis. The data sequence is represented by a small deterministic output voltage ripple. Various techniques to improve the data rate and the robustness of the communication are introduced, including line coding, a derivation of the optimum receiver in the sense of maximum-likelihood detection, and forward error correction coding as an option. Although emphasis is on a buck converter, the concept is applicable to other power converter topologies as well.

The growth and nutritional profile of the black soldier fly larvae (BSFL) is usually investigated and compared when the larvae feed on substrates that differ in the chemical composition as well as physical properties. This study compares BSFL growth on substrates that differ primarily in physical properties. This was achieved by using various fibres in the substrates. In the first experiment, two substrates with 20% or 14% chicken feed were mixed with three fibres (cellulose, lignocellulose, or straw). In the second experiment, the growth of BSFL was compared with a 17% chicken feed substrate that additionally contained straw with different particle sizes. We show that the substrate texture properties values did not influence the BSFL growth, but the bulk density of the fibre component did. The substrate mixed with cellulose led to higher larvae growth over time in comparison to substrates with higher bulk density fibres. BSFL grown on the substrate mixed with cellulose reached their maximum weight in 6 days instead of 7. Neither the fibres nor the nutrient level changed the crude protein content of BSFL and the values ranged between 33.5% and 38.3%, but an interaction between the fibre and nutrient level was observed. The size of straw particles in the substrates influenced the BSFL growth and led to a 26.78% difference in Ca concentration, a 12.04% difference in Mg concentration, and a 35.34% difference in P concentration. Our findings indicate that the BSFL-rearing substrates can be optimised by changing the fibre component or its particle size. This can improve the survival rate, reduce the cultivation time needed to reach the maximum weight, and alter the chemical composition of BSFL.

The proper treatment of cannabis agricultural wastes can reduce the environmental impact of its cultivation and generate valuable products. This study aimed to test the potential of cannabis agricultural wastes as a substrate for the rearing of black soldier fly larvae (BSFL) and yellow mealworms (MW). In the case of BSFL, replacing the fibre component (straw) in the substrate with the hemp waste can increase the nutritional value of the substrate and led to bigger larvae. The bigger larvae had lower P and Mg, and higher Fe and Ca. Crude protein also varied based on the size of larvae and/or the content of protein in the initial substrate, which was boosted by replacing straw with hemp material. No other cannabinoids than cannabidiolic acid (CBDA), cannabigerolic acid (CBGA), and cannabidiol (CBD) were found in significant amounts in the larvae. In the case of MW, the larvae grew less on the hemp material in comparison to wheat bran. Replacing wheat bran with the hemp material led to smaller larvae with higher Ca, Fe, K, and crude protein content, but lower Mg and P values. No cannabinoids were detected in the MW fed with the hemp material.

Herbal preparations of willow bark (Salix cortex) are available in many countries as non-prescription medicines for pain and inflammation, and also as dietary supplements. Currently only little information on toxicity and drug interaction potential of the extracts is available. This study now evaluated the effects of two Salix cortex extracts on human hepatocyte-like HepaRG cells, in view of clinically relevant CYP450 enzyme activity modulation, cytotoxicity and production of reactive oxygen species (ROS). Drug metabolism via the CYP450 enzyme system is considered an important parameter for the occurrence of drug-drug interactions, which can lead to toxicity, decreased pharmacological activity, and adverse drug reactions. We evaluated two different bark extracts standardized to 10 mg/ml phenolic content. Herein, extract S6 (S. pentandra, containing 8.15 mg/ml total salicylates and 0.08 mg/ml salicin) and extract B (industrial reference, containing 5.35 mg/ml total salicylates and 2.26 mg/ml salicin) were tested. Both Salix cortex extracts showed no relevant reduction in cell viability or increase in ROS production in hepatocyte-like HepaRG cells. However, they reduced CYP1A2 and CYP3A4 enzyme activity after 48 h at ≥25 μg/ml, this was statistically significant only for S6. CYP2C19 activity inhibition (0.5 h) was also observed at ≥25 μg/ml, mRNA expression inhibition by 48 h treatment with S6 at 25 μg/ml. In conclusion, at higher concentrations, the tested Salix cortex extracts showed a drug interaction potential, but with different potency. Given the high prevalence of polypharmacy, particularly in the elderly with chronic pain, further systematic studies of Salix species of medical interest should be conducted in the future to more accurately determine the risk of potential drug interactions.

Abstract Fentanyl, also known as ‘jackpot’, is a synthetic opiate that is 50–100 times more potent than morphine. Clandestine laboratories produce analogues of fentanyl, known as fentalogues to circumvent legislation regarding its production. Three pyridyl fentalogues were synthesized and then hyperpolarized by signal amplification by reversible exchange (SABRE) to appraise the forensic potential of the technique. A maximum enhancement of ‐168‐fold at 1.4 T was recorded for the ortho pyridyl 1H nuclei. Studies of the activation parameters for the three fentalogues revealed that the ratio of ligand loss trans to hydride and hydride loss in the complex [Ir(IMes)(L)3(H)2]+ (IMes=1,3‐bis(2,4,6‐trimethylphenyl)imidazole‐2‐ylidene) ranged from 0.52 to 1.83. The fentalogue possessing the ratio closest to unity produced the largest enhancement subsequent to performing SABRE at earth's magnetic field. It was possible to hyperpolarize a pyridyl fentalogue selectively from a matrix that consisted largely of heroin (97 : 3 heroin:fentalogue) to validate the use of SABRE as a forensic tool.

Fieldwork is an important part of undergraduate degrees in Geography and has been shown to be an effective pedagogic strategy. Fieldtrips are often to remote locations, both residential and shorter day trips. For institutions, field trips can be costly in terms of money and staff time and difficult to timetable. Some students may have difficulty attending due to caring commitments or employment. For some, going to a novel environment to learn new skills can be overwhelming. At Askham Bryan College a "Field and Environmental Techniques" module for Foundation Degree level students, ran in weekly two hour sessions, for 24 sessions. These were formatted to suit the College timetable and to fit with students other commitments. It resulted in a structure re-think, moving from individual lectures and longer fieldtrips to an integration of theory and fieldwork in short sessions utilizing the campus environment. Student surveys revealed this structure benefited learning as they could link theory with practice and it prepared them for carrying out future fieldwork in novel locations. In addition, students highlighted the social benefits of the module. Social aspects of fieldwork are regularly reported as a benefit of residential trips, but it was an unexpected benefit of this module.

Willow bark is traditionally used for pharmaceutical purposes. Evaluation is so far based on the salicylate content, however, health promoting effects of extracts might be attributed to the interaction of those salicylates with other compounds, which support and complement their action. So far, only S. purpurea, S. daphnoides, and S. fragilis are included in pharmaceutical extracts. Crossing with other species could result in a more diverse secondary metabolite profile with higher pharmacological value. With the help of targeted inter- and intraspecific crossing, new chemotypes were generated, whereby nine different Salix genotypes (S. alba, S. daphnoides, S. humboldtiana, S. lasiandra, S. nigra, S. pentandra, S. purpurea, S. x rubens, S. viminalis) were included in the study. Based on substances known for their health promoting potential and characteristic for Salix (selected phenolic compounds including salicylates), a targeted metabolomics analysis and clustering of 92 generated Salix clones was performed revealing four different cluster/chemoprofiles. In more specific, one group is formed by S. daphnoides clones and inter- and intraspecific hybrids, a second group by S. viminalis clones and inter- and intraspecific hybrids, a third group generally formed by S. alba, S. pentandra, S. x rubens, and S. lasiandra clones and hybrids, and a fourth group by S. purpurea clones and inter- and intraspecific hybrids. Clustering on the basis of the selected phenolic compounds can be used for identifying Salix clones with a different compound profile. New combinations of secondary plant metabolites offer the chance to identify Salix crosses with improved effects on human health.

This study investigated undergraduate chemistry students' career aspirations and how these vary from one educational system to another in different geographic regions. The participants of this study were undergraduate chemistry students from various institutions located in Australia, New Zealand and the UK. The study took place in the form of an international cross-sectional survey. Findings of this study show that undergraduate students choose to study chemistry predominantly because they are interested in it or because they like it. This study also found that, whereas undergraduate students mainly have plans of pursuing a career that uses chemistry, they seem to be aware that a chemistry course can lead to many career options and as such only a few have chosen to study it as a route to a specific career. The findings of this study may be important in informing policies to attract and retain students in chemistry courses.
