Abstract

Thiosulphate dehydrogenase was distilled to homogeneity from Thiobacillus acidophilus by a method involving ammonium sulphate precipitation, hydrophobic interaction chromatography, anion-exchange chromatography and gel filtration. The enzyme was seen to activate the oxidation of thiosulphate to tetrathionate by employing potassium ferricyanide as an artificial electron acceptor. The molecular mass of the native enzyme was obtained by gel filtration and was calculated to be 102 plus or minus 4.2 kDa. It was concluded that sulphite prevented enzyme activity.

Abstract

The filamentous fungus Aspergillus niger is widely exploited by the fermentation industry for the production of enzymes and organic acids, particularly citric acid. We sequenced the 33.9-megabase genome of A. niger CBS 513.88, the ancestor of currently used enzyme production strains. A high level of synteny was observed with other aspergilli sequenced. Strong function predictions were made for 6,506 of the 14,165 open reading frames identified. A detailed description of the components of the protein secretion pathway was made and striking differences in the hydrolytic enzyme spectra of aspergilli were observed. A reconstructed metabolic network comprising 1,069 unique reactions illustrates the versatile metabolism of A. niger. Noteworthy is the large number of major facilitator superfamily transporters and fungal zinc binuclear cluster transcription factors, and the presence of putative gene clusters for fumonisin and ochratoxin A synthesis.

Abstract

Cell-free extracts of Thiobacillus acidophilus catalysed the quantitative conversion of trithionate (S3O62-) to thiosulphate and sulphate. A continuous assay for quantification of experimental results was based on the difference in absorbance between trithionate and thiosulphate at 220 nm. Trithionate hydrolase was purified to near homogeneity from cell-free extracts of T. acidophilus. The molecular masses of the native enzyme and the subunit were 99 kDa (gel filtration) and 34 kDa (SDS/PAGE). The purified enzyme has a pH optimum of 3.5–4.5 and a temperature optimum of 70 °C. Enzyme activity was stimulated by sulphate. The stimulation of the enzyme activity by sulphate was half maximal at a concentration of 0.23 M. The Km for trithionate is 70 μM at 30 °C and 270 μM at 70°C. Enzyme activity was lost after 36 days at 0°C, 27 days at 70°C; but after 97 days at 30°C, 40% of the initial activity was still present. The enzyme activity was inhibited by mercury chloride, N-ethylmaleimide, thiosulphate and tetrathionate. Tetrathionate S4O62- was not hydrolysed by trithionate hydrolase. [ABSTRACT FROM AUTHOR]

16. Partially oxidized polycyclic aromatic hydrocarbons show an increased bioavailability and biodegradability

Abstract

Polycyclic aromatic hydrocarbons have a low water solubility and tend to adsorb on soil particles, which both result in slow bioremediation processes. Many microorganisms, known for their ability to degrade polycyclic aromatic hydrocarbons, only partially oxidize these compounds. White rot fungi, for instance, convert polycyclic aromatic hydrocarbons to more water soluble and bioavailable products. Polycyclic aromatic hydrocarbon metabolites were more readily mineralized by natural mixed bacterial cultures, like activated sludge and soil, than the parent polycyclic aromatic hydrocarbon compounds. These results suggest that sequential breakdown by white rot fungi followed by indigenous bacteria leads to an effective polycyclic aromatic hydrocarbon bioremediation process.

17. Metabolism of tetrathionate in Thiobacillus acidophilus

Abstract

Cell-free extracts of Thiobacillus acidophilus catalysed the stoichiometric conversion of tetrathionate to thiosulphate, sulphur and two protons. The pH optimum of the enzyme activity was 3.0 and its temperature optimum 40°C. The enzyme was unstable at 30 and 40°C, at which its activity decreased to zero within 100 and 20 h, respectively. Enzyme activity was not affected by incubation for 1 week on ice or by freezing and thawing of the extract. The Km for tetrathionate was 0.3 mM. Enzyme activity was stimulated by ammonium sulphate up to a concentration of 1M. The results indicate that trithionate hydrolase cannot account for the observed conversion of tetrathionate.
