Modelling complex interactions involving climatic features, socio-economic vulnerability or responses, and long impact transmissions is associated with substantial uncertainty. Physical climate storylines are proposed as an approach to explore complex impact transmission pathways and possible alternative unfoldings of event cascades under future climate conditions. These storylines are particularly useful for climate risk assessment for complex domains, including event cascades crossing multiple disciplinary or geographical borders. For an effective role in climate risks assessments, development guidelines are needed to consistently develop and interpret the storyline event analyses. This paper elaborates on the suitability of physical climate storyline approaches involving climate event induced shocks propagating into societal impacts. It proposes a set of common elements to construct the event storylines. In addition, criteria for their application for climate risk assessment are given, referring to the need for storylines to be physically plausible, relevant for the specific context, and risk-informative. Apart from an illustrative gallery of storyline examples found in literature, three examples of varying scope and complexity are presented in detail, all involving the potential impact on European socio-economic sectors induced by remote climate change features occurring far outside the geographical domain of the European mainland. The storyline examples illustrate the application of the proposed storyline components and evaluate the suitability of the criteria defined in this paper. It thereby contributes to a rigorous design and application of event-based climate storyline approaches.

INTRODUCTION: Autologous T-cells transduced to express a chimeric antigen receptor (CAR) directed against CD19 elicit high response rates in relapsed or refractory (r/r) B-cell non-Hodgkin lymphoma (B-NHL). However, r/r B-NHL remissions are durable in fewer than half of recipients of second-generation CAR T-cells. Third-generation (3G) CARs employ two costimulatory domains, resulting in improved CAR T-cell efficacy in vitro and in animal models in vivo. This investigator-initiated, phase I dose escalation trial, termed ENABLE, will investigate the safety and preliminary efficacy of WZTL-002, comprising autologous T-cells expressing a 3G anti-CD19 CAR incorporating the intracellular signalling domains of CD28 and Toll-like receptor 2 (TLR2) for the treatment of r/r B-NHL. METHODS AND ANALYSIS: Eligible participants will be adults with r/r B-NHL including diffuse large B-cell lymphoma and its variants, follicular lymphoma, transformed follicular lymphoma and mantle cell lymphoma. Participants must have satisfactory organ function, and lack other curative options. Autologous T-cells will be obtained by leukapheresis. Following WZTL-002 manufacture and product release, participants will receive lymphodepleting chemotherapy comprising intravenous fludarabine and cyclophosphamide. A single dose of WZTL-002 will be administered intravenously 2 days later. Targeted assessments for cytokine release syndrome and immune cell effector-associated neurotoxicity syndrome, graded by the American Society Transplantation and Cellular Therapy criteria, will be made. A modified 3+3 dose escalation scheme is planned starting at 5×104 CAR T-cells/kg with a maximum dose of 1×106 CAR T-cells/kg. The primary outcome of this trial is safety of WZTL-002. Secondary outcomes include feasibility of WZTL-002 manufacture and preliminary measures of efficacy. We review the natural occurrence of tail duplications and tail bifurcations in amphibians as well as experimental studies that induced tail duplications. For natural populations, we found 10 publications that mention tail duplications or bifurcations in a total of 24 individual larval amphibians belonging to 15 species, mainly from Europe and South America. Nineteen publications describe the incidence for 34 individual postlarval urodeles from 12 species from North America, Europe, and Asia. Here we add three new observations: a subadult female Triturus dobrogicus with a duplicated tail and an adult female Triturus carnifex with a bifurcated tail from Hungary, and a tadpole of Hyla arborea with a bifurcated tail tip from Greece. The causes of tail duplications in wild amphibians remain unknown. Similar tail anomalies have been obtained in the laboratory when parts of the chorda dorsalis were destroyed, when tar is injected into the tail, or when the lumbar region is irradiated.

Objectives: An immense body of knowledge has been created by establishing various job-exposure matrices (JEMs) to assess occupational exposures in community- and industry-based cohort studies. These JEMs could be made available to occupational epidemiologists using knowledge-sharing technologies, thereby saving considerable amounts of time and money for researchers investigating occupation-related research questions. In this paper, the authors give an example of how a detailed JEM can be easily transformed into a job-specific module (JSM) for use in community-based studies. Methods: OccIDEAS is operationalised as a web-based software, combining the use of JSMs with an individual expert exposure assessment to assess occupational exposures in various industries according to a set of predefined rules. The authors used a JEM focusing on endocrine-disrupting chemicals from a German study on testicular cancer in the automobile industry to create a JSM in OccIDEAS. Results: The JEM was easily translated into OccIDEAS requiring about 50 h of work by an epidemiologist familiar with the German JEM to learn about the OccIDEAS structure, establish the required set of exposure rules and to translate the JEM into OccIDEAS. Language did not represent an obstacle for translation either. To make the data available in an international context, an interpreter had to translate the German tasks and exposures after they were coded into OccIDEAS. Conclusions: JEMs which are constructed based on identifying tasks that determine exposure can be easily transformed into a JSM. Occupational epidemiologists are invited to contribute to the international scope of OccIDEAS by providing their previously established JEMs to make existing data on occupational exposures widely available to the epidemiological community.
