1. Evidence of Non-microtubule Spindle Forces in Mesostoma ehrenbergii Spermatocytes

Abstract

We tested conclusions reached in previous experiments in which Mesostoma spermatocyte chromosomes moved rapidly to a pole in the absence of microtubules: after 10 μM nocodazole (NOC) depolymerized metaphase spindle microtubules, kinetochores from each of the 3 bivalents detached from the same pole and rapidly moved to the other pole, at speeds averaging 37.7 μm/min. with some as high as 100 μm/min. We concluded that these very fast movements were due to non-microtubule forces arising from a spindle matrix. However, since the chromosomes stretch out before detaching, there is tension in the chromosomes from the stretch. Thus the movements of detached kinetochores conceivably might be due to recoil from the tension, though we argued against this possibility (Fegaras and Forer, 2018a). In this article we test whether recoil causes the movements. We cut bivalents into 2 pieces, using a femtosecond laser, before addition of NOC. When 1 bivalent was severed, all kinetochores moved to one pole in 12/15 cells; when 2 bivalents were severed, all kinetochores moved to one pole in 4/6 cells; and when all 3 bivalents were severed all kinetochores moved to one pole in 3/9 cells. The bivalent "halves" moved rapidly, with average speeds of 47 μm/min, velocities that are not significantly different from those in cells without any laser-cut bivalents (p > 0.05). Since kinetochores move at the same speeds whether they are part of bivalents or not, NOC-induced chromosome movements are not due to recoil from tension along the full-length bivalent, strongly supporting the idea that non-microtubule forces move chromosomes in Mesostoma spermatocytes.

8. Regeneration of Rhynchomesostoma rostratum

Abstract

Die Arbeit für diese Masterthesis dient zur Grundlagenforschung im Fachbereich der Regeneration. Die Spezies im Mittelpunkt dieser Forschung ist Rhynchomesostoma rostratum, ein freilebender räuberischer Platyhelminth aus der Ordnung der Rhabdocoela. Ebenso wurde während dieser Arbeit eine bisher unbestimmte Art der Gattung Mesostoma entdeckt. Dieses Mesostoma konnte anhand genetischer Daten in dieser Gattung eingeordnet werden. Eine genauere Bestimmung bedarf weiterer Untersuchung im Bereich der Histologie. Von R. rostratum und seinen weiteren Verwandten innerhalb der Rhabdocoela wird angenommen, dass sie kaum Regenerationsfähigkeiten besitzen. Durch eigene Versuche konnte gezeigt werden, dass dies nicht zutreffend ist. Daraufhin wurden weitere Regenerationsexperimente durchgeführt, die zusätzlich mit molekularen Methoden zur Färbung von Neoblasten, auch Stammzellen genannt, kombiniert wurden. Auf diese Weise konnten weitere Erkenntnisse zur Regenerationfähigkeit von Rhynchomesostoma rostratum geschaffen werden. Amputationsversuche zeigten, dass eine vollständige Regeneration des Rostrums am anterioren Körperende möglich ist, sofern das Gehirn nicht beschädigt wurde. Der Bereich posterior des Pharynx ist nur bedingt regenerierbar, da die Geschlechtsteile nicht neu gebildet werden und die Körperform nur zu einem kleinen Teil wieder hergestellt wird. Die Methode zur Färbung der Neoblasten musste auf diesen Organismus abgestimmt werden. Erfolgreiche Färbungen zeigten schließlich Stammzellen im mittleren Körperbereich um den Pharynx herum entlang der Körperwand. Es befinden sich keine Stammzellen im Kopf- und Schwanzbereich sowie im Pharynx. Kombinierte Experimente zur Regeneration mit Färbung der Neoblasten verliefen nicht erfolgreich und konnten daher keinen Aufschluss auf deren Mitwirken bei der Regeneration liefern. Die Möglichkeit, dass Neoblasten für die Regeneration dennoch verantwortlich sind, besteht weiterhin. Dies könnte durch weitere Versuchen mit der final, The work for this master’s thesis serves as basic research in the field of regeneration. The species at the center of this research is Rhynchomesostoma rostratum, a free-living predatory platyhelminth from the order of Rhabdocoela. Also, during this work a previously undetermined species of the genus Mesostoma was discovered. This Mesostoma could be classified within this genus by genetic data. A more precise determination requires further investigation in the field of histology. It is assumed that R. rostratum and its other relatives within the Rhabdocoela have little regenerative capacity. By own experiments it could be shown that this is not true. Subsequently, further regeneration experiments were conducted, which were additionally combined with molecular methods for staining neoblasts, also called stem cells. In this way, more insights of the regeneration capacity of Rhynchomesostoma rostratum could be obtained. Amputation experiments showed that a complete regeneration of the rostrum at the anterior end of the body is possible, provided that the brain is not damaged. The area posterior to the pharynx can only be regenerated to a limited extent, as the genitals could not be reformed and the body shape is only restored to a small extent. The method for staining the neoblasts had to be adapted to this organism. Successful staining finally showed stem cells in the middle part of the body along the body wall surrounding the pharynx. There are no stem cells in the head and tail area or in the pharynx. Combined experiments on regeneration with staining of the neoblasts have not yet been successful and could therefore not provide information of their involvement in regeneration. The possibility that neoblasts are nevertheless responsible for regeneration still exists. This could be shown by further experiments with the finally developed method and freshly collected animals. 9. Methods for rearing Mesostoma ehrenbergii in the laboratory for cell biology experiments, including identification of factors that influence production of different egg types.

Abstract

Mesostoma ehrenbergii spermatocytes are uniquely useful to study various aspects of cell division. Their chromosomes are large in size and few in number, with only three bivalent and four univalent chromosomes. During prometaphase, bipolar bivalents oscillate regularly to and from the poles for 1-2 hours. The univalents remain at the poles but occasionally move from one pole to the other. In addition, a precocious cleavage furrow forms during prometaphase and remains partially constricted until anaphase. Attempts to rear these animals indefinitely in laboratory conditions, however, have been mostly unsuccessful because of their reproductive strategy. M. ehrenbergii are hermaphroditic flatworms that can produce viviparous offspring (termed S eggs) and/or diapausing eggs (termed D eggs) and they follow either one of two reproductive patterns: (1) they first form S eggs and following the delivery of these eggs produce D eggs, or (2) they only produce D eggs. When only D eggs are formed, which is common under laboratory conditions, the stocks die out until the D eggs hatch, which is irregular and creates unpredictable wait times. Consequently, in order to maintain M. ehrenbergii stocks to study their spermatocytes, we examined various factors that might influence egg-type production. Feeding them daily and keeping them at 25°C favours S egg production. Currently, our cultures have reached the 53rd generation. The article discusses a study to determine the occurrence of Mesostoma tetragonum or Turbellaria in the Deepar wetlands of Assam, India. It relates the origin of the freshwater turbellarians and the growth of their population. Results of the study reveal that there were no previous record of occurrence of Mesostoma tetragonum in Deepar wetland as had appeared from other works.

Abstract

Abstract: It generally is assumed that cleavage furrows start ingression at anaphase, but this is not always true. Cleavage furrows are initiated during prometaphase in spermatocytes of the flatworm Mesostoma, becoming detectable soon after the spindles achieve bipolarity. The furrows deepen during prometaphase, but ingression soon arrests. After anaphase the pre-existing furrow recommences its ingression and rapidly cleaves the cell. Such “precocious” furrowing also commonly occurs in diatoms and other algae. The position of the “precocious” cleavage furrow changes when there are changes in the distribution of chromosomes. Each of the 4 unipolarly-oriented univalent chromosomes moves to a pole at the start of prometaphase but later in prometaphase may move to the opposite pole. The furrow position adjusts during prometaphase according to the numbers of univalents at the two poles: when there are two univalent chromosomes at each pole the furrow is symmetrical at the spindle equator, but when there are unequal numbers at the poles the furrow shifts 2–3μm toward the half-spindle with fewer univalents. Nocodazole causes spindle microtubules to disappear. After addition of nocodazole, bivalents become detached from one pole and move toward the other, which causes the furrow to shift 2–3μm toward the pole with fewer chromosomes. Furrow positioning thus is sensitive to the positioning of chromosomes in the spindle and furrow positions change in the absence of spindle microtubules. [Copyright &y& Elsevier]

Abstract

The epidermis of the free-living typhloplanids Mesostoma viaregginum and M. productum (Mesostominae) is described. In both species, the epidermis has polarized cells with nuclei located at the basal part of the cell, whereas mitochondria are in the apical one. The epidermis is entirely covered by microvilli and locomotory cilia anchored in the cytoplasm by vertical and horizontal rootlets. Rootlets exhibit distinct length and periodic structure in the two species. Furthermore, in each species vertical and horizontal rootlets possess different periodic structure. The pattern of termination of microtubules in epidermal cilia is described for the first time in the Typhloplanida; central microtubules shift along one axonemal side, doublets 1 and 6–9 lose their microtubule B, and gradually peripheral doublets become singlets. Finally, an electron-dense material caps the tip of the cilia. This pattern of termination closely resembles that of Temnocephalida, Kalytorhynchia, and Dalyelliida examined so far, but differences exist. [ABSTRACT FROM AUTHOR]

Abstract

We investigated the population dynamics of Copidodiaptomus numidicus (Copepoda: Calanoida) and Thermocyclops dybowskii (Copepoda: Cyclopoida) in Castelo-do-Bode Reservoir (Portugal). Both species in the reservoir were regulated mainly by food availability during the summers of 1993 and 1994. C. numidicus was always more abundant than T. dybowskii in the reservoir. C. numidicus produced two generations during the sampling period of both years whereas T. dybowskii produced two and three generations in 1993 and 1994, respectively. Because of high temperatures and low rainfall in 1994, the reservoir was more eutrophic than in 1993. Higher clutch size and a higher percentage of ovigerous females suggest that both species were benefiting from better feeding conditions in 1994. Despite this, population growth was reduced in 1994 relatively to 1993. Vertebrate predation and predation by copepods seem to have been minor factors in explaining this decline. However, the presence of Mesostoma sp. in the reservoir may have contributed to the reduction of both copepod populations during specific periods in 1994 although the calanoid was more affected than the cyclopoid by this predation. 14. Freshwater Rhabdocoela (Platyhelminthes) from Ephemeral Rock Pools from Botswana, with the Description of Four New Species and One New Genus.

Abstract

Four new species of freshwater rhabdocoel flatworms from ephemeral rock pools in southeastern Botswana are described and discussed. Two of them, Syringoplana kolasai n. gen. n. sp. and Mesostoma thamagai n. sp. belong to the Typhloplanidae Graff, 1905. The unique construction of the excretory system is the main characteristic of S. kolasai . M. thamagai can be separated from other Mesostoma Ehrenberg, 1837 species by the presence of a bundle of eosinophilic glands at the transition from oviduct to seminal receptacle. The other two taxa, Gieysztoria isoldeae n. sp. and G. faubeli n. sp. belong to the Dalyelliidae Graff, 1905. G. isoldeae is characterised by the presence of four separate hollow spines in the male atrium, which are connected to two accessory glandular organs. G. faubeli can be separated from other Gieysztoria Ruebush and Hayes, 1939 species by the detailed construction of the stylet. Apart from these two species the occurrence of an unidentified Microdalyellia Gieysztor, 1938 species is mentioned. [ABSTRACT FROM AUTHOR]

Abstract

Mesostoma maculatum Hofsten, 1916 Locality. Temporary pools on Mount Kavul and Mount Shekhanya, Lower Galilee. Material. Many individuals. Live observations, squash preparations, whole-mounted individual, and two sagittally-sectioned individuals deposited in the Department of Zoology at Tel Aviv University, Israel, Catalogue Number VR- 25080 and VR- 25081. Ecological features. Mesostoma maculatum lives in lentic waters, springs and brooks. In Israel, it was collected in temporary pools of Mount Kavul and Mount Shekhanya (March 2000, March 2001, January 2002) and in a temporary pool in Keren Carmel (March 2002). It tends to be negatively associated with larval Salamandra salamandra infraimmaculata, probably because Salamandra is an efficient predator of this flatworm; we have observed high predation rates in the laboratory (Eitam and Blaustein, unpublished data). Mesostoma maculatum is known from Finland (Luther, 1963), and Sweden (Steinb��ck, 1932). It was reported from Israel for the first time by Eitam et al. Abstract

We assessed the predatory impact of Mesostoma sp. (Rhabditophora) on hatching and survival of Branchipodopsis wolfi (Anostraca) under laboratory conditions. The hatching percentages of resting eggs of B. wolfi were negatively influenced to a significant degree by the presence of Mesostoma sp. or the medium in which the worms were kept before, but only when prey and predator originated from the same pool. This relationship was also not observed when eggs of the American anostracan Thamnocephalus platyurus were exposed to the Turbellaria from Botswana. There was, however, no indication of predation of B. wolfi eggs by Mesostoma sp., either by eating them totally or by sucking them out. When the same number of anostracans of different ages was offered simultaneously to Mesostoma sp., the 1- and 3-day-old specimens were eaten significantly faster than the 5-day-old juveniles and the adult B. wolfi. We conclude that Mesostoma sp. has a potentially strong influence on the population dynamics and life history characteristics of B. wolfi in temporary aquatic systems. Additionally, the feeding behaviour of Mesostoma sp. was described and the ultra structure of its rostral area was investigated.
