Abstract

The long-standing model-independent annual modulation effect measured by DAMA deep underground at Gran Sasso Laboratory with different experimental configurations is summarized and perspectives will be highlighted. DAMA/LIBRA-phase2 set-up, $\simeq$ 250 kg highly radio-pure NaI(Tl) confirms the evidence of a signal that meets all the requirements of the model independent Dark Matter annual modulation signature at high C.L.; the full exposure is 2.86 ton $\times$ yr over 22 annual cycles. The experiment is currently collecting data in the DAMA/LIBRA-phase2 empowered configuration with an even lower software energy threshold. Other recent claims are shortly commented.

Contains fulltext : 244566.pdf (Publisher’s version ) (Closed access) BACKGROUND AND AIMS: The case-mix of patients with intestinal failure due to short bowel syndrome (SBS-IF) can differ among centres and may also be affected by the timeframe of data collection. Therefore, the ESPEN international multicenter cross-sectional survey was analyzed to compare the characteristics of SBS-IF cohorts collected within the same timeframe in different countries. METHODS: The study included 1880 adult SBS-IF patients collected in 2015 by 65 centres from 22 countries. The demographic, nutritional, SBS type (end jejunostomy, SBS-J; jejuno-colic anastomosis, SBS-JC; jejunoileal anastomosis with an intact colon and ileocecal valve, SBS-JIC), underlying disease and intravenous supplementation (IVS) characteristics were analyzed. IVS was classified as fluid and electrolyte alone (FE) or parenteral nutrition admixture (PN). The mean daily IVS volume, calculated on a weekly basis, was categorized as 3 L/day. RESULTS: In the entire group: 60.7% were females and SBS-J comprised 60% of cases, while mesenteric ischaemia (MI) and Crohn' disease (CD) were the main underlying diseases. IVS dependency was longer than 3 years in around 50% of cases; IVS was infused ≥5 days/week in 75% and FE in 10% of cases. Within the SBS-IF cohort: CD was twice and thrice more frequent in SBS-J than SBS-JC and SBS-JIC, respectively, while MI was more frequent in SBS-JC and SBS-JIC. Within countries: SBS-J represented 75% or more of patients in UK and Denmark and 50-60% in the other countries, except Poland where SBS-JC prevailed. CD was the main underlying disease in UK, USA, Denmark and The Netherlands, while MI prevailed in France, Italy and Poland. CONCLUSIONS: SBS-IF type is primarily determined by the underlying disease, with significant variation between countries. These novel data will be useful for planning and managing both clinical activity and research studies on SBS.

Abstract

In questo capitolo si propone una definizione di infanzia a partire da una chiave di lettura relazionale. Il punto di osservazione proposto cioè ritiene che i bambini siano soggetti autonomi e in grado di assumere una posizione attiva, a condizione di vivere in contesti familiari e sociali in cui gli adulti, con cui sono in relazioni significative, rendano loro possibile uno spazio protetto, adeguato e stimolante, sostenuti ed aiutati da politiche pubbliche promozionali e sussidiarie. Per questo vengono analiticamente presentate, in una prospettiva di rete, familiare e comunitaria, le principali relazioni in cui sono inseriti i minori, familiari: genitori, fratelli, sorelle, nonni e sociali: educatori e gruppo dei pari. Uno sguardo, seppure veloce, è riservato alle reti virtuali che sempre più occupano l’esperienza quotidiana anche dei più piccoli. Particolare attenzione è rivolta ai servizi per la prima infanzia e alla rete virtuosa che può organizzarsi intorno ad essi per il benessere dei più piccoli, senza dimenticare il dramma della povertà educativa, condizione che coinvolge quote significative di minori, in termini di scarsità/inadeguatezza di risorse materiali e immateriali, anche nei Paesi altamente sviluppati. Inoltre, viene offerta una rapida presentazione dei principali provvedimenti normativi, a tutela dell’infanzia, che sono stati realizzati in Italia a, partire dalla Convenzione del 1989.

Abstract

The objective of this work was to determine whether the random regression model using linear splines (RRMLS) is suitable to estimate the genetic parameters for productive and reproductive traits of a multiple-breed dairy cattle population, as well as to investigate the effect of the genetic group of the progeny on the genetic merit of the sire. The multiple-trait model (MTM) and the RRMLS with one knot fitted for every genetic group were used to obtain the genetic parameters. Records of 1/2 Holstein + 1/2 Gyr (1/2HG), 5/8 Holstein + 3/8 Gyr (5/8HG), and 3/4 Holstein + 1/4 Gyr (3/4HG) crossbreed dams were considered. The RRMLS showed better fitting. The additive and residual variances estimated by the MTM and the RRMLS were similar. Heritability varied from 0.20 to 0.33 for age at first calving, from 0.09 to 0.22 for first lactation length, and from 0.15 to 0.35 for first lactation 305-day milk yield, according to the genetic composition of the dams. The RRMLS is suitable to estimate the genetic parameters for productive and reproductive traits of multiple-breed dairy cattle populations. Abstract

An assay, specifically optimized to evaluate the anti-HIV activity of antiretrovirals by flow cytometry analysis, is described. As widely used anti-HIV agents, zidovudine (AZT), abaca- vir (ABC), 20,30-dideoxyinosine (DDI), lamivu-dine (3TC), nevirapine (NVP), and efavirenz (EFV), and as drugs of recent approval raltegra- vir (RAL), etravirine (ETR), and rilpivirine (RPV), were utilized as reference drugs. HIV-1 NL4-3 virus was prepared by transfection of HEK293T cells with purified plasmid DNA and quantified by p24 antigen-capture assay. For infection, CEM-GFP cells were exposed to vehicle or to several concentrations of the drugs for 2 hr at 37°C before HIV-1 NL4-3 was added to each sample. The adsorption was prolonged for 3 hr at 37°C. After 72 hr of incubation, HIV-induced GFP expression in infected CEM-GFP cells was assessed by flow cytometry analysis and expressed as % positive cells. For comparison, p24 production in supernatants was assessed by a commercial ELISA kit. On the basis of IC50 values, the anti-HIV activity, as assayed by this method, was EFV>3TC>AZT>NVP>DDI> ABC and ETR>RPV>RAL. The comparison between the IC50 values calculated through flow cytometry and p24 production revealed overlapping results, showing that the opti- mized protocol of CEM-GFP infection with HIV NL4-3 is a suitable method to perform quanti-tative, rapid and low-expensive screening tests to evaluate the in vitro effect of new candidate anti-HIV drugs.

Abstract

The UV-inactivated herpes simplex virus 1 (HSV-1) and glycoprotein D (gD) of HSV-1 have been shown to activate nuclear factor kappaB (NF-kappaB) in U937 cells, but mechanisms involved in this activation have not been elucidated. Here we report that: (i) UV-inactivated HSV-1 induced an increased NF-kappaB activation in cells expressing human HVEM (for herpesvirus entry mediator) at surface level, naturally or following stable transfection, but not in cells in which this receptor was not detected by flow cytometry analysis, (ii) treatment with soluble gD induced a dose-dependent NF-kappaB activation in THP-1 cells naturally expressing HVEM, and a monoclonal antibody that prevents binding of gD to HVEM significantly reduced NF-kappaB activation by soluble gD in the same cells, (iii) coculture with transfectants expressing wild-type gD on their surface induced an approximately twofold increase in NF-kappaB activation in cells naturally expressing HVEM, while coculture with transfectants expressing a mutated form of gD, lacking its capability to bind HVEM, did not induce a similar effect and (iv) treatment with soluble gD induced a dose-dependent NF-kappaB activation in CHO transfectants expressing HVEM, but not in control CHO transfectants lacking any functional gD receptor. Overall, these results establish that HVEM is involved in NF-kappaB activation by HSV-1 gD.
