10. EFFECTS OF BORON SUPPLEMENTATION ON DAIRY CALVES’ HEALTH: A METABOLOMICS STUDY.

Abstract

Boron supplementation has been demonstrated to exert many beneficial effects on animals. The first metabolomic investigation into the effects of boron on dairy calf health is covered in this study. For two months, healthy newborn calves were enrolled in one control group and three experimental groups. Boron was added to the milk of the experimental groups in increasing doses. Biochemical profile, Nuclear Magnetic Resonance (NMR) metabolomic profiles on days 0, 30, and 60, and daily health scores were determined. Worsening of health score parameters was more common in the control calves, and they were prone to getting sick. Enzyme increases due to birth stress were remarkable. Significant increases in the passive colostral adequacy parameter γ-glutamyl transferase were also not maintained with boron addition. A total of 33 water-soluble and 17 lipid-soluble serum metabolites were determined. Increases in glucose, fructose, alanine, cholate, betaine, and 3-hydroxybutyrate levels, and decreases in lactate, isovaleric acid, valine, leucine, tyrosine, and 2-hydroxybutyrate levels were observed. Phosphoglyceride levels were increased, while the levels of different cholesterol types were found to be decreased. From our data, it emerged that better growth performance, increased gluconeogenesis, and liver development were associated with boron supplementation. To interpret the effects of boron supplementation on the health of dairy calves, NMR-based metabolomic assessment has outperformed biochemical analysis. [ABSTRACT FROM AUTHOR]

Abstract

Parkinson's disease (PD) is the neurological disorder showing the greatest rise in prevalence from 1990 to 2016. Despite clinical definition criteria and a tremendous effort to develop objective biomarkers, precise diagnosis of PD is still unavailable at early stage. In recent years, an increasing number of studies have used omic methods to unveil the molecular basis of PD, providing a detailed characterization of potentially pathological alterations in various biological specimens. Metabolomics could provide useful insights to deepen our knowledge of PD aetiopathogenesis, to identify signatures that distinguish groups of patients and uncover responsive biomarkers of PD that may be significant in early detection and in tracking the disease progression and drug treatment efficacy. The present work is the first large metabolomic study based on nuclear magnetic resonance (NMR) with an independent validation cohort aiming at the serum characterization of de novo drug-naive PD patients. Here, NMR is applied to sera from large training and independent validation cohorts of German subjects. Multivariate and univariate approaches are used to infer metabolic differences that characterize the metabolite and the lipoprotein profiles of newly diagnosed de novo drug-naive PD patients also in relation to the biological sex of the subjects in the study, evidencing a more pronounced fingerprint of the pathology in male patients. The presence of a validation cohort allowed us to confirm altered levels of acetone and cholesterol in male PD patients. By comparing the metabolites and lipoproteins levels among de novo drug-naive PD patients, age- and sex-matched healthy controls, and a group of advanced PD patients, we detected several descriptors of stronger oxidative stress.

Abstract

A prodromal phase of Parkinson’s disease (PD) may precede motor manifestations by decades. PD patients’ siblings are at higher risk for PD, but the prevalence and distribution of prodromal symptoms are unknown. The study objectives were (1) to assess motor and non-motor features estimating prodromal PD probability in PD siblings recruited within the European PROPAG-AGEING project; (2) to compare motor and non-motor symptoms to the well-established DeNoPa cohort. 340 PD siblings from three sites (Bologna, Seville, Kassel/Goettingen) underwent clinical and neurological evaluations of PD markers. The German part of the cohort was compared with German de novo PD patients (dnPDs) and healthy controls (CTRs) from DeNoPa. Fifteen (4.4%) siblings presented with subtle signs of motor impairment, with MDS-UPDRS-III scores not clinically different from CTRs. Symptoms of orthostatic hypotension were present in 47 siblings (13.8%), no different to CTRs (p = 0.072). Abstract

Recent findings about the differential gene expression signature of periodontal lesions have raised the hypothesis of distinctive biological phenotypes expressed by generalized chronic periodontitis (GCP) and generalized aggressive periodontitis (GAgP) patients. Therefore, this cross-sectional investigation was planned, primarily, to determine the ability of nuclear magnetic resonance (NMR) spectroscopic analysis of unstimulated whole saliva to discriminate GCP and GAgP disease-specific metabolomic fingerprint and, secondarily, to assess potential metabolites discriminating periodontitis patients from periodontally healthy individuals (HI).NMR-metabolomics spectra were acquired from salivary samples of patients with a clinical diagnosis of GCP (n = 33) or GAgP (n = 28) and from HI (n = 39). The clustering of HI, GCP, and GAgP patients was achieved by using a combination of the Principal Component Analysis and Canonical Correlation Analysis on the NMR profiles.These analyses revealed a significant predictive accuracy discriminating HI from GCP, and discriminating HI from GAgP patients (both 81%). In contrast, the GAgP and GCP saliva samples seem to belong to the same metabolic space (60% predictive accuracy). Significantly lower levels (P 0.05) of pyruvate, N-acetyl groups and lactate and higher levels (P 0.05) of proline, phenylalanine, and tyrosine were found in GCP and GAgP patients compared with HI.Within the limitations of this study, CGP and GAgP metabolomic profiles were not unequivocally discriminated through a NMR-based spectroscopic analysis of saliva.

Abstract

Metabolomics deals with the whole ensemble of metabolites (the metabolome). As one of the ‐omic sciences, it relates to biology, physiology, pathology and medicine; but metabolites are chemical entities, small organic molecules or inorganic ions. Therefore, their proper identification and quantitation in complex biological matrices requires a solid chemical ground. With respect to for example, DNA, metabolites are much more prone to oxidation or enzymatic degradation: we can reconstruct large parts of a mammoth's genome from a small specimen, but we are unable to do the same with its metabolome, which was probably largely degraded a few hours after the animal's death. Thus, we need standard operating procedures, good chemical skills in sample preparation for storage and subsequent analysis, accurate analytical procedures, a broad knowledge of chemometrics and advanced statistical tools, and a good knowledge of at least one of the two metabolomic techniques, MS or NMR. All these skills are traditionally cultivated by chemists. Here we focus on metabolomics from the chemical standpoint and restrict ourselves to NMR. From the analytical point of view, NMR has pros and cons but does provide a peculiar holistic perspective that may speak for its future adoption as a population‐wide health screening technique. Metabolomics is the omic science that best represents the molecular phenotype. Among several applications it has the potential to be translated into the clinical practice. This Review presents the NMR‐vision of metabolomics, highlighting the high degree of standardization needed along the entire workflow (pre‐analytics, NMR spectra acquisition and processing, statistical analysis and data deposition) for reliable and reproducible results. Abstract

In recent years, new formula milk (FM) products based on milk from farms that strictly adhere to the "organic farming" practices became available. However, little is known about the differences in nutritional profile of these organic formulae with respect to traditional ones. We comprehensively evaluated the metabolite profiles of FM with nuclear magnetic resonance (NMR)-based metabolomic analysis. Five commercial brands of organic and nonorganic formula liquid milk for infants (0-12 months) and toddlers (1-3 years) were analyzed, together with human milk (HM) samples. Proton NMR ( 1 H NMR) spectroscopy mapped molecular characteristics of FM linked to different production techniques, and identified differences between FM and HM samples. We performed a metabolic fingerprint analysis using multivariate and univariate statistical techniques. A clear distinction is found among different commercial brands of the FM samples. In addition, several differences in metabolomic profiles of FM have been found in comparison with HM for the first time. Notably, it was possible to identify, both in the formulations for toddlers and for infants, metabolites that vary in concentration between the formulae produced with milk obtained according to organic farming techniques, and those produced using nonorganic milk. In particular, organic and nonorganic formulations are differentiated by the levels of glucose, methionine, o -phosphocholine, butyrate, hippurate, creatine, and dimethyl sulfone. Importantly, the HM appeared to differ from both organic and nonorganic brands in a context of metabolites. These findings inform efforts to design FM in ways that closely mimic HM, and guide research to differentiate organic and traditional FM.
