Mastic asphalt workers may be exposed to polycyclic aromatic hydrocarbons (PAH) present in bitumen. We conducted a cross-shift study to determine genotoxic effects after exposure to bitumen. For this purpose, external and internal exposure of 202 mastic asphalt workers exposed to bitumen and 55 construction workers without exposure to bitumen was assessed. Exposure by inhalation to fumes of bitumen during the shift was measured by personal ambient monitoring. To assess overall exposure to bitumen (by inhalation and dermal absorption), 1-hydroxypyrene (1-OHP) and the sum of 1-, 2 + 9-,3-,4-hydroxyphenanthrene (OHPhe) were determined in pre- and postshift urine. 8-Oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodGuo) adducts, anti-benzo[a]pyrene-7,8-diol-9,10-epoxide (anti-BPDE) DNA adducts, DNA strand breaks and alkali-labile sites, and micronucleus frequencies were determined as biomarkers of genotoxic effects in white blood cells (WBC). Concentrations of fumes of bitumen were correlated with a moderate association with 1-OHP and OHPhe after work shift (r(s) = 0.25, P < 0.001 and r(s) = 0.36, P < 0.001). Significantly increased 8-oxodGuo adduct levels were observed after shift in both exposure groups (P < 0.0001). Paradoxically, decreased DNA strand break frequencies were observed after shift in both groups (P < 0.05). Postshift values in DNA strand break frequency were associated with 1-OHP (Spearman rank correlation coefficient 0.19, P = 0.01). Significantly more 8-oxodGuo adducts and DNA strand breaks were found in workers exposed to bitumen before and after shift compared with the reference group. However, no dose-dependent association was observed between exposure and genotoxic effects. Nevertheless, the findings indicate that workers exposed to bitumen exerted a higher level of DNA damage (8-oxodGuo and DNA strand breaks) in WBC compared with reference subjects. Abstract: We present results from magnetooptical investigations of large elongated single self assembled In0.3Ga0.7As quantum dots with a low surface density of . Compared to conventional In0.6Ga0.4As quantum dots the dimension of the investigated dots is enlarged by nearly one order of magnitude using a low strain In0.3Ga0.7As nucleation layer. In addition, the exciton exhibits a smaller g-factor of 0–0.4 and a larger diamagnetic coefficient of 20– in Faraday geometry, reflecting the increased extension of the exciton wavefunction, with respect to In0.6Ga0.4As quantum dots. From power dependent investigations we observe biexciton binding energies ranging from 1.7 to 1.9meV. Excited state emission appears typically 2–5meV above the ground state which is consistent with the increased dimensions of the structure. Furthermore we find linear polarization degrees of up to 0.6 from exciton emission of the elongated quantum dot structure. [Copyright &y& Elsevier]

Abstract Background Chemoradiation for cervical cancer patients is associated with considerable gastrointestinal toxicity. Intensity-modulated radiotherapy (IMRT) has demonstrated superiority in terms of target coverage and normal tissue sparing in comparison to conventional 3D planning in gynaecological malignancies. Whether IMRT in prone (PP) or supine position (SP) might be beneficial for cervical cancer patients remains partially unanswered. Methods 10 patients on FIGO stage IB-III cervical cancer, 6 patients for definitive and 4 patients for adjuvant external beam pelvic RT, were planned in PP and SP using a 7-field IMRT technique. IMRT plans for PP and SP (mean dose, Dmean 50.4 Gy) were optimized in terms of PTV coverage (1st priority) and small bowel sparing (2nd priority). A comparison of DVH parameters for PTV, small bowel, bladder, and rectum was performed. Results The comparison showed a similar PTV coverage of 95% of the prescribed dose and for target conformity in IMRT plans (PP, SP). PTV, rectum and bladder volumes were comparable for PP and SP. Significantly larger volumes of small bowel were found in PP (436 cc, + 35%, p = 0.01). PP decreased the volume of small bowel at 20-50.4 Gy (p < 0.05) and increased the rectum volumes covered by doses from 10-40 Gy (p < 0.01), the V50.4 was < 5% in both treatment positions. Bladder sparing was significant better at 50.4 Gy (p = 0.03) for PP. Conclusion In this dosimetric study, we demonstrated that pelvic IMRT in prone position for patients with cervical cancer seems to be beneficial in reducing small bowel volume at doses ≥20 Gy while providing similar target coverage and target conformity. The use of frequent image guidance with KV (kilovolt) or MV (megavolt) computertomography can reduce set-up deviations, and treatment in prone position can be done with a higher set-up accuracy. Clinical outcome studies are needed to affirm lower toxicity.

To study the associations between exposure to vapours and aerosols of bitumen and genotoxic effects, a cross-sectional and cross-shift study was conducted in 320 exposed workers and 118 non-exposed construction workers. Ambient air measurements were carried out to assess external exposure to vapours and aerosols of bitumen. Hydroxylated metabolites of naphthalene, phenanthrene and pyrene were measured in urine, whereas (+)-anti-benzo[a]pyrene-7,8-diol-9,10-epoxide ((+)-anti-BPDE), 8-oxo-7,8-dihydro-2'-deoxyguanosine (8oxodGuo) and DNA strand breaks were determined in blood. Significantly higher levels of 8-oxodGuo adducts and DNA strand breaks were found in both pre- and post-shift blood samples of exposed workers compared to those of the referents. No differences between exposed workers and referents were observed for (+)-anti-BPDE. Moreover, no positive associations between DNA damage and magnitude of airborne exposure to vapours and aerosols of bitumen could be observed in our study. Additionally, no relevant association between the urinary metabolites of PAH and the DNA damage in blood was observed. Overall, our results indicate increased oxidative DNA damage in workers exposed to vapours and aerosols of bitumen compared to non-exposed referents at the group level. However, increased DNA strand breaks in bitumen workers were still within the range of those found in non-exposed and healthy persons as reported earlier. Due to the lack of an association between oxidative DNA damage and exposure levels at the workplaces under study, the observed increase in genotoxic effects in bitumen workers cannot be attributed to vapours and aerosols of bitumen.

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous in our environment and can cause cancer. Exposure to PAHs can be assessed by protein adduct dosimetry using benzo[a]pyrene (B[a]P) as a model compound. We present an overview of analytical methods to detect B[a]P- derived protein adducts in humans, their uses in exposure assessment, and recommendations for future research. Two major methodologies, enzyme-linked immunosorbent assay (ELISA) and chemical-specific assays, could be traced in the literature but there remains limitations with both assays. ELISA is nonspecific due to cross-reactivity of the antibody with other PAHs and results are better interpreted in terms of PAH exposure. ELISA is unable to distinguish between exposed and nonexposed persons in the majority of studies. Adduct concentrations are higher by several orders of magnitude compared to those determined by chemical-specific methods. The latter methods mostly analyzed protein adducts derived by (+)-anti-B[a]P-diol epoxide. For this purpose, gas or liquid chromatography in combination with mass spectrometry or fluorescence detection were used. However, the prevalence of positive samples remained low when chemical- specific assays were used mainly due to the lack of sensitivity. Overall, data on B[a]P-derived protein adducts in humans remain inconclusive. Future research should focus on the development and standardization of a sensitive and specific method for B[a]P-derived protein adducts prior to its use in field studies. Finally, exposures of B[a]P at the workplace and via diet, a major route of exposure of the general population, can be studied. The results will contribute to the understanding of B[a]P-induced cancer and will allow for health preventive measures.

Photoelectric-enhanced radiation therapy is a bimodal therapy, consisting of the administration of highly radiation-absorbing substances into the tumor area and localized regional irradiation with orthovoltage x-rays. Irradiation can be performed by a modified computed tomography (CT) unit equipped with an additional x-ray optical module which converts the polychromatic, fan-shaped CT beam into a monochromatized and focused beam for energy-tuned photoelectric-enhanced radiotherapy. A dedicated x-ray optical module designed for spatial collimation, focusing, and monochromatization was mounted at the exit of the x-ray tube of a clinical CT unit. Spectrally resolved measurements of the resulting beam were performed using an energy-dispersive detection system calibrated by synchrotron radiation. The spatial photon fluence was determined by film dosimetry. Depth-dose measurements were performed and compared to the polychromatic CT and a therapeutic 6 MV beam. The spatial dose distribution in phantoms using a rotating radiation source (quasimonochromatic CT and 6 MV, respectively) was investigated by gel dosimetry. The photoelectric dose enhancement for an iodine fraction of 1% in tissue was calculated and verified experimentally. The x-ray optical module selectively filters the energy of the tungsten Kalpha emission line with an FWHM of 5 keV. The relative photon fluence distribution demonstrates the focusing characteristic of the x-ray optical module. A beam width of about 3 mm was determined at the isocenter of the CT gantry. The depth-dose measurements resulted in a half-depth value of approximately 36 mm for the CT beams (quasi-monochromatic, polychromatic) compared to 154 mm for the 6 MV beam. The rotation of the radiation source leads to a steep dose gradient at the center of rotation; the gel dosimetry yields an entrance-to-peak dose ratio of 1:10.8 for the quasi-monochromatic CT and 1:37.3 for a 6 MV beam of the same size. The photoelectric dose enhancement factor increases from 2.2 to 2.4 by using quasi-monochromatic instead of polychromatic radiation. An additional increase in the radiation dose by a factor of 1.4 due to the focusing characteristic of the x-ray optical module was calculated. Photoelectric-enhanced radiation therapy based on a clinical CT unit combined with an x-ray optical module is a novel therapy option in radiation oncology. The optimized quasi-monochromatic radiation is strongly focused and ensures high photoelectric dose enhancement for iodine.

Endotoxin (ET) contaminated dusts frequently can be found in the environment. Especially in agriculture, ETs represent one of the main factors in the development of respiratory diseases. In order to investigate the pathomechanisms of ET induced lung injury, the contribution of the NANC-system of vagal C-fibres to increased airway responsiveness (AR) to cholinergic agents was investigated in anesthetized rabbits. In two control groups of 6 anesthetized Wight New Zealand rabbits each, E. coli ET was infused i.v. (0.4microg/kg) (Group 1) or the aerosolized ET was inhaled for 10 min (Group 2). Before and 1 and 3 hours after ET exposure, AR to 0.2 und 2.0% acetylcholine aerosol was measured. The increased AR after ET application was compared with the ET responses in rabbits with neuropeptides depleted by subchronic capsaicin treatment on four consecutive days (Groups 3 and 4). ET-inhalation and ET-infusion both resulted in a significant increase in AR to ACH (P<0.005). The increase in dynamic elastance (E(dyn)), as a measure of airway resistance to 0.2% ACH after ET in both groups was comparable with the response to 2.0% ACH before exposure. In rabbits with capsaicin treatment, basal ACH-responsiveness was similar to the control group. After ET-Exposure, increase in AR to ACH was significantly (P<0.05) smaller compared with untreated rabbits. In conclusion, depletion of neuropeptides by capsaicin resulted in a significantly decreased ET-induced airway hyperresponsiveness (AHR) in rabbits, indicating the contribution of the NANC-system and their neuromodulators to ET-induced AHR. The results confirm the important role of the NANC-system environmental and occupational agents induced-AHR in rabbits.

We conducted a cross-shift study with 66 bitumen-exposed mastic asphalt workers and 49 construction workers without exposure to bitumen. Exposure was assessed using personal monitoring of airborne bitumen exposure, urinary 1-hydroxypyrene (1-OHP), and the sum of 1-, 2 + 9-,3-,4-hydroxyphenanthrene (OHPH). Genotoxic effects in WBC were determined with nonspecific DNA adduct levels of 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodGuo) and the formation of DNA strand breaks and alkali-labile sites. Concentration of fumes and aerosols of bitumen correlated significantly with the concentrations of 1-OHP and OHPH after shift (r(s) = 0.27; P = 0.03 and r(s) = 0.55; P < 0.0001, respectively). Bitumen-exposed workers had more DNA strand breaks than the reference group (P < 0.0001) at both time points and a significant correlation with 1-OHP and OHPH in the postshift urines (r(s) = 0.32; P = 0.001 and r(s) = 0.27; P = 0.004, respectively). Paradoxically, we measured higher levels of DNA strand breaks, although not significant, in both study groups before shift. 8-OxodGuo adduct levels did not correlate with DNA strand breaks. Further, 8-oxodGuo levels were associated neither with personal exposure to bitumen nor with urinary metabolite concentrations. Significantly more DNA adducts were observed after shift not only in bitumen-exposed workers but also in the reference group. Only low-exposed workers had significantly elevated 8-oxodGuo adduct levels before as well as after shift (P = 0.0002 and P = 0.02, respectively). Our results show that exposure to fumes and aerosols of bitumen may contribute to an increased DNA damage assessed with strand breaks.

The major DNA adducts of anti-benzo[a]pyrene diolepoxide (BPDE) were determined by high performance liquid chromatography with fluorescence detection (HPLC-FLD) in white blood cells (WBC) of workers exposed to benzo[a]pyrene (B[a]P). In addition, ambient concentrations of B[a]P at the workplace were determined by personal air sampling. Workers in a refractory setting were examined before (n=26) and 3 months after (n = 33) changing the production material (binding pitch). Furthermore, 9 coke oven workers were examined. The change in the production process in the refractory setting led to a decrease in the median of ambient B[a]P concentrations (0.14 to <0.07 microg/m3). The median of BPDE-DNA adduct levels in WBC also decreased from 0.9 adducts/10(8) nucleotides before changing the production material to <0.5 adducts/10(8) nucleotides 3 months afterwards. The B[a]P concentrations at the workplace for the coke oven workers were found to be significantly higher than in the refractory setting. However, BPDE-DNA adduct concentrations in coke oven workers and refractory setting workers showed no significant difference, which was probably due to the low number of studied subjects in the coke-oven setting. No significant differences could be observed for BPDE-DNA adduct levels between current smokers (n=21) and non-smokers (n=14; p = 0.93) from both plants. In addition, no correlation between B[a]P concentrations in the air and DNA adduct levels in refractory workers and in coke oven workers could be found (r = -0.03, p = 0.87). Because of the missing correlation between personal air sampling and BPDE-DNA adduct levels in WBC, the results may indicate that their formation is either influenced by other routes of exposure to B[a]P (e.g., skin absorption, dietary habits) or interindividual differences in their formation and repair.

Cooling lubricants are used in the metal industry during drilling or turning. Vapors and aerosols of these lubricants are suspected to induce airway hyperresponsiveness (AHR) in exposed workers. In a previous study the authors demonstrated that water-soluble lubricants induce AHR after acute exposure of rabbits to concentrations near the German MAK value (10 mg/m(3)). In the present investigation the influence of a fatty alcohol as special non-water-soluble cooling lubricant was examined to determine its influence on airway responsiveness (AR). The effects of an aerosolized non-water-soluble lubricant (40, 90, and 220 mg/m(3)) on AR to acetylcholine in a rabbit model were studied. Lubricant atmosphere analysis was performed with infrared spectroscopy. Before exposure, after 2 and 4 hours of application, AR to aerosols from 0.2 and 2% acetylcholine was tested. Basal airway and cardiovascular parameters as well as blood gases did not change during exposure. Lubricant aerosol concentration of 40 and 220 mg/m(3) for 4 hours did not significantly alter AR. Inhalation of 90 mg/m(3) lubricant increased contractile response to ACH significantly. In contrast to formerly investigated water-soluble cooling lubricants, the examined non-water-soluble lubricant did not increase AR in concentrations near the MAK; however, in higher concentrations a significant (p<.05) increase was obtained.

Metal working fluids (MWF) are used in the manufacturing process to cool and lubricate machined parts. MWF contain the coolant, mostly water-oil emulsions with soluble semi-synthetic or synthetic oils, but also emulsifiers and additives. Reaction products and both bacterial and fungal contamination occur during use. In the present review we listed literature data of dose-effect-relationships with a focus on biological substances. Most studies describe obstructive airways disease due to irritant effects of MWF. Immunoglobulin E mediated airways disease or hypersensitivity pneumonitis have rarely been described. There is no doubt that MWF exert adverse effects on human airways, but there is few data concerning dose-effect relationships. The role of bacterial and fungal contamination or endotoxin exposure cannot be defined by the available information. However, there is some evidence that biological components in MWF may represent the most hazardous substances.

Inhalational exposure to organic solvents is known to exert neurotoxic effects. Using the new multielectrode dish system (Panasonic) the effects of chronic exposure to trichloroethylene (TCE) on neuronal plasticity were assessed in different regions of the adult rat brain. Two groups of Long-Evans rats were exposed to 0 ppm or 500 ppm TCE, respectively, 6 h/day, 5 days/week for 6 months. Long-term potentiation (LTP) as well as paired-pulse potentiation/inhibition were assessed in slices from the visual cortex and the hippocampus. In addition, several behavioral tests were performed. Trichloroethanol concentrations were measured in blood and trichloroacetic acid concentrations were determined in urine. While TCE exposure impaired LTP as well as paired-pulse potentiation in hippocampal slices, no effects were seen in cortical slices. Our data demonstrate brain region specific functional changes following TCE exposure with the hippocampus being more vulnerable than the visual cortex. The behavioral measurements revealed no TCE related effects.

Airway hyperresponsiveness (AHR) to water-soluble cooling lubricants (CL) induced by aerosol administered by tracheal tube was studied in a rabbit model of occupational lung disease. Two commercial CL were examined: the first was of the boric acid amine ester type without biozide (CL-BAE), the second was of the sulfonate type with biozide (CL-SB). 50, 5.0 or 0.5 mg/m3 CL was administered over a period of twice 2 h to six different groups of rabbits. Airway responsiveness (AR) to aerosols of 0.2% and 2.0% acetylcholine solution (ACH) was measured before and after each exposure to CL. A control group A of nine animals not exposed to CL showed no significant respiratory responses following inhalation of 0.2% ACH for 1 min. Conversely, inhalation of 2.0% ACH almost doubled the dynamic elastance (Edyn) in the ACH challenge test in this animal group. Airway resistance (RI), Edyn, slope of inspiratory pressure generation (delta Pes/tI), arterial pressure (Pa) and arterial blood gas tensions (PaO2, PaCO2) were not significantly altered during and after exposures to CL. However, after CL-BAE inhalation of 50 and 5 mg/m3 over 4 h, the amplitude of the ACH-induced airway obstruction indicated by the changes in Edyn rose significantly to almost five times the control response before exposure (group C, D, p < 0.005). Similar changes in RI and delta Pes/tI were obtained. After inhalation of 0.5 mg/m3 CL-BAE (group D), no significant changes in AR were observed. Similar to CL-BAE inhalation of 50 mg/m3, CL-SB caused enlarged AR in the ACH challenge test (group E), whereas no significant changes were found after exposure to 5.0 and 0.5 mg/m3 in groups F and G. In summary, CL aerosols with and without biozide in the range of 50 and 5 mg/m3 applied via tracheal tubes increased AR to ACH within 4 h of exposure in a time- and concentration-dependent manner. It has to be assumed that this augmented AR indicates an increased risk of developing lubricant-induced obstructive lung diseases.

Clinical studies have shown that oxidizing substances like hydrogen peroxide (H2O2) and ammonium persulfate used in the hair cosmetic industry may cause airway diseases. In in vitro experiments with isolated guinea pig tracheae ammonium persulfate solutions induced an initial transient relaxation of smooth muscles. This relaxation could be measured by a decrease in isometric pressure in the cannulated trachea instilled with ammonium persulfate at a hydrostatic pressure of 2.5 kPa. In control experiments, saline caused an initial pressure decrease of less than 10% within one minute. In contrast, instillation of different ammonium persulfate solutions (9.10(-5) to 9.10(-2) M) effectively dilated tracheae and resulted in a concentration-dependent drop in intratracheal pressure to 1.53 +/- 0.62 kPa (61% of the instillation pressure). The effect of ammonium persulfate on smooth muscles is obviously mediated by nitric oxide because the relaxation could be blocked by inhibitors of nitric oxide synthase (L-NMMA 40 microM and L-NAME 200 microM). The precursor of nitric oxide, L-arginine (1 mM), the D-isomers of the inhibitors and a mixture of L-arginine and L-NAME did not affect the ammonium persulfate-induced initial intratracheal pressure decrease. The results allow us to conclude that acutely elicited tracheal muscle dilatation by ammonium persulfate is mediated by nitric oxide. However, it is possible that a continuous use of oxidizing substances may lead to epithelial damage and, therefore, reduce the production of nitric oxide, thus facilitating constrictory responses.

Airway hyperresponsiveness, manifested by increased flow resistance and resulting drop in oxygen partial pressure when conducting provocation tests, is considered an early sign of a developing obstructive airway disease, an example of which is a professionally conditioned asthma. We conducted a detailed study exploring the interrelation between the respiratory mechanical parameters and the partial pressures of oxygen and carbon dioxide (PaO2, PaCO2). Reproducibility tests for the studied cardiovascular, ventilatory and respiratory mechanical parameters at rest and under various conditions of stress (external stenoses, inhalation of hypercapnic and hypoxic gas mixtures, infusion of an acetylcholine solution) showed good reproducibility of the measured data with variation coefficients < 10%. In blood gas analyses we also found comparable variation coefficients. Four groups of experimental animals were exposed for different periods of time to various working place substances (coolants, ammonium peroxodisulfate, hair bleaches [blondizing agents], isocyanates). After the exposure we checked on the bronchial sensitivity to aerosols of 0.2% and 2% acetylcholine solutions. Concomitant with an increased response of dynamic elastance, we found an increased drop in oxygen partial pressure and an almost constant carbon dioxide partial pressure, dependent on the working place substance used and on its concentration. In untreated controls the inhalation of acetylcholine resulted in bronchoconstriction and drop in oxygen partial pressure only on provocation with 2% acetylcholine. However, in the groups exposed to coolants and ammonium peroxodisulfate there was a significant drop in oxygen partial pressure already on provocation with 0.2% acetylcholine, as well as a noticeable bronchial respiratory response. The drop in oxygen partial pressure increases further after provocation with 2% acetylcholine, whereas the oxygen partial pressure dropped to a maximum of one-third of its original level by more than 10%. Placing the drop in oxygen partial pressure provoked by acetylcholine in relation to the increase in dynamic elastance, this can be well expressed by a logarithmic formula (y = -6.2. In (x) + 0.72, r = 0.96) that does not change significantly after exposure to working place substances (y = -7.0. In (x) + 3.33, r = 0.93). The close correlation of both parameters suggests that study of the oxygen partial pressures to determine the airway hyperresponsiveness should be considered important, since a marked drop in oxygen partial pressure is seen even if obstructive respiratory response is only slightly increased (slight increase in dynamic elastance). The reason for the behaviour of the blood gases is probably an increased ventilation-perfusion imbalance due to inhomogenous peripheral bronchial reactions. In the hyperresponsive animals the reactions were merely enhanced without demonstrating any differences.

The pathogenesis of hepatitis A virus (HAV) infection was studied in owl monkeys following oral administration of the wild-type HM-175 strain of HAV. Stools were collected daily and blood and pharyngeal swabs twice weekly for viral isolation, and animals were necropsied at various intervals after inoculation. Organs were examined for the presence of virus by isolation in cell culture and for viral antigens by immunofluorescence. Monkeys excreted HAV in the stools for 1-4 days after inoculation, presumably due to the residual unabsorbed inoculum. No virus was found in stools for the next 2-3 days. HAV re-appeared on days 4-7 and then persisted through day 39. Viremia occurred on the 10th day and continued until day 35. Virus was isolated occasionally from throat swabs 1 or 2 weeks after it was detected in stools and blood, and there was no evidence that HAV replicated in the pharyngeal tissues. Animals acquired anti-HAV antibody by the 4th week, and alanine aminotransferase (ALT) was elevated 5-5.5 weeks after inoculation. HAV was isolated from liver 5 days after inoculation; however, viral antigens were first detected in Kupffer cells of the liver at 14 days and in hepatocytes at 21 days. HAV antigen was detected in epithelial cells of the intestinal crypts and in the cells of the lamina propria of the small intestine 3 days postinoculation and thereafter until the 5th week, suggesting that these cells might represent an additional site of HAV replication.

Ammonium persulfate (APS) is used as an oxidant in many industrial branches and is contained in bleaching substances used in hair cosmetic industry. In addition to skin diseases, respiratory disorders have been frequently reported. We examined whether APS can induce bronchial hyperresponsiveness (BHR) which has to be regarded as a first step in the development of obstructive respiratory disorders. In a rabbit model we tested the influence of aerosolized APS-solutions in concentrations of 0.1, 0.01 and 0.001 M (50, 5, 0,5 ppm in air) for about four hours on the contractile bronchial reaction. Exposure to APS-aerosols does not alter basal airway Resistance (RI), dynamic Elastance (Edyn), slope of inspiratory pressure generation (delta Poes/tI), arterial blood pressure and blood gases (PaO2, PaCO2). Before, after two and after four hours the influence of APS application on the bronchial reactivity to 0.2%- and 2% acetylcholine-aerosols (ACH) was tested. Exposure to an aerosol from 0.1 M APS solution, which is equivalent to a concentration of 50 ppm APS in air for two hours induces a significant increase in bronchoconstrictory responses to ACH. A nebulized 0.01 M APS solution causes an increased ACH response after four hours of exposure. However, inhalation of 0.001 M APS (0.5 ppm in air) does not alter sensitivity to ACH within four hours of exposure. In bronchoalveolar lavage an increased number of inflammatory cells was found. Our results demonstrate, aerolized APS-solutions to cause bronchial hyper-responsiveness after few hours of exposure presumably under the participation of inflammatory cells.
