Abstract

Background: The increasing incidence of dermatophytoses in the world and the side effects of the current therapies encouraged the search of alternative drugs. Hence the objective of this work was to determine antidermatophytes activity of Syzigium aromaticum formulate antidermatophytic cream. Materials and Methods: The extracts were prepared by maceration of plant materials into methanol. Three formulations of creams were made, and the best was chosen according to its physicochemical stability and appearance. The acute dermal toxicity and antidermatophytic efficacy of the cream was performed on guinea-pig. Results: The methanolic extract of S. aromaticum was incorporated in the final cream formulation. The formulation containing shea-butter 58.5%, acetylic alcohol 2.5%, stearic acid 1.5%, bee-wax 10%, borax 1.5%, polysorbate 60 2.5%, 2 drops of lactic acid and water was chosen because of its good appearance and stability. The cream with methanolic extract of S. aromaticum did not reveal any dermal toxic effect. The cream efficacy was dose-dependent. The treatment with cream at 5% methanolic extracts of S. aromaticum revealed the best potency after 14 days of treatment. Conclusion: These results show that the cream at 5% methanolic extract of S. aromaticum seed is promising in the treatment of dermatophytoses and could be used as an alternative in the development of a new therapy. 17. Anti-yeast activity of extracts and fractions from Uvariodendron calophyllum (Annonaceae)

Abstract

The resistance to available antifungals highlights the urgent need for innovative drugs to treat yeasts infections. This study aimed at evaluating the activity of extracts and fractions from Uvariodendron calophyllum against pathogenic yeasts. The ethanolic and aqueous extracts obtained by maceration were liquidliquid- partitioned using organic solvents and screened against isolates of Candida albicans, Candida glabrata, Candida parapsilosis, Cryptococcus neoformans and Candida albicans reference strains NR-29445, NR-29444, NR-29451, and NR-29450 from BEI Resources using the broth micro-dilution method. Time kill kinetic, inhibition of germ-tube, filamentation and chlamydosporulation, and biofilm formation were assessed using the best sub-fraction. Overall, the most interesting sub-fraction (FS: 237–253) showed an MIC value of 0.0625 mg/mL with cidal effect against C. albicans NR-29450 and NR-29445 at 0.25 mg/mL after 12-16 hours and 24 hours respectively. Moderate inhibitory effects were observed at 0.25 mg/mL against germ-tube formation, filamentation and chlamydosporulation of all C. albicans strains. Also, very moderate inhibition of biofilm formation by C. albicans NR-29450 at 0.25 mg/mL was obtained. The results obtained support U. calophyllum as a potential source of compounds with anti-yeast activity. Abstract

Ethnopharmacological Relevance: Ethnopharmacological surveys were conducted in two regions of Gabon. This led to highlighting some of the medicinal plants used by local populations in the management of HIV/AIDS opportunistic diseases. Two regions with the highest occurrence of HIV/AIDS cases were visited and ethnopharmarcological data was gathered. These regions were the Estuaire Province (Libreville and its neighborhood) and the Haut-Ogooué Province (Franceville and its neighborhood). The opportunistic diseases and symptomatic conditions considered during this study were: diarrhea, respiratory tract infections, cough, tuberculosis, abscesses, stomach ache, skin rashes, venereal diseases, typhoid fever, anemia, general tiredness, hepatitis and vomiting., Materials and Methods: The reported species were evaluated through three parameters: specificity, reliability and frequency. Plant parts of relevant species were harvested and extracted with an aqueous alcohol solution (ethanol/water: 1/1). The extracts obtained were submitted to phytochemical screening and in vitro microbiological assays on some clinical isolates and ATCC strains, involved in HIV/AIDS opportunistic diseases through the Agar well diffusion and Microbroth dilution methods., Results: Among the 52 species identified during this survey, Coelocaryon klainei Pierre ex Heckel (Myristicaceae), Dacryodes klaineana (Pierre) H.J. Lam (Bursecaceae), Phyllanthus diandrus Pax (Euphorbiaceae), Saccoglotys gabonensis (Baill.) Urb. (Humiriaceae) and Tetrorchidium didymostemon (Baill.) Pax & K. Hoffm. (Euphorbiaceae) were submitted to in vitro microbiological assays. Phyllanthus diandrus bark and leaves show best antibacterial activities against Pseudomonas aeruginosa and Klebsiella pneumoniae with MIC value of 0.25 respectively. Phytochemical screening revealed the presence in all the plant parts extracts of potentially bioactive molecules, including polyphenols, especially flavonoids and tannins., Conclusion: It is concluded that some of these plants might be submitted to further scientific studies, including the identification and isolation of bioactive principles, that could be developed to drugs for the treatment of HIV/AIDS opportunistic diseases., (Copyright © 2015 Elsevier Ireland Ltd.
