Background: A milestone goal of the Healthy China Program (2019–2030) is to achieve 5-year cancer survival at 43.3% for all cancers combined by 2022. To assess the progress towards this target, we analyzed the updated survival for all cancers combined and 25 specific cancer types in China from 2019 to 2021. Methods: We conducted standardized data collection and quality control for cancer registries across 32 provincial-level regions in China, and included 6,410,940 newly diagnosed cancer patients from 281 cancer registries during 2008–2019, with follow-up data on vital status available until December 2021. We estimated the age-standardized 5-year relative survival overall and by site, age group, and period of diagnosis using the International Cancer Survival Standard Weights, and quantified the survival changes to assess the progress in cancer control. Results: In 2019–2021, the age-standardized 5-year relative survival for all cancers combined was 43.7% (95% confidence interval [CI], 43.6–43.7). The 5-year relative survival varied by cancer type, ranging from 8.5% (95% CI, 8.2–8.7) for pancreatic cancer to 92.9% (95% CI, 92.4–93.3) for thyroid cancer. Eight cancers had 5-year survival of over 60%, including cancers of the thyroid, breast, testis, bladder, prostate, kidney, uterus, and cervix. The 5-year relative survival was generally lower in males than in females. From 2008 to 2021, we observed significant survival improvements for cancers of the lung, prostate, bone, uterus, breast, cervix, nasopharynx, larynx, and bladder. The most significant improvement was in lung cancer. Conclusions: Progress in cancer control was evident in China. This highlights the importance of a comprehensive approach to control and prevent cancer.

Chimeric antigen receptor (CAR)-T cell therapy has been confirmed improving remission rates in refractory patients or relapsed B-cell acute lymphoblastic leukemia (R/R B-ALL). However, the added benefits of undergoing allogeneic hematopoietic stem cell transplantation (allo-HSCT) following CAR-T therapy remain a subject of debate. In this research we investigated the efficiency and long-term outcomes of CD19 CAR-T bridging with allo-HSCT in R/R B-ALL patients. A total of 42 patients were brought into the cohort studies. Our findings revealed that patients who appected CAR-T followed by HSCT had a 1-year overall survival (OS) rate of 70 % and a 1-year leukemia-free survival (LFS) rate of 95 %. Moreover, patients who underwent this combined treatment had higher OS and LFS rates compared to those who received CAR-T therapy alone. In conclusion, the results of this clinical trial provide compelling evidence for the safety and efficacy of using CAR-T therapy as a bridging strategy to allo-HSCT in patients with R/R B-ALL.

Traditional lettuce counting relies heavily on manual labor, which is laborious and time-consuming. In this study, a simple and efficient method for localization and counting lettuce is proposed, based only on lettuce field images acquired by an unmanned aerial vehicle (UAV) equipped with an RGB camera. In this method, a new lettuce counting model based on the weak supervised deep learning (DL) approach is developed, called LettuceNet. The LettuceNet network adopts a more lightweight design that relies only on point-level labeled images to train and accurately predict the number and location information of high-density lettuce (i.e., clusters of lettuce with small planting spacing, high leaf overlap, and unclear boundaries between adjacent plants). The proposed LettuceNet is thoroughly assessed in terms of localization and counting accuracy, model efficiency, and generalizability using the Shanghai Academy of Agricultural Sciences-Lettuce (SAAS-L) and the Global Wheat Head Detection (GWHD) datasets. The results demonstrate that LettuceNet achieves superior counting accuracy, localization, and efficiency when employing the enhanced MobileNetV2 as the backbone network. Specifically, the counting accuracy metrics, including mean absolute error (MAE), root mean square error (RMSE), normalized root mean square error (nRMSE), and coefficient of determination (R2), reach 2.4486, 4.0247, 0.0276, and 0.9933, respectively, and the F-Score for localization accuracy is an impressive 0.9791. Moreover, the LettuceNet is compared with other existing widely used plant counting methods including Multi-Column Convolutional Neural Network (MCNN), Dilated Convolutional Neural Networks (CSRNets), Scale Aggregation Network (SANet), TasselNet Version 2 (TasselNetV2), and Focal Inverse Distance Transform Maps (FIDTM). The results indicate that our proposed LettuceNet performs the best among all evaluated merits, with 13.27% higher R2 and 72.83% lower nRMSE compared to the second most accurate SANet in terms of counting accuracy. In summary, the proposed LettuceNet has demonstrated great performance in the tasks of localization and counting of high-density lettuce, showing great potential for field application.

CLC-2 is a voltage-gated chloride channel that contributes to electrical excitability and ion homeostasis in many different tissues. Among the nine mammalian CLC homologs, CLC-2 is uniquely activated by hyperpolarization, rather than depolarization, of the plasma membrane. The molecular basis for the divergence in polarity of voltage gating among closely related homologs has been a long-standing mystery, in part because few CLC channel structures are available. Here, we report cryoEM structures of human CLC-2 at 2.46 – 2.76 Å, in the presence and absence of the selective inhibitor AK-42. AK-42 binds within the extracellular entryway of the Cl–-permeation pathway, occupying a pocket previously proposed through computational docking studies. In the apo structure, we observed two distinct conformations involving rotation of one of the cytoplasmic C-terminal domains (CTDs). In the absence of CTD rotation, an intracellular N-terminal 15-residue hairpin peptide nestles against the TM domain to physically occlude the Cl–-permeation pathway. This peptide is highly conserved among species variants of CLC-2 but is not present in other CLC homologs. Previous studies suggested that the N-terminal domain of CLC-2 influences channel properties via a “ball-and-chain” gating mechanism, but conflicting data cast doubt on such a mechanism, and thus the structure of the N-terminal domain and its interaction with the channel has been uncertain. Through electrophysiological studies of an N-terminal deletion mutant lacking the 15-residue hairpin peptide, we support a model in which the N-terminal hairpin of CLC-2 stabilizes a closed state of the channel by blocking the cytoplasmic Cl–-permeation pathway.

Global warming contributes to an increased frequency and severity of droughts. Drought emerges as a highly prevalent natural calamity, distinguished by its formidable disruptive impact and the capacity to trigger considerable economic setbacks. Understanding the spatiotemporal characteristics of droughts and clarifying the driving role of atmospheric circulation on droughts is vital for agricultural, hydrological, ecological, and socio-economic systems. Leveraging meteorological data from 36 stations in the middle reaches of the Yellow River Basin from 1961 to 2020, we employed the Standardized Precipitation Evapotranspiration Index (SPEI) to calculate drought occurrence. Concurrently, we explored the influence of atmospheric circulation on the SPEI. The findings of our study underscore a concerning trend of worsening drought conditions within the study area. We discovered a significant correlation between the duration and severity of drought (R = 0.83, p < 0.001); longer durations often corresponded to higher levels of severity. Turning our attention to atmospheric dynamics, the Nino Eastern Pacific index (NE) emerged as a critical driver of SPEI dynamics (the contribution of NE to SPEI was 0.22), significantly impacting drought patterns. In conclusion, the study significantly contributes to our comprehension of the evolving drought patterns under the influence of global warming. The findings can provide valuable information for water resource management and drought disaster control.

Summary: Background: Racial/ethnic disparities in prostate cancer are reported in the United States (US). However, long-term trends and contributors of racial/ethnic disparities in all-cause and cause-specific death among patients with prostate cancer remain unclear. We analysed the trends and contributors of racial/ethnic disparities in prostate cancer survivors according to the cause of death in the US over 25 years. Methods: In this retrospective, population-based longitudinal cohort study, we identified patients diagnosed with first primary prostate cancer between 1995 and 2019, with follow-up until Dec 31, 2019, using population-based cancer registries’ data from the Surveillance, Epidemiology, and End Results (SEER) Program. We calculated the cumulative incidence of death for each racial/ethnic group (Black, white, Hispanic, Asian or Pacific Islander [API], and American Indian or Alaska Native [AI/AN] people), by diagnostic period and cause of death. We quantified absolute disparities using rate changes for the 5-year cumulative incidence of death between racial/ethnic groups and diagnostic periods. We estimated relative (Hazard ratios [HR]) racial/ethnic disparities and the percentage of potential factors contributed to racial/ethnic disparities using Cox regression models. Findings: Despite a decreasing trend in the cumulative risk of death across five racial/ethnic groups, AI/AN and Black patients consistently had the highest rate of death between 1995 and 2019 with an adjusted HR of 1.48 (1.40–1.58) and 1.40 (1.38–1.42) respectively. The disparities in all-cause mortality between AI/AN and white patients increased over time, with adjusted HR 1.32 (1.17–1.49) in 1995–1999 and 1.95 (1.53–2.49) in 2015–2019. Adjustment of stage at diagnosis, initial treatment, tumor grade, and household income explained 33% and 24% of the AI/AN-white and Black-white disparities in all-cause death among patients with prostate cancer. Interpretation: The enduring racial/ethnic disparities in patients with prostate cancer, call for new interventions to eliminate health disparities. Our study provides important evidence and ways to address racial/ethnic inequality. Small cell lung cancer (SCLC) is a malignant tumor with strong invasiveness and high mortality. It has the characteristics of easy metastasis, fast growth, high degree of malignancy and strong invasiveness. The prognosis of patients is generally poor. The current clinical diagnosis of SCLC is mainly based on tissue biopsy, which is invasive, long cycle time and high cost. In recent years, liquid biopsy has been gradually applied because of its non-invasive, comprehensive and real-time characteristics that traditional tissue biopsy does not have. The main detection objects of liquid biopsy include circulating tumor DNA (ctDNA), circulating tumor cells (CTCs) and exosomes in peripheral blood. The application of liquid biopsy in the clinical treatment of SCLC will help clinicians to improve the detailed diagnosis of SCLC patients, as well as the timely control and response to the treatment response of patients.

Claudin-5 is a tight junction integral membrane protein, but it is also expressed in mature podocytes which lack tight junctions. Here the authors report that podocyte claudin-5 regulates WNT signaling activity by modulating WIF1 expression, and its downregulation contributes to kidney disease progression in mice.

The accuracy and rapidity of total iron content (TFE) analysis can accelerate iron ore production. Although the conventional TFE detection methods are accurate, its detection speed presents difficulties in meeting production requirements. Therefore, this paper proposes a method of TFE detection based on reflectance spectroscopy (wavelength range: 340–2500 nm) and remote sensing. Firstly, spectral experiments were conducted on iron ore using the HR SVC-1024 spectrometer to obtain spectral data for each sample. Then, the spectra were smoothed and dimensionally reduced by using wavelet transform and principal component analysis. To improve the detection accuracy of TFE, a two hidden layer extreme learning machine with variable neuron nodes based on an improved sparrow search algorithm and batch normalization optimization (MSSA-BNVTELM) is proposed. According to the experimental results, MSSA-BNVTELM exhibited superior detection accuracy in comparison to other algorithms. In addition, this research established a remote sensing detection model using Sentinel-2 data and MSSA-BNVTEM to detect the distribution of TFE in the mining area. The distribution of TFE in the mine area was plotted based on the detection results. The results show that the remote sensing of the mine area can be useful for detection of the TFE distribution, providing assistance for the mining plan.

Small cell lung cancer (SCLC) is a highly aggressive and fatal malignant tumor. It has the characteristics of complex etiology, low differentiation, high malignancy, fast growth, strong invasiveness, early metastasis and acquired drug resistance, resulting in poor prognosis. In recent years, with the gradual deepening understanding on the molecular mechanism of SCLC and multi-omics data, it is proposed that molecular typing can be carried out according to the differential expression of key transcription factors, including SCLC-A, SCLC-N, SCLC-P and SCLC-I subtypes. Molecular typing of SCLC and its clinical application will help doctors to further optimize the detailed diagnosis and treatment plan of SCLC patients, so as to prolong the survival time and improve the quality of life of patients.

Pancreatic ductal adenocarcinoma (PDAC) is an extremely aggressive cancer, characterized by a high metastatic burden. RIO Kinase 3 (RIOK3) has been shown to promote invasion and metastasis of PDAC by cytoskeleton remodeling, but the exact mechanism is still unknown. In this study, we analyzed transcriptome sequencing data from RIOK3 stable knockdown PANC-1 cells and TCGA-PDAC data and discovered that RIOK3 was substantially related to focal adhesion signaling in PDAC. Additionally, silencing RIOK3 dramatically decreased Focal Adhesion Kinase (FAK) protein expression and phosphorylation (Tyr397 and Tyr925 sites). Immunoprecipitation assay verified the interaction of RIOK3 and FAK. Furthermore, RIOK3 considerably increased the protein stability of FAK protein but not FAK-Y925F protein. The biological function of RIOK3 in increasing PDAC cell invasion and migration was shown to be dependent on FAK activation. Moreover, we discovered that RIOK3 mutations were mainly characterized by amplification. RIOK3 mRNA was found to be significantly elevated in PDAC tissues and was associated with a poor prognosis. Furthermore, RIOK3 mRNA was significantly upregulated in later T-stage, pre-existing lymph node metastases, and later pathological stage samples. Overall, our study found that RIOK3 promotes PDAC cell invasion and metastasis by stabilizing FAK protein expression and upregulating its phosphorylation. This also provides a new target for therapeutic modalities targeting FAK.

BackgroundAnti-Embolism (AE) devices therapy is an additional antithrombotic treatment that is effective in many venous diseases, but the correlations between this medical compression therapy and cardiovascular arterial disease or comorbid diabetes mellitus (DM) are still controversial. In this study we investigated the association between compression therapy and intensive care unit (ICU) mortality in patients with a first acute myocardial infarction (AMI) diagnosis complicated with type II DM.MethodsThis retrospective cohort study analyzed all patients with AMI and type II DM in the Medical Information Mart for Intensive Care-IV database. We extracted the demographics, vital signs, laboratory test results, comorbidities, and scoring system results of patients from the first 24 h after ICU admission. The outcomes of this study were 28-day mortality and ICU mortality. Analyses included Kaplan–Meier survival analysis, Cox proportional-hazards regression, and subgroup analysis.ResultsThe study included 985 eligible patients with AMI and type II DM, of who 293 and 692 were enrolled into the no-AE device therapy and AE device therapy groups, respectively. In the multivariate analysis, compared with no-AE device therapy, AE device therapy was a significant predictor of ICU mortality (HR = 0.48, 95% CI = 0.24–0.96, P = 0.039) and 28-day mortality (HR = 0.50, 95% CI = 0.27–0.90, P = 0.021). In addition to age, gender and coronary artery bypass grafting surgery, there were no significant interactions of AE device therapy and other related risk factors with ICU mortality and 28-day mortality in the subgroup analysis.ConclusionsSimple-AE-device therapy was associated with reduced risks of ICU mortality and 28-day mortality, as well as an improvement in the benefit on in-hospital survival in patients with AMI complicated with type II DM.

Abstract Most human diseases are systems diseases, and systems biomarkers are better fitted for diagnostic, prognostic, and treatment monitoring purposes. To search for systems biomarker candidates, lactate dehydrogenase (LDH), a housekeeping protein expressed in all living cells, was investigated. To this end, we analyzed the serum LDH activities from 172,933 patients with 48 clinically defined diseases and 9528 healthy individuals. Based on the median values, we found that 46 out of 48 diseases, leading by acute myocardial infarction, had significantly increased (p 0.8) for hepatic encephalopathy and lung fibrosis.

Therapeutic antibodies-F(ab’)2 obtained from hyperimmune equine plasma could treat emerging infectious diseases rapidly because of their high neutralization activity and high output. However, the small-sized F(ab’)2 is rapidly eliminated by blood circulation. This study explored PEGylation strategies to maximize the half-life of equine anti-SARS-CoV-2 specific F(ab’)2. Equine anti-SARS-CoV-2 specific F(ab’)2 were combined with 10 KDa MAL-PEG-MAL in optimum conditions. Specifically, there were two strategies: Fab-PEG and Fab-PEG-Fab, F(ab’)2 bind to a PEG or two PEG, respectively. A single ion exchange chromatography step accomplished the purification of the products. Finally, the affinity and neutralizing activity was evaluated by ELISA and pseudovirus neutralization assay, and ELISA detected the pharmacokinetic parameters. The results displayed that equine anti-SARS-CoV-2 specific F(ab’)2 has high specificity. Furthermore, PEGylation F(ab’)2-Fab-PEG-Fab had a longer half-life than specific F(ab’)2. The serum half-life of Fab-PEG-Fab, Fab-PEG, and specific F(ab’)2 were 71.41 h, 26.73 h, and 38.32 h, respectively. The half-life of Fab-PEG-Fab was approximately two times as long as the specific F(ab’)2. Thus far, PEGylated F(ab’)2 has been prepared with high safety, high specificity, and a longer half-life, which could be used as a potential treatment for COVID-19.

Over fertilization has resulted in serious soil compaction and acidification, as well as nonpoint pollution. A strategy for variable fertilization according to Site-Specific Management Zones (SSMZ) is urgently needed. We developed an object-oriented method using image segmentation for SSMZ delineation for a farming cooperative founded in 2013 in northeastern China. The method is based on Normalized Difference Vegetation Index (NDVI) during the crop growth period from 2011 to 2016. We analyzed and validated SSMZ results with plant sampling data, NDVI, soil temperature and moisture. The results indicated that (i) SSMZs derived from NDVI and image segmentation enhanced the difference between SSMZs and the homogeneity within SSMZs; (ii) crop dry biomass was consistent with NDVI, and NDVI showed a clear change in the regular SSMZ pattern with terrain variation after 2013; and (iii) SSMZ patterns were mainly affected by different farming practices or by topographic factors before and after the cooperative was founded. This new approach for SSMZ delineation will accelerate the application of precision agriculture in relief areas. If the study results are applied to other areas, the main factors of SSMZ need to be determined because of climate differences as well as specific soil and terrain conditions.

The pollution characteristics of multi-size atmospheric particulates in Xinxiang, which was one of the most polluted cities across China, are still unclear even through air quality in Xinxiang has been improved in recent years. PM10, PM2.5, and PM1 samples were synchronously collected from 21 December 2019 to 17 January 2020 to explore pollution levels and reveal sources of PM in Xinxiang. The average mass concentrations of PM10, PM2.5, and PM1 were as high as 155.53 μg m−3, 120.07 μg m−3, and 85.64 μg m−3 during the observation period, respectively. Almost all of the chemical compositions in PM10, PM2.5 and PM1 increased continuously and obviously with the aggravation of the pollution level. Compared with the clean period, the enhancement of sulfate (23–27%) in PM was obvious higher than nitrate (19–22%) during the pollution period, which demonstrated that sulfate was the main contributor to the high concentration of PM in this study. Similar source distributions for PM10, PM2.5, and PM1 were also found, including traffic source, combustion source, secondary aerosols, industrial source, and fugitive dust, by using the positive matrix factorization (PMF) model. Furthermore, the contributions of the combustion source and secondary aerosol were found to be higher in smaller particles (PM2.5 and PM1), while the contribution of fugitive dust was higher in PM10. Moreover, dust and sand were entrained by air masses from the northwest that increased the contribution of dust in PM at the observation site. The potential source contribution function (PSCF) analysis illustrated that regional emission sources in northern and eastern Xinxiang might be important potential contributors to PM pollution in Xinxiang.

A large number of neuroimaging studies have detected brain abnormalities in first-episode schizophrenia both before and after treatment, but it remains unclear how these abnormalities reflect the effects of antipsychotic treatment on the brain. To summarize the findings in this regard and provide potential directions for future work, we reviewed longitudinal structural and functional imaging studies in patients with first-episode schizophrenia before and after antipsychotic treatment. A total of 36 neuroimaging studies was included, involving 21 structural imaging studies and 15 functional imaging studies. Both anatomical and functional brain changes in patients after treatment were consistently observed in the frontal and temporal lobes, basal ganglia, limbic system and several key components within the default mode network (DMN). Alterations in these regions were affected by factors such as antipsychotic type, course of treatment, and duration of untreated psychosis (DUP). Over all we showed that: (a) The striatum and DMN were core target regions of treatment in schizophrenia, and their changes were related to different antipsychotics; (b) The gray matter of frontal and temporal lobes tended to reduce after long-term treatment; and (c) Longer DUP was accompanied with faster hippocampal atrophy after initial treatment, which was also associated with poorer outcome. These findings are in accordance with previous notions but should be interpreted with caution. Future studies are needed to clarify the effects of different antipsychotics in multiple conditions and to identify imaging or other biomarkers that may predict antipsychotic treatment response. With such progress, it may help choose effective pharmacological interventional strategies for individuals experiencing recent-onset schizophrenia.

DNA nanostructures of increasing complexity have been designed using scaffolded origami or single-stranded tiles and bricks. Here the authors demonstrate the construction of complex structures using a wireframe approach that overcomes limitations of using scaffolding.

Crop biomass is an agricultural indicator of productivity, and knowledge of the temporal and spatial variation in biomass in different topography is critical for the application of precision management techniques. This study integrates microtopography with multitemporal remote sensing observations to reveal biomass- and yield-limiting variables. Three SPOT-6 high spatial resolution images and six topographic variables were combined to model the spatial variation in soybean biomass at multiple stages during the growing season. The results showed the following. (i) The multiple regression model for biomass estimation that combines topographic variables with vegetation indices can achieve higher accuracy than a vegetation index model. (ii) Biomass varied dramatically with topography during the growing season. (iii) Microtopographic variables, such as curvature and slope, had distinctive impacts on crop conditions over a growing season. Early in the growing season, sunny upper slopes produced more biomass than shady lower slopes, whereas this trend reversed over the season. Gentle concave slopes (–1.2 m−1 < curvature < 0 m−1) showed greater productivity later in the season, while slopes with high concavity (curvature < –1.2 m−1) or high convexity (curvature > 0.75 m−1) suppressed crop growth. These conclusions can be used directly to precisely manage nutrients and water applications.

Metal-organic frameworks (MOFs), which have become popular in recent years as excellent carriers of drugs and biomimetic materials, have provided new research ideas for fighting pathogenic bacterial infections. Although various antimicrobial metal ions can be added to MOFs with physical methods, such as impregnation, to inhibit bacterial multiplication, this is inefficient and has many problems, such as an uneven distribution of antimicrobial ions in the MOF and the need for the simultaneous addition of large doses of metal ions. Here, we report on the use of MIL-101(Fe)@Ag with efficient metal-ion release and strong antimicrobial efficiency for co-sterilization. Fe-based MIL-101(Fe) was synthesized, and then Ag+ was uniformly introduced into the MOF by the substitution of Ag+ for Fe3+. Scanning electron microscopy, powder X-ray diffraction (PXRD) Fourier transform infrared spectroscopy, and thermogravimetric analysis were used to investigate the synthesized MIL-101(Fe)@Ag. The characteristic peaks of MIL-101(Fe) and silver ions could be clearly seen in the PXRD pattern. Comparing the diffraction peaks of the simulated PXRD patterns clearly showed that MIL-101(Fe) was successfully constructed and silver ions were successfully loaded into MIL-101(Fe) to synthesize an MOF with a bimetallic structure, that is, the target product MIL-101(Fe)@Ag. The antibacterial mechanism of the MOF material was also investigated. MIL-101(Fe)@Ag exhibited low cytotoxicity, so it has potential applications in the biological field. Overall, MIL-101(Fe)@Ag is an easily fabricated structurally engineered nanocomposite with broad-spectrum bactericidal activity.

The incidence of myocardial infarction (MI) increases every year worldwide. Better diagnostic and prognostic biomarkers for clinical applications are the consistent pursuit of MI research. In addition to electrocardiogram, echocardiography, coronary angiography, etc., circulating biomarkers are essential for the diagnosis, prognosis, and treatment effect monitoring of MI patients. In this review, we assessed both strength and weakness of MI circulating biomarkers including: (1) originated from damaged myocardial tissues including current golden standard cardiac troponin, (2) released from non-myocardial tissues due to MI-induced systems reactions, and (3) preexisted in blood circulation before the occurrence of MI event. We also summarized newly reported MI biomarkers. We proposed that the biomarkers preexisting in blood circulation before MI incidents should be emphasized in research and development for MI prevention in near future.

Nanomaterial technology has attracted much attention because of its antibacterial and drug delivery properties, among other applications. Metal-organic frameworks (MOFs) have advantages, such as their pore structure, large specific surface area, open metal sites, and chemical stability, over other nanomaterials, enabling better drug encapsulation and adsorption. In two examples, we used the common pathogenic bacterium Staphylococcus aureus and highly infectious influenza A virus. A novel complex MIL-101(Fe)-T705 was formed by synthesizing MOF material MIL-101(Fe) with the drug favipiravir (T-705), and a hot solvent synthesis method was applied to investigate the in vitro antibacterial and antiviral activities. The results showed that MIL-101(Fe)-T705 combined the advantages of nanomaterials and drugs and could inhibit the growth of Staphylococcus aureus at a concentration of 0.0032 g/mL. Regarding the inhibition of influenza A virus, MIL-101(Fe)-T705 showed good biosafety at 12, 24, 48, and 72 h in addition to a good antiviral effect at concentrations of 0.1, 0.2, 0.4, 0.8, 1.6, and 3 μg/mL, which were higher than MIL-101(Fe) and T-705.

The root tissues play important roles in water and nutrient acquisition, environmental adaptation, and plant development. In this study, a diversity panel of 388 wheat accessions was collected to investigate nine root system architecture (RSA) traits at the three-leaf stage under two growing environments: outdoor pot culture (OPC) and indoor pot culture (IPC). Phenotypic analysis revealed that root development was faster under OPC than that under IPC and a significant correlation was observed between the nine RSA traits. The 660K single-nucleotide polymorphism (SNP) chip was used for a genome-wide association study (GWAS). Significant SNPs with a threshold of −log10 (p-value) ≥ 4 were considered. Thus, 36 quantitative trait loci (QTLs), including 13 QTL clusters that were associated with more than one trait, were detected, and 31 QTLs were first identified. The QTL clusters on chromosomes 3D and 5B were associated with four and five RSA traits, respectively. Two candidate genes, TraesCS2A01G516200 and TraesCS7B01G036900, were found to be associated with more than one RSA trait using haplotype analysis, and preferentially expressed in the root tissues. These favourable alleles for RSA traits identified in this study may be useful to optimise the root system in wheat.

Both the chemical reaction mechanism and rate can largely explain the formation mechanism of the by-products of SF6. By understanding this mechanism, we can develop and improve models of the chemical kinetics of SF6 decomposition under discharge. Using quantum chemistry, this study compares the reaction mechanisms and rates of SF6−, SF5−, SF4−, and SF3− and H2O under discharge at 298 K–12 000 K and reveals the formation mechanism of their anionic by-products, i.e., SOF4−, SOF3−, SOF2−, and SOF−. The key parameters such as the reaction equation and reaction rate to improve the chemical kinetic models under partial and arc discharges were then obtained. In this study, the structural optimizations, vibrational frequencies, and zero-point energies of the reactants, products, complexes, intermediates, and transition states were calculated at the B3LYP/6-311G(d,p) level. The single-point energies of all species were calculated at the CCSD(T)/aug-cc-PVTZ level. The strengths and sites of weak interactions were determined from the electrostatic potential of the molecular surface, and the reaction rates were obtained using transition state theory. It has been found that SF6−, SF5−, SF4−, and SF3− combined with H2O to form weak-interaction complexes dominated by hydrogen bonding, thus providing the initial conditions for R1, R2, R3, and R4, respectively. All four reactions were composed of multiple elementary reactions with the first step being the rate-determining step. Moreover, compared to their corresponding reactions of SF5, SF4, SF3, and SF2 with H2O, they achieved lower potential energy barriers and higher reaction rates. Note that the reaction rates decreased in the following order: R3 > R2 > R1 > R4.

Human herpesvirus 6 (HHV-6) is an important immunosuppressive and immunomodulatory virus worldwide. However, whether and how HHV-6 infection influences the metabolic machinery of the host cell to provide the energy and biosynthetic resources for virus propagation remains unknown. In this study, we identified that HHV-6A infection promotes glucose metabolism in infected T cells, resulting in elevated glycolytic activity with an increase of glucose uptake, glucose consumption and lactate secretion. Furthermore, we explored the mechanisms involved in HHV-6A-mediated glycolytic activation in the infected T cells. We found increased expressions of the key glucose transporters and glycolytic enzymes in HHV-6A-infected T cells. In addition, HHV-6A infection dramatically activated AKT-mTORC1 signaling in the infected T cells and pharmacological inhibition of mTORC1 blocked HHV-6A-mediated glycolytic activation. We also found that direct inhibition of glycolysis by 2-Deoxy-D-glucose (2-DG) or inhibition of mTORC1 activity in HHV-6A-infected T cells effectively reduced HHV-6 DNA replication, protein synthesis and virion production. These results not only reveal the mechanism of how HHV-6 infection affects host cell metabolism, but also suggest that targeting the metabolic pathway could be a new avenue for HHV-6 therapy.

Freezing of gait (FOG) is a disabling phenomenon that is described by patients with Parkinson’s disease (PD). Not only gait may be involved in the freezing phenomenon, but also some nonmotor symptoms, such as freezing of urination (FOU) in this case. The characters of urinary dysfunctions in this case resemble “off” freezing: (1) abrupt difficulty in starting or continuing in urination; (2) the urinary dysfunctions fluctuated with medication state; and (3) the urinary dysfunctions could be alleviated dramatically by an external cueing. Urinary dysfunctions in this patient (and maybe more PD patients) are associated not only with the classical “nonmotor symptoms” but also the freezing phenomenon. FOU could be a part of the spectrum of freezing symptoms. The subtypes of the freezing phenomenon will shed light on the PD pathophysiology and clinical treatment.

Abstract Lake anchovy (Coilia ectenes taihuensis) is a sedentary, dominant fish species that forms an unmanaged fishery in Taihu Lake, eastern China. The environment and developmental stage of lake anchovy are likely important drivers of their gut microbiome, which is linked to host health and development. To investigate the relationship between the gut microbiome and three defined factors (fish sex, fish body size, and the local habitat), high‐throughput sequencing of the 16S ribosomal RNA gene was used to study the microorganisms of 184 fish samples and four water samples collected in Taihu Lake. Four dominant bacterial phyla (Proteobacteria, Firmicutes, Planctomycetes, and Cyanobacteria) were present in all fish samples. We compared the microbial communities of males and females and found that the relative abundance of Corynebacteriaceae was significantly higher in males than in females, while the opposite trend was detected for Sphingomonadaceae. We also discovered that the relative abundance of Firmicutes was positively correlated with fish body size and that the proportions of Proteobacteria and Tenericutes were lower in larger fish than in fish of other sizes. Finally, we found that the difference in microbial richness between eastern and northern Taihu Lake was the most marked. Lake anchovy was rich in Lactobacillus and Clostridium in the eastern site, while those in the northern site had the highest abundance of Sphingomonas and Methylobacterium, suggesting that the local habitat may also influence the intestinal microbiome. These findings will not only help researchers understand the community composition of the intestinal microflora of lake anchovy but also contribute to the protection of fish resources in Lake Taihu and the sustainable use of lake anchovy.

SF(6) has been widely used as an insulating and arc extinguishing medium in electric power equipments. Detection of gas decomposition products is an important means for condition monitoring and fault diagnosis. Trace H(2)O and O(2) have a significant influence on the species and concentrations of SF(6) decomposition products. It is important to explore the influence mechanism of trace H(2)O and O(2) on SF(6) decomposition products under arcing conditions. A series of SF(6) decomposition tests were carried out on a 40.5kV SF(6) circuit breaker, and a gas chromatograph and an electrochemical sensor were used to measure decomposed components. First, the H(2)O content remained constant, and the O(2) content and arcing energy changed in the tests. It has been found that the decomposition products of SF(6) are SOF(2), SO(2), CO, CO(2), CF(4), C(2)F(6) and C(3)F(8). Then, the O(2) content remained constant, and the H(2)O content changed. The decomposition products are mainly SOF(2), SO(2), CO and CO(2). Concentrations of all these decomposition products increase first and then tend to be stable slowly over time, but change differently with the increase of H(2)O and O(2) contents, because H(2)O and O(2) play different roles in the formation process of each decomposition characteristic components.

Objective: It has been established that reduced vitamin B12 serum levels are associated with cognitive decline and mental illness. The chronic use of methamphetamine (MA), which is a highly addictive drug, can induce cognitive impairment and psychopathological symptoms. There are few studies addressing the association of MA with vitamin B12 serum levels. This study examined whether the serum levels of B12 are associated with MA addiction.Methods: Serum vitamin B12, homocysteine (Hcy), glucose and triglyceride concentrations were measured in 123 MA addicts and 108 controls. In addition, data were collected on their age, marital status, level of education and Body Mass Index (BMI) for all participants. In the patient group, the data for each subject were collected using the Fagerstrom Test for Nicotine Dependence (FTND), the Alcohol Use Disorders Identification Test (AUDIT), and a drug use history, which included the age of onset, total duration of MA use, the number of relapses and addiction severity.Results: Our results showed that MA addicts had lower vitamin B12 levels (p < 0.05) than those of healthy controls, but Hcy levels were not significantly different between the two groups (p > 0.05). Serum B12 levels were negatively correlated with the number of relapses in the MA group. Furthermore, binary logistics regression analysis indicated that the B12 was an influencing factor contributing to addiction severity.Conclusion: The findings of this study suggest that some MA addicts might have vitamin B12 deficiency, and serum B12 levels may be involved in the prognosis of MA addiction.

Glycerol-3-phosphate acyltransferases (GPATs) play an important role in glycerolipid biosynthesis, and are mainly involved in oil production, flower development, and stress response. However, their roles in regulating plant height remain unreported. Here, we report that Arabidopsis GPAT1 is involved in the regulation of plant height. GUS assay and qRT-PCR analysis in Arabidopsis showed that GPAT1 is highly expressed in flowers, siliques, and seeds. A loss of function mutation in GPAT1 was shown to decrease seed yield but increase plant height through enhanced cell length. Transcriptomic and qRT-PCR data revealed that the expression levels of genes related to gibberellin (GA) biosynthesis and signaling, as well as those of cell wall organization and biogenesis, were significantly upregulated. These led to cell length elongation, and thus, an increase in plant height. Together, our data suggest that knockout of GPAT1 impairs glycerolipid metabolism in Arabidopsis, leading to reduced seed yield, but promotes the biosynthesis of GA, which ultimately enhances plant height. This study provides new evidence on the interplay between lipid and hormone metabolism in the regulation of plant height.

The research on decomposition characteristics of SF6 and its by-products have great significance to the operation, maintenance, condition assessment and fault diagnosis of power equipment. In this paper, the particle composition models of SF6, SF6/polytetrafluoroethylene (PTFE), SF6/PTFE/O2, SF6/PTFE/H2O, and SF6/PTFE/O2/H2O were established by using Gibbs free energy minimization method, and the effects of trace H2O and O2 impurities and PTFE vapour on SF6 by-products were studied by the models. In order to verify the correctness of the simulation results, a series of breaking experiments were carried out on a 40.5 kV SF6 circuit breaker, and a gas chromatograph was used to detect and analyse the SF6 by-products. It was found that when PTFE vapour is involved in the arc plasma, the main by-product after arc quenching is CF4, and the molar fractions of C2F6 and C3F8 are very low. When O2 is involved, the main by-products are SOF2, SO2 and SO2F2, and a small amount of CO and CO2 was also produced. When H2O is involved, the main by-products in simulation are SOF2, SO2 and HF, and a small amount of SO2, CO2, CO, SO2F2 and H2 was also produced. The experimental results are in good agreement with the above results.

The redistribution of solar radiation, temperature, soil moisture and heat by topography affects the physical and chemical properties of the soil and the spatial distribution characteristics of crop growth. Analyses of the relationship between topography and these variables may help to improve the accuracy of digital elevation models (DEMs). The purpose of correcting Shuttle Radar Topography Mission (SRTM) data is to obtain high-precision DEM data in cultivated land. A typical black soil area was studied. A high-precision reference DEM was generated from an unmanned aerial vehicle (UAV) and extensive measured ground elevation data. The normalized differential vegetation index (NDVI), perpendicular drought index (PDI) extracted from SPOT-6 remote sensing images and potential solar radiation (PSR) extracted from SRTM. The interactions between topography and NDVI, PDI, and PSR were analyzed. The NDVI, PDI and PSR in June, July, August and September of 2016 and the SRTM were used as independent variables, and the UAV DEM was used as the dependent variable. Linear stepwise regression (LSR) and a back-propagation neural network (BPNN) were used to establish an elevation prediction model. The results indicated that (1) The correlation between topography and NDVI, PSR, PDI was significant at 0.01 level. The PDI and PSR improved the spatial resolution of SRTM data and reduce the vertical error. (2) The BPNN (R21 = 0.98, root mean square error, RMSE1 = 0.54) yielded a higher SRTM accuracy than did the studied linear model (RMSE1 = 1.00, R21 = 0.90). (3) A series of significant improvements in the SRTM were observed when assessed with the reference DEMs for two different areas, with RMSE reductions of 91% (from 14.95 m to 1.23 m) and 93% (from 15.6 m to 0.94 m). The proposed method improved the accuracy of existing DEMs and could provide support for accurate farmland management.

Jumonji domain-containing protein 6 (JMJD6) is a homolog of hypoxia-inducible factor (HIF) asparaginyl hydroxylase, an inhibitor of HIF. HIF-1α is known to participate in neuropathic pain (NPP) during chronic constriction injury (CCI); however, the roles of JMJD6 in NPP have not been systematically investigated. In this study, we examined the temporal distribution and cellular location of JMJD6 in the spinal cord during CCI. In addition, we assessed behavioral changes representative of NPP in rats. Following CCI, lentiviral vectors (LV-JMJD6) were intrathecally administered to observe the changes in the expression of JMJD6, HIF-1α, and its downstream factor caspase-3. Co-immunoprecipitation was used to detect potential interactions between JMJD6 and HIF-1α. We found that JMJD6 was decreased in rats following CCI, which was accompanied by significant NPP–associated behavioral changes. JMJD6 was mainly expressed in neurons. Intrathecal injection of LV-JMJD6 following CCI alleviated the thermal and mechanical hyperalgesia, normalized JMJD6 protein expression, and decreased HIF-1α protein expression with a corresponding reduction in caspase-3 protein expression. Furthermore, the co-immunoprecipitation analyses showed that JMJD6 and HIF-1α protein immunoprecipitated with each other, indicating an interaction between these two proteins. Taken together, the results suggest that JMJD6 may serve as a sensor in neurons of the adult rat spinal cord during the CCI state. Furthermore, JMJD6 may exert its function in NPP by regulating HIF-1α in rats exposed to CCI.

Background/Aims: Pulmonary arterial endothelial plexiform lesions are a basic pathological change associated with pulmonary vascular remodeling and are characterized by the formation of tumorlets as a result of over-growth of endothelial cells. Accumulating evidence suggests that platelet-derived growth factor (PDGF) participates in regulating the progression of pulmonary arterial hypertension. However, whether PDGF promotes the survival of pulmonary arterial endothelial cells (PAECs), as well as the specific molecular mechanisms that underlie its actions, remains unknown. Methods: MTT assays, caspase-3 and caspase-9 activity assays and western blot analysis were performed. Results: We found that both the mRNA and protein expression of PDGF-B was induced by hypoxia and that the inhibitory effects exerted by hypoxia on apoptosis were attenuated by inhibitors of PDGF beta. Moreover, PDGF-B inhibited apoptosis in a dose-dependent manner by stimulating the phosphorylation of both Akt and Stat3, and the PI3K/AKT pathway serves as an up-stream participant in the Stat3 activation stimulated by PDGF-B. Additionally, the anti-apoptotic effects of PDGF-B were abolished when PAECs were treated with either an inhibitor or small interfering RNA targeting Stat3. Conclusions: These observations suggest that PDGF-B is induced by hypoxia and protects against apoptosis via the PI3K/Akt/Stat3 signaling pathway.

An innovative array of magnetic coils (the discrete Rogowski coil—RC) with the advantages of flexible structure, miniaturization and mass producibility is investigated. First, the mutual inductance between the discrete RC and circular and rectangular conductors are calculated using the magnetic vector potential (MVP) method. The results are found to be consistent with those calculated using the finite element method, but the MVP method is simpler and more practical. Then, the influence of conductor section parameters, inclination, and eccentricity on the accuracy of the discrete RC is calculated to provide a reference. Studying the influence of an external current on the discrete RC’s interference error reveals optimal values for length, winding density, and position arrangement of the solenoids. It has also found that eccentricity and interference errors decreasing with increasing number of solenoids. Finally, a discrete RC prototype is devised and manufactured. The experimental results show consistent output characteristics, with the calculated sensitivity and mutual inductance of the discrete RC being very close to the experimental results. The influence of an external conductor on the measurement of the discrete RC is analyzed experimentally, and the results show that interference from an external current decreases with increasing distance between the external and measured conductors.

For nonstationary time series consisting of multiple time-varying frequency (TVF) components where the frequency of components overlaps in time, classical linear filters fail to extract components. The G-filter based on time deformation has been developed to extract components of multicomponent G-stationary processes. In this paper, we explore the wide application of the G-filter for filtering different types of nonstationary processes with multiple time-frequency structure. Simulation examples illustrate that the G-filter can be applied to filter a broad range of multicomponent nonstationary process where TVF components may in fact overlap in time.

FILIA is a member of the recently identified oocyte/embryo expressed gene family in eutherian mammals, which is characterized by containing an N-terminal atypical KH domain. Here we report the structure of the N-terminal fragment of FILIA (FILIA-N), which represents the first reported three-dimensional structure of a KH domain in the oocyte/embryo expressed gene family of proteins. The structure of FILIA-N revealed a unique N-terminal extension beyond the canonical KH region, which plays important roles in interaction with RNA. By co-incubation with the lysates of mice ovaries, FILIA and FILIA-N could sequester specific RNA components, supporting the critical roles of FILIA in regulation of RNA transcripts during mouse oogenesis and early embryogenesis.

The classical linear filter can successfully filter the components from a time series for which the frequency content does not change with time, and those nonstationary time series with time-varying frequency (TVF) components that do not overlap. However, for many types of nonstationary time series, the TVF components often overlap in time. In such a situation, the classical linear filtering method fails to extract components from the original process. In this paper, we introduce and theoretically develop the G-filter based on a time-deformation technique. Simulation examples and a real bat echolocation example illustrate that the G-filter can successfully filter a G-stationary process whose TVF components overlap with time.
